(a)HAの少なくとも1つのヒドロキシル基の活性化を得るため、極性非プロトン性溶媒中のヒアルロン酸(HA)塩を、炭酸フェニルエステルまたはギ酸フェニルエステルの間で選択される炭酸化剤と接触させるステップであって、前記HAは、前記極性非プロトン性有機溶媒に可溶な塩の形態であるステップ;
(b)求核置換によってHA−EDAを得るため、ステップ(a)で得られた活性化HA塩をエチレンジアミン(NH2−CH2−CH2−NH2、EDAと表記される)と接触させるステップ;
(c)求核置換によってHA−EDA−MAを得るため、ステップ(b)で得られたHA−EDAをメタクリル酸無水物(MAと表記される)と接触させるステップ;
を含み、
好ましくは上記のすべてのステップは同じ容器で行われる
手順は、本発明のさらなる主題である。
・ リン酸塩緩衝溶液pH7.2に単独で溶解させたPEP FMは、低温、たとえば4℃で保存しても経時的にその活性を徐々に失う。
・ リン酸塩緩衝溶液pH7.2に単独で溶解させたPEP FMは、凍結保護物質の存在下でも溶液の凍結乾燥後、完全にその活性を失う;
・ リン酸塩緩衝溶液pH7.2に単独で溶解させ、366nmで10分間光照射したPEP FMは、光照射の間にその活性を維持するが、溶液の凍結乾燥後、凍結保護物質の存在下でも完全にその活性を失う;
・ HA−EDA−MA誘導体の存在下でリン酸塩緩衝溶液pH7.2に溶解させ、366nmで10分間光照射したPEP FMは、凍結保護物質の非存在下および存在下の両方で凍結乾燥後もその活性を維持する;
・ 光照射の間にPEP FMを導入し、ゲルとして回収したHA−EDA−MAのヒドロゲルは、それらの調製直後に解析した場合、人工腸液pH7.2中で約70%の酵素を24時間まで活性型として持続的に放出することができる。HA−EDA−MAゲルに残存するPEP FMの量は、完全にその活性を維持する。
・ 光照射の間にPEP FMを導入し、ゲルとして回収したHA−EDA−MAのヒドロゲルは、4℃で10日間保存後に解析した場合、人工腸液pH7.2中で約60%の酵素を24時間まで活性型として持続的に放出することができ、活性の消失が起きるのはごく一部である(約20%)。
・ 凍結保護物質の非存在下で光照射の間にPEP FMを導入し、凍結乾燥粉末として回収したHA−EDA−MAのヒドロゲルは、それらの調製直後に解析した場合、人工腸液pH7.2中で約70%の酵素を24時間まで活性型として持続的に放出することができる。HA−EDA−MAヒドロゲルに残存するPEP FMの量は、完全にその活性を維持する。
・ 凍結保護物質の非存在下で光照射の間にPEP FMを導入し、凍結乾燥粉末として回収したHA−EDA−MAのヒドロゲルは、4℃または−20℃で10日間保存後に解析した場合、人工腸液pH7.2中で約50%の酵素を24時間まで活性型として持続的に放出することができ、活性の消失が起きるのはごく一部である(約30%)。これは、凍結乾燥後にPEP FMは、単独すなわちHA−EDA−MAヒドロゲルの非存在下で完全にその活性を失うため、非常に良好な結果である。
・ ポリマーの重量に対して1.5%w/wのトレハロース(凍結保護物質の例として)の存在下で光照射の間にPEP FMを導入し、凍結乾燥粉末として回収したHA−EDA−MAのヒドロゲルは、それらの調製直後に解析した場合、人工腸液pH7.2中で約60%の酵素を24時間まで活性型として持続的に放出することができる。HA−EDA−MAヒドロゲルに残存するPEP FMの量は、完全にその活性を維持する。
これらのサンプルは4℃で10日間保存される場合、人工腸液pH7.2中で24時間まで活性型として約50%のPEP FMをなお持続的に放出することができ、活性の消失が起きるのはごく一部である(約30%)。
これらのサンプルは−20℃で10日間保存される場合、人工腸液pH7.2中で24時間まで活性型として約50%のPEP FMをなお持続的に放出することができ、PEP FMは完全にその活性を維持する。
・ ポリマーの重量に対して3%w/wのトレハロース(凍結保護物質の例として)の存在下で光照射の間にPEP FMを導入し、凍結乾燥粉末として回収したHA−EDA−MAのヒドロゲルは、それらの調製直後に解析した場合、人工腸液pH7.2中で約50%の酵素を24時間まで活性型として持続的に放出することができる。HA−EDA−MAヒドロゲルに残存するPEP FMの量は、完全にその活性を維持する。
これらのサンプルは4℃または−20℃で1および2ヶ月保存される場合、人工腸液pH7.2中で24時間まで活性型として約50%のPEP FMをなお持続的に放出することができ、PEP FMは完全にその活性を維持することが立証される。
− 導入した酵素を凍結乾燥プロセスから保護することができる;
− 導入した酵素を、凍結保護物質の存在下で全体として、凍結乾燥粉末の保存中の変化から保護することができる;
− 人工胃液(酸活性酵素の場合)および人工腸液(すべての酵素の場合)において活性型としての酵素の持続的な放出を可能にすることができる、
− セリアック病患者のグリアジンペプチドを解毒できる活性型としての酵素の経口投与および放出のため、腸溶コーティングを用いてまたは用いずに顆粒、カプセルおよび錠剤のような従来の経口剤形を調製するのに使用することができる。
ワンポット合成によるHA−EDA−MA誘導体の調製
水酸化テトラブチルアンモニウム溶液を用いてヒアルロン酸溶液の中和により調製したヒアルロン酸のテトラブチルアンモニウム塩(HA−TBA)1gを、90mlの無水ジメチルスルホキシド(DMSO)に溶解させた(ヒアルロン酸の重量平均分子量260kDa)。
HA−EDA−MA誘導体の光架橋
実施例1に従い得られた30mgのHA−EDA−MA誘導体を、6%w/vに等しい最終濃度を有すべく、500μlの0.05Mリン酸塩緩衝溶液pH7.2に室温で溶解させ、真空下で脱気した。次いで、この溶液をパイレックス(登録商標)管に入れ、Rayonet photoreactor UVを用いて366nmの波長で10分間照射した(図2を参照)。この後、得られたヒドロゲルをゲルとして回収するか、または凍結乾燥して粉末を得た。
PEP FM活性の評価
フラボバクテリウム・メニンゴセプチカム(Flavobacterium meninosepticum)(FM)に由来するPEPの活性を、当該酵素がその特異的基質、すなわちカルボベンゾキシ−Gly−Pro−p−ニトロアニリド(Z−Gly−Prp−pNA)と反応する比色アッセイにより測定した。
リン酸塩緩衝溶液pH7.2中のPEP FMの、時間の関数としての安定性
0.05M リン酸塩緩衝溶液pH7.2中の0.2U/mlのPEP FMのアリコート(1.0ml)を調製し、その経時的な安定性を評価するため5日間まで4℃で冷蔵庫に保管した。1日後、2日後、3日後、4日後および5日後、酵素の活性を、実施例3で報告したように比色アッセイにより評価した。各実験は3回ずつ行った。結果を図3に示す。
リン酸塩緩衝溶液pH7.2中のPEP FMの凍結乾燥およびその活性の評価
トレハロース(7.5μg/mlまたは15μg/ml)の非存在下または存在下、0.05M リン酸塩緩衝溶液pH7.2中の0.2U/mlのPEP FM1mlを凍結乾燥した。次いで酵素の活性を実施例3で報告したように比色アッセイにより評価した。各実験は3回ずつ行った。すべての場合において、活性は認められなかった。
リン酸塩緩衝溶液pH7.2中のPEP FMの光照射およびその活性の評価
0.05M リン酸塩緩衝溶液pH7.2中の0.2U/mlのPEP FM1mlを、366nmに等しい波長で様々な時間(1〜20分)光照射した。
リン酸塩緩衝溶液pH7.2中のPEP FMの光照射後の凍結乾燥およびその活性の評価
0.05M リン酸塩緩衝溶液(PBS)pH7.2中の0.2U/mlのPEP FM1mlを、366nmに等しい波長で10分間光照射した。
トレハロースの存在下でのリン酸塩緩衝溶液pH7.2中のPEP FMの光照射後の凍結乾燥およびその活性の評価
0.05M リン酸塩緩衝溶液pH7.2中の0.2U/mlのPEP FM200μlを、0.05M リン酸塩緩衝溶液pH7.2中の15μg/mlのトレハロース800μlと混合した。この溶液を366nmに等しい波長で10分間光照射した。
PEP FMを導入したHA−EDA−MAゲルの調製
30mgのHA−EDA−MA誘導体を、400μlの0.05M リン酸塩緩衝溶液pH7.2に溶解させた。この溶液を真空下で脱気し、次いで0.05M リン酸塩緩衝溶液pH7.2中の0.4U/mgのPEP FM100μlを加えた。ここにおいて最終ポリマー濃度は、6%w/vに等しかった。この溶液を366nmに等しい波長で10分間光照射した。
PEP FMを導入した凍結乾燥粉末としてのHA−EDA−MAヒドロゲルの調製
0.4U/mgのポリマーのPEP FMを導入したHA−EDA−MAヒドロゲルを、実施例9で報告したように調製した。
1.5%w/wのトレハロースの存在下でPEP FMを導入した凍結乾燥粉末としてのHA−EDA−MAヒドロゲルの調製
30mgのHA−EDA−MA誘導体を、350μlの0.05M リン酸塩緩衝溶液pH7.2と、0.05M リン酸塩緩衝溶液pH7.2中の4.5mg/mlのトレハロース100μlとの混合物に溶解させた。この溶液を真空下で脱気し、次いで0.05M リン酸塩緩衝溶液pH7.2中の0.4U/mgのPEP FM50μlを加えた。この溶液を366nmに等しい波長で10分間光照射した。
3%w/wのトレハロースの存在下でPEP FMを導入した凍結乾燥粉末としてのHA−EDA−MAヒドロゲルの調製
30mgのHA−EDA−MA誘導体を、350μlの0.05M リン酸塩緩衝溶液pH7.2と、0.05M リン酸塩緩衝溶液pH7.2中の9mg/mlのトレハロース100μlとの混合物に溶解させた。この溶液を真空下で脱気し、次いで0.05M リン酸塩緩衝溶液pH7.2中の0.4U/mgのPEP FM50μlを加えた。この溶液を366nmに等しい波長で10分間光照射した。
人工腸液pH7.2中の実施例9のサンプルからの放出試験
PEP FMを導入したHA−EDA−MAゲルのアリコート(15mg)を、10mlの人工腸液pH7.2を含むバイアルに24時間入れた(100rpm、37℃)。所定の時間間隔で、0.15mlの放出媒体を採取し、アッセイにより解析して実施例3に記載されているように酵素活性を判定した。等量の新鮮な媒体を加え、シンク条件を、実験を通して維持した。酵素の量は、検量線(y=1,5836×+0,0537、r2=0.9981)を用いて判定した。
人工腸液pH7.2中の実施例10のサンプルからの放出試験
凍結保護物質の非存在下でPEP FMを導入した凍結乾燥粉末としてのHA−EDA−MAヒドロゲルからのPEP FM(実施例10)の放出試験を、実施例13に記載されているように行った。放出実験の結果を図6に示す。
人工腸液pH7.2中の実施例11のサンプルからの放出試験
トレハロース(1.5%w/w)の存在下でPEP FMを導入した凍結乾燥粉末としてのHA−EDA−MAヒドロゲルからのPEP FM(実施例11)の放出試験を実施例13に記載されているように行った。放出実験の結果を図7に示す。
リン酸塩緩衝溶液pH7.2中の実施例12のサンプルからの放出試験
トレハロース(3%w/w)の存在下でPEP FMを導入した凍結乾燥粉末としてのHA−EDA−MAヒドロゲルからのPEP FM(実施例12)の放出試験を実施例13に記載されているように行った。放出実験の結果を図8、9および10に示す。
Claims (11)
- プロリルエンドペプチダーゼ(PEP)、エンドプロテアーゼ(EP)およびそれらの組み合わせからなる群から選択され、光架橋型メタクリルヒアルロン酸誘導体(HA−EDA−MA)ヒドロゲルに包括された少なくとも1つの外来酵素を含む組成物であって、前記ヒアルロン酸誘導体は50,000〜1,500,000ダルトンの分子量のヒアルロン酸(HA)またはその塩を含み、少なくとも1つのヒドロキシル基が、炭酸フェニルエステルまたはギ酸フェニルエステルの間で選択される炭酸化剤で活性化後、エチレンジアミン(EDA)との反応およびその後のメタクリル酸無水物(MA)との反応で官能基化されている、組成物。
- 前記酵素は1mU/mg〜100U/mgのポリマー濃度で前記ヒドロゲルに包括される、請求項1に記載の組成物。
- 前記組成物はゲルの形態である、請求項1〜2のいずれか一項に記載の組成物。
- 前記組成物は凍結乾燥粉末の形態である、請求項1〜2のいずれか一項に記載の組成物。
- ポリマーの重量に対して0.1〜10%w/wの濃度で少なくとも1つの凍結保護物質をさらに含み;前記凍結保護物質は好ましくはトレハロースである、請求項4に記載の組成物。
- 前記酵素は、PEPである場合、フラボバクテリウム・メニンゴセプチカム(Flavobacterium meningosepticum)(FM)、ミキソコッカス・ザンサス(Myxococcus xanthus)(MX)、スフィンゴモナス・カプスラータ(Sphingomonas capsulata)(SC)もしくはアスペルギルス・ニガー(Aspergillus niger)(AN)またはそれらの組み合わせからなる群から選択される微生物に由来し、EPである場合、オオムギエンドプロテアーゼである、請求項1〜5のいずれか一項に記載の組成物。
- セリアック病の処置に使用するための、請求項1〜6のいずれか一項に記載の組成物。
- 請求項1〜6のいずれか一項に記載の組成物を含む経口投与用の医薬製剤。
- 請求項1〜6のいずれか一項に記載の組成物を調製するためのプロセスであって、1%w/v〜20%w/vの濃度の水溶液中にて、180〜800nmの範囲の波長での光照射によってHA−EDA−MA誘導体を光架橋するステップを含み、
前記光架橋は、1mU/mg〜100U/mgのポリマー濃度の少なくとも1つの外来酵素の存在下で行われるか;または
前記光架橋は前記外来酵素の非存在下で行われ、その後前記外来酵素は、得られたヒドロゲルを1mU/mg〜100U/mgのポリマー濃度の外来酵素の溶液と接触させることにより導入され;
前記酵素はプロリルエンドペプチダーゼ(PEP)、エンドプロテアーゼ(EP)およびそれらの組み合わせからなる群から選択され;
前記HA−EDA−MA誘導体は50,000〜1,500,000ダルトンの分子量のヒアルロン酸(HA)またはその塩を含み、
少なくとも1つのヒドロキシル基がエチレンジアミン(EDA)との反応およびその後のメタクリル酸無水物(MA)との反応で官能基化されている、プロセス。 - 以下のステップ:
(a)ヒアルロン酸(HA)の少なくとも1つのヒドロキシル基の活性化を得るため、極性非プロトン性溶媒中のHA塩を、炭酸フェニルエステルまたはギ酸フェニルエステルの間で選択される炭酸化剤と接触させるステップであって、前記HAは前記極性非プロトン性有機溶媒に可溶な塩の形態である、ステップ;
(b)求核置換によってHA−EDAを得るため、前記ステップ(a)で得られた活性化HA塩をエチレンジアミン(NH2−CH2−CH2−NH2、EDAと表記される)と接触させるステップ;
(c)求核置換によってHA−EDA−MAを得るため、前記ステップ(b)で得られたHA−EDAをメタクリル酸無水物(MAと表記される)と接触させるステップ、
によりHA−EDA−MA誘導体を調製するステップを含む、請求項9に記載のプロセス。 - 前記HA−EDA−MA誘導体を調製するステップはすべて続いて同じ容器で行われる、請求項10に記載のプロセス。
