Abstract
Description
i.十分な水中において0.9モル当量〜1モル当量のアルキルアミンと、1.0モル当量〜1.5モル当量の塩基とを混合して水溶液中に0.1M〜1.0Mの濃度であってpH8〜14のアルキルアミンを得る工程、
ii.上記混合物を50℃〜90℃の範囲の温度まで加熱して、アルキルアミンの加熱した水性塩基性溶液を得る工程、
iii.1モル当量のアダマンタンカルボニルクロリドを上記アルキルアミンの加熱した水性塩基性溶液に添加して二相性混合物を得る工程、
iv.上記反応が完了するまで、上記アルキルアミンと上記アダマンタンカルボニルクロリドの二相性混合物を撹拌して50℃〜90℃の範囲の温度に維持する工程、
v.上記反応混合物を撹拌して30℃を下回るまで冷却し、上記反応混合物から上記アダマンタンアミド生成物を沈殿させる工程、並びに
vi.上記アダマンタンアミド生成物を上記反応混合物から分離する工程、
を含む。
一般的には、本発明の方法によって製造され得るアダマンタンアミドに関して限定はない。本発明の方法は、アダマンタンアミドを製造するため開始材料として使用され得る多くのアダマンタン誘導体のうち、1−アダマンタンカルボニルクロリドが最も一般的であり、バルク量で商業的に入手可能であることから、1−アダマンタンアミドの合成に最も有用である。1−アダマンタンカルボニルクロリドとアミンとの反応により、1−アダマンタンアミドが得られる。しばしば、本発明の方法により製造されたアダマンタンアミドは、式I又は式IIを有する化合物によって表される。式Iのアダマンタンアミドは、一般的には本発明者らの目的の生物学的標的に対して試験した場合に式IIのアダマンタンアミドよりも更に強力であり、優れた機能的利益をもたらすことから、式Iのアダマンタンアミドが好ましい。
式中、各R1は独立して水素又はC1〜4のアルキルである。

本発明の方法の工程(i)は、アルキルアミンと水性塩基溶液を混合することを含む。アルキルアミンは、二級又は一級であってもよく、直鎖若しくは分岐鎖又は環状であってもよく、置換又は非置換であってもよい。アルキルアミンは、アルキルアミンのアルキル部分が所望のアダマンタンアミドの対応するアミド部分を形成するように選択される。
実験方法
全ての試薬及び溶媒を商業的な供給元から得て、更なる精製を行わずに使用した。
ヘキサン中10%酢酸エチルを使用する薄層クロマトグラフィー及び/又は三フッ化ホウ素:メタノール誘導体化を使用するガスクロマトグラフィー。後者の方法について、反応アリコート(100マイクロリットル)を様々な時間点で移し、クロロホルム中15%イソプロパノール:1N塩酸(500マイクロリットル:500マイクロリットル)に分割し、固体になるまで有機層を分離及び蒸発させて処理する。該固体材料(10ミリグラム)を三フッ化ホウ素:メタノール溶液(1ミリリットル)に溶解し、100℃で5分間加熱し、室温まで冷却して、ヘキサン:水(2ミリリットル:1ミリリットル)で希釈した。有機層を分離し/蒸発させ、残渣をアセトン(1ミリリットル)に溶解し、ガスクロマトグラフィー分析用に100万分の20部(parts per million)まで希釈した。モニタリングを様々な時間点(例えば、0分、15分、30分、60分、120分)で行って反応の完了を判断した。
Claims (10)
- アダマンタンアミドの製造方法であって、
i.十分な水中において0.9モル当量〜1モル当量のアルキルアミンと、1.0モル当量〜1.5モル当量の塩基とを混合して水溶液中に0.1M〜1.0Mの濃度であってpH8〜14のアルキルアミンを得る工程、
ii.前記混合物を50℃〜90℃の範囲の温度まで加熱して、アルキルアミンの加熱した水性塩基性溶液を得る工程、
iii.1モル当量のアダマンタンカルボニルクロリドを前記アルキルアミンの加熱した水性塩基性溶液に添加して二相性混合物を得る工程、
iv.前記反応が完了するまで、前記アルキルアミンと前記アダマンタンカルボニルクロリドの二相性混合物を撹拌して50℃〜90℃の範囲の温度に維持する工程、
v.前記反応混合物を撹拌して30℃を下回るまで冷却し、前記反応混合物から前記アダマンタンアミド生成物を沈殿させる工程、並びに
vi.前記アダマンタンアミド生成物を前記反応混合物から分離する工程、
を含む、アダマンタンアミドの製造方法。 - 前記方法が、前記反応混合物中の残留アダマンタンカルボン酸塩を再利用し、アダマンタンカルボニルクロリドへと変換する追加工程を更に含む、請求項1に記載の方法。
- 前記塩基が無機塩基である、請求項1又は2に記載の方法。
- 前記塩基がアルカリ金属水酸化物、炭酸塩、及びそれらの混合物からなる群から選択される、請求項3に記載の方法。
- 前記塩基が水酸化ナトリウムである、請求項3に記載の方法。
- 前記温度が55℃〜80℃の範囲である、請求項1〜5のいずれか一項に記載の方法。
- 工程(i)における前記pHが10〜12の範囲である、請求項1〜6のいずれか一項に記載の方法。
- 前記反応の完了が15分〜2時間を要する、請求項1〜7のいずれか一項に記載の方法。
- 工程(iii)で添加されるアダマンタンカルボニルクロリドが予め融解されている、請求項1〜8のいずれか一項に記載の方法。
- 工程(iii)で添加されるアダマンタンカルボニルクロリドが固体として徐々に添加される、請求項1〜8のいずれか一項に記載の方法。
