Description
Qa/QA+Qb/QB=SIであり、
ここで、
QA=化合物Aの濃度(ppm)、単独作用がエンドポイントを生成し、またはエンドポイントが確立できなかった場合、試験した最高濃度をエンドポイントとして計算に使用し、SIは値の「未満または<」で記録する。
Qa=混合物中の化合物Aの濃度(ppm)であり、エンドポイントを生成した。
QB=化合物Bの濃度(ppm)、単独作用がエンドポイントを生成し、またはエンドポイントが確立できなかった場合、試験した最高濃度をエンドポイントとして計算に使用し、SIは値の「未満または<」で記録する。
Qb=混合物中の化合物Bの濃度(ppm)であり、エンドポイントを生成した。
第二の殺生物剤とのMBIT相乗作用を、微生物の増殖を抑制するのに必要とされる最小殺生物剤または殺生物剤混合濃度を評価することによって決定した。96ウェルのマイクロタイタープレートフォーマットを使用して、全試験を実施した。全試験について、MBIT単独、第二の殺生物剤単独、両方の殺生物剤化学物質の組み合わせの様々な濃度を含む200μlの微生物増殖培地を、マイクロタイタープレートの個別のウェルに加えた。特に、細菌(E.coli、ATCC#8739)にはトリプチックソイブロス(TSB)を、イースト(カンジダ・アルビカンス、ATCC#10231)にはイースト麦芽エキスブロス(YMB)を、およびカビ(クロコウジカビ、ATCC#16404)にはポテトデキストロースブロス(PDB)を使用した。最終濃度が104CFU/mLまたは104胞子/mLの試験微生物を、並行実験の各ウェルに適用し、MIC評価を開始した。殺生物剤を含まない増殖培地を、各実験装置の対照として使用し、各微生物の増殖生存可能性を確認した。これらの殺生物剤濃度の64個の可能な組み合わせの他に、個別の殺生物剤それぞれの8つの濃度(2倍希釈)を微生物増殖阻害試験で評価した。個別の殺生物剤の濃度の評価には、相乗作用インデックスを計算するために、抑制濃度のエンドポイントを達成することを必要とした。有機体を加えた後に、96ウェルのマイクロタイタープレートを、25℃で48時間、または殺生物剤が含まれていない対照ウェル内に増殖が観察されるまでインキュベートした。目に見える有機体増殖による濁度に基づき、個々のウェルに、増殖ありまたは増殖なしとして、スコアを付けた。微生物の増殖を阻害した組み合わせられた殺生物剤の濃度の他に、MBITと第二の殺生物剤の両方について、有機体が増殖しなかった単一の活性殺生物剤の最低濃度を、相乗作用インデックスのために記録した。相乗作用を表す2つの殺生物剤の比を表1〜6に示す。
MIC評価では特定の殺生物剤化学物質を評価することができないため、MKC評価を使用し、共殺生物剤DXN(6−アセトキシ−2、4−ジメチル−m−ジオキサン)およびTHNM(2−ヒドロキシメチル−2−ニトロ−1、3−プロパンジオール)とMBITの相乗作用を特定した。前述の相乗的な組み合わせを評価するために以下の試験方法を設計した。結果は表7および表8で見ることができる。
96ウェルのマイクロタイタープレートの各ウェルに、2.0%のTSBを補充した無菌水を適用することによって、試験を開始した(第0日目)。様々な濃度のMBIT単独、共殺生物剤単独、または両方の殺生物剤化学物質の組み合わせをマイクロタイタープレートの個別のウェルに加え、次に、最終濃度が5x104CFU/mLの大腸菌(ATCC#8739)を加えた。試験の第7、14および21日目に等量の播種を行った。細菌の殺傷の程度を確認するために、第1、7、8、14、15、21、22日目に各試料の一定分量(20μl)を取り出し、トリプチックソイブロス(180μl)に移した。30℃でインキュベーションしてから48時間後、TSB内の濁度の存在または喪失によって、細菌殺傷(最大検出限界は5x101CFU/mL)を視覚的に確認した。

96ウェルのマイクロタイタープレートの各ウェルに、2.0%のポテトデキストロースブロスで補充した無菌水を適用することによって、試験を開始した(第0日目)。様々な濃度のMBIT単独、共殺生物剤単独、または両方の殺生物剤化学物質の組み合わせをマイクロタイタープレートの個別のウェルに加え、次に、最終濃度が5x104胞子/mLのクロコウジカビ(ATCC#16404)を加えた。試験の第7および14日目に等量の播種を行った。カビ殺傷の程度を確認するために、第1、7、8、14、15、21日目に各試料の一定分量(20μl)を取り出し、ポテトデキストロースブロス(180μl)に移した。30℃でインキュベーションしてから48〜96時間後、PDB内の濁度の存在または喪失によって、カビ殺傷(最大検出限界は5x101胞子/mL)を視覚的に確認した。
Claims (8)
- 以下を含む殺菌性組成物
(a)N−メチル−1,2−ベンゾイソチアゾリン−3−オンおよび
(b)ドデシルグアニジン、ポリ(ヘキサメチレンビグアニド塩酸塩)、ナトリウムピリチオン、オルト−フェニルフェン酸ナトリウム、テルブトリン、ジメトキサンおよび2−(ヒドロキシメチル)−2−ニトロプロパン−1,3−ジオールから成る群から選択される少なくとも1つの殺菌剤。 - 少なくとも1つの前記殺菌剤は、ドデシルグアニジンであり、N−メチル−1,2−ベンゾイソチアゾリン−3−オンとドデシルグアニジンの比が1:1〜1:650である、請求項1に記載の殺菌性組成物。
- 少なくとも1つの前記殺菌剤は、ポリ(ヘキサメチレンビグアニド塩酸塩)であり、N−メチル−1,2−ベンゾイソチアゾリン−3−オンとポリ(ヘキサメチレンビグアニド塩酸塩)の比が4:1〜2:1または1:533〜1:1067である、請求項1に記載の殺菌性組成物。
- 少なくとも1つの前記殺菌剤は、ナトリウムピリチオンであり、N−メチル−1,2−ベンゾイソチアゾリン−3−オンとナトリウムピリチオンの比が21:1〜1:40である、請求項1に記載の殺菌性組成物。
- 少なくとも1つの前記殺菌剤は、オルト−フェニルフェン酸ナトリウムであり、N−メチル−1,2−ベンゾイソチアゾリン−3−オンとオルト−フェニルフェン酸ナトリウムの比が1:60〜1:135である、請求項1に記載の殺菌性組成物。
- 少なくとも1つの前記殺菌剤は、テルブトリンであり、N−メチル−1,2−ベンゾイソチアゾリン−3−オンとテルブトリンの比が1:100〜1:110,000である、請求項1に記載の殺菌性組成物。
- 少なくとも1つの前記殺菌剤は、ジメトキサンであり、N−メチル−1,2−ベンゾイソチアゾリン−3−オンとジメトキサンの比が1:1〜1:650である、請求項1に記載の殺菌性組成物。
- 少なくとも1つの前記殺菌剤は、2−(ヒドロキシメチル)−2−ニトロプロパン−1,3−ジオールであり、N−メチル−1,2−ベンゾイソチアゾリン−3−オンと2−(ヒドロキシメチル)−2−ニトロプロパン−1,3−ジオールの比が3:1〜1:5である、請求項1に記載の殺菌性組成物。
