Description
PTEN腫瘍抑制因子(参照により全体として本明細書に援用される国際公開第WO98/34624号を参照されたい)は、重要な二次メッセンジャーであるホスファチジルイノシトール3,4,5−三リン酸を脱リン酸化する細胞質ホスファターゼである(MaehamaおよびDixon 1998年)。この活性は、抗アポトーシス経路、細胞周期の進行および細胞代謝の増加を含むAktのPIP3活性化により開始される多くの腫瘍形成性のシグナルを下方制御する(SulisおよびParsons 2003年)。癌におけるPTENの役割は、多くの異なる腫瘍タイプにおいて遺伝的なまたは機能的な頻繁な欠損から明らかである(BonneauおよびLongy 2000年)。最初にグリア癌において欠失していることが発見され、以後は前立腺、乳房、子宮内膜、メラノサイト、腎臓および肺の腫瘍形成と関係づけられてきた。また、PTENの生殖系列の変異は、カウデン症候群などの遺伝性の癌素因症候群とも関連した(Eng 2003年)。PTEN欠損のマウスモデルは、多くの異なる組織タイプにおいてヘテロ接合性マウスおよび組織特異的ノックアウトの双方で腫瘍抑制因子としての役割を再現した(Di Cristofano,Pesceら.1998年;Kwabi−Addo,Giriら.2001年;PetrocelliおよびSlingerland 2001年;You,Castrillonら.2002年;Fraser,Zhuら.2004年)。
(i)生体膜を通過してカーゴ分子(cargo molecule)を輸送するための配列番号1に記載の配列の連続したアミノ酸残基22〜173、または配列番号1のアミノ酸残基22〜173の一部分を含むペプチドと、(ii)カーゴ分子とを含む組成物であって、カーゴ分子が配列番号4に記載の配列を有するアミノ酸残基を含むペプチドではない組成物。
第1シリーズの実験
PTEN腫瘍抑制因子は、癌において最も一般的に変更される遺伝子の1つである。ホスファチジルイノシトール3,4,5−三リン酸の脂質ホスファターゼとして機能し、次にホスファチジルイノシトール3−キナーゼ(PI3K)およびAktからの腫瘍形成のシグナル伝達を抑制する。PTEN mRNAを検査することによって、5’非翻訳領域(UTR)が770bpについてPTENのオープンリーディングフレーム(ORF)とインフレームであることが明らかとされた。このUTR ORF内に、標準のAUG開始コドンの513塩基対上流に弱いKozak配列内に代替のCUG開始コドンが存在する。標準のPTEN ORFの発現により約55kDaに移動するタンパク質が産生されるが、5’UTRを含んでいるPTEN cDNAの発現によりPTENロングと称される70kDaの第二のタンパク質を産生することができる。開始部位の変異は、55kDa PTENが標準の開始コドンでの翻訳から産生されるが、他方でPTENロングは上流の代替の開始部位から開始されることを指示する。異なるPTEN抗体での免疫ブロットによって、複数の細胞株にPTENロングが内因性に存在することが実証された。マウスES細胞におけるノックダウンおよびノックアウト研究によって、この大きなタンパク質が実際にPTENであることが確認された。付加されたN末端タンパク質配列がシグナルペプチドおよび切断部位をコードし、PTENロングが分泌経路に進入することを指示している。PTENロングは優先的にレクチンコンカナバリンAと結合し、グリコシル化されることが示される。さらにまた、PTENロングは、PTENに対する抗体とヘパラン硫酸の両方を用いる親和性精製により馴化培地から精製できる。また、PTENロングは、インビボでのプロテアーゼ保護アッセイにおける分解に感受性であるが、正常なPTENは感受性ではなく、PTENロングが細胞膜の外側に局在することを示している。
PTEN mRNAは上流に代替の開始部位を有する
NCBIに寄託されたPTEN mRNA(LiおよびSun 1997年;Steck,Pershouseら.1997年)は、広範囲にわたる5’UTRを含む。5’UTR領域中の約770bpの連続配列は、開始コドンとインフレームである。メチオニンは、この領域でコードされない;しかしながら、標準の開始コドンから−519で始まるいくつかの代替の開始CUGコドンが存在する。この配列の翻訳からscansite(scansite.mit.edu)およびprosite(www.ebi.ac.uk/ppsearch)に従って同定可能なドメインは明らかとされなかった。この全体領域の翻訳によって、173アミノ酸がPTENに加えられ、その分子量を約70キロダルトンに増加させる(図2)。
PTENロングのN末端配列は、膜貫通配列またはシグナルペプチドのいずれかの指標であり得るアラニンの長いストレッチを含む。SignalIP 3.0を用いた翻訳配列の分析によって、高い確率(>95%)で前記配列がシグナルペプチドを含むことが予測された(図5)。シグナルペプチドは、特徴的に塩基性アミノ酸と続く疎水性のストレッチとから構成される。推定上の疎水性の膜貫通ヘリックスは、プロリンにより破壊され、やや極性の配列が続く。また、配列が切断されることも予測され、このタンパク質がERの内腔に放出されることを指摘している。
PTENは、いくつかのプロテオグリカン(例えば、シンデカンおよびグリピカン)と結合し、膜の外側のリーフレット(leaflet)に付着することが見出される。これらのプロテオグリカンは、最もヘパラン化した細胞外分子のうちの2つである(Blero,Zhangら.2005年)。PTENは、以前に高度に陰性に荷電した種に高い親和性を有することが示され、PTENの特性により高度に陰イオン性のPIP3を好むことが導かれる(Das,Dixonら.2003年)。ヘパランは、最も陰性に荷電した生体分子の1つであるため、ヘパランがPTENの細胞外基質への結合を媒介することが可能であった。マウスの肝臓からのタンパク質抽出物を用いて、PTENがヘパランと高親和性に結合することが発見された。さらにまた、ヘパリンアガロースカラムから1M NaClを用いたPTENの連続的な溶出によって、PTENロングも溶出された(図7)。
細胞表面上のPTENロングの存在は、このタンパク質の部分が可溶性であり、細胞環境に放出される可能性は排除しない。ヘパリンセファロースをHEK293細胞で馴化した血清不含培地からPTENロングを親和性精製するために使用した。カラムの溶出によって、培地中に50kDaの分子量に移動するPTENの存在が明らかとされた(図9)。PTENロング特異的な抗体での免疫ブロットは同じ50kDa種を明らかにし、このタンパク質が代替の開始部位から翻訳される配列およびPTENモノクローナル抗体のC末端エピトープの配列を保持することを示している。これは、55kDaであることが観察されたPTENの部分が実際に上流の開始コドンに由来する切断された翻訳産物であることを暗示する。
生理的な分泌物の最良の供給源の1つは血清である。ヘパリンセファロースを用いてヒト血清からPTENを親和性精製した。ヒト血清を遠心分離し、ろ過して粒子状物質を除去した。次に、BC150に1:5に希釈し、広くプロテインA/Gで事前に清澄化してIgGを除去した。血清を少量のヘパリンセファロースを用いてバッチでインキュベーションした。ヘパリンセファロースをlaemmli緩衝剤で溶出し、溶出液をPTENについてブロットし、重鎖の混入を排除するため二次抗体単独でブロットした。PTENおよびPTENロングはともにヒト血清に見出された(図10)。
PTENロングの抗血管形成の役割を以下に示す:(1)PTENロングは、通常、マウスの発生している網膜において弱く発現するが、新生児の発生の間で退行/細胞死を経験している血管に高レベルの発現が認められる(図11);(2)PTENロングは、腫瘍中のアポトーシスしている血管に認められる。さらに、PTENロングで処理された上皮細胞は、トランスフェクトされた細胞から部分的に精製され、細胞の移動を阻害し、アポトーシスを誘導した(図12)。また、精製されたPTENロングは、カスパーゼ−3切断により測定された通り、培養においてU87、HUVEC内皮細胞、または293細胞のアポトーシスの活性化と関連する細胞死も誘導できる。
図13は、PTENロングでのマウスの処理を示す。(A)マウス(n=5)に神経膠芽細胞腫細胞株U87が注射され、異種移植片が乳房の脂肪パッドの2部位(左および右)に形成された。腫瘍移植後、1つの腫瘍にPTENロングを直接注射し、反対側の腫瘍には注射されなかった(w/PTENロング)。また、5匹のマウスの対照セットは、空ベクターでトランスフェクトされた細胞から由来する疑似の精製タンパク質の調製物を注射された(空ベクター)。再び、反対側の腫瘍は注射されなかった(w/空ベクター)。マウスを1〜11日および13〜14日に処理した。示された日にノギスで最大の直径(cm)を測定した。腫瘍容量が≧1cmに達した時、マウスを屠殺した。(B)タンパク質をPTENロング発現ベクターを293細胞にトランスフェクションすることにより調製し、V5親和性樹脂を用いて部分的に精製し、V5ペプチドで溶出した。図14は、14日間のPTENロングの対照注射で処理したマウスの生存率を示す(日数)。
p7のマウス網膜中のPTENロングおよび血管の染色によって、PTENロングがこのポイントでマウスの網膜の血管発生において最初に退行する硝子体管を選択的に染色することが明らかとされた。PTENロングに対する抗体は、エピトープ:N−PRHQQLLPSLSSFFFSHRLPD−C(配列番号3)に対するものである。血管の染色をBS1−レクチンで行った。
PTENロングの精製のための1つの方法において、293細胞をATG/ATG PTENロングでトランスフェクトし、細胞溶解産物をNi+アフィニティーカラムに通過させた。PTENロングをAKTA Purifier上のNi+カラムによりイミダゾール溶出緩衝剤を用いて一貫して精製した。
PTEN(orf 403アミノ酸)またはPTENロングのいずれかで注射前にトランスフェクトされたU87細胞の異種移植片。注射7日後、PTENを過剰発現しているコホート(n=4のうち4)と比較してPTENロングを過剰発現しているコホートにおいて乳房血管の減少が認められた。これはPTENロングが腫瘍環境に影響し得ることを示唆する。
図18および21は、無傷な細胞に適用された場合に、細胞内AKTリン酸化を減少する、PTENではなく、PTENロングの能力を示す。
カーゴ分子は、配列番号1の残基22〜173に記載されている配列を有する連続したアミノ酸残基を含む第2のペプチドにペプチド結合を介して共有結合される。細胞膜にこの組成物を接触させると、カーゴ分子ペプチドは、細胞膜を通過して輸送され、細胞内に送達される。
細胞溶解産物および組織からのPTEN免疫ブロットにおいて二番目のより大きいタンパク質バンドを定期的に観察した。より大きなバンドがPTENであることが確認される証拠には、次の事項が含まれる:大きなタンパク質バンドは、異なるPTENモノクローナル抗体により検出された;細胞がPTENに対するsiRNAで処理されるまたはPTEN遺伝子座がマウスにおいてノックアウトされる場合、大きなタンパク質は存在しない。PTENの5’UTRは、PTENの古典的な開始コドンと700塩基対以上にわたりインフレームであることが観察された。さらに、PTENの522塩基対上流のCUGコドンが存在し、翻訳される場合にPTEN免疫ブロットにおける大きなタンパク質のバンドのサイズを説明することができた。強いコザック配列とは関連しないが、−1シトシンおよび+1グアノシン配列が保持される。翻訳され、PTEN ORFに付加される場合、約70kDaのタンパク質が産生されるはずであり、その分子量は観察された大きなPTENバンドの分子量である。
MSES PTENロングの生成
大腸菌(e.coli)における精製を目的として、PTENロングのオープンリーディングフレームを改変し、最初の21個のアミノ酸を欠失させ、真核生物のシグナルペプチド配列を除き、これを最初の4つのアミノ酸に基づいてMSESバージョンと称した(図28)。このタンパク質は、PTENロングに独特の代替の翻訳領域の153アミノ酸と、PTENロングとPTENの間で共有される403アミノ酸を有し、IPTGを含む誘導性発現ベクターを用いて大腸菌に導入され、ニッケルおよびヘパリン親和性カラムを用いて抽出物から精製された。細胞侵入のための6個のアルギニンリピートの重要性を調べることを目的として、この配列はインフレーム(R6)で欠失させ、このタンパク質を上記の通り精製した。
精製されたMSES PTENロング野生型またはR6突然変異タンパク質は、1マイクログラム/mlの濃度で、培養中の増殖されたMDA−MB−468ヒト細胞の培地に添加された。並行した同一条件下で、細胞はまた、代替の翻訳領域を欠損しているオリジナルの403アミノ酸のPTEN、またはRFPタンパク質のみを発現する細胞から調製される疑似処理を用いて処理された。細胞は、細胞分画のために細胞を単離する前に、37℃にて30分間インキュベートされた。本発明者らは、細胞質および核画分を単離した。分画の有効性は、細胞質および核画分に対して、それぞれ対照のためチューブリンおよびBAF180を用いたウエスタンブロットによって測定された。野生型PTENロングだけが細胞に侵入し、細胞質および核に存在していた。重要なことには、PTENおよびR6について欠失されたPTENロングはいずれも細胞に侵入することができなかった。これらのデータは、R6配列が効率的な細胞侵入に必要とされ、最初の21アミノ酸は必要とされないことを指示する。
上記研究は、図2中の構築物の最初の153アミノ酸が細胞内に別のタンパク質配列を送達するために使用され得ることを示唆した。この質問に答えるために、MSES PTENロング構築物の最初の153アミノ酸は、Pten−Long LeaderについてPLと呼ばれ、ヒトp53の393アミノ酸に融合され、PL−p53融合タンパク質を生成した(図30)。V5−Hisエピトープタグは、精製および検出の目的で使用するためにC末端に融合された。
PL−p53融合タンパク質は、IPTGを用いて大腸菌に導入され、ニッケルおよびヘパリン親和性カラムを用いて細菌溶解物から精製された。PL−p53タンパク質は、MDA−MB−468またはH1299ヒト癌細胞とともに1時間インキュベートし、その後、分画のために細胞を回収し、融合タンパク質が細胞に侵入するかどうかを決定した。増加したp53が、処理されたH1299細胞およびMDA−MB−468細胞の核において検出され得たが、疑似処理されたこれらの細胞では検出されず、PUMAおよびp21(両者ともにp53転写因子の標的である)の発現における増加と関連付けられた(図31)。MDA−MB−468異種移植片の毎日の処理によって、10日間にわたって腫瘍体積が減少したが、RFP(赤色蛍光タンパク質)を発現する大腸菌由来の疑似処理された対照では減少しなかった。
マウスは、細菌発現させたMSES PTENロング(Long)、RFP(赤色蛍光タンパク質疑似対照)、IgG対照、および細胞内への取り込みをブロックする抗PTEN抗体138G6で処理された。PTENロングは他の処理と比べて血糖値を減少させたが、抗PTEN抗体138G6はIgG対照と比較してグルコースを増加させた(図32)。このデータは、PTENロングが血糖値を減少させ得ることを実証している。マウスは、グルコースで処理する前にIP注射された。
