上記で要約したように、本開示の態様は、フローサイトメータにおいて事象を検出するための方法を含む。ノイズのない細胞事象の取り込みは、光学サイドローブ、流体ドリフト、ベースラインドリフト、フローセル汚染などの現象によって難しいものとなる。これらの現象は、小細胞事象に似ている望ましくない信号を生じる。これらの望ましくない信号は、より大きい有効な信号が生成される前及び/又は後にしばしば生成される。これらの誤って取り込まれた望ましくない信号は、有効な細胞事象、例えば、小細胞事象の特徴抽出を汚染(contaminate)する。

実施例1:事象検出のための望ましくない細胞事象信号の除去
この事象検出の例では、粒子は細胞であり、クリーンな血小板(PLT)細胞ヒストグラムを得るべく望ましくない細胞事象信号が除去される。
この例では、デジタルデータ取り込みサブシステムは、細胞事象カウント並びにアッセイの持続時間の追跡を維持する。アッセイにおける所定の時点で、データ取り込みシステムが、或る細胞事象信号(例えば、細胞事象からの前方散乱光)が期待値を下回ると判定する場合、光電子信号の可変利得設定が増加される。データ取り込みサブシステムが、アッセイの初めに固定の時間(約300ms)内に不飽和の事象に比べて多くの飽和した細胞事象を見つける場合、光電子信号の可変利得設定が減少される。
理論的には、10MSPSで2マイクロ秒の持続時間の理想ガウシアンパルスは、1.75MHzの帯域幅を占有する。理論値を用いる代わりに、既存の電子装置でクリーンなフローセル上でとられた実験データから帯域幅を計算した。実験データは、クリーンなフローセルにおける関心ある帯域幅はおよそ1.65MHzであり、したがって、理論上のものに非常に近いことを示す。理想ガウシアンパルスのパワースペクトル及びクリーンなフローセルからとられた実験データのパワースペクトルが、それぞれ、図13及び図14に示される。

- フローサイトメータにおいて事象を検出するための方法であって、
粒子を分析器のフローセルに流すこと、
前記フローセルを通って流れる粒子を光学的にインテロゲートすること、
推定上の事象の特徴を抽出すること、
前記推定上の事象にタイムスタンプを付与すること、
推定上の以前の事象と推定上の現在の事象との時間差を求めること、および、
前記時間差を持続時間閾値と比較すること、を含み、
前記時間差が前記持続時間閾値を上回る場合、前記推定上の現在の事象を現在の事象として記憶し、
前記時間差が前記持続時間閾値を下回る場合、前記推定上の現在の事象のピーク高さ特徴及び前記推定上の以前の事象のピーク高さ特徴をピーク高さ閾値と比較し、
前記推定上の現在の事象のピーク高さ特徴が前記ピーク高さ閾値を下回る場合、前記推定上の現在の事象を廃棄し、

- フローサイトメータにおいて細胞を検出するための方法であって、
細胞を含む細胞サンプルをフローサイトメータのフローセルに流すこと、
前記フローセルを通って流れる細胞からの光信号を第1の利得設定で検出すること、および、
前記フローセルを通って流れる細胞からの光信号を第2の利得設定で検出すること、を含み、前記第2の利得設定が、前記第1の利得設定とは異なる、方法。 - 前記第2の利得設定が前記第1の利得設定よりも高い、請求項24に記載の方法。
- 前記第2の利得設定が、高い細胞濃度を有する細胞サンプルを考慮に入れるためになされる、請求項25に記載の方法。
- 前記第2の利得設定が前記第1の利得設定よりも低い、請求項24に記載の方法。
- 前記第2の利得設定が、低い細胞濃度を有する細胞サンプルを考慮に入れるためになされる、請求項27に記載の方法。
- 前記第1及び第2の利得設定が、フォトダイオードの利得設定、光電子増倍管(PMT)の利得設定、又はその両方を含む、請求項24から28のいずれか一項に記載の方法。
- 前記細胞サンプルが血液サンプルである、請求項24から29のいずれか一項に記載の方法。
- 第1の細胞タイプを検出するべく前記第1の利得設定で検出された光信号を解析すること、および、
第2の細胞タイプを検出するべく前記第2の利得設定で検出された光信号を解析すること、を含む、請求項24から30のいずれか一項に記載の方法。 - 前記第1の利得設定が前記第2の利得設定よりも高く、前記第1の利得設定で血小板が検出され、前記第2の利得設定で赤血球(RBC)が検出される、請求項31に記載の方法。
- 前記第1の利得設定が前記第2の利得設定よりも低く、前記第1の利得設定で赤血球(RBC)が検出され、前記第2の利得設定で血小板が検出される、請求項31に記載の方法。
- 前記第1の利得設定での光信号の検出が、前記第2の利得設定での光信号の検出と同じ持続時間にわたる、請求項24から33のいずれか一項に記載の方法。
- 前記第1の利得設定での光信号の検出が、前記第2の利得設定での光信号の検出とは異なる持続時間にわたる、請求項24から33のいずれか一項に記載の方法。
- 前記第1及び第2の利得設定での光信号の検出が、0.1〜10秒から独立に選択された持続時間にわたる、請求項24から33のいずれか一項に記載の方法。
- 前記第1及び第2の利得設定での光信号の検出が、0.5〜5秒から独立に選択された持続時間にわたる、請求項24から33のいずれか一項に記載の方法。
- コンピュータデバイスによって実行されるときに、前記コンピュータデバイスに、
前記フローセルを通って流れる細胞サンプルの細胞からの光信号を第1の利得設定で検出させ、
前記利得設定を前記第1の利得設定から第2の利得設定に変化させ、
前記フローセルを通って流れる細胞からの光信号を前記第2の利得設定で検出させる、
命令を記憶する非一時的コンピュータ可読媒体。 - 前記第2の利得設定が前記第1の利得設定よりも高い、請求項38に記載の非一時的コンピュータ可読媒体。
- 前記第2の利得設定が、高い細胞濃度を有する細胞サンプルを考慮に入れるためになされる、請求項39に記載の非一時的コンピュータ可読媒体。
- 前記第2の利得設定が前記第1の利得設定よりも低い、請求項38に記載の非一時的コンピュータ可読媒体。
- 前記第2の利得設定が、低い細胞濃度を有する細胞サンプルを考慮に入れるためになされる、請求項41に記載の非一時的コンピュータ可読媒体。
- 前記第1及び第2の利得設定が、フォトダイオードの利得設定、光電子増倍管(PMT)の利得設定、又はその両方を含む、請求項38から42のいずれか一項に記載の非一時的コンピュータ可読媒体。
- 前記細胞サンプルが血液サンプルである、請求項38から43のいずれか一項に記載の非一時的コンピュータ可読媒体。
- 前記第1の利得設定が前記第2の利得設定よりも高く、前記第1の利得設定で血小板が検出され、前記第2の利得設定で赤血球(RBC)が検出される、請求項44に記載の非一時的コンピュータ可読媒体。
- 前記第1の利得設定での光信号の検出が、前記第2の利得設定での光信号の検出と同じ持続時間にわたる、請求項38から45に記載の非一時的コンピュータ可読媒体。
- 前記第1の利得設定での光信号の検出が、前記第2の利得設定での光信号の検出とは異なる持続時間にわたる、請求項38から45に記載の非一時的コンピュータ可読媒体。
- 前記第1及び第2の利得設定での光信号の検出が、0.1〜10秒から独立に選択された持続時間にわたる、請求項38から45に記載の非一時的コンピュータ可読媒体。
- 前記第1及び第2の利得設定での光信号の検出が、0.5〜5秒から独立に選択された持続時間にわたる、請求項38から45に記載の非一時的コンピュータ可読媒体。
- フローセルと、
前記フローセルに光学的に結合された光学検出システムと、
請求項38から49のいずれか一項に記載の非一時的コンピュータ可読媒体と、を備えるフローサイトメータ。 - フローセルにおける汚染のレベルを判定する方法であって、
粒子を含むサンプルをフローセルに流すこと、
流している間に生粒子事象データを収集すること、
前記生粒子事象データ内の粒子事象の数をカウントすること、
フィルタされた信号を生成するべく逆ガウシアンフィルタ係数で前記生粒子事象データをフィルタすること、前記逆ガウシアンフィルタ係数は、期待パワースペクトル分散に基づいており、
前記フィルタされた信号のエネルギーを求めること、および
前記フローセルにおける汚染のレベルを判定するべく、前記フィルタされた信号の求めたエネルギーからクリーンなフローセルのベースラインエネルギーを差し引くこと、を含む、方法。 - 前記粒子事象の数をカウントすることが、アナログ−デジタル変換器から固定の時間間隔で生データを抽出すること、DCオフセットを判定すること、および固定の閾値を下回る生データをゼロに置き換えることを含む、請求項51に記載の方法。
- 前記閾値がフルダイナミックレンジの1%〜5%である、請求項52に記載の方法。
- 前記閾値がピーク振幅の1/e2である、請求項52に記載の方法。
- 前記サンプルが、前記フローセル内のシース流体のストリームへ流し込まれ、前記期待パワースペクトル分散が、前記シース流体の圧力に基づく、請求項51から54のいずれか一項に記載の方法。
- 前記粒子が細胞である、請求項51から55のいずれか一項に記載の方法。
- 前記サンプルが全血サンプルである、請求項56に記載の方法。
- コンピュータデバイスによって実行されるときに、前記コンピュータデバイスに、
粒子を含むサンプルが前記フローセルを通って流れる際に生粒子事象データを収集させ、
前記生粒子事象データ内の粒子事象の数をカウントさせ、
フィルタされた信号を生成するべく逆ガウシアンフィルタ係数で前記生粒子事象データをフィルタさせ、前記逆ガウシアンフィルタ係数は、期待パワースペクトル分散に基づいており、
前記フィルタされた信号のエネルギーを求めさせ、
前記フローセルにおける汚染のレベルを判定するべく、前記フィルタされた信号の求めたエネルギーからクリーンなフローセルのベースラインエネルギーを差し引かせる、
命令を記憶する非一時的コンピュータ可読媒体。 - 前記粒子事象の数をカウントすることが、アナログ−デジタル変換器から固定の時間間隔で生データを抽出すること、DCオフセットを判定すること、および固定の閾値を下回る生データをゼロに置き換えることを含む、請求項58に記載の非一時的コンピュータ可読媒体。
