Abstract

Description
本発明は、ギャップ結合関連タンパク質発現の調整のための薬剤、組成物、および化合物の使用方法に関する。これらの薬剤、組成物、および方法は、例えば、インビボおよびインビトロでの組織工学(例えば、皮膚、角膜組織、および眼の外科的手順に伴う結合が含まれる)に有用である。
疾患または損傷による組織不全または臓器不全は、臓器または組織の移植以外に完全に回復する選択肢がほとんどない世界的に主な健康上の問題である。しかし、適切なドナーを見出す際の問題は、大部分の患者がこの選択肢を利用できないことである。組織工学または再造形による完全に機能を果たすか所望の機能を果たす合成または半合成の組織または器官の模倣物は、代替手段として現在調査されている。

(材料と方法)
(レーザー処置)
雌Wistarラット(d32〜34)を、実験動物の使用に関するARVO規則と一致した条件下で飼育した。動物を、Hypnorm(商標)(10mg/ml、Jasen Pharmaceutica,Belgium)とHypnovel(登録商標)(5mg/ml、Roche products Ltd,New Zealand)との1:1混合物の0.83ml/100g体重の用量での腹腔内投与によって麻酔した。
角膜レーザー治処置前後に、各動物を、Confoscan 2(Fortune Technologies America,USA)スリットスキャニングインビボ共焦点顕微鏡を使用して臨床的に観察した。Confoscan 2は、収集時間毎に画像を直接デジタル化するという明確な利点を有するスリットスキャニングテクノロジーの改良型である。動物を麻酔し、それぞれ収集ヘッドの前にインビボ強焦点顕微鏡の対物レンズの位置がくるように調整した特別にデザインしたプラットフォームに置いた。
スリットスキャニングインビボ共焦点顕微鏡を使用して収集した全画像を、ハードディスクドライブに保存し、その後、NAVISプロプラエタリソフトウェア(Confoscan 2,Nidek Co Ltd)によって分析した。

適切な角膜切片を、異なる目的のための異なるマーカーで免疫組織化学的に染色した。核染色剤Hoechst33 258を使用して染色を使用して、角膜中心または周辺角膜の上皮細胞数および間質細胞数を評価した。この目的のために、AnalySIS(登録商標)3.2ソフトウェア(Soft Imaging System,USA)を使用して、目的の領域を、最初に適切な角膜領域のTIFFファイル画像上にフリーハンドで描き、領域の値をソフトウェアによって自動的に得た。次いで、手動計数オプションを使用して、その領域内の細胞を計数し、単位領域あたりで示す。
30%プルロニック酸ゲル(BASF Corp)を含むリン酸緩衝化生理食塩水(分子グレードの水)を使用して、未修飾のα1コネキシン(コネキシン43)特異的アンチセンスODNをレーザーフォトリフラクティブケラテクトミー後の麻酔ラットの結膜下に送達させた。FITCタグを使用した予備試験では、処方物は24時間まで眼の前房に残存することが示された(示さず)。
アンチセンス適用後、レーザー手術から2時間、12時間、24時間、48時間、72時間、1週間、および2週間後の角膜を、スリットスキャニングインビボ共焦点顕微鏡を使用して試験した。コントロールラットは、レーザー手術のみを受けた。
ODN=AS ODN処置した眼(レーザー手術後1回投与)。
抗コネキシン43 ODNでの処置により、上皮回復が促進された。AS ODN処置角膜の90%で、PRKレーザー手術から12時間以内に擦過性上皮細胞が認められ、コントロールでは認められなかった(図1B)。この段階で、30%のコントロール角膜で停止内皮細胞のみが認められた(図1A)。24時間までに、全コントロール細胞およびアンチセンス処置角膜で上皮細胞が認められたが、コントロールの61%に対して処置群の72%が活発な擦過性細胞が認められた。これは、再上皮形成は、コントロールよりもコネキシン43特異的AS ODN処置角膜で迅速に進行することを示す。
選択された測定点でのコントロールとODN処置角膜との間の間質細胞数を比較するための時間とMann Whitneyノンパラメトリック統計的検定との関数としての各群内間質前部および間質後部の細胞密度を比較するための反復測定を使用した両側サンプルt検定を使用して、レーザー手術から24時間後のみで統計的に有意な結果が得られた(表2)。この測定点で、コントロールおよびODN処置群では、間質前部の間質細胞密度は、術前値と比較して非常に増加した(p値<0.05)。コントロール角膜の間質後部では、間質細胞密度はまた、術前値と比較して増加したが(p値<0.05)、ODN処置角膜の間質後部では、間質細胞密度は術前値と統計的に有意に異ならない(p値>0.05)。間質前部および間質後部の2群間の間質細胞密度を比較した場合、ODN処置角膜は、常にコントロール角膜よりも間質細胞密度が低かった(p値<0.05)。これは、コントロール角膜と比較してODN処置角膜がより少数の細胞が間質再造形またはエンジニアリングに関与するという概念を支持する。これは、抗コネキシン43 ODNの適用によって手術部位の細胞過形成が減少することを示す最初の報告である。エキソビボ組織化学分析(実施例II)は、この細胞過形成が望ましくない間質マトリクス(stromal matrix)再造形および瘢痕を誘導する筋肉線維芽細胞に関連することを示す。
(材料と方法)
(組織学:組織の採取および固定)
適正な数の動物(Wistrarラット)を、レーザーフォトリフラクティブケラテクトミー後の選択された測定点で屠殺し、上記の実施例1に記載のようにDB1抗コネキシンODNを麻酔ラットに投与し、組織学的分析のために角膜切片を調製した。コントロールラットには、レーザー手術のみを受けさせた。眼全体およびコントロール組織を、Oxoid PBSでリンスし、その後にTissue−Tek(登録商標) OCT(Sakura Finetek,USA)に包埋し、液体窒素中で凍結させた。必要に応じて(いくつかの抗体の使用のために)、凍結組織を、冷アセトン(−20℃)中で5分間固定し、クリオカットで切断した。
低温切片作製手順は以下のように行った:非固定組織の凍結塊を−80℃での貯蔵庫から取り出し、Leica CM3050Sクルオスタットに約20分間入れて、クリオスタットと同一の温度(すなわち、−20℃)に平衡化した。組織の平衡化が達成された時点で、標本をTissue Tek(登録商標)OCTを備えた標本皿に置いた。厚さが12μmの切片(H/E染色のため)または25μm(免疫標識のため)の切片に切断し、Superfrost(登録商標)Plus slides(Menzel−Gleser,Germany)上に置いた。低温切断直後に、組織塊を−80℃の貯蔵庫に戻し、低温切片を支持するスライドを直後に使用するか、−80℃で保存した。眼の光軸と平衡に切片にした。
一連の異なる染色試薬への浸漬を容易にするために、スライドをガラスラックに入れた。表面張力を下げるための試薬にラックを入れて震盪し、各溶液を交換する毎に排出させた。Gill’sIIヘマトキシリン/エオシン染色前に、−80℃で保存していたスライドを、最初に1〜2分間室温まで加温し、冷アセトン中で固定し、そして/または直後に水道水に短期間浸して水和した。スライドをGill’sIIヘマトキシリン中で2分間染色し、その後、過剰な染色を水道水でリンスオフした。Scottの水道水代替物(STWS)への4秒間の浸漬によって識別染色(stain differentiation)を行った。次いで、流水で1分間リンスし、30分間の継続的な浸漬により1%エオシンで染色した。最後に、切片を水道水で即座にリンスし、95%、100%EtOHで脱水し、キシレンで洗浄し、DPX封入剤(Sigma)を使用してマウントした。核の対比染色(H/Eまたは免疫組織化学分析と並行)のために、Hoechst 33
258(Sigma)を使用した。H/E染色切片に対して角膜の厚さを測定した。
切片を、部位特異的モノクローナル抗体を使用してコネキシン43について免疫標識し、α平滑筋アクチンを認識する抗体を使用して筋線維芽細胞について免疫標識し、抗ラミニン−1抗体を使用して基底膜蓄積について免疫標識した。さらに、抗ビメンチン抗体を使用して、角膜間質細胞と筋線維芽細胞とを識別し、Ki−67抗体を使用して細胞増殖を示した。
結果は、エキシマレーザー光焼灼によって作製された病変が術後24時間までに閉じることを示した(図2)。角膜の周囲および中心への単核/多核および/または円形、卵形細胞による間質への典型的な侵入が両群で認められ、術後24時間で最も顕著であるが、アンチセンスODN処置群は、コントロール群と比較してこれらの細胞数が有意に少なかった(図2A、B、C)。これは、実施例1で認められたインビボ共焦点顕微鏡写真由来の所見に匹敵する。上皮の厚さは、レーザー誘導性病変部位の図2で認められるコントロール角膜(図A、B)、焼灼領域下の瘻孔(図A、B)、および周囲間質(図C)で様々であり、コントロール角膜中の円形細胞による広範な侵入(細胞過形成)が認められた。図Bおよび図Cで顕著な間質水腫も認められた。アンチセンスODN処置角膜では、上皮は均一な厚さであり(図D、E)、中央領域(図D)および周囲間質(図E)では、間質水腫の徴候はほとんどなかった。さらに、間質では、円形細胞はほとんど存在しなかった。図2のスケールバーは、20ミクロンを示す。
手術に供していない角膜の中央間質の厚さは250μmであるが、エキシマレーザー手術を使用して、70μmの角膜組織(上皮および間質の一部を含む)を除去した。正常な角膜上皮の厚さは50μm(平均)であるので、20μmの間質組織をレーザー手術によって除去した。したがって、創傷から24時間後、48時間後、および72時間後の中央間質の厚さと術前の中央間質の厚さとを統計的に比較するために、除去した厚さと術前の中央間質の厚さとを調整して使用した(250−20=230μm)。
顕微鏡による観察により、コントロール角膜と比較してODN処置角膜中に存在するコネキシン43レベルの減少が認められた。図3は、顕微鏡画像の組み合わせを示す。上の列はコントロール角膜を示し、下の列はアンチセンスODN処置角膜を示す。円形細胞による典型的な間質の侵入が、縁部、周囲、および中央領域で24時間以内に両群で認められた。しかし、ODN処置角膜における円形細胞の密度はより低かった。両群の縁部で、抗コネキシン43は間質に均一に分布したが(3A、D)、処置群は、コントロール(3B)と比較して周囲(3E)のレベルが低かった。この段階では、コネキシン43レベルは、両群の上皮で正常に戻ったが、コントロール群では、瘢痕様間質(3C)または過形成(以下の図4を参照のこと)が認められたのに対し、アンチセンス処置角膜では、正常レベルのコネキシン43を有する正常な上皮が認められた(3F)。スケールバーA、D、E、Fは、10ミクロンを示し、BおよびCは20ミクロンを示す。これらの図では、コネキシン43は、白色の点状標識として認められ、細胞核が灰色で出現する。図3に示した結果により、コネキシン43タンパク質レベルが抗コネキシン43ODNでの処置後に減少し、それにより、間質中の細胞動員度がより小さくなることが示唆される。さらに、コントロール角膜の31%(術後24時間で25%、術後48時間で67%、術後72時間で0%)と比較して、たった7%(術後24時間で0%、術後48時間で0%、術後72時間で20%)のODN処置角膜しか上皮過形成の徴候を示さない。これを、H/E染色切片およびKi−67標識切片に対して評価した。
ビメンチン抗体での標識は、AS ODN処置角膜と比較したコントロール角膜の間質中の細胞数の増加が未分化の角膜間質細胞起源ではないことを示したので、α平角筋アクチン抗体を使用して標識を行った。この標識は、コントロール角膜が手術部位下により多数の筋線維芽細胞を有するが、周辺の周囲間質にも有することを示した。この筋線維芽細胞数および罹患領域の増加は、術後24時間で顕著になり、48時間および72時間から少なくとも1週間にわたって持続した(表4)。図4は、レーザー手術から1週間後の筋線維芽細胞標識(抗α平滑筋アクチン)を示す。図4A、B、およびCはコントロールであり、図4D、E、およびFはアンチセンス処置角膜である。創傷後1週間までに、コントロール角膜では、少数から中程度の数の筋線維芽細胞が周囲間質の前半部に存在し(4A)、中程度から密集レベルで中間周囲間質領域に存在し(4B)、中程度のレベルで中央領域の間質の後半部に認められる(4C)。対照的に、処置角膜では、非常に少数の筋線維芽細胞が周囲間質(4D)または中間周囲間質(4E)に存在し、中程度から少数が中央間質に存在する(4F)。中央間質のいくつかの場合、筋線維芽細胞は、上皮の真下の領域に密集している(示さず)。したがって、上記実施例1(細胞過形成)および図2で認められた細胞数の増加は、筋線維芽細胞の分化および侵入に起因するようである。筋線維芽細胞は、瘻孔中の瘢痕組織蓄積、それに伴う結晶蓄積の減少および創傷コラーゲンIII分泌の減少を担うことが公知である(Ahmadi A.J.and Jakobiec F.A.;2002;Int Ophthalmol Clin.Summer;42(3):13− 22.)。
レーザーフォトリフラクティブケラテクトミー後、再生上皮と共に基底膜が再形成される。ラミニン−1に対する抗体での標識は、再形成された基底膜が不連続であり、上皮−間質が不規則に結合していることを示す(図5)。24時間後、コントロールは焼灼領域の縁部(図5A)およびより中央の領域(図5B)にラミニン蓄積はほとんどなく、そして/または不規則であるのに対して、アンチセンス処置角膜は、これらの両領域でより規則的なラミニン蓄積を示した(図5C、図5D)。48時間後、コントロールは連続的にラミニンを蓄積せず(図5E−焼灼領域の縁部;図5F−中央)、非常に不規則であった(図5E)。対照的に、アンチセンスODN処置角膜は、創傷縁部(図5G)および中央(図5H)で連続的且つ比較的平らな基底膜を有していた。図5中の全スケールバーは、20ミクロンを示す。コネキシン43アンチセンス処置角膜は、24時間以内にあまり不規則でないより密集したより連続的な基底膜を形成した。
角膜を、エキソビボ臓器培養モデルおよびアンチセンスODNを使用して調整した特異的コネキシンに置いた。2つのコネキシン(コネキシン43およびコネキシン31.1)がこれらの実験で標的化された。コネキシン43の下方制御を使用して、コネキシンをインビトロで制御することができることを証明し、コネキシン31.1は、角膜から剥がれ落ちようとしている細胞中の角膜の上皮外層中に発現するので、コネキシン31.1が標的化された。コネキシン31.1発現の減少によって上皮組織の肥厚を操作することを目的とした。
30〜34日齢のWistrarラットを、Nembutalまたは二酸化炭素で安楽死させ、ラットの全眼を切り出した。眼表面を切り出し、0.1mg/mlペニシリン−ストレプトマイシンで5分間殺菌し、滅菌PBSでリンスした。次いで、全眼を、角膜を上にして60mmの培養皿中の滅菌ホルダー上に移した。眼を、空気−培地界面に角膜上皮を曝露させてマウントし、無血清培地(Opti−MEM,インビトロgen)中にて34℃の加湿5%CO2インキュベーター中で48時間培養した。上皮を湿らせるために、100μlの培地を8時間〜12時間毎に表面に滴下した。培地レベルを、結膜縁部レベルに維持した。
成体動物モデルにおける脳または脊髄損傷後のギャップ結合タンパク質コネキシン43に特異的なアンチセンスオリゴヌクレオチドの適用により、病変の拡大が遮断され、炎症反応およびその後の瘢痕形成が軽減される。本発明者らは、本発明者らのアンチセンスアプローチを、確立された病変のための修復ストラテジーを精査するための脊髄セグメントおよびインタクトな脊髄についてのエキソビボ培養モデルのためになおさらに開発した。
末梢神経移植のために、本発明者らは、移植時にコネキシン43特異的アンチセンスオリゴデオキシヌクレオチドで組織を再処置し、それにより、移植部位からの病変の拡大および宿主組織の神経集団が単離されてニューロン修復が制限される小グリアの活性化を予防する。
脊髄セグメントを、35mm皿中の培養インサート(Millipore Millicell)に入れ、セグメント上にメニスカス(miniscus)が形成されるまで培養培地レベルを増加させた。コネキシン43特異的アンチセンスオリゴデオキシヌクレオチド(30量体、濃度1μM)を含む30%プルロニックゲルを、直ちに脊髄組織上に注ぐ。生理学的温度に加温されるとゲルは固化し、アンチセンスオリゴヌクレオチドが徐放される。この処置により、24時間と48時間との間のコネキシン43タンパク質レベルが減少し、処置から6〜8時間後に最も減少する。培養で安定化し、その後切創に再曝露したこのような脊髄セグメントは、インビボでの手術介入と同一の症状(切断領域への病変拡大および組織腫脹が含まれる)を示す。この効果を、切開時のコネキシン43特異的アンチセンスオリゴマーでの処置によって防止することができ、したがって、切断末端は鋭いままであり、浮腫または組織腫脹の明らかな徴候は認められない。
al.,1999,Exp Neurol.159:131−138;Aguayo,A.J.et al.,1981,J:Exp.Biol.95:231−240)または肋間神経(Cheng,H.,et al.,1996,Science,273:510−513)移植が軸索延長およびニューロンの生存を誘導する高い可能性を有することが示されている。
処置セグメントの間の移植物のために、軸索再生の強力なプロモーターであることが示されているので、シュワン細胞を選択した(Xu,X.M.et al.,1999,J.Neurosci.11:1723−1740;Keirstead,H.S.et al.,1999,Exp.Neurol.159:225−236)。
移植片組織に対する即時型応答、遅延型応答、または連続的応答が存在することを確立するために末梢移植片および移植物の両方についての経時変化実験を行う。いくつかのマーカーを使用して、ニューロン応答および修復潜在性を評価する。これらには、以下が含まれる:ニューロン(Neuronal−Nに対する抗体)、神経細繊維(MAP−2およびSMI−31に対する抗体)、および細胞質マーカー(CMFDA)、および膜色素(Di−IまたはAxon grease−Molecular Probes,Oregon,USA)。ニューロン新芽形成の増加、軸索移動距離の増加(距離移動比に対する橋長)、および移植片に向かうか通過する軸索成長数の増加を特にモニタリングする。細胞特異的マーカー(樹状細胞についてはGFAP、小グリア細胞についてはIsolectin−B4、およびシュワン細胞についてはS−100)。グリア細胞の分布および密度、ならびに髄鞘形成レベルを評価する。脊髄セグメントから再生した軸索とは対照的に、抗CGRP(末梢神経マーカー)を使用して、末梢神経起源の軸索を識別する。GAP−43(成長関連タンパク質)抗体を使用して、ニューロン成長錐体を同定する。トルイジンブルー染色半薄切片および移植片断面の電子顕微鏡法を、形態学的分析のために使用する。
Unit Leica TCS 4DおよびSP2共焦点レーザースキャニング顕微鏡を使用して分析する。HItachi H−7000電子顕微鏡にて電子顕微鏡法を行う。画像分析プログラム(AnalySISまたはNIH Image J)を使用して、コントロールと処置移植片との間の装置を定量する。
(材料)
本明細書中で使用した材料には、当該分野で認識されている抗体およびプラスミド(例えば、ラットコネキシン43(T7291)およびコネキシン26のプラスミド;マウスコネキシン43およびコネキシン26(インビボgen,USA)、マウス抗ラットコネキシン43およびウサギ抗ラットコネキシン26(Zymed)(51−2800)のプラスミド;ならびにMolecular Probes,Eugene ORのヤギ抗マウスAlexa 488およびヤギ抗ウサギAlexa 568二次抗体など)が含まれる。Hoechst33258色素(Sigma)を使用して核を染色した。全てのデオキシリボザイムおよびオリゴデオキシヌクレオチドを、脱塩オリゴマーとしてSigma
Genosys,Australiaから購入した。TaqMan標識オリゴマーを、Applied Biosystems,USAから購入した。全てのオリゴデオキシヌクレオチドを、未修飾ホスホジエステルオリゴデオキシヌクレオチドとして購入した。
デオキシリボザイムのデザインおよび教示は、以前の研究で記載されているものに類似していた(Santoro,S.W.and Joyce,G.F.Proc.NatlAcad.Sci.USA,(1997),94,4262−4266 and Cairns,M.J.et al.,(1999)Nat.Biotech 17,480−486)。簡潔に述べれば、標的コネキシンのmRNA配列中の全てのAUおよびGU部位を、AまたはGの各端上の8または9ヌクレオチドを使用して選択した。デオキシリボザイムは、「A」または「G」が「10〜23」触媒コア「ggctagctacaacga」に置換されたセンスコード配列の相補物である。コントロールデオキシリボザイムは、1つの点変異(g→A)を有する欠損触媒コア「ggctaActacaacga」を有していた(S.W.and Joyce,G.F.Biochem 37,13330−13342)。本発明者らはまた、以下の3つの要件を満たさないAUおよびGUデオキシリボザイムによって残存するギャップを対象とするためのGCおよびAC特異的デオキシリボザイムをデザインした。各デオキシリボザイムを、ATG開始コドンからの「A」または「G」の位置にしたがって命名した。インビトロアッセイのために選択したデオキシリボザイムは、以下の3つの要件を満たさなければならなかった。
マウスコネキシン43およびコネキシン26のcDNAを、NcoIおよびNheIを使用してpORFベクター(インビボgen)から切り出し、インビトロ転写の前にpGEM−T(Promega)にサブクローン化した。全長2.4kbラットコネキシン43cDNAおよび200ヌクレオチドの5’非翻訳領域を含む全長コード1.4kbラットコネキシン43cDNAを、インビトロ転写のために使用した。全長mRNAを、Promega Riboprobeキットを使用して、線状化プラスミドDNAから転写した。得られたmRNAを、PCRスピンカラム(Qiagen)を使用して精製した。濃度を、260nmのODの分光光度計読み取りによって決定した。次いで、デオキシリボザイム(最終濃度40μM)およびmRNA(0.01〜0.05μg/μl総mRNA)を、2×切断緩衝液(100mM Tris 7.5;20 mM MgCl2;300 mM NaCl;0.02% SDS)を使用して37℃5〜10分間個別に予め平衡化した。次いで、mRNAおよびデオキシリボザイムとの混合物を、37℃で1時間インキュベートし、その後、10×Bluejuice(インビトロgen)を添加して切断反応を停止させ、混合物を氷上に保持した。次いで、反応混合物を、1×TBE緩衝液および7M尿素を含む予備泳動4%ポリアクリルアミドゲル(19:1アクリル:ビス比、BioRad)にロードし、2時間まで泳動した。ゲルを、10000倍希釈のSYBR green II(Mol Probes,USA)を含むTBE bufferで染色し、BioRad Chemi Doc systemを使用して画像化した。
インビトロで首尾よくmRNAを切断するデオキシリボザイム結合アームの20ヌクレオチド配列に基づいて、アンチセンス配列を選択した。選択した配列を、30量体のオリゴのデザインで使用するために選択した(Brysch,W.(1999).Antisense Technology in the Ventral Nervous System,ed.H.A.Robertson;Oxford University
Press 21− 41)および(Walton S.,et al.,(2002)Biophysical Journal 82,366−377)。簡単に述べれば、無関係の遺伝子(GGGGおよびCpGモチーフ)との8〜10CG対合の部分的配列相補性などの配列に関連する悪影響を回避した。できれば誤プライミングを防止するために、最後の5ヌクレオチド中にチミジンまたは3つを超えるCまたはGを有する3’末端を有するアンチセンスも回避する。ホモ二量体、回文モチーフ、または二次ヘアピン構造などの安定な二次構造を形成するオリゴマーは、標的mRNAに結合するオリゴマーを妨害する。交差ハイブリッド形成、非特異的タンパク質結合、および毒性に起因する化学的に関連する悪影響の可能性を評価するために、コントロールオリゴマー(センス、スクランブル、逆、およびミスマッチオリゴマーが含まれる)もデザインした。

製造者のプロトコールにしたがってTRIzol試薬(GIBCO,インビトロgen,USA)を使用して、単離ラット角膜から総RNAを抽出した。RNAサンプルの質を、臭化エチジウム染色アガロースゲルでの電気泳動によって評価し、18Sおよび28S rRNAバンドをUV照明下で視覚化した。抽出収率を、260nmでの分光光度法で定量した。リアルタイムPCRのために、cDNAを、20μlの最終反応体積でのオリゴdTおよびsuperscript II RnアーゼH逆転写酵素(Life Technologies,インビトロgen,USA)の使用によって5μgの総RNAから調製した。製造者の説明書に従ってTaqMan Universal PCR Master Mix(Applied Biosystems,USA)を使用して、50μlの体積の各サンプルについて100ngの総RNAのcDNA等価物を使用した96ウェル光学反応プレート中で定量的PCR反応を行った。ABI PRISM 7700配列検出システム装置(Applied Biosystems,USA)にてPCRを実施した。サーマルサイクリング条件は、製造者の標準的プロトコールを使用して、95℃で10分間の最初の変性、50サイクルの2工程PCR(95℃で15秒間の変性、60℃で1分間のアニーリング−伸長が含まれる)を含んでいた。全実験を、3連で繰り返した。各標的についてのネガティブコントロールのモニタリングでは、キャリーオーバーは認められなかった。
Cy3およびTaqMan(Fam,Tamra)標識オリゴマーを使用して、透過および安定性を評価した。Cy3標識オリゴマー(Sigma Genosys)およびTaqMan(FAM,TAMRA)標識オリゴマー(Applied Biosystems)を、プルロニックゲルと共に適用して、オリゴマーの安定性および角膜上皮への透過の両方を測定した。処置した角膜を、4%パラホルムアルデヒド中で20分間固定し、1%寒天にマウントし、40×浸水レンズ下でホールマウントとして観察した。オリゴマー透過の深さを、LeicaSP2共焦点顕微鏡のZ−スキャンオプションおよび測定したz距離に対する強度のプロットを使用して測定した。TaqManオリゴマーの破壊を、共焦点のLamdbaスキャンオプションを使用して測定した。FAM(ドナー)とTAMRA(受容体)分子との間の蛍光共鳴エネルギー転移(またはFRET)は、インタクトな30量体オリゴマー中で起こる。オリゴマーが破壊される場合、FAMおよびTAMRAはもはや極めて接近せず、FRETはもはや生じない。
前に記載のように、角膜切片に対するコネキシンの免疫標識を行った。簡潔に述べれば、切片を、10%ヤギ血清でブロッキングし、250倍希釈(マウス抗ラットコネキシン43)または500倍希釈(ウサギ抗ラットコネキシン26)の一次抗体と4℃で一晩インキュベートした。次いで、切片をPBSで洗浄し、400倍希釈のAlexa488標識二次抗体と室温で2時間インキュベートし、その後、4%パラホルムアルデヒドで固定し、0.2μMヨウ化プロピジウムまたは50倍希釈のHoechst 332358で10分間対比染色を行った。切片を、Citifluro antifade倍地(Agarscientific UK)にマウントした。Leica TCS−4DまたはLeica SP2共焦点レーザースキャニング顕微鏡を使用して全画像を収集し、TIFファイルとして保存した。全画像を、一定の電圧(520〜540V)およびオフセット(−2)設定を使用して収集した。コントロール組織の画像のグレースケールを最大にするようにグロー−垂直二連式ディスプレイ(glow−over−under display)オプションを使用して、電圧およびオフセットを設定した。次いで、、同一実験内の全サンプルについて、同一の設定を使用した。
げっ歯類コネキシン43mRNAに対して特異的に66種のデオキシリボザイムをデザインした(表5)。マウスおよびラットコネキシン43mRNAの両方を認識するために、22種のデオキシリボザイムをデザインした。本発明者らはまた、ネガティブコントロールとして、「10−23」触媒コア中に1つの点変異を有する2つの欠損デオキシリボザイムを購入した。デオキシリボザイム切断結果は、ラットコネキシン43mRNA(1.1Kb長)(示さず)およびラットコネキシン43mRNA(2.4Kb長)(図11A)と類似していた。両ラット(図11A)およびマウスk(図11B)では、コネキシン43mRNAは、デオキシリボザイムに接近可能な類似の領域を有するようである。結果は、曝露されてデオキシリボザイム切断に利用可能なげっ歯類コネキシン43mRNA上の4つの領域を示す。これらの領域は、ATG開始コドンからおよそ367〜466塩基、526〜622塩基、783〜885塩基、および1007〜1076塩基に位置する。2つの欠損デオキシリボザイム(df605およびa1df783)は、げっ歯類コネキシン43mRNAの切断は認められなかった。ラットコネキシン43mRNAの非翻訳領域の200塩基対に対してデザインされたデオキシリボザイムも、いかなる切断活性も示さなかった。
ラット角膜は、臓器培養におけるコネキシン43およびコネキシン26の発現を維持し、30%プルロニックF−127ゲルによるアンチセンスオリゴマーの送達に容易に利用可能である。したがって、ラット角膜臓器培養を、インビトロモデル由来のアンチセンスオリゴデオキシヌクレオチドデザインの有効性を試験するためのモデル系として選択した。本発明者らは、ラット角膜を24時間培養し、内皮がインタクトなままであることを見出した。しかし、48時間を超える培養時間は、角膜の不透明度に影響をあたえるようであり、それにより、使用しなかった。Cy3標識オリゴマーを使用して、培養角膜へのオリゴマーの透過度を決定した。インタクトな角膜全体の共焦点光学スライドは、蛍光シグナルがCY3標識オリゴマーと共に存在することを示し、図13Aは、最初の処置から1時間後の培養角膜の蛍光シグナルの深さが10μmであることを示す。オリゴデオキシヌクレオチドに抱合したTaqManプローブを使用して、30%プルロニックゲルを含むインタクトなオリゴデオキシヌクレオチドの角膜上皮への送達を測定し、照明した。有意なオリゴマーの比率は、治療1時間後にインタクトなままであった(図13B、13C)。インタクトなオリゴマーの点状シグナル(図13Cで起こるFRET)は、赤色(グレースケールで示す)の波長として認められる一方で、分解オリゴマー由来のシグナル(FRETなし)は、緑色(グレースケールで示す)の発光スペクトルで出現する(図13B)。
予備実験では、本発明者らは、本発明者らのポジティブコントロール(DB1)を1回適用したラット角膜を試験し、8時間後にコネキシン43タンパク質発現の有意な変化は認められなかった。8時間間隔で3回適用した24時間後に明らかなタンパク質ノックダウンが認められた。デオキシリボザイム切断アッセイ由来の結果に一部基づいて、本発明者らは、インビボで一定のアンチセンスオリゴマー(DB1、r43as605、r43as783、r43as885、r43as953、およびr43aslO76)、非機能的と予想されるアンチセンスオリゴマー(r43asl4、r43as769、およびr43as892)、およびネガティブコントロール(DB1センス)を試験した。本発明者らは、コントロールと比較した治療から24時間後のコネキシン43タンパク質レベルのノックアウトを見出し(図14A)、決定された全アンチセンスオリゴマーが陽性のはずである(図14C、14E、14G)。陰性と予想される3つ全てのオリゴマーおよびネガティブコントロールオリゴマーは、コネキシン43発現に影響を与えなかった。(図14B、14D、4F、4H)。DB1(as885の30量体)は、より短いas885と類似のノックダウン率を示した(ほぼ50%ノックダウン)本実験で同定されたより良好なアンチセンスオリゴマーの1つは、タンパク質レベルを64%減少させるas605のようであった。これらの定量結果のまとめを、図15に示す。
リアルタイムPCRを使用して、mRNAレベルに対するアンチセンスオリゴデオキシヌクレオチドの効果を決定した。コネキシン43タンパク質発現をノックダウンするアンチセンスオリゴデオキシヌクレオチド(as605、as885、DB1)はまた、処置後8時間以内にコントロール角膜と比較してコネキシン43mRNAレベルが低いことが確認された。コネキシン43mRNAの相対レベルの減少率は、コネキシン43タンパク質の減少レベルと十分に相関した。ネガティブアンチセンスオリゴマー(as769)およびネガティブコントロール(DB1センス、ゲルのみ)は、コントロール角膜と比較してコネキシン43mRNAレベルが変化しなかった。
