Abstract

Description
式Aの化合物と式Bの化合物を反応させて式Cの化合物を形成し、
X1はクロロ、ブロモ、トリフルオロメタンスルホネート又はヒドロキシであり、
X2はクロロ、ブロモ又はヨードであり、X1がクロロである場合、X2はブロモ又はヨードであり、X1がブロモである場合、X2はヨードであり、X1がヒドロキシである場合、X2はクロロ、ブロモ又はヨードであり、X1がトリフルオロメタンスルホネートである場合、X2はブロモ又はヨードであり、
X3はボロン酸又はボロン酸エステルであり、
XはCH又はNであり、XがCHである場合、少なくとも1つのR2がピリジルであり、
X4はクロロ、ブロモ、トリフルオロメタンスルホネート又はヒドロキシであり、

式Fの化合物を1−クロロ−3,5−ジブロモベンゼンと反応させて式Gの化合物を形成し、
式Aの化合物と式Bの化合物を反応させて式Cの化合物を形成し、
式Cの化合物及び式Dの化合物の一方を第1のホウ素エステル化試薬と反応させてボロン酸又はボロン酸エステルを生成させ、これを式Cの化合物及び式Dの化合物の他方と反応させて式Eの化合物を形成する工程を含む方法に関する。
X1はクロロ、ブロモ、トリフルオロメタンスルホネート又はヒドロキシであり、
X2はクロロ、ブロモ又はヨードであり、X1がクロロである場合、X2はブロモ又はヨードであり、X1がブロモである場合、X2はヨードであり、X1がヒドロキシである場合、X2はクロロ、ブロモ又はヨードであり、X1がトリフルオロメタンスルホネートである場合、X2はブロモ又はヨードであり、

[定義]
ここで用いる用語「芳香族基」は、少なくとも1つの芳香族部分を有する原子価1以上の原子の配列を示す。少なくとも1つの芳香族部分を有する原子価1以上の原子の配列は、窒素、硫黄、セレン、ケイ素、酸素などのヘテロ原子を含んでも、或いは炭素及び水素のみから構成されてもよい。ここで用いる用語「芳香族基」は、フェニル、ピリジル、フラニル、チエニル、ナフチル、フェニレン及びビフェニル基を含むが、これらに限らない。上記の通り、芳香族基は少なくとも1つの芳香族部分を有する。芳香族部分は常に、(4n+2)つの「非局在化」電子を有する環状構造であり、ここでnは1以上の整数であり、具体的にはフェニル基(n=1)、チエニル基(n=1)、フラニル基(n=1)、ナフチル基(n=2)、アズレニル基(n=2)、アントラセニル基(n=3)などがある。芳香族基は非芳香族成分を含有してもよい。例えば、ベンジル基はフェニル環(芳香族部分)とメチレン基(非芳香族成分)とを含む芳香族基である。同様に、テトラヒドロナフチル基は、芳香族部分(C6H3)が非芳香族成分−(CH2)4−に縮合した芳香族基である。記述の便宜上、用語「芳香族基」はここでは、アルキル基、アルケニル基、アルキニル基、ハロアルキル基、ハロ芳香族基、共役ジエニル基、アルコール基、エーテル基、アルデヒド基、ケトン基、カルボン酸基、アシル基(例えばエステル、アミドなどのカルボン酸誘導体)、アミン基、ニトロ基などの広い範囲の官能基の導入を包含すると定義する。例えば、4−メチルフェニル基は、メチル基を有するC7芳香族基であり、ここでメチル基はアルキル基である官能基である。同様に、2−ニトロフェニル基は、ニトロ基を有するC6芳香族基であり、ここでニトロ基は官能基である。芳香族基はハロゲン化芳香族基を含み、例えば4−トリフルオロメチルフェニル、ヘキサフルオロイソプロピリデンビス(4−フェニ−1−イルオキシ)(即ち、−OPhC(CF3)2PhO−)、4−クロロメチルフェニ−1−イル、3−トリフルオロビニル−2−チエニル、3−トリクロロメチルフェニ−1−イル(即ち、3−CCl3Ph−)、4−(3−ブロモプロピ−1−イル)フェニ−1−イル(即ち、4−BrCH2CH2CH2Ph−)などがある。芳香族基の別の例には、4−アリルオキシフェニ−1−オキシ、4−アミノフェニ−1−イル(即ち、4−H2NPh−)、3−アミノカルボニルフェニ−1−イル(即ち、NH2COPh−)、4−ベンゾイルフェニ−1−イル、ジシアノメチリデンビス(4−フェニ−1−イルオキシ)(即ち、−OPhC(CN)2PhO−)、3−メチルフェニ−1−イル、メチレンビス(4−フェニ−1−イルオキシ)(即ち、−OPhCH2PhO−)、2−エチルフェニ−1−イル、フェニルエテニル、3−ホルミル−2−チエニル、2−ヘキシル−5−フラニル、ヘキサメチレン−1,6−ビス(4−フェニ−1−イルオキシ)(即ち、−OPh(CH2)6PhO−)、4−ヒドロキシメチルフェニ−1−イル(即ち、4−HOCH2Ph−)、4−メルカプトメチルフェニ−1−イル(即ち、4−HSCH2Ph−)、4−メチルチオフェニ−1−イル(即ち、4−CH3SPh−)、3−メトキシフェニ−1−イル、2−メトキシカルボニルフェニ−1−イルオキシ(例えば、メチルサリチル)、2−ニトロメチルフェニ−1−イル(即ち、2−NO2CH2Ph)、3−トリメチルシリルフェニ−1−イル、4−t−ブチルジメチルシリルフェニ−1−イル、4−ビニルフェニ−1−イル、ビニリデンビス(フェニル)などがある。用語「C3〜C10芳香族基」は炭素原子数3個以上10個以下の芳香族基を含む。芳香族基1−イミダゾリル(C3H2N2−)は、C3芳香族基の1例である。ベンジル基(C7H7−)はC7芳香族基の1例である。

式Oの化合物の合成
−3,5−ジイル)ジピリジン(化合物N)、1.31g(5.15mmol、1.0当量)のピナコールジボラン、1.0g(10mmol、2当量)の乾燥KOAc、180mg(0.44mmol、8%)のリガンドPCyBin及び90mg(0.1mmol、2%)のPd2(dba)3を仕込んだ。シュレンク管を排気し、アルゴンで充填することを3回繰り返した。シュレンク管をアルゴン雰囲気下においた。20mLの無水ジオキサンを添加した。その後、シュレンク管をアルゴン雰囲気下110℃で5時間加熱した。アリコート(0.5mL)を取り、濾過し、EtOAcで洗い、濃縮乾固した。このアリコートを1H−NMR分光法により分析したところ、すべての一臭化物が対応するホウ素エステルに転化されたことが明らかになった。

Claims (25)
- 式Aの化合物と式Bの化合物を反応させて式Cの化合物を形成し、
式中、R1、R2及びR3は各々独立にC1〜C20脂肪族基、C3〜C20芳香族基又はC3〜C20脂環式基であり、
X1はクロロ、ブロモ、トリフルオロメタンスルホネート又はヒドロキシであり、
X2はクロロ、ブロモ又はヨードであり、X1がクロロである場合、X2はブロモ又はヨードであり、X1がブロモである場合、X2はヨードであり、X1がヒドロキシである場合、X2はクロロ、ブロモ又はヨードであり、X1がトリフルオロメタンスルホネートである場合、X2はブロモ又はヨードであり、
X3はボロン酸又はボロン酸エステルであり、
XはCH又はNであり、XがCHである場合、少なくとも1つのR2がピリジルであり、
X4はクロロ、ブロモ、トリフルオロメタンスルホネート又はヒドロキシであり、
X5はクロロ、ブロモ又はヨードであり、X4がクロロである場合、X5はブロモ又はヨードであり、X4がブロモである場合、X5はヨードであり、X4がヒドロキシである場合、X5はクロロ、ブロモ又はヨードであり、X4がトリフルオロメタンスルホネートである場合、X5はブロモ又はヨードであり、
a及びcは各々独立に0〜4の範囲の整数であり、
bは0〜3の範囲の整数である。 - さらに、3−(3−ブロモフェニル)ピリジンをピナコールジボランと反応させて式Fの化合物を形成する工程を含む、請求項10記載の方法。
- 式Aの化合物が1−クロロ−3,5−ジブロモベンゼンであり、式Bの化合物が3−ピリジンボロン酸である、請求項15記載の方法。
- さらに、3−(3−ブロモフェニル)ピリジンをピナコールジボランと反応させて式Fの化合物を形成する工程を含む、請求項24記載の方法。
