Abstract
Description
本発明は改善された腫瘍崩壊ワクシニアウィルスを提供する。発明者は、腫瘍組織では、先行技術と比較して、不活性化されたN1L遺伝子を備えたTK−欠乏ワクシニアウィルス株が、高められた選択性および反腫瘍効能を示すことを見いだした。本発明はさらにワクシニアウィルス・ベクターを提供する。
本発明の最初の態様によれば、少なくとも3つのワクシニアウィルス・プロモータを含む核酸配列であって、該少なくとも3つのプロモータが核酸配列内で同じオリエンテーションに配置されるものが提供される。線形のDNAは2つの可能なオリエンテーション、5’から3’方向と、3’から5’方向を有している。例えば、同じポリヌクレオチド分子/鎖内で第1のプロモータが5’から3’方向に配置され、第2のプロモータも5’から3’方向に配置される場合、2つのプロモータは同じオリエンテーションに配置されている。
用語は、さらにDNAのより長い部分を格納することができる酵母人工染色体およびバクテリア人工染色体を含んでいる。
使用される腫瘍細胞株はすべて、発明者の研究所、ATCCまたは癌研究英国細胞株サービスユニットのいずれか、あるいは共同研究者の研究所に貯蔵されている。ヒト癌細胞株はすべてSTR分析によって遺伝子型が決定された。この研究の中で使用されるネズミ科の腫瘍細胞株は次のものを含んでいる:CMT93(結腸直腸)、LLC(ルイス肺癌)およびB16−F10(転移性黒色腫)が、C57BL/6株に由来する一方、結腸直腸癌細胞株CT26はBALB/c株に由来した。SCC7はC3H/Henハツカネズミからの頭頸部癌を派生した扁平上皮癌で、Dr Osam Mazda(Department of Microbiology,Kyoto Prefectural University of Medicine,Japan)によって寄贈された。MOSECはネズミ科の卵巣癌細胞株である。Panc02は化学的に引き起こされたネズミ科の膵癌細胞株である;DT6606(膵癌)は、K−Rasでコンディショニングされた膵臓の中の突然変異を有するC57BL/6株トランスジェニックマウスに由来する。これは、David Tuveson(CRUK,Cambridge Research Institute,Cambridge,UK)教授の親切な贈り物だった。さらに、我々のグループは、若いオバルブミン遺伝子(DT6606−ova)を含むプラスミドを備えたDT6606細胞株を安定してトランスフェクトした。最後に、CV1はATCC、VA、アメリカから得られたアフリカ緑ザルの「正常な」腎臓細胞株であり、ウイルスの滴定分析およびウイルスの大量製造を促進するために在庫の細胞株として使用された。本発明の中で使用されるヒト癌細胞株は次のものを含んでいる:人間の膵癌細胞株SUIT−2、MIAPaCa2、PANC1、PT−45およびCapan−2;人間の結腸直腸癌細胞株HT29、HCT116およびSW620、胃の腺癌MKN45および人間の卵巣癌細胞株A2780。
VVL15は、Timiryasova TM et al.Biotechniques.2001;31:534,6,8−40に記載されている、生体外の細胞内の組換え技術をそれぞれ使用して、合成初期/後期とp7.5プロモータの管理の下で、ワクシニアウィルスのLister株のTK領域の中へのlacZリポーターおよびホタルルシフェラーゼ遺伝子の挿入によって構築された。VVL15はTK−欠乏である。
原性のウイルスのエクスパンジョンは、急速に2度凍結融解され、CV1細胞(80−90%のコンフリューエンス)を含むフラスコに、36−40のT175を感染させるのに必要な、5%のFCS CMの必要な体積へ薄められた。48時間後に、感染されたCV1細胞を収穫し、そして5分(4℃)間、2,000毎分回転数の速度で遠心分離を繰り返し、単一のペレットに集められた。ペレットはPBSの中で洗われ、10mMのトリス−HCl(pH9)バッファーの12mlに懸濁され、後日の浄化のために−80℃で貯蔵された。
上記で得られた濃縮ウイルスの溶解物懸濁液は2度凍結融解され、ダウンス型ホモジナイザー(Thermofisher)に移され、60ストロークで均質化された。その後、それは30秒間超音波処理に供された。5分間4℃で2,000毎分回転数での遠心分離に続いて、上澄み(リリースされたビリオン粒子状物質を含む)は集められ、10mMトリス−HClバッファーで30mlの全容積に薄められた。その溶液は4つのアリコートに分割された;それぞれは、36mlのBeckman超遠心分離機チューブ中の36%グルコース溶液の17mlの上に優しく層にされ、4℃で80分間13,500毎分回転数で遠心分離機にかけられた。結果として生じるペレット剤は、10mMトリス−HClの中に合計16mlまで再懸濁され、再び4つに分割され、各チューブの表面近傍で25%w/mから底部で40%に勾配形成され、4つのグルコース勾配上に注意深く層にされた。第2ラウンドの超遠心分離が行なわれた。これはハツカネズミへ静脈内に投与された時それは有毒になりえる微粒子の細胞残屑を削除するのに必要である。最終ペレット剤は、ウイルスの再懸濁液バッファー(PBS;10%グリセロール;138mM NaCl;pH7.4)の1−4ml中に再懸濁された。清浄化されたウイルスのサンプルは下記に述べられるようにTCID50分析によって滴定された。
細胞は2〜4×105細胞/ウエルでシードされ、成長速度によって、10%FCSを有するミディアム中の6−ウエルプレートの3つのウエルに、1PFU/細胞のワクシニアウィルスを16−18時間後に感染させた。サンプルは、72時間まで24時間の間隔で3回繰り返して収穫された。
ウィルス複製は、Wang et al(J Clin Invest,2009,119:1604−1615)に述べられているようにTCID50(50%組織培養感染量)によって測定された。
もし、言及されない場合には、Graphpad Prism5が相対的な統計分析に使用された。2重の条件比較はアンペアードスチューデントt検定を使用してなされた。1つを超える条件、あるいは補足変数、たとえば時間については、1あるいは2ウエイ分散分析(ANOVA)がそれぞれ行なわれた。ポストホックテスト(1ウエイ分散分析用のKnewman−Keulsおよび2ウエイ分散分析用のBonferroni)は、実験内の特定の組の条件を比較した。生存データは、ログランクのKaplan−Meierプロットとして示され、グループ間のすべての違いも統計的に有意だったか否かを示した。
例1:pUC19−N1Lシャトルベクターの構築
発明者は、L1024とL025の31bp(左アーム)を含むフラグメント、およびL026とL025の22bp(右アーム)を含むフラグメントによりフランクされた特定の発現カセットを含む図3に示されたpUC19−N1Lシャトルベクターを構築した。発現カセットは次の特徴を有する:
(1)3つのワクシニアウィルスmH5プロモータがある。また、各プロモータには、任意の興味のある遺伝子の容易な挿入のためのクローニング制限酵素サイトがある;
(2) レポーター遺伝子RFPは組換えウイルスの正の淘汰のために1つのmH5プロモータによって進行される;
(3)mH5プロモータは、左から右に挿入された遺伝子の発現のみを進行する。
本発明の中で使用される相同組換え戦略は、L025(N1L)座のコード配列の全体をほとんど交換することを目指した。
L024/25とL025/26の交点の配列分析は、L025のORFs上流(22bp)および下流(31bp)が完全なままであることを確認した。
図4は、発明者によって作成されたワクシニアウィルス・Lister株および様々なワクシニアウィルス構成物を表す。pUC19スーパーシャトルベクターはそれぞれ、VVL15(1個の細胞当たり0.1PFU)にあらかじめ感染した(2時間早く)CV1細胞へ(Effectene−basedプロトコル−Qiagenを使用して)トランスフェクトされ。48時間後に、蛍光顕微鏡の下の、細胞質のプラスミドあるいは相同組換えが成功した比較的わずかのウイルスからの赤い蛍光の存在により、適切なカセットの発現を確認した。これらの後者は以下のように選択された。細胞と上澄みは皿から細胞をこすり落とし、2度凍結融解されることにより収穫された。この溶解物の1μlは、80−90%のコンフルエンスに成長したCV1細胞を有する6つのウェルプレートのすべてに感染させるために使用された。この低いウイルス負荷は、よく分離されたプラークの発生を保証するだろう。
細胞とメディアは48時間後にこすり取られて収穫され、「原性のウイルスのエクスパンジョン(primary viral expansion)」として維持された。
例3:組み換えのVVLsの中のN1Lの欠失の確認:
N1L遺伝子はすべての新規なVVL組み換え個体の中で削除された(図5a)。センスおよびアンチセンスN1L遺伝子プライマーは、感染したCV1細胞から抽出されたPCRウィルスDNAを介して増幅するために使用された。VVL15ウイルスだけがこの遺伝子を含んでいた。プライマー対にわたるセグメントを含むN1L遺伝子が、およそ750bpにわたると予想された。A52R遺伝子はすべてのVVL組み換え個体の中にあった。センスおよびアンチセンスA52R遺伝子プライマーは、感染したCV1細胞から抽出されたDNAからのこの座をPCR増幅に使用された。プライマー対にわたるA52R遺伝子セグメントがおよそ880bpにわたると予想された(図5b)。
各遺伝子組み換えアームド組み替えウィルス(transgene−armed recombinant virus)の最終のプラーク浄化ラウンドからの上澄みのサンプルは、メーカーのプロトコル(ebioscience、Biolegend)に従って、適切なサイトカインに特有のELISAキットを使用して分析された。腫瘍細胞感染の際に適切な組み替えウィルスによってサイトカイン遺伝子組み換えが発現したかどうか評価するために、上記のウィルス複製分析に述べられているものと同じ実験がセット・アップされた。ウィルス感染の後24、48および72時間で、上澄みはウェルの個々の複製のセットから集められた。また、サイトカインの濃度はメーカーの指示にしたがってELISAによって決定された。対照試料はVVL15−N1L感染したウェルから集められた上澄みだった。
血清を含まないDMEMの100μlの中の2x106 CT26細胞が、7週齢の雌のBALB/cハツカネズミの剃毛された右わき腹へ皮下注射された。腫瘍がおよそ100mm3の時、それらは2つのグループへ無作為に分けられた。VVL15あるいはVVL15−N1Lのいずれかの1x108 PFUのIV投与量は、尾静脈に注射された。ウイルス注射の1、3、7および10日後に、CO2吸入によって各グループからの3匹のハツカネズミを殺した。血液はあらかじめヘパリン処理された1.5mlのエッペンドルフチューブの中へ、心臓の穴あけによって集められた。収穫された腫瘍、脳、肺、肝臓、ひ臓、腎臓および卵巣は、冷やされた(−80℃に)イソペンタンに浮かぶペトリ皿上で直ちにスナップ凝固(snap−frozen)された。融解された日の後に、それらは血清非含有DMEMの中で計量され、均質化された(あるいは血液の場合には攪拌された)。サンプルは、1mg当たり5μl(あるいは血液の1μl当たり5μl)で薄められた。さらなる凍結融解サイクルに続き、組織ホモジェネートは、TCID50分析を使用して、生存ウイルスPFUについて滴定された。生物学的分布実験は、IV−デリバードウイルスが腫瘍に播種されたか否かを決定し、オフターゲット複製(off−target replication)の決定(つまり腫瘍選択性およびしたがって安全性の程度の決定)をするために行われた。CT26の皮下のひ腹モデルは方法セクションに述べられているようにして、このセクションのために利用された。尾静脈注入に続いて、注入の後少なくとも10日まで、腫瘍組織から両方のウイルスを回収した。ピーク・タイターは3〜7日である。予想外に、VVL15N1Lのウイルスの回収はVVL15と比較して、腫瘍組織で低減されなかった(図6a)。
腫瘍細胞がワクシニアウィルスによって殺すことができる多数の機構がある。これらは、生得の宿主側防御機構により引き起こされた、アポトーシス、ウイルスを媒介とした細胞のバーストからの死、免疫的生体防御反応を含む。ウイルスが細胞へ過度に細胞毒性の場合、それは腫瘍の全体にわたって自分で繁殖するのに十分な後代を生成しないかもしれない。更に、ウイルスのより多くのコピーが存在するので、模写するその能力がその治療の遺伝子組み換えの発現と関連すると予想されることができる。
VVL15−N1Lは、一連のネズミ科の癌細胞株の細胞毒性についてVVL15−RFPと生体外で比較された。細胞は成長速度によって、96−ウエルプレート内で、1ウエルあたり1x103または1x104の細胞で接種され、16−18時間後にウイルスに感染させた。ウィルス感染の後の6日目の細胞生存はMTS分析によって決定された。また、Wang et al(J Clin Invest,2009,119:1604−1615)に述べられている方法により、EC50値(腫瘍細胞の50%を殺すウイルスの投与量)が計算された。分析はすべて、少なくとも3回行なわれた。EC50値(つまり細胞の50%を殺すのに必要なPFU)に基づいて、CT26とDT6606の細胞には2つのウイルス間の細胞毒性に有意差はなかった。対照的に、CMT93、LLCおよびSCC7細胞を殺すことでVVL15と比較して、VVL15−N1Lは著しくより有力だった(図8)。
3つの共通遺伝子の生体内の腫瘍モデルが使用された:
C3H/HeN株ハツカネズミ中のSCC7細胞;
C57BL/6株ハツカネズミの中のDT6606−ova細胞、および
C57BL/6株ハツカネズミの中のLLC細胞。
ウイルスおよび腫瘍特異性のT細胞の適時の生成を保証するために、ひ臓は感染の14日後に収穫された。IFN−γ放出分析が続いて生成された脾細胞懸濁液について行なわれた。
(1) 皮下のひ腹腫瘍ひ臓共通遺伝子(syngeneic subcutaneous flank tumours)からは、下記に述べられる(さらに表2を参照)適切な腫瘍細胞株について確立された。腫瘍が体積でおよそ100mm3だった時、ハツカネズミは3つのグループへ無作為に分類された。PBSの50μl中のVVL15あるいはVVL15−N1Lウイルスのいずれかの1x108PFUは、29ゲージ針を付けられた1mlのインスリン用注射筒を使用して腫瘍内(IT)に注射された。針は、広い普及のためにウイルス配置中に腫瘍の全体にわたり異なる方向で多数回を刺された。第3のグループは、ビヒクルバッファー(つまりPBSの50μl)の等価な体積を注射された。感染の14日後に、動物はCO2吸入によって安楽死させられた。ひ臓は無菌条件の下で収穫され、2mlの注射器のプランジャーの平面端部を使用して、70μm細胞ストレーナ(Becton Dickinson Falcon)によってつぶされて、50mlの三角フラスコの中へT細胞培地(TCM)(RPMI−1640、10%のFCS、1%のストレプトマイシン/ペニシリン、1%のナトリウム・ピルベート)とともに注入した。小球になった脾細胞は、5mlのRBC細胞融解バッファー(シグマ−Aldrich)中に再懸濁され、1,200毎分回転数で遠心分離され、5分間氷の上に置かれた。洗浄遠心分離サイクルの後、それらは、5x106細胞/mlの終末濃度へTCMで再懸濁された。
細胞培養培地(CM)中の刺激物細胞(つまり適切な目標またはコントロール腫瘍細胞株−SCC7、LLCあるいはDT6606o−ova)の5x106/mlの単個細胞浮遊液は、50mlの三角フラスコ中で調製された。100μg/mlの終末濃度を達成するために、マイトマイシンC(MMC)(Roche)の1mg/mlの溶液がこの懸濁液に加えられ、1時間、37℃で、5%のCO2空気中で、湿潤インキュベータ中でインキュベートされた。細胞は、40mlのPBSで続いて2度洗われ、40mlのCMの中で再懸濁され、接種の準備ができるまで培養された(30−60分以内)。5x105細胞/mlの終末濃度を達成するために、成長阻止された刺激物細胞は、TCMの中で再懸濁された。
この分析は、記憶T細胞がそれらの同系統のエピトープ−MHC複合体によって活性化される時のIFN−γの放出に基づく。脾細胞プールはCD8+ T細胞の刺激に必要な細胞型(例えばAPCs、Th細胞)のすべてを含んでいるべきである。(1)からの脾細胞懸濁液の各々の100μlは、丸底の96ウエルプレートの三つのウェルの中で目標腫瘍刺激物細胞懸濁液の100μlと共培養された(つまり、5x105脾細胞と5x104成長阻止腫瘍細胞)。脾細胞のみのコントロールは、200μl TCMの中の5x105脾細胞を含んでいた。適切な場合、脾細胞も、TCM(5μg/mlの終末濃度を達成)の中の卵子ペプチド(H−2Kb/SIINFEKL、Proimmune)の100μlと共培養されるか、あるいは5x104 MHC相容性、成長阻止コントロール腫瘍細胞(C56BL/6マウス由来腫瘍モデルが使用された時B16−F10)を含むTCMの100μlで培養された。
5x106CMT93細胞あるいは3x106DT6606細胞は、C57BL/6オスのハツカネズミの剃毛された右ひ腹の皮下に上記のように注入された。一旦腫瘍体積がほぼ100mm3に達したならば、それらは、3つのグループに無作為に分けられ、50μlのPBS中のウイルスの1x108PFUの投与、または50μl PBSビヒクルバッファコントロールが、表3(スケジュール1および2)に概説された治療スケジュールごとに注射された。腫瘍体積は毎週2度のカリパス測定によってモニターされた。また、ハツカネズミは毎週計量された。腫瘍増殖は毎週2度追跡された。また、腫瘍体積が1000mm3に接近した時、ホームオフィス・ガイドラインによって管理され、動物は安楽死させられた。DT6606ひ腹腫瘍モデルの処理の際のVVL15−N1L作用薬は、腫瘍成長速度の減少および延長された生存について統計的に有意であった(図13a−b)が、CMT93モデルではウイルス剤のIT適用に続く腫瘍成長速度間の違いはなかった。ただし両方ともPBSグループより著しく遅い成長であった(図13c−d)。
静脈内に投与された時、ウイルスが効果的かどうか評価するために、同所性の肺癌モデルが利用された。100μl PBSの中の5x106LLC細胞は、7週齢の雌のC57BL/6ハツカネズミの尾静脈に注射された。
LLCの皮下のひ腹注射に続く肺に転移する傾向を備えた非常に積極的な腫瘍モデルである。確かに、皮下に成長したLLC腫瘍の外科的切除が、肺転移の割合を高めたことは報告された。これは恐らく切除によって、血管形成阻害剤の分泌の回避が主原因であろう。
それはよい合併療法であるかもしれない。
VVL15N1Lの反腫瘍効能を高めるために、GM−CSFおよびIL−12がVVL15N1LベクターのN1L領域に挿入された。これらの組み換え個体の各々の潜在能は、共通遺伝子のDT6606の皮下のひ腹モデルに対して生体内でテストされた(表3、スケジュール1)を参照)。腫瘍体積が100mm3の平均に達した時、ウイルス(VVL15−N1L、VVL15−N1L−mGMCSFあるいはVVL15−N1L−mIL12)の1x108PFU1日用量(合計5日)あるいはビヒクル・バッファコントロール(PBSの50μl)の等価なボリュームがIT注射された(1つのグループ当たりn=7)。毎週2度のカリパス測定によって腫瘍増殖がフォローされた(図16a)。腫瘍体積が1,000mm3を超過した時、対応するKaplan Meir生存曲線(図16b)は人道的な動物犠牲の必要に基づいた。図16a−bは、GMCSFの遺伝子組み換えアームドウイルスだけがVVL15−N1Lよりも著しくよくはなかったことを実証する。しかし、IL12−アームドウイルスは実験の終わりに6/7匹のハツカネズミの治癒および100%の生存を導き、重要な潜在能を実証した。この結果の普遍性を確立するために、これらのアームドウイルスは、他のモデルの中でテストされるだろう。
IL−21の構築
物質と方法:
ネズミ科の膵臓癌モデル:
DT6606の皮下の共通遺伝子の腫瘍はオスのC57BL/6ハツカネズミの中で確立された。腫瘍が直径で5−6mmに達した時、PBS、VV−ΔTkΔN1L−mIL−21、VV−ΔTkΔN1L−hIL−21あるいはコントロール・ウイルスVV−ΔTkΔN1Lは、1、3、7、9および11日目に腫瘍内に(5x107pfu/注入)適用された。腫瘍増殖は毎週2度測定された。また、動物生存がモニターされた。生存データはプリズム(登録商標)(GraphPad Software,CA,USA)を使用して、比較された。また、ログランク(Mantel Cox)テストは生存差の有意性を決定するために使用された。有意性はアンペアードスチューデントTテスト(unpaired students T test)(*p<0.05;**p>0.01;***p<0.001)を使用して決定された。
HPD−1NR腫瘍を有するシリアンハムスター − 1x106HPD−1NR細胞は、HPD−1NR腫瘍を有するシリアンハムスターの右ひ腹の中への皮下注射によって接種された。腫瘍が313mm3に達した時、PBS、VV−ΔTkΔN1L−mIL−21、VV−ΔTkΔN1L−hIL−21あるいはコントロール・ウイルスVV−ΔTkΔN1Lは、1、3、7、9および11日目に腫瘍内に(5x107pfu/注入)適用された。腫瘍は毎週2度測定された。また、動物生存がモニターされた。生存データはプリズム(登録商標)(GraphPad Software,CA,USA)を使用して、比較された。また、ログランク(Mantel Cox)テストは生存差の有意性を決定するために使用された。有意性はアンペアードスチューデントTテスト(*p<0.05;**p>0.01;***p<0.001)を使用して決定された。
1x107SHPC6細胞はシリアンハムスターの右下の腹膜腔へ接種された。4日後に、1つのグループ当たり10のハムスターに各々500μl PBS、および異なるVVsの2x107PFUを、4、6および8日に腹膜内(IP)に注射した。ハムスターの生存率がモニターされた。生存データはプリズム(登録商標)(GraphPad Software,CA,USA)を使用して、比較された。また、ログランク(Mantel Cox)テストは生存差の有意性を決定するために使用された(* p<0.05,**p<0.01,***p<0.001)。
人間とマウスのIL−21 cDNAシーケンスが、puc19N1Lシャトルベクター(図3に示されたように)に挿入された。標準の相同組換えは、結果として生ずるプラスミドpShuttleN1LmIL−21またはpShuttleN1L− hIL−21を、0.05PFU/細胞のVVL15にあらかじめ感染したCV−1(アフリカ緑ザル腎臓)細胞と共同トランスフェクトすることにより、Lister株ワクシニアウィルス(VVL15)のTK−欠乏バックボーンの中で行なわれた。トランスフェクトされたCV−1細胞溶解物は、プラーク検定に供された。5つのプラーク浄化ラウンドが単一のクローンのウイルスを繁殖させるために行なわれた。また変性はN1Lgene欠失についてPCRによって再確認された。結果として生ずるウイルスはVV−ΔTkΔN1L−mIL−21およびVV−ΔTkΔN1L−hIL−21と命名された。これらのウイルスの潜在能をテストするために、細胞死分析(MTS分析)がネズミ科の膵癌細胞株(DT6606)内の3つのワクシニアウィルスの細胞毒性を発見するために使用された。それは突然変異体のRas−p53の遺伝子組み換えの膵臓癌モデルに由来する。
実験は3回行われた。
IL21がVV−ΔTkΔN1Lの反腫瘍効能を高めることができるかどうかテストするために、これらのウイルス(マウスIL21あるいはヒトIL21)の各々の潜在能が、共通遺伝子のDT6606の皮下のひ腹モデルに対して生体内でテストされた。
Claims (26)
- 少なくとも3つのワクシニアウィルス・プロモータを含み、該少なくとも3つプロモータが同じオリエンテーションで配置される核酸配列。
- 該プロモータがウイルスの生活環の初期および/または後期に活性を有するプロモータであることを特徴とする請求項1に記載の核酸配列。
- mH5、H5、P7.5およびPE/Lから成る群からプロモータが選ばれる請求項2に記載の核酸配列。
- 該核酸配列が異種ポリペプチドをコードすることを特徴とする請求項1に記載の核酸配列。
- ポリペプチドがサイトカインであることを特徴とする請求項4に記載の核酸配列。
- GM−CSF、IL−12およびIL−21から成る群からサイトカインが選ばれることを特徴とする請求項5に記載の核酸配列。
- 前記核酸配列が1つ以上の制限部位を含むことを特徴とする請求項1〜6のうちのいずれか1項記載の核酸配列。
- SalI、BglII、HindIII、SmaI、BamHIおよびMluIから成る群から1つ以上の制限部位が選ばれることを特徴とする請求項7に記載の核酸配列。
- ポリペプチドがリポーター・ポリペプチドであることを特徴とする請求項4に記載の核酸配列。
- 前記核酸配列が図1に表された核酸配列に本質的に一致するシーケンスを含むことを特徴とする請求項1〜9のうちのいずれか1つによる核酸配列。
- 請求項1〜10のうちのいずれか1つの核酸配列を含むワクシニアウィルス・ベクター。
- 請求項1〜11のうちのいずれか1つのベクターまたは核酸配列を含み、N1L遺伝子にベクターまたは核酸配列が挿入されていることを特徴とするワクシニアウィルス。
- 不活性化されたN1L遺伝子を含むTK−欠乏ワクシニアウィルス。
- 異種のポリペプチドをコード化する核酸配列の挿入によってN1L遺伝子が不活性化されることを特徴とする請求項13に記載のTK−欠乏ワクシニアウィルス。
- ポリペプチドがサイトカインであることを特徴とする請求項14に記載のTK−欠乏ワクシニアウィルス。
- GM−CSF、IL−12およびIL−21から成る群からポリペプチドが選ばれることを特徴とする請求項14または15に記載のTK−欠乏ワクシニアウィルス。
- 核酸配列が少なくとも3つのプロモータを含み、該プロモータが同じオリエンテーションに配置されることを特徴とする請求項14〜16のうちのいずれか1項記載のTK−欠乏ワクシニアウィルス。
- mH5、H5、P7.5およびPE/Lから成る群からプロモータが選ばれることを特徴とする請求項17に記載のTK−欠乏ワクシニアウィルス。
- 癌治療で使用される請求項1〜18のうちのいずれか1つのワクシニアウィルス、ワクシニアウィルス・ベクターあるいは核酸配列。
- TK−欠乏ワクシニアウィルスを含む組成物であって、ウイルスは不活性化されたN1L遺伝子を含み、該N1L遺伝子は異種のポリペプチドをコードするベクターまたは核酸配列の挿入によって不活性化され、任意に薬学的に受理可能なキャリアーあるいは補形薬を含む組成物。
- ポリペプチドがGM−CSF、IL−12およびIL−21から成る群から選ばれたサイトカインであることを特徴とする請求項20に記載の組成物。
- 癌治療で使用される請求項20または21に記載の組成物。
- 癌の治療のための薬物の製造における、請求項12〜18のうちのいずれか1つによるワクシニアウィルスの使用。
- TK−欠乏ワクシニアウィルスを対象に投与することを含む癌の治療方法であって、ウイルスが不活性化されたN1L遺伝子を含み、該N1L遺伝子は異種のポリペプチドをコードする核酸配列またはベクターの挿入によって不活性化される方法。
- ポリペプチドがサイトカインであることを特徴とする請求項24に記載の方法。
- GM−CSF、IL−12およびIL−21から成る群からサイトカインが選ばれることを特徴とする請求項25に記載の方法。
