R-A1A2-アミドベンジル-カルボナート

NaHCO3水溶液中のアミノ酸のカップリングおよびシリル基脱保護を除き、すべての反応を新鮮無水溶媒を用いて無水条件で実施した。カラム精製は、明記した溶離剤を用いてシリカゲル上で行った。質量スペクトル(MS)はエレクトロスプレーイオン化を用いて得た。

i)DME(40mL)中のFmoc-Phe-NHS(4.96g、10.26mmol)をH2O(40mL)およびTHF(20mL)の混合物中のL-シトルリン(1.80g、10.26mmol)およびNaHCO3(0.86g、10.26mmol)を含む溶液に加えた。反応混合物を15時間撹拌した。活性化からの残存DCCをろ過し、有機溶媒をロータリーエバポレーターで除去した。残渣にH2O(100mL)およびiPrOH(10mL)を加えた。懸濁液を5%KHSO4でpH=3まで酸性化し、生成物をEtOAc:iPrOH=9:1溶液(3×、500mL)で抽出し、食塩水:iPrOH=9:1の混合物(2×、50mL)で洗浄し、乾燥(Na2SO4)し、ろ過して濃縮し、オイルポンプで乾燥した。エーテルで粉砕して、純粋な生成物1eを得た。収量3.84g(68%)。MS: 545.6 [M+Na]+; 528.5 [M-H2O]+; 306.3 [M-Fmoc+H2O]+。
生成物1a〜hをp-アミノベンジルアルコール(PABA)と2-エトキシ-1-エトキシカルボニル-1,2-ジヒドロキノリン(EEDQ)存在下でカップリングし、2a〜hを生成した。PABAに1つのアミノ酸だけが結合している4つの代表的化合物3j〜lも調製した。

NAG(R1,R2,R3)-L-AA-PABC-PNP(表2、3)の調製
R1、R2およびR3が保護基であり、Lがガラクトサミン部分(NAG)とジペプチド(AA)との間の連結である、NAG(R1,R2,R3)-L-AA-PABC-PNPの調製を、NAG-L-CO2H酸6、10a、b、13および17の調製から開始し、これらをNHSエステルに変換した後、H-AA-PABA 2をアシル化するために用いた。塩基感受性ポリマーのために設計したカルボナート21a〜fにおいて、保護基をポリマー修飾の間に除去しなければならなかった。このために、10a、b、13および17の調製において、GAL部分のAc-保護基を不安定なトリエチルシリル(TES)およびtert-ブチルジメチルシリル(TBDMS)基で置き換えた。これらの基は、塩基感受性PNP-カルボナート部分を損なうことなく、H2O中70%トリフルオロ酢酸(TFA)溶液を0℃で用いて除去することができる。

10a、bの調製
条件:(i)H2、Pd/C(10%)、THF。(ii)無水コハク酸、Et3N、DCM、1時間。
7の調製のために、NAG 4(2g、3.52mmol)をMeOH(10mL)、H2O(32mL)、およびEt3N(25mL)の溶液中で10時間撹拌することによりO-脱アセチル化した。すべての揮発性物質をロータリーエバポレーターにより40℃で除去し、残渣を反応混合物からトルエンの蒸発を2回行って乾燥した。生成物7を次の段階で直接用いた。MS: 544.3 [M+Et3N+1]+; 443.7 [M+1]+; 204 [脱グリコシル化生成物]+。
17を、ペンタ酢酸エステル14[3〜5]をPEG4モノ-tBuエステルでグリコシル化して(TMSOTf、DCE/HO-PEG4-CO2tBu、SnCl4、DCM)15を得ることにより調製した。15を加水分解して(HCO2H、10時間)酸16を得、次いでこれをO-脱アセチル化し(Et3N、MeOH、H2O(5:7:6)10時間)、TBDMSClで処理して(DMF中TBDMSCl(3当量)、イミダゾール、10時間)ビス-シリル化NAG-酸17を得た。
条件:(i)トリフルオロメタンスルホン酸トリメチルシリル(TMSOTf)、ジクロロエタン(DCE)(ii)12-ヒドロキシ-4,7,10-トリオキサドデカン酸t-ブチル(HO-PEG4-CO2tBu)、SnCl4、ジクロロメタン(DCM)。
条件:(i)NHS、DCC、DXCM、10時間。
条件:(i)DIEA、DMF、5〜10時間。(ii)(PNP)2CO、ジオキサンまたはDCM、25〜60℃、16〜48時間。
生成物19a(R1=R2=R3=OAc、L=L1、AA=GlyGly)のために、DMF中のNAG-NHSエステル18a溶液(0.05M)(0.282mmol)をDMF中の3aの0.1M溶液(0.282mmol)およびDIEA(59μL、0.338mmol)で処理した。3時間で、すべての揮発性物質をロータリーエバポレーターにより40℃/オイルポンプ減圧下で除去し、Et2Oで粉砕し、カラムにより、溶離剤:EtOAc:CHCl3:MeOH=8:7:5〜8:7:6の勾配で精製した。収量114mg(53%)。MS: 754.4 [M+1]+。
生成物20a(R1=R2=R3=OAc、L=L1、AA=GlyGly)のために、ジオキサン(5mL)中の19a(100mg、0.132mmol)、(PNP)2CO(202mg、0.663mmol)およびDIEA(0.07mL、0.396mmol)の懸濁液を暗所、40℃で8時間撹拌した。さらに(PNP)2CO(121mg、0.397mmol)およびDIEA(0.04mL、0.226mmol)を加え、撹拌を40℃でさらに8時間続けた。すべての揮発性物質をロータリーエバポレーターで除去し、生成物をカラムにより、溶離剤:CHCl3:EtOAc:MeOH=7:8:3で精製した。収量84mg(69%)。
21a〜fの生成。塩基感受性ポリアクリラートの修飾のために、NAGおよびジペプチドAA上の保護基を除去した後、20f〜lをTFA:H2O=3:1の混合物で処理することによりポリマーに結合し(TFA/H2O=3:1、5℃、2〜3時間)、NAG-ジペプチドマスキング試薬21a〜fを提供した。

A. メリチンのプロテアーゼ切断可能なマスキング剤による修飾。1×mgのメリチンペプチドおよび10×mgのHEPES塩基を1〜10mg/mLペプチドで、2〜6×mgのNAG含有プロテアーゼ切断可能基質のアミン反応性p-ニトロフェニルカルボナートまたはN-ヒドロキシスクシンイミドカルボナート誘導体の添加によりマスクした。次いで、動物への注入前に、溶液を室温(RT)で少なくとも1時間インキュベートした。

DPCを前述のとおりに調製した。6〜8週齡のマウス(C57BL/6またはICR系統、それぞれ約18〜20g)をHarlan Sprague Dawley(Indianapolis IN)から入手した。マウスを注入前に少なくとも2日間ケージに収容した。給餌はHarlan Teklad Rodent Diet(Harlan、Madison WI)で適宜行った。DPCを本明細書に記載のとおりに合成した。結合体溶液(0.4mL)を尾静脈への点滴により注入した。組成物は生理的条件で可溶性かつ非凝集性であった。溶液を尾静脈への点滴により注入した。他の血管への注入、例えば、眼窩後注入も等しく有効であると予想される。Wistar Hanラット、175〜200gをCharles River(Wilmington、MA)から入手した。ラットを注入前に少なくとも1週間ケージに収容した。ラットの注入量は典型的には1mlであった。特に記載がないかぎり、注入の48時間後に血清試料を採取し、および/または肝試料を採取した。
マウスを4時間(ラットの場合は16時間)絶食させた後に顎下採血により血清を採取した。ラットの場合、血液を頚静脈から採取した。血清ApoBタンパク質レベルを、標準のサンドイッチELISA法によって定量した。簡単に言うと、ポリクローナルヤギ抗マウスApoB抗体およびウサギ抗マウスApoB抗体(Biodesign International)をそれぞれ捕捉および検出抗体として用いた。その後、HRP結合ヤギ抗ウサギIgG抗体(Sigma)を適用してApoB/抗体複合体を結合した。次いで、テトラメチルベンジジン(TMB、Sigma)発色の吸光度をTecan Safire2(Austria、Europe)マイクロプレート読み取り器により450nmで測定した。
動物からの血漿試料を、標準の手順に従い、血液(9倍量)(マウスの場合は顎下採血により、またはラットの場合は頸静脈から)を0.109mol/Lクエン酸ナトリウム抗凝固剤(1倍量)を含む微小遠沈管に採取することにより調製した。血漿中のF7活性を、BIOPHEN VIIキット(Hyphen BioMed/Aniara、Mason、OH)を製造者の推奨に従って用い、色素産生法により測定した。発色の吸光度をTecan Safire2マイクロプレート読み取り器により405nmで測定した。
100mMのpH7.5 HEPES緩衝液中のポリアクリラートAnt-41658-111を活性化ジスルフィド試薬のスクシンイミジルオキシカルボニル-アルファ-メチル-アルファ(2-ピリジル-ジチオ)トルエン(SMPT)(Pierce)で0.5重量%修飾し、続くsiRNAの結合のためのチオール反応性基を提供した。次いで、チオール反応性ポリマーを60mg/mL HEPES塩基中で5mg/mLに希釈した。この溶液に10mg/mLの様々な記載の酵素切断可能なマスキング試薬を加えた。この量はポリマーアミン1に対しマスキング剤2のモル比であった。ポリマー修飾反応のために、ポリマーアミンのマスキング試薬に対する好ましいモル比は1:1から1:5である。より好ましい比は1:2から1:4である。より好ましい比は1:2である。1時間後、酢酸エステル保護チオール内因性齧歯類第VII因子siRNA(ポリマーに対し0.1から0.2重量当量)をポリマー溶液に加えた。終夜インキュベートした後、無水マレイン酸のN-アセチルガラクトサミン誘導体(NAG-CDM;表5)を加えることにより、結合体をさらに修飾した。NAG-CDMを25mg/mLまで加え、30分から4時間インキュベートした。
式中、Rはポリマーであり、R1はASGPrリガンド(例えば、N-アセチルガラクトサミン)を含む。表5に示すとおり、第VII因子発現はジペプチド剤でマスクしたDPCで処置した動物では49〜85%低減した。

両親媒性膜活性ポリ(ビニルエーテル)ポリアミンDW1360を10×重量当量のジペプチド切断可能マスキング剤で、ポリマー修飾中にマスキング剤が保護基を保持している以外はポリアクリラートAnt-41658-111について記載したとおりに修飾した。ポリマー修飾後、アミノ酸保護基をTFAにより除去し、NAG酢酸エステル保護基を30体積%トリエチルアミン、50%メタノールおよび20%水の溶液存在下でのインキュベーションにより除去した。酢酸エステル脱保護溶液をロータリーエバポレーションにより除去した。マスクしたポリマーをマウスにコレステロール-ApoB siRNA結合体と同時注入した。

メリチンを前述のとおり示した量の酵素的に切断可能なマスキング剤で可逆的に修飾した。次いで、200〜300μgのマスクしたメリチンを50〜100μgのApoB siRNA-コレステロール結合体と同時注入した。ApoBレベルへの影響を前述のとおりに判定した。ペプチダーゼ切断可能なジペプチド-アミドベンジルカルバマートで修飾したメリチンは有効なsiRNA送達ペプチドであった。D型メリチンペチドの酵素的に切断可能なマスキング剤と組み合わせての使用が好ましい。ペプチドマスキングはより安定であったため、同じレベルの標的遺伝子ノックダウンにはより多くのペプチドが必要とされるが、治療指数は変化または改善のいずれもしなかった(CDM-NAGによる同じペプチドのマスキングに比べて)。

A)標的遺伝子ノックダウン測定
以下に示すすべての試験について、Aha1遺伝子転写物に特異的なsiRNA、標的遺伝子。高感度緑色蛍光タンパク質(EGFP)に対するsiRNAをオフターゲット対照として用いた。Aha1 siRNAはヒトおよびマウス遺伝子の両方において100%相同であるAha1の配列モティーフに相補的であった。したがって、Aha1 siRNAのヒト異種移植片における宿主の細胞または腫瘍細胞のいずれかへの送達はmRNA切断および分解を引き起こす。マウスおよびヒトAha1遺伝子で異なる配列モティーフを用いて、細胞型の混合集団を含む組織試料中のヒトAha1およびマウスAha1 mRNAレベルの両方の定量的測定を可能にするPCRプライマーを設計した。siRNA送達後24、48または72時間の時点で、いくらかの健常マウス肝組織が付着した腫瘍を採取し、全RNA単離のためにTri-Reagent(Invitrogen)中で処理した。次いで、ヒトおよびマウスAha1 mRNAレベルの両方をqPCR検定により、内部参照遺伝子としてヒトCyc-Aおよびマウスβ-アクチンを用いて測定した。模擬注入した動物、またはオフターゲット対照GFP siRNAを注入したマウスからの動物におけるAha1 mRNAレベルを100%と考えた。結果は対照に対するAha1 mRNAレベルのパーセントで表し、以下の表に示す。
HegG2、Hep3B、またはHuH7細胞肝細胞癌に2つの発現ベクター、pMIR85ヒト胎盤分泌アルカリ性ホスファターゼ(SEAP)ベクターおよびpMIR3ネオマイシン/カナマイシン耐性遺伝子ベクターを同時形質移入して、安定にSEAPを発現する細胞株を生じた。細胞を、10%FBSおよび300ug/ml G418を補足したDMEM中で培養し、回収し、計数し、マトリゲル(BD Biosciences)(50体積%)と混合した。無胸腺ヌードマウスまたはScidベージュマウスを約3%イソフルランで麻酔し、胸骨臥位で置いた。剣状突起のすぐ下で小さい1〜2cmの正中線腹部切開を行った。湿った綿棒を用いて、肝左葉をゆっくり体外に出した。肝左葉をゆっくり退縮させ、シリンジ針を左葉の真ん中に挿入した。シリンジ針の斜端を肝臓被膜のすぐ下約0.5cmに挿入した。細胞100,000個を含む細胞/マトリゲル混合物10μlをシリンジポンプを用いて肝臓に注入した。針を肝臓内にしばらく(15〜20秒間)挿入したままにして、確実に注入を完了した。SEAP-HepG2細胞を無胸腺ヌードマウスに注入した。SEAP-Hep3BおよびSEAP-HuH7細胞をScidベージュマウスに注入した。次いで、シリンジを肝臓からの針から除去し、綿棒を細胞の漏出または出血を防ぐために注入部位上に置いた。マトリゲル/細胞混合物は目に見える塊を形成し、針の除去後も消失しなかった。次いで、肝葉をゆっくり腹部に戻し、腹壁を縫合した。腫瘍植え込み後、1週間に1回血清を採取し、SEAP検定にかけて腫瘍成長をモニターした。ほとんどの試験で、腫瘍測定値がSEAP値に基づき約4〜8mmと推定される、移植の4〜5週間後に腫瘍マウスを用いた。
HT29細胞を10%FBSを補足したMcCoy's 5a培地中で培養し、回収し、計数し、マトリゲル(BD Biosciences)(50体積%)と混合した。無胸腺ヌードマウスを約3%イソフルランで麻酔し、胸骨臥位で置いた。剣状突起のすぐ下で小さい1〜2cmの正中線腹部切開を行った。湿った綿棒を用いて、肝左葉をゆっくり体外に出した。肝左葉をゆっくり退縮させ、シリンジ針を左葉の真ん中に挿入した。シリンジ針の斜端を肝臓被膜のすぐ下約0.5cmに挿入した。細胞約40,000個を含む細胞/マトリゲル混合物5μlをシリンジポンプを用いて肝臓に注入した。針を肝臓内にしばらく(15〜20秒間)挿入したままにして、確実に注入を完了した。次いで、シリンジを肝臓から除去し、綿棒を細胞の漏出または出血を防ぐために注入部位上に置いた。マトリゲル/細胞混合物は目に見える塊を形成し、針の除去後も消失しなかった。次いで、肝葉をゆっくり腹部に戻し、腹壁を縫合した。移植の4〜5週間後に腫瘍マウスを用いた。
前述のとおり、(2011062805)Ant-129-1ポリマーDPCを18×重量過剰のPEG24-Phe-Citマスキング剤(またはPEG24-Val-Citマスキング剤)または7×PEG550-CDMのいずれかで修飾(マスク)した。前述のとおり、Aha1-siRNA(RD-09070)またはGFP-siRNA(RD-05814;オフターゲット対照)をポリマーに結合した(4:1の重量比)。DPCを送達前にゲルろ過で精製せず、標的指向リガンドを加えなかった。動物ごとに等張グルコース200μl中のDPC結合体320μg(ポリマー重量)を尾静脈注入により投与した(群ごとにn=3)。24時間後、等張グルコース200μl中のDPC結合体320μg(ポリマー重量)の二回目の注入を動物に投与した。二回目の注入の48時間後、血清試料を採取して、肝酵素(ALTおよびAST)および血中尿素窒素(BUN)レベルを測定することにより毒性を評価し、続いて組織を採取し、qPCR分析を行った。
前述のとおり、Ant-129-1ポリマーを5×Dig-PheCit(Dig-FCit)マスキング剤で修飾した。次いで、siRNAを結合体に結合した。最後に、Dig-FCit-Ant-129-1-siRNA結合体を8×(重量)PEG12-FCitでさらに修飾した。Aha1-siRNA(RD-09070)またはGFP siRNA(RD-05814)を4:1のポリマー:siRNA重量比で結合した。PEG12-FCit DPCをSephadex G50スピンカラムで精製して、未結合の試薬を除去した。
DPCを、a)PEG12-FCitの代わりにPEG24-FCitを用い、かつb)Dig-PEG12-NHSを用いてDigをポリマーに結合したこと以外は、前述のとおりに調製した。PEG24-FCit DPCはPEG12-FCit DPCよりも凝集が少なく、その大きさがより小さく、より均質であった。連結が不安定でないことに加えて、Dig-PEG12-NHSはより長いPEGも含んでいた。DPCはbsAbとの複合体であり、動物に前述のとおりに注入した。血清および組織採取を注入の24時間後または48時間後のいずれかに行った。表12に示すとおり、DPCの1回用量(250μgポリマー重量)は、注入の24時間後に46〜56%のヒトAha1ノックダウンを引き起こした。
Ant-129-1ポリマーを5×モル過剰のDig-PEG12-NHSおよび8×重量過剰のPEG24-FCitで修飾した。Aha1 siRNAまたはGFP siRNAを修飾ポリマーに4:1のポリマー:siRNA重量比で結合した。DPCをSephadex G50スピンカラムで精製して、未結合の試薬を除去した。注入前に、Dig-DPCを滅菌PBS中、等モル量のIGF1R-Dig bsAbもしくはCD33-Dig bsAbと、またはbsAbなしで、少なくとも30分間複合体形成した。HT29腫瘍細胞(ヒト結腸直腸腺癌;ATCC番号HTB-38)を含む動物にDPCを注入した。HT29細胞はインスリン様成長因子-1受容体タンパク質(IGF1R)を過剰発現し、IGF1R-Dig二重特異性抗体に結合し、これを内部に取り入れることができる。動物(n=3)にDPC(320μgポリマー重量)を投与した。24時間後に注入を繰り返した。血清および組織試料を2回目の投与の48時間後に採取した。腫瘍細胞におけるヒトAha1のノックダウンは26〜38%であった(表13)。CDM-DPCに比べて、FCit-DPCは低いオフターゲット肝Aha1ノックダウンを示した(78〜83%に対して24〜36%)。FCit-DPCはCDM-DPCに比べて肝蓄積の低減も示した。
400μgのLau41648-106を8×(重量)PEG12-ValCitまたは16×PEG24-PheCitで修飾した。100μgのAha1 siRNAまたは100μgのEg5対照siRNAを、前述のとおり、修飾ポリマーに結合した。Hep3B-SEAP腫瘍細胞を含む動物にDPCを注入した。血清および組織試料を注入の48時間後に採取した。腫瘍細胞におけるヒトAha1ノックダウンは26〜38%であった。
Lau24ABポリアクリラート(100μg)を50mM HEPES pH8.0緩衝液中、125I-ボールトン-ハンター(BH)試薬(50μCi)により、室温で1時間処理した。標識ポリマーを2mlの水中Sephadex QEAスピンカラムで精製した。標識ポリマーの溶液を4℃で保存した。200gのラット1匹あたり約1mgの0.2μCiを有するポリマーを注入するために、非標識ポリマーに125I標識ポリマーを補足した。標識および非標識ポリマーの混合物(約3.5匹の動物に対して計算)をPEG24-FCitまたはPEG-CDMで前述のとおりに修飾した(2mg/mlポリアクリラート、14mg/ml PEG-CDM試薬、14mg/ml NAG-PEG-CDM試薬、16mg/ml PEG24-FCit試薬)。インキュベーション時間1時間。次いで、反応混合物を等張グルコースで希釈して、1mlの量の動物ごとの注入用量を得た。3匹/群に注入した。所与の時間に動物から採血した(0.1〜0.2ml)。試料中のポリマーの量をガンマ計数器で計数することにより求めた。図4に示すとおり、プロテアーゼ切断可能なマスキング剤で修飾したポリマーはpHに不安定な無水マレイン酸マスキング剤でマスクしたポリマーよりも排出が遅かった。循環時間の延長は非肝組織への標的指向のために有益である。
A)ポリ(ビニルアクリラート)
i)N-Boc-エチルエトキシアクリラートおよびメタクリル酸プロピルのRAFT共重合:
式中:Aはboc保護エチル-エトキシアミノアクリラートであり
Bはメタクリル酸プロピルであり
CはRAFT剤CPCPA(4-シアノ-4-(フェニルカルボノチオイルチオ)ペンタン酸)であり
かつnおよびmは整数である。
合成後のboc保護基の除去によりアミンモノマーを生じる。
2,2'-アゾビス(2-メチルプロピオニトリル)(AIBN、ラジカル開始剤)、4-シアノ-4-(フェニルカルボノチオイルチオ)ペンタン酸(CPCPA、RAFT剤)および酢酸ブチルはSigma Aldrichから購入した。メタクリル酸プロピルモノマー(Alfa Aesar)はろ過して阻害剤を除去した。
乾燥し、沈澱した生成物をDCMに溶解した(100mg/mL)。ヘキサンを、曇り点に達する直後まで加えた(約20ml)。得られた乳状溶液を遠心分離した。下層(約60%のポリマーである濃稠な液体)を抽出し、ヘキサン中に完全に沈澱させた。残りの上部の溶液も、ヘキサンをさらに加えることにより完全に沈澱させた。両方の分画を遠心分離し、その後、ポリマーを単離し、減圧下で乾燥した。分画1:Mw 87,000(PDI 1.5);分画2:Mw 52,000(PDI 1.5〜1.6)。
約10mgのポリマーを0.5mLの89.8%ジクロロメタン、10%テトラヒドロフラン、0.2%トリエチルアミンに溶解した。分子量および多分散性(PDI)を、Shimadzu Prominence HPLCに接続したWyatt Helos II多角光分散検出器により、Jordi 5μ 7.8×300 Mixed Bed LS DVBカラムを用いて測定した。粗製ポリマー:MW:73,000(PDI 1.7)、分画1:MW 87,000(PDI:1.5)、分画2:MW 52,000(PDI 1.5〜1.6)
モノマーのモル供給比は80 BAEEA:20メタクリル酸プロピル(CAS:2210-28-8)およびモノマーの合計モルに基づき3%のAIBN触媒であった。BAEEA(6.53g、25.2mmol)(A)、メタクリル酸プロピル(0.808g、6.3mmol)(B)、AIBN(0.155g、0.945mmol)、およびジオキサン(34.5ml)を、撹拌子を入れた50mlのガラスチューブに加えた。化合物AおよびBは実施例16Aiで前述したとおりに調製した。反応は三つ組で計画した。各溶液に長いピペットを用いて窒素を1時間通気した。ピペットを除去し、各チューブに注意深く栓をした。次いで、各溶液を油浴を用いて60℃で3時間加熱した。各溶液を室温まで戻し、丸底中で混合した。粗製ポリマーを減圧下で乾燥した。MALSで得た分子量:55,000(PDI 2.1);H1NMRで得たポリマー組成:アミン:アルキル=74:26。
乾燥した粗製ポリマーを75%ジクロロメタン、25%テトラヒドロフラン、および0.2%トリエチルアミン中、50mg/mlで溶解した。次いで、ポリマーをJordi Gel DVB 104Å−500mm/22mmカラムで、流速5ml/分および注入10mlを用いて分別した。前半の分画を15〜17分で回収し、後半の分画を17〜19分で回収した。分画15〜17:Mw 138,000(PDI 1.1);分画17〜19:Mw 64,000(PDI 1.2)。

X mol%アミン保護ビニルエーテル(例えば、2-ビニルオキシエチルフタルイミド)を、乾燥器で乾燥した丸底フラスコの無水ジクロロメタン中に窒素雰囲気下で加える。この溶液に、Y mol%の低級疎水性基(例えば、プロピル、ブチル)ビニルエーテルおよび任意にZ mol%の高級疎水性基(例えば、ドデシル、オクタデシル)ビニルエーテルを加える(図1)。溶液を-50から-78℃の浴に入れ、2-ビニルオキシエチルフタルイミドを沈澱させる。この溶液に10mol%BF3・(OCH2CH3)2を加え、反応を-50から-78℃で2〜3時間進行させる。メタノール中の水酸化アンモニウム溶液を加えて重合を停止する。ポリマーを減圧下で乾燥し、次いで1,4-ジオキサン/メタノール(2/1)に加える。フタルイミドに対して20mol当量のヒドラジンを加えて、アミンから保護基を除去する。溶液を3時間還流し、次いで減圧下で乾燥する。得られた固体を0.5mol/LのHClに溶解し、15分間還流して、ポリマーの塩酸塩を生成し、蒸留水で希釈し、さらに1時間還流する。次いで、溶液をNaOHで中和し、室温(RT)まで冷却し、分子セルロースチューブに移し、蒸留水に対して透析し、凍結乾燥する。ポリマーをサイズ排除または他のクロマトグラフィを用いてさらに精製することができる。ポリマーの分子量を、分析用サイズ排除クロマトグラフィおよび多角光分散によるサイズ排除クロマトグラフィ(SEC-MALS)を含む、標準の手順に従ってのカラムを用いて推定する。
アミン/ブチル/オクタデシルポリ(ビニルエーテル)ターポリマーを、2-ビニルオキシエチルフタルイミド(5g、23.02mmol)、ブチルビニルエーテル(0.665g、6.58mmol)、およびオクタデシルビニルエーテル(0.488g、1.64mmol)モノマーから合成した。2-ビニルオキシエチルフタルイミドを、磁気撹拌子を含む200mLの乾燥器で乾燥した丸底フラスコの無水ジクロロメタン36mL中に窒素雰囲気下で加えた。この溶液に、ブチルビニルエーテルおよびn-オクタデシルビニルエーテルを加えた。モノマーを室温(RT)で完全に溶解して、澄明な均質溶液を得た。次いで、澄明溶液を含む反応容器を、ドライアイスをACS等級の変性アルコールおよびエチレングリコールの1:1溶液に加えることにより生成した-50℃の浴に入れ、フタルイミドモノマーの目に見える沈澱を生成させた。約1.5分間冷却した後、BF3・(OCH2CH3)2(0.058g、0.411mmol)を加えて、重合反応を開始した。フタルイミドモノマーは重合開始後に溶解した。反応を-50℃で3時間進行させた。メタノール中の1%水酸化アンモニウム5mLを加えて重合を停止した。次いで、溶媒をロータリーエバポレーションにより除去した。
すべてのメリチンペプチドを、当技術分野において標準のペプチド合成技術を用いて作成した。適切なメリチンペプチドは全L型アミノ酸、全D型アミノ酸(インベルソ)でありうる。LまたはD型とは独立に、メリチンペプチド配列を反転することができる(レトロ)。
Claims (20)
- 両親媒性膜活性ポリアミンを可逆的に修飾するための化合物であって、ジペプチド-アミドベンジル-カルボナートに共有結合している立体安定化剤または標的指向リガンドを含む、化合物。
- 下記によって表される構造を有する、請求項1記載の化合物:

R4は非荷電であって、ポリエチレングリコールまたは標的指向リガンドを含み、
R5は2、4、または6位にあって、-CH2-O-C(O)-O-Zであり、ここでZは

R6は、R5が占有している位置を除き、2、3、4、5、または6位のそれぞれで独立に水素、アルキル、-(CH2)n-CH3(ここでn=0〜4)、-(CH2)-(CH3)2、またはハロゲン化物である。 - R4が立体安定化剤を含む、請求項2記載の化合物。
- 立体安定化剤がポリエチレングリコール(PEG)である、請求項3記載の化合物。
- 標的指向リガンドがアシアロ糖タンパク質受容体(ASGPr)リガンドを含む、請求項2記載の化合物。
- ASGPrリガンドが、ラクトース、ガラクトース、N-アセチルガラクトサミン、ガラクトサミン、N-ホルミルガラクトサミン、N-プロピオニルガラクトサミン、N-n-ブタノイルガラクトサミン、およびN-イソ-ブタノイル-ガラクトサミンからなるリストより選択される、請求項5記載の化合物。
- R1が-CH2-フェニルである、請求項7記載の化合物。
- R2が-(CH2)3-NH-C(O)-NH2である、請求項7記載の化合物。
- 立体安定化剤がPEGである、請求項10記載の送達ポリマー。
- 標的指向リガンドがASGPrリガンドを含む、請求項10記載の送達ポリマー。
- ASGPrリガンドが、ラクトース、ガラクトース、N-アセチルガラクトサミン、ガラクトサミン、N-ホルミルガラクトサミン、N-プロピオニルガラクトサミン、N-n-ブタノイルガラクトサミン、およびN-イソ-ブタノイル-ガラクトサミンからなるリストより選択される、請求項12記載の送達ポリマー。
- 両親媒性膜活性ポリアミンが、ランダム、ブロック、または交互合成ポリマーからなるリストより選択される、請求項10記載の送達ポリマー。
- 両親媒性膜活性ポリアミンがメリチンペプチドである、請求項10記載の送達ポリマー。
- 両親媒性膜活性ポリアミンがRNAiポリヌクレオチドにさらに共有結合している、請求項16記載の送達ポリマー。
- インビボでRNAiポリヌクレオチドを哺乳動物の細胞に送達する方法であって、RNAiポリヌクレオチドを哺乳動物に請求項10記載の送達ポリマーと同時投与する段階を含む、方法。
- RNAiポリヌクレオチドが送達ポリマーに共有結合している、請求項18記載の方法。
- 細胞が肝実質細胞または肝臓癌細胞である、請求項18記載の方法。
