Abstract
Description
本発明の第1形態において、アンチセンス鎖は、RISC(RNA-induced silencing complex)で標的mRNAと相補的に結合して標的mRNAを分解するRNAi活性を示す鎖であり、センス鎖は、アンチセンス鎖と相補的な配列を有する鎖を意味する。
本発明による第2形態の課題を果たすための本発明は下記構造のASO−高分子接合体を提供する:
リガンドが結合した二本鎖オリゴRNA構造体を製造するために、二本鎖オリゴRNA構造体に結合可能なリガンド物質を製造した。
N−アセチルガラクトサミン(N-Acetyl Galactosamine、NAG)を二本鎖オリゴRNA構造体に結合するために、下記反応式Iに示すように、1−ヘキサ(エチレングリコール)−N−アセチルガラクトサミン−ホスホラミダイト(1-Hexa(Ethylene Glycol)-NAG-Phosphoramidite)を製造した。
出発物質の塩酸ガラクトサミン(SIGMA Aldrich社製、米国)(2g、9.27mmol)、アセトニトリル(Acetonitrile、SAMCHUN社製、韓国)(31mL)、トリエチルアミン(SIGMA Aldrich社製、米国)(15.42mL、111.24mmol)を混合して1時間還流する。徐々に常温に冷却し、冷却水を用いて0℃に冷却した後、無水酢酸(SIGMA Aldrich社製、米国)(8.76mL、92.70mmol)を10分間滴加した。滴加が完了した後、冷却水を除去し、常温で24時間攪拌した。反応が完了すると、重炭酸ナトリウム(SAMCHUN社製、韓国)水溶液にpHが中性になるまで徐々に投入した。pHが中性になってから常温で2時間攪拌して生成された固体を濾過し、濾過物をエチルアセテート(SAMCHUN社製、韓国)(100mL×2)、蒸留水(100mL×2)、エチルアセテート(100mL×1)の順に洗浄した。固体を真空乾燥して、1,3,4,6−テトラアセチル−N−アセチルガラクトサミン(1.82g、52.3%)を得た(図3参照)。
前記実施例1−1−1で得られた1,3,4,6−テトラアセチル−N−アセチルガラクトサミン(1.81g、4.82mmol)、塩化鉄(III)(SIGMA Aldrich社製、米国)(1.02g、6.27mmol)、メチレンクロライド(SAMCHUN社製、韓国)(48mL)を混合して常温で攪拌した。10分間攪拌した後、ヘキサ(エチレングリコール)(SIGMA Aldrich社製、米国)(1.58mL、4.82mmol)を投入して2時間還流した。反応が完了すると、セライト(SIGMA Aldrich社製、米国)を使用して濾過し、濾過物をメチレンクロライド(50mL×2)で洗浄した。濾液を減圧濃縮し、エチルアセテート(100mL)と蒸留水(100mL)を投入して水層を抽出した。取得した水層をメチレンクロライド(100mL×3)で抽出して有機層を集めて無水硫酸マグネシウム(SAMCHUN社製、韓国)で乾燥して濾過した。濾液を減圧濃縮し、真空乾燥して、3,4,6−トリアセチル−1−ヘキサ(エチレングリコール)−N−アセチルガラクトサミン(2.24g、74.9%)を得た(図4参照)。
前記実施例1−1−2で得られた(2.22g、3.71mmol)、メチレンクロライド(37mL)、トリエチルアミン(0.94mL、6.75mmol)を混合して常温で攪拌した。10分間攪拌した後、2−シアノエチルN,N−ジイソプロピルクロロホスホラミダイト(2-Cyanoethyl N,N-diisopropylchlorophosphoramidite、SIGMA Aldrich社製、米国)(0.75mL、3.38mmol)を投入して45分間攪拌した。反応が完了すると、減圧濃縮し、エチルアセテート(100mL)と蒸留水(100mL)を投入して有機層を抽出した。取得した有機層を無数硫酸マグネシウムで乾燥し、濾過した。濾液を減圧濃縮し、カラムで分離して、1−ヘキサ(エチレングリコール)−N−アセチルガラクトサミン−ホスホラミダイト(1.14g、42.2%)を得た(図5参照)。
葉酸を二本鎖オリゴRNA構造体に結合するために、下記反応式IIに示すように、NHS−葉酸を製造した。
ペプチド化合物がα−アミノ酸を含んでいるため、これと同様な構造を有しているペプチド誘導体とPEGに存在するアミン官能基との結合反応を行った。この結合反応の前提條件としてPEGとペプチド誘導体に存在するアミン官能基は、ペプチドに存在するカルボン酸との結合反応の際に交互競争反応を行うため、ペプチド化合物に存在するアミン官能基を保護基で置換する先行過程が必要であった。ペプチド化合物に存在するアミン基は、9−フルオレニルメチルオキシカルボニル(9-fluorenylmethyloxycarbonyl;Fmoc)またはt−ブチルオキシカルボニル(tert-Butyloxycarbonyl;t−BOC)の保護基で置換されてペプチドのカルボン酸との反応性を除去してPEGに結合可能な形態のペプチド化合物を製造した。
前記実施例1で製造された結合可能なリガンド物質は、実施例1−1のNAGホスホラミダイトのようにホスホラミダイト形態に構成されて、通常の化学的オリゴ合成過程(遮断除去(deblocking)、結合(coupling)、キャッピング(capping)および酸化(oxidation)からなる過程)によりPEG末端に結合するか、実施例1−2のNHS−葉酸のようにNHS−リガンド形態に構成されて、PEGにアミン基を結合した後、アミンPEGとエステル反応により5’末端にリガンドが結合したPEG−RNAの形態に結合することができる。合成された5’末端にリガンドが結合したPEG−RNA構造体は、相補的な配列の疎水性残基が結合した形態である5’C24−RNA構造体とアニーリングして、5’末端にリガンドが結合した二本鎖オリゴRNA構造体を合成した。
以下、実施例では、サバイビンを抑制するためにサバイビン二本鎖オリゴRNAを使用した。サバイビンは、今までテストされたほとんどの新生腫瘍や形質転換された細胞株において共通して発現するタンパク質であって、抗ガン治療において重要な標的になると予測されている(Survivin; a new target for anti-cancer therapy. Cancer Treat Rev. 2009 Nov; 35(7); 553-62)。本発明のサバイビン二本鎖オリゴRNAは、配列番号1と記載されるセンス鎖とこれに相補的な配列であるアンチセンス鎖からなっており、対照群として使用される二本鎖オリゴRNAは、配列番号2と記載されるセンス鎖とこれに相補的な配列であるアンチセンス鎖からなっている。本実施例に使用される二本鎖オリゴRNAは、下記の塩基配列からなっている。

実施例2−1で合成されたRNAにPEGホスホラミダイト試薬を処理して、通常のRNA合成過程により5’PEG−RNA構造体を製造した(図7参照)。
実施例2−3−1:N−アセチルガラクトサミンホスホラミダイトを用いたNAG−PEG−RNA構造体の製造
実施例2−2で合成された5’PEG−RNA構造体に実施例1−1で合成されたN−アセチルガラクトサミンホスホラミダイト試薬を用いて、通常のRNA合成過程により、N−アセチルガラクトサミンをPEG−RNA構造体にホスホジエステル結合した。N−アセチルガラクトサミンリガンドは、デンドリマリンカー(dendrimer linker)を活用して、PEGに単分子あるいはそれ以上の分子のN−アセチルガラクトサミンを結合することができる。
実施例2−2で合成されたPEG−RNA構造体に、実施例1−1で合成されたN−アセチルガラクトサミンホスホラミダイト試薬を用いて、通常のRNA合成過程により、N−アセチルガラクトサミンをPEG−RNA構造体にホスホジエステル結合して、5’モノ−NAG−PEG−RNA構造体を合成した(図8参照)。
実施例2−2で合成されたPEG−RNA構造体に、デンドリマホスホラミダイト(Dendrimer Phosphoramdite)(Trebler Phosphoramidte、Glen research社製、米国)試薬で合成した後、実施例1−1で合成されたNAGホスホラミダイト試薬を用いて、通常のRNA合成過程により、3個のNAGをPEG−RNA構造体にホスホジエステル結合で5’トリ−NAG−PEG−RNA構造体を合成した(図9参照)。
実施例2−2で合成されたPEG−RNA構造体に、アミンホスホラミダイト試薬を用いて、通常のRNA合成過程により、アミン基をPEG−RNA構造体にホスホジエステル結合でアミン−PEG−RNA構造体を合成した。合成されたアミン−PEG−RNA構造体に、実施例1−2で合成されたNHS−葉酸をエステル反応により5’葉酸−PEG−RNA構造体を合成した(図10参照)。
実施例1−3で製造された保護されたペプチド化合物と実施例2−3−2のアミン−PEG−RNA構造体のPEGのアミン基とペプチドのカルボキシル基の結合反応は、BOP(Benzotriazole-1-yl-oxy-tris-(dimethylamino)-phosphonium hexafluorophosphate、SIGMA Aldrich社製、米国)とHOBT(1-Hydroxybenzotriazole、SIGMA Aldrich社製、米国)の条件下で行われた。前記結合反応が行われた後、保護基を除去するために、ピペリジン(SIGMA Aldrich社製、米国)処理を施して、5’ペプチド−PEG−二本鎖オリゴRNA構造体を製造した。
実施例2−3の5’リガンドが結合したPEG−二本鎖オリゴRNA構造体配列と相補的な配列のRNA構造体は、実施例2−1のRNA合成方法により合成した後、ジスルフィド結合が含まれている炭素数24個のテトラドコサン試薬を用いて、通常のRNA合成過程により、C24をRNA構造体にホスホジエステル結合で結合して、5’C24−RNA構造体を合成した(図11参照)。
実施例2−1,2−2,2−3および2−4で合成されたRNA一本鎖は、60℃の恒温槽で28%(v/v)のアンモニア処理を施して合成されたRNAおよびRNA構造体をCPGから分離した後、脱保護反応により保護残基を除去した。保護残基が除去された5’末端にリガンドが結合したPEG−二本鎖オリゴRNA構造体および5’C24−二本鎖オリゴRNA構造体は、70℃のオーブンでN−メチルピロリドン、トリエチルアミンおよびトリエチルアミントリヒドロフルオリド(triethylamine trihydrofluoride)を10:3:4の体積割合で処理して、2’TBDMS(tert-butyldimethylsilyl)を除去した。
アミン−CPGにPEGホスホラミダイト試薬を用いて3’アミン−PEG−RNAの形態に合成した後、実施例1−2のNHS−葉酸のようにNHS−リガンド形態からなる結合可能なリガンド物質とエステル反応により3’末端にリガンドが結合したPEG−二本鎖オリゴRNA構造体を合成することができる。合成された3’末端にリガンドが結合したPEG−二本鎖オリゴRNA構造体は、疎水性物質であるC24が結合して3’末端にリガンドが結合したPEG−RNA−C24を形成するようになり、これは、相補的な配列のRNAとアニーリングされて3’末端にリガンドが結合した二本鎖オリゴRNA構造体に合成される。
アミン−CPGにポリエチレングリコールホスホラミダイト試薬を用いて、通常のRNA合成過程により、3’CPG−アミン−PEGに合成された。3’CPG−アミン−PEGは、実施例2−1のRNA合成過程により、所望の配列のRNAを有する3’CPG−アミン−PEG−RNA構造体に合成された(図12参照)。
実施例3−1で合成された3’CPG−アミン−PEG−RNA構造体は、ジスルフィド結合が含まれている炭素数24個のテトラドコサン試薬を用いて、通常のRNA合成過程により、C24をRNAにホスホジエステル結合して構造体を合成した(図13参照)。
実施例3−2で合成された3’アミン−PEG−RNA−C24構造体は、60℃の恒温槽で28%のアンモニア処理を施して合成された3’アミン−PEG−二本鎖オリゴRNA構造体および3’アミン−PEG−RNA−C24構造体をCPGから分離した後、脱保護反応により保護残基を除去した。保護残基が除去された3’アミン−PEG−RNA−C24構造体は、70℃のオーブンでN−メチルピロリドン、トリエチルアミンおよびトリエチルアミントリヒドロフルオリドを10:3:4の体積割合で処理して、2’TBDMS(tert-butyldimethylsilyl)を除去した。分離した3’アミン−PEG−RNA−C24構造体は、NHS−リガンド形態からなる結合可能なリガンド物質とエステル反応により、3’末端にリガンドが結合したPEG−RNA−C24構造体に合成された。
実施例3−2で合成された3’アミン−PEG−RNA−C24構造体は、実施例1−2で合成されたNHS−葉酸とエステル反応により、3’葉酸−PEG−RNA−C24構造体を合成した(図14参照)。
実施例3−3の3’末端にリガンドが結合したPEG−RNA−C24配列と相補的な配列のRNA一本鎖は、実施例2−1のRNA合成方法で合成した。合成されたRNA一本鎖は、60℃の恒温槽で28%のアンモニア処理を施して合成されたRNAをCPGから分離した後、脱保護反応により保護残基を除去した。保護残基の除去されたRNAは、70℃のオーブンでN−メチルピロリドン、トリエチルアミンおよびトリエチルアミントリヒドロフルオリドを10:3:4の体積割合で処理して、2’TBDMSを除去した。
前記反応物を高速液体クロマトグラフィー(LC−20A Prominence、SHIMADZU社製、日本)によりRNAおよび3’リガンドが結合したPEG RNA−C24構造体を分離して精製し、これをMALDI−TOF質量分析装置(MALDI TOF−MS、SHIMADZU社製、日本)で分子量を測定して、合成しようとする塩基配列および3’末端にリガンドが結合したPEG RNA−C24構造体と対応するかを確認した。
実施例2および3で合成された5’末端にリガンドが結合した二本鎖オリゴRNA構造体および3’末端にリガンドが結合した二本鎖オリゴRNA構造体は、二本鎖オリゴRNAの末端に結合した疎水性物質間の疎水性相互作用によりリガンドが結合した二本鎖オリゴRNA構造体からなるナノ粒子、すなわちミセルを形成する(図1参照)。実施例2で合成された5’葉酸−リガンド−二本鎖オリゴRNA構造体からなるナノ粒子の大きさおよび臨界ミセル濃度、透過型電子顕微鏡(Transmission Electron Microscope、TEM)分析によりナノ粒子の形成を確認した。
ゼータ電位測定装置により前記ナノ粒子の大きさを測定した。具体的に、前記5’葉酸−二本鎖オリゴRNA構造体を1.5mLのDPBS(Dulbecco’s Phosphate buffered saline)に50μg/mLの濃度で溶かした後、超音波分散機(Wiseclean、DAIHAN社製、韓国)でナノ粒子の大きさを均質化(700W;振幅20%)した。均質化したナノ粒子は、ゼータ電位測定装置(Nano−ZS、MALVERN社製、英国)で大きさを測定したが、物質に対する屈折率は1.459、吸収率は0.001とし、溶媒であるPBSの温度25℃およびそれによる粘度は1.0200および屈折率は1.335の値を入力して測定した。1回の測定は、大きさを20回繰り返して測定することで行われ、これを3回繰り返した。
単分子に親油基と親水基がともに存在している両親媒性物質は、界面活性剤になることができるが、界面活性剤は、水溶液に溶解されると、疎水性物質は水との接触を避けるために中央に集まり、親水性物質は外側に向かってミセルを形成する。この際、最初にミセルを形成する濃度を臨界ミセル濃度と言う。蛍光染料を用いてCMCを測定する方法は、ミセルが形成される前/後に蛍光染料の蛍光値グラフ曲線の傾斜が急激に変化する特性に基づくものである。
葉酸−二本鎖オリゴRNA構造体からなるナノ粒子が形成された形態を確認するために、透過型電子顕微鏡により観察した。
実施例2で合成された5’葉酸−二本鎖オリゴRNA構造体からなるナノ粒子(葉酸−SAMiRNA)の生体外条件下での改善した二本鎖オリゴRNAの効能を確認するために、葉酸受容体が過剰発現する細胞株であるKB細胞株を葉酸が存在する条件であるか否かに応じて、トランスフェクション物質が存在しない状態で処理して結合したリガンドによる改善したSAMiRNAの伝達効率および標的遺伝子の発現阻害効果を確認した。
米国合衆国培養細胞系統保存機関(American type Culture Collection、ATCC)から手に入れたヒト口腔上皮ガン細胞(KB)株は、葉酸を含まないRPMI−1640培養培地(Gibco、米国)に、10%(v/v)のウシ胎児血清、ペニシリン100units/mL、ストレプトマイシン100μg/mLを添加して、37℃、5%(v/v)CO2の条件下で培養した。
前記実施例5−1で培養された腫瘍細胞株(1.3×105/ウェル)を前記実施例の5−1条件下で6−ウェルプレートで葉酸を含まないRPMI−1640培養培地で18時間培養してから培地を除去した後、各ウェル当たり同量のOpti−MEM(商標)培地を分注した。
前記実施例5−2でトランスフェクションされた細胞株から全体RNAを抽出してcDNAを合成した後、リアルタイムPCRによりサバイビンのmRNA発現量を韓国公開特許第2009−0042297号公報の方法にしたがって相対定量した。
実施例2で合成された5’葉酸−二本鎖オリゴRNA構造体からなるナノ粒子(葉酸−SAMiRNA)の生体内条件下での二本鎖オリゴRNAの効能を確認するために、葉酸受容体が過剰発現する細胞株であるKB細胞株からなる腫瘍を有するマウスに投与し、伝達された葉酸−SAMiRNAおよびSAMiRNAの腫瘍組織内における標的遺伝子の発現阻害効果を確認した。
実施例5−1で培養されたKB細胞株を1×106ずつ5週齢のヌードマウス(BALB/C nu)の皮下組織に投与した。投与後、2日おきに腫瘍長軸および短縮の長さを測定して腫瘍の成長を観察し、投与後、2週経過した時点で、約150〜200mm3ほどに腫瘍が成長していることを確認した。
実施例6−1で準備したKB xenograft modelに実施例2で合成された配列番号1を含む5’葉酸−二本鎖オリゴRNA構造体とリガンドが結合していない二本鎖オリゴRNA構造体を実施例4−1と同じ方法で超音波分散機により二本鎖オリゴRNA構造体からなるナノ粒子を均質化した。均質化したナノ粒子は、5mg/kgの体重の量で尾静脈投与によりKB xenograft modelに1回投与し(n=4)、投与後、48時間または72時間が経過した時点で腫瘍組織を採取した。採取した腫瘍組織は、全体RNAを抽出してcDNAを合成した後、リアルタイムPCRによりサバイビンmRNAの発現量を韓国公開特許第2009−0042297号公報の方法にしたがって相対定量した(図17参照)。
本発明の実施例では、サバイビン(Survivin)を抑制するためにサバイビン(Survivin)ASOを使用した(Biol. Proced. Online 2004; 6(1); 250-256)。サバイビンは、今までテストされたほとんどの新生腫瘍や形質転換された細胞株で共通して発現されるタンパク質であって、抗ガン治療において重要な標的になると予測されている(Abbrosini G. et al. Nat. Med. 3(8); 917-921, 1997)。

本実施例で使用されたASOは、DNA骨格構造をなしているリン酸基がホスホチオエート基で置換されているS−オリゴを用いてホスホチオエート結合方法で連結されたものである。
前記実施例7および実施例8で合成、分離したASO−高分子接合体が結合していない最初の形態のASOに比べてASOの安定性が向上したか否かを確認した。高分子接合体が結合していないASOとASO−高分子接合体を生体内条件をまねした形態である30および50%(v/v)のFBSが添加された培地でそれぞれ0、3、5、7および10日間培養した後、最初のASOに比べて分解された程度を電気泳動法または重合酵素連鎖反応法(PCR)により検証した。
ASO−高分子接合体は、ASOの末端に付与された疎水性物質間の疎水性相互作用によってASO−高分子接合体からなるナノ粒子を形成する(図18参照)。ゼータ電位測定装置によりASO−高分子接合体からなるナノ粒子の臨界ミセル濃度および大きさを測定した。
実施例8で製造した配列番号3のASO−高分子接合体のナノ粒子の大きさをゼータ電位測定装置で測定した。前記ASO−高分子接合体50μgを1mLのDPBSに溶かした後、超音波分散機(Wiseclean、DAIHAN社製、韓国)でナノ粒子の大きさを均質化(700W;振幅20%)した。均質化したナノ粒子は、ゼータ電位測定装置(Nano−ZS、MALVERN社製、英国)で大きさを測定したが、物質に対する屈折率は1.454、吸収率は0.001とし、溶媒である水の温度25℃およびそれによる粘度および屈折率を入力して測定した。1回の測定は、大きさを20回繰り返して測定することで行われ、これを3回繰り返した。

実施例8で製造した何も接合されていないASOとASO−高分子接合体を用いて腫瘍細胞株であるヒト大腸ガン細胞株(SW480)をトランスフェクションさせ、前記トランスフェクションされた腫瘍細胞株のサバイビンの発現パターンを分析した。
米国合衆国培養細胞系統保存機関(American type Culture Collection、ATCC)から手に入れたヒト大腸ガン細胞株(SW480)は、生長培地(Leibovitz’s L−15培養培地、GIBCO/Invitorgen;米国)に10%(v/v)のウシ胎児血清、ペニシリン100units/mL、ストレプトマイシン100μg/mLを添加して、37℃、5%(v/v)の二酸化炭素(CO2)の条件下で培養した。
前記実施例8で製造した何も接合されていないASOとASO−高分子接合体を用いて腫瘍細胞株であるヒト大腸ガン細胞株(SW480)をトランスフェクションさせ、前記トランスフェクションされた腫瘍細胞株のサバイビンの発現パターンを分析した。
実施例11−2でトランスフェクションされた細胞株から全体RNAを抽出してcDNAを合成した後、リアルタイムPCRによりサバイビン遺伝子のmRNA量を韓国公開特許第2009−0042297号公報の方法にしたがって相対定量した(図21参照)。
実施例7および実施例8で製造したそれぞれのASO−高分子接合体を用いて腫瘍細胞株であるヒト大腸ガン細胞株(SW480)をトランスフェクションさせ、前記トランスフェクションされた腫瘍細胞株のサバイビンの発現パターンを分析した。
米国合衆国培養細胞系統保存機関(American type Culture Collection、ATCC)から手に入れたヒト大腸ガン細胞株(SW480)は、生長培地(Leibovitz’s L−15培養培地、GIBCO/Invitorgen;米国)に10%(v/v)のウシ胎児血清、ペニシリン100units/mL、ストレプトマイシン100μg/mLを添加し、37℃、5%(v/v)二酸化炭素(CO2)の条件下で培養した。
実施例12−1で培養された腫瘍細胞株の1つのウェル当たり1.3×105を前記実施例5−1条件下で6−ウェルプレートで18時間生長培地(Leibovitz’s L−15培養培地、GIBCO/Invitorgen;米国)で培養してから培地を除去した後、各ウェル当たり800μLのOpti−MEM培地を分注した。
実施例12−2でトランスフェクションされた細胞株から全体RNAを抽出してcDNAを合成した後、リアルタイムPCRによりサバイビン遺伝子のmRNA量を韓国公開特許第2009−0042297号公報の方法にしたがって相対定量した。

- 下記式(1)の構造を有するリガンドが結合した治療薬物−高分子構造体:
- 前記治療用薬物は、二本鎖オリゴRNAまたは、抗ガン剤であることを特徴とする請求項1に記載のリガンドが結合した治療薬物−高分子構造体。
- 前記リガンドは、標的細胞特異的に結合して受容体媒介エンドサイトーシスをする、標的特異的抗体、アプタマー、ペプチド及び受容体特異的化学物質からなる群より選択されることを特徴とする請求項1に記載のリガンドが結合した治療薬物−高分子構造体。
- 前記受容体特異的化学物質は、葉酸、N−アセチルガラクトサミン及びマンノースから選択されることを特徴とする請求項3に記載のリガンドが結合した治療薬物−高分子構造体。
- 前記二本鎖オリゴRNAは、19〜31個のヌクレオチドからなる請求項2に記載のリガンドが結合した治療薬物−高分子構造体。

- (1)官能基−親水性物質が結合した固形支持体をベースとし、RNA一本鎖を合成する段階と、
(2)前記官能基−親水性物質が結合したRNA一本鎖の5’末端に疎水性物質を共有結合する段階と、
(4)前記官能基−RNA−高分子構造体および別に合成した相補的な配列のRNA一本鎖を固形支持体から分離する段階と、

(6)前記リガンドが結合したRNA−高分子構造体と相補的な配列のRNA一本鎖をアニーリングして二本鎖を形成する段階と、を含むリガンドが結合した二本鎖オリゴRNA構造体の製造方法。 - (1)固形支持体をベースとし、RNA一本鎖を合成する段階と、
(2)前記RNA一本鎖の5’末端に親水性物質を共有結合する段階と、
(3)前記RNA一本鎖の親水性物質にリガンドを結合する段階と、
(4)固形支持体からリガンドが結合したRNA−親水性高分子構造体および別に合成した相補的な配列のRNA−疎水性高分子構造体の一本鎖を分離する段階と、
(5)前記リガンドが結合したRNA−親水性高分子構造体と相補的な配列のRNA疎水性高分子構造体をアニーリングして二本鎖を形成する段階と、
を含むリガンドが結合した二本鎖オリゴRNA構造体の製造方法において、
(1)〜(4)段階の間に、(1)段階のRNA一本鎖と相補的な配列からなるRNA一本鎖を合成した後、疎水性物質を共有結合して、疎水性物質が結合した形態のRNA−疎水性高分子構造体一本鎖を合成する段階をさらに含むことを特徴とする製造方法。 - (1)官能基が結合している固形支持体をベースとし、RNA一本鎖を合成する段階と、
(2)段階(1)で得られた物質に親水性物質を共有結合する段階と、
(3)段階(2)で得られた物質にリガンドを共有結合する段階と、
(4)段階(3)で得られた物質を固形支持体から分離する段階と、
(5)段階(4)で得られた物質に、3’末端に結合した官能基により疎水性物質を共有結合する段階と、
(6)段階(5)で得られた物質と相補的な配列のRNA一本鎖をアニーリングして二本鎖を形成する段階と、
を含むリガンドが結合した二本鎖オリゴRNA構造体の製造方法。 - 請求項1〜15のいずれか一項に記載のリガンドが結合した治療薬物−高分子構造体を含むナノ粒子。
- 請求項1〜15のいずれか一項に記載のリガンドが結合した治療薬物−高分子構造体を含む薬剤学的組成物
- 請求項19に記載のナノ粒子を含む薬剤学的組成物。
- 下記式(5)の構造を有するアンチセンスオリゴヌクレオチド(ASO)と高分子物質の接合体;
- 前記ASOは10〜50個のオリゴヌクレオチドからなることを特徴とする請求項22に記載の接合体。
- 前記オリゴヌクレオチドは一つ以上のヌクレオチド内の糖構造の2’炭素位置で−OH基が−CH3(メチル)、−OCH3、−NH2、−F(フッ素)、−O−2−メトキシエチル、−O−プロピル、−O−2−メチルチオエチル、−O−3−アミノプロピル、−O−3−ジメチルアミノプロピル、−O−N−メチルアセトアミドまたは−O−ジメチルアミドオキシエチルへの置換による修飾;ヌクレオチド内の糖構造内の酸素が硫黄で置換された修飾;ヌクレオチド間の結合のホスホロチオエートまたはボラノリン酸、メチルホスホネート結合への修飾が、一つ以上または二つ以上に組み合わせて修飾されるか、または、PNAまたはLNA形態へ修飾されることを特徴とする請求項23に記載の接合体。
- 前記親水性物質の分子量は200〜10,000である高分子化合物であることを特徴とする請求項22に記載の接合体。
- 前記親水性物質はポリエチレングリコール、ポリビニルピロリドン及びポリオキサゾリンからなる群より選択されることを特徴とする請求項25に記載の接合体。
- 前記疎水性物質の分子量は250〜1,000であることを特徴とする請求項22に記載の接合体。
- 前記疎水性物質はC12〜C50の炭化水素またはコレステロールであることを特徴とする請求項27に記載の接合体。
- 前記共有結合は非分解性結合または分解性結合であることを特徴とする請求項22に記載の接合体。
- 前記非分解性結合は、アミド結合またはリン酸結合であることを特徴とする請求項29に記載の接合体。
- 前記分解性結合は、ジスルフィド結合、酸分解性結合、エステル結合、無水物結合、生分解性結合及び酵素分解性結合からなる群より選択されることを特徴とする請求項29に記載の接合体。
- 請求項22〜31のいずれか一項に記載のASO―高分子接合体の親水性物質にリガンドが結合されることを特徴とするリガンド結合ASO−高分子接合体。
- (a)固形支持体に親水性物質を共有結合する段階と、
(b)前記親水性物質を含む固形支持体をベースとし、ASOを合成する段階と、
(c)前記固形支持体上のASOの5’末端に疎水性物質を共有結合する段階と、
(d)前記親水性物質と疎水性物質が結合したASO−高分子接合体を固形支持体から分離および精製する段階を含むASO−高分子接合体の製造方法。 - (a)官能基が付着している固形支持体をベースとし、ASOを合成する段階と、
(b)前記ASO5’末端に親水性物質を共有結合する段階と、
(c)前記親水性物質が結合したASO接合体を固形支持体から分離する段階と、
(d)前記固形支持体から分離したASO3’末端に疎水性物質を共有結合する段階と、を含むASO−高分子接合体の製造方法。 - (a)官能基が付着している固形支持体に親水性物質を結合する段階と、
(b)前記官能基−親水性物質が結合している固形支持体にASOを合成する段階と、
(c)前記ASO5’末端に疎水性物質を共有結合する段階と、
(d)前記親水性物質と疎水性物質が結合したASO−高分子接合体を固形支持体から分離する段階と、
(e)前記固形支持体から分離したASO−高分子接合体の親水性物質の末端にリガンドを結合する段階と、を含むASO−高分子接合体の製造方法。 - (a)官能基が付着している固形支持体をベースとし、ASOを合成する段階と、
(b)前記ASOの末端に親水性物質を共有結合する段階と、
(c)前記親水性物質が結合したASO接合体にリガンドを共有結合する段階と、
(d)前記官能基が付着したASO−親水性高分子−リガンド接合体を固形支持体から分離する段階と、
(e)前記固形支持体から分離したASOの3’末端に疎水性物質を共有結合する段階と、を含むASO−高分子接合体の製造方法。 - 請求項22〜31のいずれか一項に記載のASO−高分子接合体を含むナノ粒子。
- 請求項32に記載のリガンド結合ASO−高分子接合体を含むナノ粒子。
- 請求項22〜31のいずれか一項に記載のASO−高分子接合体の薬学的有効量を含む薬剤学的組成物。
- 請求項37に記載のナノ粒子を薬学的有効量を含む薬剤学的組成物。
- 請求項32に記載のリガンド結合SO−高分子接合体の薬学的有効量を含む薬剤学的組成物。
- 請求項38に記載のナノ粒子を薬学的有効量を含む薬剤学的組成物。
