Abstract
Description
本願は、その参照により本明細書に含められる2010年6月21日付け出願の米国特許出願第12/820,063号の優先権を主張するものである。
[基材をコーティングするために使用可能なリパーゼ含有材料の生産]
材料:ポリアクリレート樹脂Desmophen A870 BA、及びヘキサメチレンジイソシアネート(HDI)系多官能脂肪族ポリイソシアネート樹脂Resmodur N3600、ポリエステル樹脂Bayhydrol XP7093及びBayhydur 302は、Bayer Corp.(Pittsburgh,PA)から入手される。界面活性剤BYK−333は、BYK−Chemic(Wallingrord,CT)から入手される。Bacillus subtilis由来のα−アミラーゼ KLEISTASE SD80(EC3.2.1.1)、Burkholderia cepacia由来のリパーゼPS及びAspergillus niger由来のリパーゼAPI2は、天野エンザイム株式会社(名古屋、日本)から入手される。酢酸ブチル、ブラッドフォード試薬、ウシ血清由来のウシ血清アルブミン(BSA)、ポテト由来のデンプン、小麦由来のデンプン、マルトース、酒石酸ナトリウムカリウム、3,5−ジニトロサリチル酸、Na2(PO4)、NaCl、K2(PO4)、カゼイン、トリクロロ酢酸、フォーリン及びチオカルトのフェノール試薬、Na2(CO3)、酢酸ナトリウム、酢酸カルシウム、チロシン、パルミチン酸p−ニトロフェニル、エタノール、イオジン、グルコース、マルトース、マルトトリオース、マルトヘキソース、分子量が異なる(10kDa、40kDa)デキストリンは、Sigma Chemical Co.、St.Louis、MO、U.S.A.から入手した。アルミニウムパネル及び8パス・湿潤フィルムアプリケータは、Paul N.Gardner Company、Inc.(Pompano Beach、FL)から入手される。
約5mg/mLのタンパク質濃度(ブラッドフォード法により決定される)で、脱イオン水中に7.5gの粗製リパーゼ粉末を含む150mL溶液からリパーゼAP12を精製する。全ての溶液を氷上に保つ。30kDaの遮断膜を用いて55psiの圧力で、150mL容量のAmiconセル(Millipore,Billerica,MA)を使用して限外濾過を実施する。各ラン後150mLの体積になるまで残った濃縮溶液にDI(脱イオン)水を添加することによって、限外濾過を3回反復する。最終的に残った精製済み溶液を用いて、典型的に200mg/mLのリパーゼ濃度でコーティングを調製する。
[指紋除去]
例1のリパーゼ含有コーティング済みパネルに、顔又は前腕の皮膚に触れてヒトの指紋を付着させる。指紋のついたパネルを室温で少なくとも24時間インキュベートする。対照のパネルを、酵素を含まない例1のコーティングで被覆する。この第1のインキュベーション期間の後、コーティングされた基材を65℃以上の温度のオーブン内で1〜6時間インキュベートする。
指紋除去速度は、対照に比べ、酵素含有基材の場合の方が速い。図2は、例2で記述されているように2つのコーティングの間の界面に指紋が付着された例1に記載の酵素含有タンパク質−ポリマーコーティング又は対照でコーティングされたアルミニウムプレートを明示している。図2Aは、周囲温度で3日間のインキュベーションの後に残留している指紋を明示している。プレートをその後65℃で2.5時間インキュベートし、図2Bに示されているようにさまざまな間隔で写真を撮影する。図3は、指紋除去の速度上昇が、指紋の由来源によって左右されることを実証している。
Claims (18)
- 基材又はコーティング上の指紋の除去を容易にする方法であって、
基材又はコーティングを用意する工程、及び
リパーゼ又はその類似体を前記基材又は前記コーティングと結合させて、前記リパーゼ又はその類似体が指紋の成分を酵素分解できるようにする工程
を含む、方法。 - 前記リパーゼ又はその類似体が前記基材又は前記コーティングに共有結合により付着される、請求項1に記載の方法。
- 前記リパーゼ又はその類似体が前記基材又は前記コーティングに非共有結合的に接着されるか又はその中に混合される、請求項1に記載の方法。
- 前記基材を加熱するか又は前記基材の表面に熱を加える工程を含む、請求項1に記載の方法。
- 前記加熱が少なくとも30分間行われる、請求項4に記載の方法。
- 前記基材又は前記コーティングが有機架橋性ポリマー樹脂を含む、請求項1に記載の方法。
- 前記有機架橋性ポリマー樹脂が、アセトアセテート、酸、アミン、カルボキシル、エポキシ、ヒドロキシル、イソシアネート、シラン、ビニル、又はそれらの組み合わせの官能基を含む、請求項6に記載の方法。
- 前記有機架橋性ポリマー樹脂が、アミノプラスト、メラミンホルムアルデヒド、カルバメート、ポリウレタン、ポリアクリレート、エポキシ、ポリカルボネート、アルキド、ビニル、ポリアミド、ポリオレフィン、フェノール樹脂、ポリエステル、ポリシロキサン、又はそれらの組み合わせである、請求項6に記載の方法。
- 前記有機架橋性ポリマー樹脂がヒドロキシル官能化アクリレート樹脂である、請求項6に記載の方法。
- 前記リパーゼが、リポタンパク質リパーゼ、アシルグリセロールリパーゼ、ホルモン感受性リパーゼ、ホスホリパーゼA1、ホスホリパーゼA2、ホスホリパーゼC、ホスホリパーゼD、ホスホイノシチドホスホリパーゼC、リソホスホリパーゼ、ガラクトリパーゼ、又はそれらの類似体である、請求項1に記載の方法。
- 前記リパーゼがトリアシルグリセロールリパーゼである、請求項1に記載の方法。
- 指紋除去を容易にするための組成物であって、
基材又はコーティング、及び
指紋の成分を分解させることができるリパーゼ又はその類似体であって、前記基材又は前記コーティングと結合されるリパーゼ又はその類似体、
を含む、組成物。 - 前記基材又は前記コーティングが、アセトアセテート、酸、アミン、カルボキシル、エポキシ、ヒドロキシル、イソシアネート、シラン、ビニル、又はそれらの組み合わせの官能基を有する有機架橋性ポリマー樹脂を含む、請求項12に記載の組成物。
- 前記有機架橋性ポリマー樹脂が、アミノプラスト、メラミンホルムアルデヒド、カルバメート、ポリウレタン、ポリアクリレート、エポキシ、ポリカルボネート、アルキド、ビニル、ポリアミド、ポリオレフィン、フェノール樹脂、ポリエステル、ポリシロキサン、又はそれらの組み合わせである、請求項12に記載の組成物。
- 前記有機架橋性ポリマー樹脂がヒドロキシル官能化アクリレート樹脂である、請求項12に記載の組成物。
- 前記リパーゼが、リポタンパク質リパーゼ、アシルグリセロールリパーゼ、ホルモン感受性リパーゼ、ホスホリパーゼA1、ホスホリパーゼA2、ホスホリパーゼC、ホスホリパーゼD、ホスホイノシチドホスホリパーゼC、リソホスホリパーゼ、ガラクトリパーゼ、又はそれらの類似体である、請求項12に記載の組成物。
- 前記リパーゼがトリアシルグリセロールリパーゼである、請求項12に記載の組成物。
