Abstract
Description

材料及び方法
AON設計は、(部分的に)m−foldプログラムにより予見される標的エクソンRNAの重複オープン二次構造に、また(部分的に)ESE−finderソフトウエアにより予見される重複推定SR−タンパク質結合部位に基づいて行った。AONは、Prosensa Therapeutics B.V.(ライデン、オランダ)により合成し、これは2’−O−メチルRNA及び全長ホスホロチオエート(PS)骨格を含む。
健常個体(「ヒトコントロール」)(例1、3及び4;エクソン43、50、52スキッピング)又はエクソン45欠失を有するDMD患者(例2;エクソン46スキッピング)由来の筋管培養物を以前記載した通りに処理した(Aartsma−Rusら、Neuromuscul.Disord.2002;12:S71〜77及びHum Mol Genet2003;12(8):907〜14)。AONのスクリーニングのため、筋管培養物を50nM及び150nM(例2)、200nM及び500nM(例4)又は500nMのみ(例1及び3)の各AONでトランスフェクションした。トランスフェクション試薬UNIFectylin(Prosensa Therapeutics BV、オランダ)は、AON1□g当たり2□lのUNIFectylinを用いた。エクソンスキッピング効率は、標的エクソン(43、46、50、51、52又は53)側方のエクソンに存在するプライマーを用いたネステッドRT−PCR解析により決定した。配列検証のため、アガロースゲルからPCR断片を単離した。定量化のため、DNA1000LabChipsキットを用いてAgilent2100bioanalyzer(Agilent Technologies、米国)においてPCR産物を解析した。
DMDエクソン43スキッピング
エクソン43内の配列を標的とする一連のAONを設計し、健常なコントロール筋管培養物にトランスフェクションした。続く単離RNAのRT−PCR及び配列解析は、本明細書では配列番号2として定義されているエクソン43内の一続きのヌクレオチドを標的とするほぼ全てのAONが、エクソン43スキッピングを確実に誘導できることを示した。図1に示す通り、PS237(配列番号65)は、500nMで最高レベルのエクソン43スキッピング(最大66%)を再現可能に誘導した。比較のため、それぞれ最大13%及び36%のエクソン43スキッピングレベルを誘導する、PS238及びPS240も示す(図1)。エクソン43の正確なスキッピングは、新規のより小さな転写産物断片の配列解析によって確認した。エクソン43スキッピングは、未処置細胞(NT)には観察されなかった。
エクソン46内の配列を標的とする一連のAONを設計し、DMD遺伝子にエクソン45欠失を有するDMD患者由来の筋管培養物にトランスフェクションした。このような突然変異を有する患者のため、アンチセンスが誘導するエクソン46スキッピングは、疾病の1又複数種の症状を確実に緩和することができる、新規BMD様ジストロフィンタンパク質の合成を誘導した。続く単離RNAのRT−PCR及び配列解析は、本明細書では配列番号3として定義されているエクソン46内の一続きのヌクレオチドを標的とするほぼ全てのAONが、50nMと比較的低濃度のAONであってもエクソン46スキッピングを確実に誘導できることを示した。図2に示す通り、PS182(配列番号117)は、最高レベルのエクソン46スキッピング(50nMで最大50%、150nMで74%)を再現可能に誘導した。比較のため、150nMでそれぞれ最大55%、58%及び42%のエクソン46スキッピングレベルを誘導する、PS177、PS179及びPS181も示す(図2)。エクソン46の正確なスキッピングは、新規のより小さな転写産物断片の配列解析によって確認した。エクソン46スキッピングは、未処置細胞(NT)には観察されなかった。
エクソン50内の配列を標的とする一連のAONを設計し、健常なコントロール筋管培養物にトランスフェクションした。続く単離RNAのRT−PCR及び配列解析は、本明細書では配列番号4として定義されているエクソン50内の一続きのヌクレオチドを標的とするほぼ全てのAONが、エクソン50スキッピングを確実に誘導できることを示した。図3に示す通り、PS248(配列番号127)は、最高レベルのエクソン50スキッピング(500nMで最大35%)を再現可能に誘導した。比較のため、500nMで最大14〜16%のエクソン50スキッピングレベルを誘導する、PS245、PS246及びPS247も示す(図3)。エクソン50の正確なスキッピングは、新規のより小さな転写産物断片の配列解析によって確認した。エクソン50スキッピングは、未処置細胞(NT)には観察されなかった。
エクソン51内の配列を標的とする一連のAONを設計し、健常なコントロール筋管培養物にトランスフェクションした。続く単離RNAのRT−PCR及び配列解析は、本明細書では配列番号5として定義されているエクソン51内の一続きのヌクレオチドを標的とするほぼ全てのAONが、エクソン51スキッピングを確実に誘導できることを示した。配列番号180を有するAONは、最高レベルのエクソン51スキッピングを再現可能に誘導した(データなし)。
エクソン52内の配列を標的とする一連のAONを設計し、健常なコントロール筋管培養物にトランスフェクションした。続く単離RNAのRT−PCR及び配列解析は、本明細書では配列番号6として定義されているエクソン52内の一続きのヌクレオチドを標的とするほぼ全てのAONが、エクソン52スキッピングを確実に誘導できることを示した。図4に示す通り、PS236(配列番号299)は、最高レベルのエクソン52スキッピング(200nMで最大88%、500nMで91%)を再現可能に誘導した。比較のため、500nMを投与した場合、それぞれ最大59%及び10%レベルのエクソン52スキッピングを誘導するPS232及びAON52−1(Aartsma−Rusら、Oligonucleotides2005により以前刊行された)も示す(図4)。新規のより小さな転写産物断片の配列解析によって、エクソン52の正確なスキッピングを確認した。エクソン52スキッピングは、未処置細胞(NT)には観察されなかった。
エクソン53内の配列を標的とする一連のAONを設計し、健常なコントロール筋管培養物にトランスフェクションした。続く単離RNAのRT−PCR及び配列解析は、本明細書では配列番号7として定義されているエクソン53内の一続きのヌクレオチドを標的とするほぼ全てのAONが、エクソン53スキッピングを確実に誘導できることを示した。配列番号328を有するAONは、最高レベルのエクソン53スキッピングを再現可能に誘導した(データなし)。

から選択されるヌクレオチド配列のうちの一つの配列内の一続きの少なくとも8ヌクレオチドと結合する分子。 - 表1〜6に示されている配列番号8〜358及び/又は配列番号529〜535から選択されるアンチセンスヌクレオチド配列を含む又はからなる、請求項1に記載の分子。
- 配列番号16、配列番号65、配列番号70、配列番号91、配列番号110、配列番号117、配列番号127、配列番号165、配列番号166、配列番号167、配列番号246、配列番号299及び配列番号357から選択されるアンチセンスヌクレオチド配列を含む又はからなる、請求項2に記載の分子。
- 2’−O−アルキルホスホロチオエートアンチセンスオリゴヌクレオチドを含む、請求項1から2までのいずれか一項に記載の分子。
- 2’−Oメチルホスホロチオエートリボースを含む、請求項4に記載の分子。
- 請求項1から5までのいずれか一項に記載の分子の発現を駆動する発現カセットを含む、ウイルスに基づくベクター。
- 好ましくは、DMD若しくはBMD患者のDMD mRNA前駆体のスプライシングを調節するため、又はDMD若しくはBMD患者を治療するための薬剤として使用される、請求項1から5までのいずれか一項に記載の分子又は請求項6に記載のウイルスに基づくベクター。
- 患者のDMD mRNA前駆体のエクソン6、7、11、17、19、21、43、44、45、50〜53、55、57、59、62、63、65、66、69又は75のうちの少なくとも1個のスキッピングを誘導又は促進することができる分子と任意選択で組み合わせた、請求項1から5までのいずれか一項に記載の分子及び/又は請求項6に記載のベクター、薬学的に許容される担体を含む、医薬組成物。
- 患者、好ましくは患者の単離細胞におけるDMD mRNA前駆体のエクソン43、エクソン46、エクソン50〜53のうちの少なくとも1個のスキッピングを誘導及び/又は促進するための方法であって、前記細胞及び/又は前記患者に請求項1から5までのいずれか一項に記載の分子又は請求項6に記載のベクターを提供するステップを含む上記方法。
- 患者のDMD mRNA前駆体のエクソン6、7、11、17、19、21、43、44、45、50〜53、55、57、59、62、63、65、66、69又は75のうちの少なくとも1個のスキッピングを誘導又は促進することができる追加的な分子が使用される、請求項9に記載の方法。
- DMD mRNA前駆体のスプライシングを調節するための、又はDMD若しくはBMD患者を治療するための薬剤を調製するための、請求項1から5までのいずれか一項に記載の分子、請求項6に記載のベクター又は請求項8に記載の医薬組成物の使用。
