複数のハフニア(HfO2)ナノストローが貫通するメンブレンと、
前記円筒壁の基部全体にわたって前記メンブレンを接続してウェルを形成する生体適合性接着剤とを有し、前記ナノストローが、前記ウェル内に0.1マイクロメートルより大きく突出する、ナノストローウェルインサートデバイス。 - 前記円筒壁が、円形横断面を有する、請求項9に記載のデバイス。
- 前記円筒壁が、ポリカーボネート管、アクリル管、ポリカーボネート管又はポリエチレンテレフタレート(PET)管のうちの1つを含む、請求項9に記載のデバイス。
- 前記円筒壁と前記メンブレンの間に接着剤を更に有する、請求項9に記載のデバイス。
- 前記円筒壁と前記メンブレンの間にアクリル接着剤を更に有する、請求項9に記載のデバイス。
- 前記複数のハフニアナノストローが、10nm〜1000nmの孔径を有する、請求項9に記載のデバイス。
- 前記複数のハフニアナノストローが、前記ウェル内に0.1〜25マイクロメートル突出する、請求項9に記載のデバイス。
- 前記複数のナノストローがそれぞれ、前記デバイスの外側から、前記メンブレンを通って前記ウェル内に延在する連続チャネルを有する、請求項9に記載のデバイス。
- 前記円筒壁の外径が、1.5cm〜0.5cmである、請求項9に記載のデバイス。
- 細胞を培養しトランスフェクトする方法であって、
ナノストローウェルインサートデバイス内で1つ以上の細胞を培養するステップであって、前記ナノストローウェルインサートデバイスが、円筒壁と、前記円筒壁の基部全体に延在してウェルを形成するメンブレンとを有し、前複数のナノストローが、前記メンブレンを通って前記ウェル内に0.1マイクロメートルより大きく突出するステップと、
前記ナノストローウェルインサートデバイスをアダプタの基部に入れて、前記複数のナノストローが、前記基部内のリザーバ貯蔵部と流体連通するようにするステップであって、前記リザーバ貯蔵部が、前記基部内の底面電極と電気通信するステップと、
カバーを前記基部の上に配置するステップであって、前記カバーが上部電極を有し、それにより、前記上部電極が、前記ナノストローウェルインサートデバイス内に延在し、前記上部電極が、間の前記ナノストローウェルインサートによって前記底面電極から離されるステップと、
前記底面電極と前記上部電極の間に電圧を印加して、前記リザーバ貯蔵部から物質を、前記複数のナノストローを通して前記1つ以上の細胞内に導入するステップと、
前記アダプタから前記ナノストローウェルインサートを除去し、前記ナノストローウェルインサート内の前記1つ以上の細胞を培養するステップとを含む方法。 - 前記配置するステップが、前記基部の上に前記カバーを配置して、前記上部電極が、前記底面電極から0.25〜1.25cm離れるようにするステップを含む、請求項18に記載の方法。
- 前記電圧を印加するステップが、前記アダプタの外面上の第1の電気接点と、前記アダプタの表面の外側上の第2の電気接点との間に前記電圧を印加するステップを含み、前記第1の電気接点が、前記アダプタ内で前記上部電極と電気通信し、前記第2の電気接点が、前記アダプタ内の前記底面電極と電気通信する、請求項18に記載の方法。
- 前記電圧を印加するステップが、約0.1V〜20Vを印加するステップを含む、請求項18に記載の方法。
- 前記電圧を印加するステップが、15V未満を印加するステップを含む、請求項18に記載の方法。
- 前記ナノストローウェルインサートデバイス内の前記細胞を撮像するステップを更に含む、請求項18に記載の方法。
- 前記電圧を印加するステップが、10〜500マイクロ秒のパルス幅を1Hz〜10kHzのパルス周波数で印加するステップを含む、請求項18に記載の方法。
- 前記細胞を培養するステップが、複数のハフニア(HfO2)ナノストロー上で前記細胞を培養するステップと含む、請求項18に記載の方法。
- 前記ナノストローウェルインサートデバイスを前記基部に入れる前に前記物質を前記リザーバ貯蔵部に装填するステップを更に含む、請求項18に記載の方法。
