体外サイトカイン濾過、組み換えヒト活性化プロテインC、コルチコステロイド、ヒト組み換えラクトフェリン、および免疫調節など、敗血症の実験的補助治療を評価する多くの研究が行われてきた。免疫調節は広く予測されていたが、患者母集団の異質性および敗血症の病原性の複雑さによって、これらの実験的手法の進歩が限定された。インターロイキン−1(IL−1)または腫瘍壊死因子−α(TNF−α)など単一のメディエーターのブロックにより、敗血症生存率の改善の見込みはほとんど示されなかった。サイトカイン除去は、動物試験において有望な結果を示した。しかし、様々な結果は、サイトカイン除去の直接的影響ではなく他の下流機構を調節する結果であると考えられる。エンドトキシンのクリアランスの利点は、動物試験においても有効性を示したが、敗血症患者の転帰の改善はまだ実証されていない。

細菌を捕捉および除去するための抗生物質で官能化されたマイクロ流体デバイス
本実施例において、流れている流体から細菌を単離するデバイスおよび方法を開発した。細菌で汚染された試料を、1つまたは複数の流路から構成されるマイクロ流体デバイス(図1Aおよび1B)の少なくとも1つの入口に導入した。細菌に特異的に結合し、高親和性で流れている流体から捕捉する生体適合性リガンドであるコリスチン−PEG 1000−シラン(Col−PEG−Si)(図2A、2B、および2C)で、流路壁を連続的に官能化した。流路はそれぞれ、慣性力に影響を与えるように設計されたアスペクト比および湾曲を有し、その慣性力によって、細菌細胞および細菌サイズの粒子がマイクロ流路の壁に沿って押しやられた(図3)。これは、官能化された流路壁に沿って細菌を捕捉することになる(図4A、4B、および4C)。血球を含めて、他の細胞型はすべて、細菌標的化リガンドによって捕捉されず、流路の第2の部分に沿って自由に流された。これによって、汚染されていない流体が流路出口(単数または複数)を通って出ていった。マイクロ流路内のコリスチン表面提示の特徴付けを行い、その結果から、細菌とCol−PEG−Siとの急速な結合が明らかになった。
血液で運ばれる病原体を急速検出および捕捉するための慣性ベースの流体プラットフォーム
慣性ベースの流体装置を使用した病原体およびエンドトキシンの体外抽出を、敗血症に関連した罹患率と死亡率を低減する次世代技術として使用することができる。
NHS−PEG−シラン(N−ヒドロキシスクシンイミド−ポリエチレングリコール1000−シラン、Nanocs,Inc.、ロット#160429)(30mg/mL)を、エタノール/水、pH5(50/50v/v%、酢酸でpH調整)に溶解した。35mg/mLのコリスチン硫酸塩(Sigma−Aldrich、ロット#SLBN5158V)を、NHS−PEG−シラン溶液に添加した。溶液を30秒間ボルテックスし、次いで21℃で2時間反応させた。NHSを使用して、ヘテロ二官能性NHS−PEG−シランリンカーをコリスチンの同様な反応性を示す5つのL−α−ジアミノ酪酸(Dab)残基の1つにカップリングさせ、コリスチン−PEG−シランを得た。
PEG−シラン(ポリエチレングリコール1000−シラン、Nanocs,Inc.、ロット#160706OH)(30mg/mL)を、エタノール/水、pH5(50/50v/v%、酢酸でpH調整)に溶解した。溶液を30秒間ボルテックスし、次いで21℃で2時間インキュベートさせた。
マイクロ流体デバイスパターンを設計し、AutoCADソフトウェア(AutoCAD 2014、AutoDesk,Inc.)で描いた。設計は、1つの入口および1つの出口を設けた6ループ二重螺旋状マイクロ流路からなるものであった。マイクロ流路は、6ループについて時計回りに回転し、S字ジャンクションを通って方向を変え、次いで反時計回りに回転して、二重螺旋を形成する。二重螺旋状マイクロ流路は、406mm(L)、300μm(W)、15μm(H)の寸法である。隣接している2つのループ間の間隔は500μmである。湾曲の最外半径は9.8mmである。標準フォトリソグラフィー技法を使用して、SU8−2010(MicroChem Corp.)層(WS−400 Lite Series Spin Processor、Laurell Technologies Corp.)で4インチのシリコンウェハ(Nova Electronic Materials)にスピンコーティングしたシリコンマスターから鋳型を生成した。95℃で5分間ソフトベーキングした後、マスクアライナー(MJB 3 Mask Aligner、Suss MicroTech)をUV光(Novacure 2100、Exfo Inc.)およびネガ型フォトマスク(Infinite Graphics,Inc.)と共に使用して、SU−8層をパターン形成した。続いて、95℃で5分間の後露光ベーキングステップの後、SU−8現像薬(MicroChem Corp.)を使用して、得られたウェハを現像した。最終のハードベーキングを150℃で5分間行った。ウェハをマスター鋳型として使用して、マイクロ流体流路を注入成形した。脱気したPDMS(ポリジメチルシロキサン、PDMSベースと硬化剤を10:1の比で混合、Sylgard 184、Dow Corning Inc.)を鋳型に注入成形し、65℃で4時間ベーキングした。流路が埋め込まれたPDMSを、続いて安全かみそりの刃で切断し、マスター鋳型から取り出した。1.5mmの生検用パンチ(Integra Miltex)を使用して、PDMSに1つの入口および1つの出口をあけた。次いで、PDMSスラブを、酸素プラズマ処理(PDC−001 Plasma Cleaner、Harrick Plasma)後のガラス基板(43mm×50mm、Ted Pella,Inc.)に結合させた。プラズマ処理およびガラス製カバースリップへの結合の直後に、外径1/16インチのタイゴンマイクロボアチュービング(Cole Parmer Corp.)を、入口ポートと出口ポートとから挿入した。コリスチン−PEG−シランリンカーおよびシラン化学的性状を使用して、コリスチンを二重螺旋状マイクロ流体デバイス流路壁に繋ぎ止めた(図19)。1mLのルアーロック式使い捨てシリンジ(Becton Dickinson)を使用して、コリスチン−PEG−シラン溶液を、酸化PDMSマイクロ流路に流した。十分に確立したシラン処理の原理に従って、コリスチン−PEG−シラン溶液とマイクロ流路表面を65℃で30分間接触させた。ヘテロ二官能性のコリスチン−PEG−シランリンカーの使用は、細菌およびエンドトキシンとのコリスチン相互作用の立体障害を低減することを意図したPEG分子スペーサーを含むマイクロ流路壁のコリスチン修飾を可能にするように設計された。生理学的なpH値の範囲において、コリスチンの第一級アミン基はプロトン化され、したがって正に荷電された。コリスチンの正電荷によって、グラム陰性病原体およびエンドトキシンの負に荷電された外膜への結合が可能になる。まさに同様の手法を使用して、PEG−シランで官能化されたデバイスも合成し、コリスチン修飾を欠いたPEG化対照群として使用した。リガンドを添加せずに、官能化されていない二重螺旋状マイクロ流体デバイスを得た。
コリスチン化二重螺旋状マイクロ流体デバイス内のコリスチンの蛍光標識は、ATTO 488 NHSエステル(Sigma−Aldrich、ロット#BCBQ4012V)を使用して行った。ATTO 488 NHSエステル(2mg/mL)を、使用直前にジメチルスルホキシド(DMSO、Sigma−Aldrich)に溶解した。その蛍光剤をコリスチン化マイクロ流路に添加し、続いて室温で30分間インキュベートすることによって標識した。次いで、染色されたマイクロ流路をリン酸緩衝生理食塩水(PBS、Gibco、ロット#1806048)、pH7.4で洗浄した。同じプロトコールを、対照のPEG化マイクロ流路に対して使用した。次いで、蛍光染色したマイクロ流路を、蛍光顕微鏡(EVOS FL、Invitrogen)を使用して画像処理した。Image−Jソフトウェアを使用して、捕捉された画像の流路幅にわたって蛍光強度を定量化した。官能化されていないマイクロ流体デバイス内のATTO 488 NHSエステルの希釈液を参照として使用して、ATTO 488 NHSエステル標準蛍光較正曲線を作成した。コリスチン化マイクロ流路およびPEG化マイクロ流路の蛍光強度を、ATTO 488 NHSエステル標準曲線の蛍光強度と比較し、二重螺旋状マイクロ流路1つ当たりのコリスチン分子の数を定量化することが可能になった。
コリスチン−PEG−シランリガンドが流れ条件下で二重螺旋状マイクロ流体デバイスの表面から解体する可能性を、MaxSignal(登録商標)コリスチン酵素結合性免疫吸着アッセイ(ELISA)試験キット(BIOO Scientific、ロット#109501081415$626311708249)を使用して評価した。PBS、pH7.4を、0.2mL/分でコリスチン化二重螺旋状マイクロ流体デバイスに2時間流した。試料をデバイス出口から様々な時点(1、2、4、5、10、20、30、45、60、120分)で回収し、分析した。各試料中の全コリスチン含有量を、コリスチンELISAにより製造業者の指示書に従って定量化した。マイクロタイタープレートリーダーを使用して、吸光度を450nmで測定し、これらの値を使用して、試料中のコリスチン濃度を較正曲線に従って算出した(アッセイ検出範囲、0.5〜50ng/ml)。コリスチンリガンド保持の特徴付けの結果によって、二重螺旋状マイクロ流体デバイスをそれぞれ、実験で使用する前に70%エタノールで連続的に30分間洗浄することが促され、捕捉調査時においてコリスチンの遊離は無視できるほどであった。
細菌培養物(すなわち、アシネトバクター・バウマニ(Acinetobacter baumannii) ATCC17978、コリスチン耐性アシネトバクター・バウマニ(Colistin-resistant Acinetobacter baumannii) 19606R41、コリスチン耐性アシネトバクター・バウマニ(Colistin-resistant Acinetobacter baumannii) 「患者2」、肺炎桿菌(Klebsiella pneumoniae) ATCC700603、枯草菌(Bacillus subtilis) 1A578 (Bacillus Genetic Stock Center)、黄色ブドウ球菌(Staphylococcus aureus) ATCC29213)をLB寒天(Sigma−Aldrich、ロット#SLBR1403V)に蒔き、37℃で終夜増殖させた。次いで、各株の単一コロニーをLB培地(1倍)(Gibco、ロット#1803272)に播種し、150rpmで振動させて37℃で18時間増殖させた。細胞濃度をOD595分光測定(Infinite F500、Tecan)で測定した。
平均直径10.2μm±4μmおよび2μm±0.3μmのポリスチレンマイクロスフェア(Spherotech Inc.)を使用して、マイクロ流路内の粒子集約を分析した。1vol%のTween−20界面活性剤(Sigma−Aldrich)を含む1w/v%の懸濁液を用意して、粒子凝集を防止した。脱イオン(DI)水を使用して、等体積の懸濁液2つを0.1w/v%に希釈した。1mLのルアーロック式チップシリンジおよび外径1/16インチのタイゴン入口チュービングを通ってマイクロ流路の入口ポートに連結された注入シリンジポンプ(Nexus 3000、Chemyx Inc.)を使用して、蛍光粒子を個別に、官能化されていない二重螺旋状マイクロ流路に0.2mL/分で導入した。蛍光顕微鏡(EVOS FL、Invitrogen)を使用して、二重螺旋状流路内の粒子の軌道および集約をリアルタイムで可視化した。この方法は、SYTO(登録商標)9緑色蛍光核酸標識(DMSO中5mM溶液、分子プローブ、ロット#1687876A)A.バウマニ(A. baumannii) ATCC17978を使用して繰り返された。SYTO(登録商標)9を用いて5分間インキュベートした後、細菌を、RNase不含超純水(USB Corp.、ロット#4235512)で3回洗浄し、約107コロニー形成単位(CFU)/mLの濃度に再懸濁した。注入シリンジポンプを使用して、蛍光標識された細菌懸濁液を0.2mL/分でPEG化二重螺旋状マイクロ流体デバイスに流した。蛍光顕微鏡(EVOS FL、Invitrogen)を使用して、二重螺旋状流路内の細菌の軌道および集約をリアルタイムで可視化した。
細菌株のコリスチン最小発育阻止濃度(MIC)を標準寒天平板希釈法で確認した。臨床検査標準協会の感受性限界点に従って、結果を解釈した。
「慣性集約の可視化」方法の節で既に記載したように、A.バウマニ(A. baumannii) ATCC17978をSYTO(登録商標)9で標識した。注入シリンジポンプを使用して、蛍光標識された細菌懸濁液を、0.2mL/分でコリスチン化およびPEG化された二重螺旋状マイクロ流体デバイスに流した。次いで、デバイスをPBS、pH7.4で4回洗浄して、弱く結合しているいずれの細菌も除去した。蛍光顕微鏡(EVOS FL、Invitrogen)を使用して、マイクロ流路の蛍光画像を得た。この方法は、EDTA抗凝固処理されたヒト全血に添加された蛍光標識されたA.バウマニ(A. baumannii) ATCC17978および黄色ブドウ球菌(S. aureus) ATCC29213を使用して繰り返された。バンダービルト大学被験者(Vanderbilt University Human Subjects)施設内倫理委員会(IRB)(プロトコール番号111251)に従って十分な説明に基づく同意を得て、健常ドナーからヒト血液を得た。
コリスチン化およびPEG化された二重螺旋状マイクロ流体デバイスの病原体捕捉能力の試験は、まず、A.バウマニ(A. baumannii) ATCC17978細菌懸濁液をPBS、pH7.4中約8×107CFU/mLに調整することによって実施した。対照として働く最初の懸濁液の系列希釈により、約102CFU/mLの濃度を得た。細菌試料をコリスチン化二重螺旋状マイクロ流体デバイスに0.2ml/分で注入し、試料をデバイス出口から異なる5つの時点(1、2、3、4および5分)で回収した。この方法を、PEG化二重螺旋状マイクロ流体デバイスでも繰り返した。二重螺旋状マイクロ流体デバイスの出口から回収された溶液を対照群と同じように希釈した。病原体捕捉能力を定量化するために、回収された試料および対照をLB寒天ペトリ皿に3回繰り返して蒔いた(100μL)。プレートを37℃で24時間インキュベートし、ペトリ皿に形成されたコロニーを計数した。最初の細菌および回収された細菌の数は、複製物から計数された平均コロニー数にそれらのそれぞれの希釈因子を掛けることによって算出した。回収された細菌の数を最初の細菌数(すなわち、無処置対照群)から差し引くことによって、捕捉された細菌の数を決定した。捕捉された細胞の数を最初の細胞の数で割ることによって、捕捉能力を算出した。PBS、pH7.4中の以下の細菌株の捕捉能力を定量化した:A.バウマニ(A. baumannii) ATCC17978、コリスチン耐性A.バウマニ(colistin-resistant A. baumannii) 19606R、コリスチン耐性A.バウマニ(colistin-resistant A. baumannii) 「患者2」、肺炎桿菌(K. pneumoniae) ATCC700603、および枯草菌(B. subtilis) 1A578。この方法を、EDTA抗凝固処理されたヒト全血に添加された蛍光標識されたA.バウマニ(A. baumannii) ATCC17978でも繰り返した。
Pierce LAL発色性エンドトキシン定量化キット(Thermo Scientific、ロット#RG236327)を製造業者のプロトコールに従って使用して、コリスチン化およびPEG化された二重螺旋状マイクロ流体デバイスのエンドトキシン捕捉能力をそれぞれ定量化した。簡潔に言えば、大腸菌(E. coli)エンドトキシン標準品(011:B4)をエンドトキシン不含水中に1EU/mL(エンドトキシン単位)の濃度で復元し、注入シリンジポンプを使用して、その溶液を0.2ml/分でコリスチン化二重螺旋状マイクロ流体デバイスに流した。試料をデバイス出口から異なる5つの時点(1、2、3、4および5分)で回収した。この方法を、PEG化二重螺旋状流体デバイスで繰り返した。次いで、回収された試料を、製造業者のプロトコールに従ってエンドトキシン濃度について分析し、450nmにおいて結果を読み取った。
二重螺旋状マイクロ流体デバイスでの処置が血液学的パラメータを有意に変化させるかどうか決定するために、1mLのEDTA抗凝固処理されたヒト全血を、0.2ml/分で血液浄化デバイスに流した後、コリスチン化デバイス出口から回収した。この方法を、PEG化デバイスで繰り返した。次いで、赤血球溶解を分析して、血漿中における遊離ヘモグロビンの放出を決定した。血漿ヘモグロビンを、十分に確立したプロトコールに従って測定した。デバイスに通した後、血液試料を遠心し(500×g)、Triton−X陽性対照に対する溶血(%)を決定するために、プレートリーダー(Tecan、Infinite M1000 Pro)を使用して、上澄液をヘモグロビン放出について451nmで分光光度分析した。Triton X−100(Sigma−Aldrich)を脱イオンH2Oに希釈して、20v/v%のTriton−X洗浄剤濃度が得られ、それは、陽性対照として働いた。使用された陰性対照は、二重螺旋状マイクロ流体デバイスに流さなかった(すなわち、無処置の)EDTA抗凝固処理されたヒト全血であった。ヘモグロビン放出(%)を次式に従って算出した:ヘモグロビン放出(%)=[(試料451nm−陰性対照451nm)/(陽性対照451nm−陰性対照451nm)]×100%。各試料(すなわち、無処置、コリスチン化デバイス、PEG化デバイス)の全血球計算をバンダービルト大学トランスレーショナルパソロジー共有資源コア(Vanderbilt University Translational Pathology Shared Resource Core)によって行った。
既に記載した方法を使用して、最初の二重螺旋状マイクロ流体デバイスのより大きいバージョンを製作および官能化した。デバイスのスケールアップしたバージョンは、1つの入口および1つの出口を設けた4ループ二重螺旋状マイクロ流路からなるものであった。スケールアップしたバージョンの寸法は、長さ(L)323mm、幅(W)750μm、高さ(H)15μmであった。隣接している2つのループ間の間隔は500μmであり、湾曲の最外半径は11.2mmであった。スケールアップしたデバイスの寸法を、容積対表面積の比が最初のデバイス設計の比に匹敵するように調整した。スケールアップしたデバイスの細菌捕捉能力は、捕捉能力を小さい方のデバイスの捕捉能力と比較するように特徴付けられた。スケールアップしたデバイスを使用した「細菌捕捉の定量化」アッセイを、A.バウマニ(A. baumannii) ATCC17978で行った。
測定値はすべて、エラーバーが指し示すように少なくとも3通りの試料の平均値(average mean)±標準偏差(SD)として報告されている。有意差は、P値<0.01によって定義されるように、一元配置ANOVAと、その後に続けてポストホックテューキーの多重比較検定を使用して決定した。
マイクロ流路壁へのコリスチンコンジュゲーション
蛍光標識および化学量論的分析を使用して、1.76μgのコリスチンがコリスチン化二重螺旋状デバイスの壁に沿って官能化されたと推定された(図20)。このデバイスの表面に結合しているコリスチンの量はヒトに無毒である。ELISAを行って、二重螺旋状マイクロ流体デバイスから放出されたコリスチン含有分子の量を推定した。2時間の連続流れによって、流路から約300ngのコリスチンが放出された(図21)。
官能化されていない二重螺旋状マイクロ流体流路内で連続流れ条件下における蛍光ポリスチレンマイクロ粒子を画像処理した。体積流量0.2ml/分では、対応するRe=14.9の場合、2μmサイズの粒子はマイクロ流路の内壁の近くに集約され(図14a)、10.2μmサイズの粒子はマイクロ流路幅の中心の近くに集約された(図14b)。混成の蛍光画像は、10.2μm(赤色流)および2μm(緑色流)の粒子の軌道を示し、大きい方の粒子は、より大きい血球(すなわち、赤血球および白血球)の軌道に近づく。マイクロ粒子は、直径に応じて平衡位置で慣性集約した。細菌サイズの粒子が血球サイズの粒子からはっきり分離することは明白であった。A.バウマニ(A. baumannii) ATCC17978は、直径2μmの球状粒子より流路壁にはるかに近くに慣性集約した。さらに、細菌は、二重螺旋状デバイスの両方の側部表面の近くに現れた。これらの挙動は、正味の慣性揚力(FL)およびディーン抗力(FD)から誘導される慣性設計と一致したものであった。
コリスチン化およびPEG化された二重螺旋マイクロ流体デバイスの細菌捕捉能力を定性的および定量的に評価するための標準供試体として、A.バウマニ(A. baumannii) ATCC17978細菌細胞を選択した。緑色の蛍光標識されたA.バウマニ(A. baumannii) ATCC17978は、マイクロ流体デバイスを0.2mL/分で通過し、続いて洗浄した後、コリスチン化マイクロ流路壁に結合された(図4)。おそらくコリスチン表面官能化がないため、PEG化二重螺旋マイクロ流体デバイス内では、著しい細菌捕捉が行われなかった(図4)。
EDTA抗凝固処理され、A.バウマニ(A. baumannii) ATCC17978で実験的に汚染されている新鮮なヒト全血を使用して、二重螺旋状マイクロ流体デバイスの血液浄化能力を評価した。単一のコリスチン化二重螺旋状マイクロ流体デバイスは、流速0.2ml/分でデバイスを1回通過させて、約107コロニー形成単位の細菌を全血から除去した(図16a)。流れている血液からの細菌細胞捕捉も、蛍光顕微鏡を使用して確認された。緑色の蛍光標識されたA.バウマニ(A. baumannii) ATCC17978は、感染血液をマイクロ流体デバイスに0.2mL/分で通し、続いて洗浄した後、コリスチン化マイクロ流路壁に結合される(図16bおよび16c)。しかし、ヒト全血に添加されたグラム陽性黄色ブドウ球菌(S. aureus) ATCC29213は、コリスチン化マイクロ流路壁に結合せず、コリスチンで官能化されたデバイスにおけるグラム陰性病原体への特性が確認された(図16d)。ヒト全血をコリスチン化およびPEG化された二重螺旋状マイクロ流体デバイスに流しても、血液組成に検出可能な影響を及ぼすこともなく(表2)、赤血球溶血を誘導することもなかった(表3)。
コリスチン化二重螺旋状マイクロ流体デバイスは、約107CFUの生A.バウマニ(A. baumannii) ATCC17978(図15a)および同様な量の肺炎桿菌(K. pneumoniae) ATCC700603(図15b)をPBSからデバイスを作動させて5分以内に除去した。さらに、A.バウマニ(A. baumannii)のコリスチン耐性株(すなわち、A.バウマニ(A. baumannii) 19606RおよびA.バウマニ(A. baumannii) 「患者2」)を、野生型コリスチン感受性A.バウマニ(A. baumannii)株と等しい有効性で捕捉した(図17)。統計分析により、コリスチン化二重螺旋状マイクロ流体デバイスが、PBSにおける場合と有意差がない能力でグラム陰性細菌をヒト全血から捕捉したことが確認された(図17)。
グラム陰性細菌の外膜の主成分の1つであるエンドトキシンも、敗血症の特徴である全身性炎症反応に寄与する。したがって、二重螺旋状マイクロ流体デバイスを使用した、流れている流体からのエンドトキシンの除去を評価した。エンドトキシンをエンドトキシン不含水に添加し、0.2mL/分で5分間、合計1mLをデバイスに連続的に流した。エンドトキシンは、コリスチン化二重螺旋状マイクロ流体デバイスを通っている流体から急速に捕捉され、エンドトキシン捕捉効率は、1回の通過操作で100%に近づいた(図23)。一方、PEG化デバイスを使用すると、エンドトキシンは効果的には捕捉されず、二重螺旋におけるエンドトキシン保持で重大な意味をもつコリスチンの必要性が明らかになった。
記載されたコリスチン化二重螺旋状マイクロ流体デバイスの捕捉能力は、約107CFUであり(図17)、敗血症患者を治療するための要件を3桁より高く上回っている。しかし、潜在的な臨床移行のためには、現行のデバイス設計に関連する流速を高めて、より大きい動物モデルおよびヒトの治療を可能にしなければならない。したがって、二重螺旋状流体設計のより大きいバージョンを開発して、現行の設計のスケールアップの実現可能性を明らかにした。スケールアップしたバージョンのコリスチン化二重螺旋状マイクロ流体デバイスは、最初の設計で使用されていた流速より3倍高い流速で作動し、1回の通過操作で107を超えるA.バウマニ(A. baumannii) ATTC17978 CFUを捕捉した(図8)。スケールアップしたデバイスの寸法は、適切な流体力学的粒子集約特性を維持しながら、容積対表面積の比が元のデバイスの比と同様となるように選択した。閉導管における体液輸送の支配的な原理を使用して、処置容積の増加に適した性能特性を備えたより大きい二重螺旋状デバイスを設計することができる。

- 前記流路が、健全な材料に比べた疾患材料の前記サイズに基づいて前記流路の横断面の少なくとも一部分に沿って疾患材料を単離するように適合されているアスペクト比を規定する長さ、横断面、および水力直径を有する、請求項26から50のいずれか一項に記載の方法。
- 疾患材料を生体液から捕捉および除去する方法であって、
前記生体液を流体デバイスに導入するステップであって、前記流体デバイスが少なくとも1つの入口と、少なくとも1つの出口と、前記少なくとも1つの入口と前記少なくとも1つの出口の間の多方向流路とを含む、ステップと、
前記生体液を前記流体デバイスに流して、前記疾患材料を前記流体デバイス内の前記流路の内壁に沿って官能化された疾患材料標的化リガンドに集約および曝露するステップと、
前記疾患材料と前記流路の前記内壁に結合している疾患材料標的化リガンドとの結合により、前記疾患材料を捕捉するステップと、
前記疾患材料を前記生体液から除去するステップと
を含む方法。 - 前記流体デバイスが螺旋状流路を含む、請求項52に記載の方法。
- 前記生体液が血液である、請求項52または53に記載の方法。
- 流体デバイスを流れてきた生体液を含む組成物であって、前記流体デバイスが少なくとも1つの入口と、少なくとも1つの出口と、前記少なくとも1つの入口と前記少なくとも1つの出口の間の多方向流路とを含み、前記流路が内壁を含み、前記内壁の少なくとも一部分がリガンドで被覆されており、そのようなリガンドが疾患材料に結合し、前記生体液から除去する、組成物。
- 前記生体液が血液である、請求項55に記載の組成物。
- 前記流体デバイスが螺旋状流路を含む、請求項55または56に記載の組成物。
- 疾患材料を生体液から捕捉および検出する方法であって、
血液を、流路を含む流体デバイスに流し込むステップであって、前記流路が、前記流体デバイス内の前記流路の内壁に沿って官能化された疾患材料標的化リガンドを有する、ステップと、
前記疾患材料の前記疾患材料標的化リガンドへの結合により前記疾患材料を捕捉するステップと、
前記疾患材料を単離するステップと、
前記疾患材料を培養するステップと
を含む方法。 - 前記生体液が血液である、請求項58に記載の方法。
- 前記流路が螺旋状流路である、請求項58または59に記載の方法。
- 疾患材料を生体液から捕捉および検出する方法であって、
血液を、流路を含む流体デバイスに流し込むステップであって、前記流路が、前記流体デバイス内の前記流路の内壁に沿って官能化された疾患材料標的化リガンドを有する、ステップと、
前記疾患材料の前記疾患材料標的化リガンドへの結合により前記疾患材料を捕捉するステップと、
前記疾患材料を単離するステップと、
前記疾患材料を、ポリメラーゼ連鎖反応(PCR)、蛍光インサイツハイブリダイゼーション(FISH)、およびマトリックス支援レーザー脱離/イオン化飛行時間(MALDI−TOF)からなる群から選択される手順によって同定するステップと
を含む方法。 - 前記生体液が血液である、請求項61に記載の方法。
