Claims (15)
- ポリメラーゼ−鋳型複合体を調製する方法であって、
(a)ポリメラーゼを提供するステップ、および、
(b)0.8μM〜2.2μMの濃度のヌクレオチドを含む35℃〜45℃の温度の溶液中で前記ポリメラーゼをポリヌクレオチド鋳型と接触させ、それにより前記ポリメラーゼ−鋳型複合体を調製するステップ、
を含む、前記方法。 - 前記溶液を、前記ポリメラーゼ−鋳型複合体の前記ポリメラーゼで飽和させるステップをさらに含む、請求項1に記載の方法。
- 鋳型−ポリメラーゼ複合体の処理能力を増大させる方法であって、
0.8μM〜2.2μMの濃度のヌクレオチドを含む35℃〜45℃の温度の溶液中でポリメラーゼ−鋳型複合体を形成させるステップを含み、
ここで、前記高温溶液中で形成された前記ポリメラーゼ−鋳型複合体の処理能力が、対照である室温のポリメラーゼ−鋳型複合体溶液から得られる処理能力より大きいものである、前記方法。 - 前記ポリメラーゼ−鋳型複合体を形成させるステップが、前記溶液を前記ポリメラーゼ−鋳型複合体の前記ポリメラーゼで飽和させるステップを含むものである、請求項3に記載の方法。
- 前記ポリメラーゼ−鋳型複合体を形成させた後、前記溶液中のヌクレオチドの濃度を上昇させるステップをさらに含む、請求項1〜4のいずれか1項に記載の方法。
- 前記ポリメラーゼ−鋳型複合体の前記ポリメラーゼが配列番号14として示されるアミノ酸配列と少なくとも85%、90%、95%、98%もしくはそれ以上の配列同一性を有し、またはこれと同一である、請求項1〜5のいずれか1項に記載の方法。
- ポリヌクレオチド鋳型のナノポアに基づくシーケンシングのための方法であって、
0.8μM〜2.2μMの濃度のヌクレオチドを含む溶液中でポリメラーゼ−鋳型複合体を形成させるステップであって、前記溶液が高い温度を有するステップ、
前記形成されたポリメラーゼ−鋳型複合体をナノポアと組み合わせて、ナノポア−シーケンシング複合体を形成させるステップ、
タグ付きヌクレオチドを前記ナノポアシーケンシング複合体に提供して、35℃〜45℃の高い温度で前記鋳型の鋳型依存的ナノポアシーケンシングを開始するステップ、および、
前記ナノポアを利用して、前記タグ付きヌクレオチドのそれぞれが前記ポリメラーゼと会合しつつ前記タグ付きヌクレオチドのそれぞれが組み込まれている間に、前記タグ付きヌクレオチドのそれぞれと会合したタグを検出し、それにより前記ポリヌクレオチド鋳型の配列を決定するステップ、
を含む、前記方法。 - 前記ポリメラーゼ−鋳型複合体を形成させるステップが、前記溶液を前記ポリメラーゼ−鋳型複合体の前記ポリメラーゼで飽和させるステップを含むものである、請求項7に記載の方法。
- 前記ポリメラーゼ−鋳型複合体の前記ポリメラーゼが配列番号14として示されるアミノ酸配列と少なくとも85%、90%、95%、98%もしくはそれ以上の配列同一性を有し、またはこれと同一である、請求項7または8に記載の方法。
- 前記ポリメラーゼがSpyCatcher配列(配列番号10)に直接または間接的に連結されている、請求項7〜9のいずれか1項に記載の方法。
- ポリメラーゼ−鋳型複合体を調製する方法であって、
(a)ポリメラーゼを提供するステップ、および、
(b)少なくとも100mMの濃度の塩を含みヌクレオチドを含まない溶液中で前記ポリメラーゼをポリヌクレオチド鋳型と接触させ、それにより前記ポリメラーゼ−鋳型複合体を調製するステップ、
を含む、前記方法。 - 鋳型−ポリメラーゼ複合体の処理能力を増大させる方法であって、
前記方法が、少なくとも100mMの濃度の塩を含みヌクレオチドを含まない溶液中で鋳型−ポリメラーゼ複合体を形成させるステップを含み、
ここで、前記鋳型−ポリメラーゼ複合体の処理能力が同様に高い濃度の塩を含む溶液中、ヌクレオチドの存在下で形成された場合の同じ鋳型−ポリヌクレオチド複合体の処理能力より大きいものである、前記方法。 - 鋳型依存的DNA合成を行う方法であって、
(a)少なくとも100mMの濃度の塩を含みヌクレオチドを含まない溶液中でポリメラーゼ−鋳型複合体を提供するステップ、および、
(b)ヌクレオチドを前記溶液に添加することによって前記鋳型依存的DNA合成を開始するステップ、
を含む、前記方法。 - 高い塩濃度におけるナノポアシーケンシングのための方法であって、
(a)少なくとも100mMの濃度の塩を含む溶液中でポリメラーゼ−鋳型複合体を提供するステップであって、前記溶液がヌクレオチドを実質的に含まないステップ、
(b)前記ポリメラーゼ−鋳型複合体をナノポアと組み合わせてナノポア−シーケンシング複合体を形成させるステップ、
(c)タグ付きヌクレオチドを前記ナノポアシーケンシング複合体に提供して、前記高い塩濃度で前記鋳型の鋳型依存的ナノポアシーケンシングを開始するステップ、および、
(d)前記ナノポアを利用して、前記ヌクレオチドのそれぞれが前記ポリメラーゼと会合しつつ前記ヌクレオチドのそれぞれが組み込まれている間に、前記タグ付きヌクレオチドのそれぞれと会合したタグを検出し、それにより前記ポリヌクレオチド鋳型の配列を決定するステップ、
を含む、前記方法。 - 少なくとも100mMの塩の溶液中にポリメラーゼ−鋳型複合体を含む、保存または反応組成物。
