Abstract
Description
表面はまた、曲面、凸面、または凹面であってもよい(例えば、粒子の表面)。検体は(存在する場合)、前記2つの物体の一方または両方に結合し、それにより、テザー粒子の運動を変化させる。その結果、検体の存在下では、空間座標パラメータの性質が変化する。したがって、測定された座標パラメータの性質の変化に基づき、検体の存在及び/または濃度を判定することができる。
Claims (7)
- テザー粒子運動を用いて検体をセンシングするための方法であって、
検体を含んでいるマトリックスを、表面と、第1の端部が前記表面に結合され、かつ第2の端部が機能化粒子に結合されたテザー分子とを有するセンサデバイスに接触させるステップであって、前記機能化粒子が、該粒子が前記表面に結合された第1の状態と、該粒子が前記表面に結合されていない第2の状態とを有し、該粒子が、前記検体の存在の有無に応じて前記第1の状態と前記第2の状態との間で切り替わり、これにより、前記検体の存在の有無に応じて機能化粒子の運動特性が変化するステップと、
前記表面に対する前記機能化粒子の空間座標パラメータを測定するステップと、
前記測定された空間座標パラメータの変化に基づき前記検体の存在/濃度を判定するステップとを有する方法。 - 前記空間座標パラメータを測定する前記ステップが、
前記機能化粒子及び/または表面を照明するステップと、
前記機能化粒子及び/または表面からの光放射を検出するステップと、
前記光放射から、前記表面に対する前記機能化粒子の位置、方向、及び/または速度を判定するステップとを含むことを特徴とする請求項1に記載の方法。 - 前記空間座標パラメータを測定する前記ステップが、
前記機能化粒子及び/または表面の自由電荷キャリアを励起するステップと、
前記機能化粒子及び/または表面からの光放射を検出するステップとを含み、
前記励起及び/または検出が、前記機能化粒子及び/または表面のプラズモン共鳴の近傍の波長で実施され、
前記検体の存在/濃度を判定する前記ステップが、前記検出された光放射の変化を測定するステップを含むことを特徴とする請求項1に記載の方法。 - 前記検出された空間座標パラメータの変化に基づき前記検体の存在/濃度を判定する前記ステップが、
粒子局在化の分布、局在化パターンの面積の変化、または粒子局在化間のステップサイズの変化に基づき前記検体の存在/濃度を判定するステップを含むことを特徴とする請求項1に記載の方法。 - 前記空間座標パラメータを測定する前記ステップが、
5nmよりも大きい線幅を有する光源または高輝度発光ダイオードによって前記機能化粒子及び/または表面を照明するステップを含むことを特徴とする請求項1に記載の方法。 - 前記検体の存在/濃度を判定する前記ステップが、
背景ノイズを抑制し、特異性を向上させるために、ヒストグラム及び/またはヒストグラム処理を実施するステップを含むことを特徴とする請求項1に記載の方法。 - 前記機能化粒子の前記空間座標パラメータを測定するステップが、
前記粒子の位置、前記粒子の方向、前記粒子の角速度、または前記粒子の線速度を測定するステップを含むことを特徴とする請求項1に記載の方法。
