Abstract
Description
本明細書に提供された実施例の理解を容易にするために、特定の頻繁に使用される方法及び/用語を説明するものである。
「特異的ハイブリダイゼーション」は、本明細書において第1のポリヌクレオチドと第2のポリヌクレオチド(例えば、第1のポリヌクレオチドとは明らかに異なるが、実質的に同一の配列)とのハイブリッド形成として定義され、前記混合物中では実質的に関連性のないポリヌクレオチド配列はハイブリッドを形成しない。
様々な方法を利用して、1若しくはそれ以上の進化される治療用タンパク質候補を発見、作成、および/または選択することができる。選択された分子は、既存の組み換えタンパク質である可能性があり、または酵素、ホルモン、および抗体を含む組み換えタンパク質の一群に由来する可能性もある。治療用タンパク質には、クローンヒト酵素およびホルモンを含む可能性がある。組み換え抗体は、いかなる数の利用できる発生およびスクリーニング基盤を用いても発見することができる。抗体は一本鎖、完全ヒト、Fab、Fv、Fc、二官能性、キメラ、ヒト化、または完全ヒト抗体またはそのフラグメントを含むいずれの形態であってもよい。1つの好適な態様では、全長抗体分子を発見するために前記CIAO法が利用される。組み換え抗体ライブラリは、最適化または非最適化哺乳類宿主の選択またはスクリーニング系により作成およびスクリーニングし、進化の候補を生み出すことができる。例えば、哺乳類または酵母細胞系など、いくつかの真核生物発現系が発表されている。
タンパク質進化のすべての方法は、タンパク質の性能および発現を最適化する同時進化を目的として採用することができる。タンパク質性能の最適化には、親和性、薬物動態学的特徴、組織ターゲッティング、タンパク質−タンパク質凝集など、様々な特徴の改善、アッセイのばらつきが大きいことへの対処、および他のin vivo特徴の修正を含む可能性がある。
図1を参照し、単純な例として直鎖ペプチドを利用すると、第1段階において、位置1からn(nはポリペプチド鎖の残基数に相当する)の各コドンにおける一連の天然型アミノ酸変異体(またはそのサブセット、またはアミノ酸誘導体)は、本明細書で包括的位置進化(CPE(商標))と呼ばれるプロセスにより作成される。この方法は、標的分子の各ポリペプチド鎖に繰り返される。最小のアミノ酸変異体セットには、天然アミノ酸19種類それぞれのコドン1つのみが含まれる。しかし、各発現系はコドンのバイアスを受けている可能性があることが認められており、この場合、不十分なtRNAプールにより、翻訳の行き詰まり、早まった翻訳終結、翻訳フレームシフト、およびアミノ酸の誤った取り込みが行われる可能性がある。従って、発現を最適化するため、各セットが停止コドンを含め、最高63種類のコドンを含む。次の段階では、新しい各分子のシークエンシングにより、突然変異を確認する。他の確認方法を採用することもできる。

好ましくは、例えば、機能および発現の改善など、複数の特徴を同時にスクリーニングする。
この方法では、タンパク質の機能(例えば、抗体の場合は抗原/受容体の結合)を失わずに、変化が起こる可能性のないすべての部位を同定することが可能である。また、機能に影響せずに変化が起こる場所も示す。さらに、宿主系で発現しない分子が生じる変化を同定するため、変異の影響を評価しない。
本発明の1つの実施形態では、図2に示すとおり、相乗的進化のEvoMap(商標)が作成され、利用される。相乗的進化では、2〜20箇所の選択された部位で同時変異を組み合わせ、コンビナトリアルな効果を生み出すことができる。前記テンプレートポリペプチドのEvoMap(商標)を利用し、特定の単一アミノ酸点突然変異を選択し、複数部位のポリペプチド変異に集合させる。
別の実施形態では、前記CPE/EvoMapを利用し、完全に突然変異する部位を同定、開発する。1つの態様では、複数の完全に突然変異する部位の開発がフレックス進化と呼ばれ、糖鎖付加(例えば、N−またはO−結合型糖鎖付加をするアミノ酸のコドン、コンセンサス配列Asn−Aa−Ser−ThrまたはSer/Thr内のAsn)および化学的結合の導入など、標的とする変化を起こすために利用する。プロテアーゼ開裂部位のデザイン、精製および/または検出用のタグ導入、部位特異的ラベリングなどに利用することもできる。さらに、サイレント変異のコドン最適化は、タンパク質発現の改善に利用することができる。この実施形態では、タンパク質発現後のフレックス進化と呼ばれる進化、作成された変異ポリペプチドライブラリを、テンプレートポリペプチドと比較し、少なくとも1つの所定の特性、特徴、または活性について、再スクリーニングする。1つの態様では、前記所定の特性に、タンパク質−タンパク質凝集の抑制、タンパク質安定性の向上、またはタンパク質溶解性の向上が含まれる。1つの態様では、前記変異ポリペプチドライブラリを、1若しくはそれ以上の特性について同時にスクリーニングする。別の態様では、真核生物発現系において、例えば、哺乳類、植物、酵母、および昆虫細胞株など、糖鎖付加部位の導入にグリコシル化を利用できる。
1つの実施形態では、当該開示により、テンプレートポリペプチドから、またはこれに基づき形成した変異ポリペプチドを同定、マッピングする方法を提供する。図8を参照し、単純な例として直鎖ペプチドを利用すると、第1段階では、位置2からn(nはポリペプチド鎖の残基数に対応する)の各コドンにおける一連の天然型アミノ酸変異体(またはそのサブセット、またはアミノ酸誘導体)は、本明細書で包括的位置挿入(CPI(商標))進化と呼ばれるプロセスにより作成される。
Comprehensive Positional Deletion Evolution(CPD(商標))は、テンプレート・ポリペプチドから形成される、または、テンプレート・ポリペプチドに基づく変異体ポリペプチドを同定する、及びマッピングする方法に関するものである。CPD進化は、一つの位置のタンパク質を介して同時に全てのアミノ酸を欠失させる。典型的には、そのポリペプチドはnアミノ酸残基を有し、その方法は、(a)n−1(最初の残基がメチオニンの場合にはn−2)分離ポリペプチドを生成する工程、ここにおいて、各ポリペプチドは単一の所定位置が欠けているという点でテンプレート・ポリペプチドとは異なるものであり、少なくとも一つの所定の性質、特徴、または活性を分析する工程、と(b)各メンバーにおいて、テンプレート・ポリペプチドに関連する前記性質、特徴または活性における任意の変化を同定する工程とを有する。
Combinatorial Protein Synthesis(CPS(商標))は、二つまたはそれ以上の突然変異を結合させるために、CPE、CPI、CPDまたは任意の他の進化の技術から個々のヒットを結合させることを含む。CPSは、最適化された遺伝子及びタンパク質の特徴においてスクリーニングされた結合された突然変異とタンパク質を合成するのに使用される。CPS(商標)の図解は図3に示されている。一態様において、結果としてアップ変異体または中立突然変異となる二つまたはそれ以上の点突然変異はCPSにおいて結合される。
一実施形態において、真核細胞系は、CHO、HEK293、IM9、DS−1、THP−1、Hep G2、COS、NIH 3T3、C33a、A549、A375、SK−MEL−28、DU 145、PC−3、HCT 116、Mia PACA−2、ACHN、Jurkat、MM1、Ovcar 3、HT 1080、Panc−1、U266、769P、BT−474、Caco−2、HCC 1954、MDA−MB−468、LnCAP、NRK−49F及びSP2/0細胞株;及びマウス脾細胞及びラビットPBMCからなる群の1つから選択される哺乳類系である。
後発生物製剤は、特許でもはや保護されていない承認された医療用医薬品として同一のアミノ酸配列(つまり化学的組成物)を有するタンパク質ベースの治療薬である。一態様において、CIAO法は特に後発生物製剤に関連がある。等価な製剤及び組成物において治療的タンパク質を生成するため、市場において競争的であるために重要であるので、後発生物製剤はできる限り急速に、及び安くつくらなければならない。細胞培養培地及びプロセスの開発は、後発生物製剤を調製し、製造する中で最もコストと時間のかかる部分の一部である。
この実施例は、フローサイトメトリー分離とELISAの組合せを用いて、標的抗原への結合活性を持つヒト抗体を単離するために、哺乳類細胞表面提示系のヒト抗体ライブラリを作成しスクリーニングする方法について記載するものである。
1. ヒト抗体ライブラリを、下記の付録1.1、1.3、及び1.4に記載されているような哺乳類細胞に安定に組み込む。
2. フローサイトメトリー解析の前に、安定完全ヒト抗体ライブラリのクローンを増大させる。
3. フローサイトメトリー解析の日に、1×PBSで1×107個の細胞を洗浄する。
4. Detachin細胞剥離培地を用いて細胞を剥離し、1×PBS中に細胞を収集する。
5. 3000rpmで5分間遠心して細胞を沈殿させる。上清を除去する。
6. 細胞沈殿物を1mlの冷却1×PBS中に再懸濁し、3000rpmで5分間遠心する。
7. 上清を除去し、精製ヒト001タンパク質を2μg/mlで含む冷却1×PBS500μl中に、細胞沈殿物を再懸濁する。
8. 手動で時々かき混ぜながら、1時間氷上でインキュベートする。
9. 3000rpmで5分間細胞を遠心沈殿させる。上清を除去する。
10. 細胞沈殿物を1mlの冷却1×PBS中に再懸濁し、3000rpmで5分間遠心する。
11. 工程7及び8を繰り返す。
12. 上清を除去し、ウサギ抗ヒト001ポリクローナル抗体を1g/mlで含む冷却10%ヤギ血清添加1×PBS500μl中に、細胞沈殿物を再懸濁する。
13. 手動で時々かき混ぜながら、1時間氷上でインキュベートする。
14. 3000rpmで5分間細胞を遠心沈殿させる。上清を除去する。
15. 細胞沈殿物を1mlの冷却1×PBS中に再懸濁し、3000rpmで5分間遠心する。
16. 工程7及び8を繰り返す。
17. 上清を除去し、ヤギ抗ウサギ抗体のFITCコンジュゲート及びヤギ抗ヒトFc抗体のピレトリン(pyroerthrin)コンジュゲートを含む冷却10%ヤギ血清添加1×PBS500μl中に、細胞沈殿物を再懸濁する。
18. 手動で時々かき混ぜながら、1時間氷上でインキュベート(incubate)する。
19. 3000rpmで5分間細胞を遠心沈殿させる。上清を除去する。
20. 細胞沈殿物を1mlの冷却1×PBS中に再懸濁し、3000rpmで5分間遠心する。
21. 工程7及び8を繰り返す。
22. 上清を除去し、冷却2%ヤギ血清添加1×PBS中に細胞沈殿物を再懸濁する。
23. Dako社MoFlo(商標)を用いてフローサイトメトリー解析を次に行う。
24. PE/FITC蛍光強度比が上位0.1%となる細胞が含まれるように分離窓(sort window)を描く。この分離窓の中に入った細胞を収集し、100μlの成長培地の入った96ウェルプレートに入れる。
1. 前記クローンを96ウェルプレートから6ウェルプレートに拡張する。この6ウェルプレートの中で、細胞が80%の密度(confluence)に達したとき、Qiagen社DNeasy Tissue Kitを用いてゲノムDNA単離を次に行う。
2. 細胞から培地を吸引除去する。500mlの1×PBSを各6ウェルに加える。
3. 細胞を3000rpmで5分間遠心する。
4. 上清を除去し、細胞沈殿物を200μlの1×PBS中に再懸濁する。
5. 20μlのプロテアーゼK及び200μlのバッファーALをサンプルに加え、ボルテックスして(vortex)完全に混合し、56℃で10分間インキュベートする。
6. 200μlのエタノールを加え、ボルテックスして完全に混合する。
7. 工程6で得た混合物をピペットでスピンカラムに移す。8000rpmで1分間遠心分離を行う。得られた通過液を捨てる。
8. 500μlのバッファーAW1を加え、8000rpmで1分間遠心分離を行う。得られた通過液を捨てる。
9. 500μlのバッファーAW2を加え、14000rpmで2分間遠心分離を行う。得られた通過液を捨てる。再度14000rpmで1分間遠心分離を行う。カラムの膜が完全に乾燥していることを確認する。
10. スピンカラムを滅菌済の微量遠心チューブに入れ、200μlのバッファーAEを直接膜上にピペットで入れる。
11. 室温で1分間インキュベートし、8000rpmで1分間遠心分離してゲノムDNAを溶出させる。
12. 1.5ml微量遠心チューブに以下の反応をセットアップすることで、ゲノムDNAの品質確認をする:
gDNA 5μl
10×サンプルローディングバッファー 5μl
総体積 10μl
0.5g/mlのエチジウムブロマイド入り0.8%アガロースTAEゲル上にロードする。1kB DNAラダーを基準として用いる。1×TAEバッファー中、100Vで20〜30分間、ゲル電気泳動を行う。
13. 滅菌済のPCRチューブ中で以下のPCR反応をセットアップする:
gDNA 1μl
2×HotStar Taq Master Mix 12.5μl
可変領域の順方向プライマー* 0.5μl
可変領域の逆方向プライマー* 0.5μl
H 2 O 10.5μl
総体積 25μl

14. このPCRチューブをサーマルサイクラー中に置き、サイクリングのプログラムを開始させる:
初期活性化段階(initial activation step):15分、95℃
3段階サイクリング
変性(denaturation):40秒、94℃
徐冷(anneealing):40秒、55℃
伸長(extension):2分、72℃
サイクル数:30回
最終伸長段階(final extension step):10分、72℃
12. 1.5ml微量遠心チューブに以下の反応をセットアップすることで、PCR反応物の品質確認をする:
PCR反応物 5μl
10×サンプルローディングバッファー 5μl
総体積 10μl
0.5g/mlのエチジウムブロマイド入り1%アガロースTAEゲル上にロードする。1kB DNAラダーを基準として用いる。1×TAEバッファー中、100Vで20〜30分間、ゲル電気泳動を行う。
16. Invitrogen社のTOPO 2.1 Kitを用いて、1.5ml微量遠心チューブに以下のクローニング反応をセットアップする:
PCR反応物 4μl
塩溶液(Salt Solution) 1μl
TOPOベクター 1μl
総体積 6μl
17. 反応物を穏やかに混合し、室温で5分間インキュベートする。
18. 工程17のTOPOクローニング反応物を2μl取って、One Shot ケミカルコンピテントE.Coliに加え、穏やかに混合する。
19. 氷上で30分間インキュベートする。
20. この細胞に42℃で30秒間、熱ショックを与える。
21. このチューブを氷上に移し、2分間インキュベートする。
22. 室温のSOC培地を250μl加える。
23. このチューブを37℃、200rpmで1時間、水平方向に振盪する。
24. この形質転換体(transformation)10μlを、再度温めたLB−カルベニシリンのプレート上に播く。
25. プレートを37℃で一晩インキュベートする。
26. それぞれの形質転換体から、シーケンシング(sequencing)のために6クローンを採取する。
27. 重鎖及び軽鎖の可変領域配列を解析する。ELISA法を用いた2段階目のスクリーニングを開始する。
反応は選択する制限酵素に依存するものであり、製造者の使用説明書に従うものである。幾つかの例を本明細書に記載する。
氷上の微量遠心チューブに、以下の切断反応を準備する:

7. ボルテックスして混合し、簡単に遠心する。

9. 12000×gで1分間遠心し、コレクションチューブ(collection tube)から液体をデカント(decant)する。
10. 全てのサンプルが処理されるまで繰り返す。
11. 750μlのPEバッファー(エタノールを加える)で洗浄する。
9. 12000×gで1分間遠心し、コレクションチューブから液体をデカント(decant)する。
13. カラムをコレクションチューブに戻して、再度遠心する。
14. カラムを新しい遠心チューブに入れ、50μLのEBバッファーで溶出する。

制限酵素により切断された完全ヒト抗体クローン 100μl
10×サンプルローディングバッファー 3μl

2. 0.5g/mlのエチジウムブロマイド入り1%アガロースTAEゲル上にロードする。1kB DNAラダーを基準として用いる。1×TAEバッファー中、100Vで20〜30分間、ゲル電気泳動を行う。
3. 重鎖(HC:heavy chain)及び軽鎖(LC:light chain)の可変領域に対応するバンドを切り出し、QIAquick Gel Extraction Kitを用いて精製する。
4. ゲル1体積に対して3体積のバッファーQGを加える。
5. ゲル切片が完全に溶解するまで、50℃で10分間インキュベートする。ゲル1体積分のイソプロパノールをサンプルに加えて混合する。
6. 添付の2mlコレクションチューブに、QIAquickスピンカラムをセットする。
7. QIAquickカラムにサンプルをアプライ(apply)し、1分間遠心する。
8. 通過液を捨て、QIAquickカラムを同じコレクションチューブに戻す。
9. 0.75mlのバッファーPEをQIAquickカラムに加え、1分間遠心する。
10. 通過液を捨て、QIAquickカラムを17900×g(13000rpm)でさらに1分間遠心する。
11. QIAquickカラムを、滅菌済の1.5ml微量遠心チューブにセットする。
12. 52μlのバッファーEBを、QIAquick膜の中心に加え、このカラムを1分間遠心する。1分間カラムを立てたままにし、次いで1分間遠心する。
以下のライゲーション(ligation)反応を氷上の微量遠心チューブ中に用意した:
切断されたベクターDNA(100ng) xμl
ヒト重鎖及び軽鎖の可変領域 yμl
5×T4リガーゼバッファー 4μl
ヌクレアーゼフリーの水 19μlまでの必要量
T4リガーゼ(2000U/μl) 1μl
総反応体積 20μL
1. 微量遠心チューブ中で穏やかに混合し、少しの間(5秒間)遠心する。
2. 室温で2時間、又は16℃で一晩インキュベートする。
3. 各ライゲーション反応混合物を、スーパーコンピテントE.Coli細胞中に入れて形質転換する。
4. 14mlのBD社Falcon(商標)ポリプロピレン丸底チューブを幾つか、氷上で予冷しておく。SOC培地を42℃で用意する。
5. 前記スーパーコンピテント細胞を氷上で解凍する。解凍後穏やかに混合し、前記予冷しておいたチューブ中に、それぞれ100μlずつ等分して入れる。
6. 等分した細胞に、それぞれ1.7μlのβ−メルカプトエタノールを加える。この細胞を氷上で10分間、2分毎に穏やかに回し混ぜながらインキュベートする。
7. 2μlのライゲーション反応混合物を、等分した細胞の1つに加える。このチューブを穏やかにはじいて混ぜる。
8. このチューブを氷上で30分間インキュベートする。
9. このチューブに42℃の湯浴で45秒間、熱パルスを与える。
10. このチューブを氷上で2分間インキュベートする。
11. 予熱したSOC培地を900μl加え、このチューブを225〜250rpmで振盪しながら37℃で1時間インキュベートする。
12. 20μl及び200μlの形質転換体混合物を、カルベニシリン含有LB寒天プレート上に播く。
13. このプレートを37℃で一晩インキュベートする。
14. プレート上のコロニーを計数し、PCRスクリーニング及びシーケンシングのために6コロニーを採取する。
15.正しい配列を持つクローンを1つ選び、プラスミドDNAを調製し、293F細胞への形質移入(transfection)を次に行う。
1. 形質移入の1週間前に、293F細胞を血清添加ダルベッコ変法イーグル培地(D−MEM:Dulbecco's Modified Eagle Medium)中の単層培養に移す。
2. 形質移入の1日前に、96ウェルの形式で、形質移入サンプル毎に、100μlの血清添加D−MEM中に0.1×105の細胞を播く。
3. 各形質移入サンプルのために、DNA−リポフェクタミン複合体を調製する。
4. DNA0.2μgを、50μlのOpti−MEM血清使用量低減培地で希釈する。穏やかに混合する。
5. リポフェクタミン0.125μlを、50μlのOpti−MEM血清使用量低減培地で希釈する。穏やかに混合し、室温で5分間インキュベートする。
6. 前記希釈DNAと前記希釈リポフェクタミンとを合わせる。穏やかに混合し、室温で20分間インキュベートする。
7. 100μlの前記DNA−リポフェクタミン複合体を、細胞及び培地の入ったウェルにそれぞれ加える。このプレートを前後に揺すって穏やかに混合する。
8. この細胞を5%CO2のインキュベーター中37℃でインキュベートする。
9. 6時間後、各ウェルに100μlの血清添加D−MEMを加える。この細胞を5%CO2のインキュベーター中37℃で一晩インキュベートする。
10. 各ウェルの培地を吸引除去する。100μlの4mM L−グルタミン入り293 SFM IIで各ウェルを洗浄する。100μlの4mM L−グルタミン入り293 SFM IIを各ウェルに加える。
11. 形質移入の96時間後、ELISAのために上清を収集する。

付録1.2:完全ヒト抗体ライブラリの作製
全ての機能的なヒト生殖細胞系列重鎖(VH)及びκ軽鎖(Vk)のV領域は、V base(http://vbase.mrc−cpe.cam.ac.uk/)から得て、アライメントすることができる。このアライメントを、多様性に関して、特にフレームワーク3(framework three)領域において、次いで解析することができる。結果得られた配列クラスターから、所望の数のVH及びVkの遺伝子を、ライブラリ作成のために次いで選択することができる。
重鎖及び軽鎖V領域の遺伝子(フレームワーク1、2、及び3、並びにCDR1、CDR2、及びCDR3を含む)を、2つの遺伝子特異的なプライマーを用いて、ヒトゲノムDNAから増幅する。次いで、部分的なV領域の遺伝子を、オーバーラップPCRによって結合する。全長の軽鎖V領域に、哺乳類発現ベクター中にクローニングする前に、ネステッドPCRによってリンカーを追加する。クローニングされた軽鎖可変領域の配列を確認する(軽鎖Vクローンを産生)。重鎖可変領域をTOPOクローニングし、配列を確認する。(重鎖V TOPOクローン)。配列を確認した重鎖V領域を、対応するプラスミドから増幅し、リンカーを3'末端に加え、得られたPCR産物を、前記軽鎖可変領域クローンにクローニングし、Vk/VHの連結物を形成し、哺乳類ベクター中にクローニングする。
所望の哺乳類ベクターにおける全長κ軽鎖及びIgG1重鎖の発現は、例えばCMVプロモーターのような単一プロモーターで駆動させることができる。それぞれの鎖の前には、新生ポリペプチド鎖を小胞体(ER:endoplasmatic reticulum)に標的させる分泌シグナルを付ける。アンカーシグナルは、重鎖のC末端に結合させることができる。このシグナルは切断除去され、分泌後に前記全長IgGを細胞膜の外側に付着させるアンカーに置き換わる。
1. 形質移入の1週間前に、CHO−S細胞を血清添加ダルベッコ変法イーグル培地(D−MEM:Dulbecco's Modified Eagle Medium)中の単層培養に移す。
2. 形質移入の1日前に、10cmの組織培養プレート中に、形質移入サンプル毎に、15mlの血清添加D−MEM中に6×106の細胞を播く。10枚の10cmプレートを用意する。
3. それぞれの10cmプレートに、工程4〜7に従ってDNA−リポフェクタミン複合体を調製する。
4. マキシプレップ(maxi−prep)した完全ヒト抗体ライブラリのプラスミドDNA25μgを、1.5mlのOpti−MEM血清使用量低減培地で希釈する。穏やかに混合する。
5. 60μlのリポフェクタミンを、1.5mlのOpti−MEM血清使用量低減培地で希釈する。穏やかに混合し、室温で5分間インキュベートす
6. 前記希釈DNAと前記希釈リポフェクタミンとを合わせる。穏やかに混合し、室温で20分間インキュベートする。
7. 3mlの前記DNA−リポフェクタミン複合体を、細胞及び培地の入ったプレートにそれぞれ加える。このプレートを前後に揺すって穏やかに混合する。
8. この細胞を5%CO2のインキュベーター中37℃でインキュベートする。
9. 各プレートの培地を15mlの血清添加D−MEMに換える。この細胞を5%CO2のインキュベーター中37℃で48時間インキュベートする。
10. Detachin(商標)細胞剥離培地を用いて細胞を剥離し、血清添加D−MEM中に細胞を再懸濁する。
11. 1枚の10cm組織培養プレート中の、800μg/mlのG418入り血清添加D−MEM10mlに、0.4×106の細胞を播く。形質移入した全ての細胞を移動し、結果150枚の10cmプレートを得る。
12. 1枚の10cm組織培養プレート中の、800μg/mlのG418入り血清添加D−MEM10mlに、0.4×106の形質移入していないCHO−S細胞を播く。
13. 4日毎に、800μg/mlのG418入り血清添加D−MEMを細胞に与える。
14. 14日後、形質移入していないCHO−S細胞のプレートを観察する。プレートには生きた細胞が無いはずである。
15. 残った形質移入した細胞を、Detachin(商標)細胞剥離培地を用いて剥離し、冷凍用培地中に1×107cells/mlで細胞を冷凍する。
1. 形質移入の1日前に、10cmの組織培養プレート中に、形質移入サンプル毎に、15mlの血清添加D−MEM中に6×106のEcoPack2(商標)293細胞を播く。10枚の10cmプレートを用意する。
2. MBS添加培地を調製する。この作業は形質移入のすぐ前に行う。それぞれの10cm組織培養プレートについて、12mlのMBS入り培地を調製しなければならない。
3. 12mlのMBS添加培地をそれぞれの10cmプレートに加え、このプレートを37℃のインキュベーターに戻す。この作業は、DNA懸濁液を添加する20〜30分前に行わなければならない。
4. 10gのマキシプレップした完全ヒト抗体ライブラリのプラスミドDNAペレットを、450μlの滅菌済H2O中に懸濁し、このDNAを5mlのBD社Falcon(商標)ポリプロピレン丸底チューブに分注する。
5. このDNAに、Stratagene社Transfection MBS Mammalian Transfection Kitの溶液Iを50μl、溶液IIを500μl加える。
6. このDNA懸濁液中の沈殿物については、500μlにセットしたピペットを使って、沈殿物を吸い上げたり戻したりすることで穏やかに再懸濁する。
7. このDNA懸濁液を室温で10分間インキュベートする。
8. インキュベーターから10cmプレートを形質移入のために取り出し、前記DNA懸濁液をこのプレートに滴下する形で加え、細胞がプレートから飛び出さないように、またDNA懸濁液が均等に行き渡るように、穏やかに回し混ぜる。
9. 細胞培養プレートを、37℃のインキュベーターに戻す。
10. 3時間インキュベートした後、このプレートから培地を除去し、4mlの25μMクロロキン添加血清添加D−MEMに換える。このプレートを37℃のインキュベーターに戻す。
11. さらに6〜7時間インキュベートした後、25μMクロロキン入りの前記成長培地を除去し、4mlのクロロキン無添加血清添加D−MEMに換える。
12. このプレートを37℃のインキュベーターで一晩インキュベートする。
13. このプレートから培地を除去し、3.0mlの新しい血清添加D−MEMに換える。このプレートを37℃のインキュベーターに戻す。
14. このインキュベーターからこのウイルス産生パッケージング細胞を取り出す。
15. 最初のプレートからウイルスを含有する上清を収集し、0.45μmフィルターを通して滅菌済の50mlコニカルチューブに入れる。
16. このウイルス上清を1.5mlのクライオバイアルに分注し、ドライアイス/エタノール浴で瞬間冷凍する。このウイルス上清を−80℃で保管する。
17. 1枚の10cm組織培養プレート中の、D−MEM10mlに、0.5×106のNIH−3T3細胞を播く。
18. 前記上清を、37℃のH2O浴で素早く攪拌させながら急速解凍する。
19. このウイルスを、DEAE−デキストラン溶液入りウシ血清添加DMEMで、0.3×105ウイルス粒子/mlの力価になるよう希釈する。100mmプレート毎に、感染用に3mlの希釈ウイルスを用意する(細胞の20%が感染する)。DAEA−デキストラン入り成長培地である「偽混合物(mock cocktail)」を用意し、ネガティブコントロールとして用いる。
20. NIH−3T3細胞から培地を吸引除去し、各プレートに3.0mlの希釈ウイルスを細胞の上に均等に播く。プレートを37℃のインキュベーターに戻して3時間置く。
21. 3時間インキュベートした後、追加で7.0mlのウシ血清添加D−MEMを各プレートに加え、プレートを37℃のインキュベーターに戻す。プレートを48時間インキュベートする。
22. 10cm組織培養プレートのそれぞれの培地を、10mlの800μg/mlのG418入り血清添加D−MEMに換える。
23. 4日毎に、800μg/mlのG418入り血清添加D−MEMを細胞に与える。
24. 14日後、偽感染のNIH−3T3細胞のプレートを観察する。プレートには生きた細胞が無いはずである。
25. 残った形質移入した細胞を、Detachin(商標)細胞剥離培地を用いて剥離し、冷凍用培地中に1×107cells/mlで細胞を冷凍する。
変異導入反応
1組のプライマー(プライマー混合物1及びプライマー混合物2)を、挿入するそれぞれのコドンについて設計する。設計は遺伝子配列に依存し、Sequencher(商標)(Gene Codes Corporation社)やVectorNTI(登録商標)(Life Technologies社)などの配列解析データベースを、前記プライマーの設計に使用することができる。CPE(商標)においては、一組のプライマーを、変異させるそれぞれのコドンについて設計する。 目標の縮重コドン(NNK又はNNN)は中間に配置し、その両側に20塩基を配置する(プライマー全長:43塩基、固有の変異をシーケンシングして同定するために9fクローン)。テンプレートDNAは目的の遺伝子を持つベクターDNAである。

1. 1つの96ウェルプレート毎に、1つのネガティブコントロール反応(プライマーをTEバッファーに置換)を用意する。
2. 穏やかに混合し、卓上遠心分離機中で少しの間(5秒間)遠心する。
3. 下に概要を記したサイクリングパラメーターを用いて、サイクル反応を行う。
1. 増幅反応物の品質確認をするために、96ウェル薄壁PCRプレート又は0.2ml薄壁PCRチューブに、以下の反応をセットアップする:
変異誘導反応物 5μl
水 4μl
サンプルローディングバッファー 1μl
体積 10μl
2. 0.5g/mlのエチジウムブロマイド入り1%アガロースTAEゲル上にロードする。1kB DNAラダーを基準として用いる。1×TAEバッファー中、100Vで20〜30分間、ゲル電気泳動を行う。
1. 0.5μlの制限酵素DpnI(10U/μl)を直接、各反応物に加える。
2. 穏やかに混合し、卓上遠心分離機中で少しの間(5秒間)遠心する。
3. PCR機器の中で、37℃で2時間インキュベートする。
4. 各96ウェルプレートから6つの反応混合物を、XLI Blueスーパーコンピテント細胞に形質転換する。反応物の残りは−20℃で保管する。
5. 0.2mlPCRチューブを幾つか、氷上で予冷しておく。SOC培地を42℃まで温める。
6. 前記XLI Blueスーパーコンピテント細胞を氷上で解凍する。解凍後に穏やかに混合し、前記予冷しておいたチューブ中に、それぞれ50μlずつ等分して入れる。
7. 等分した細胞に、それぞれ0.8μlのβ−メルカプトエタノールを加える。この細胞を氷上で10分間、2分毎に穏やかに回し混ぜながらインキュベートする。
8. 2μlの反応混合物を、等分した細胞の1つに加える。このチューブを穏やかにはじいて混ぜる。
9. このチューブをコールドブロックの上で30分間インキュベートする。
10. このチューブに42℃の湯浴で45秒間、熱パルスを与える。
11. このチューブを氷上で2分間インキュベートする。
12. 予熱したSOC培地を100μl加え、このチューブを225〜250rpmで振盪しながら37℃で1時間インキュベートする。
13. 全ての形質転換体混合物を、カルベニシリン含有LB寒天プレート上に播く。
14. このプレートを37℃で一晩インキュベートする。
15. プレート上のコロニーを計数し、ミニプレップ及びシーケンシングのために12コロニーを採取する。
1. XLI Blueスーパーコンピテント細胞を氷上で解凍する。96の反応のために20のコンピテント細胞のチューブを解凍する。解凍後、4μlのβ−メルカプトエタノールを、250μlのコンピテント細胞の各チューブに加える。この細胞を氷上で10分間、2分毎に穏やかに回し混ぜながらインキュベートする。
2. 0.2mlPCRチューブを幾つか、氷上で予冷しておく。SOC培地を42℃まで温める。
3. 前記予冷しておいたチューブ中に、それぞれ50μlずつ等分して入れる。
4. 2μlの反応混合物を、等分した細胞の1つに加える。このチューブを穏やかにはじいて混ぜる。
5. このチューブをコールドブロックの上で30分間インキュベートする。
6. このチューブに42℃の湯浴で45秒間、熱パルスを与える。
7. このチューブを氷上で2分間インキュベートする。
8. 予熱したSOC培地を100μl加え、このチューブを225〜250rpmで振盪しながら37℃で1時間インキュベートする。
9. 全ての形質転換体混合物を、カルベニシリン含有LB寒天プレート上に播く。
10. このプレートを37℃で一晩インキュベートする。
11. ミニプレップのために、96ウェルブロック中で細胞を培養する。
12. QIAVac96 kitを用いて、製造者の使用説明書に従って、ミニプレップDNAを調製する。
形質移入
形質移入の1週間前に、293F細胞を血清添加ダルベッコ変法イーグル培地(D−MEM:Dulbecco's Modified Eagle Medium)中の単層培養に移す。形質移入の1日前に、96ウェルの形式で、形質移入サンプル毎に、100μlの血清添加D−MEM中に0.2×105及び0.4×105の細胞を播く。
1. 各形質移入サンプルのために、DNA−リポフェクタミン複合体を調製する。
2. DNA0.2μgを、50μlのOpti−MEM血清使用量低減培地で希釈する。穏やかに混合する。
3. リポフェクタミン0.125μlを、50μlのOpti−MEM血清使用量低減培地で希釈する。穏やかに混合し、室温で5分間インキュベートする。
4. 前記希釈DNAと前記希釈リポフェクタミンとを合わせる。穏やかに混合し、室温で20分間インキュベートする。
5. 100μlの前記DNA−リポフェクタミン複合体を、細胞及び培地の入ったウェルにそれぞれ加える。このプレートを前後に揺すって穏やかに混合する。
6. この細胞を5%CO2のインキュベーター中37℃でインキュベートする。
7. 6時間後、各ウェルに100μlの血清添加D−MEMを加える。この細胞を5%CO2のインキュベーター中37℃で一晩インキュベートする。
8. 各ウェルの培地を吸引除去する。100μlの4mM L−グルタミン入り293 SFM IIで各ウェルを洗浄する。100μlの4mM L−グルタミン入り293 SFM IIを各ウェルに加える。
9. 形質移入の96時間後、ELISAのために上清を収集する。
1. Nunc−Immuno Maxisorp96ウェルプレートを、抗原2μg/mlが入ったコーティング溶液100μlでコートする。
2. プレートをシーラーで覆い、4℃で一晩インキュベートする。
3. プレートをデカントし、残った液体を軽く叩いて出す。
4. 200μlの洗浄溶液を加える。室温で5分間、200rpmで振盪する。
5. プレートをデカントし、残った液体を軽く叩いて出す。
6. 200μlのブロッキング溶液を加える。室温で1時間、200rpmで振盪する。
7. プレートをデカントし、残った液体を軽く叩いて出す。
8. コントロール抗体(2μg/ml)入りのブロッキング溶液を100μl/ウェルで二つ組、プレートに加える。
9. 形質移入体(SOP 5A)の上清100μlを二つ組、プレートに加える。
10. 室温で1時間、200rpmで振盪する。
11. プレートをデカントし、残った液体を軽く叩いて出す。
12. 200μlの洗浄溶液を加える。室温で5分間、200rpmで振盪する。
13. 工程11〜12を3回繰り返す。
14. アフィニティー精製ヤギ抗ヒト抗体とHRPのコンジュゲートを、ブロッキング溶液中に1:5000で希釈した溶液100μlを、各ウェルに加える。
15. 室温で1時間、200rpmで振盪する。
16. プレートをデカントし、残った液体を軽く叩いて出す。
17. 200μlの洗浄溶液を加える。室温で5分間、200rpmで振盪する。
18. 工程17〜18を3回繰り返す。
19. Sigma社TMB基質100μlを各ウェルに加える。室温でインキュベートし、2〜5分毎にチェックする。
20. 反応を停止させるために100μlの1N HClを加える。
21. 450nmで読み取る。

本実施例は、Fcコドン変異体ライブラリを作成する方法と、Fcポリペプチドの親型に比べて、生産宿主細胞における発現量向上について最適化されたFc変異体を得るためのスクリーニング方法を提供するものである。
標的領域(この場合においてはヒトIgG1分子のFc部位)の各コドンについて、標的コドン及びその両側に20塩基を含むように、1組の縮重プライマー(順行性と逆行性)を設計する。標的コドンの3つ目の位置(ゆらぎの位置)は混合の塩基(表3)を含むので、同じコドンを用いて標的位置において全てのサイレント変異を作成できる(例A)。対応するアミノ酸が別のコドンによってエンコードされうる場合は、同じコドン位置について2つ目の縮重プライマーのセットを設計する(例B)。
対応する順行性及び逆行性の縮重プライマーは1:1の割合で混合され、テンプレートとアニールし、熱安定性DNAポリメラーゼを用いた鎖置換によって伸長して全長の生産物となる。テンプレートをDpnIによって切断し、全長の伸張産物をE.coli中に形質転換する。変異誘導反応毎に12コロニーまでを、シーケンシングする。配列が確認された変異体は96ウェルプレート中に配列し、グリセロールストックにする。このグリセロールストックは、プラスミドDNAをミニプレップし、哺乳類細胞へ形質移入し、スクリーニングするために使われる。
Fcコドン変異体ライブラリからのクローンを、哺乳類細胞株中へ形質移入した。
全長のIgGが生産され、培地の中に分泌された。発現したFcコドン変異体の上清について、ELISAアッセイを用いて、IgGの発現レベルが親クローンよりも高いものをスクリーニングした。このELISAデータを、形質移入効率を測定するβ−ガラクトシダーゼアッセイを用いて標準化した。最初のスクリーニングにおいて同定した高順位のものについて、3回繰り返し形質移入及びスクリーニングを行い、発現レベルの高さを確認した。

- 真核細胞産生宿主における抗体或いは抗体断片の選択、進化及び発現の方法であって、前記方法は、
a.抗体細胞表層ディスプレイを用いて真核細胞産生宿主において抗−抗原抗体ライブラリを作成する工程と、
b.少なくとも1つの既定の性質、特徴或いは活性に対して前記ライブラリをスクリーニングする工程と、
c.前記ライブラリから鋳型抗体を選択する工程と、
d.抗体細胞表層ディスプレイを用いて前記真核細胞産生宿主において1セットの変異抗体を生成するために前記鋳型抗体を進化させる工程と、
e.前記少なくとも1つの既定の性質、特徴或いは活性に対して前記変異抗体をスクリーニングする工程と、
f.前記鋳型抗体と比較した場合、前記少なくとも1つの既定の性質、特徴或いは活性の最適化に基づいて前記1セットの変異抗体からアップ突然変異抗体を選択する工程と、

a.m×nの別々のセットの抗体を産生する工程であって、各セットはCDRの単一の既定の位置にX個の異なる既定のアミノ酸残基を持つメンバー抗体を有するものであり、ここにおいて各セットの抗体は、単一の既定の位置が異なり;産生された異なるメンバー抗体の数はm×n×Xに等しいものである、前記抗体を産生する工程を有するものである、方法。 - 請求項5記載の方法において、mは6である、方法。

前記スクリーニングする工程(e)は、
i.少なくとも1つの既定の性質、特徴或いは活性に対して各メンバーポリペプチドをアッセイする工程と、
j.前記鋳型ポリペプチドと比較して前記メンバーポリペプチドの性質、特徴或いは活性におけるあらゆる変化を同定する工程と、
k.機能マップを作成する工程であって、ここにおいて前記機能マップは、前記鋳型ポリペプチドと比較して、アップ突然変異及び/若しくはサイレント変異を生じる、前記変異ポリペプチドにおける位置及び変異を同定するために使用されるものである、前記機能マップを作る工程とを有する、前記スクリーニングする工程を有するものである、方法。 - 請求項8記載の方法において、前記Xは19の天然由来のアミノ酸残基を示すものであり、前記鋳型ポリペプチドの所定の位置を表したものではない、方法。
- 請求項9記載の方法において、前記産生する工程は、
i.変異誘発された各コドンに対する64重縮重オリゴヌクレオチドを用いて、ポリメラーゼ−ベース複製へ、前記鋳型ポリペプチドをコード化しているコドン−含有ポリヌクレオチドを暴露する工程であって、ここにおいて、1セットの子孫ポリヌクレオチドを産生するために、前記各64重縮重オリゴヌクレオチドは、第一の相同配列及び変性N,N,Nトリプレット配列を有するものである、前記暴露する工程と、
ii.前記子孫ポリヌクレオチドによってコード化されたポリペプチドを発現させるように、クローン複製へ前記1セットの子孫ポリヌクレオチドを暴露する工程とを有するものである、方法。 - 請求項8記載の方法において、前記機能マップは、(a)前記鋳型ポリペプチドと比較して、前記変異ポリペプチドの活性に影響を与えない位置及び変異;(b)前記鋳型ポリペプチドと比較して、完全に変異可能な部位;及び(c)前記鋳型ポリペプチドと比較して、アップ突然変異を生じる位置及び変異、から成る群の1若しくはそれ以上を同定するために使用されるものである、方法。
- 請求項1記載の方法において、前記既定の性質、特徴或いは活性は、タンパク質−タンパク質凝集の軽減、タンパク質安定性の増強、タンパク質可溶性の増加、グリコシル化部位の導入、共役部位の導入、免疫原性の軽減、タンパク質発現の増強、抗原親和性における増加、抗原親和性の減少、結合親和性における変化、免疫原性における変化、或いは特異性の増強から選択されるものである、方法。
- 哺乳類細胞産生宿主における抗体を進化及び発現させる方法であって、
前記方法は、
a.鋳型抗体を選択する工程と、
b.抗体細胞表層ディスプレイを用いて、哺乳類細胞産生宿主において1セットの変異抗体を生成するために前記鋳型抗体を進化させる工程と、
c.少なくとも1つの既定の性質、特徴或いは活性に対して前記変異抗体をスクリーニングする工程と、
d.前記鋳型抗体と比較して、前記少なくとも1つの既定の性質、特徴或いは活性の最適化に基づいて前記1セットの変異抗体からアップ突然変異抗体を選択する工程と、

e.あらゆる工業規模に対しても、工程bと同じ真核細胞産生宿主において前記アップ突然変異抗体を発現させる工程とを有するものである、方法。 - 請求項1記載の方法において、前記哺乳類細胞産生宿主は、CHOK1SV或いはNSO宿主細胞株から選択されるものである、方法。
- 請求項13記載の方法において、前記スクリーニングする工程は、機能マップを作成する工程であって、ここにおいて前記機能マップは、前記鋳型ポリペプチドと比較して、アップ突然変異及び/若しくはサイレント変異を生じる、前記変異ポリペプチドにおける位置及び変異を同定するために使用されるものである、前記機能マップを作成する工程を有するものである、方法。
- 細胞産生宿主においてヒトタンパク質を進化及び製造させる方法であって、
前記方法は、
a.細菌或いは真核生物産生宿主から成る群の1つから選択された細胞産生宿主において、ヒトタンパク質ライブラリを作成する工程と、
b.少なくとも1つの既定の性質、特徴或いは活性に対して前記ライブラリをスクリーニングする工程と、
c.前記少なくとも1つの既定の性質、特徴或いは活性に基づいて前記ライブラリから鋳型ヒトタンパク質を選択する工程と、
d.前記細胞産生宿主において1セットの変異ヒトタンパク質を産生するために前記鋳型ヒトタンパク質を進化させる工程と、
e.前記少なくとも1つの既定の性質、特徴或いは活性に対して前記1セットの変異ヒトタンパク質をスクリーニングする工程、及び修飾された発現に対してスクリーニングする工程と、
f.(1)前記少なくとも1つの既定の性質、特徴或いは活性の最適化、及び(2)前記鋳型ヒトタンパク質と比較した場合、修飾された発現に基づいて、前記1セットの変異ヒトタンパク質からアップ突然変異ヒトタンパク質を選択する工程と、
g.前記作成する工程(a)と同じ産生宿主において前記アップ突然変異ヒトタンパク質を発現させる工程を有するヒトタンパク質を製造する工程とを有するものである、方法。 - 請求項22記載の方法において、前記修飾された発現は、改善された発現である、方法。
- 請求項23記載の方法において、前記進化させる工程は、包括的位置進化(CPE);包括的位置挿入進化(CPI);包括的位置欠失進化(CPD);コンビナトリアルタンパク質合成(CPS)に続く包括的位置進化(CPE);或いはコンビナトリアルタンパク質合成(CPS)に続く包括的位置欠失進化(CPD)の1つを有するものである、方法。
- 請求項23記載の方法において、前記ヒトタンパク質は、抗体である、方法。
- 請求項25記載の方法において、前記抗体は、完全長抗体である、方法。
- 請求項26記載の方法において、前記スクリーニングする工程(e)における前記少なくとも1つの既定の性質、特徴或いは活性は、前記鋳型抗体と比較して、1若しくはそれ以上の(1)サイレント変異に対してスクリーニングする工程、及び(2)ミスセンス変異に対してスクリーニングする工程を有するものである、方法。
- 請求項26記載の方法において、Fc及びFvから選択される1若しくはそれ以上の群;及び1若しくはそれ以上のCDRsは、前記鋳型ヒト抗体と比較して、前記アップ突然変異ヒト抗体において修飾されているものである、方法。
- 請求項22記載の方法において、前記スクリーニングする工程(e)は、前記少なくとも1つの既定の性質、特徴或いは活性に対して前記1セットの変異ヒトタンパク質をスクリーニングする工程と、修飾された発現に対してスクリーニングする工程とを同時に有するものである、方法。
- 請求項22記載の方法において、前記ヒトタンパク質は、酵素サイトカイン、受容体、DNA結合タンパク質、キレート剤或いはホルモンから選択されるものである、方法。
- 請求項22記載の方法において、前記細胞産生宿主は、真核生物産生宿主であり、前記進化させる工程は、細胞表層ディスプレイを用いて前記真核細胞産生宿主において1セットの変異ヒトタンパク質を生成するために前記鋳型ヒトタンパク質を進化させる工程を有するものである、方法。
- 請求項26記載の方法において、前記細胞産生宿主は、真核生物産生宿主であり、前記進化させる工程は、細胞表層ディスプレイを用いて前記真核細胞産生宿主において1セットの変異ヒトタンパク質を生成するために前記鋳型ヒトタンパク質を進化させる工程を有するものである、方法。
- 真核細胞産生宿主においてヒトタンパク質の増強した発現及び製造のために進化させる方法であって、
前記方法は、
a.進化のために鋳型ヒトタンパク質を選択する工程と、
b.前記産生宿主において1セットの変異ヒトタンパク質を産生するために、前記鋳型ヒトタンパク質をコード化する変異コドンの産生を有する前記鋳型ヒトタンパク質を進化させる工程と、
c.少なくとも1つの既定の性質、特徴或いは活性に対して前記1セットの変異ヒトタンパク質をスクリーニングし、前記鋳型ヒトタンパク質と比較した場合、増強した発現に対してスクリーニングする工程と、
d.(1)前記少なくとも1つの既定の性質、特徴或いは活性の保持或いは最適化、及び(2)前記鋳型ヒトタンパク質と比較した場合、増強した発現に基づいて、前記1セットの変異ヒトタンパク質からアップ突然変異ヒトタンパク質を選択する工程と、
e.前記進化させる工程と同じ産生宿主において前記アップ突然変異ヒトタンパク質を発現させる工程を有する、前記アップ突然変異ヒトタンパク質を製造する工程とを有する、方法。 - 請求項33記載の方法において、前記アップ突然変異ヒトタンパク質の変異コドンは、少なくとも1つのサイレント変異及び/若しくはミスセンス変異を生じるものである、方法。
- 請求項34記載の方法において、前記アップ突然変異ヒトタンパク質の変異コドンは、少なくとも1つのサイレント変異及び/若しくはミスセンス変異を生じるものである、方法。
- 請求項35記載の方法において、前記鋳型ヒトタンパク質は、倫理的に承認されたタンパク質治療薬であり、前記アップ突然変異ヒトタンパク質は、バイオ後続品である、方法。
- 請求項33記載の方法において、前記選択する工程(d)は、(1)前記少なくとも1つの既定の性質、特徴或いは活性の最適化、及び(2)前記鋳型ヒトタンパク質と比較した場合、増強した発現に基づいて、前記1セットの変異ヒトタンパク質からアップ突然変異ヒトタンパク質を選択する工程を有するものである、方法。
- 標的ヒトタンパク質を同定し生成する方法であって、前記方法は、
a.タンパク質細胞表層ディスプレイを用いて真核細胞産生宿主においてヒトタンパク質ライブラリを作成する工程と、
b.少なくとも1つの既定の性質、特徴或いは活性に対して前記ライブラリをスクリーニングする工程と、
c.前記少なくとも1つの既定の性質、特徴或いは活性に基づいて前記ライブラリから標的ヒトタンパク質を同定する工程と、
d.標的ヒトタンパク質を生成するために、前記作成する工程と同じ真核細胞産生宿主において前記標的ヒトタンパク質を発現させる工程とを有するものである、方法。 - 請求項38記載の方法において、前記標的ヒトタンパク質は、抗体である、方法。
- 請求項39記載の方法において、前記抗体は、完全長抗体である、方法。
- 製造宿主におけるヒトタンパク質を進化させる方法であって、前記方法は、
a.製造宿主において1セットの変異ヒトタンパク質を製造するために鋳型ヒトタンパク質を変異させる工程と、
b.少なくとも1つの既定の性質、特徴或いは活性に対して1セットの変異子孫タンパク質をスクリーニングする工程とを有するものである、方法。 - 請求項41記載の方法であってさらに、
c.前記少なくとも1つの既定の性質、特徴或いは活性に基づいて、前記1セットの変異ヒトタンパク質からアップ突然変異ヒトタンパク質を選択する工程とを有するものである、方法。 - 請求項42記載の方法であって、前記方法はさらに、
d.前記変異させる工程(a)と同じ産生宿主において前記アップ突然変異ヒトタンパク質を発現させる工程を有する、前記アップ突然変異ヒトタンパク質を製造する工程を有するものである、方法。 - 請求項42記載の方法において、前記選択する工程(c)はさらに、
(1)前記鋳型ヒトタンパク質と比較した場合、前記少なくとも1つの既定の性質、特徴或いは活性の最適化、及び(2)前記鋳型ヒトタンパク質と比較した場合、修飾された発現に基づいて、前記1セットの変異ヒトタンパク質からアップ突然変異ヒトタンパク質を選択する工程を有するものである、方法。 - 請求項44記載の方法において、前記修飾された発現は、増強された発現である、方法。
- 細胞製造宿主におけるヒトタンパク質を進化及び製造する方法であって、前記方法は、
a.製造宿主において1セットの変異ヒトタンパク質を生成するために鋳型ヒトタンパク質を変異させる工程と、
b.少なくとも1つの既定の性質、特徴或いは活性に対して前記1セットの変異ヒトタンパク質をスクリーニングする工程、及び修飾された発現に対してスクリーニングする工程と、
c.(1)前記少なくとも1つの既定の性質、特徴或いは活性の最適化、及び(2)前記鋳型ヒトタンパク質と比較した場合、修飾された発現に基づいて、前記1セットの変異ヒトタンパク質からアップ突然変異ヒトタンパク質を選択する工程と、
d.工程a.と同じ製造宿主において前記アップ突然変異ヒトタンパク質を発現させる工程を有する、前記ヒトタンパク質を製造する工程とを有するものである、方法。 - 真核細胞産生宿主においてヒトタンパク質の増強された発現及び製造のために進化させる方法であって、前記方法は、
a.製造宿主において1セットの変異ヒトタンパク質を生成するために、鋳型ヒトタンパク質をコード化している変異コドンの産生を有する、前記鋳型ヒトタンパク質を変異させる工程と、
b.少なくとも1つの既定の性質、特徴或いは活性に対して前記1セットの変異ヒトタンパク質をスクリーニングし、前記鋳型ヒトタンパク質と比較した場合、増強された発現に対してスクリーニングする工程と、
c.(1)前記少なくとも1つの既定の性質、特徴或いは活性の保持或いは最適化、及び(2)前記鋳型ヒトタンパク質と比較した場合、増強された発現に基づいて、前記1セットの変異ヒトタンパク質からアップ突然変異ヒトタンパク質を選択する工程と、
d.前記工程(a)と同じ製造宿主において前記アップ突然変異ヒトタンパク質を製造する工程とを有するものである、方法。 - 請求項47記載の方法において、前記スクリーニングする工程(b)は、前記少なくとも1つの既定の性質、特徴或いは活性に対して前記1セットの変異ヒトタンパク質をスクリーニングする工程と、増強された発現に対してスクリーニングする工程とを同時に有するものである、方法。
