- 前記N−末端ドメインとしてKIF5B蛋白質は、NM_004521のポリヌクレオチドによって暗号化されるKIF5B蛋白質の1番目アミノ酸から329番目アミノ酸の連続的な少なくとも329個のアミノ酸を必須的に含む、請求項2に記載の融合蛋白質。
- 前記N−末端ドメインとしてKIF5B蛋白質は、KIF5B蛋白質の329番目アミノ酸から始まる2つ以上のKIF5Bコイルドメインを追加で含む、請求項3に記載の融合蛋白質。
- 前記N−末端ドメインとしてKIF5B蛋白質は、NM_004521のポリヌクレオチドの1番目エクソンから16番目エクソンのポリヌクレオチド、1番目エクソンから15番目エクソンのポリヌクレオチド、または1番目エクソンから23番目エクソンのポリヌクレオチドによって暗号化されるアミノ酸配列を必須的に含む、請求項2乃至4のうちのいずれか一項に記載の融合蛋白質。
- 前記C−末端ドメインとしてRET蛋白質は、NM_020630またはNM_020975のポリヌクレオチドの12番目エクソンの1番目アミノ酸から始まって、NM_020630またはNM_020975のポリヌクレオチドによって暗号化されるRET蛋白質のC−末端に向かって、連続的な300個以上のアミノ酸を必須的に含むものである、請求項2に記載の融合蛋白質。
- 前記C−末端ドメインとしてRET蛋白質は、NM_020630またはNM_020975の12番目エクソンの開始点に該当するアミノ酸から始まって、NM_020630またはNM_020975のポリヌクレオチドによって暗号化されるRET蛋白質のC−末端に向かって、連続的な300乃至450個のアミノ酸を必須的に含む、請求項2乃至6のうちのいずれか一項に記載の融合蛋白質。
- 前記融合蛋白質は、
配列番号3、7、11または15のアミノ酸配列を有するか、または
配列番号3、7、11または15番のアミノ酸配列と90%以上のアミノ酸配列同一性(sequence identity)を有するアミノ酸配列を有する、請求項2に記載の融合蛋白質。 - 請求項1乃至8のうちのいずれか一項によるKIF5B−RET融合蛋白質を暗号化する融合遺伝子。
- 前記融合遺伝子は、
配列番号1、5、9または13のヌクレオチド配列を有するか、または
配列番号1、5、9または13のヌクレオチド配列と90%以上のヌクレオチド配列同一性(sequence identity)を有するヌクレオチド配列を有する、請求項9に記載の融合遺伝子。 - 10番染色体上逆位または転座を含む、RET−関連染色体の再配列;
請求項1乃至8のうちのいずれか一項によるN−末端ドメインとして融合パートナーとC−末端ドメインとしてRET蛋白質を含む融合蛋白質;
融合蛋白質を暗号化する融合遺伝子;及び
癌にかからない個別対象から得られた標準時料と比較してRET過発現からなる群より選択された少なくとも1種以上を、対象から得られた試験試料で探知する段階を含み、
前記少なくとも1種以上が試験試料から検出される場合、前記対象を癌患者と決定する、
肺ガン診断の情報を提供する方法。 - 前記RET−関連染色体の再配列は、10番染色体上逆位である、請求項11に記載の肺ガン診断の情報を提供する方法。
- 前記融合蛋白質を暗号化する融合遺伝子は、大規模並列配列分析法(massive parallel sequencing technology)を用いた全ゲノム(whole−transcriptome、RNA)及び全ゲノム(whole−genome、DNA)併合技術で探知及び検証される、請求項11に記載の肺ガン診断の情報を提供する方法。
- 前記融合蛋白質を暗号化する融合遺伝子は、配列番号1、5、9または13でそれぞれ配列番号2、6、10または14の融合領域を含む連続的な100乃至200個のヌクレオチドを含むポリヌクレオチド断片を生成できるプライマー対、及び配列番号2、6、10または14の融合領域、及び配列番号4、8、12または16の融合領域に結合する抗体またはアプタマーと混成化可能なポリヌクレオチドからなる群より選択された1種以上を用いて探知される、請求項13に記載の肺ガン診断の情報を提供する方法。
- 前記融合蛋白質を暗号化する融合遺伝子は、
5’−GTGAAACGTTGCAAGCAGTTAG−3’(配列番号20)と5’−CCTTGACCACTTTTCCAAATTC−3’(配列番号21)のプライマー対、または5’−TAAGGAAATGACCAACCACCAG−3’(配列番号22)と5’−CCTTGACCACTTTTCCAAATTC−3’(配列番号21)のプライマー対を用いて探知され、
前記10番染色体上逆位は5’−CAGAATTTCACAAGGAGGGAAG−3’(配列番号18)と5’−CAGGACCTCTGACTACAGTGGA−3’(配列番号19)のプライマー対を用いて探知される、請求項11に記載の肺ガン診断の情報を提供する方法。 - 配列番号2、6、10または14からなる、融合領域と混成化可能なポリヌクレオチド、
配列番号1、5、9または13でそれぞれ配列番号2、6、10または14の融合領域を含む連続的な100乃至200個のヌクレオチドを含むポリヌクレオチド断片を生成できるプライマー対、
10番染色体上逆位領域と混成化可能なプローブ、
10番染色体上逆位領域を含む連続的な100乃至200個のヌクレオチドを含むポリヌクレオチド断片を生成できるプライマー対、及び
配列番号4、8、12または16の融合領域に結合する抗体またはアプタマーからなる群より選択された少なくとも1種以上を含む、
肺ガン診断用組成物。 - 前記プライマー対は、
請求項1による融合蛋白質を暗号化するKIF5B−RET融合遺伝子を探知するための、5’−GTGAAACGTTGCAAGCAGTTAG−3’(配列番号20)と5’−CCTTGACCACTTTTCCAAATTC−3’(配列番号21)のプライマー対、または5’−TAAGGAAATGACCAACCACCAG−3’(配列番号22)と5’−CCTTGACCACTTTTCCAAATTC−3’(配列番号21)のプライマー対、または
10番染色体上逆位を探知するための、5’−CAGAATTTCACAAGGAGGGAAG−3’(配列番号18)と5’−CAGGACCTCTGACTACAGTGGA−3’(配列番号19)のプライマー対である、
請求項16に記載の肺ガン診断用組成物。 - 請求項1乃至8のうちのいずれか一項による融合蛋白質に対する1種以上の融合蛋白質抑制剤、前記融合蛋白質を暗号化する融合遺伝子に対する1種以上の融合遺伝子抑制剤、RET暗号化遺伝子に対する1種以上の抑制剤、またはこれらの混合物を、活性成分として含む、肺ガン予防または治療用組成物。
- 前記融合蛋白質抑制剤は、融合蛋白質に特異的に結合するアプタマー、融合蛋白質に特異的に結合する抗体、ソラフェニブ(sorafenib、4−[4−[[4−chloro−3−(trifluoromethyl)phenyl]carbamoylamino]phenoxy]−N−methyl−pyridine−2−carboxamide)、及びカボザンチニブ(Cabozantinib、N−4−(6,7−Dimethoxyquinolin−4−yl)oxy)phenyl)−N−4−fluorophenyl)cyclopropane−1,1−dicarboxamide)からなる群より選択された1種以上であり、
前記融合遺伝子抑制剤またはRET暗号化遺伝子抑制剤は、
融合遺伝子またはRET暗号化遺伝子に特異的に結合可能なsiRNA、shRNA、miRNA、及びアプタマーからなる群より選択された1種以上である、
請求項18に記載の肺ガン予防または治療用組成物。 - 融合蛋白質を発現する細胞に試料化合物を接触させ;及び細胞内融合蛋白質の発現水準を測定する段階を含み、
前記試料化合物を処理した細胞内融合蛋白質の発現水準が、試料化合物を処理する前の細胞または試料化合物を処理しない細胞の融合蛋白質の発現水準に比べて減少した場合、前記試料化合物を抗ガン剤候補物質と決定する、抗ガン剤のスクリーニング方法。
