Description
[技術的課題]
そこで、本発明者らは、ヒトのAIMP2−DX2からexon2が欠損した部位のexon1とexon3の接合部分(junction)について、Fabファージライブラリ(phage library)をスクリーニングすることにより、野生型AIMP2には結合せず、AIMP2−DX2にのみ特異的に結合する抗体を選別して、本発明を完成した。
配列番号1で表示されるアミノ酸配列を含むポリペプチドを抗原決定部として認識する、AIMP2−DX2に特異的に結合する抗体又は抗体の断片を提供するものである。

(a)前記抗体又は抗体の断片をコードするポリヌクレオチドを含む組換え発現ベクターで宿主細胞を形質転換する段階;
(b)形質転換された宿主細胞を培養して抗体又は抗体の断片を生産する段階;及び
(c)宿主細胞から生産された抗体又は抗体の断片を収得する段階を含むAIMP2−DX2に特異的に結合する抗体又はその断片を製造する方法を提供するものである。
がん診断用製剤を製造するための前記抗体又は抗体の断片の使用を提供するものである。
(a)個体から生物学的試料を収得する段階;
(b)前記抗体又は抗体の断片を利用して前記生物学的試料内のAIMP2−DX2タンパク質の水準を測定する段階;及び
(c)前記AIMP2−DX2タンパク質の水準を正常個体のAIMP2−DX2タンパク質の水準と比較する段階を含む個体のがんの診断方法を提供するものである。
前記のような目的を達成するために本発明は、
配列番号1で表示されるアミノ酸配列を含むポリペプチドを抗原決定部として認識する、AIMP2−DX2に特異的に結合する抗体又は抗体の断片を提供する。
前記抗体又は抗体の断片を含むがん診断用組成物を提供する。
前記抗体または抗体の断片をコードするポリヌクレオチドを提供する。
前記ポリヌクレオチドを含む組換え発現ベクターを提供する。
前記組換え発現ベクターで形質転換された細胞を提供する。
(a)前記抗体又は抗体の断片をコードするポリヌクレオチドを含む組換え発現ベクターで宿主細胞を形質転換する段階;
(b)形質転換された宿主細胞を培養して抗体又は抗体の断片を生産する段階;及び
(c)宿主細胞から生産された抗体又は抗体の断片を収得する段階を含むAIMP2−DX2に特異的に結合する抗体又はその断片を製造する方法を提供する。
がん診断用製剤を製造するための前記抗体又は抗体の断片の使用を提供する。
(a)個体から生物学的試料を収得する段階;
(b)前記抗体又は抗体の断片を利用して前記生物学的試料内のAIMP2−DX2タンパク質水準を測定する段階;及び
(c)前記AIMP2−DX2タンパク質水準を正常個体のAIMP2−DX2タンパク質水準と比較する段階を含む個体のがん診断方法を提供する。
(a)本発明による抗体又は抗体の断片をコードするポリヌクレオチドを含む組換え発現ベクターで、宿主細胞を形質転換する段階;
(b)形質転換された宿主細胞を培養して、抗体又は抗体の断片を生産する段階;及び
(c)宿主細胞から生産された抗体又は抗体の断片を収得する段階を含むAIMP2−DX2に特異的に結合する抗体又はその断片を製造する方法を提供する。
(a)個体から生物学的試料を収得する段階;
(b)前記抗体又は抗体の断片を利用して前記生物学的試料内のAIMP2−DX2タンパク質水準を測定する段階;及び
(c)前記AIMP2−DX2タンパク質水準を正常個体のAIMP2−DX2タンパク質水準と比較する個体のがん診断方法を提供する。
従って、本発明は、AIMP2−DX2タンパク質に特異的に結合する抗体又は抗体の断片、これを含むがん診断用組成物、さらに、前記抗体をコードするポリヌクレオチドと組換え発現ベクター、これを利用して形質転換された細胞及び前記抗体又は抗体の断片を製造する方法を提供する。本発明で提供する抗体と抗体の断片は、野生型AIMP2には反応せず、exon2が欠損したAIMP2−DX2突然変異タンパク質に特異的に結合するため、がん、炎症性疾患など多様な病理に関連しているAIMP2−DX2タンパク質の存在と発現程度を確認するのに効果的である。
キメラFabライブラリ製作
AIMP2−DX2に特異的に結合する抗原結合部位(antigen-binding site)をスクリーニングするためのウサギ/ヒトキメラ(chimeric)Fabライブラリーを構築した。ウサギの免疫化(immunization)、ウサギ脾臓細胞(splenocyte)からのmRNAの分離とcDNA合成、さらに、ウサギ/ヒトキメラFab抗体ライブラリの構築は、基本的に“Phage Display:A Laboratory Manual”(Barbas, C.F. 3rd. et al. Eds. Cold Spring Harbor Laboratory Press)によって公知にされた方法に基づいて行った。

<2−1>ファージ提示Fabライブラリパニング
AIMP2−DX2抗原ペプチドに特異的に結合する抗原結合部位を有するFabを選別するために、前述した実施例で製作した抗体(Fab)ライブラリパニング(panning)実験を、公知の方法(Bai, X. et al. PLoS ONE 2015 doi:10.1371/journal.pone.0141045)により実施した。
実施例<2−1>のパニングで選別されたコロニーのファージが発現する抗体(Fab)が、実際にAIMP2−DX2抗原ペプチドに特異的に結合するか否かを確認した。

H5抗体又は断片(Fab)のAIMP2−DX2に対する結合特異性をさらに詳しく確認した。
Claims (15)
- 配列番号1で表示されるアミノ酸配列を含むポリペプチドを抗原決定部(epitope)として認識する、AIMP2−DX2に特異的に結合する抗体又は抗体の断片。
- 前記抗体又は抗体の断片は、配列番号3で表示されるアミノ酸配列を含む重鎖相補性決定部位(CDR)1、配列番号5で表示されるアミノ酸配列を含む重鎖CDR2、及び配列番号7で表示されるアミノ酸配列を含む重鎖CDR3を含む重鎖可変領域;及び配列番号9で表示されるアミノ酸配列を含む軽鎖CDR1、配列番号11で表示されるアミノ酸配列を含む軽鎖CDR2及び配列番号13で表示されるアミノ酸配列を含む軽鎖CDR3を含む軽鎖可変領域を含むことを特徴とする請求項1記載の抗体又は抗体の断片。
- 前記抗体又は抗体の断片は、配列番号15で表示されるアミノ酸配列を含む重鎖可変領域;及び配列番号17で表示されるアミノ酸配列を含む軽鎖可変領域を含むことを特徴とする請求項1記載の抗体又は抗体の断片。
- 前記抗体は、配列番号19で表示されるアミノ酸配列を含む重鎖;及び配列番号21で表示されるアミノ酸配列を含む軽鎖を含むことを特徴とする請求項1記載の抗体又は抗体の断片。
- 前記抗体は、IgG、IgA、IgM、IgE及びIgDからなる群から選ばれたことを特徴とする請求項1記載の抗体又は抗体の断片。
- 前記抗体の断片は、ダイアボディ、Fab、Fab’、F(ab)2、F(ab’)2、Fv及びscFvからなる群から選ばれたことを特徴とする請求項1記載の抗体又は抗体の断片。
- 請求項1乃至6のうちいずれか一項に記載の抗体又は抗体の断片を含むがん診断用組成物。
- 前記がんは結腸がん、大腸がん、肺がん、肝臓がん、胃がん、食道がん、膵臓がん、胆嚢がん、腎臓がん、膀胱がん、前立腺がん、精巣がん、子宮頸部がん、子宮内膜がん、絨毛がん、卵巣がん、乳がん、甲状腺がん、脳がん、頭頸部がん、悪性黒色腫、リンパ腫、再生不良性貧血及び血液がんからなる群から選ばれたことを特徴とする請求項7記載のがん診断用組成物。
- 請求項1乃至6のうちいずれか一項に記載の抗体又は抗体の断片をコードするポリヌクレオチド。
- 前記ポリヌクレオチドは、配列番号16又は配列番号18で表示される塩基配列を含むことを特徴とする請求項9記載のポリヌクレオチド。
- 請求項10に記載のポリヌクレオチドを含む組換え発現ベクター。
- 請求項11に記載の組換え発現ベクターで形質転換された細胞。
- (a)請求項12に記載の組換え発現ベクターで宿主細胞を形質転換する段階;
(b)形質転換された宿主細胞を培養して抗体又は抗体の断片を生産する段階;及び
(c)宿主細胞から生産された抗体又は抗体の断片を収得する段階を含むAIMP2−DX2に特異的に結合する抗体又はその断片を製造する方法。 - がん診断用製剤を製造するための請求項1乃至6のうちいずれか一項に記載の抗体又は抗体の断片の使用。
- (a)個体から生物学的試料を収得する段階;
(b)請求項1乃至6のうちいずれか一項に記載の抗体又は抗体の断片を利用して、前記生物学的試料内のAIMP2−DX2タンパク質水準を測定する段階;及び
(c)前記AIMP2−DX2タンパク質水準を、正常個体のAIMP2−DX2タンパク質水準と比較する段階を含む個体のがん診断方法。
