Abstract
本発明は、脂質異常症の治療ならびに予防、特に、心臓血管にリスクを有する患者においてＨＤＬコレステロールを増加させるのに有用である、チョウセンアザミの葉の抽出物の調製物に関する。脂肪と炭水化物のバランスを正常化するため、ならびに、特に、薬物により誘発された脂質異常症を有する梗塞後の患者におけるＬＤＬ／ＨＤＬ比を良好に変化させることでＨＤＬコレステロール量を著しく増加させるために、前記抽出物は有用である。

選択されたチョウセンアザミの栽培品種、好ましくはチョウセンアザミあるいはカルドン（Ｃｙｎａｒａ ｃａｒｄｕｎｃｏｌｕｓ）、好ましくは、チョウセンアザミの若芽が、本発明の抽出物の調製に使用される。発芽苗の植え替え後、あるいは、種の発芽後、約一か月の葉を使用することが好ましい。ＨＤＬリポタンパク質の生成の調節に係わる活性な脂溶性成分の含有率を同時に損なうことなく駆除薬を除去することは不可能であるため、駆除薬を使用せず、また、生育培地中における駆除薬の不存在を監視する、有機農業の手法を使用して該チョウセンアザミ属の種を作り出すことが好ましい。
特に好ましいチョウセンアザミの品種は、チョウセンアザミの有棘のムラサキ色の品種である。茎葉の高さが２０ｃｍに達するまで生育させた植体から、６０℃〜１００℃、好ましくは８０℃の温度において、容易に乾燥される植物のバイオマスを取得する。
好ましくは、選択された栽培品種は、３．８％の含有率のカフェオイルキナ酸、１．５％の含有率のルテオリン フラボノイド、ならびに、３％の含有率のシナロピクリンを含まなければならない。乾燥後、植物のバイオマスは、例えば、WO 2007/006391、ならびに、WO 2008/107183中に記載される、公知の手法で抽出される。
得られる抽出物は、３０〜４５％、好ましくは、３５±２％の含有率のカフェオイルキナ酸、８〜１６％、好ましくは、１２±２％の含有率のフラボノイド、ならびに、１０〜１８％、好ましくは、１３±２％の含有率のシナロピクリンを含む。
前記抽出物は、全コレステロール、ＬＤＬ コレステロールおよび血糖の低減、ならびに、ｃＨＤＬの増加において、予期せぬ活性を示した。より具体的には、抽出物は、有意に、空腹時の血糖、全コレステロールおよびＬＤＬ コレステロールの濃度を２０％低減し、利尿を増進し、従って、血圧の低減に役立ち、そして、脂肪肝の軽減に寄与する。
特に、高脂血症の（ｈｙｐｅｒｌｉｐａｅｍｉｃ）患者と、スタチン治療またはコレステロール低下薬に起因してＨＤＬ コレステロールの正常値を下回っている患者の双方において、ＨＤＬ コレステロールの予期せぬ高い増加が観測されることを強調すべきである。チョウセンアザミの抽出物の投与に続く、ＨＤＬ コレステロールにおける中程度の不規則な増加は文献に報告されているが、その再現性ならびに整合性の証拠は付されていない。事実、以下の研究（Natur. Med.,13、17-24、1998; Arzneim-Forschung, 50, 260-65, 2000, The Cochrane Library, 2002, Issue 3）は、実質的な関連性が低い相反するデータを報告している。
逆に、本発明の組成物を用いた実験は、全コレステロールとＬＤＬ コレステロールにおいて約２０％の低減と、ＨＤＬ コレステロールにおいて１９％の有意な増加を示している。２００〜２８０ｍｇ／ｄｌの範囲の全コレステロールを示す患者の事例において、その期間にわたり変化しないことが見出されたこの増加は、公知の薬草調製物では観測されていない。
また、本発明の抽出物は、メタボリック症候群を患っている患者における種々の指標に対して有効であることを証明し、すなわち、血糖、脂質の指標、および高血圧等の指標の正常化が観測された。
本発明の抽出物が、カプセル剤、あるいは、通常または胃保護型の錠剤等の適合する投与形態に製剤化されるであろう。
好ましい担体は、抽出物中に存在するシナロピクリンの吸収を促進するω−３ 脂肪酸に富む油である。
さらなる形態においては、本発明の組成物は有用な、あるいは、補完的な活性を有する他の物質と組み合わせて、投与することができる。
本発明の組成物は、例えば、“Remington‘s Pharmaceutical Handbook”, Mack Publishing Co., N.Y., USA,に記載される従来手法に従って、製剤化されるであろう。特に、付形剤ならびにカプセルの母材との相互干渉の回避に特に注意を要する従来の植物成分の製剤化方法に従って、本発明の組成物を製剤化できるだろう。経口製剤の例は、錠剤、糖衣錠、ソフトおよびハード・ゼラチン・カプセル、ならびに、セルロース・カプセルである。平均的投与量は、１日、１〜３回、１００〜５００ｍｇの抽出物に相当する。
下記の実施例は、本発明をより詳細に説明する。

Claims (9)
- ３０〜４５％の含有率のカフェオイルキナ酸、８〜１６％の含有率のフラボノイド、ならびに、１０〜１８％の含有率のシナロピクリンを含む、チョウセンアザミ属の種の品種の葉の抽出物。
- カフェオイルキナ酸の含有率は３５±２％であり、フラボノイドの含有率は１２±２％であり、ならびに、シナロピクリンの含有率は１３±２％である、請求項１に記載の抽出物。
- 選別されたチョウセンアザミの栽培品種、あるいは、カルドンの栽培品種の若芽から得られる、請求項１または２に記載の抽出物。
- チョウセンアザミの栽培品種から得られる、請求項３に記載の抽出物。
- 前記栽培品種は、有棘のムラサキ色の品種である、請求項４に記載の抽出物。
- 高血糖症、高コレステロール血症、高血圧症、ならびに、脂肪肝の薬理学的治療に使用するための、請求項１〜５に記載の抽出物。
- ＨＤＬコレステロールの増加に使用するための、請求項１〜５に記載の抽出物。
- 少なくとも一つの付形剤または担体と混合した状態で、請求項１〜５に記載の抽出物を活性成分として含有する医薬組成物。
- 担体は、ω−３不飽和脂肪酸を含有する油である、請求項８に記載の組成物。
