Abstract
Description
本発明は、AIMP2のエクソン2が欠損した変異体AIMP2DX2およびその利用に係り、具体的には、AIMP2DX2、その診断マーカーとしての用途およびその抑制剤を使用した抗癌用途に関する。
癌による死亡が世界的に増加しつつある。去る数年間、癌に対する集中的な研究が行われたにも拘わらず、癌は依然として全世界的な主要死亡原因である。
本発明の目的は、AIMP2タンパク質のエクソン2の領域が欠失したAIMP2DX2タンパク質を提供することにある。
図1a〜図1eは、TGF−βシグナル伝達におけるAIMP2の機能的重要性およびAIMP2とSmad2/3との相互作用を示す。AIMP2+/+およびAIMP2−/−MEFsにおいてTGF−βが細胞増殖(図1a)、コロニー形成(図1b)、細胞周期の進行(図1c)、並びにSmad2およびSmad3の核移動(図1d)に及ぼす効果を測定した。図1aでは、0、2および4ng/mlのTGF−β濃度で6時間細胞を培養した。TGF−β無処理細胞におけるチミジン挿入(thymidine incorporation)を1としたとき、数値は4つの独立的実験値の平均を示す。図1bでは、AIMP2+/+およびAIMP2−/−MEFs(14.5d)をTGF−β(2ng/ml)の存在下に4時間培養し、ホルムアルデヒドで固定した後、Giemsa染色によりコロニーを視覚化した。図1cでは、MEFsをTGF−βで24時間処理した後、G0/G1相細胞の占有程度をフローサイトメトリー(flow cytometry)で検査した。図1dでは、MEFSをTGF−β(2ng/ml)で1時間処理した。Smad2およびSmad3をその特異的抗体で反応させた後、FITCコンジュゲート抗体(緑色)で視覚化した。核はPI(赤色)で染色された。図1eでは、AIMP2とSmad2およびSmad3との相互作用を、Smad2およびSmad3に対する抗体を用いた共同免疫沈殿(coimmunoprecipitation)によって検査した。
AIMP2(ARS-interacting multi-functional protein 2)は、アミノアシル−tRNA合成酵素複合体(Aminoacyl-tRNA synthetase:ARSs)の形成に関連したタンパク質の一つであって、p38/JTV−1またはp38と呼ばれる。
(細胞培養、化合物および細胞周期の分析)
本研究に使用された細胞株は、10%FBS含有RPMI−1640で維持した。マウス胚児線維芽細胞(MEFS:Mouse embryonic fibroblasts)は、12.5日〜14.5日目の胚児から単離し、20%FBS含有DMEMで培養した。TGF−βが細胞周期に及ぼす影響を確認するために、細胞を無血清または1%FBS含有培地で2ng/mlのTGF−βで処理して培養し、細胞を集めてFACS分析した。また、細胞増殖は[3H]チミジン挿入(thymidine incorporation)で測定した。細胞を、TGF−βを処理または無処理した状態の無血清培地で20時間培養した後、[3H]チミジン1μCi/mlの存在下に4時間培養した。挿入されたチミジンは、液体閃光計算器を用いて文献(Kim et al., Nat. Genet. 34, 330-336(2003))によって定量した。TGF−βはR&D systemから購入し、抗−Smad2およびSmad4抗体はSnata Cruzから購入した。抗−AIMP2抗体は実験室で製造した。単離したAIMP2ンパク質500μg(1mg/ml)をウサギ(male、NewZealand)にcomplete adjuvant(Sigma)500μlと混ぜて皮下に注射した。7日後、AIMP2タンパク質500μgをincomplete adjuvant(Sigma)500μlと混ぜてウサギの皮下に注射し、これを7日間隔で2回さらに行った。3日後、ウサギの血液から抗体を採取する工程によって抗−AIMP2抗体を製造した。
細胞をTGF−βで一定の時間処理し、プロテアーゼ含有RIPA緩衝液を用いて細胞からタンパク質を抽出し、10〜12%SDS−PAGEを用いて単離した後、特異的抗体としてECLシステムを用いて免疫ブロットした。免疫沈殿のために、細胞溶解物をIgGおよびアガロース−コンジュゲートされたタンパク質Aで処理した。遠心分離の後に、上澄み液を特異的抗体およびアガロース−コンジュゲートされたAで2時間培養した。冷たいPBSで2回、RIPAで1回洗浄した後、結合したタンパク質を特異的抗体で沈殿させ、溶出してウエスタンブロット分析を行った。
全RNA(total-RNA)を製造社(Qiagen)のプロトコールに従って単離した。新鮮に準備された組織(3×3×3mm)を小さい欠片に切り、350μlの溶解緩衝液と混合し、ホモジナイザーまたは注射器で均質化させた。350μlの70%エタノールを添加した後、溶解物(lysate)を多数回上下に揺らしてカラムにロードし、13,000rpmで15秒間遠心分離した。カラムを洗浄緩衝液で2回洗浄した後、RNAを40μlのRNase−free DWで溶出した。逆転写のために、1μgの単離されたRNAをAIMP2−特異的プライマー(図8b)の鋳型として使用した。逆転写の後、DWで3倍に希釈させ、1μlを、0.5μl dMTP(各2.5mM)、0.5μlの表示されたプライマー(各10pM)、1.5μl DMSOおよび0.1μl Taqポリメラーゼ(5U/μl)を含む30μlPCR反応に使用した。
AIMP2DX2の発現を抑制するために、AIMP2DX2に対するsiRNAを製作した。AIMP2DX2に対するsiRNAをコーディングする配列を製作し、IMG−700(Imgenex)ベクターのSalIおよびXbaI部位と連結させ、クローニングをDNAシーケンシングで確認した。H322細胞を、si−DX2発現するベクター2μgで形質転換させた。1mlの無血清培地で3時間DNA−リポソーム複合体とインキュベーションした後、20%FBSを含む1mlのRPMI−1640をH322細胞に投与した。さらに20時間培養した後、無血清条件または指示された時間だけTGF−βを処理した。
ヒトAIMP2およびSmad2(およびその欠失断片)をコーディングするcDNAを、特異的プライマーを用いるPCRによって獲得した。SmadおよびAIMP2のPCR産物をEcoRIおよびXhoIで切断してそれぞれpEG202(LexA)およびpJG4−5(B42)のようなサイトに結合させた(Gyuris et al, a human G1 and S phase protein phosphatase that associates with Cdk2. Cell 75, 791-803, 1993)。Lex−Smad2断片とB42−AIMP2間の相互作用は、X−galを含む酵母培地で生存する能力で分析した(Rho et al, Proc Natl Acad Sci U S A 96, 4488-93, 1999)。
野生型MEFsをpcNDA−AIMP2DX2またはpcDNAで形質転換させ、形質転換体を、400μg/ml G418を含むDMEM培地で選択した。形質転換されていない細胞を除去した後、形質転換体を、G418を含まない培地で3日間培養した後、2%PFAで固定させ、Giemsaで染色した。
冷凍した組織スライドを2%パラホルムアルデヒドで固定し、冷たいPBSで3回洗浄した。0.2%トリトンX−100(PBST)および1%BSA含有PBSでブロッキングおよび浸潤(permeablization)した後、スライドを抗−AIMP2抗体で2時間培養した。PBSで洗浄した後、抗−ウサギ山羊IgG−FITCおよびプロピジウムヨウ化物(propidium iodide)(PI50μg/ml)で1時間培養し、その後PBSで洗浄し、固定して共焦点顕微鏡(confocal microscopy)で観察した。
AIMP2DX2を発現するプラスミドを構築するために、AIMP2DX2のcDNAをpcDNA3.1−mycにクローニングした。まず、AIMP2DX2をH322 cDNAにおいてEcoRIおよびXhoIリンカー付きプライマーを用いて増幅させた後、EcoRIおよびXhoIを用いてpcDNA3.1−mycにクローニングした。AIMP2DXを発現するpcDNA3−myc/AIMP2DX2ベクター(図13)は、寄託名をEscherichai coli DH5alpha/AIMP2DX2として、2004年10月25日付けで、KCTC(Korean Collection for Type Cultures、韓国大田市儒城區魚隠洞52番地韓国生命工学研究院)に寄託番号KCTC 10710BPで寄託した。
マウスゲノムDNA上の突然変異は、方法によるgene trap方法に従った(Zambrowicz et al, Nature, 9, 608-611, 1998)。gene trapベクターは、129/SvEvBrdマウスに由来した胚児幹細胞の無作為的突然変異ライブラリ(OmniBank library, Lexicon Genetics, USA)の生成に使用された。ライブラリにおいて、AIMP2遺伝子が破壊されたOST7994クローンを同定した。前記クローンを用いて、Lexicon Genetics社の標準プロトコールに従ってC57/BL6異型接合のマウスを生成した。同型接合の突然変異マウスを得るために、異型接合のマウスを交配させた。単離された13.5日目のマウスの腹をrazor bladeで切り、トリプシン酵素処理してペトリ皿に培養のためのプレーティングを行った。マウス胚児線維細胞(embryonic fibroblast cells)から単離されたゲノムDNAを用いてサザンブロットおよびPCR分析して突然変異を確認した。AIMP2発現をノーザンブロットおよびウエスタンブロットで確認した。gene trapの挿入位置をゲノムDNAシーケンシングで確認した。サザンブロットのために、マウス尻からゲノムDNAを単離し、SacIで分解した。切断されたDNA断片を変性ゲル電気泳動(denaturing gel electrophoresis)で単離し、放射学的にラベルされたAIMP2遺伝子の1.2kb PCR産物を配列番号17と18のプライマーでハイブリッドさせた。ハイブリッドされたバンドを蛍光イメージ分析器(phosphorimage analyzer)を用いて検出した。単離されたゲノムDNAを配列番号19と21、配列番号20と21の特異的プライマーでPCR分析に使用した。
AIMP2+/−マウス32匹と(雄19匹と雌13匹)AIMP2+/+マウス25匹(雄14匹および雌11匹)を実験に使用した。マウス腹腔内にベンゾピレン(100mg/kg)を単一注射した。対照区として、AIMP2+/+マウス3匹(雄2匹および雌1匹)およびAIMP2+/−マウス5匹(雄4匹および雌1匹)をバイカル溶液(生理食塩水に10%DMSOおよび35%PEG40が溶けた溶液)で注射した。
前記で製作したAIMP2ノックアウト(KO)型と野生型(WT)マウス総33匹をWT雄対照区(3匹)、WT雄処理区(6匹)、WT雌対照区(2匹)、WT雌処理区(6匹)、KO雄対照区(2匹)、KO雄処理区(6匹)、KO雌対照区(3匹)、KO雌処理区(5匹)に分類した。
TGF−βシグナル伝達においてAIMP2が重要に作用することを考察するために、TGF−βで誘導された細胞成長停止、Smad2/3の核移動およびSmad2/3とAIMP2との相互作用に対するAIMP2+/_+およびAIMP2−/−MEFsの反応を調べた。チミジン挿入、コロニー形成およびフローサイトメトリーで確認した結果、正常細胞の成長はTGF−βに抑制されるのに比べ、AIMP2欠乏細胞はこのようなシグナルに反応を示さなかった(各図1a、bおよびc)。MEFsをTGF−βで処理したとき、正常細胞ではSmad2およびSmad3が核に移動したが、AIMP2−/−細胞ではそうではなかった(図1d)。このような全ての結果は、Smad2および3を経由するTGF−βシグナル伝達におけるAIMP2の機能的重要性を示す。
癌形成とAIMP2との関連性を調査するために、相異なる各種癌細胞株におけるAIMP2の発現程度を観察した。観察した6つの細胞株のうち3つはウエスタンブロット(図3a)およびFACS分析(図3b)において低いAIMP2水準を示した。低いAIMP2水準を示す細胞株全てはTGF−βタイプII受容体が正常水準で発現し、キナーゼ活性も保有しているという事実は、低いAIMP2水準が受容体の機能異常による結果ではないことを意味する。AIMP2水準差が転写差異によるものかを確認するために、他の組み合わせのAIMP2特異的プライマーを用いてRT−PCRを行った。AIMP2遺伝子は4つのエクソンから構成されている(図8a)。エクソン3および4を貫通するAIMP2 cDNAを生成させる配列番号3と4のプライマーを使用したとき、減少したAIMP2水準を示す細胞からAIMP2転写の減少は観察されなかった(図3c、第1行)。これは低いAIMP2水準の低い転写の結果ではないことを意味する。エクソン1から3までの転写物を生成させる配列番号5と6のプライマーを使用したとき、予想した大きさの転写物だけでなく、小さい大きさの転写物も得ることができた(図3c、第2行)。小さい大きさの転写物の配列分析結果、AIMP2の69個のアミノ酸をコーディングするエクソン2が欠損していることが分かった(図8b)。この小さい大きさの転写物の生成を再確認するために、エクソン2欠損によって生成されるエクソン1およびエクソン3の接合配列をターゲッティングする配列番号8のプライマー(図8b、プライマーDX−B)を製作し、配列番号7プライマーを共に用いてRT−PCRを行った。低い水準のAIMP2を発生する細胞株は、小さい大きさの転写物(DX2と命名し、図3c、第2行および第3行)を生成した。
DX2生成とAIMP2抑制との一般な関連性を理解するために、DX2でDU145細胞を形質転換した後、AIMP2水準の変化を調査した。ウエスタンブロット(図4a、第1行)およびFACS分析(図9b)の結果、DX2で形質転換された細胞においてAIMP2は減少した。また、AIMP2のターゲットであるc−mycの発現がDX2の導入によって増加した(図4a、第3行)。DX2はTGF−βによる成長停止を軽減させた(図4b、図9c)。
DX2をMEFsにトランスフェクションした後、DX2が細胞成長に及ぼす効果を顕微鏡分析およびコロニー形成によって観察した。細胞成長は、DX2導入によって著しく増加した(図5a)。また、DX2に特異的に結合できる3種のsiRNA(si−DX2)を(図5b)発現するそれぞれのベクターを製造し、DX2を発現するH322およびH460細胞を形質転換した後、正常的AIMP2およびsmad2のリン酸化に及ぼす影響を観察した(図5c)。4番または5番siRNAを発現するベクターをH322細胞にトランスフェクションしたとき、TGF−βによる細胞死滅が増加し(左)、細胞周期の停止(右)が起こることをフローサイトメトリーで観察することができた(図5d)。DX2を発現するH322細胞に、DX2の転写を特異的に抑制する4番siRNA(si−DX2)を発現するベクターを組み込み、siRNAによってAIMP2の正常的水準およびTGF−ベータシグナル伝達が復旧されるかをチェックした。si−DX2はDX2転写物を除去させ(図5eの上)、AIMP2の正常水準およびTGF−βにより誘導されたSmad2のリン酸化(図5eの下および図5f)、Smad2の核移動(図5f)、TGF−β依存的レポーターの発現(図5g)、および成長停止(図5h)を復旧させた。前記全ての結果は、TGF−βシグナル伝達および細胞成長調節に及ぶDX2の崩壊的な効果を証明することである。
AIMP2の減少は様々な癌細胞株からよく発見され(図3a、bおよびd)、AIMP2の損失は肺細胞の過増殖をもたらすため(Kim et al., Nat. Genet. 34, 330-336, 2003)、本発明者はAIMP2の異常が肺癌形成と関連しているかを調べた。このような可能性を検討するために、化学的発癌物質、BP(benzo-(α)-pyrene)(Wang et al, Cancer Res. 63, 4389-4395, 2003)をAIMP2+/+およびAIMP2+/−マウス腹腔内に投与して肺腫瘍を誘導した後、肺における腫瘍形成を観察した。BP投与後6週から、AIMP2+/−マウスでは50〜70%の頻度で、正常マウスでは約30%の頻度で肺腫瘍が観察された(図6a)。これは、異型接合マウス(heterozygous mice)が、BPにより誘導された腫瘍形成にさらに敏感であることを意味する。BP注射マウスの肺におけるDX2生成有無を調査した。3匹のAIMP2+/+マウスのうち1匹のみが腫瘍を発生させるのに比べ、AIMP2+/−マウスから単離した4つの肺のうち3つから腫瘍が生成された。全てのこのような腫瘍はDX−2を生成させた(図10)。これは、肺癌形成とDX2との関連性があることを意味する。
癌発生とAIMP2、AIMP2DX2との連関性を多様な組織および癌細胞株におけるAIMP2DX2発現で検査した。
2つの肝細胞癌(hepatocellular carcinoma)試料から、正常と癌組織におけるAIMP2DX2の生成有無をRT−PCRで観察した。癌組織からAIMP2DX2の形成が観察された(図11a)。その他の多様な癌細胞株におけるAIMP2DX2の生成を調べた。細胞株を6ウェルプレートに1×105ずつシードし、24時間後に細胞を収穫した。収穫した細胞を、1%β−メルカプトエタノールを含むGSS溶液を用いて溶解させ、酸性フェノール(pH4.3)とクロロホルムを含む4%イソアミルアルコールを入れて混合した。これを遠心分離し、上澄み液をイソプロパノールで沈殿させ、再び遠心分離してペレットを得た。ペレットをエタノールで洗浄し、DWに溶かしてRNAを得た。RNAをM−MLV逆転写酵素(invitrogen)を用いて42℃で1時間逆転写させ、配列番号16のプライマーと配列番号6のプライマーを用いて、95℃で1分、60℃で1分、72℃で1分を1サイクルとした条件で38サイクルのPCRを行った。その結果、肺癌細胞株(H226、H460、H322およびH290)、胚児腎臓癌細胞株(293)、骨肉腫細胞株(SaOS2)、皮膚癌細胞株(HaCAT)、乳癌細胞株(MCF7)からAIMP2DX2が発現した(図11b)。
AIMP2遺伝子をノックアウトさせたマウスにおける発癌程度を、AIMP2発現が正常な野生型マウスと比較してみた。その結果、AIMP2ノックアウトマウスにおいて、皮膚腫瘍が発生するマウス数とマウスの腫瘍の数が確然に増加することが分かった。具体的に、マウスにDMBAにより癌を誘発し、TPAで癌の誘発を促進させる場合、AIMP2ノックアウト雌マウスは雌対照区マウスに比べて腫瘍発生率が約1.3倍増加し、AIMP2ノックアウト雄マウスは雄対照区マウスに比べて腫瘍発生率が約2倍増加した(図12b)。また、腫瘍発生マウスに発生した腫瘍の数も、AIMP2ノックアウト雌マウスは雌対照区マウスに比べて約4倍増加し、AIMP2ノックアウト雄マウスは雄対照区マウスに比べて約2倍増加した(図12cおよび図12d)。
本発明の癌に特異的に発現するAIMP2DX2(タンパク質、mRNA)を癌診断マーカーとして用いて癌を正確且つ容易に診断することができ、AIMP2DX2タンパク質に対する抗体、siRNA、アンチセンス核酸などのAIMP2DX2の抑制剤を用いて癌を効果的に予防または治療することができる。
Claims (27)
- AIMP2(ARS-interacting multi-functional protein 2)タンパク質のエクソン2領域が欠失したAIMP2DX2タンパク質。
- 配列番号2のアミノ酸配列を有する、請求項1に記載のAIMP2DX2タンパク質。
- 癌診断マーカーである、請求項1に記載のAIMP2DX2タンパク質。
- 請求項1のAIMP2DX2タンパク質をコーディングする核酸分子。
- 配列番号1のヌクレオチド配列を有する、請求項4に記載の核酸分子。
- 請求項1のAIMP2DX2タンパク質をコーディングする核酸分子を含む組み換えベクター。
- 請求項6の組み換えベクターで形質転換された形質転換体。
- 請求項7の形質転換体を培養して請求項1のAIMP2DX2タンパク質を製造する方法。
- 請求項1のAIMP2DX2タンパク質に特異的な抗体。
- 請求項1のAIMP2DX2タンパク質に特異的な抗体を含む癌診断キット。
- (a)分析する試料を提供する段階と、
(b)前記試料から、請求項1のAIMP2DX2タンパク質をコーディングするヌクレオチド配列の発現を検出する段階とを含んで、癌診断マーカータンパク質AIMP2DX2を検出する方法。 - 段階(b)において、請求項1のAIMP2DX2タンパク質に特異的な抗体と接触させて検出する、請求項11に記載の方法。
- 段階(b)において、増幅反応またはハイブリッド化反応によって検出する、請求項11に記載の方法。
- 増幅反応が、AIMP2とAIMP2DX2のmRNAを区分するプライマーを用いるRT−PCR(reverse transcription-polymerase chain reaction)により行われるか、或いは、ハイブリッド反応が、AIMP2とAIMP2DX2のmRNAを区分するプローブを用いて行われる、請求項13に記載の方法。
- 配列番号5と6、配列番号7と8、または配列番号16と6のヌクレオチド配列を有するプライマー、または配列番号8または配列番号16のヌクレオチド配列を有するプローブを用いる、請求項13に記載の方法。
- 配列番号3、配列番号4、配列番号5、配列番号6、配列番号7、配列番号8または配列番号16のヌクレオチド配列を有する核酸分子。

- (a)試験物質を、請求項1のAIMP2DX2タンパク質をコーディングする遺伝子を発現する細胞と接触させる段階と、
(b)試験物質によるAIMP2DX2タンパク質をコーディングする遺伝子の発現抑制またはAIMP2DX2タンパク質の分解促進を測定する段階とを含んで、請求項1のAIMP2DX2タンパク質をコーディングする遺伝子の発現を抑制またはAIMP2DX2タンパク質の分解を促進する抗癌剤をスクリーニングする方法。 - RT−PCRまたはウエスタンブロットによりスクリーニングする、請求項26に記載の方法。
