Abstract
【解決手段】(a)フラビンタンパク質、キノンタンパク質及びそれらの組み合わせからなる群から選択される酵素、及び(b)フェノチアジン、フェノキサジン及びそれらの組み合わせからなる群から選択される媒介物を含む、分析対象物を検出するための試薬。

Description
液状試薬を、100mMリン酸ナトリウムpH7.4中、20単位/μlピロロキノリンキノン−グルコースデヒドロゲナーゼ(PQQ−GDH)及び24mM媒介物Iになるように調製した。試薬の第一の成分を、媒介物を100mMホスフェートpH7.4に溶解し、pHを調節して7.4に戻し、溶液をWhatman0.45ミクロンPTFEシリンジフィルタに通ってろ過することによって製造した。凍結乾燥させたPQQ−GDH(Toyobo製品番号GLD-321)を20単位/μlの活性まで加えることにより、試薬を完成させた。
スクリーン印刷法を使用して電気化学的バイオセンサを構築した。センサは、炭素作用電極及び銀/塩化銀参照電極で構成した。100mMリン酸緩衝液(pH7.4)中に12mM媒介物Iを含有する、酵素PQQ−グルコースデヒドロゲナーゼ(10単位/μl)の溶液(150〜800nl)を作用電極の表面に被着させ、室温で5分間乾したのち、乾燥させた。電極を、センサへのサンプル溶液の接種を可能にする小さな毛管間隙を有するフォーマットとして組み立てた。
5種のバイオセンサ試薬処方(表1)を調製し、Titan Scan Technologies(カリフォルニア州サンディエゴ)でSureBeam(登録商標)滅菌技術を使用して電子ビーム照射に付した。処方Iは25kGy、50kGy及び100kGyで照射したが、処方II〜Vそれぞれは25kGyのみで照射した。表1の右寄り二列の見出し中、CMCはカルボキシメチルセルロースを意味し、PEOはポリエチレンオキシドを意味する。
Claims (10)
- 表面を有する第一の電極、及び前記第一の電極に結合された第二の電極を含み、
前記第一の電極の表面が、
フラビンタンパク質、キノンタンパク質及びそれらの組み合わせからなる群から選択される酵素、並びに
二つの酸化還元等価物を運ぶ能力を有する一以上の媒介物
を含む試薬を含む、電気化学的センサ。 - 一以上の媒介物が、3−(4′−クロロフェニルイミノ)−3H−フェノチアジン、3−(4′−ジエチルアミノフェニルイミノ)−3H−フェノチアジン、3−(4′−エチルフェニルイミノ)−3H−フェノチアジン、3−(4′−トリフルオロメチルフェニルイミノ)−3H−フェノチアジン、3−(4′−メトキシカルボニルフェニルイミノ)−3H−フェノチアジン、3−(4′−ニトロフェニルイミノ)−3H−フェノチアジン、3−(4′−メトキシフェニルイミノ)−3H−フェノチアジン、7−アセチル−3−(4′−メトキシカルボニルフェニルイミノ)−3H−フェノチアジン、7−トリフルオロメチル−3−(4′−メトキシカルボニルフェニルイミノ)−3H−フェノチアジン、3−(4′−ω−カルボキシ−n−ブチルフェニルイミノ)−3H−フェノチアジン、3−(4′−アミノメチルフェニルイミノ)−3H−フェノチアジン、3−(4′−(2″−(5″−(p−アミノフェニル)−1,3,4−オキサジアゾイル)フェニルイミノ)−3H−フェノチアジン、3−(4′−β−アミノエチルフェニルイミノ)−3H−フェノチアジン、6−(4′−エチルフェニル)アミノ−3−(4′−エチルフェニルイミノ)−3H−フェノチアジン、6−(4′−〔2−(2−エタノールオキシ)エトキシ〕エトキシフェニル)アミノ−3−(4′−〔2−(2−エタノールオキシ)エトキシ〕エトキシフェニルイミノ−3H−フェノチアジン、3−(4′−〔2−(2−エタノールオキシ)エトキシ〕エトキシフェニルイミノ−3H−フェノチアジン、3−(4′−フェニルイミノ)−3H−フェノチアジンボロン酸、(3−(3′,5′−ジカルボキシフェニルイミノ)−3H−フェノチアジン、3−(4′−カルボキシフェニルイミノ)−3H−フェノチアジン、3−(3′,5′−ジカルボキシフェニルイミノ)−3H−フェノキサジン、3−(3′,5′−フェニルイミノ)−3H−フェノチアジンジスルホン酸、3−(3−フェニルイミノ)−3H−フェノチアジンスルホン酸及びそれらの組み合わせからなる群から選択される、請求項1記載の電気化学的センサ。
- 前記フラビンタンパク質が、FAD−グルコースオキシダーゼ、フラビン−ヘキソースオキシダーゼ、FAD−グルコースデヒドロゲナーゼ、[FAD]−乳酸オキシダーゼ、[FAD]−コレステロールオキシダーゼ、[FAD]−アルコールオキシダーゼ、[FAD]−D−アミノ酸オキシダーゼ、[FAD]−コリンオキシダーゼ及びそれらの組み合わせからなる群から選択される、請求項1記載の電気化学的センサ。
- 前記酵素が、FAD−グルコースオキシダーゼ、PQQ−グルコースデヒドロゲナーゼ及びそれらの組み合わせからなる群から選択される、請求項1記載の電気化学的センサ。
- カルボキシメチルセルロース、ポリエチレンオキシド及びそれらの組み合わせからなる群から選択されるポリマーをさらに含む、請求項1記載の電気化学的センサ。
- 前記第二の電極が銀/塩化銀参照電極である、請求項1記載の電気化学的センサ。
