B−(L−E)k
[式中、Bは、結合部分であり;Lは、切断可能な結合であり;Eは、分子タグである]により表すことができる。ホモジニアスアッセイでは、切断可能な結合Lが、易酸化結合であり得、より好ましくは、それが一重項酸素により切断され得る結合である。部分「−(L−E)k」は、単一の結合化合物が、切断可能な結合を介して複数の分子タグを結合させ得ることを示す。一態様では、kが1以上の整数であるが、他の実施形態では、kが数百を超える、例えば、100〜500の場合もあり、または、kが数百を超えて最大数千もの数である、例えば、500〜5000である。通常、複数の異なる種類の結合化合物の各々は、異なる分子タグEを有する。切断可能な結合、例えば、易酸化結合と、分子タグEとは、通常の化学反応によりBに結合している。
様々なレベルのHER3タンパク質を発現している安定な細胞系の生成
全長HER3(NM_001982.2)に対する市販のcDNA(Origene Technologies,Inc.)を制限酵素Not IおよびXba Iで消化し、得られた断片をpcDNA.3 1 Zeocin選択発現ベクターにサブクローニングした。得られたプラスミドを細菌に導入して形質転換し、スクリーニングして正しい挿入を検証した。陽性クローンを配列検証し、細菌で増殖させ、プラスミドをQiagen Maxi−prepキットを使用して精製した。ヒト胎児腎臓細胞(HEK−293)をAmerican Type Culture Collectionから購入し、5%CO2中、37℃において、10%FBS、1×ペニシリン−ストレプトマイシン(100Xは10,000U/mlのペニシリン−Gと10,000μg/mlのストレプトマイシンである)、およびGlutamax(GIBCO)を補充したDMEMで維持した。トランスフェクトするより前の日に、細胞をおよそ25〜30%集密に分割し、ペニシリン−ストレプトマイシンを含まない培地で一晩インキュベートした。次いで細胞を、製造者の説明書に従ってFugene HD(Roche)でトランスフェクトした。次の日に培地を新鮮な完全培地と交換し、細胞を完全培地で48時間インキュベートした後、400mg/mLのZeocin(Invitrogen)を加えた。Zeocinの濃度は、トランスフェクトされたプラスミドを含まない野生型293細胞に対して様々な濃度のZeocinを使用して死滅曲線を実行することによって決定した。クローニングリングを使用しておよそ16個のZeocin耐性クローンを単離し、液体窒素で凍結させた。元のクローンのサブセットを首尾よく増殖させることができ、
HER3特異的ELISAキット(R&D Systems,Inc)を使用して、製造者の説明書に従って試験し、過剰発現しているHER3を検証した(図1)。ELISAによって実証された、HER3を高度に発現している1つのクローン(293−H3クローン1)を、最適化アッセイに使用するための対照として選択した。
HER3に対する抗体の生成およびスクリーニング
HER3の細胞質末端に対してエピトープ特異性を持つHER3特異的なモノクローナル抗体(Ab−6)をLabVisionから購入した。VERATAG(登録商標)レポーター(Pro11)およびストレプトアビジンをコンジュゲートしたメチレンブルー(「分子はさみ」)を以前記載されたプロトコールに従って合成し、精製した(例えば、任意の図を含め、上記、および本明細書に参照により組み込まれている米国特許第7,105,308号を参照されたい)。抗体−VERATAG(登録商標)コンジュゲートおよび抗体−ビオチンコンジュゲート、すなわち、Ab6−Pro11およびB9A11−ビオチンを、sulfo−NHS−LC−LC−ビオチン(Pierce)を製造者のプロトコールに従ってリンカーとして使用して作製し、コンジュゲート産物をHPLC(Agilent)によって精製した。一連の所有の抗体を以下の通り生成した。固定した293クローン13細胞またはHER3タンパク質のC末端領域に含有される種々のエピトープを提示している一連のペプチドでマウスを免疫した(図2A)。免疫の期間中、各マウスに、4週間にわたって数回の免疫を受けさせた。ハイブリドーマを作出し、限界希釈物を用いてクローンを単離した。CellSpot(商標)またはELISAスクリーニングを含めた一連のアッセイによって個々のクローンからの馴化培地をプロファイリングし、その後目盛アップし、次いで免疫組織化学的検査(IHC)によってさらにスクリーニングし(図2B)、その後、最終的に化学物質遊離法(メチレンブルー)(図2C)または二重抗体光遊離法(図2D)のいずれかを用いたVERATAG(登録商標)技術によってスクリーニングした。これらの方法を使用してうまく機能したいくつかの抗体を、図2Aに記載している。とりわけ1つの抗体、B9A11は最も強力であり、最大のダイナミックレンジと感受性を示し、実施例5において下記されているアッセイの最終フォーマットの開発へ進展させた。
FACSおよびELISAによる、様々なレベルのHER3を発現している細胞系パネルからのブロックおよびFFPE切片の生成
HER3タンパク質の発現が様々である3つの細胞系、MDA−MB−453、MDA−MB−468およびSKOV−3を、American Type Cell Culture Collectionから購入した。MDA−MB−453およびMDA−MB−468細胞系を、37℃、5%CO2において、ダルベッコ改変イーグル培地(Dulbecco’s modified Eagle medium(DMEM))、10%FBS、1×ペニシリン−ストレプトマイシンおよび1×Glutamaxで維持した。SKOV3細胞を、37℃、5%CO2において、10%FBS、1×ペニシリン−ストレプトマイシンおよび1×Glutamax(GIBCO)を補充したマッコイ5a培地(McCoy’s 5a Media)で維持した。293−H3クローン1細胞を、37℃、5%CO2において10%FBS、1×ペニシリン−ストレプトマイシンおよび1×Glutamaxを補充したDMEMで維持した。各細胞系に対して少なくとも10個の500mmの培養皿において、細胞をほぼ集密になるまで増殖させた。培地を取り除いた後、細胞を冷たい1×PBSで1回洗浄し、各皿に1×Pen−fix(Thermo Scientific)15mLを加えた。細胞を掻き取り細胞溶液を4℃で一晩(>16時間)固定した。一晩固定した後、細胞を3200×gで15分間遠心分離した。細胞ペレットをゴム製のOリングに移し、濾紙で包み、処理カセットに置いた。処理するために自動Tissue−Tek処理装置を使用した。簡単に記載すると、細胞ペレットを、濃度を次第に上昇させたアルコール、Clear−rite(キシレン代替品)、そしてパラフィンに曝露させた。処理後、パラフィン包埋ステーション(paraffin embedding station)を使用してペレットをブロックに包埋した。細胞ペレットを処理するために使用したすべての溶媒は、Richard−Allen Scientificから入手した。以下の方法によってフローサイトメトリーを用いて、HER3受容体数について同じロットの細胞の割合を試験した。市販のELISAキットを使用し、製造者の推奨に従うことにより(Human ErbB3−DuoSet ELISA;R&D systems)、HER3受容体のレベルを定量化するために細胞の調製物から全細胞溶解物を調製した。ミクロトーム(LEICA)を用いて厚さ7μmの切片を切り取り、正電荷を帯びたガラススライド(VWR)に置いた。スライドを30分間風乾し、次いで70℃に設定した加熱したオーブンで1.5時間焼いた。すべての試料スライドを将来のアッセイのために4℃で貯蔵した。ELISA、フローサイトメトリー、IHCおよびVERATAG(登録商標)比較の結果を図3に示す。
市販の乳癌FFPE切片からの切片の生成
FFPE乳癌ブロックをAsterandから購入した。ミクロトーム(LEICA)を用いて厚さ5μmの切片を切り取り、正電荷を帯びたガラススライド(VWR)に置いた。切片を30分間風乾し、次いでオーブンで、70℃で1.5時間焼いた。すべての試料スライドを将来のアッセイのために4℃で貯蔵した。臨床材料から以前切片化された乳腺腫瘍もこれらの試験に使用した。ガラススライドでH&E染色した腫瘍の例を図4に示す。
ホルマリン固定パラフィン包埋(FFPE)された細胞系および乳房組織における、Her−3 VERATAG(登録商標)アッセイ
FFPE試料を、一連の溶媒を使用して脱パラフィン/再水和させた。簡単に記載すると、スライドをキシレン(2×、5分)、100%エタノール(2×、5分)、70%エタノール(2×、5分)および脱イオン水(2×、5分)に順次浸漬した。再水和した試料の熱誘導性エピトープ回復(Heat−induced epitope retrieval)を、圧力釜(Biocare)を使用して、1×DAKO(pH9.0)(Lab Vision)250mLを含有するスライドホルダーで行った。室温で30分間冷却した後、スライドを脱イオン水で1回すすいだ。疎水性ペン(Zymed)を使用してスライドに疎水性の円を描いて試薬をスライド上に保持した。次いで、1×PBS中1%マウス血清、1.5%BSAおよびプロテアーゼ阻害剤とホスファターゼ阻害剤のカクテル(Roche)を含有するブロッキングバッファーで試料を1時間ブロッキングした。吸引してブロッキングバッファーを取り除いた後、ブロッキングバッファーで調製した、VERATAG(登録商標)をコンジュゲートした抗体(Ab−6:LabVision、1μg/mL)およびビオチンをコンジュゲートした抗体(B9A11;Monogram proprietary、2μg/mL)の混合物を加え、結合反応物を加湿チャンバー内、4℃で振とうしながら一晩インキュベートした。抗体混合物を吸引し、試料を、1×PBS中0.25%TritonX−100を含有する洗浄バッファーで洗浄し、ストレプトアビジンをコンジュゲートしたメチレンブルーを1×PBS中、2.5μg/mLの濃度で加えた。室温で1時間インキュベートした後、過剰のストレプトアビジン−メチレンブルー試薬を吸引し、試料を洗浄バッファーで1回洗浄した後、脱イオン水を3回交換して洗浄した。0.01×PBS中、3pMのフルオレセインおよび2種のCE内部マーカー(MFおよびML)を含有するイルミネーションバッファー(Illumination buffer)を試料切片に加えた。電子冷却ブロック(Torrey Pine Scientific)を備えた自家LEDアレイイルミネーターを使用して、結合したVERATAG(登録商標)を約4℃で光活性化切断によって遊離させた。照射後、水酸化ホウ素ナトリウムを加えることによってVERATAG(登録商標)中間体を還元して、定量化可能な形態にする。遊離したVERATAG(登録商標)レポーターを含有するCE試料を上記スライド上の組織切片上から回収し、CE試料における遊離したVERATAG(登録商標)レポーターを、30℃で6kV、50秒のCE注入条件下、ABI3130 CE機器(22cmのキャピラリーアレイ;Applied Biosystems)で分離し、検出した。臨床検査室における一般的なH3Tアッセイのワークフローを図5に示す。
CEピークの分析、腫瘍面積の正規化およびバッチの正規化
VERATAG(登録商標)Informerソフトウェアを使用してVERATAG(登録商標)の同定および定量化を行った(例えば、米国特許出願公開第2007−0203408−A1号を参照されたい)。未加工のCE電気泳動図においてVERATAG(登録商標)シグナルを分析するために、2種のCE内部マーカー、MF(最初のマーカー)およびML(最後のマーカー)を使用して、VERATAG(登録商標)ピークを、それらの電気泳動移動度、または2種のマーカーに対する移動時間t、すなわち[t(VERATAG(登録商標))−t(MF)]/[t(ML)−t(MF)]に従って同定した。次いで、同定されたVERATAG(登録商標)ピークを、各VERATAG(登録商標)についてピーク面積を算出することによって定量化した。組織切片からのVERATAG(登録商標)回収におけるばらつき、およびキャピラリーアレイにわたるCE注入効率および/または検出感度における処理のばらつきを補正するために、フルオレセイン(3pM)をイルミネーションバッファーに含め、各試料の処理における内部参照対照として一緒に電気泳動した。次いで、各VERATAG(登録商標)ピークの面積を、VERATAG(登録商標)ピーク(VERATAG(登録商標)ピーク面積)を内部フルオレセインピーク(フルオレセインピーク面積/3pMに対して面積正規化することによってRFUまたはRPAとして報告した。これは、可変腫瘍試料に対するRPA*IB 容積/TA(=相対ピーク面積×試料切片にローディングされたイルミネーションバッファーの容積(IB)÷腫瘍面積mm2(RPA*IB 容積/TA=pモル/L*L/mm2=pモル/mm2)として定量化される。詳細には、VERATAG(登録商標)レポーターのCE蛍光シグナル強度、またはピーク面積(PAVeraTag(登録商標))は、時間に対して積分した相対蛍光単位(シグナルの高さ)で得られる(RFU−S/S)。相対ピーク面積(RPAVeraTag(登録商標))は、VERATAG(登録商標)ピーク面積(PAVeraTag(登録商標))を既知濃度の内部フルオレセイン標準(PAF)に関して正規化することによって測定することができ、したがって、測定される検体の最初の濃度に比例する。VERATAG(登録商標)アッセイのシグナルは腫瘍含量に比例する定量的な読み出し情報であるので、臨床試料にわたってVERATAG(登録商標)アッセイシグナルを正確に比較するには、このパラメータにおける差異を調整する必要がある。したがって、試料のVERATAG(登録商標)アッセイシグナルデータ収集に対して腫瘍含量を測定し、その腫瘍含量を使用してVERATAG(登録商標)アッセイの結果を正規化する。腫瘍試料が非常に小さいまたは非常に大きい場合、反応体積(すなわち、抗体、ストレプトアビジンをコンジュゲートしたメチレンブルーおよびイルミネーションバッファー試薬の体積)を調整し、この調整を腫瘍含量の正規化に反映させることに注意するべきである。移動時間、フルオレセイン、イルミネーションバッファーおよび腫瘍面積についてVERATAG(登録商標)ピーク面積を調整した後、対照および試料を、それらの調整されたピーク面積にそれぞれの算出されたバッチ正規化係数(BNF)を掛けることによって正規化する。調整されたRPA値のそれぞれに、それぞれのBNFを掛けて正規化RPA値を得る。この正規化RPA値は定義上単位をもたないので、その値はVERATAG(登録商標)単位で示される。所与の試料についての報告できる(役に立ち、飽和していない)正規化された値すべてを平均してその試料についての最終的な値を決定する。
ダイナミックレンジを増大させるための抗体濃度の最適化
Ab−6抗体およびB9A11抗体の最適濃度を、アッセイのダイナミックレンジ全体にわたる細胞系パネル(293H3−クローン1、MDA−MB−453、MDA−MB−468およびSKOV3)に対するVERATAG(登録商標)HER3トータルアッセイ(上記)における最終濃度を変化させることによって決定した。次いで、これらの結果を、同じ細胞系のFFPEブロック調製物からのHER3のフローサイトメトリーおよびELISAの結果に基づいて予想される倍率変化と比較した。2:1のB9A11対Ab−6の比を丸で囲んだ図6に示した通り、同じ細胞系セットにおけるELISAおよびフローサイトメトリーと比較して予想される倍率変化と比較したHER3の正確な検出に基づいて、最適濃度、2mg/mLのB9A11−ビオチンおよび1mg/mLのAb−6 Pro−11が、実行するために選択された。
HER3トータルVERATAG(登録商標)アッセイの正確度
4つのよく特徴付けられた細胞系(293H3−クローン1、MDA−MB−453、MDA−MB468、MDA−MB−231およびSKOV3)から首尾よく複製された3つの複製物を使用して、1バッチのHER3トータルVERATAG(登録商標)アッセイを行い、次いでVERATAG(登録商標)測定の正確度を自家で生成したフローサイトメトリーおよびELISAのデータと比較した。結果の100%が、293H3−クローン1>MDA−MB−453>MDA−MB468>SKOV3という点においてフローサイトメトリーおよびELISAからの自家データと一致した。4つの細胞系試料のいずれについてもシグナルレベル間にオーバーラップは観察されなかった、すなわち、各細胞系が完全に分離された。ELISAおよびフローサイトメトリーの両方によりHER3の総レベルに対する内部データセットを生成した。4つの正確度の細胞系についての結果を図7に示す。HER3のフローサイトメトリーからの結果とHER3のELISAからの結果は、これらの交差検定技術を使用することによって同じ順位序列(same rank order)保存性が実証されたという点において、非常に類似していた。
HER3トータルVERATAG(登録商標)アッセイの感度
HER3トータルVERATAG(登録商標)アッセイの感度を、低いHER3発現の対照細胞系であるMDA−MB−468の8つの複製物を含有する1バッチを、低/陰性HER3発現の対照細胞系であるSKOV3の8つの複製物と比較することによって決定した。MDA−MB−468の各複製物についての値を、対比較によってSKOV3複製物と比較して感度を決定した。MDA−MB−468とSKOV3との間で対比較したうちの100%(64/64)が、MDA−MB−468>SKOV3という結果になった(図8)。
HER3トータルVERATAG(登録商標)アッセイの再現性
アッセイ間の再現性を、4つのよく特徴付けられた細胞系(293H3−クローン1、MDA−MB−453、MDA−MB−468およびSKOV3)に対して、種々の機器類(CE、イルミネーター)、数人のオペレーターを使用し、4週間にわたってアッセイを行って、上記の通りHER3トータルVERATAG(登録商標)アッセイを8つの別々のバッチで行うことによって決定した。バッチ正規化の手順の後、データを8バッチにわたって比較して再現性を確認した。ダイナミックレンジにわたる再現性は、8〜15%であった(図9参照)。
HER3トータルVERATAG(登録商標)アッセイの精度
アッセイ内の再現性を、1バッチ/細胞系のHER3トータルVERATAG(登録商標)アッセイを行い、3つの対照細胞系(293H3−クローン1、MDA−MB−453、MDA−MB−468)のそれぞれの15個の複製物の動作を比較することによって決定した。各バッチの15個の複製物について対比較を行ってアッセイの精度を決定した。293H3−クローン1のデータの95%が1.2倍以内であり、MDA−MB−453のデータの100%が1.23倍以内であり、MDA−MB−468のデータの95%が1.37倍以内であった。結果を図10に示す。
HER3トータルVERATAG(登録商標)アッセイの直線性
HER3 VERATAG(登録商標)の結果の直線性を、よく特徴付けられた細胞系対照である293H3−クローン1、MDA−MB−453、MDA−MB−468)を取得し、連続的な「縮小(cut−down)」実験を行って以下の寸法、1、1/2、1/4、1/16のFFPE切片を創出することによって決定した。次いで、切片サイズに関してアッセイの直線性を理解するために、これらの「縮小」切片を、H3T VERATAG(登録商標)アッセイにおいて処理し、最終的な切片面積の正規化データを対様式で比較した(図11)。MDA−MB−453は、元の切片サイズのおよそ1/16に至るまで直線であり、一方MDA−MB−468は、元の切片サイズのおよそ1/2であるはずのところに至るまで直線であった。結果を図11に示す。
HER3トータルVERATAG(登録商標)アッセイの特異性
患者由来の腫瘍試料および細胞系対照を、VERATAG(登録商標)HER3トータルアッセイを用いて、アイソタイプ対照抗体を使用して処理した。HER3 Ab−6−Pro11に対して、一致したアイソタイプ対照はIgG1−Pro11であった。HER3 B9A11−ビオチンに対して、一致したアイソタイプ対照は同様にIgG1−ビオチンであった。これらの反応からのシグナルは抗原特異的ではなく、非特異的なバックグラウンドを表す。試料を、以下の3つのフォーマットのそれぞれにおいて処理し、同じバッチ内で並列に処理した:
フォーマット1:HER3 Ab6−Pro11/B9A11−ビオチン(通常のフォーマット)
フォーマット2:HER2 Ab6−Pro11/IgG1−ビオチン
フォーマット3:IgG1−Pro11/B9A11−ビオチン
各IgG1フォーマット(フォーマット2およびフォーマット3)からの試料の結果を、各バッチに存在する陰性対照であるSKOV3(比較パラメータA)と比較した。試料の結果を、それぞれの実際のHER3総シグナル(フォーマット1)とも比較した。結果を図12に示す。
臨床的な乳腺腫瘍の測定とダイナミックレンジ
試験集団は、1999〜2006年の間の単一の施設におけるトラスツズマブに基づく療法(the International Serum Her−2/neu Study Group trial)の間に前向きに観察された患者(n=105)で構成された。HER−2/neuを過剰発現している(HercepTest;DAKO Diagnostics、Austriaによって決定された腫瘍細胞IHC3+が>10%)および/またはERBB2が増幅した(すべてのIHC2+の症例において義務があるFISH試験で陽性を示す)MBCを持つ外来患者(ECOG PS 0〜2、年齢>18歳、推定寿命>12週間)のみを含めた。さらに、患者にはトラスツズマブ未処置であり、治療開始前4週間以内に2次元的に測定可能な疾患の進行を有すること(以前に放射線照射した病変は除外)が要求された。この試験群の試行実験からの試料を、8つの別々のバッチでのH3T VERATAG(登録商標)アッセイにおいて処理した。各バッチについて、腫瘍面積分析の他にCEピーク分析も行い、それに応じてバッチを正規化した。アッセイにおいて処理した105個の患者試料のうち、85個の試料が、検出限界を超える測定可能なH3Tレベルを有し、8つの患者試料が検出可能な腫瘍を有さず、1つの試料について、データを除いてフルオレセイン不足が実証され、最終的に、8つの患者試料がアッセイの検出限界を下回り、低/陰性H3T発現と見なされた。結果を図13に示す。
トラスツズマブへの反応に対する最適なカットオフの決定
位置走査分析を用いて最適なカットオフを決定し、それにより、統計的に有意なカットオフを上回る患者は、このカットオフを下回る患者と比較して好ましくない無増悪期間(TTP)を有した(図14)。カットオフは、試験した集団の結果のおよそ中央値であった(0.158、HR=2.3、p=0.0004。TTPは、トラスツズマブを含有する治療の開始から増悪(SWOG)または打ち切りまでの時間として定義され、OSはトラスツズマブを含有する治療の開始から死亡または打ち切りまでの時間として定義された。この患者集団の全生存期間に着目した場合、有意なカットオフを決定することはできなかったが、0.158カットオフを使用したOSにトレンドがあった(HR=1.7;p=0.059)。
TTPについてのカプラン・マイヤー(KM)分析
以前報告されたH2Tカットオフ(logH2T≧1.14)を使用して、患者を、HER2正常群(N=26、TTP中央値=4.1mos)とHER2過剰発現群(N=55、TTP中央値=11.1mos、HR=0.43、p=0.0002)とに細分した。HER2過剰発現群において、位置走査によって定義された最適なカットオフ(H3T>0.158;図14)を上回るH3T発現のレベルでは、カットオフを下回るH3T発現(N=30、TTP中央値=13.1mos、HR=2.7、p=.0002)と比較して短い無増悪期間中央値(N=25、TTP中央値=6.1mos)が予測された。HER2過剰発現サブグループを調査する単変量コックス比例ハザード分析により、TTPの最も重要な予測因子としてH3Tが同定された(特定のカットオフを上回る対下回る)(HR=2.98、p=0.0004)。これらの結果を図15に示す。
他の悪性腫瘍におけるVERATAG(登録商標)によるHER3の総発現
H3T VERATAG(登録商標)アッセイを、結腸腫瘍、卵巣腫瘍、滑膜の腫瘍を含めた乳癌以外の多くの異なる悪性腫瘍に対して行った(図16)。これらの癌のすべてについて、以下の範囲の順位序列で同様のダイナミックレンジが観察された:卵巣癌>滑膜癌≧結腸癌。これらの腫瘍におけるダイナミックレンジは、癌に応じて0.5〜1.5logにわたる。
P95と併せたHER3の測定およびトラスツズマブへの反応との相関
以前報告されたP95のカットオフ(米国特許出願第12/629,037号)を使用して、上記のHER−2が過剰発現している患者を、そのHER3レベルに基づいて最初に細分した後、さらに層別化して、図17に示した4つの患者サブグループを生じさせた。位置走査によって予測された適切なカットオフを下回ると定義された、P95およびHER3が低レベルである患者は、他のどのサブグループよりも長い無増悪期間中央値(TTP=15.0mos)を有した(トレンドに対するログランク検定、p<0.0001)。P95およびHER3が高レベルである患者は、最短の無増悪期間中央値(TTP=3.2mos)を有した。結果を図17に示す。
HER2、HER3およびP95の測定ならびにトラスツズマブへの反応との相関
実施例17における患者集団を、図18に示した通り、そのHER2、P95およびHER3のレベルによって細分した。位置走査の方法論によって定義された、高いHER2、低P95および低いHER3である患者は、高レベルのHER2および高レベルのHER3またはP95またはその両方を有する患者よりも長い無増悪期間を有し、後者はH2T VERATAG(登録商標)アッセイによって決定された、低いHER2値を有する患者と同様の無増悪期間を示した。HER2発現レベルが正常であるサブグループ(H2Tlo)内で、FISH陽性群およびFISH陰性群は同様の無増悪期間を経験した。
Claims (31)
- 患者に由来する試料におけるHer−3または複合体化したHer−3の存在および/または量を測定および/または定量化する方法であって、試料を供給するステップと、前記試料におけるHer−3および/または複合体化したHer−3の存在および/または量を決定するステップを含む方法。
- Her−3レベルが高ければ、標的化療法が前記患者に奏効する可能性が低い、請求項1に記載の方法。
- Her−3レベルが低ければ、前記標的化療法が前記患者に奏効する可能性がある、請求項1に記載の方法。
- 前記試料が腫瘍試料である、請求項1に記載の方法。
- 前記試料が、FFPE試料または可溶化FFPE試料である、請求項1に記載の方法。
- 前記試料が、乳癌試料を含む、請求項1に記載の方法。
- 前記測定が、広範なダイナミックレンジにわたり定量的であり得る、請求項1に記載の方法。
- 前記試料を結合化合物と混合するステップと、Her−3および/または複合体化したHer−3に結合した前記結合化合物の存在および/または量を決定するステップを含む、請求項1に記載の方法。
- 前記結合化合物が、Her−3に特異的に結合する、請求項8に記載の方法。
- 前記結合化合物が、抗体を含む、請求項8に記載の方法。
- 前記抗体が、配列番号1〜8を有するペプチドのうちの1つに対して産生させた抗体である、請求項10に記載の方法。
- 前記抗体が、(a)それぞれ、配列番号13、14、および15に示される配列を有するCDR1、CDR2、およびCDR3を含む軽鎖可変領域と、(b)それぞれ、配列番号16、17、および18に示される配列を有するCDR1、CDR2、およびCDR3を含む重鎖可変領域を含むモノクローナル抗体;または(a)それぞれ、配列番号19、20、および21に示される配列を有するCDR1、CDR2、およびCDR3を含む軽鎖可変領域と、(b)それぞれ、配列番号22、23、および24に示される配列を有するCDR1、CDR2、およびCDR3を含む重鎖可変領域を含むモノクローナル抗体のうちの少なくとも1つである、請求項10に記載の方法。
- 前記抗体が、前記軽鎖および前記重鎖のアミノ酸配列が、それぞれ、配列番号9および11を有する抗体、または前記軽鎖および前記重鎖のアミノ酸配列が、それぞれ、表1に示される配列番号10および12を有する抗体のうちの少なくとも1つである、請求項10に記載の方法。
- (i)Her−3および/または複合体化したHer−3を含有し得る試料と;(ii)有効近接距離を有する、Her−3に結合することが可能な近接プローブと;(iii)Her−3に結合することが可能であり、1つ以上のシグナル伝達分子が結合している少なくとも1つの結合化合物とを混合するステップを含み、ここで、前記有効近接距離内において前記近接プローブおよび前記結合化合物が結合すると、前記Her−3および/または複合体化したHer−3の存在および/または量と相関するシグナルが分子タグから生成される、請求項1に記載の方法。
- 前記近接プローブおよび/または前記結合化合物が、Her−3または少なくとも1つの他の検体に特異的に結合することが可能である、請求項14に記載の方法。

- 標的化療法による治療が、癌を有する被験体に奏効する可能性があるかどうかを判定し、疾患の時間経過を予測し、かつ/または前記被験体の癌の時間経過において重要事象が生じる確率を予測する方法であって、前記被験体の癌に由来する生物学的試料においてHer−3の量を測定するステップを含み、ここで、前記方法が、Her−3レベルに依存する、方法。
- 前記Her−3レベルが高ければ、標的化療法が前記患者に奏効する可能性が低い、請求項18に記載の方法。
- 前記Her−3レベルが低ければ、前記標的化療法が前記患者に奏効する可能性がある、請求項18に記載の方法。
- 前記被験体の癌が、乳癌、結腸直腸癌、卵巣癌、非小細胞肺癌、または胃癌である、請求項18に記載の方法。
