(発明を実施するための形態)

細胞及びウイルス:HEK293T及びMDCK細胞を、American Type Culture Collection(Manasass,VA)から入手し、10%ウシ胎児血清及びペニシリン−ストレプトマイシンを含むダルベッコ改変イーグル培地(Gibco)において維持した。インフルエンザA/PR/8/34(PR8)H1N1ウイルスを、8プラスミド逆遺伝学システムを使用して作製した。組織培養された馴化インフルエンザA/Hong Kong/8/68(HK68)H3N2ウイルスを、ATCC(ATCC−VR−1679)から入手した。ウイルスは、10日齢特定病原体不在ニワトリ胚において35℃で増殖及び増幅させた(Charles River Laboratories、SPAEAS)。

IAVセグメントPB2パッケージングシグナルのSHAPE特徴分析は保存構造を同定する
プライマー伸長により分析される選択的2’−ヒドロキシルアシル化(SHAPE)及び計算的モデリングを、IAVセグメントPB2ゲノムvRNAに適用し、構造化RNAドメインを検索した。株A/Puerto Rico/8/1934(H1N1)「PR8」からのインビトロ転写全長(−)センスPB2 vRNAを、溶液中で折り畳み(Pang et al.,2011)、柔軟な一本鎖状態で存在するヌクレオチドと優先的に反応する求電子性SHAPE試薬を使用して調査した(Wilkinson et al.,2006)(図1)。この分析は、PB2を含む、全てのセグメントについての(−)センスRNAに対して(+)センスでのRNA二次構造保存について高い可能性を見出した最近の生物情報学研究(Priore et al.,2012、Moss et al.,2011)と一貫して、2341ntのvRNAのほとんどが広く構造化されていないことを明らかにした(図2)。これらの以前の研究は、末端コード領域(TCR)を分析せず、代わりにPB2 5’TCRの末端の手前の80個のヌクレオチドで止まっている。SHAPE誘導モデリングは、ヌクレオチド34〜87((−)センス表記)を含む、安定なRNA二次構造、最も注目すべきは、本明細書でパッケージングステムループ2(PSL2)(図1A)と称される、ステムループモチーフを含有するこの領域中のいくつかのエリアを示した。このセグメントは、未解明のメカニズムを介した変異分析を通したPB2パッケージングにおいて以前に示されたヌクレオチドのセットを含んだ(図1A〜1B、環状ヌクレオチド参照)(Gao et al.,2012、Marsh et al.,2008、Liang et al.,2008、Gog et al.,2007)。これらの従来の変異がPSL2構造の破壊を通して作用するという仮説を支持し、変異体のSHAPE分析は、全てが野生型PSL2構造を破棄した異なる立体構造をもたらした(図1C、図3)。PSL2を包含する60ヌクレオチド領域は、異なるサブタイプ及び種由来の季節性及びパンデミック株の間で単一ヌクレオチドレベルでの100%近い配列保存を示し(図4)、インタクトなPSL2構造を維持するための厳しい生物学的要件の存在を示す。PB2 vRNA内の異なる下流配列はPSL2の二次構造を変え得るので、PSL2の構造的保存は、高病原性鳥H5N1及びパンデミック1918H1N1株を含む、様々なIAV株及びサブタイプから単離される全長野生型PB2 vRNAに対してSHAPE分析を行うことにより探究された。ステムループ内の2つの分岐するヌクレオチドの存在及び隣接する配列における顕著な分岐にかかわらず、PSL2ステムループ構造は、これらの多様な種及びサブタイプにわたるPB2RNAのSHAPE誘導モデリングにおいて回収された(図1D〜1F)。
PSL2RNA構造のSHAPE分析をさらに試験し、インビボ試験に必要な追加の情報を提供する変異を明らかにするために、多次元化学マッピング(Kladwang and Das,2010)方法をPSL2セグメントに適用した。第一に、mutate−and−map(M2)測定は、PB2パッケージングにとって重要であることが以前に見出されているヌクレオチドでの変化を含む、各ステム残基の全体的変異後の化学反応性パターンの破壊を確認した(図5A、備考欄参照)(Marsh et al.,2008、Gog et al.,2007)。これらのM2データに基づく自動化計算分析は、SHAPE誘導PSL2構造を高い信頼度で回収し(図1C、図3、図5B〜図5D)、構造モデルをさらに検証した。第二に、予測試験として、補償的変異は、初期パッケージング欠損変異により破壊された野生型ステムループ構造における塩基対を復元するように設計された(図6、パネルA〜B)。これらの変異−レスキュー多様体は、実際に、PSL2SHAPEパターンを復元し、モデル化構造のインビトロ検証における塩基対解像を提供し、PSL2構造の役割をインビボで試験するための配列多様体を提案した。
PSL2構造の治療設計及び標的化は、IAV感染をインビトロ及びインビボで阻害する
PSL2媒介性ウイルスパッケージングを標的とする治療可能性を探究するために、ホスホロチオエートヌクレオシド間結合を含有する9つのロックド核酸(LNA)(Vester and Wengel,2004)を、要素の全体的なRNA二次構造を破壊し、これによりウイルス産生を阻害することが予測される主要残基に対して設計した(図15、パネルA)。設計されたLNAのうちの2つ、LNA8及びLNA9は、配列において、それぞれ、LNA6及びLNA7と同一であるが、RNase−H活性化に最適化された6〜7個の非修飾(非ロックド)DNAヌクレオチドを有する。第一に、PSL2RNA二次構造に対してLNA結合が有する影響を評価するために、トウプリンティング及びSHAPE化学マッピングを、LNAの存在下でPB2 vRNAについて行った。抗ウイルスアッセイ結果での実証では、LNA6〜9にコードされる配列は、野生型PSL2構造を結合及び破壊する最大の能力を示した(図16)。
インビボ有効性実験:長期単回用量予防
Balb/C雌マウス(5マウス/グループ)を、致死用量の野生型PR8ウイルスでの感染の3日前(−3日目)または感染の1日前(−1日目)のいずれかに、単回用量の20ugLNA9で予め鼻腔内処理した。マウスを、体重減少、臨床スコア、及び生存について毎日モニタリングした。図17、パネルAは、時間に伴うマウスの生存パーセントを示す。図17、パネルBは、投与後の時間に伴う体重減少パーセントを示す。
薬物の存在下での連続継代後の、オセルタミビル及びLNA9に対するインフルエンザウイルスの感受性:薬物選択実験
オセルタミビル(タミフル)は、市場で最も広く使用され、備蓄されるニューラミニダーゼ阻害剤(NAI)である。全てのNAIと同様に、オセルタミビルは、薬物を収容するウイルスニューラミニダーゼ(NA)タンパク質における立体的構造再配置を必要とする。この再配置に影響を及ぼすNAタンパク質にけるいずれかの変異は、オセルタミビルの結合親和性を低減し、よって薬物有効性を低減する。とりわけ、(用語体系に応じてH275Y変異としても知られる)H274Y変異体は、オセルタミビル耐性と最もよく関連付けられる。免疫低下患者におけるH274Y変異の急速な選択は、最終手段のNAI薬、ぺラミビルの臨床的失敗をもたらし得、免疫低下宿主における多剤耐性ウイルスについての選択が、以前に考えられたよりも一般的であり得ることを示す。これは、最近のオセルタミビル耐性及びNAI耐性ウイルスの拡散と共に、一般的なNAIの使用を再評価する必要性を示す。新たなクラスの抗ウイルスの開発は、ヒトの健康に及ぼし得る未来のインフルエンザパンデミックの有害な影響を低減するためには急務である。

PSL2モチーフに特異的に結合する薬剤が、細胞中のインフルエンザA型ウイルスを阻害する、方法。
項40.候補薬剤が、小分子、核酸、及びポリペプチドから選択される、項39に記載の方法。
項41.決定ステップが、細胞パラメータを検出することを含み、細胞中のパラメータの、候補薬剤と接触していない細胞中と比較した変化が、候補薬剤がPSL2モチーフに特異的に結合することを示す、項40に記載の方法。
項42.PSL2モチーフに特異的に結合する薬剤が、インフルエンザA型ウイルス感染を有する対象を治療する、項39〜41のいずれか1つに記載の方法。

- PB2 vRNA領域と相補的なオリゴヌクレオチド配列を含むオリゴヌクレオチド化合物であって、前記領域が、前記PB2 vRNAの5’末端コード領域の(−)センス表記中のヌクレオチド34〜87を含む、オリゴヌクレオチド化合物、またはその塩。
- 前記PB2 vRNAの領域のパッケージングステムループ2(PSL2)モチーフの領域と相補的な少なくとも8個のヌクレオシドサブユニットを含むオリゴヌクレオチド配列を含む、請求項1に記載の化合物。
- 前記オリゴヌクレオチドが、ホスホロチオエート、ホスホロジチオエート、ホスホルアミデート、及びチオホスホルアミデート結合から選択されるヌクレオシド間結合を含む、請求項1に記載の化合物。
