トール様受容体(TLR)は、先天性免疫系の制御において重要な役割を果たすセンチネル細胞において発現されるタンパク質のクラスである。哺乳動物の免疫系は、2つの一般的なストラテジーを使用して感染性の疾患と戦う。病原体曝露は、免疫賦活性サイトカイン、ケモカイン及び多反応性IgM抗体の産生によって特徴づけられる先天性免疫応答を迅速にトリガーする。先天性免疫系は、感染性微生物の多様な群によって発現される病原関連分子パターン(PAMP)に対する曝露によって活性化される。PAMPの認識は、受容体のトール様ファミリーのメンバーによって媒介される。特異的オリゴヌクレオチドに対して応答するTLR 4、TLR 8及びTLR 9などのTLR受容体は、特別な細胞内コンパートメントに位置して、エンドソームを呼ばれる。TLR 4、TLR 8及びTLR9受容体の調整の機構は、DNA−タンパク質相互作用に基づいている。

全ての試薬は、最高の純度で供給元から得て、及び任意のさらなる精製を伴わずに使用した。HPLCは、Varian Prostarシステムで行った。UV/Viは、Varian Cary 300分光光度計で記録した。蛍光スペクトルは、SPEX FluoroLog蛍光光度計で得た。
オリゴヌクレオチドは、自動DNAシンセサイザー(ABI 3400、Applied Biosystems、Inc)で1.0マイクロモル濃度のスケールで合成した。水酸化アンモニウム水溶液(55℃、14時間)での切断及び脱保護の後、DNAを逆相HPLCによって精製して、UV分光計によって定量化した。
脂質単量体(クロロホルムに溶解した40μmolの1,2−ジオレオイル−sn−グリセロ−3−ホスホコリン(DOPC))を20mLバイアルに添加して、次いで一晩凍結乾燥前に蒸発させて、溶媒を除去して薄い脂質フィルムを生じる。次いで、フィルムをHBS緩衝液(50mL、20mM Hepes緩衝液、pH 7.4にて150mM NaCl)で再水和して、続いて激しく混合してリポソーム懸濁液を形成し、次いでパルス化することなく30分間氷浴においてプローブ音波破砕した。次いで、生じる懸濁液を104,986g及び90分間4℃にて超遠心分離機にした。リン脂質濃度は、元素分析を使用して算出した。
LSNAの細胞取り込みを視覚化するために、HeLa細胞を一晩Lab−Tek(登録商標);II Chamber #1.5 German Coverglass System(Nalge Nunc International)で培養し、及びCy5−標識したLSNA(DNA濃度の0.1μM)と共にインキュベートした。16時間のインキュベーションの後、培地を新鮮な培地で置き換えて、生細胞を、製造業者の説明書に従ってヘキスト33342(Invitrogen)で染色した。全ての画像は、Mai Tai 3308レーザー(Spectra−Physics)を使用して40x倍率にてZeiss 510 LSMで得た。蛍光発光は、390−465nm及び650−710nmにて、それぞれ729及び633nmでの励起(図5)で収集した。図5の左パネルは、HeLa細胞へのリポソームフルオレセインの侵入を示し、その一方で、図5の右パネルは、フルオレセインの共局在性を示し、及びCy5は、細胞内への全てのリポソームの送達を示唆する。
リポソーム粒子の細胞毒性は、Alamar Blue(登録商標)Assay(Invitrogen)で評価した。簡潔には、HeLa細胞を200μLの培地中の96ウェルプレートに播種して、24時間インキュベートした。次いで、細胞をFITCカプセル化されたそのままのSUV及びLSNA機能化DNAで種々の濃度のリン脂質濃度(0、32.5、65、162.5μM)にて処理した。16時間後、培地を除去し、細胞を3回PBSで洗浄して、次いで90μLの新鮮な培養培地を10μLのアラマーブルー試薬を加えて4時間インキュベートした。次いで、560nmにて励起及び590nmにて発光を確認することによってこれらを解析した。
材料
1,2−ジオレオイル−sn−グリセロ−3−ホスホコリン脂質単量体(DOPC)は、乾燥粉末形態で、またはクロロホルム溶液中のいずれかでAvanti Polar Lipids, Inc.から購入し、さらに精製することなく使用した。ホスホラミダイト及びその他のDNA合成試薬は、Glen Research, Incから最高の純粋にて購入し、及び製造業者から受けとったときに使用した。
凍結乾燥は、Freezone Lyohilizer(Labconco、Kansas City、MO)を使用して実施した。音波処理は、パルスにすることなく20kHzの40%強度にてチタン−合金固体プローブ超音波処理器(500ワットのVibra−Cell(商標)VC 505、Sonics &Materials,Inc.、Newtown、CT)セットを使用して行った。超遠心分離は、Beckman−Coulter Avanti J−30I(Beckmann−Coulter,Inc.、Indianapolis、IN)を使用して実施した。透過型電子顕微鏡法(TEM)は、日立−2300 STEM電子顕微鏡を使用して行った。動的光散乱法(DLS)は、Malvern Zetasizer Nano−ZS(Malvern Instruments、UK)を使用して収集した。MALDI−ToF解析は、Bruker Autoflex III SmartBean質量分析計(Bruker Daltonics Inc、MA、USA)を使用して行った。蛍光測定は、Fluorlog−3システム(HORIBA Jobin Yvon Inc、NJ、USA)で実施した。UV−Vi分光法は、Cary 5000 UV−Vi分光光度計を使用して収集した。(Varian Inc., CA, USA)。
オリゴヌクレオチドは、自動化固体支持体ホスホラミダイト合成を使用してExpedite 8909 Nucleotide Synthesis System(MM48 Synthesizer、Bioautomation)で活性化因子としてDCIを使用して合成した。トコフェロールホスホラミダイトは、延長した15分の結合時間を使用して自動化されたプロトコルによって結合した。固相合成の完了後、オリゴヌクレオチド鎖を水性水酸化アンモニウム(28−30%の水溶液、Aldrich)での一晩の処理を使用して固体支持体から切断し、その後に窒素ガス(貯蔵窒素を使用した)の穏やかな流れを使用して、過剰アンモニアを除去した。オリゴヌクレオチドを、TEAA(トリエチルアンモニウムアセテート)緩衝液及びアセトニトリルの勾配(勾配:30分にわたって10%v/vから100%v/vアセトニトリル)を使用する逆相高圧液体クロマトグラフィー(HPLC、Varian)でMicrosorb C18カラムを使用して精製した。生成物を含む収集した画分を凍結乾燥器で濃縮した。得られたオリゴヌクレオチドをナノピュア水に再懸濁して、純度を、MALDI−TOF及び変性アクリルアミドゲル電気泳動技術を使用して解析した。
脂質単量体保存液の量(25−50mg)を20mLバイアルに添加して、25mLガラスバイアルに置き、及び溶媒を、窒素気流を使用して慎重に蒸発した。得られた脂質単量体は、残留するクロロホルムを除去するために真空下でさらに一晩乾燥させた。次いで、生じる脂質フィルムを20mM HBS(5.0mL)で水化し、続いてバイアルをボルテックスしてリポソーム懸濁液を形成した。この懸濁液を10℃より下(氷水浴で冷却)に混合脂質の温度を保持して、さらに30分間プローブ音波破砕した。音波処理の後、懸濁液を12℃にて90分間100,000xgにて超遠心分離に供した。遠心分離の後、所望の小単層小胞(SUV)を含む透明な上清を収集して、ペレットを捨てた(図1)。より狭いサイズ分布で粒子を得るために、得られたSUV粒子をポリカーボネート膜(30nm孔径)を通してさらに押し出した。
リポソームSNAを調製するために、15μMの所望の3’−トコフェロール修飾オリゴヌクレオチドをSUV溶液に添加し(1.3mMの[リン脂質])、及び一晩振盪させた。次いで、生じる溶液を架橋されたセファロースカラム(Separose CL 4B、Aldrich)でのゲル濾過クロマトグラフィーを介して精製した。粒径分布は、DLSを使用して解析した。TEMを使用してリポソームSNAを観察するために、試料をプラズマ洗浄炭素TEMグリッド上に置き、及び酢酸ウラニル(2%w/v)の溶液でさらに染色した(2分間染色し、次いで水で洗浄し、及び乾燥)。次いで、乾燥したグリッドを日立−2300 STEM電子顕微鏡下で撮像した。
全てのゲル電気泳動実験は、1×TBE(トリスボラート、EDTA)緩衝液中で1%のアガロースゲルにおいて行った。試料は、充填薬剤としてグリセロール(30%v/v、5μL)を用いて、ウェルに充填した。ゲルチャンバーを1×TBEで満たして、氷で予冷した。ゲルを10℃にて1時間70Vにて泳動して、ゲルの画像をCy5フィルタでFluorchem Qで記録した。
リポソームの表面上のDNAの充填を決定するために、Cy5標識した3’トコフェロール修飾DNAの濃度を増大して固定された濃度のSUVと共に一晩インキュベートした(1.3mM[P])。次いで、リポソームSNAをゲル電気泳動を使用して解析した。SUVで機能化したDNA密度を定量化するために、構築物を1%のSDS溶液に溶解して、吸光度を260nmにて収集して、それぞれのDNA鎖の吸光係数を使用して算出した。対応する溶液におけるリポソームの数は、リポソームのリン脂質濃度が機能付与の後一定のままであるという仮定で理論方程式を使用して算出した。
2ナノ粒子成分系は、表1に記載されているようにリンカー鎖に相補的な鎖で機能化したリポソームSNAを使用して形成した。凝集体は、2つのDNA機能性リポソームSNAの添加及び1:1の比(総DNA濃度1.5μM、容積1mL)でリンカー鎖にこれらをハイブリダイズすることによって形成した。リンカーをもつリポソームSNAについての吸光度スペクトルをCary 5000 UV−Vi分光計を使用して収集して、リンカーのないリポソームSNAの吸光度スペクトルと比較した。次いで、凝集体を0.25℃/分の割合にて20から65℃までの温度の緩やかな増大に供して、吸光度を凝集体について260nmにてモニターした。
乾燥DOPC単量体(25mg)をHBS(5mL)中の20mMスルホローダミンB溶液に再懸濁した。生じる懸濁液を100nm、80nm、50nm、30nmサイズの一連のポリカーボネート膜を通して徐々に押し出した。ローダミン含有リポソームを架橋されたセファロース(セファロースCL−4B、Aldrich)でのゲル濾過クロマトグラフィーを介して遊離ローダミンから分離した。得られた粒子を、上で記述した手順を使用してDNA−トコフェロール抱合体で機能化した。構築物の血清安定度を解析するために、ローダミン含有リポソーム及びリポソーム−SNAを、HBS中の10%のウシ胎児血清溶液に懸濁して、420nmにて試料を励起し、及び480nmにて強度を測定して、色素の放出をFluorlog−3システムでモニターした。
SKOV−3細胞は、アメリカン・タイプ・カルチャー・コレクション(ATCC)から購入して、10%の熱不活化ウシ胎児血清、100Uペニシリン及び50μgストレプトマイシンを伴うMcCoy’s 5A培地中で培養し、及びATCC説明書に従って5%のCO2で37℃にて維持した。細胞研究のためには、細胞を50%の集密度にて処理の24時間前にプレートにまいた。
リポソームSNAの細胞内部移行の視覚化のためには、SKOV3細胞を50%の集密度にて35mmのFluoroDish(商標)チャンバーにまいた。細胞を20時間培地においてCy5標識されたリポソームSNA(0.1μMのDNA濃度)と共にインキュベートし、続いて0.01%(容積によって)のトゥイーン−20を含む1×PBSで3回洗浄し、次いで新鮮な培地で置換した。核を、製造業者のプロトコルに従ってヘキスト3342(Invitrogen)で染色した。次いで、生細胞をMai Tai 3308レーザー(Spectra−Physics)でのZeiss LSM510 倒立レーザー走査共焦点顕微鏡で40×拡大にて撮像した。ヘキストを780nmにて励起して、390−495nmにて収集して、640nmにて励起し、及び650−710nmにて発光。
リポソームSNAの細胞の取り込みを遊離DNA鎖と比較するために、細胞を、100μLの培地中の96ウェルにまき、及び0.1μM濃度の遊離DNAまたはリポソームSNAと共に、及び24時間インキュベートした。未処理細胞を実験のための負の対照として使用した。インキュベーション期間の後、細胞を0.01%(容積の)のTween−20を含む1×PBSで3回洗浄して、次いでトリプシン処理して懸濁液を形成した。フローサイトメトリーは、背景強度に関して未処理細胞からのシグナルを使用して、Guava easyCyte 8HT(Millipore, USA)でCy5強度チャンネルを使用して細胞の懸濁液で行った。エラー値は、単一の試料を表する異なるウェルからの中央シグナルの平均値の標準誤差を使用して算出した。
リポソームSNAの細胞毒性を評価するために、SKOV−3細胞を実験の24時間前に96ウェルにまいた。細胞をDNAの種々の濃度のリポソームSNAで24時間処理した。リポソームSNAの細胞毒性をDharmaFECT(登録商標)1(Dharmacon)(市販のトランスフェクション薬剤)と比較した。細胞には、製造業者のトランスフェクションプロトコルに従ってDharmaFECT(登録商標)1でトランスフェクトした種々の濃度のDNAをトランスフェクトした。処理を受けなかった細胞は、負の対照として使用した。24時間のインキュベーション期間の後、細胞を1×PBSで3回洗浄して、alamarBlue(登録商標)溶液(Thermo Fisher Scientific Inc.)と共にインキュベートして、4時間5%のCO2中で37℃にてインキュベートした。590nmにおける蛍光発光を、BioTek, Synergy H4 Hybrid Readerを使用して記録した。
SKOV−3細胞を6穴プレートにまき、及び5%のCO2中で37℃にて一晩インキュベートした。細胞を抗HER2アンチセンスリポソームSNA及びスクランブル化リポソームSNAと共にインキュベートした。24時間の処理の後、培地を新鮮な培地で置換して、細胞をさらなる48時間増殖させた。HER2タンパク質ノックダウンを解析するために、細胞を収集して、プロテアーゼ及び脱リン酸化酵素阻害薬を含む100μLの哺乳動物細胞溶解緩衝液(Cell Signaling、MA、USA)に再懸濁した。(Thermo Scientific, IL, USA)。細胞可溶化液中のタンパク質濃度は、BCA Protein Assay Kit(Pierce、IL、USA)を使用して決定した。同量(20μg)のタンパク質を4−20%のPrecast勾配ゲル(Bio−Rad)によって分画して、ニトロセルロース膜(Thermo Scientific、IL、USA)へ転写した。膜をトリス−緩衝食塩水(TBS)中の5%の乾燥脱脂乳溶液(w/v)を使用してブロックした。タンパク質をHER2(1:1000)に対する一次ウサギ抗体及びGAPDH(1:500)続いて抗ウサギ二次抗体(1:10,000)(LI−COR Biosciences、NE、USA)で検出した。蛍光シグナルは、Odyssey(登録商標)Infrared Imaging System(LI−COR Biosciences、NE、USA)を使用して記録した。
典型的なリポソームSNAは、2工程で合成した(図7)。第1の工程は、脂質単量体から30nm直径単層小胞の調製を含む。このサイズ粒子は、SNAトランスフェクションの見地から理想的であり、及びより高い血液循環を最大にするために、及び腎臓を介したクリアランスを最小限にするために、適切な範囲にある。残念なことに、このサイズ状況のリポソームは、たいてい不安定であり、及び融合してより大きな構造を形成する。したがって、この研究の目標は、このような構造を合成し、及びこのような粒子成長経路を回避する方法を決定することであった。
Ramos−Blue(商標)細胞は、NF−κB/AP−1リポーターBリンパ球細胞である。Ramos−Blueは、NF−κB/AP−1−誘導性SEAP(分泌型胚性アルカリ性ホスファターゼ)リポーター遺伝子を安定して発現するBリンパ球株化細胞である。刺激したときに、これらは、上清にSEAPを産生し、これはQUANTI−Blueアッセイを使用して容易にモニターすることができる。QUANTI−Blueは、SEAPの存在下において青くなるSEAP検出媒体である(図14)。
本開示の組成物の有効性をさらに証明するために、リポソーム粒子を、HIF1−α及びBAXを個々にターゲットするようにデザインした。実験には、成長の早いマウス神経芽腫株化細胞であるNeuro−2a(N2A)株化細胞を利用した。HIF1−α(図18)及びBAX(図19)をターゲットするリポソーム粒子とN2A細胞を接触すると、標的遺伝子産物の量における有意な減少を示した。それぞれの実験において、mRNA発現の相対量は、6穴プレートにおけるN2A細胞の処理を開始した72時間後の定量的PCR(qPCR)によって決定した−細胞は、リポソーム粒子の除去及びMEM及び10%ウシ胎児血清(FBS)との培地の置換の前に、OptiMEM中で24時間リポソーム粒子で処理した。
Claims (48)
- 実質的に球状形状を有するリポソーム粒子であって、前記リポソーム粒子は:
複数の脂質基を含む脂質二重層;及び
オリゴヌクレオチド、
を含む、前記リポソーム粒子。 - 請求項1の前記リポソーム粒子であって、前記複数の脂質基は、脂質のホスファチジルコリン、ホスファチジルグリセロール及びホスファチジルエタノールアミンファミリーからなる群より選択される脂質を含む、前記リポソーム粒子。
- 請求項2の前記リポソーム粒子であって、前記脂質は、1,2−ジオレオイル−sn−グリセロ−3−ホスホコリン(DOPC)、1,2−ジミリストイル−sn−ホスファチジルコリン(DMPC)、1−パルミトイル−2−オレオイル−sn−ホスファチジルコリン(POPC)、1,2−ジステアロイル−sn−グリセロ−3−ホスホ−(1’−rac−グリセロール)(DSPG)、1,2−ジオレオイル−sn−グリセロ−3−ホスホ−(1’−rac−グリセロール)(DOPG)、1,2−ジステアロイル−sn−グリセロ−3−ホスホコリン(DSPC)、1,2−ジパルミトイル−sn−グリセロ−3−ホスホコリン(DPPC)、1,2−ジ−(9Z−オクタデセノイル)−sn−グリセロ−3−ホスホエタノールアミン(DOPE)及び1,2−ジヘキサデカノイル−sn−グリセロ−3−ホスホエタノールアミン(DPPE)からなる群より選択される、前記リポソーム粒子。
- 請求項1−3のいずれか1項の前記リポソーム粒子であって、前記オリゴヌクレオチドは、親油性連結基を含むオリゴヌクレオチド−脂質抱合体であり、前記親油性連結基は、脂質二重層の中に吸着される、前記リポソーム粒子。
- 請求項4の前記リポソーム粒子であって、前記親油性連結基は、トコフェロールまたはコレステロールを含む、前記リポソーム粒子。
- 請求項5の前記リポソーム粒子であって、トコフェロールは、トコフェロール誘導体、アルファ−トコフェロール、ベータ−トコフェロール、ガンマ−トコフェロール及びデルタ−トコフェロールからなる群より選択される、前記リポソーム粒子。
- 請求項1−6のいずれか1項の前記リポソーム粒子であって、前記オリゴヌクレオチドは、RNAまたはDNAを含む、前記リポソーム粒子。
- 請求項7の前記リポソーム粒子であって、前記RNAは、非コードRNAである、前記リポソーム粒子。

- リポソーム粒子を作製する方法であって:
溶媒にリン脂質を添加して第1の混合物を形成することであって、前記第1の混合物は、複数のリポソームを含むこと;
前記複数のリポソームを破壊して第2の混合物を作り出すことであって、前記第2の混合物は、リポソーム及び小単層小胞(SUV)を含むこと;
前記第2の混合物から前記SUVを単離することであって、前記SUVは、約20ナノメートルと50ナノメートルとの間の粒径を有すること;
単離されたSUVにオリゴヌクレオチドを添加してリポソーム粒子を作製すること、
を含む、方法。 - 請求項17の方法であって、前記第1の混合物中の複数のリポソームの粒径は、約100ナノメートル〜150ナノメートルの間にある、前記方法。
