背景技术

在起始密码子AUG处发生的翻译起始之前进行5'UTR的核糖体扫描。尽管核糖体决定适当起始密码子的实际方式仍未完全被理解,但是mRNA本身和指示起始前复合物将在何处减慢扫描并且开始翻译的序列都具有某些特性。典型的“科扎克序列(Kozaksequence)”CCACCATGG(其中加下划线的ATG是起始密码子)已显示为对起始最有利的序列情形(科扎克(Kozak)1991)。mRNA二级结构有可能通过起始前复合物扫描的实际减慢也促进起始,这需要螺旋酶在读取之前溶解二级结构(科扎克(Kozak)1990)。
在某些转录物中,翻译起始可从非AUG密码子开始发生。这通常只包含从转录物翻译的总蛋白的较小部分并且结果是在N末端变化的混合种类的蛋白质。科扎克(Kozak)描述了从非AUG密码子开始的翻译起始效率并且发现了GUG和CUG都能在试管内起始翻译,但效率低得多(科扎克(Kozak)1989)。进一步研究已显示甲硫氨酸的有效性可通过一种尚不明朗的机制来改变翻译起始的混乱性,但是可能涉及由营养敏感性激酶使43S起始前复合物的成分eIF2磷酸化(荷西(Hershey)1991;汉恩(Hann)1994)。
许多蛋白质已显示从替代性的起始密码子开始翻译。转录因子c-myc具有替代性的上游CUG起始密码子,当所述起始密码子被翻译时,添加14个氨基酸到蛋白质的N末端(汉恩(Hann)和以色曼(Eisenman)1984)。这种替代性的同功体(isoform)已显示在伯基特氏淋巴瘤(Burkitt's lymphoma)中选择性地被破坏(汉恩(Hann),金(King)等人1988)。在组织培养中,当甲硫氨酸处于低浓度时,较长形式的myc主要在高细胞密度下转录(汉恩(Hann),斯隆布朗(Sloan-Brown)等人1992)。进一步研究已揭示较长形式的c-myc是生长抑制性的并且具有与典型的c-myc蛋白质不同组的转录目标(汉恩(Hann),迪克斯特(Dixit)等人1994)。(弗洛尔凯维奇(Florkiewicz)和索玛(Sommer)1989)(普拉茨(Prats),克伽德(Kaghad)等人1989。)
另外,已知蛋白质的实际亚细胞定位可由替代性的起始密码子来指示。在小鼠的情况下,从上游CUG密码子起始的原癌基因int-2替代物编码核定位,而AUG密码子编码信号肽以定位到分泌途径(阿克兰德(Acland),狄克逊(Dixon)等人1990)。类似现象在人类FGF3中有描述,其中从AUG翻译的蛋白质以分泌途径为目的地,而从上游CUG翻译的蛋白质定位到细胞核(凯富(Kiefer),艾克兰德(Acland)等人1994)。此外,在一些真核细胞蛋白质,如TEF-1和PRPS-3中,蛋白质完全从CUG密码子起始(泰勒(Taira),艾扎瑟(Iizasa)等人1990;肖(Xiao),大卫德森(Davidson)等人1991)。
以分泌为目的的蛋白质通过称为信号肽的一段疏水性氨基酸而靶向内质网(布罗布(Blobel),沃特(Walter)等人1979)。通常在蛋白质的N末端处发现,信号肽在翻译时结合信号识别颗粒(SRP),并且使核糖体停止并易位到粗面内质网,在所述粗面内质网处核糖体结合SRP受体。一旦核糖体停泊,即释放SRP-SRP受体复合物,并且通过Sec61易位子穿过ER的内腔而重新开始翻译。然后,在可溶性蛋白质从Sec易位子释放蛋白质的情况下,裂解出信号肽。在蛋白质跨越膜的情况下,跨膜螺旋体充当ER易位的信号肽。这些蛋白质在高尔基体中通过糖基化而广泛地被修饰并且穿梭到分泌泡囊中的质膜(艾伯特(Alberts)2002)。
存在许多在癌症中已显示出重要性的分泌型蛋白质。举例来说,Wnt信号传导途径已显示在肺癌中有变化。Wnt是卷曲蛋白受体家族的分泌型配体。Wnt激活卷曲蛋白引起散乱,使得包含APC的□-连环蛋白(catenin)降解复合物离解,从而允许□-连环蛋白的水平上升并且易位到细胞核,在所述细胞核中□-连环蛋白可与TCF转录因子相互作用并且反式激活TCF转录因子。APC中的失活突变和□-连环蛋白中的激活突变在遗传性和偶发性结肠癌中已有详细描述。另外,许多细胞外配体拮抗剂(如SFRP和Wnt-5a)竞争相同的卷曲蛋白受体(如Wnt)。SFRP和Wnt-5a两者都显示是肿瘤抑制剂;SFRP基因剔除小鼠产生淋巴肿瘤并且在黑素瘤中已检测到Wnt-5a的后生沉默。
如本文所揭示,新颖的经差异性翻译的蛋白质的前导序列,即PTEN-long,能够类似于HIV TAT充当细胞穿透肽。
发明内容

从培养基中纯化PTEN肝素-在15cm培养皿中的10%FBS DMEM中使HEK293细胞生长达到汇合。将所述细胞与15ml不含FBS的DMEM一起培育整夜。从20个培养盘中收集培养基并且通过0.45微米过滤器过滤。在4℃下使用4ml/min的流速用AktaPrime(安玛西亚生物科学公司(AmershamBioscience))以DMEM平衡1ml肝素HP柱。随后使条件培养基以1ml/min穿过所述柱。用10体积的BC200(200mM NaCl,50mM Tris pH 7.4,1mM EDTA,0.2%曲拉通X-100)洗涤所述柱。用5ml的1MNaCl以1ml/min洗脱1ml洗脱份中的蛋白质。由280nm下的OD来确定各洗脱份的蛋白质浓度。用20%三氯乙酸使各洗脱份的一半沉淀,用真空干燥的冷丙酮洗涤。在20μl拉米利溶解缓冲液中重构蛋白质并且使用PTEN和PTEN-long的抗体进行免疫墨点点渍。
从血清中纯化PTEN-来自AB血浆的人类血清从希格玛公司(Sigma)获得。通过0.45微米过滤器过滤1ml血清并且使用蛋白质A/G琼脂糖预先清除抗体而用于1小时培育。将肝素-琼脂糖与预先清除的血清以及琼脂糖对照一起培育整夜并且在第二天用BC150(150mM NaCl,25mM Tris pH 7.4,1%NP-40,0.25%脱氧胆酸钠,1mM EDTA)洗涤。用拉米利样品缓冲液洗脱蛋白质并且进行免疫墨点点渍而用于PTEN分析或二次处理(只对于重链污染)。
伴刀豆球蛋白A下拉-用BC500(500mM NaCl,20mM Tris pH 7.4,1%曲拉通X-100,1mM MnCl2,1mM CaCl2,l×蛋白酶抑制剂混合物)溶解次汇合的HEK293细胞。使细胞溶解产物离心并且过滤。在4℃下用20微升伴刀豆球蛋白A琼脂糖(希格玛公司(Sigma))进行下拉,持续1小时。用BC500洗涤树脂并且用拉米利样品缓冲液洗脱蛋白质。
结果
PTEN mRNA具有上游替代性起始部位
存放在NCBI中的PTEN mRNA(李(Li)和孙(Sun)1997;斯泰克(Steck),波斯豪斯(Pershouse)等人1997)含有外延的5'UTR。5'UTR区域中约770bp的相连序列在起始密码子的框内。在此区域中没有甲硫氨酸被编码,但是,有在典型的起始密码子的-519处开始的若干个替代性的起始CUG密码子。根据scansite(scansite.mit.edu)和prosite(www.ebi.ac.uk/ppsearch),此序列的翻译没有揭示可鉴别的结构域。此完整区域的翻译将添加173个氨基酸到PTEN中,从而使它的分子量增加到约70,000道尔顿(图2)。
其他PTEN直系同源物的比对揭示智人UTR的翻译序列可在来自各种物种的PTEN的开放阅读框架中找到。黑猩猩、黄牛、意蜂和秀丽隐杆线虫都含有与智人5'UTR的翻译产物同源的蛋白序列(图3)。此外,智人5'UTR和小家鼠PTEN 5'UTR的比对显示了广泛的核苷酸同源性(未图示)。与智人5'UTR的翻译相比,小家鼠5'UTR对于522个碱基对在典型起始密码子的框内翻译并且揭示了高度同源的蛋白序列(图5.3)。5'UTR的同源性和来源于智人5'UTR的氨基酸序列在其他物种的翻译蛋白质中的实际存在指示了此序列的进化重要性。
PTEN mRNA可从替代性的上游部位起始翻译。PTEN ORF的过度表达产生了55kDa的单个蛋白质色带。包含5'UTR产生了约70kDa的第二个较大蛋白质色带。PTEN免疫墨点中的较大蛋白质色带也内生地存在于许多细胞系中并且可由不同单克隆抗体检测(图4)。此较大蛋白质色带也存在于小鼠野生型ES细胞中并且在PTEN基因剔除小鼠ES细胞中不存在并且在稳定地表达PTEN shRNA的小鼠ES克隆体中减少(图4)。使用siRNA在HEK293细胞中使人类PTEN蛋白的基因表达阻断还引起70kDa蛋白质的基因表达阻断。
在剔除PTEN的PC3细胞系中编码PTEN的ORF的质粒的表达使得55kDa蛋白质产生。当也编码5'UTR的质粒被过度表达时,产生70kDa蛋白质。上游假定的起始密码子从CTG突变为ATG(图1,“ATG/ATG”)主要使免疫墨点模式转变成70kDa形式(图4)。55kDa色带也被确认为通过突变而源于ATG起始密码子(图4,“CTG/ATA”)。
因此,5'UTR足以起始较长形式的PTEN的翻译。因此,5'UTR被称为替代性翻译的区域的5'ATR并且检测到的较大蛋白质被称为“PTEN-long”。
产生针对从5'ATR翻译的氨基酸的经亲和纯化的多克隆抗体并且所述多克隆抗体用于确认过度表达研究中重组PTEN-long的产生,以及HEK293细胞中内生形式的产生(图4)。从HEK293细胞的全细胞溶解产物免疫墨点分析和PC3细胞的过度表达研究中,似乎存在多种形式的PTEN-long,从而指示了潜在的翻译后修饰、无文献记载的剪接形式或甚至是5'ATR中的替代性起始密码子。
PTEN-long编码N末端信号肽
PTEN-long的N末端序列含有一段较长的丙胺酸,所述丙胺酸可指示跨膜序列或信号肽。使用SignalIP 3.0对翻译序列进行的分析以高度可能性(>95%)预测所述序列含有信号肽(图5)。信号肽在特征方面包含碱性氨基酸,接着是疏水段。假定的疏水性跨膜螺旋体被脯氨酸破坏并且接着被略带极性的序列破坏。所述序列也预测被裂解,从而指示蛋白质应释放到ER的内腔中。
分泌蛋白质和细胞外蛋白质的特点之一是添加复合糖部分到高尔基体中,一种称为糖基化的过程。糖可通过N-糖基化而在一致序列N-X-S/T(X不能是脯氨酸)处添加到天冬酰胺中(古帕塔(Gupta)和布鲁纳克(Brunak)2002);丝氨酸、苏氨酸和酪氨酸的羟基组还可以是O-糖基化的目标(尤莱纽斯(Julenius),摩尔加德(Molgaard)等人2005)。PTEN具有多个O-糖基化部位,但是只有一个N-糖基化部位。在下拉分析法中使用结合糖部分的植物凝集素伴刀豆球蛋白A,以判定HEK293细胞中PTEN补体的一部分是否被糖基化。从这些细胞中纯化PTEN-long约为50%的PTEN混合物(图6)。这显示了PTEN-Long被糖基化并且细胞质的55kDa形式的PTEN被糖基化或PTEN-Long在细胞外裂解。
PTEN-long结合类肝素并且在细胞表面上被发现
PTEN结合许多蛋白聚糖,如多配体蛋白聚糖(syndecan)和磷脂酰肌醇蛋白聚糖(glypican),所述蛋白聚糖被发现连接到膜的外叶上。这些蛋白聚糖是两种类肝素化程度最高的细胞外分子(布来诺(Blero),张(Zhang)等人2005)。先前已显示,PTEN对带较高负电荷的物质具有高亲和力,PTEN的这种特性使它偏好高度阴离子性的PIP3(达斯(Das),狄克逊(Dixon)等人2003)。由于类肝素是带最大负电荷的生物分子之一,因此有可能的是,类肝素可介导PTEN与细胞外基质的结合。使用来自小鼠肝脏的蛋白质提取物,发现PTEN以高亲和力结合类肝素。此外,使用1M NaCl对来自肝素琼脂糖柱的PTEN进行连续洗脱,也洗脱PTEN-long(图7)。
粘着于细胞膜外表面上的PTEN-long应对蛋白酶降解敏感。在蛋白酶保护分析法中,将活细胞与蛋白酶一起培育并且只有细胞外蛋白质被降解,这是因为脂质膜对蛋白酶是不可渗透的并且用以保护所有细胞内蛋白质。通过与PBS一起轻轻搅动而从附着培养物中移除HEK293细胞并且使所述细胞与渐增浓度的蛋白酶K一起悬浮。用PMSF使反应停止并且用拉米利缓冲液溶解细胞。PTEN-long显示了对蛋白酶K和E-钙粘蛋白(一种已知细胞外蛋白质)处理的敏感性(图8)。另一方面,PTEN显示适度的蛋白酶敏感性,这指示了55kDa物质的某个部分也在细胞外(因为它被糖基化)或在使细胞质PTEN暴露于蛋白酶K的分析法期间发生了一定程度的细胞溶解。包括使用膜渗透性曲拉通的对照来证实PTEN在暴露于蛋白酶K时可降解。有待了解的是,这是PTEN所特有的还是以55kDa迁移并且保留PTEN抗体的C末端抗原决定基的PTEN-long裂解形式。此资料指示PTEN-long在细胞表面上。
可溶性PTEN-long分泌到培养基中
细胞表面上PTEN-long的存在不排除一部分蛋白质是可溶的并且释放到细胞环境中的可能性。使用肝素琼脂糖从针对HEK293细胞调节的无血清培养基中亲和纯化PTEN-long。柱洗脱揭示培养基中存在以50kDa分子量迁移的PTEN(图9)。使用PTEN-long特异性抗体的免疫墨点分析揭示相同的50kDa物质,从而指示这种蛋白质保留从替代性起始部位翻译的序列和来自PTEN单克隆抗体的C末端抗原决定基的序列。这强有力地暗示了所观察到的55kDa的PTEN部分实际上是从上游起始密码子处开始裂解的翻译产物。
通过在用ATG/ATG构建体转染的HEK293细胞中过度表达PTEN-long来进一步确认PTEN分泌到培养基中。使用这些细胞来产生无血清条件培养基整夜并且使用PTEN单克隆抗体6H2.1使PTEN从1ml培养基中免疫沉淀。较大PTEN色带和较低的55kDa色带成功地从培养基中免疫沉淀。由于蛋白质被过度表达,因此对蛋白质的适当处理可能不会发生,这使得完整大小的70kDa PTEN分泌出。
PTEN在人类血清中被发现
生理分泌物质的最佳来源之一是血清。肝素琼脂糖用于对来自人类血清的PTEN进行亲和纯化。使人类血清短暂离心并且过滤以移除颗粒物质。随后用BC150以1:5稀释所述血清并且用蛋白质A/G充分地预先清除以移除IgG。将所述血清与少量肝素琼脂糖一起分批培育。用拉米利缓冲液洗脱肝素琼脂糖并且对洗脱液进行墨点点渍而用于PTEN分析并且只用于二级抗体处理以排除重链污染。PTEN和PTEN-long都在人类血清中被发现(图10)。
PTEN-long的抗血管生成活性
PTEN-long的抗血管生成作用显示如下:(1)PTEN-long通常在小鼠的发育中的视网膜中较弱地表达但在新生仔发育期间在经历衰退/细胞死亡的血管中有高水平的表达(图11);(2)PTEN-long在肿瘤的凋亡性血管中被发现。此外,用部分地从转染细胞纯化的PTEN-long处理上皮细胞,抑制细胞迁移并且诱导细胞凋亡(图12)。纯化的PTEN-long也可诱导与U87、HUVEC内皮细胞或培养中的293细胞的细胞凋亡的激活相关的细胞死亡,如由卡斯蛋白酶3裂解所测量。
PTEN-long的活体内抗肿瘤和抗血管生成活性
图13显示了用PTEN-long处理小鼠。(A)用成胶质细胞瘤细胞系U87注射小鼠(n=5)以在乳腺脂肪垫的2个部位(左侧和右侧)处形成异种移植物。在肿瘤移植之后,用PTEN-long直接注射一个肿瘤并且不注射对侧肿瘤(w/PTEN-long)。也用来源于空载体转染的细胞的空白纯化蛋白质的制剂(空载体)注射5只小鼠的对照组。再次,不注射对侧肿瘤(w/空载体)。第1-11天和第13-14天处理小鼠。在指定天数用测径器测量最大直径(cm)。当肿瘤体积≥1cm时,处死小鼠。(B)通过将PTEN-long表达载体转染到293细胞中来制备蛋白质并且使用V5亲和树脂部分地纯化,接着用V5肽洗脱。图14显示了用PTEN-long对照注射液处理14天的小鼠的存活分数(以天计)。
视网膜染色
在p7鼠类视网膜中对PTEN-long和血管的染色揭示PTEN-long选择性地染色玻璃体血管,所述玻璃体血管在鼠类视网膜血管发育中的此时刻开始退化。PTEN-long抗体针对以下抗原决定基:N-PRHQQLLPSLSSFFFSHRLPD-C(SEQ ID NO:3)。用BS1-植物凝集素进行血管染色。
纯化
在一种纯化PTEN-long的方法中,用ATG/ATG PTEN-long转染293细胞并且使细胞溶解产物穿过Ni+亲和柱。在使用咪唑洗脱缓冲液的AKTA纯化器上使用Ni+柱一致地纯化PTEN-long。
肿瘤退化
在用PTEN(orf403氨基酸)或PTEN-long注射之前转染U87细胞的异种移植物。在注射后7天,与过度表达PTEN的群组(n=4,总数是4)相比,过度表达PTEN-long的群组中的乳腺血管减少。这表明PTEN-long可影响肿瘤环境。
通过PTEN-long前导序列的跨膜递送
图18和图21显示了PTEN-long(而非PTEN)当应用于完整细胞时减少细胞内AKT磷酸化的能力。

讨论
有规律地观察到来自细胞溶解产物和组织的PTEN免疫墨点中的第二个较大蛋白质色带。确认较大色带是PTEN的证据包括:较大蛋白质色带由不同PTEN单克隆抗体检测到;当用针对PTEN的siRNA处理细胞或在小鼠中基因剔除PTEN基因座时不存在较大蛋白质。观察到PTEN的5'UTR对于超过700个碱基对在PTEN的典型起始密码子的框内。此外,在PTEN上游有CUG密码子522个碱基对,这些碱基对如果被翻译,则可计入PTEN免疫墨点中的较大蛋白质色带的尺寸中。尽管与强科扎克序列无关,但是它保留-1胞嘧啶和+1鸟嘌呤核苷序列。当被翻译并且添加到PTEN ORF中时,应产生约70kDa的蛋白质,这是观察到的较大PTEN色带的分子量。
此序列的翻译已存在于其实际编码序列内的许多PTEN直系同源物中。也检查小鼠5'UTR,因为已观察到小鼠组织溶解产物中的类似色带。小鼠5'UTR核苷酸序列与智人5'UTR高度同源并且类似地在起始密码子的框内。两个潜在的替代性起始密码子存在于-522和-516处,并且从那些部位开始的此序列的翻译揭示氨基酸序列90%以上与智人序列同源。此假定蛋白的保守性是显著的并且显示了对此序列的进化重要性。为了更好地描述此序列,将其重新命名为5'ATR或PTEN的替代性翻译的区域,以描述其翻译潜力。
构建质粒,其中PTEN的开放阅读框架与5'ATR一起克隆,并且将此重组PTEN的表达与单独的典型的产生403个氨基酸的开放阅读框架进行比较。与只含有PTEN的典型ORF的表达质粒(产生以55kDa迁移的单个色带)相比,包含5'ATR会产生以约70kDa迁移的第二个较高PTEN蛋白质色带。较大蛋白质只占翻译的总蛋白质的较小部分;然而,假定起始部位突变为ATG使蛋白质比率向主要的较大形式转变。
来自5'ATR的蛋白序列的保守性指示它不只是进化的人造品。N末端含有一段脂肪族氨基酸,预测所述脂肪族氨基酸为跨膜序列。使用Prosite和Signal 3.0IP预测PTEN-long的N末端是具有紧随其后的蛋白酶裂解部位的信号肽。
活体内蛋白酶保护分析法用于测试PTEN-long是否位于细胞的细胞外表面上。随着细胞外蛋白酶的量逐渐增加,PTEN-long显示不断降解,而PTEN并不如此,从而指示至少一些PTEN-long在细胞外并且至少部分连接到细胞膜的外叶上。考虑牵涉到在细胞膜外叶上的活性脂质磷酸酶,这是最令人感兴趣的结果。结合外膜的蛋白聚糖的两个家族(磷脂酰肌醇蛋白聚糖和多配体蛋白聚糖)先前在PTEN蛋白复合物中识别。
PTEN存在于细胞表面上不排除可溶的分泌型PTEN的可能性。多配体蛋白聚糖和磷脂酰肌醇蛋白聚糖是两种类肝素化程度最高的蛋白聚糖。类肝素是带较高负电荷的葡糖胺聚糖并且PTEN已显示对阴离子具有亲和力,从而部分地解释了选择带较高负电荷的PIP3作为其受质。从小鼠肝脏中纯化PTEN的优化实验揭示PTEN和PTEN-long都可以使用肝素琼脂糖柱来纯化。此外,使用肝素琼脂糖柱,从针对HEK293细胞调节的无血清培养基中纯化约50kDa的蛋白质。由对PTEN具特异性的单克隆抗体和针对PTEN-long中存在的独特氨基酸残基的多克隆抗体来识别纯化蛋白。先前PTEN-long抗体只识别约70kDa的蛋白质色带。两种抗体都可以识别这一个色带的观察结果指示蛋白水解处理可能发生并且通过免疫墨点观察到的蛋白质是保留两种抗体的抗原决定基的PTEN片段。PTEN和PTEN-long也可以使用肝素亲和纯化而从人类血清中纯化。
在过去10年内积累了大量呈现PTEN序列的文献。此处证实PTEN新形式的存在,所述新形式从替代性部位开始翻译并且分泌到外叶和细胞外空间中。
活体内结果显示PTEN-long是新的抗肿瘤化合物,所述化合物通常存在于人类血清中并且具有抗血管生成和促细胞凋亡特性。
第二系列实验
产生MSES PTEN-Long
出于在大肠杆菌中纯化的目的,改变PTEN-Long开放阅读框架以删除前21个氨基酸,从而移除真核信号肽序列,所述真核信号肽序列基于其前四个氨基酸而被称为MSES型式(图28)。在大肠杆菌中使用具有IPTG的可诱导性表达载体来诱导具有PTEN-Long所特有的替代性翻译区域的153个氨基酸和PTEN-Long与PTEN之间共享的403个氨基酸的此蛋白质,并且使用镍和肝素亲和柱从提取物中纯化。出于检查六个精氨酸重复对于细胞进入的重要性的目的,框内删除此序列(R6)并且如上纯化蛋白质。
MSES PTEN-Long进入细胞中
将纯化的MSES PTEN-Long野生型或R6突变型蛋白质加入在培养中以1微克/毫升的浓度生长的MDA-MB-468人类细胞的培养基中。在平行的相同条件下,还用缺少替代性翻译区域的最初403氨基酸PTEN或从只表达RFP蛋白的细胞制备的空白处理试剂来处理细胞。在分离细胞以用于细胞分部分离之前,在37℃下培育细胞30分钟。我们分离细胞质和细胞核部分。通过使用分别控制细胞质和细胞核部分的微管蛋白和BAF180进行西方墨点分析来测量分部分离的有效性。只有野生型PTEN-Long进入细胞中并且存在于细胞质和细胞核中。重要的是,删除R6的PTEN和PTEN-Long都不能进入细胞中。这些资料指示有效的细胞进入需要R6序列,而并不需要初始21个氨基酸。
产生PTEN-Long-p53融合蛋白
以上研究表明图2中的构建体的前153个氨基酸可用于将另一蛋白序列递送到细胞中。为提出这个问题,MSES PTEN-Long构建体的初始153个氨基酸(称为Pten-Long前导子的PL)与人类p53的393个氨基酸融合,以产生PL-p53融合蛋白(图30)。V5-His抗原决定基标签与C末端融合以用于纯化和检测的目的。

-p53进入细胞中,激活基因表达并且抑制肿瘤生长
在大肠杆菌中使用IPTG诱导PL-p53融合蛋白并且使用镍和肝素亲和柱从细菌溶解产物中纯化。在收集细胞用于分部分离之前,将PL-p53蛋白与MDA-MB-468或H1299人类癌细胞一起培育1小时,以判定融合蛋白是否可进入细胞中。可在经处理而非空白处理的H1299细胞和MDA-MB-468细胞的细胞核中检测到增加的p53并且与PUMA和p21(p53转录因子的两个目标)的表达增加相关联(图31)。MDA-MB-468异种移植物的每日处理使得经过10天肿瘤体积减小,但是从表达RFP(红色荧光蛋白)的大肠杆菌制备的空白处理对照并不如此。
用细菌表达的MSES PTEN-Long处理的小鼠的葡萄糖耐量测试
用细菌表达的MSES PTEN-Long(Long)、RFP(红色荧光蛋白空白对照)、IgG对照和阻断吸收到细胞中的抗PTEN抗体138G6处理小鼠。PTEN-long相对于其他处理降低葡萄糖水平,而抗PTEN抗体138G6相对于IgG对照增加葡萄糖(图32)。此资料显示PTEN-long可减少血糖。在用葡萄糖处理之前腹膜内注射小鼠。

Claims (5)
1.一种融合蛋白,该融合蛋白包括在其氨基末端的序列是SEQ ID NO:1的氨基酸22-173序列的连续氨基酸,该连续氨基酸通过肽键结合到序列是非PTEN肿瘤抑制蛋白序列的连续氨基酸上,所述非PTEN肿瘤抑制蛋白选自人类P53、p16、ARF、VHL、SMAD4、ARID1A、BAF180、BRCA1、BRCA2、RB和LKB1组成的组。
2.根据权利要求1所述的融合蛋白,其特征在于,所述非PTEN肿瘤抑制蛋白是人类p53蛋白。
3.一种融合蛋白,其特征在于,其序列如SEQ ID NO:8中所述。
4.如权利要求1或2所述的融合蛋白在制备用于将所述非PTEN肿瘤抑制蛋白递送到细胞中的药物中的用途。
5.如权利要求1-3任意一项所述的融合蛋白在制备用于治疗肿瘤的药物中的用途。
