Abstract

<P>PROBLEM TO BE SOLVED: To provide a skin care preparation for external use, obtained by formulating black vinegar or an extract of the black vinegar. <P>SOLUTION: The black vinegar is obtained by fermenting/maturing cereals such as a rice and a wheat for two or more months regardless of the cleaning degree. 本発明は、黒酢及び/または黒酢抽出物を有効成分として含有する各種剤、更にこれを配合してなる、皮膚炎症疾患や、肌荒れ、敏感肌といった皮膚及び頭皮のトラブルの予防ならびにその改善に有効で、安全性の高い皮膚外用剤に関するものであり、例えば、化粧品類(人及びその他の動物用に用いる各種製剤)、医薬品類、医薬部外品類等に用いることが出来る。
【0002】本発明は、抗老化作用、美白作用、抗炎症作用、高保湿作用、抗酸化作用を目的とした各種剤、及び化粧料に関する。

【従来の技術】
酢とは、調味料の一つで、3〜5%の酢酸を含む酸味のある液体を言う(例えば非特許文献1参照)。更に、酢はその原材料により、りんご酢、レモン酢、ワイン酢等の果実酢、米酢等の穀物酢等に分類されるが、その内容成分は大きく異なる。
従来、抗老化剤として果実酢を化粧用皮膚組成物に応用した例が知られている(例えば特許文献1参照)が、穀物酢をについてはあまり知られていない。

黒酢は穀物酢の一種であるが、一般の酢、もしくは米酢等との違いは食酢協会では半年以上の発酵、熟成されたものと定義されている。中国産の場合は特に香酢と呼ばれているが日本産、中国産の違いは日本産の場合、主に玄米を使うことが多く、中国産の場合はもち米、麦胚芽を使用し発酵、熟成される。また発酵期間については2ヵ月から8年が一般的である。
【0005】黒酢と一般的な米酢とでは、その含有アミノ酸量が大きく異なると言われている。http://www.foodrink.co.jpによると100ml中の主要アミノ酸量(mg)は表1のであるとされている。

【0007】黒酢の食品としての効用としては疲労回復、食欲増進、血圧降下、コレステロール低下、肝機能強化、肥満解消、便器解消が挙げられているが、皮膚外用剤として黒酢、その成分の有効利用について未だ報告されていない。本発明者等は黒酢の独自の新規有用的な利用方法・作用の検討、追求を試み鋭意研究を進めた結果、黒酢、もしくは黒酢抽出物が抗老化作用、美白作用、抗炎症作用、高保湿作用、抗酸化作用を有し、皮膚外用剤に応用することが非常に有効であることを見出し、本発明を完成するに至った。

【発明が解決しようとする課題】本発明の課題は、黒酢及び/または黒酢の抽出物を配合した新規の皮膚外用剤を提供することにある。

【課題を解決するための手段】本発明で使用する黒酢とは、米、麦等の穀物類をその精米度に関係なく2ヶ月以上発酵・熟成した酢をいう。特に日本産、中国産を限定するものではなく、また黒酢の原料はうるち米、もち米、麦胚芽、もろみなど黒酢製造に使用される原料であれば特に限定されるものではない。発酵に用いる菌の種類も特に限定されない。黒こうじ菌、酵母等発酵に用いることの出来る菌であれば良い。また発酵期間においても特に限定しないが、熟成期間をかければかけるほど色味が濃くなり、それと共に有機酸、ビタミン、ミネラル、アミノ酸などの成分が増えるため、好ましくは長いほうが良い。

本発明で使用する黒酢抽出物は、黒酢をそのまま或いは凍結乾燥等の前処理を行ったものを水、アルコール、多価アルコールまたはそれらの混合物を使用して、抽出することによって調製することが可能であり、それらを溶液状で使用することも可能であるが、それら溶液を乾燥して得られる粉体も皮膚外用剤に使用することが出来る。また、黒酢の抽出においては、水のかわりに緩衝液や、希酸および希アルカリを使用することも可能である。

抽出された黒酢抽出物はそのままでも使用可能であるが、さらに濾過、遠心分離等の工程により精製するのが好ましい。多くの場合は、そのままの状態で利用できるが、必要ならば、その効果に影響のない範囲で更に脱臭、脱色などの精製処理を加えても良く、脱臭・脱色等の精製処理手段としては、活性炭カラムなどを用いれば良く、抽出物質により一般的に適用される通常の手段を任意に選択して行えば良い。また、得られた黒酢抽出物を凍結乾燥法、スプレードライ法などの熱乾燥法により乾燥物として得ることも可能である。

本発明の黒酢及び/または黒酢抽出物が配合される各種剤の剤型は特に限定されず、例えば、液体、粉末、顆粒、クリーム、乳液、ジェル、軟膏、パップ、プラスター、スティック、シート等とすることができる。また化粧料としては、化粧水、乳液、美容液、一般クリーム、クレンジングクリーム等の洗顔料、パック、髭剃り用クリーム、日焼けクリーム、日焼け止めクリーム、日焼け止めローション、日焼けローション、化粧石鹸、ファンデーション、おしろい、パウダー、口紅、リップクリーム、アイライナー、アイクリーム、アイシャドウ、マスカラ、シャンプー、リンス、染毛料、頭髪用化粧品、浴用剤等、各種化粧品に利用可能である。黒酢の配合量は直接皮膚に塗布する組成物では、組成物中、0.00001〜100.0重量%含むことができるが、好ましくは0.001重量%〜10.0重量%であり、特に好ましくは0.01重量%〜5.0重量%である。また、浴用剤の場合では、200〜300Lの浴湯に投じるときに適当な濃度になる処方として、その配合量は0.01重量%〜70.0重量%とすることが好ましい。黒酢抽出物の場合、直接皮膚に塗布する組成物では、組成物中、乾燥重量で0.00001重量%〜100.0重量%配合可能であるが、好ましくは0.0001〜1.0重量%、特に好ましくは0.001〜0.5%である。また、浴用剤の場合では、200〜300Lの浴湯に投じるときに適当な濃度になる処方として、0.001〜10.0重量%とすることが好ましい。

本発明の各種剤及び化粧料には、以下に示すような通常、皮膚外用剤、化粧料において使用されている各種成分や添加剤の中から用途に適したものを任意に選択、併用して製造することができる。例えば保湿剤、ホルモン類、金属イオン封鎖剤、pH調整剤、キレート剤、防腐・防バイ剤、清涼剤、安定化剤、乳化剤、動・植物性蛋白質及びその分解物、動・植物性多糖類及びその分解物、動・植物性糖蛋白質及びその分解物、血流促進剤、消炎剤・抗アレルギー剤、細胞賦活剤、角質溶解剤、創傷治療剤、増泡剤、増粘剤、口腔用剤、消臭・脱臭剤、苦味料、酵素などが上げられ、これらとの併用によって、相加的及び相乗的な各種の効果が期待できる。

【発明の実施の形態】以下、本発明を実施例により説明するが、本実施例によって本発明が限定されるものではない。

黒酢100gを真空凍結乾燥して黒酢凍結乾燥物を6.0g得た。

黒酢を真空凍結乾燥し、黒酢凍結乾燥物10gを50%エタノール水溶液100mLに浸漬し60℃で24時間抽出し、沈殿物を遠心分離(10000G、10分)後、ろ過し、黒酢抽出物を0.1g得た。
【0019】実施例1〈抗酸化作用の測定方法(ロダン鉄法による)〉
2×10−2Mリノール酸原液1mlに0.1%供試料液(製造例1、製造例2で調製した黒酢抽出物)0.2mlと0.1Mリン酸緩衝液(pH7.0)0.8mlとを加え、この混合液を密封して37℃で72時間反応させた後、ロダン鉄法により過酸化物量を測定した。前記反応後の混合液の0.1mlを取り出し、これに75%エタノール4.7mlと30%チオシアン酸アンモニウム液0.1mlとを加えて撹拌し、さらに2×10−2M塩化第一鉄を含む3.5%塩酸溶液0.1mlを加え撹拌後、500nmにおける吸光度(As)を測定した。陰性対照として、前記0.1%供試料液の代わりに蒸留水を使用した場合の500nmにおける吸光度(Ac)を測定した。また、陽性対照として0.01%イーミックスD液(Tween
20を加えて水に溶解したもの)を添加した場合の500nmにおける吸光度(Ab)を測定する。そして、抗酸化活性率(%)は、数1により算出される。

【0024】実施例2〈コラゲナーゼ阻害効果の測定方法〉
まず、次の(1)0.25unit/ml精製I型コラゲナーゼ、(2)0.25mg/mlフルオレセインイソチオシアネート(以下FITCと称する)標識I型コラーゲン、(3)40mMO−フェナントロリンを調製する。
(1)0.25unit/ml 精製I型コラゲナーゼ:正常ヒト線維芽細胞由来精製I型コラゲナーゼをトリス塩酸緩衝液(pH7.5)で0.25unit/mlに調製する。
(2)0.25mg/ml FITC 標識I型コラーゲン:FITC標識I型コラーゲングルコースをトリス塩酸緩衝液(pH7.5)で0.25mg/mlに調製する。
(3)40mM O− フェナントロリン: O−フェナントロリンをトリス塩酸緩衝液(pH7.5)で40mMに調製する。

FITC標識I型コラーゲン、トリス塩酸緩衝液(pH7.5)を加え、35℃で2時間インキュベートし、次いで反応液に40mM O−フェナントロリンを添加し、35℃で30分間処理した後、エタノール沈殿法により分解されたコラーゲン含有上清を得る。この上清中に含有されるコラーゲンの蛍光強度を蛍光分光光度計により、励起波長:Ex=495nm、蛍光波長:Em=520nmにおける蛍光強度(Es)を測定する。また、「酵素検体(B)」は、各供試料液の代わりにブランクについて、前記「酵素・供試料検体」の測定と同様の操作を行って、Ex=495nm、Em=520nmにおける蛍光強度(Eb)を測定する。

【0030】これらの結果より、この発明にかかる黒酢または黒酢抽出物にコラゲナーゼ阻害効果が認められる。コラーゲンの分解酵素であるコラゲナーゼを抑制することから、黒酢または黒酢抽出物には弾性繊維であるコラーゲンの分解を低減させる効果、すなわち抗老化作用があることがわかる。また、コラゲナーゼ阻害物質は炎症性疾患の治療薬にも使われていることから、黒酢または黒酢抽出物にも抗炎症作用を有することがわかる。

表4に示す処方に従い、(1)〜(10)を攪拌、溶解し、化粧水を製造した。得られた化粧水はいずれも37℃の条件下において3ヶ月間安定であった。また、しみ、そばかすに悩むパネラーに適用し、その美白効果を調べた。パネラーは30〜50代の女性を1群10名として1つの化粧水を3ヵ月1日2回顔面に塗布し、期間終了時にパネラーに対する問診により、その美白効果を判定した。判定は、下記の基準に従ってスコアをつけ、平均スコアとして表した。
