Description
同じサブタイプ又は異なるサブタイプのインフルエンザウイルスの株と交差反応する中和するモノクローナル抗体を、周期的投与によって作製する方法が本明細書に提供される。また、このような抗体を含む組成物、及びこのような抗体を用いてインフルエンザウイルス疾患を診断、予防、又は治療する方法が本明細書に提供される。
インフルエンザウイルスは、オルトミクソウイルス科のファミリーに属する封入されたRNAウイルスである(Palese及びShaw「オルトミクソウイルス科:ウイルス及びその複製第5版、フィールズのウイルス学(Orthomyxoviridae: The Viruses and Their Replication, 5th ed. Fields' Virology)」(B. N. Fields、D. M. Knipe、及びP. M. 本明細書で使用する場合、用語「約」又は「およそ」は、数と共に使用する場合、引用される数の0.25%、0.5%、1%、5%、又は10%内の任意の数を指す。用語「約」又は「およそ」は、アミノ酸の位置に対する引用で使用する場合、N末端方向又はC末端方向のいずれかにおける、配列における特定のアミノ酸位置又は、アミノ酸位置の5、4、3、2、若しくは1の残基内である任意のアミノ酸を指す。
(5.1 抗体の作製)
インフルエンザウイルス抗原に結合するモノクローナル抗体を作製する方法が本明細書に提供される。このようなモノクローナル抗体は、インフルエンザウイルス粒子上のインフルエンザウイルス抗原、例えば赤血球凝集素に結合し得る。具体的な実施態様において、モノクローナル抗体は、インフルエンザウイルス粒子に結合する。別の具体的な実施態様において、モノクローナル抗体は、当該技術分野で公知の技術、例えば、ELISA、ウェスタンブロット法、FACs、又はビアコアによって評価されるように、非インフルエンザウイルス赤血球凝集素抗原に対するインフルエンザウイルスの1、2、3、又はそれより多くの株によって発現する赤血球凝集素に選択的に結合する。換言すれば、モノクローナル抗体は、当該技術分野で公知の技術、例えば、ELISA、ウェスタンブロット法、FACs、又はビアコアによって評価されるように、非インフルエンザウイルス赤血球凝集素タンパク質よりも高い親和性で、インフルエンザウイルスの1、2、3、又はそれより多くの株によって発現する赤血球凝集素に結合する。
ある実施態様において、非ヒト対象に投与されるインフルエンザウイルスは不活性化される。当業者に公知の技術を用いて、ウイルスを不活性化してよい。一般的な方法は、不活性化のためにホルマリン、熱、又は界面活性剤を用いる。例えば、すべての内容が引用により本明細書に組み込まれている米国特許第6,635,246号を参照されたい。他の方法には、すべての内容が引用により本明細書に組み込まれている米国特許第5,891,705号;大5,106,619号、及び第4,693,981号において説明されたものを含む。
インフルエンザウイルスに由来し又はそれから得られる抗原をコードする核酸は、例えば発現ベクターなど、ベクターの一部として非ヒト対象に投与され得る。加えて、インフルエンザウイルスに由来し又は得られる抗原は、このような抗原をコードする核酸を宿主細胞にトランスフェクトすることによって作製され得、このような核酸はベクターの一部であり得る。具体的な実施態様において、ベクターは、インフルエンザウイルスに由来し又はそれから得られる抗原をコードする核酸の発現を指示できる発現ベクターである。発現ベクターの非限定的な例には、複製欠損レトロウイルス、アデノウイルス、アデノ随伴ウイルス、ニューカッスル病ウイルス、ワクシニアウイルス、及びバキュロウイルスなどのプラスミド及びウイルスベクターを含むが、これらに限定されない。標準的な分子生物学技術を用いて、インフルエンザウイルスに由来し又はそれから得られる抗原をコードする核酸を発現ベクターに導入してよい。
インフルエンザウイルスの抗原的に異なる株に結合しかつ中和する、本明細書に説明される方法に従って作製されるモノクローナル抗体が、本明細書に提供される。具体的な実施態様において、当業者に公知の技術、例えば、結合についてのELISA又はウェスタンブロット法、及び後掲の実施例6に説明されるような微量中和アッセイによって測定されるような、インフルエンザA型ウイルスのH3サブタイプの抗原的に異なる株に結合しかつ中和する、本明細書に説明される方法に従って作製されるモノクローナル抗体が本明細書に提供される。
インビボで延長した(extended)(又は延長した(increased))半減期を有するよう修飾された抗体が、本明細書に提供される。とくに、対象、好ましくは哺乳類、最も好ましくはヒトにおける約3日間〜約180日間(又はそれより多い)、及びいくつかの実施態様において、3日間超、7日間超、10日間超、15日間超、20日間超、25日間超、30日間超、35日間超、40日間超、45日間超、50日間超、少なくとも約60日間、75日間超、90日間超、105日間超、120日間超、135日間超、150日間超、165日間超、又は180日間超の半減期を有する修飾された抗体が本明細書に提供される。

一態様において、本明細書に説明される中和する抗体を投与することによって対象におけるインフルエンザウイルス疾患を予防、管理、及び/又は治療する方法が本明細書に提供される。具体的な実施態様において、対象におけるインフルエンザウイルス疾患を予防又は治療する方法は、対象に、有効量の本明細書に説明される中和する抗体、又はその医薬組成物を投与することを含む。別の実施態様において、対象におけるインフルエンザウイルス疾患を予防、管理、又は治療する方法は、対象に、有効量の本明細書に説明される中和する抗体、又はその医薬組成物及び別の療法を投与することを含む。特定の実施態様において、中和する抗体は、モノクローナル抗体である。具体的な実施態様において、予防、管理、又は治療されるインフルエンザウイルス疾患は、第2群インフルエンザウイルスとして特徴づけられるインフルエンザウイルスによって生じる。別の具体的な実施態様において、予防、管理、又は治療されるインフルエンザウイルス疾患は、H3サブタイプのインフルエンザウイルスとして特徴づけられるインフルエンザウイルスによって生じる。
一実施態様において、本明細書に提供される方法に従って治療又は予防される患者は、未処置の対象であり、すなわち、インフルエンザウイルス感染に起因する疾患を有しない対象、又はインフルエンザウイルス感染に罹ったことがなくかつ現に感染していない対象である。別の実施態様において、本明細書に提供される方法に従って治療又は予防される患者は、インフルエンザウイルス感染を獲得する危険にある対象である。別の実施態様において、本明細書に提供される方法に従って治療又は予防される患者は、インフルエンザウイルス感染を獲得する危険にある未処置の対象である。別の実施態様において、本明細書に提供される方法に従って治療又は予防される患者は、インフルエンザウイルス疾患を罹患している又は罹患することが予期される患者である。別の実施態様において、本明細書に提供される方法に従って治療又は予防される患者は、インフルエンザウイルス感染又はそれに付随する疾患と診断された患者である。いくつかの実施態様において、本明細書に提供される方法に従って治療又は予防される患者は、インフルエンザウイルス疾患のどの症状も顕在化していない、インフルエンザウイルスに感染した患者である。
本明細書に説明される抗体又は組成物は、種々の経路によって対象に送達され得る。これらには、鼻内、気管内、経口、皮内、筋肉内、腹腔内、経皮、静脈内、結膜、及び皮下の経路を含むが、これらに限定されない。また、肺への投与は、例えば、吸入器又はネブライザーの使用、及びスプレーとしての使用のためのエアゾール剤を用いた製剤によって採用することができる。
種々の実施態様において、本明細書に説明される抗体若しくは本明細書に提供される方法に従って作製される抗体、又はこのような抗体をコードする核酸は、1以上の他の療法(例えば、抗ウイルス療法又は免疫調節療法)との併用で対象に投与され得る。いくつかの実施態様において、本明細書に説明される医薬組成物は、1以上の療法との併用で対象に投与され得る。1以上の他の療法は、インフルエンザウイルス疾患の治療若しくは予防において有益であり得、又はインフルエンザウイルス疾患に付随する容態を寛解させ得る。
複数のインフルエンザウイルス株に対する免疫応答を生じることのできる免疫原性組成物(例えば、ワクチン)が本明細書に提供される。操作に関する任意の特定の理論によって拘束されるよう意図するものではないが、本明細書に説明される抗体又は本明細書に説明されるプロセスによって作製される抗体によって結合するインフルエンザ抗原内の抗原結合領域は、複数のインフルエンザ株と交差反応することのできる免疫応答を生じるために、宿主免疫系に1以上の比較的保存された抗原領域を提示するのに有用であると考えられている。1以上の抗原領域が、インフルエンザサブタイプにわたって十分に保存されているので、このような免疫応答は、インフルエンザウイルスのいくつかのサブタイプと交差反応し得る。
本明細書に説明される抗体又は本明細書に提供される方法に従って作製される抗体は、インフルエンザウイルスを検出し及びインフルエンザウイルス感染を検出、診断、又はモニターする診断目的のために用いることができる。具体的な実施態様において、抗体は、特定のインフルエンザウイルスが存在するか又は特定のインフルエンザウイルスサブタイプが生物検体(例えば、痰、鼻汁、他の流体、細胞、又は組織試料)に存在するかを決定するために用いることができる。
(5.8.1 抗体活性を試験するアッセイ)
抗体は、当業者に公知である種々の方法で特徴づけることができる(例えば、ELISA、表面プラズモン共鳴ディスプレイ(ビアコア動態)、ウェスタンブロット法、免疫蛍光検査、免疫染色、及び/又は微量中和アッセイ)。いくつかの実施態様において、抗体は、インフルエンザウイルス抗原(例えば、赤血球凝集素ポリペプチド)、又はインフルエンザウイルスに結合する能力についてアッセイされる。
抗体又はその組成物は、抗ウイルス活性についてインビトロで評価することができる。一実施態様において、抗体又はその組成物は、インフルエンザウイルスの増殖に及ぼすそれらの効果についてインビトロで試験される。インフルエンザウイルスの増殖は、(例えば細胞培養における)当該技術分野で公知であるか又は本明細書に説明される任意の方法によって評価することができる。具体的な実施態様において、細胞は、0.0005及び0.001、0.001及び0.01、0.01及び0.1、0.1及び1、若しくは1及び10の感染効率(MOI)、又は0.0005、0.001、0.005、0.01、0.05、0.1、0.5、1、5若しくは10の感染効率で感染し、本明細書に説明されるモノクローナル抗体又は本明細書に提供される方法に従って作製されるモノクローナル抗体を補充された無血清培地と一緒にインキュベートされる。ウイルス力価は、赤血球凝集素プラーク、又は本明細書に説明される任意の他のウイルスアッセイによって上清中で判定される。ウイルス力価を評価することができる細胞には、MDCK細胞、EFK‐2細胞、Vero細胞、一次ヒト臍静脈内皮細胞(HUVEC)、H292ヒト上皮細胞株、及びHeLa細胞を含むが、これらに限定されない。インビトロアッセイには、当該技術分野で周知であるか又は本明細書に説明される方法を用いて、インビトロの培養細胞における(例えば、プラーク形成で測定されるような)変化したウイルス複製、又は(例えば、ウェスタンブロット分析で測定されるような)ウイルスタンパク質の生成、又は(例えば、RT‐PCR若しくはノーザンブロット分析で測定されるような)ウイルスRNAを測定するものを含む。
抗体又はその組成物への曝露の後の細胞(感染済み又は未感染)又は細胞株の生存度を評価し、それにしたがって抗体又はその組成物の細胞傷害性を測定するために、当該技術分野で周知の多くのアッセイを用いることができる。例えば、細胞増殖は、ブロモデオキシウリジン(BrdU)の取込み(例えば、Hoshinoらの文献(1986、Int. J. Cancer 38, 369);Campanaらの文献(1988, J. Immunol. Meth. 107:79)を参照)、(3H)チミジン取込み(例えば、Chen, J.の文献(1996、Oncogene 13:1395-403);Jeoung, J.の文献(1995、J. Biol. Chem. 270:18367 73)参照)を測定することによって、細胞の直接計数によって、あるいは癌原遺伝子(例えば、fos、myc)などの公知の遺伝子の転写、翻訳若しくは活性の変化、又は細胞周期マーカー(Rb、cdc2、サイクリンA、D1、D2、D3、Eなど)を検出することによってアッセイすることができる。このようなタンパク質及びmRNA並びに活性のレベルは、当該技術分野で周知である任意の方法によって測定することができる。例えば、市販の抗体を含む抗体を用いて、ELISA、ウェスタンブロット法、又は免疫沈降などの公知の免疫診断方法によってタンパク質を定量化することができる。当該技術分野で周知でありかつ常用される方法を用いて、例えば、ノーザン分析、RNアーゼ保護、又は逆転写と連結したポリメラーゼ連鎖反応を用いて、mRNAを定量化することができる。細胞生存度は、トリパンブルー染色又は当該技術分野で公知である他の細胞生死マーカーを用いて評価することができる。具体的な実施態様において、細胞生存度を判定するために、細胞ATPレベルが測定される。
好ましくは、抗体及びその組成物は、ヒトで用いる前に所望の治療的又は予防的活性についてインビボでアッセイされる。例えば、抗体若しくはその組成物及び/又は別の療法を投与することが好ましいかどうかを判定するために、インビボアッセイを用いることができる。例えば、インフルエンザウイルス疾患を予防するための抗体又はその組成物の使用を評価するために、動物をインフルエンザウイルスに感染させる前に抗体又は組成物を投与することができる。あるいは、又は加えて、動物をインフルエンザウイルスに感染させるのと同時に、抗体又はその組成物を動物に投与することができる。インフルエンザウイルス感染又はそれに付随する疾患を治療するための抗体又はその組成物の使用を評価するために、動物をインフルエンザウイルスに感染させた後に抗体又は組成物を投与することができる。具体的な実施態様において、抗体又はその組成物は、動物に2回以上投与される。
一実施態様では、インフルエンザウイルスの複製を調節する抗体又はその組成物が、感染ヒト対象において評価される。この実施態様によると、抗体又はその組成物がヒト対象に投与され、ウイルス複製に及ぼす抗体及び/又は組成物の効果は、例えば生体試料(例えば、血清又は血漿)中のウイルス又はウイルス核酸のレベルを分析することによって判定される。ウイルス複製を変化させる抗体又はその組成物は、対照で処置された対象又は対象群におけるウイルス複製レベルを、抗体又はその組成物で処置された対象又は対象群におけるウイルス複製レベルと比較することによって特定することができる。あるいは、ウイルス複製の変化は、抗体又はその組成物の投与の前後に対象又は対象群におけるウイルス複製レベルを比較することによって特定することができる。生体試料を得て、mRNA又はタンパク質の発現を分析するために、当業者に公知である技術を用いることができる。
本明細書で提供される1以上の抗体など、本明細書に説明される医薬組成物の成分の1以上を充填した1以上の容器を含む医薬パック又はキットが、本明細書で提供される。医薬又は生物製剤の製造、使用、又は販売を規制する政府機関によって規定される形式での通知は、このような容器(複数可)に任意に添付することができ、前記通知は、ヒト投与のために、製造、使用、又は販売の前記機関による認可を反映する。
以下の実施例は、説明のために提示されるものであり、限定のために提示されるものではない。
(6.1.1 プラスミドの調製)
3の抗原的に異なるインフルエンザA型ウイルスH3サブタイプ:A/香港/1/1968(プラスミド1)、A/アラバマ/1/1981(プラスミド2)、及びA/北京/47/1992(プラスミド3)の各々に由来する赤血球凝集素(HA)分子をコードする3のDNAプラスミドを作製した。そうするために、各ウイルス由来のHAのオープンリーディングフレームを親ウイルス株から増幅し、哺乳類細胞におけるタンパク質発現を支持するニワトリアクチンプロモーターを含む哺乳類発現ベクターであるpCAGGS(Niwaらの文献(1991, Gene 108:1993-199))へとクローニングした。
10のC57/BL6マウス及び10のBalb/cマウスを免疫化した。第一の免疫化の3日前に、マウスにふくらはぎの筋肉において0.5%ブピバカインを注射した。各免疫化は、3週間間隔で実施し、以下の免疫化スケジュールに従って、100μLの各プラスミド調製物(プラスミド1〜3)の筋肉(ふくらはぎの筋肉)内注射で行った:

モノクローナル抗体を、当該技術分野で公知のハイブリドーマ技術を用いて作製した(例えば、Harlowらの文献「抗体:実験室マニュアル(Antibodies; A Laboratory Manual)」(Cold Spring Harbor Laboratory Press, 第2版, 1988);Hammerlingらの文献「モノクローナル抗体及びT細胞ハイブリドーマ(Monoclonal Antibodies and T-Cell Hybridomas)」(563 681, Elsevier, N.Y., 1981)参照)。簡潔には、免疫化したマウス由来の脾臓を摘出し、脾細胞を単離した。次に、脾細胞をミエローマ細胞と融合させた。融合後、融合から結果として生じるハイブリドーマを96ウェルプレートに分配した(1のマウスあたり10のプレート、1のウェルあたりおよそ5の生ハイブリドーマ)。融合1週間後、(十分な抗体産生を可能にするために)上清を取り出し、ブロット法によって及びELISAによって、インフル円zなA型ウイルス株A/香港/1/1968との反応性についてスクリーニングした。スクリーニングは、ブロットアッセイ又はELISAのいずれかによって測定されるように、A/香港/1/1968に対する活性を有するモノクローナル細胞集団が得られるまで、陽性ウェルのサブクローニングと交互の反復ラウンドで実施した。
H3サブタイプのインフルエンザA型ウイルスを中和する第6.1.3節において作製されたモノクローナル抗体の能力を、微量中和アッセイを用いて評価した。
H3サブタイプのインフルエンザA型ウイルスを中和する、第6.1.3節において作製されたモノクローナル抗体の能力を、プラーク減少アッセイを用いてさらに評価した。
H3サブタイプのインフルエンザA型ウイルスと宿主細胞との間の融合を阻害する第6.1.3節において作製されたモノクローナル抗体の能力を、赤血球融合アッセイを用いて評価した。
本明細書に提供される交差反応する中和するモノクローナル抗体を作製する方法は、互いに抗原的に異なるH3サブタイプのインフルエンザA型ウイルス株由来の赤血球凝集素と交差反応するモノクローナル抗体の作製を成功裏に生じる。その上、これらの抗体は、複数のアッセイによって判定されるように、試験したH3サブタイプのインフルエンザA型ウイルス株すべてを中和することができる。
インフルエンザA型ウイルスの負荷からマウスを保護する、本明細書に説明された方法に従って作製された交差反応する中和するモノクローナル抗体の能力を、マウスにおける抗体の受動的転移によって評価した。
本明細書に説明される方法に従って作製される交差反応する中和するモノクローナル抗体は、受動免疫をインビボで与えることができる。
本明細書に説明される方法に従って作製される交差反応する中和するモノクローナル抗体の結合領域を決定するために、A/香港/1/1968(H3)赤血球凝集後の異なる断片に融合した緑色蛍光タンパク質(GFP)のコード配列を含む融合タンパク質をコードする核酸コンストラクトを作製した。HA断片をポリメラーゼ連鎖反応によって作製し、その後、pCAGGS-GFPベクターへとクローニングした。種々の核酸コンストラクトによってコードされた融合タンパク質の線図については図8を参照されたい。各核酸コンストラクト(50ng/6ウェル皿)を293T細胞へとリポフェクタミン2000(Invitrogen)を用いてトランスフェクトし、細胞によって発現する融合タンパク質に結合するモノクローナル抗体の能力を、標準的な技術を用いるウェスタンブロット法によって評価した(例えば、Sean Gallagherの文献「タンパク質科学における最新プロトコール(Current Protocols in Protein Science)」(Hoefer Scientific Instruments, San Francisco, California, 1996)参照)。抗GFP抗体をタンパク質発現についての陽性対照として用いた。
(6.4.1 材料及び方法)
(6.4.1.1 動物)
Jackson Laboratory製の6週齢の雌BALB/cマウスをすべての実験に用いた。
ハイブリドーマ上清を、固相酵素免疫検定法(ELISA)による、及びウェスタンブロット法による、反応性についてのモノクローナル抗体のスクリーニングに用いた。他のアッセイについては、受容体破壊酵素を用いて処理した精製済みモノクローナル抗体又は腹水調製物(例えば、Jordanらの文献(J Immunol, 1954;72(3):229-35)参照)を用いた。受容体破壊酵素で処理した腹水を、ELISAアッセイ、微量中和アッセイ、プラーク減少アッセイ、及び融合アッセイによる結合の測定に用いた。抗体を、すでに説明した方法によって精製した(例えば、Hawlow E参照の文献「レーンD 抗体:実験室マニュアル(Lane D. Antibodies: a laboratory manual)」(Cold Spring Harbor, NY:Cold Spring Harbor Laboratory;1988. Xiii, 726p))。アイソタイプにおける差のため、プロテインA‐アガロース(Roche)をモノクローナル抗体7A7及び39A4の精製に用いたのに対し、プロテインG‐アガロース(Roche)をモノクローナル抗体(mAb)12D1の精製に用いた。
6週齢のBALB/cマウスを、pCAGGSプラスミドにおけるインフルエンザウイルス赤血球凝集素のオープンリーディングフレームをコードするDNAコンストラクトで免疫化した(例えば、Baslerらの文献(Proc Natl Acad Sci U S A 98:2746-2751)参照)。個々の免疫化は、筋肉内に3週間間隔で与えられ、100μg DNA含有100μL PBSからなった。免疫化スケジュールにおいて利用される赤血球凝集素を、以下の親ウイルスからクローニングした‐一次免疫:A/香港/1/1968、二次免疫:A/アラバマ/1/1981、三次免疫:A/北京/47/1992のHA。融合3日後、マウスを50μgの精製済みA/ワイオミング/3/2003ウイルスで追加免疫した。B細胞ハイブリドーマを、すでに説明された方法によって作製した(例えば、de StGrothらの文献(J Immunol Methods 35:1-21)参照)。
ハイブリドーマ上清を、A/香港/1/1968ウイルスとの反応性について、ブロット法によって及びELISAによってスクリーニングした。ELISAについては、5μg/mLの精製済みウイルス(ウェルあたり50μL)で被覆したウェルに対する直接的な結合を評価した。ブロットアッセイについては、10μgの精製済みウイルスをニトロセルロースストリップに吸着させ、個々のストリップをハイブリドーマ上清とともにインキュベートした。ELISAアッセイ及びブロットアッセイについては、ウイルスに対する抗体の結合を、ヤギ抗マウスγ鎖セイヨウワサビペルオキシダーゼ二次抗体(SouthernBiotech, Birmingham, Al)を用いて検出した。A/香港/1/1968ウイルスに対するいずれかのアッセイにおいて活性を有するすべてのウェルを反復してサブクローニングし、ハイブリドーマ集団の単クローン性を確実にした。
ブロットを、すでに説明した方法によって作製した(例えば、Towbinらの文献(Proc Natl Acad Sci U S A 1979;76(9):4350-4)参照)。SDS泳動緩衝液を電気泳動に用いた。非還元的ゲル条件のために、試料を、SDSを有するが還元剤を有さない深緩衝液において調製し、煮沸しなかった。
MDCK細胞をウイルスに1の感染効率で感染させ、37℃で6時間インキュベートした。感染済み細胞及び非感染細胞を1μg/mLのモノクローナル抗体とともに室温で1時間インキュベートした。ヤギ抗マウスフルオレセイン抱合体(SouthernBiotech)をモノクローナル抗体結合の検出に用いた。
A/香港/1/1968ウイルス又はA/パナマ/2007/1999ウイルスのHAを発現する2の安定した細胞株を作製した。いずれかの細胞株のHAを発現する偽型ウイルスを、A/WSN/33ウイルス由来の7のセグメント(HAセグメントを除くすべて)及びウミシイタケルシフェラーゼをコードする1のセグメントを有するウイルスによる細胞の感染によって作製した。A/香港/1/1968ウイルス又はA/パナマ/2007/1999ウイルスのHAを発現する偽型ウイルスを中和標的として用いた。ウイルスをモノクローナル抗体とともに室温で30分間、緩徐に揺らしながらインキュベートした。精製済みポリクローナルマウスIgG(Invitrogen)を陰性対照に用いた。次に、ウイルス及びモノクローナル抗体を含む混合物を、MDCK細胞を集密になるまで蒔種した96ウェルプレートのウェルに転移し、37℃で12時間インキュベートした。個々の決定要因を三つ組で実施した。インキュベーション後、細胞溶解液におけるルシフェラーゼ活性を、ウイルス感染の読み出しとして測定した(ウミシイタケルシフェラーゼアッセイシステム、Promega)。
抗体及びウイルス(〜50pfu/ウェル)を室温で30分間緩徐に揺らしながら共インキュベートした。MDCK細胞を蒔種した6のウェルプレートPBSで1回洗浄し、200μLのウイルス及びモノクローナル抗体を各ウェルに添加した後、37℃で20分間インキュベートした。ウイルスをモノクローナル抗体とともに細胞から吸引し、抗体を含む寒天重層物を各ウェルに添加した。プレートを37℃で3日間インキュベートし、プラークをクリスタルバイオレット染色によって計数した。精製済みマウスIgG(Invitrogen)を陰性対照に用いた。
感染の前に、マウスをケタミン(75mg/体重kg)/キシラジン(15mg/体重kg)の腹腔内投与によって麻酔した。6週齢のBALB/cマウスに、10LD50のA/香港/1/1968、A/PR/8/34リアソータントウイルス、又は2700pfuのA/ジョージア/1981ウイルスによる負荷の1時間前、24時間後、又は48時間後のいずれかに30mg/kgモノクローナル抗体を腹腔内で与えた(肺力価実験)。精製済みマウスIgG(Invitrogen)を陰性対照に用いた。ウイルスをPBSに懸濁し、50μLで鼻内投与した(鼻孔あたり25μL)。マウスを毎日体重測定し、開始体重の75%にまで減少した場合、屠殺した。肺力価実験のために、マウス肺をA/ジョージア/1981による感染の4日後に摘出し、肺ホモジネートにおけるウイルス力価をプラークアッセイによって判定した。肺の損傷の組織学的評価のために、肺をA/香港/1/1968‐A/PR/8/34リアソータントウイルスによる感染の4日後に摘出した。組織をパラフィン包埋し、切片をヘマトキシリン及びエオシンで染色した。
マウスモノクローナル抗体を、ニワトリ赤血球及びA/香港/1/1968ウイルスを用いる標準的な赤血球凝集阻害アッセイ(例えば、Cohenらの文献(Virology 1963;20:518-29)参照)において試験した。赤血球融合アッセイのために、ウイルスをニワトリ赤血球(2%赤血球終濃度)とともに氷上で10分間インキュベートした。抗体の希釈物を添加し、試料を氷上で30分間インキュベートした。次に、クエン酸ナトリウム緩衝液(pH4.6)を添加して、最終的なpHを5.0にし、試料を室温で30分間インキュベートした。試料を3000rpmで3分間遠心分離して、赤血球をペレットにした後、上清をELISAプレートに転移し、工学密度測定(340nm)によってNADPH含有量を決定した。NADPHは、融合誘発性赤血球溶解の関数として上清に存在した。
緑色蛍光タンパク質に融合したA/香港/1/68ウイルス赤血球凝集素の切断物をコードするDNAコンストラクトをpCAGGSプラスミドにおいて作製した。すべてのコンストラクトを配列決定及び確認した。次に、293T細胞に、リポフェクタミン2000(Invitrogen, Inc.)を用いてHA-GFP融合遺伝子をコードする種々のpCAGGSをトランスフェクトした。細胞溶解液を4〜20%トリス-HCl SDS‐PAGEゲル(Bio‐Rad Laboratories)で分離し、タンパク質をProtranニトロセルロース膜(Whatman)へとブロットした。GFP及び切断型HA断片を、ウサギ抗GFP(Santa Cruz Biotechnology, Inc)及び抗H3モノクローナル抗体12D1をそれぞれ用いて検出した。二次抗体は、抗ウサギIgG HRP(Dako)及び抗マウスIg HRP(GE Healthcare)であった。
(6.4.2.1 広範に反応する抗H3モノクローナル抗体の単離)
交差反応性抗体の特異性の提供を高めるために、マウスを、およそ10年間隔で生じるH3ウイルス:A/香港/1/1968、A/アラバマ/1/1981、A/北京/47/1992由来の赤血球凝集素をコードするDNAによる連続投与によって免疫化した。融合3日前に、マウスをH3ウイルスA/ワイオミング/3/2003で追加免疫した。ウイルスの追加免疫直後に融合を実施することによって、赤血球凝集素特異的B細胞のみが融合時に脾臓に存在することを確実にした。選択された赤血球凝集素は、数十年間にわたって生じるウイルスに由来し、したがって、複数のH3抗原クラスターを提示した(例えば、Smithらの文献(Science 2004;305(5682):371-6)参照)。融合後、ハイブリドーマ上清を、A/香港/1/1968を結合する能力について、ウェスタンブロット法によって又はELISA法によってスクリーニングし、サブクローニングの連続的なラウンドを、モノクローナルハイブリドーマ集団を単離するまで陽性上清に関して実施した。

抗H3モノクローナル抗体をまず、H3インフルエンザウイルスを中和する能力について微量中和アッセイによって評価した。このアッセイにおいて用いられるウイルスは、ウイルス赤血球凝集素の代わりにホタルルシフェラーゼをコードする遺伝子セグメントを含んでおり;赤血球凝集素は、特にH3赤血球凝集素タンパク質を安定して発現する細胞におけるウイルスの増殖によりウイルス外皮膜上に存在する。A/香港/1968又はA/パナマ/99ウイルスの赤血球凝集素を発現するルシフェラーゼウイルスを作製した。抗H3モノクローナル抗体によるウイルスの中和は、単一周期の複製後のルシフェラーゼ活性に基づいて判定された。微細中和によって、3の抗H3モノクローナル抗体は、A/香港/1968及びA/パナマ/99の両方の赤血球凝集素を中和すると判定された(図12)。
3のモノクローナル抗体を、H3ウイルス感染によって生じた疾患における受動的転移療法としての使用のために、インビボで試験した。マウスに30mg/kgのモノクローナル抗体を、10のマウスLD50リアソータントH3ウイルスによる負荷の1時間前、24時間後、又は48時間後のいずれかに腹腔内で与えた(A/香港/68リアソータントウイルスは、マウス適応性A/PR/8ウイルス由来の6の非赤血球凝集素、非ノイラミニダーゼセグメントを含んでいる。)。マウスを毎日体重測定し、開始体重の75%に到達する場合に屠殺した。モノクローナル抗体12D1による予防的又は治療的のいずれかのマウスの処置は、100%保護的であった。モノクローナル抗体39A4は、予防的処置による有効性について評価され、インビボで同様に100%保護的であった。モノクローナル抗体7A7により予防的に処置されたマウスは、A/香港/68リアソータントウイルスに対して40%しか保護されなかった(図14)。
抗H3モノクローナル抗体によるウイルス中和の機序を決定するために、ニワトリ赤血球のウイルス赤血球凝集を阻害するモノクローナル抗体の能力を検討した。3のモノクローナル抗体のうちのいずれも赤血球阻害活性を有さず、前記モノクローナル抗体が宿主細胞に対するウイルスの結合を妨害することによって作用しないことを示唆した。
12D1又は39A4の特異性を反映する微細な特異性を有する抗体を誘発し得るH3赤血球凝集素の領域の同定を検討した。抗H3モノクローナル抗体12D1又は39A4の存在下でのA/香港/1968ウイルスの16の継代は、結合エピトープの同定を援助し得るエスケープ変異体を生じなかった。プラークアッセイにおいて50μg/mLのモノクローナル抗体12D1又は39A4とのA/香港/1968ウイルスのインキュベーション後に存在する16のプラークの赤血球凝集素を配列決定し、野生型赤血球凝集素からの変化を認めなかった。モノクローナル抗体12D1インフルエンザ疾患に対する保護をインビボで仲介し、かつウェスタンブロット法による変性した赤血球凝集素単量体との反応性によって明らかにされたように、ウイルス赤血球凝集素の連続的なエピトープと反応するので(三量体構造を必要としない。)、12D1結合エピトープに焦点を合わせた。GFPに融合した種々の長さの赤血球凝集素セグメントからなる赤血球凝集素切断突然変異体を作製した。GFP発現を利用して、トランスフェクトした293T細胞におけるコンストラクトの発現を評価した。切断突然変異体の分析により、12D1パラトープが、アミノ酸30〜106の領域におけるHA2サブユニットとの優勢な相互作用をすることが判定された。106〜184切断による結合の損失とともに76〜184切断及び91〜184切断におけるGFP発現が低減しない12D1結合の低減は、12D1結合が、HA2の76〜106領域におけるアミノ酸との接触に依存することを示唆した(図18)。これら30のアミノ酸は、HA2の長いアルファ‐ヘリックスの膜遠位半分内に収まる。12D1パラトープは、ウェスタンブロット法による結合に必要とされない(HA1又はHA2における)この領域の外側のアミノ酸と追加的な接触を有し得る。
インビトロ及びインビボでH3インフルエンザウイルスを広範に中和する抗体を誘発する免疫化スケジュールが開発された。
モノクローナル抗体66A6を、抗体7A7、12D1、及び39A4の作製について用いる第6.1節に説明したのと同じアプローチを用いて作製した。
第6.1.3節に説明されるELISAアプローチを用いて、インフルエンザA型ウイルス株A/香港/1/1968、A/ブリズベーン/10/2007、及びA/パナマ/2007/1999を結合するモノクローナル抗体66A6の能力を評価した。図23に示すように、モノクローナル抗体66A6は、インフルエンザウイルス株の各々を結合した。
インフルエンザA型ウイルス株A/香港/1/1968の融合を阻害するモノクローナル抗体66A6の能力を、第6.1.6節に説明される通り評価した。図24に示すように、モノクローナル抗体66A6は、A/香港/1/1968赤血球凝集素と赤血球との低pH融合を阻害しない。
モノクローナル抗体66A6の結合領域を、第6.3節に説明されたアプローチを用いて決定した。図25に示すように、モノクローナル抗体66A6は、ウェスタンブロット法によって評価されるように、インフルエンザウイルス株A/香港/1/1968(H3)の赤血球凝集素タンパク質のHA1領域に結合する。
プラーク減少アッセイを第6.1.6節に説明されるように用いて、H3サブタイプのインフルエンザA型ウイルスを中和するモノクローナル抗体66A6の能力を評価した。図26に示すように、モノクローナル抗体66A6は、インフルエンザA型ウイルス株A/パナマ/2007/1999(H3)及びA/アラバマ/1/1981(H3)を中和する。
インフルエンザA型ウイルスによる負荷からマウスを保護するモノクローナル抗体66A6の能力を評価した。この方法によると、5の群におけるBALB/cマウスに、20mgの66A6モノクローナル抗体(腹腔内‐第1群)又は対照(PBS‐第2群)を、A/香港/1/1968(H3)由来の赤血球凝集素遺伝子セグメント及びノイラミニダーゼ遺伝子セグメント並びにマウスインフルエンザA型ウイルスA/PR/8/34由来の6の他のインフルエンザウイルス遺伝子セグメント(赤血球凝集素及びノイラミニダーゼではない。)を含む10LD50 X31キメラウイルスによる負荷の1時間前に投与した。マウスの体重を毎日測定し、66A6モノクローナル抗体を投与するマウスの体重を、対照(PBS)を投与するマウスの体重と比較した。
(6.6.1 免疫化及びハイブリドーマ作製)
周期的免疫化戦略を用いて、インフルエンザA型ウイルスの抗原的に異なるH1サブタイプ及びH3サブタイプと交差反応するモノクローナル抗体を作製した(図30)。6週齢のBALB/cマウスを、pCAGGSプラスミドにおけるインフルエンザウイルス赤血球凝集素のオープンリーディングフレームをコードするDNAコンストラクトで免疫化した(例えば、Baslerらの文献(Proc Natl Acad Sci U S A 98:2746-2751)参照)。個々の免疫化は、筋肉内に3週間間隔で与え、100μg DNA含有100μL PBSからなった。免疫化スケジュールにおいて利用される赤血球凝集素を、以下の親ウイルスからクローニングした‐一次免疫:A/香港/1/1968(H3)、二次免疫:A/ソ連/92/77(H1)、三次免疫:A/カリフォルニア/1/88(H3)、四次免疫:A/カリフォルニア/04/09(H1)。最終免疫の3週間後及びハイブリドーマの作製の3日前に、マウスに50μgの精製済みA/ブリズベーン/59/07様(H1)ウイルス及び50μgの精製済みA/ブリズベーン/10/07様(H3)ウイルスを含む組成物を用いて、静脈内に追加免疫した。B細胞ハイブリドーマを、すでに説明した方法によって作製した(例えば、de StGrothらの文献(J Immunol Methods 35:1-21)参照)。
ハイブリドーマ上清を、第6.4.1.5節に説明されるようなELISAによって、A/香港/1/1968ウイルス及びA/カリフォルニア/04/09(H1)との反応性についてスクリーニングした。いずれかの株と反応するハイブリドーマを選択し、それには、抗体1、抗体2、抗体3、及び抗体4を産生するものを含んだ。
MDCK細胞に1又は0.5の感染効率でウイルスを感染させ、37℃で12時間インキュベートし、トリプシンの不在下で固定した。感染済み細胞及び非感染細胞を1μg/mLのモノクローナル抗体とともに室温で1時間インキュベートした。ヤギ抗マウスフルオレセイン抱合体(SouthernBiotech)をモノクローナル抗体結合の検出に用いた。抗体1及び抗体2は、免疫蛍光によって3のH1インフルエンザウイルス由来のHAを認識する(図31)。抗体4は、免疫蛍光によって2のH3インフルエンザウイルス由来のHAを認識する(図31)。
ウェスタンブロットを、すでに説明した方法によって作製した(例えば、Towbinらの文献(Proc Natl Acad Sci U S A 1979;76(9):4350-4)参照)。試料を、SDS及び0.6M DTTを含む負荷緩衝液において100℃で5分間煮沸した。SDS泳動緩衝液を電気泳動に用いた。変性条件及び還元条件下で、抗体1及び抗体3はHAを認識しない(図32)。抗体4は、HAのHA2サブユニットを認識する(図32)。
(6.7.1 免疫化及びハイブリドーマ作製)
周期的な免疫化戦略を用いて、抗原的に異なるインフルエンザA型ウイルスH1サブタイプと交差反応するモノクローナル抗体を作製した(図33)。6週齢のBALB/cマウスにpCAGGSプラスミドにおけるインフルエンザウイルス赤血球凝集素のオープンリーディングフレームをコードするDNAコンストラクトで免疫化した(例えば、Baslerらの文献(Proc Natl Acad Sci U S A 98:2746-2751)参照)。個々の免疫化は、筋肉内に3週間間隔で与え、100μg DNA含有100μL PBSからなった。免疫化スケジュールにおいて利用される赤血球凝集素を、以下の親ウイルスからクローニングした‐一次免疫:A/サウスカロライナ/1918(H1)、二次免疫:A/ソ連/92/77(H1)、三次免疫:A/カリフォルニア/04/09(H1)。最終免疫の3週間後及びハイブリドーマ作製の3日前に、マウスに50μgの精製済みA/ブリズベーン/59/07様(H1)ウイルスを用いて静脈内に追加免疫した。B細胞ハイブリドーマを、すでに説明した方法によって作製した(例えば、de StGrothらの文献(J Immunol Methods 35:1-21)参照)。
ハイブリドーマ上清を、第6.4.1.5節に説明されるように、A/ソ連/92/77(H1)ウイルス及びA/カリフォルニア/04/09(H1)ウイルスとの反応性についてELISAによってスクリーニングした。1のハイブリドーマ由来の起こり得る(potential)クローンと2のH1ウイルスとの反応性を図34に示す。
交差反応する抗H1モノクローナル抗体の結合を評価するために、ウェスタンブロット法を実施することができる。ウェスタンブロットは、当該技術分野で公知の方法によって作製することができる(例えば、Towbinらの文献(Proc Natl Acad Sci U S A 1979;76(9):4350-4)参照)。試料は、SDS及び0.6M DTTを含む負荷緩衝液において100℃で5分間煮沸することができる。SDS泳動緩衝液は、電気泳動に用いることができる。
交差反応する抗H1モノクローナル抗体の結合を評価するために、免疫蛍光法を実施することができる。MDCK細胞に1又は0.5の感染効率でウイルスを感染させ、37℃で12時間インキュベートし、トリプシンの不在下で固定することができる。感染済み細胞及び非感染細胞は、1μg/mLのモノクローナル抗体とともに室温で1時間インキュベートすることができる。ヤギ抗マウスフルオレセイン抱合体(SouthernBiotech)は、モノクローナル抗体結合の検出に用いることができる。次に、ウイルスを結合する、交差反応する抗H1モノクローナル抗体の能力は、免疫蛍光アプローチを用いて評価することができる。
Claims (50)
- 同じか又は異なるサブタイプのインフルエンザA型ウイルス赤血球凝集素(HA)の2以上の株に結合しかつ中和するモノクローナル抗体を作製する方法であって、2、3、4、又はそれより多くの免疫原性組成物を非ヒト対象に、各免疫原性組成物の投与を一定時間離して投与すること、前記対象由来のB細胞ハイブリドーマを作製すること、及び、インフルエンザA型ウイルスHAサブタイプの2以上の株に結合しかつ中和するモノクローナル抗体を産生するハイブリドーマクローンを選択することを含み、この中で、各免疫原性組成物が、不活性型インフルエンザウイルス、弱毒インフルエンザウイルス、弱毒インフルエンザウイルス以外の生インフルエンザウイルス、インフルエンザウイルスに由来し若しくはそれから得られる抗原、又はインフルエンザウイルスに由来し若しくはそれから得られる抗原をコードする核酸を含み、かつこの中で、1の免疫原性組成物は、別の免疫原性組成物とは、前記インフルエンザウイルス、又は、前記HA若しくはその断片又は前記HA若しくはその断片をコードする核酸が由来し又は得られるインフルエンザウイルスが、抗原的に異なるという点において異なる、前記方法。
- 2以上のインフルエンザA型ウイルスHAサブタイプの株に結合しかつ中和するモノクローナル抗体を作製する方法であって、2、3、4、又はそれより多くの免疫原性組成物を非ヒト対象に、各免疫原性組成物の投与を一定時間離して投与すること、前記対象由来のB細胞ハイブリドーマを作製すること、及び、インフルエンザA型ウイルスHAサブタイプの2以上の株に結合しかつ中和するモノクローナル抗体を産生するハイブリドーマクローンを選択することを含み、この中で、各免疫原性組成物が、不活性型インフルエンザウイルス、弱毒インフルエンザウイルス、弱毒インフルエンザウイルス以外の生インフルエンザウイルス、インフルエンザウイルスに由来し若しくはそれから得られる抗原、又はインフルエンザウイルスに由来し若しくはそれから得られる抗原をコードする核酸を含み、かつこの中で、1の免疫原性組成物は、別の免疫原性組成物とは、前記インフルエンザウイルス、又は前記HA若しくはその断片又は前記HA若しくはその断片をコードする核酸が由来し又は得られるインフルエンザウイルスが、抗原的に異なるという点において異なる、前記方法。
- 前記モノクローナル抗体を単離することをさらに含む、請求項1又は2記載の方法。
- 前記2以上のインフルエンザA型ウイルスが、H3サブタイプ、又はサブタイプH1及びH3のものである、請求項1記載の方法。
- 前記インフルエンザA型ウイルスサブタイプがH1及びH3である、請求項2記載の方法。
- インフルエンザA型ウイルスHAサブタイプの2以上の株に結合しかつ中和するモノクローナル抗体を作製する方法であって、(i)非ヒト対象に、第一の不活性型インフルエンザウイルス、第一の弱毒インフルエンザウイルス、弱毒インフルエンザウイルス以外の第一の生インフルエンザウイルス、第一のインフルエンザウイルス若しくはその断片に由来するか又はそれから得られるHA、あるいは第一のインフルエンザウイルス若しくはその断片に由来し又はそれから得られるHAをコードする核酸を含む第一の免疫原性組成物を投与すること;(ii)第一の時間の後、前記対象に、第二の不活性型インフルエンザウイルス、第二の弱毒インフルエンザウイルス、弱毒インフルエンザウイルス以外の第二の生インフルエンザウイルス、第二のインフルエンザウイルス若しくはその断片に由来するか又はそれから得られるHA、あるいは第二のインフルエンザウイルス若しくはその断片に由来し又はそれから得られるHAをコードする核酸を含む第二の免疫原性組成物を投与し、この中で、前記第二のインフルエンザウイルスが前記第一のインフルエンザウイルスとは抗原的に異なること;及び(iii)第二の時間の後、前記対象由来のB細胞ハイブリドーマを作製し、かつインフルエンザA型ウイルスHAの2以上の株に結合しかつ中和するモノクローナル抗体を発現するハイブリドーマクローンを選択することを含む、前記方法。
- インフルエンザA型ウイルスHAサブタイプの2以上の株に結合しかつ中和するモノクローナル抗体を作製する方法であって、(i)非ヒト対象に、第一の不活性型インフルエンザウイルス、第一の弱毒インフルエンザウイルス、弱毒インフルエンザウイルス以外の第一の生インフルエンザウイルス、第一のインフルエンザウイルス若しくはその断片に由来するか又はそれから得られるHA、あるいは第一のインフルエンザウイルス若しくはその断片に由来し又はそれから得られるHAをコードする核酸を含む第一の免疫原性組成物を投与すること;(ii)第一の時間の後、前記対象に、第二の不活性型インフルエンザウイルス、第二の弱毒インフルエンザウイルス、弱毒インフルエンザウイルス以外の第二の生インフルエンザウイルス、第二のインフルエンザウイルス若しくはその断片に由来するか又はそれから得られるHA、あるいは第二のインフルエンザウイルス若しくはその断片に由来し又はそれから得られるHAをコードする核酸を含む第二の免疫原性組成物を投与し、この中で、前記第二のインフルエンザウイルスが前記第一のインフルエンザウイルスとは抗原的に異なること;並びに(iii)第二の時間の後、前記対象に、第三の不活性型インフルエンザウイルス、第三の弱毒インフルエンザウイルス、弱毒インフルエンザウイルス以外の第三の生インフルエンザウイルス、第三のインフルエンザウイルス若しくはその断片に由来し又はそれから得られるHA、あるいは第三のインフルエンザウイルス若しくはその断片に由来し又はそれから得られるHAをコードする核酸を含む第三の免疫原性組成物を投与し、この中で、前記第三のインフルエンザウイルスが、前記第一の及び前記第二のインフルエンザウイルスとは抗原的に異なること;並びに(iv)第三の時間の後、前記対象由来のB細胞ハイブリドーマを作製し、かつインフルエンザA型ウイルスHAサブタイプの2以上の株に結合しかつ中和するモノクローナル抗体を発現するハイブリドーマクローンを選択することを含む、前記方法。
- インフルエンザA型ウイルスHAサブタイプの2以上の株に結合しかつ中和するモノクローナル抗体を作製する方法であって、(i)非ヒト対象に、第一の不活性型インフルエンザウイルス、第一の弱毒インフルエンザウイルス、弱毒インフルエンザウイルス以外の第一の生インフルエンザウイルス、第一のインフルエンザウイルス若しくはその断片に由来するか又はそれから得られるHA、あるいは第一のインフルエンザウイルス若しくはその断片に由来し又はそれから得られるHAをコードする核酸を含む第一の免疫原性組成物を投与すること;(ii)第一の時間の後、前記対象に、第二の不活性型インフルエンザウイルス、第二の弱毒インフルエンザウイルス、弱毒インフルエンザウイルス以外の第二の生インフルエンザウイルス、第二のインフルエンザウイルス若しくはその断片に由来するか又はそれから得られるHA、あるいは第二のインフルエンザウイルス若しくはその断片に由来し又はそれから得られるHAをコードする核酸を含む第二の免疫原性組成物を投与し、この中で、前記第二のインフルエンザウイルスが前記第一のインフルエンザウイルスとは抗原的に異なること;(iii)第二の時間の後、前記対象に、第三の不活性型インフルエンザウイルス、第三の弱毒インフルエンザウイルス、弱毒インフルエンザウイルス以外の第三の生インフルエンザウイルス、第三のインフルエンザウイルス若しくはその断片に由来し又はそれから得られるHA、あるいは第三のインフルエンザウイルス若しくはその断片に由来し又はそれから得られるHAをコードする核酸を含む第三の免疫原性組成物を投与し、この中で、前記第三のインフルエンザウイルスが、前記第一の及び前記第二のインフルエンザウイルスとは抗原的に異なること;(iv)第三の時間の後、前記対象に、第四の不活性型インフルエンザウイルス、第四の弱毒インフルエンザウイルス、弱毒インフルエンザウイルス以外の第四の生インフルエンザウイルス、第四のインフルエンザウイルス若しくはその断片に由来し又はそれから得られるHA、あるいは第四のインフルエンザウイルス若しくはその断片に由来し又はそれから得られるHAをコードする核酸を含む第四の免疫原性組成物を投与し、この中で、前記第四のインフルエンザウイルスが、前記第一の、前記第二の、及び前記第三のインフルエンザウイルスとは抗原的に異なること;並びに(v)第四の時間の後、前記対象由来のB細胞ハイブリドーマを作製し、かつインフルエンザA型ウイルスHAサブタイプの2以上の株に結合しかつ中和するモノクローナル抗体を発現するハイブリドーマクローンを選択することを含む、前記方法。
- 前記第一の時間が2〜4週間又は2〜6週間であり、かつ第二の時間が2〜5日間又は5〜10日間である、請求項6記載の方法。
- 前記第一の及び第二の時間が2〜4週間又は2〜6週間であり、かつ前記第三の時間が2〜5日間又は5〜10日間である、請求項7記載の方法。
- 前記第一、第二、及び第三の時間が2〜4週間又は2〜6週間であり、かつ第四の時間が2〜5日間又は5〜10日間である、請求項8記載の方法。
- 前記HAサブタイプがH3である、請求項6、7、又は8記載の方法。
- 前記第一のインフルエンザウイルスが、A/香港/1/1968型であり、第二のインフルエンザウイルスが、A/アラバマ/1/1981型であり、第三のインフルエンザウイルスが、A/北京/47/1992型であり、かつ第四のインフルエンザウイルスが、A/ワイオミング/3/2003型である、請求項8記載の方法。
- 前記モノクローナル抗体を単離することをさらに含む、請求項6、7、又は8記載の方法。
- 請求項1、6、7、又は8記載の方法によって作製される、同じサブタイプ又は異なるサブタイプのインフルエンザA型ウイルスHAの2以上の株に結合しかつ中和する、単離されたモノクローナル抗体。
- 請求項2記載の方法によって作製される2以上のインフルエンザA型ウイルスHAサブタイプ由来の株に結合しかつ中和する、単離されたモノクローナル抗体。
