「細菌人工染色体(BAC)」の用語は、大腸菌などの細菌細胞内にDNA塩基配列をクローン化するために使用されるプラスミドDNA構築物を言う。典型的に、30,000個から約300,000個にわたる塩基対のDNA塩基配列が、BACに挿入され得る。挿入されたDNAを有するBACは、細菌細胞によって取り込まれ得る。細菌細胞が成長して分裂するにつれ、BAC DNAは、細菌細胞内において、非常に低い細菌細胞ごとのコピー数、好ましくは細胞毎に3コピーを超えない数、たとえば細胞毎に単一のコピーで安定して維持される。BACの複製は、複製起点(ori)配列、典型的にはoriS配列において開始される。この複製は、BACによってコードされる、概してrepEおよび/またはrepFという、遺伝子産物によって厳重に規制される。BACはさらに、parA、B、およびCなどの蛋白質をコードし、分裂時におけるBACコピーの配分を娘細胞に指示する。典型的に、BACベクターは、青色選択を可能にするlacZなど、抗生物質耐性またはリポーター酵素マーカーなどの選択可能マーカーをさらに含む。一般に使用されるBACの例はpBeloBac11(Shizuya et al. (1992) Proc. Natl . Acad. Sci. USA 89, 8794-8797)である。このベクターの配列は、GenBank登録番号U51113において報告された。pBeloBac11は、円形のプラスミドであって、oriSと、oriSにおける複製を開始および規制する蛋白質を産生するrepE遺伝子と、配分遺伝子parA、B、およびCとを含む。選択のために、pBeloBac11は、クロラムフェニコール抵抗コード遺伝子を含む。また、ベクターは、挿入断片がBACにおいてクローン化された時に妨害され得るもしくはベクターから除去されるlacZa遺伝子を含む。
動物
インターフェロンタイプIおよびIIレセプタ―の両方のノックアウトを伴う129/Svマウス(AG129マウス、B&K Universal Ltd/UK)がハウスにおいて育成された。
Vero−B(アフリカミドリザルの腎臓;American Type Culture Colletion(ATCC)CCL−81)およびBHK−21(ベビーハムスター腎臓細胞;ATCC CCL−10)細胞がATCCから取得された。すべての細胞は、本質的に記載のように培養された(De Burghgraeve et al. (2012) PLoS ONE 7, e37244)。黄熱ウイルスワクチン株17D(スタマリル(登録商標))は、ベルギー国ブリュッセルのSanofi Pasteur MSDから購入された。
pShuttle−BAC生殖のルーチンクローニングおよび生殖に使用されるバクテリア菌株は、それぞれ、大腸菌Top10(Invitrogen)およびEpi300−T(Epicenter)であった。全長フラビウイルスcDNAプラスミドpACNR−FL17DII、pACNR−DENV2、およびp4を用いて形質転換させたバクテリア(以下を参照)は、慣例的に28℃で成長させ、プラスミドDNAの収率は、クロラムフェニコール増幅によって増加させた。Epi−300T細胞を含むシャトルプラスミドは、以下に記載するように37℃で成長させて増幅された。酵母株サッカロミセスセレビシエYPH500(遺伝子型:MATα ura3-52 lys2-801_amber ade2-101_ochre trp1-Δ63 his3-Δ200 Ieu2-Δ1)は、Difco-BD BiosciencesおよびSigma-Aldrichwasからの選択培地上において成長させた。コンピテント酵母細胞の形質転換は、リチウム酢酸塩法を使用して行なわれ、酵母は28℃で成長させた。
YFV−17D、デング熱タイプ2(DENV2)、およびデング熱タイプ4(Denv4)のウイルスcDNAを含む本発明に係る細菌人工染色体は、以下に記載するように製造された。
プラスミド構築物(細菌人工染色体)
すべてのプラスミド構築物は、標準的な技術によって生成され、サンガー配列決定技法によって確認された。誘導性シャトルベクターpShuttleBAC/Pmeは、以下のいくつかのステップにおいて生成された(図1)。まず、アラビノース誘導性pBeloBAC/oriV(Wild et al. (2002) Genome Res. 12, 1434-1444)の誘導体であるpBAC/LacZ(Addgeneプラスミド#13422)内に存在するlacZ遺伝子が、(i)ヒグロマイシンBに対する抵抗性を有するhph遺伝子を駆動するシミアンウイルス40(SV40)プロモーター/起点(配列番号1)と、(ii)デルタ肝炎ウイルス(HDrz)のゲノムリボザイムの合成cDNA(Chadalavada et al. (2007) RNA 13, 2189-2201)(配列番号2)と、(iii)サッカロミセスセルビシエエピソーム2μプラスミド起点および(iv)トリプトファンを栄養原体増殖させるTRP1遺伝子とを含む合成DNAカセットによって置き換えられる。ビルディングブロック(i)は、プライマー#334および#425を使用してpBABEハイグロ(Morgenstern et al. (1990) Nucleic Acids Res. 18, 3587-3996;Addgeneプラスミド#1765)から増幅されたPCRであった。ビルディングブロック(ii−iv)は、プライマー#426および#231を使用してpJet(-)/Trp1_2micron-YF3'_HDrz_BstEから増幅されたPCRであった。プラスミドpJet(-)/Trp1_2micron-YF3'_HDrz_BstEは、YFV−17D cDNAの3′末端(ウイルスnt9466から10862を含む)と(ii)HDrz(DNAヌクレオチド#109、110、および111から組み立てられる)との融合体と、PCRプライマー#194および#195を使用してpRE637(Esteban & Fujimura, (2003) Proc. Natl. Acad. Sci. USA 100, 2568-2573)から取り出された(iii+iv)2μ−TRP1配列とを含む、pJet1.2/blunt(CloneJET PCRクローニングキット、Fermentas)の誘導体である。ビルディングブロック(i)は、pBAC/LacZのSalIサイトへクローン化される前にオーバーラップ伸展PCRによって(ii−iv)に融合され、pShuttleBAC/SV40_Hygro_HDrz中間構築物を生じさせた。続いて、hph遺伝子スタッファー要素が、プライマー#474および#475を使用した全長プラスミドにわたる逆PCR、およびT4 DNAリガーゼによる再円化によって、大腸菌における形質転換およびクローン化の前に、最終pShuttleBAC/Pmeシャトルベクター(図1A)における多重クローニングサイト(SfiI−PmeI−BstEII)のためのpShuttleBAC/SV40_Hygro_HDrzから置き換えられた。
一連の合成DNA構築物(pShuttleBAC)は、いくつかのDNAビルディングブロックから組み立てられ、RNAウイルス発現プラスミドとして機能した(図1)。共通のベクター構築物pShuttleBAC/Pmeは、大腸菌における状態増幅のための第2の起点(oriV)と、酵母2μ起点およびサッカロミセスセルビシエにおけるエピソーム複製のためのTRP1栄養要求性マーカーとを含む細菌人工染色体である(図1A)。YFV−17DワクチンのcDNAは、pShuttleBAC/Pme内に存在するSV40プロモーターとHDVリボザイムとの間の相同によってpShuttle/YF17Dを得るために挿入された(図1B)。pShuttle/YF17Dのマップが図8に示される(配列番号4)。他のフラビウイルスのcDNAもまた挿入された。pACNR・FLYF17DIIなどの先行技術のフラビウイルスcDNAクローンとは対照的に、pShuttle/YF17Dは、大腸菌における優位な遺伝子安定性を示し(図2)、高いプラスミドDNA収率を生成するように誘導され得る。
本発明に係る細菌人工染色体から発現されるYFV−17Dの特徴は原生の原生ワクチンのものと同一であることが分かった(複製の効率、ウイルス収率、およびプラーク表現型など)。また、この裸YFV−71DプラスミドDNAがAG129マウスの腹腔内に注射されると、親ウイルスと同様の病理、病的状態、および死亡率が結果として現れる。この簡便、堅牢、かつ再現可能な系は、真核細胞培養または有胚鶏卵を必要とすることなく低コストでYFVのDNAワクチンを提供する。これは、もはやコールドチェーンを必要とせず、針を用いずに投与することができる。
インビトロ転写、インビトロキャッピング、および電気穿孔
全長YFV−17D cDNAを含むプラスミドpACNR−FLYF17DII(Bredenbeek et al. (2003) J. Gen. Virol. 84, 1261-1268)は、AflIIを用いて線形化され、プロテイナーゼK消化、フェノールクロロホルム抽出、およびエタノール沈澱によって精製された。代替的に、インビトロ転写(IVT)のための全長cDNA鋳型がプライマー#173および#55ならびにKAPA高忠実度DNAポリメラーゼを使用したpACNR−FLYF17DIIのPCRによって作られ、プラスミド収率の制限が克服された。ランオフRNA転写産物は、インビトロにおいてSp6 RNAポリメラーゼを使用して産生された(Ribomax体規模RNA産生キット、Promega)。転写産物は、精製されたワクシニアウイルス7−メチルグアノシントランスフェラーゼ(Scriptcap7mGキャッピング系、Epicenter)を使用してキャッピングされ、キャリアRNAとしてBHK−21細胞から抽出された過剰な全RNAを使用したBHK−21細胞の電気穿孔のために使用された。細胞培養培地は、トランスフェクトされた細胞がほぼ完全な細胞病原性を示した時に摘出された。遠心分離によってセルデブリスから媒体が取り除かれ、続いてBHK−21細胞上にウイルスストックを準備するために使用された。
Vero−B細胞は、10%ウシ胎仔血清を含む媒地において6ウェルプレートに70%の培養密度まで接種され、1:3のDNA対ビヒクル比率でTransit−LTI試薬(Mirus)を使用して2.5μgのプラスミドDNAを用いて後日トランスフェクトされた。プラスミドpmRFP1は、1:10の比率で合わせて共にトランスフェクトされ、等しいトランスフェクション効能を確定するための視覚制御として機能した。長期にわたるウイルスストックの維持および産生のために、媒地は、2%のみの血清を含むように一夜の培養期間で変更された。
pShuttleBACベクター由来ウイルスRNAからの適切なプロセシングは、cDNA末端の急速増殖(RACE)によって分析された。新しい逆連結反応/増幅プロトコルの後の5′RACEは、かなり詳細に開示された(Dallmeier & Neyts (2013) Anal. Biochem. 434, 1-3)。非ポリアデニル化YFV−17DゲノムRNAの3′RACEは、本質的に以前に記載されたように行われた。簡単に、5μLのデオキシリボヌクレアーゼIで処理されたpShuttle/YF17DがトランスフェクトされたVero細胞の全RNA(RNAのうちの約1μg)は、16℃で一夜における10μLの全反応量における15%ポリエチレングリコール(PEG)−8000がある場合におけるT4 RNAリガーゼ2(New England Biolabs)のK227Q変異体の活動により、5′アデニル化および3′ジデオキシトシン(ddC)変質リンカー#393(miRNAクローニングリンカー1,IDT DNA技術)に連結された。連結反応生成物は、One−Step RT−PCR(Qiagen)によってYFV−17Dおよびリンカー特定プライマー#391および#394を使用してそれぞれ増幅された。265bpのアンプリコンは、Taq DNAポリメラーゼによって生成されてサンガー配列決定法によって分析された3′アデニンオーバーハングを精白した後に、ゲル精製され、pJet1.2/blunt(CloneJet PCRクローニングキット、Fermentas)にクローン化された。
アガロースゲル電気泳動を変性させた後のノーザンブロット法およびウイルス複製中間体の検出は、採用されたゲル系およびプローブ設計に関して僅かに変更を加えて本質的に記載のように行われた(Dallmeier et al. (2008) PLoS Pathog. 4, e1000230)。簡単に、3μgの全RNAは、20mMの3−(N−モルホリノ) プロパンスルホン酸(MOPS)、pH7.0、5mMの酢酸ナトリウム、ならびに1%ホルムアミドおよび0.01μgのエチジウムブロマイドmL−1を含む2mMのメチレンジアミンテトラ酢酸(EDTA)中の1%アガロースゲルを介して熱変性されて分離され、その後に20×SSC(3M塩化ナトリウム、300mMクエン酸ナトリウム,pH7.0)において正の電荷を有するナイロン膜(Roche Diagnostics)上に毛細転移された。YFV−17D(nt7637−8136)のNS5領域の5′末端に対して相補的な鎖特定DIGラベル一本鎖DNAプローブが、Knuchel et al. (2000) J. Histochem. Cytochem. 48, 285- 294により、プライマー#947および#948を使用して生成された。ハイブリダイゼーションおよび免疫検出は、フィルターハイブリダイゼーション(Roche Diagnostics)のDIG適用マニュアルによるものである。
トランスフェクトされた細胞から放出された感染性ウイルスは、 以前に開示された方法で(Kaptein et al. (2010) Antimicrob Agents Chemother. 54, 5269-8520)0.5×保存培地に置かれる1%微結晶セルロース(Avicell)を使用してBHK−21細胞の合流単層に対する感染7日のウイルスプラーク測定法によって定量化された。感染したマウス組織におけるウイルスRNA負荷の定量化のために、スナップフローズン検死が行なわれた。ここでは、RLTバッファーにおけるPrecellysビード粉砕機(Rneasy、Qiagen)において破砕され、その後に全RNAが製造者の説明書に従って抽出された。NS3遺伝子の約150nt延長を標的としたプライマーおよび基準としてプローブの使用および同じクローン化されたYFV−17D cDNA断片の系列希釈により、開示されたものと全く同じ方法で(Kaptein et al. (2010)上記)YFV−17D RNAのための定量的逆転写酵素PCR(RT−qPCR)が行なわれた。
トランスフェクション7日後のpShuttle/DV2トランスフェクトされたBHK−21細胞の取り除かれた上澄みが使用され、サブコンフルエントなVero−B培養を接種するために使用された。Vero−B培養は、4%パラホルムアルデヒドにおいて5日後に固定され、本質的に以前に開示された方法で(De Burghgraeve et al. (2012) PLoS ONE 7, e37244)免疫染色された。細胞内E蛋白質発現は、DENV2血清型特定単クローン抗体(mAb)3H5.1(ミリポア)、およびAb分類された二次Alexa Lour-488(ミリポア)によって検出された。DAPI着色の後、細胞はFLoid Cell Imaging station(Life Technologies)を使用して視覚化された。
10μgから20μgのプラスミドDNAが33%プロピレングリコール中においてキャリア(Fumoto et al. (2012) Mol. Pharm. 9, 1962-1970)としての20μg炭酸カルシウムマイクロフラワーと混合され、成体(約20g)のAg129マウスの腹膜中に注射された。代替的に、弱毒化YFV−17Dワクチン(スタマリル(登録商標)、Sanofi Pasteur MSD、ブリュッセル)の半分の投与量が腹膜中に注射された。重量および挙動が毎日監視された。
哺乳動物細胞内にトランスフェクトされた場合、適切にプロセシングされたYFV−17D RNAの転写は、細胞内の自律ウイルス複製(図3A)を開始するpShuttle/YF17D(図3B)から行なわれる。pShuttle/YF17Dがトランスフェクトされた細胞は、最終的に感染性ビリオンを分泌する。感染性ビリオンは、組織培養内における細胞変性効果を誘導する能力(図4Aから図4C)、ウイルス収率、およびプラーク表現型(図4D+図4E)に関し表現型が親YFV−17Dウイルスとは区別することができない。産生された組み換えウイルスは、このため、生物学上同一(定量的および定性的)であると考えられる。同様に、pShuttle/DV2がトランスフェクトされた細胞は、組み換え感染性DENV2ニューギニア株C(図5)を産生する。
腹腔内経路以外のpShuttle/YF17D(DNA−YFVax)の適用経路の実現可能性を評価するために、9週齢の雄AG129マウス(雄、重量22gから25g)のグループ(n=3)に対し、100μLの燐酸塩緩衝食塩水 (PBS)中の25μgのDNA−YFVaxが、(i)注射器およびG27針を使用して皮下(s.c.)に、または(ii)たとえばインシュリンもしくは麻酔薬のヒトへの医療的使用のための経皮的適用が許可された針無し噴射注射器(Injex-30、Injex Pharma GmbH、ベルリン、ドイツ)によって経皮的に(t.d.)注射された。上記のように200μLの33%プロピレングリコール中の炭酸カルシウム微細結晶を用いて作られた25μgのDNA−YFVaxが腹腔内に注射されたAG129は、対照群としての役割を果たした。病的状態および死亡率は上記のように評価され、試験は30日後に終了した(表2を参照)。
DNA-YFVaxによるYFVに対する防護免疫の誘導を評価するために、症状発現前のシリアンゴールデンハムスター(Mesocricetus auratus)モデルが使用された(Tesh et al. (2001) J Infect Dis. 183, 1431-1436)。このため、雌ハムスターのグループ(8週齢から10週齢、90gから100g)は、スタマリル(登録商標)の1/5投与量(100μL)、または上記のように200μLの33%プロピレングリコール中の炭酸カルシウム微細結晶を用いて形成された20μgのpShuttle/YF17D(DNA−YFVax)のいずれかの腹腔内(i.p.)注入によって免疫化された。代わりに、繰り返しで10μgのDNA−YFVaxが適用された。血液は、完全な外科麻酔下において心臓穿刺によって週ごとに採られ、血清は、遠心分離によって採取され、−80℃の冷凍で保存された。2匹の未処置のハムスターが、正常血清としての役割を果たした。関連する免疫保護を評価するために、(i)間接免疫蛍光法分析(IIFA)、および(ii)プラーク減少中和テスト(PRNT)によって結成が分析された。
(a)開始材料:pACNR−FLYF17DII(Bredenbeek et al. (2003)上記)およびpShuttle/YFV−17Dの大規模プラスミド製剤が標準的な技術を使用して行なわれた。このため、pACNR−FLYF17DIIは、標準的な大腸菌K12誘導体株において形質転換され、100μg/mLアンピシリンを含むLB寒天培地上で平板培養され、好ましいプラスミド安定性となるように28℃(37℃の代わり)で一夜にわたって成長された。小さなコロニーが2日間の連続的な一夜培養において強度の振動下において28℃で100μg/mLを含むLBにおいて拡大および成長し、最終的に1Lバッチ培養に到達した。このバッチは、28℃で一夜で成長し、最終的に、クロラムフェニコールを20μg/mLの最終濃度となるまで添加することによってさらに28℃で8時間にわたって増幅された。同様に、pShuttle/YFV−17Dは、大腸菌株EPI300−T細胞(Epicentre)において形質転換され、20μg/mLクロラムフェニコールを含むLB寒天培地上で平板培養され、さらに37℃で一晩で成長した。後者のプラスミドは、これに伴って拡大し、さらにすべての増殖が20μg/mLクロラムフェニコールの存在下において37℃で行なわれた。最終的な一夜のバッチ培養は、20μg/mLクロラムフェニコールおよび0.01%Lアラビノースを含む新鮮なLB媒体に6分の1に希釈化され、6時間以下の時間にわたって成長された。プラスミドは、標準的なカラム親和性精製(Qiagen)を使用して精製され、TE(10mM トリス−HCl、1mM EDTA)において1μg/mLの最終濃度に溶解され、−20℃において冷凍保存された。
YFV−17D cDNA含有プラスミドのクローン遺伝子安定性に対処するために、実施例6に記載される両方のプラスミドは、大腸菌EPI300−Tに形質転換され、上記のように選択培地として適切な抗体を含有するMacConkey寒天培地上に筋状で摂取された(実施例6bを参照)。pACNR−FLYF17DIIクローンは、28℃で24時間にわたって培養され、pShuttle/YF17Dクローンは、37℃で16時間にわたって培養された。
pASおよびpALシリーズの初期経路からのプラスミドの直接配列(低い経路番号P0からP3)(表6)は、元のクローンプラスミド製剤において13%という高い変異頻度を示した(実施例6aを参照)。見付かったほぼすべての変異は、未熟停止コードン(PMSt)および単一のヌクレオチド欠失/挿入の導入によるノンセンスもしくはフレームシフト変異(FS)であった。これらの変異は、全長YFV−17D読取枠(ORF)の明らかに完全な切除発現であった。pSLシリーズにおいて、同様の変異はP1において見られたが、頻度は低かった。P0におけるpSSシリーズにおいて、ウイルスORFの発現を消失させないミスセンス変異が見付かった。
pALおよびpASシリーズからのプラスミドが経路P10において分析された時、プラスミドの大部分は、大きな構造的な再配列を含み、これにより、48のうちの10(21%)および48のうちの44(92%)のプラスミドクローンが、nt2500−3600領域の増幅に完全に失敗した(表8)。数キロベースに至るDNAがプラスミドから消失するという、後者の変異体プラスミドに異常な制限パターンが伴った。注目すべきは、試験されたすべての変異体クローンは、依然としてnt1−940領域(表6)を含み、無関係の汚染物ではなく元のpACNR−FLYF17DII(表8)の子孫である可能性が高かった。このような毒性のcDNA断片の欠失は予期されており、先行技術において報告されている(Yamshchikov et al. (2001)上記)。
YFV−17Dの組み換えキメラ型誘導体が開発され、非対応抗原のためのベクター(Guy et al. (2010) Vaccine 28, 632-49; 米国特許出願公開第20100278773号)、たとえば日本脳炎ウイルス(JEV)および西ナイルウイルス(WNV)のprMおよびE蛋白質などの他の病原性フラビウイルス抗原の表面糖蛋白質のためのベクターとしてYFV−17Dが機能するワクチンとして使用される。これらは、ChimeriVax−JE(Imojev(登録商標) Sanofi Pasteur-MSD)およびChimeriVax−WN20としてそれぞれ開発された。本発明に係るpShuttle/YFV−17D(DNA−YFVax)は、トランスフェクトされたプラスミドDNAからこれらの上記のChimeriVaxワクチンウイルスを直接的に開始することができるように変更され得て、生ウイルスを含有する前記弱毒化ChimeriVax−JE(Imojev(登録商標))およびChimeriVax−WN20ワクチンと完全に置き換えることができる。
エンテロウイルス属は、センス方向におけるRNAゲノムを有する小さい無外膜ウイルスのピコナウイルスのファミリーに属する(+)−RNAウイルスである。典型的に、ピコナウイルスのゲノムは、キャッピングされないが、代わりに5′末端が共有結合で取り付けられたVPg蛋白質を担持する。キャップ構造の無い状態で、内部のリポソーム導入部位(IRES)は、ウイルス蛋白質発現のためのウイルスRNAへの細胞翻訳機構を採用する。原則的に、ピコルナウイルスにおける感染性ウイルス後代の複製および産生は、ウイルスゲノムの異種転写の後に細胞内で開始される。先行技術において、ウイルスゲノムは、 ファージプロモーターの制御下におけるcDNAから発現され、同族のファージRNAポリメラーゼがプロデューサ―セル(2プラスミド系)に共トランスフェクトされ、発現した場合にのみ転写される。同様に、前記ファージポリメラーゼは、組み換えバキュロウイルス(Yap ef al. ( 1997) Virology. 231, 192-200)などのヘルパーウイルスを使用したcDNAの形質導入時に細胞内で発現され得る。

- ワクチンの製造のための細菌人工染色体(BAC)の使用であって、前記BACは、
−細菌細胞ごとに10より大きい数のコピーに前記BACを増幅させるための誘導性細菌ori配列と、
−弱毒化RNAウイルスゲノムのcDNAを含み、哺乳動物細胞における前記ウイルスcDNAの転写および感染性RNAウイルスへの転写されたRNAのプロセシングのためのシス調節因子を含む、ウイルス発現カセットとを備える、使用。 - 弱毒化RNAウイルスゲノムの前記cDNAは、RNAウイルスゲノムのキメラ型ウイルスcDNA構築物であり、異種のDNA塩基配列が挿入される、または生来ウイルス配列が削除、短縮、もしくは変異される、請求項1に記載の使用。

- 前記酵母ori配列は、2μプラスミド起点またはARS1(自己複製配列1)またはその機能的に同種の誘導体である、請求項8に記載の使用。
- 前記RNAポリメラーゼ駆動プロモーターはRNAポリメラーゼIIプロモーターである、請求項3に記載の使用。
- 前記RNAポリメラーゼIIプロモーターは、サイトメガロウイルス最初期(CMV−IE)プロモーター、シミアンウイルス40プロモーター、またはその機能的に同種の誘導体である、請求項10に記載の使用。
- 前記RNAポリメラーゼ駆動プロモーターは、RNAポリメラーゼIもしくはIIIプロモーターである、請求項3に記載の使用。
- 前記自己開裂用要素は、デルタ肝炎ウイルスのゲノムリボザイムのcDNA、または機能的に同種のRNA要素である、請求項3から11のいずれか1項に記載の使用。
- 前記ウイルス発現カセットは、 弱毒化YFV−17DワクチンのcDNAを含み、ビリオン表面蛋白質のためのcDNAコードのうちの1つ以上が欠失、短縮、または変異され、YFV−17Dのこのような機能的ビリオン表面蛋白質が発現されず、異種の蛋白質についてのcDNA配列コードがYFV−17D cDNAに挿入される、請求項6に記載の使用。
- 前記異種の蛋白質は、フラビウイルスのビリオン表面蛋白質である、請求項14に記載の使用。
- 前記ウイルス発現カセットは、弱毒化YFV−17DワクチンのcDNAを含み、1つ以上の無関連のcDNA配列が、前記ウイルス性ポリ蛋白質内での1つ以上の異種の蛋白質として発現されるように挿入される、請求項14または15に記載の使用。
- 前記ウイルス発現カセットは、ウイルスcDNAを含み、外部cDNA配列が前記組み換えウイルスによって異種に発現されるように挿入される、請求項1から16のいずれか1項に記載の使用。
- RNAウイルスに対するワクチンを製造する方法であって、
a)BACが移入される細菌宿主を提供するステップを備え、前記BACは、
−細菌細胞ごとに10より大きい数のコピーに前記BACを増幅させるための誘導性細菌ori配列と、
−弱毒化RNAウイルスゲノムのcDNAを含み、哺乳動物細胞における前記ウイルスcDNAの転写および転写されたRNAから感染性ウイルスRNAへのプロセシングのためのシス調節因子とを含むウイルス発現カセットとを含み、方法はさらに、
b)前記誘導性oriを活性化させる化合物を加えることによって前記BACを増幅させるステップと、
c)前記増幅されたBACを単離するステップと、
d)前記BACをワクチンに形成するステップとを備える、方法。 - BACであって、
−細菌細胞ごとに10より大きい数のコピーに前記BACを増幅させるための誘導性細菌ori配列と、
−弱毒化RNAウイルスゲノムのcDNAを含み、哺乳動物細胞における前記ウイルスcDNAの転写および転写されたRNAから感染性ウイルスRNAへのプロセシングのためのシス調節因子とを含むウイルス発現カセットとを備え、

−細菌細胞ごとに10より大きい数のコピーに前記BACを増幅させるための誘導性細菌ori配列と、
−弱毒化RNAウイルスゲノムのcDNAを含むウイルス発現カセット、または
−弱毒化RNAウイルスゲノムのcDNAを含み、哺乳動物細胞における前記ウイルスcDNAの転写および転写されたRNAから感染性ウイルスRNAへのプロセシングのためのシス調節因子とを含むウイルス発現カセットとを備え、
RNAウイルス感染の予防に使用される、BAC。 - 生DNAワクチンとしての請求項1から18のいずれか1項に記載の細菌人工染色体の使用。
- 生来もしくは組み換えRNAウイルスゲノムのcDNAの維持、および前記cDNAからの生来もしくは組み換えウイルスの増殖のための、請求項1から18のいずれか1項に記載の細菌人工染色体の使用。
- ワクチンの製造のための細菌人工染色体(BAC)であって、前記BACは、
−細菌細胞ごとに10より大きい数のコピーに前記BACを増幅させるための誘導性細菌ori配列と、
−弱毒化RNAウイルスゲノムのcDNAを含み、哺乳動物細胞における前記ウイルスcDNAの転写および転写されたRNAから感染性ウイルスRNAへのプロセシングのためのシス調節因子とを含むウイルス発現カセットとを備える、細菌人工染色体。
