Abstract
Description
かかる状況を鑑み、本発明は、マイクロダスト貪食促進剤を提供することを課題とする。
て、前記植物はアイブライト(Euphrasia officinalis L.)であることが好ましい。
また、本発明の別の態様は、本発明のマイクロダスト貪食促進剤を含有する、肌ダメージ改善用の組成物である。
で知られるハーブの一種である。
抽出は、アイブライト等のハマウツボ科コゴメグサ属に属する植物の植物体、地上部、根茎部、及び/又は種子の乾燥物1質量部に対して溶媒を1〜30質量部加え、室温であれば数日間、溶媒の沸点付近の温度であれば数時間浸漬することにより行うことができる。浸漬後は、室温まで冷却し、所望により不要物を除去した後、溶媒を減圧濃縮等により除去すればよい。しかる後、シリカゲルやイオン交換樹脂を充填したカラムクロマトグラフィーなどで分画生成して、所望の抽出物を得ることができる。
抽出に用いる溶媒としては、極性溶媒が好ましく、例えば、水;エタノール、イソプロピルアルコール、ブタノール等のアルコール類;1,3−ブタンジオール、ポリプロピレングリコール等の多価アルコール類;アセトン、メチルエチルケトン等のケトン類;及びジエチルエーテル、テトラヒドロフラン等のエーテル類から選択される1種又は2種以上が好ましく挙げられる。
ここで、マイクロダストとは、肌に悪影響を与える微小物質全般を言い、特に限定されないが、例えば自動車、火力発電所、焼却炉、暖炉、タバコなどの排煙、火山噴火による噴出物、土壌粒子などが由来の粒子状物質(PM)、粉塵、硫黄酸化物(二酸化硫黄など)、窒素酸化物(二酸化窒素など)などの排出ガス、石綿などの、化学物質や生体異物を言う。また、通常、マイクロスケールの粒子であり、例えば長径20μm以下のものである。
込むことが知られているが、マクロファージやランゲルハンス細胞と同様にマイクロダスト等の20μm以下の微小粒子に対しても貪食作用を発揮し、細胞内に取り込むことが知られている(V. B. Morhenn et al., Dermatologic Surgery, vol. 28, no. 6, (2002) 484-490)。
本発明のマイクロダスト貪食促進剤は、ケラチノサイトのマイクロダストに対する貪食作用を、非適用時に比べて促進(増強)させ、その取り込み量を増加させることができる。
マイクロダストを貪食により取り込んだケラチノサイトは、ターンオーバーの過程で、通常、マイクロダストを抱えたまま外層に向けて上昇していき、剥離して体外へ排出される。
本発明のマイクロダスト貪食促進剤の適用量は、特に限定されないが、例えば、前記植物の抽出物の固形分として、1日当たり1〜200mgを1回又は数回に分けて摂取されることが好ましい。
ニウム塩等のカチオン界面活性剤類、ソルビタン脂肪酸エステル、脂肪酸モノグリセライド、これらのポリオキシエチレン付加物、ポリオキシエチレンアルキルエーテル、ポリオキシエチレン脂肪酸エステル等の非イオン界面活性剤類、ポリエチレングリコール、グリセリン、1,3−ブタンジオール等の多価アルコール類、糖アルコール類、増粘・ゲル化剤、酸化防止剤、紫外線吸収剤や紫外線散乱剤等の紫外線防御剤、色剤、防腐剤、pH調整剤、粉体、美白成分や抗シワ成分等の各種有効成分、各種植物エキス、などが好適に例示できる。
アイブライト抽出物によるマイクロダスト貪食促進作用を、以下の手順で評価した。
ヒト正常ケラチノサイト(凍結NHEK 新生児由来、クラボウ社製品)を、4ウェルチャンバープレートにそれぞれ7.0×104個/ウェルとなるように播種し、HuMedia-KG2 クラボウ製増殖用培地(クラボウ社製品)を用いて37℃、5%CO2環境下で培養した。播種24時間後、直径0.5μmの蛍光マイクロビーズ(TetraSpeckTM Microspheres,
0.5μm, fluorescent、ThermoFisher社製)を5×106個/ウェル、及びアイブライ
トエキス(香栄興業株式会社製品)を最終濃度0質量%又は0.3質量%となるように添加し、18時間インキュベーションした。その後PBS(−)で細胞を洗浄して培地中のマイクロビーズを除去し、4%パラホルムアルデヒドで細胞を固定し、封入した。それぞれについて共焦点顕微鏡にて蛍光観察した。
図1に示される結果の通り、アイブライトエキスを添加したケラチノサイトでは、マイクロダストの貪食作用が促進されたことが認められた。
Claims (3)
- ハマウツボ科(Orobanchaceae)コゴメグサ属(Euphrasia)に属する植物の抽出物からなる、マイクロダスト貪食促進剤。
- 前記植物が、アイブライト(Euphrasia officinalis L.)である、請求項1に記載のマイクロダスト貪食促進剤。
- 請求項1又は2に記載のマイクロダスト貪食促進剤を含有する、肌ダメージ改善用の組成物。
