Abstract
Description
ピロフェオホルビド-脂質の合成
ピロフェオホルビド(ピロ)は、以前記述されたように(Zheng et al., Bioconjugate Chemistry, 13-392, 2002)、Spirulina pacifica藻類(シアノテック社)から得た。標準的な反応において、107 mgのピロ(200 umol)を、98.7 mgの16:0リゾホスファチジルコリン(1-パルミトイル-2-ヒドロキシ-sn-グリセロ-3-ホスホコリン、アヴァンティ・ポーラー・リピッド社 # 855675)、76.3 mgのEDC(シグマ社)、および48.7 mgのDMAP(シグマ社)と、5 mLのアミレン安定化クロロホルム(シグマ社)中で混合した。反応混合物を、室温にてアルゴン下で24時間撹拌した。得られたピロ-脂質位置異性体産物は、2695 HPLC/MSマイクロマスZQ 2000システム(ウォーターズ社)を用いて、質量分析を伴う逆相HPLCにより分析した。4.6 mm×75 mmの3.5μmサンファイアーC8 HPLCカラム(ウォーターズ社)を、60℃におけるカラム加熱、0.1%TFA中15%アセトニトリルから0.1%TFA中95%アセトニトリルまでの勾配を用いて、流速0.5 mL/分で15分間にわたって使用した。ピロ-脂質位置異性体をさらなる分析に付し、あるいは、40℃の加熱を伴う回転式蒸発器を用いて減圧下でクロロホルムを除去してさらなる酵素的位置異性体選択を実施した。
酵素スクリーニングのために、シグマ社から入手した以下の酵素を使用した:Arachis hypogaea(ピーナッツ)由来のホスホリパーゼD-II型(P0515);Burkholderia由来のリポタンパク質リパーゼ(L9656);Candida rugosa由来のリパーゼ、VII型(L1754);Clostridium perfringens由来のホスホリパーゼC、I型(P7633);ブタ肝臓由来のエステラーゼ(E3019);ブタ膵臓由来のホスホリパーゼA2(P6534);ブタ膵臓由来のリパーゼ(L3126);Thermomyces lanuginosus由来のホスホリパーゼA1(L3295);Bacillus stearothermophilusの組換えエステラーゼ(69509);Pseudomonas cepacia由来のリパーゼ(62309);Rhizomucor miehei由来のリパーゼ(L4277);Thermomyces lanuginosus由来のリパーゼ(L0777);ミツバチ毒由来のホスホリパーゼA2(P9279)。酵素スクリーニングのためには、酵素を反応あたり2 nmolのピロ-脂質とともに加え、10 ugの乾燥酵素(ホスホリパーゼD、ホスホリパーゼC、リポタンパク質リパーゼ、Candida rugosa由来リパーゼ、ブタ肝臓由来エステラーゼ、ホスホリパーゼA2、ブタ肝臓リパーゼ)、あるいは液体もしくは懸濁形態で得られた酵素(上記以外の全ての酵素)については1 uLを用いて、50 uLの0.025% Triton(登録商標)X100および5 mM Tris pH 8中で37℃で3時間インキュベートした。インキュベーション後、サンプルを50 uLのDMSOと組み合せ、遠心分離して沈殿タンパク質を除去し、上記のようにHPLC/MSに付した。ミツバチ毒由来のホスホリパーゼA2およびThermomyces lanuginosus由来のリパーゼをそれぞれsn-1およびsn-2コンジュゲート化ピロ-脂質異性体を切断するための最良の酵素として同定した後、反応あたり3 nmolのピロ-脂質を使用して、表示された時間点にてインキュベートして、これらの2つの酵素を再評価した。sn-2位にピロが結合したピロ-脂質の標準的な精製のためには、乾燥した混合ピロ-脂質を、1 mM CaCl2、50 mM Tris pH 8、0.5% Triton(登録商標)X-100、および10% MeOH中に、5 mg/mLのピロ-脂質となるように直接再懸濁した。それからミツバチ毒由来のPLA2を0.1 mg/mLの濃度にて加え、この溶液を37℃にて24時間インキュベートした。sn-1位にピロが結合したピロ-脂質の調製のためには、sn-1およびsn-2ピロ-脂質の位置異性体混合物をまず精製し、それから0.5% Triton(登録商標)X-100および1 mM CaCl2中で2.5 mg/mLのピロ-脂質にてインキュベートし、10 mLのピロ-脂質溶液あたり200 uLのThermomyces lanuginosus由来調製リパーゼ溶液を加えることによって消化を行なった。反応状態はHPLC/MSを用いて調査し、数日間にわたって実行され、1日毎にさらに100 uLのリパーゼを追加した。反応後、HPLC/MSを使用して、望まない位置異性体が消失したことを確認することにより、完了を確証した。さらなる2体積のクロロホルムおよび1.25体積のメタノールを加え、ピロ-脂質を有機相から抽出し、40℃、減圧にて回転式蒸発器を用いて溶媒を除去した。それからピロ-脂質をDCM中に再懸濁し、ジオールシリカ(ソーブテック社、#52570)で充填したフラッシュクロマトグラフィーカラムに適用した(100 mgのピロ-脂質あたり約10 gの乾燥シリカ粉末)。ピロ-脂質に対してソレラ・フラッシュクロマトグラフィー・システム(バイオテージ社)を使用し、DCM中0〜10%のメタノール勾配を用いた。溶出画分をまとめ、減圧にて回転式蒸発器を用いて溶媒を除去した。最後に、ピロ-脂質を10 mLの20%水および80% t-ブタノール中に溶解し、液体窒素中で凍結し、2〜3日間にわたって凍結乾燥した。sn-1ピロ-脂質については、NMR解析結果は以下の通りであった:
異性体として純粋なピロ-脂質、コレステロール(アヴァンティ・ポーラー・リピッド社)、およびジステアロイル-sn-グリセロ-3-ホスホエタノールアミン-N-メトキシ(ポリエチレングリコール)(PEG-2000-PE、アヴァンティ・ポーラー・リピッド社)をクロロホルム中に溶解した。解析研究のために、ホウケイ酸試験管中における分散、窒素流下における乾燥、それに続く真空下における残余クロロホルム除去によって、95%ピロ-脂質(表示された位置異性体組成のもの)および5% PEG-2000-PEの0.5 mgの薄膜を形成した。脂質薄膜をリン酸緩衝食塩水(150 mM NaCl、10 mMリン酸pH 7.4)で再水和させ、液体窒素を用いて凍結させた後に65℃温水槽にて解凍することにより、5回の凍結-解凍サイクルに付した。その後、ミニ押出し機(アヴァンティ・ポーラー・リピッド社)を用いて、100 nmポリカーボネート膜を通して15回、ポルフィソームを押し出した。サイズ測定はナノサイザー(マルバーン社)にて実施した。蛍光自己消光はフルオロマックス蛍光光度計(ホリバジョバンイボン社)を使用して記録した。ポルフィソーム溶液をPBS中で2.5 ug/mLに希釈し、2 nmスリット幅を用いて420 nmにて励起した。発光は1 nmスリット幅を用いて600から800 nmまで測定し積分した。バックグラウンドの減算は、等濃度の100 nm卵ホスファチジルコリン:コレステロール(3:2モル比)リポソームを用いて行なった。透過型電子顕微鏡観察は、H-7000透過型電子顕微鏡(日立製作所)を使用して、1%酢酸ウランでネガティブ染色したポルフィソームを用いて実施した。インビボ光熱研究のためには、65モル%のポルフィリン-脂質を、クロロホルム中に溶解した30モル%コレステロールおよび5モル% PEG-2000-PEと組み合せることにより、5 mgの脂質薄膜を調製し、これを窒素ガスの気流下で穏やかに乾燥させ、真空下で1時間さらに乾燥させた。それからこの脂質薄膜を1 mLのPBS中で再水和させ、5回の凍結-解凍サイクルに付してポルフィソーム懸濁液を得た。この懸濁液を、10 mLのLIPEX(登録商標)サーモバレル押出し機(カタログ番号T.005、ノザンリピッズ社、カリフォルニア州)を使用して、700 psi(4826 kPa)の窒素ガス下で、孔径100 nmのポリカーボネート膜(アヴァンティ・ポーラー・リピッド社)を通して10回押し出した。温度は、循環温水槽を用いて70℃に正確に制御した。
光熱治療のためには、2x106の細胞をマウスの右側腹部に注射することにより、KB腫瘍をメスのヌードマウス中で増殖させた。腫瘍の直径が4〜5mmに達したとき、30モル%コレステロールおよび5モル% PEG-2000 DSPEを含有する42 mg kg-1のポルフィソームを尾静脈から注射した。注射24時間後に、2% (v/v)イソフルオレンを用いてマウスを麻酔し、671 nmにおける出力700 mW、直径8 mmのスポットサイズを有するレーザー(671 nm 2W DPSSレーザー、レーザーグロー・テクノロジーズ社)で腫瘍を照射し、赤外線カメラ(ミクロスペック社)を用いて腫瘍温度を記録した。腫瘍体積を1日毎に測定し、腫瘍の直径が10 mmに達したらマウスを安楽死させた。
1:1:2:2のピロ:脂質:DMAP:EDCモル比を使用したアシル化反応において、ピロフェオホルビド-a(ピロ)をリゾリン脂質1-パルミトイル-2-ヒドロキシ-sn-グリセロ-3-ホスホコリンにコンジュゲート化した。この反応は一晩で完了状態まで進行したが、2つの位置異性体、すなわち1つはピロをsn-2位に有し、1つはピロをsn-1位に有し、それぞれsn-2ピロ-脂質およびsn-1ピロ-脂質と呼ばれるものを産生した(図1A)。これら異性体の存在は、60℃におけるカラム加熱を用いたHPLC-MSを使用して、近接して一緒に溶出する2つのピークとして明らかにされた(図1B)。これらのピークはどちらも、ピロ-コンジュゲート化脂質の同じ予測分子量吸収スペクトルを示した(図7)。異性体のアイデンティティは、酵素切断されたコンジュゲートにおけるグリセロール骨格の中央炭素上の水素の1H化学シフトを調べることによって確認し、この水素はエステルまたは第一級アルコールのどちらかに連結していた(酵素切断は下記において説明し、切断産物および未消化位置異性体のNMRスペクトルは図8に示している)。ピロ対リン脂質の出発比の調節は、得られる位置異性体産物の比の変化をもたらした。1:1の比を使用した場合には、産物の80%超がsn-2ピロ-脂質異性体(すなわち、sn-2ヒドロキシルにピロがコンジュゲート化したもの)であった。1:7に比を上昇させた場合には、sn-2ピロ異性体産物は35%に減少し、sn-1ピロ異性体は65%に増加した。この観察の1つの説明は、リゾリン脂質のわずかな一部が、ピロと反応する前にアシル基転位を受けたということである。アシル基転位リゾリン脂質はより反応性が高い第一級アルコールを含んでいたため、反応において迅速に消費され、従ってリゾリン脂質のより大きな出発比はより多くのsn-1ピロ-脂質の生成をもたらしたのである。従って、反応条件を変化させることによりある程度の位置異性体選択を達成することができたが、より改善された異性体純度を達成するためには他のアプローチが必要であった。
Claims (51)
- ポルフィリン-リン脂質コンジュゲートの二重層を含むナノベシクルであって、各々のポルフィリン-リン脂質コンジュゲートは、1つのリン脂質のsn-1位またはsn-2位のうちの1つにおける脂質側鎖に共有結合した1つのポルフィリン、ポルフィリン誘導体、またはポルフィリン類似体を含み、前記ナノベシクルは、規定されたsn-1:sn-2ポルフィリン-リン脂質コンジュゲートの位置異性体比を有するナノベシクル。
- 位置異性体的に実質的に純粋である、請求項1に記載のナノベシクル。
- ナノベシクルにおける前記ポルフィリン-リン脂質コンジュゲートの位置異性体としての純度が>97%である、請求項2に記載のナノベシクル。
