本発明は、1つまたはそれ以上のターゲティング部分と1つまたはそれ以上のイメージング部分、診断部分、または治療部分とが取り付けられたスキャフォールドとしてのヒト血清アルブミンドメインIIIを含むコンストラクト、それらの組成物、およびそれらの使用に関する。
この10年間に、コンビナトリアルライブラリー技術は、さまざまなターゲットまたは組織に高い特異性およびアフィニティーで結合するように選択されたペプチド、アプタマーおよび小化学分子を含む数多くの分子をもたらしてきた(Ainaら, 2007;BarbasおよびWhite, 2009;Bembenekら, 2009)。しかし生体内に投与した場合、これらの分子は、一過性の血清内残留と、イメージング応用または治療応用にとって十分なレベルになるまでターゲット部位に蓄積できないこととを特徴とする、最適とはいえない薬物動態(PK)を示すことが多い。この問題に対処するために、それらのターゲティング分子をスキャフォールドに取り付けることができる。最近の総説にいくつかのスキャフォールドが記載されている(GronwallおよびStahl, 2009;NuttallおよびWalsh, 2008)。しかし、それらは非ヒト由来であるか(例えばブドウ球菌プロテインAに由来するアフィボディ(affibody)(Friedmanら, 2007)、ラクダ科動物およびサメの単一ドメイン抗体アイソタイプ(Saerensら, 2008)、植物サイクロチドに由来するシステイン・ノット・ミニタンパク(Simonsenら, 2008))、または制御可能なPKを与えることができない(アンキリン、アドネクチン(adnectin)、アビマー(avimer)、リポカリンおよびアンチカリン(anticalin)(NuttallおよびWalsh, 2008))。

本発明は、HSA-DIIIと、HSA-DIIIにコンジュゲートされた1つまたはそれ以上の小分子ターゲティング剤およびHSA-DIIIにコンジュゲートされたイメージング部分または治療部分の1つまたはそれ以上とを含むコンストラクトの作製における、スキャフォールドとしてのHSA DIIIの使用を提供する。FcRn受容体への結合に影響を及ぼすか、ターゲティング部分、治療部分およびイメージング部分の取り付け部位を追加することになるような、ドメインIII内のアミノ酸配列改変を選択することにより、HSA-DIIIスキャフォールドまたはHSA-DIII担体は、個別に調整された薬物動態(PK)を有するコンストラクトが得られるように改変することができ、また、多価性および/または多重特異性の可能性をもたらし、機能性基を取り付けるための残基も提供する。

「処置する」または「処置」という用語には、
(1)疾患を防止すること、すなわち、その生物(organism)にばく露されているかもしれないが、疾患の症状をまだ経験または呈示していない哺乳動物において、疾患の臨床症状が発生しないようにすること、
(2)疾患を阻害すること、すなわち疾患またはその臨床症状の発生を抑止または低減すること。これには、観察される剥離(detachment)の程度または剥離を有する対象の数もしくは対象が剥離を有するリスクを低減することが含まれる。
(3)疾患を軽減すること、すなわち、疾患またはその臨床症状の退縮を引き起こすこと
が含まれる。
以下の実施例は、本願請求項の発明を例示するために提供するものであって、それを限定するものではない。CEAとドメインIIIスキャフォールドとの融合タンパク質(図8参照)を使って本発明を例示するが、スキャフォールドをターゲティング剤および/またはイメージング剤および治療剤にコンジュゲートする方法は、他にも考えられる。
本発明者らは、がん胎児性抗原(CEA)をターゲットとする詳細に研究された抗体フラグメントと、HSA DIII野生型(WT、非突然変異型)またはH535、H510もしくはH464のアラニン残基への突然変異を組み込んだ3つのHSA DIII変異体の一つとからなる融合タンパク質を調べた。異種移植無胸腺ヌードマウスに124I標識Db-DIIIまたはDbタンパク質を注入し、インビボでDbの血清内残留を調整するHSA DIIIの能力を評価するために、連続小動物PET/CTイメージング研究を行った。加えて、本発明者らは、アルブミンのFcRn結合および循環内残留にとってのH535、H510およびH464残基の相対的重要性に関する結論を引き出すこともできた。
Db-DIIIコンストラクトの作製
テンプレートとして市販のHSA cDNA(OriGene Technologies、メリーランド州ロックビル)を使用し、5'SpeI制限部位と3'EcoRI制限部位を導入するプライマーを使って、ポリメラーゼ連鎖反応(PCR)により、HSA DIII遺伝子を増幅した。プライマー配列は次のとおりとした:

DIIIにおけるヒスチジン残基H535、H510またはH464のアラニンへの突然変異は、Quick-Change突然変異誘発キット(Stratagene、カリフォルニア州ラホーヤ)を適当な突然変異誘発プライマー(フォワードプライマーだけを掲載する)と共に使用することにより、部位特異的突然変異誘発法によって導入した:

完全なDIII(WT、H535A、H510AおよびH464A)遺伝子をpCR2.1-Topoベクター(Invitrogen、カリフォルニア州カールズバッド)にクローニングしてから、抗CEA Db(Wuら, 1999)を既に含有しているpUC18ベクター(New England Biolabs、マサチューセッツ州ベバリー)に移した。そのpUC18ベクターからDb-DIII遺伝子全体を切り出し、pEE12哺乳類発現ベクター(Bebbingtonら, 1992)に、XbaI部位とEcoRI部位を使ってライゲーションした。
NS0マウス骨髄腫細胞(Sigma-Aldrich、ミズーリ州セントルイス)を、5%熱失活透析ウシ胎仔血清(FBS;Omega Scientific Inc.、カリフォルニア州ターザナ)、1%v/vの200mM L-グルタミン(Mediatech, Inc.、バージニア州マナッサス)および1%v/vのペニシリン-ストレプトマイシン(10,000IU/mlペニシリン、10,000μg/mlストレプトマイシン;Mediatech Inc.)を補足した非選択的グルタミン非含有ダルベッコ変法イーグル培地(DME/High Modified;SAFC Biosciences、カンザス州ルネクサ)で維持した。以前に記述されているように(Kenanovaら, 2005)、対数増殖期のNS0細胞1×107個に、SalI(New England Biolabs、マサチューセッツ州イプスウィッチ)による消化で線状化した10μgのpEE12-Db-DIII DNAを、エレクトロポレーションによってトランスフェクトした。
精製Db-DIIIタンパク質を、非還元(NR)条件下および還元(R)条件下のドデシル硫酸ナトリウムポリアクリルアミドゲル電気泳動(SDS-PAGE)、ウェスタンブロット、質量分析ならびにサイズ排除クロマトグラフィーで分析した。タンパク質を還元するために、1Mジチオスレイトール(DTT)を最終濃度が0.2Mになるように加えた。SDS-PAGEには、4-20%勾配のトリス-HClレディーゲル(ready gel)(Bio-Rad Laboratories、カリフォルニア州ハーキュリーズ)で泳動を行い、Instant Blue Coomassieベース溶液(Expedion Protein Solutions、英国ケンブリッジ)で発色させた。ウェスタンブロットにおけるDb-DIIIタンパク質の検出は、APコンジュゲートヤギ抗マウスFab特異的mAb(Sigma-Aldrich)で、ニトロブルーテトラゾリウム(NBT)および5-ブロモ-4-クロロ-3-インドリル-ホスフェート(BCIP)(Promega、ウィスコンシン州マディソン)AP基質を使って達成するか、セイヨウワサビペルオキシダーゼ(HRP)コンジュゲートプロテインL(Sigma-Aldrich)を、4-クロロ-1-ナフトール/3,3'-ジアミノベンジジン(CN/DAB)基質キット(Thermo Scientific、イリノイ州ロックフォート)で呈色させることによって達成した。
精製Db-DIII WT、H535A、H510AおよびH464Aを、以前に記述されたIodogen法(Olafsenら, 2006)を使って、陽電子放射体124I(0.02M NaOH中のヨウ化ナトリウム;IBA Molecular、バージニア州スターリング)で放射性ヨウ素標識した。標識反応(0.114〜0.130ml)は0.1mgの精製タンパク質と12.9〜18.0MBqのNa124Iとを含有した。標識効率は、以前に記述されているように(Olafsenら, 2006)、インスタント薄層クロマトグラフィー(ITLC)により、モノクローナル抗体ITLCストリップキット(Biodex Medical Systems、ニューヨーク州シャーリー)を使って測定した。
フィルタ補正逆投影(FBP)アルゴリズム(Defriseら, 1997)を使って全てのイメージを再構築し、AMIDEソフトウェア(LoeningおよびGambhir, 2003)によって表示した。全てのイメージに同じカラー閾値(color threshold)を適用した。関心領域(ROI;楕円体(ellipsoid)、深さ0.4mm、n=4)を、CEA陽性腫瘍の領域および下半身の低放射能軟組織領域(筋)に描いた。腫瘍対軟組織(T:ST)比を個々のマウスについて決定し、各時点およびコンストラクトについて平均した。各イメージにおいて心臓上にROI(n=4)を描き、入力関数(input function)として注入量(単位MBq)およびシリンダファクター(cylinder factor)(単位MBq/cc/イメージユニット)を入力した後に、血液の%ID/gをAMIDEソフトウェアで計算した。ADAPTIIソフトウェアパッケージを使って、各タンパク質の平均滞留時間(MRT)をその血中放射能曲線から計算した(D'argenioおよびSchumitzky, 1979)。全ての比および%ID/g値についてSEを計算し、図示した(エラーバー)。全てのT:ST ROI比および血中放射能曲線を、それぞれ、対応のないスチューデントのt検定を使って比較し、有意差に調べた。0.05以下の両側p値を統計的に有意と見なした。
Db-DIIIタンパク質の製造および生化学的特徴付け
a.作製、発現および精製
Db-DIIIコンストラクトは約1.4キロ塩基対の長さを有し、XbaI制限部位とEcoRI制限部位に挟まれている(図1A)。工学的に作製されたDb-DIII分子は、ELISAで決定したところ、トランスフェクションされたNS0細胞の終末期培養物(terminal culture)において、10〜16μg/mlの密度で発現した。プロテインLにはDb-DIIIタンパク質を結合する能力があったが、プロテインAによる捕捉の方が効率がよかった。そこで、精製にはプロテインAアフィニティークロマトグラフィーを選択した。Dbは、2つのscFv分子の非共有結合的二量体であるから、各Db分子はそのC末端に結合した2つのDIIIタンパク質を有し、分子質量計算値が約101kDである融合タンパク質をもたらす(図1B)。
精製Db-DIII WTおよび変異体をNR条件下およびR条件下のSDS-PAGEで分析した(図2A)。Db-DIIIタンパク質は、NR条件下で、約101kDaというその予想分子質量に相当する主要バンドをもたらした(図2A、レーン1、2および3)。SDS-PAGEクーマシー染色ゲル(図2A、レーン1、2および3)と、抗マウスFab特異的抗体でプローブしたウェスタンブロット(図2B、レーン2)の両方に、低分子質量の弱いバンドも2本認められた。還元すると、主要バンド[(scFv-DIII)2;101kDa]は、scFv-DIIIフラグメント(約48kDa)とDIII分子(約23kDa)に相当する2本のバンドに分かれた(図2A、レーン5)。融合タンパク質のDIII部分をポリクローナル抗HSA抗体で検出しようという試みは成功しなかったので、ウェスタンブロットではその代わりに、Db-DIIIタンパク質のDb構成要素に結合するHRPコンジュゲートプロテインLを使用した(図2B、レーン3)。上側のバンド[(scFv-DIII)2;101kDa]と真ん中のバンド(scFv-DIII;48kDa)だけが検出された。したがって、還元タンパク質SDS-PAGEゲル上の約23kDaの下側のバンド(図2A、レーン5)は、DIIIドメインのみを表すはずである。精製タンパク質が実際にDb-DIIIであることを確認するために、質量分析を使用した。DbまたはDIIIアミノ酸配列に合致するペプチドがいくつか決定され、タンパク質の実体が確認された(データ未掲載)。
サイズ排除クロマトグラフィーは、Db-DIII WT(101kDa)が単一のピークとして、28.17分の溶出時間で溶出することを示した(図2C)。同じ条件下で、Db-DIII H535A、H510AおよびH464Aタンパク質は、28.2分の平均溶出時間によって特徴付けられ、凝集も多量体化も検出されなかった。サイズ排除クロマトグラフィーピークの積分により、1段階のプロテインAアフィニティーカラム精製後に、約98%のタンパク質純度が明らかになった。
HSAの結晶構造(Sugioら, 1999)に基づいてHSA DIIIの構造モデルを作成した(図3A)。DIIIは、ループで互いに接続された10個のαヘリックス(DIIIa中に6個およびDIIIb中に4個)から構成されている。残基H464(DIIIa中)、H535およびH510A(共にDIIIb中)が描かれている。HSAとFcRnの結晶構造(Martinら, 2001)を使って、DIIIとFcRnのドッキングモデルも作成した(図3B)。ZDOCKアルゴリズムは、FcRn残基H161およびH166(Andersenら, 2006)ならびにHSA DIII H535、H510およびH464を含む相互作用に偏った。これにより11個の候補構造が得られた。潜在的な強いpH依存的結合に関して、PyMOLを使ってこれらの構造を分別し、分析した。分析から得られた総合的所見は、大半の予想構造においてそうであったように、FcRn残基H166およびH161を取り巻く保存された芳香族残基が、DIII H510およびH535残基の2つと接触すると思われるというものだった。FcRn H166およびH161は、低pH環境においてヒスチジンがプロトン化された時にアフィニティーが増加するであろうDIII上のグルタミン酸残基と相互作用するようだった。また、FcRn残基D102とN101は提案された構造の多くにおいて相互作用し、役割を果たしていると思われる。ZDOCKによって与えられた10番目の構造が、強いpH依存的結合を示す可能性が最も高いと思われた。この構造は、DIII H535とFcRn F157;DIII H510とFcRn W51およびY60;DIII H464とFcRn D101およびN102またはK123のどちらか;FcRn H166とDIII E505;ならびにFcRn H161とDIII E531の間の潜在的相互作用を含んでいた。最後に、T84.66ダイアボディの結晶構造(Carmichaelら, 2003)を使って、Db-DIII分子のモデルを作成した(図3C)。2つのDIII分子は、各二量体型ダイアボディに18アミノ酸リンカーを介して取り付けられており、これが比較的フレキシブルな分子構造をもたらすはずである。
Db-DIII融合タンパク質に関する124I標識効率は63.9〜81.5%の範囲にあり、注入された比放射能は13.0〜18.0GBq/μmolだった。連続小動物PETイメージングにより、腫瘍ターゲティングおよび循環における残留に関して、インビボでDb-DIII融合タンパク質とDb単独とを比較することが可能になった(図4)。イメージから、5つのタンパク質は全てLS174T(CEA陽性)腫瘍をターゲットにすることが明らかになった。腫瘍の解剖学的位置はCTイメージ上に明確に見える。ターゲティングは、DbおよびDb-DIII H464Aについては、4時間で早くも認められ、Db-DIII WT、H535AおよびH510A分子については20時間で認められた。CEA陽性腫瘍におけるシグナルが、どのタンパク質についても、全試験期間(51時間)を通して持続したのに対し、バックグラウンド(循環放射能)は変動した。さまざまな時点におけるDb-DIIIとDbタンパク質のT:ST ROI比の統計的比較(図5A)は、4時間および20時間の時点では、全てのタンパク質が互いに有意差のないT:ST比(P>0.05)を示すことを表している。28時間から44〜51時間までは、Db T:ST比が、全てのDb-DIIIタンパク質より有意に大きい状態が続いた(0.04〜0.01の範囲のP値)。51時間時点において、H535A(P=0.03)、H510A(P=0.02)およびH464A(P=0.01)変異体は、軟組織シグナル減弱の速度が速いために、WTと比較して有意に高いT:ST比を示した。しかし、H510A T:ST比にはH535Aとの有意差がなかった(P=0.1)。H464A T:ST比にもH510A T:ST比との有意差がなかった(P=0.07)。H464A T:ST比はH535Aと有意に異なった(P=0.02)。各時点における血中放射能(%ID/g)のPETイメージ定量により、血中放射能曲線(図5B)の作成と、血中での各タンパク質のMRT(表1)の計算が可能になった。Db-DIII WTは、全てのヒスチジン突然変異体およびDb単独と比較して、有意に遅い血中クリアランス速度を示した(P<0.05)。Db-DIII H535AとH510AおよびH510AとH464Aの血中放射能曲線は互いに有意差がなかったが(それぞれP=0.09およびP=0.08)、H535Aは、H464A変異体と比較して有意に長い血中残留を特徴とした(P=0.01)。したがって、血清MRTが最も長いものから最も短いものへの順序は、Db-DIII WT>H535A≧H510A≧H464A>Dbであり、ここで、Db-DIII H535Aは、H464Aと比較して有意に長い循環滞留時間を有する。51時間における体内分布により、血清内残留の順序が確認された(表2)。Db-DIIIタンパク質については血中で測定される放射能が4.0〜1.6%ID/gの範囲であったのに対し、LS174T腫瘍取り込みは、Dbの0.5%ID/gと比較して2.5〜1.3%ID/gだった。以前の研究により、放射性ヨウ素化T84.66 Dbは、注入の2時間後に最大腫瘍取り込み(13.68±1.49%ID/g)に到達し、その後は、腫瘍中の放射能が減弱し始めることが示されている(Wuら, 1999)。腫瘍塊は、LS174T腫瘍とC6腫瘍に関して、それぞれ平均で161mgおよび126mgだった。長い血清中滞留時間は一般に高いLS174T腫瘍取り込みと関連することが認められた。CEA陽性対陰性腫瘍取り込み比は、51時間の時点で、Db単独では13であったのに対して、Db-DIIIタンパク質では1.5〜2.2の範囲にあった。
最初のステップとして、またHSA DIIIが調整可能なPKを有するタンパク質スキャフォールドとして作用しうることの原理証明として、本発明者らは、2つの構成要素からなる融合タンパク質を設計した。1つは、インビボで詳細に研究された小さい二価抗体フラグメントである抗CEA T84.66 Dbとした。この抗CEA Dbは、LS174T(CEA陽性)腫瘍担持マウス(Yazakiら, 2001)において、2.89時間(123I)〜3.04時間(111In)の消失β半減期を示す。このDbは、他のタンパク質(すなわちウミシイタケまたはガウシア・ルシフェラーゼ)との融合にも成功しており、そのインビボ・ターゲティング能を保っていた(Venisnikら, 2007;Venisnikら, 2006)。したがって、Dbは、原理証明研究にとって良いモデルターゲティング分子になる。第2の構成要素は、未知の特徴を有するもの、すなわちHSA DIII WTまたは突然変異したH535、H510もしくはH464残基を有するその変異体の一つである。適正なフォールディングを保証するためにDb-DIII融合タンパク質を哺乳動物細胞で発現させた。発現レベルは妥当であり、アフィニティー精製により、計算値101kDaと合致する分子質量のタンパク質が得られた(図2A)。Dbは、SDS-PAGE条件下では互いに分離されて25kDa付近に移動する2つのscFv分子の非共有結合的二量体である(Wuら, 1999)。DIII内でもscFv内でもシステイン残基は全て対を形成しているので(Curryら, 1998;Dugaiczykら, 1982;Wu, 1999)、本発明者らは、Db-DIII分子が、scFv-DIII(約48kDa)分子種として移動するであろうと予想した。興味深いことに、非還元条件下では、タンパク質の大部分は二量体型[(scFv-DIII)2;101kDa]のままであり、SDS条件下で増加した構造安定性を示した。Db-DIIIコンピュータモデル(図3C)をさらに詳しく調べた後、DbとDIIIの間のリンカー長を18アミノ酸から5アミノ酸に減らした(データ未掲載)。この改変は、タンパク質の移動パターンに影響を及ぼさなかった([(scFv-DIII)2];図2A)。この挙動は予想外だったので、SDS-PAGEゲル(図2A)からDb-DIIIタンパク質バンドを切り出し、抽出されたタンパク質を質量分析で分析した(データ未掲載)。その結果、約101kDaのタンパク質は確かにDb-DIIIであることが確認された。SDS安定性および熱安定性の上昇は、β-グリコシダーゼ(Gentileら, 2002)の場合と同様に、2つのscFv-DIII分子間の極性相互作用、イオン相互作用の結果かもしれない。Db-DIIIタンパク質の分子サイズは、生理的条件下でのサイズ排除クロマトグラフィーによって確認した。Db-DIIIの溶出時間は、同じ条件下で約27.3分(Kenanovaら, 2005)に溶出する類似する分子量の別のタンパク質(scFv-Fc、105kDa)に近い。Db-DIIIはわずかに小さいので28.2分付近に溶出したが、Db単独では38.2分に溶出する(Kenanovaら, 2005)。クロマトグラム上のシングルピーク(図2C)から、高純度であることに加えて、Db-DIIIタンパク質の完全性と、それが単一の分子種として存在することも明らかになった。DIII-FcRnドッキングモデル(図3B)の分析も、H535、H510およびH464残基の考えうるFcRn相互作用パターンを明確にするのに役立ち、FcRn結合にとってのそれらの重要性を明確にすることもできた。ただし、DIIIヒスチジン残基の実際のランキングは、インビボ分子イメージングによって決定した。
Alexa Fluor 647コンジュゲートDIIIタンパク質による結合研究
Alexa Fluor 647タンパク質標識キット(Invitrogen、オレゴン州ユージーン)を製造者の説明書に従って使用することにより、発蛍光団Alexa Fluor 647(1.25kDa)をHSA、DIII WT、H535A、H510AおよびH464Aタンパク質にコンジュゲートした。0.316〜3160nMの範囲の各蛍光タンパク質の希釈液(三つ一組)を、ヒトFcRnを発現するコンフルエントな293ヒト胎児腎臓細胞(Petkovaら, Int Immunol. 2006;18:1759-1769)と共に、丸底96ウェルプレート中、pH6.5でインキュベートした。また、Alexa Fluor 647コンジュゲートHSAの希釈液を、FcRn発現を欠く293細胞と共にインキュベートした(対照反応)。1×PBS(pH6.5)による洗浄ステップ後に、Maestro(商標)インビボ蛍光イメージングシステム(CRi、マサチューセッツ州ウォーバーン)により、ディープ・レッド(Deep Red)(671〜705nm)励起フィルタおよびレッド(Red)(700nmロングパス)蛍光フィルタを使って、細胞をイメージングした。同じサイズの関心領域(ROI)を各ウェルに描き、蛍光シグナルを測定し、各希釈液について平均した。Alexa-Fluor 647コンジュゲートDIIIタンパク質濃度の関数としての平均蛍光を使って結合曲線を作成した。50%蛍光が測定されるDIII濃度により、FcRnに関するDIIIタンパク質の相対的結合アフィニティーが示された(図6参照)。
図7に、発蛍光団コンジュゲートHSAおよびDIIIタンパク質の結合曲線を描く。より左側にシフトした曲線(HSAおよびDIII WT)は、100nM程度の相対的結合アフィニティーで、FcRn発現293細胞への強い結合を表し、続いてDIII H535AおよびH510A(それぞれ約200nMおよび300nM)、そして1μMという最も低い相対的結合アフィニティーを有するDIII H464Aである。Alexa Fluor 647コンジュゲートHSAは(FcRnを欠く)293細胞に結合しなかったことから、FcRnとの特異的相互作用が示唆される。細胞結合研究に基づいて、ヒトFcRnへの結合アフィニティーの順番を高い方から低い方に並べると次のとおりになる:HSA>DIII WT>DIII H535A>DIII H510A>DIII H464A。
HSAおよびDIIIタンパク質に関する131I標識効率は39.6〜93.6%の範囲にあり、注入された比放射能は1.5〜3.1μCi/μgだった。インタクトなHSAと全てのDIIIタンパク質の血中放射能曲線(図10)は、Db-DIII融合タンパク質で観察されたものと同じ排除順序を示す(この図にはDIIIaとDIIIbが付け加えられている)。さらにまた、FcRnに対するHSAおよびDIIIタンパク質の相対的結合アフィニティーの低下(図7)は、循環残留量の減少に比例している。表3に血中半減期の推定値を要約する。血中クリアランスの順序は、遅いものから速いものへと順に、次のとおりである:HSA>DIII WT>DIII H535A>DIII H510A>DIII H464A>DIIIa>DIIIb。最も遅く消失するタンパク質(DIII WT)から最も速く消失するタンパク質(DIIIb)までの緩速相(slow phase)(β)半減期の幅は約2倍であり、t1/2βは15.3〜6.9時間の範囲にある。循環滞留時間のこのスペクトルにより、所望する応用(例えば治療、イメージング)に最も適合しうるDIIIプラットフォームを選択することが可能になる。
アプタマー分子の作製と選ばれたDIIIスキャフォールドへのコンジュゲーション
ヌクレアーゼ耐性ピリミジン2'-フルオロUTPおよび2'F CTPを含有する修飾ターゲット特異的アプタマーは、T7 RNAポリメラーゼプロモーターを担持する二本鎖DNAテンプレートから、ランオフ転写によって作製することができる。転写反応は、Y639F突然変異型T7 RNAポリメラーゼを使って行うことができる。反応に使用されるヌクレオチドは、ATP、GTP、2'F dCTPおよび2'F dUTPからなるであろう。アプタマーをDIIIスキャフォールドにコンジュゲートするために、スクシンイミジル6-ヒドラジノニコチンアミドアセトンヒドラゾン(SANH)を、DIIIスキャフォールドのリジン残基と反応させることができる(下図)。2つの分子間のビス-アリールヒドラゾン結合は354nMでUVトレースが可能であるので、コンジュゲーション比を分光学的に決定することができる。精製後に、コンジュゲートされた生成物の全てを、インビトロ(細胞)でターゲットに結合する能力について評価し、次にインビボ(異種移植マウス)でターゲットに結合する能力について評価することができる。
抗CEAペプチド-DIIIbコンジュゲートの評価:
C末端にHyNic基を有するCEA特異的環状ペプチド(SDWVCEFIKSQWFCNVLASG、Kd=160nM)を業者に委託して合成した(SoluLink)。水溶液中に溶解すると、ペプチドが沈殿した。このペプチドは水への溶解性がないのでそのままではインビボ応用には不適当である。コンジュゲーションに先だって、HyNic修飾ペプチドをジメチルホルムアミド(DMF)有機溶媒に溶解した。SoluLinkによって提供されたプロトコールに従い、精製DIIIbを修飾して、ランダムなリジン残基に4FBを組み込んだ。コンジュゲーション後に、ペプチド/DIII比をA354nmを測定することによって決定したところ、各DIIIb分子につき平均2個のCEA特異的ペプチドであり、このコンジュゲートは水溶液に可溶であった。精製には、Superdex 200カラム(GE Healthcare、ニュージャージー州ピスカタウェイ)によるサイズ排除クロマトグラフィーを使用した。次に、精製された抗CEAペプチド-DIIIbコンジュゲートを124Iで放射性標識し、LS174T(CEA陽性)およびC6(CEA陰性)腫瘍を担持する4匹の無胸腺ヌードマウスに静脈内注入した。小動物PET/CTにより、4、20および27時間の時点で、マウスをイメージングし、その後、マウスを安楽死させて、解剖し、組織/臓器をガンマウェルカウンターでカウントした。下記表4に、注入の27時間後における、1グラムあたりのパーセント注入量計算値(%ID/g)を示す。

- a)ヒト血清アルブミンのドメインIII、ドメインIIIa、もしくはドメインIIIb、またはドメインIII、ドメインIIIa、もしくはドメインIIIbに対して実質的な配列同一性を有するポリペプチドを含む、タンパク質スキャフォールド;
b)タンパク質スキャフォールドに共有結合で連結されているターゲティング部分;および
c)タンパク質スキャフォールドに共有結合で連結されている治療部分またはイメージング部分
を含むコンストラクト。 - ターゲティング部分が、ターゲット組織またはターゲット細胞の受容体に結合するリガンドである、請求項1に記載のコンストラクト。
- ターゲティング部分が、腫瘍特異的抗原に結合する抗体または免疫学的に活性なそのフラグメントである、請求項1に記載のコンストラクト。
