本発明に係る皮膚弾力改善効果を確認するために、前記実施例1〜3及び比較例1〜4の化粧水を用いて、下記のようにして試験した。30〜40代の女性35名をそれぞれ5名ずつ7組に分けてそれぞれ前記実施例1、実施例2、実施例3、実施例4、比較例1、比較例2、比較例3及び比較例4の化粧水をそれぞれ個別に毎日2回(朝、夕方)ずつ4週間顔面に塗布した後、皮膚弾力測定機(Cutometer SEM 575,C+K Electronic Co.,Germany)を用いて皮膚弾力改善効果を測定し、その結果を下記表2に示す。表2の結果値は、皮膚弾力測定機の皮膚粘弾性を意味する。

ヒト繊維芽細胞を24ウェル平板培養器で培養した後、下記表3の濃度で前記製造例1のツバキ酢エキス、製造例2のスイレン酢エキス、ツバキ酢エキスとスイレン酢エキスとの混合エキス、比較製造例1の通常のツバキエキス、比較製造例2の通常のスイレンエキス、及び通常のツバキエキスと通常のスイレンエキスとの混合エキスをそれぞれ個別に含有する培地に入れ替えた。培養3日目に、10%のウシ胎児血清入りダルベッコ変法イーグル培地(DMEM)を各ウェル当たりに0.5mlずつ添加した後、L[2,3,4,5−3H]−プロリン10μCiを添加した。24時間経過後に、各ウェルに入っている培地と細胞を掻き集めて5%のトリクロロ酢酸(TCA;Trichloroacetic acid)溶液に入れて水洗した後、2本の試験管に分注し、一方の試験管にはタイプIコラゲナーゼ(type I collagenase)1unit/μlを入れ、37℃において90分間培養し、他方の試験管は4℃において保管した。次いで、全ての試験管に50%のトリクロロ酢酸を0.05mlずつ添加し、4℃において20分間放置した後、それぞれ12,000rpmにて10分間遠心分離して、それぞれの上澄液と沈殿物を液体シンチレーション計数法(LSC;Liquid Scintillation Counter)で計数毎分(CPM;Count Per Minute)値を得て、下記数式1に基づいて対照群と実験群に対してコラーゲン生合成割合(RCB;Relative Collagen Biosynthesis)を測定し、その結果を下記表3に示す。
