Description
発現された遺伝子の総数は、40,000〜150,000を超える範囲にわたると見積もられる。この数は、コードされるタンパク質の数の正確な反映ではない。なぜなら、多くの場合、これらの遺伝子からは、このmRNA(転写物)から1つより多いスプライシング改変体が生じるからである。再度、見積もりは変化するが、おそらく500,000程度の多さの異なるmRNAがヒトにおいて産生される。ヒト遺伝子の少なくとも30%がいくつかのスプライシング改変体を有すると推定される。Mironovら(1999)Genome Research 9:1288〜1293)。これらの数は、ある程度保存されるべきであると考えられる。類似の数がマウスおよびラットについても正しく、そして下等な生物体(例えば、Drosophila melanogaster、およびCaenorhabditis elegans)でもまたオルタネーティブスプライシングが生じると考えられる。異なるスプライシング改変体(バリアント)から翻訳されたタンパク質は、研究論文の数が増加していることによって証明されるように、有意に異なる機能を有し得る。異なるスプライシング改変体が、異なる組織、異なる発生段階、および異なる疾患状態で発現され得る。

1.抗原結合フラグメントおよびそれらの組成物
本発明は、DCを特異的に認識する抗原結合フラグメントを含む。すなわち、抗原は、DC上で見出され、その結果、抗原を認識する抗原結合フラグメントは優先的にDCまたはそれらのサブセットを認識するか、または結合する。あるいは、BDCA−4によるように、抗原は、他の細胞型で見出される;しかし、造血細胞内で、抗原はDC上に主に発現する。

e)抗原提示細胞を得るためのインビトロでのT細胞刺激。
a)自己免疫あるいは至適のおよび選択的な取り込み/トランスフェクションについてDCに対する抗原または核酸(ウイルス、プラスミドDNA、RNAなど)のインビボ標的化に関与するDC(例えば)のリガンド−またはリガンド媒介免疫治療を模倣する抗体。BDCA−2が、抗原取り込みおよびプロセシング機能を有する分子であるらしいのと同様に、この文脈において特に有用であり得る。
本発明は、血清および他の供給源におけるBDCA−2(可溶性BDCA−2(そのアイソフォームを含む)を含む)を測定するための、抗DC特異的抗原結合フラグメントを含むキットを包含する。これらのキットを使用する診断手順は、診断室、実験室、医師、または個人によって行われ得る。臨床サンプルは、試験される標的の富化のために、必要に応じて前処理される。次いで、使用者は、キットに含まれる試薬を適用して、その診断成分における変化したレベルまたは変更を検出する。

(DC特異的モノクローナル抗体(mAb)の生成)
6〜8週齢の5匹の雌性Balb/cマウス(Simonsen Laboratories、Gilroy、CA)に、麻酔下で、右足蹠に約5×105〜1×106の精製HLA−DR+lin-血液DCを0日目、4日目、7日目、11日目および14日目に、そして左足蹠に約1×106のHLA−A2+Bristol−8 Bリンパ芽球腫を、−3日目、0日目、4日目、7日目、11日目、および14日目に接種した。両方の細胞型を、室温で10分間1:100PHA(Gibco/BRL、Gaithersburg、MD)とともにインキュベートし、そして注射前にPBSで洗浄した。この処置は、非特異的アジュバント効果を提供し、そしてフロイントアジュバントのようなアジュバントについての必要性を除去する。
正常な健常なボランティアからのバフィコートを、the Institute for Transfusionmedicine、Hospital Merheim、Cologne、Germanyから得た。PBMCを、標準的Ficoll−Paque(Pharmacia、Uppsala、Sweden)密度勾配遠心分離によりバフィコートから調製した。
「未成熟」単球由来DC(Mo−DC)の生成のために、精製CD14+単球を、培地[2mmol/L L−グルタミン、10% FCS(Sigma)、100mmol/Lピルビン酸ナトリウム(Gibco/BRL)、100 U/mlペニシリン(Gibco/BRL)および100μg/mlストレプトマイシン(Gibco/BRL)を補充した、RPMI 1640(Gibco/BRL)]中で5×105〜1×106細胞/mlの細胞密度で、500〜1000U/ml rIL−4(PeproTech、Rocky Hill、NJ)および100ng/ml rGM−CSF(PeptoTech)の存在下で、加湿5%CO2含有雰囲気下で37℃で7日間培養した。「成熟」Mo−DCの生成のために、「未成熟」Mo−DCを1回洗浄し、そして20ng/ml TNF−α(PeproTech)の存在下でさらに3日間培地中で培養した。CD34+造血前駆細胞由来DC(CD34−DC)の生成のために、精製CD34+細胞を、100ng/ml rFlt3−Ligand(PeproTech)、0.5ng/mL rTGF−β1(PeproTech)、10ng/ml rTNF−α、20ng/ml rSCF(PeproTech)および100ng/ml rGM−CSFの存在下で、培地中で11日間、細胞密度5×104細胞/mlで培養した。新鮮に単離したCD4+lin-血液DCを、10ng/ml rIL−3(PeproTech)の存在下で48時間まで、培地中で細胞密度5×105〜1×106細胞/mlで培養した。単離したCD1c発現DC、BDCA−2発現DC、およびBDCA−3発現DCを、いかなるサイトカインも含まないか、または10ng/ml rIL−3、20ng/ml IL−4(PeproTech)および100ng/ml GM−CSFの存在下で、培地中で48時間まで、細胞密度5×105〜1×106細胞/mlで培養した。
(血液DCのフローサイトメトリー分析)
FACScalibur(BD Biosciences)を、1色フローサイトメトリー、2色フローサイトメトリー、3色フローサイトメトリーまたは4色フローサイトメトリーのために使用した。5×103〜2×105細胞/サンプルのデータを、リスト様式で得、そしてCellQuestソフトウェア(BD Biosciences)を使用して分析した。
細胞を、細胞遠心機(Cytospin 3,Shandon,Pittsburgh,PA)において、スライド上にスピンダウンさせた。蛍光顕微鏡検査のために、細胞遠心分離の後に、スライドを一晩風乾し、そしてFluoromount G(Southern Biotechnology Associates)で封入した。メイ−グリュンヴァルト/ギームザ染色のために、スライドを、細胞遠心分離の後に少なくとも2時間に間にわたって風乾し、メイ−グリュンヴァルト/ギームザ溶液(Merck,Darmstadt,Germany)中で2分間、室温で染色し、蒸留水中で徹底的にリンスし、ギームザ溶液(Merck)中で15分間、室温で染色し、蒸留水中で繰り返し洗浄し、そして少なくとも2時間風乾した。Zeiss Axioscop(顕微鏡)(Zeiss,Oberkochen,Germany)を、分析のために使用した。デジタル写真を、Xillix Microlmager Ml1400−12X(Xillix,Vancouver,Canada)を使用して作成した。
(交差阻害、同時キャッピングおよび同時インターナリゼーションの分析)
2つの異なるmAbクローンが同じ(かまたは密接に関連する)抗原エピトープを認識するか否かを分析するために、交差阻害結合アッセイを実施した。1×106と2×106との間の細胞を、これら2つのmAbクローンのうちの一方とともに、約100μg/mlの濃度で10分間、4℃で予備インキュベートし、次いで、他方のmAbクローンのPE結合体で、その最適な力価でさらに5分間、4℃で染色した。PBMCを使用して、BDCA−2、BDCA−3、およびBDCA−4に特異的なmAbクローンの交差阻害を分析し、そしてMOLT−4細胞を使用して、CD1c特異的mAbクローンの交差阻害を分析した。細胞染色を、フローサイトメトリーによって分析した。

血液DCサブセットのエンドサイトーシスを評価するために、精製したCD1c+、BDCA−2+およびBDCA−3+血液DC、ならびに(コントロールとして)精製したCD3+T細胞を、37℃で、1mg/ml Luciferイエロー(LY)を含む培地中で、0分間、15分間、45分間、および75分間インキュベートした。その後、細胞を氷冷PBS/EDTA/BSA中で3回洗浄し、そしてフローサイトメトリーによって分析した。
(単離された血液DCの非培養血液細胞との反応性)
表1に列挙するmAbの血液細胞との反応性によって、これらを以下の4つの群に分割し得る:(1)AC144、AD5−13A11およびADB−4B8;(2)AD5−17F6;(3)AD5−5E8およびAD5−14H12;ならびに(4)AD5−8E7。

新たに単離されたプラスマ細胞様CD11c-血液DCは、生存および変異に関してIL−3に依存するが、一方でCD11c+血液DCの生存および変異は、サイトカイン依存性がはるかに低い。CD11c-およびCD11c+血液DCにおける、BDCA−2、BDCA−3およびBDCA−4の発現を、rIL−3の存在下での全血液DCの培養の0時間、1時間、3時間、6時間、9時間、12時間、18時間、24時間、36時間、および48時間後に、分析した。この結果を図4に示す。BDCA−2の発現は、CD11c-血液DCにおいて、48時間以内に完全にダウンレギュレーションされる。対照的に、BDCA−4は、CD11c-血液DCにおいてなおさらにアップレギュレートされ、そしてBDCA−2とは異なり、これはまた、全てでなければ大部分のCD11c+DCにおいて、高レベルに発現される。BDCA−3の発現は、CD11c-血液DCにおいて迅速に誘導され、24時間後に最も高い発現レベルに達する。その後、BDCA−3発現は、再度ダウンレギュレートされるようである。CD11c+血液DCにおけるBDCA−3の発現の分析は、BDCA−3-CD11cbrightおよびBDCA−3+CD11cdimサブセットが培養の開始時に存在するという事実によって、複雑になる。BDCA−3の発現は、BDCA−3+CD11cdim血液DC集団における培養の少なくとも6時間まで変化しないままであり、そしてBDCA−3-CD11cbright血液DCサブセットの少なくともいくらかの細胞において、3時間以内で誘導される。
機能的CD1a+DCを、エキソビボで、単球およびCD34+造血前駆細胞から生成した。Benderら(1996);Picklら(1996);Romaniら(1994);Sallustoら(1994);Cauxら(1992);Mackensenら(1995);Szabolcsら(1995);Herbstら(1996);de Wynterら(1998);およびStrunkら(1996)。図7は、Mo−DC(これらは、rGM−CSFの存在下で単球を7日間培養することによって生成した)ならびにIL−4およびCD34−DC(rFlt3リガンド、rTGF−β1、rTNF−α、rSCFおよびrGM−CSFの存在下でCD34+造血前駆細胞を11日間培養することによって生成した)が、CD1a、CD1cおよびBDCA−4を発現するが、BDCA−2もBDCA−3もいずれも発現しないことを示す。
(抗BDCA−2 mAb媒介架橋の際のBDCA−2のインターナリゼーション)
抗DCA−2 mAb標識BDCA−2+細胞の37℃でのインキュベーションがmAbインターナリゼーションを生じる可能性を、FITC結合体化AC144 mAb(IgG1)でのPBMCの染色によって、検討した。次いで、37℃でのインキュベーションに続いて、残っている細胞表面関連mAbを、PE結合体化ラット抗マウスIgG1 mAbでの染色によって検出した。図8に示すように、細胞を37℃でインキュベートした場合に、ラット抗マウスIgG1−PE染色の強度は、バックグラウンドレベルまで非常に迅速に低下する。対照的に、AC144−FITC染色の強度は、約50%のレベルまで一時的にのみ低下するが、その後、ほぼインキュベーション前のレベルまで戻る。このことは、BDCA−2が、レセプター媒介エンドサイトーシスと類似の速度論で、抗BDCA−2 mAb架橋の際にインターナリゼーションされることを実証する。AC144−FITC染色強度の一時的な低下は、恐らく、エンドサイトーシス前のBDCA−2/抗BDCA−2 mAb複合体のパッチングおよびキャッピングに起因する。
(単離されたCD1c+、BDCA−2+およびBDCA−3+血液DCの形態)
CD1c+、BDCA−2+およびBDCA−3+細胞を、PE結合体化一次mAbおよび抗PE Ab結合体化マイクロビーズで間接的に磁気によって標識すること、ならびに標識した細胞をMACSによって濃縮することによって、PBMCから単離した(図9)。細胞遠心分離スライドのメイ−グリュンヴァルト/ギームザ染色(図9)の際に、新たに単離されたBDCA−2発現細胞は、血液および扁桃由来のCD11c-CD4+lin-DCの代表的なリンパプラスマ細胞様形態を示す:すなわち、楕円形または湾入した核を有する、中程度の大きさの円形細胞。対照的に、新たに単離されたCD1c+血液DCと新たに単離されたBDCA−3+血液DCとの両方が、血液または扁桃由来のCD11c+CD4+lin-DCの代表的な形態学的特性を示す:すなわち、短い細胞プロセスおよびさらに過度に小裂片に分かれた核を有する、あまり円形ではない細胞。CD1c+BDCに加えて、小さな休止リンパ球の代表的な形態を有するCD1c+B細胞は、単離されたCD1c+PBMCの細胞遠心分離スライドにおいて、見られ得る。高度に純粋なCD1c+BDCは、CD1c+細胞の濃縮の前に、CD19+B細胞が磁気的にPBMCから除去された場合に、得られる。

BDCA−2+およびBDCA−3+血液DCの表現型を、PE結合体化mAbおよびFITC結合体化mAbを用いる二色免疫蛍光によって分析した。CD1c+血液DCの分析のために、B細胞の排除のために、CD19−Cy5を使用して、三色染色を実施した。表現型分析の結果を表2に示し、そして以下のように要約し得る:血液DCサブセットのいずれも、CD1a、CD8、CD15、CD16、CD19、CD20、CD23、CD25、CD27、CD34、CD61、CD69、CD7.1、CD77、CD80、CD83、グリコホリンA(GPA)、TCRαβ、AC133、IgD、IgMおよびCMRF−56抗原を発現しない。全ての血液DCサブセットが、CD43、CD44、CD54およびMHCクラスI分子を、類似のレベルで発現する。BDCA−2+血液DCは、これらがCD45RAおよび少量のCD10除いて、CD13、CD40、CD45RO、CD56を発現しない点、ならびにこれらが、より高レベルのCD4を除いて、より低レベルのCD18、CD38、CD58、CD98、CD116およびCLAを発現する点で、他の2つのサブセットとは異なる。CD1c+血液DCは、これらが、より低レベルのCD62Lを除いて、CD2、より高レベルのMHCクラスII分子を発現する点、ならびにこれらが、FcレセプターCD32、CD64およびFcεR1を発現する点で、他の2つのサブセットとは異なる。恐らく、Fcレセプター発現に起因して、CD1c+血液DCはまた、IgG、κおよびλに対してポジティブである。さらに、いくらかのCD1c+DCは、CD14およびCD11bに対してポジティブであり、これによって、発現レベルは、CD1c発現とCD2発現との両方のレベルに逆に相関する。BDCA−3+血液DCは、これらがCD36をずっと低いレベルで発現する点、およびこれらが低レベルのCD5を発現するようである点で、他の2つのサブセットとは異なる。最後に、CD11cおよびCD123を除いて、少なくとも1つのさらなる抗原(CD33)は、3つ全てのサブセットの識別に有用である:CD33は、BDCA−2+DCにおいて低レベルで、BDCA−3+DCにおいて中程度のレベルで、そしてCD1c+DCにおいて高レベルで、発現される。

新たに単離されたCD1c+血液DCおよびBDCA−3+血液DCを、いかなるサイトカインをも補充しない培地中で1日間培養し、そして新たに単離されたBDCA−2+血液DCを、IL−3およびCD40 mAbを補充した培地中で、CD32トランスフェクトした線維芽細胞上で2日間培養した。この培養期間の後に、細胞を、CD1a、CD80、CD83、CD86およびHLA−DRの発現に関して分析した。比較のために、いわゆる「未熟」Mo−DC(単球を7日間、GM−CSFおよびIL−4の存在下で培養することによって生成した)およびいわゆる「成熟」Mo−DC(「未熟」Mo−DCの、3日間のTNF−αの存在下での培養によって生成した)もまた、含めた。Sallustoら(1995)J.Exp.Med.182:389−400;ならびにSallustoら(1998)J.Immunol.28:2760−2769。図10に示すように、「未熟」Mo−DCおよび「成熟」Mo−DCとは対照的に、血液DCサブセットのいずれも、この培養期間の後に、CD1aを発現しない。しかし、同時刺激分子であるCD80およびCD86、DC活性化抗原CD83(Zhouら(1995);Zhouら(1992)J.Immunol.149:735−742;およびZhouら(1996)Proc.Natl.Acad.Sci.USA 93:2588−2592)、ならびにHLA−DR分子は、培養の際に、3つ全ての血液DCサブセットにおいて、成熟Mo−DCと比較して類似のレベルまで、アップレギュレートされる。これらの結果は、3つ全ての血液DCサブセットをIL−3、IL−4およびGM−CSFで補充した培地中で2日間培養した別の実験と有意には異ならなかった。血液および扁桃由来のCD11-cCD4+lin-DCに関して以前に示されたように、BDCA−2+血液DCは、IL−3を含まない培地中で培養される場合に、急速に死滅する。
(新たに単離されたCD1c+、BDCA−2+およびBDCA−3+血液DCのエンドサイトーシス能力)
精製したCD1c+、BDCA−2+およびBDCA−3+血液DC、ならびにコントロールとして、精製したCD3+T細胞のエンドサイトーシス能力を、これらの細胞を37℃で、LYの存在下で培養し、そして種々の時間の後のLYの取込みをフローサイトメトリーによって分析することによって、試験した。図11に示すように、精製したCD3+T細胞とは異なり、精製したCD1c+血液DC、BDCA−3+血液DC、およびいくらかの程度まではBDCA−2+血液DCもまた、LYをエンドサイトーシスする能力を有する。類似の結果が、FITC−デキストランを使用して得られた。全ての血液DC集団のエンドサイトーシス能力は、Mo−DCと比較した場合に、ずっと低い。

(材料および方法):
BDCA−2+BDCA−4+BDCおよびBDCA−2でトランスフェクトしたかまたはトランスフェクトしていないU937細胞における、細胞質ゾルカルシウムの測定。BDCA−2+BDCA−4+血液DCおよびBDCA−2でトランスフェクトしたかまたはトランスフェクトしていないU937細胞を、Indo−1 AM(Sigma,St.Louis,MO)とともに、Valituttiら(1993)Eur.J.Immunol.23:790−795によって記載されるように、装填した。抗BDCA−2(AC144、IgG1)または抗BDCA−4(AD5−17F6、IgG1)mAbを、それぞれ新たに単離されたBDCA−2+BDCA−4+BDCおよびBDCA−2でトランスフェクトされたかまたはトランスフェクトされていないU937細胞に添加し、続いて架橋剤としてラット抗マウスIgG1 mAb(X56)を添加したか、または添加しなかった。細胞を、フローサイトメーターで分析して、Ca2+フラックスを検出した。生存細胞(散乱基準に基づく)およびIndo−1で標識した細胞(405nm対525nmの発光スペクトルに基づく)のみを、この分析に含んだ。

CD4+CD123brightCD11c-プラスマ細胞様DCは、エンベロープされたウイルス、細菌、および腫瘍細胞に応答して、主要なI型インターフェロンプロデューサーであることが示された。Fitgerald−Bocarslyら(1993)Pharmacol.Ther.60:39−62;Siegalら(1999)Science 284:1835−1837;Cellaら(1999)Nature Med.5:919−923。この理由によって、これらはまた、天然I型インターフェロン産生細胞(NIPC)と呼ばれてきた。プラスマ細胞様DCは、BDCA−2およびBDCA−4を発現する。図14に示すように、プラスマ細胞様DCにおける表面BDCA−2の、特定のmAbとの連結、続いて二次架橋mAb(ラット抗マウスIgG1、X56)との連結は、インフルエンザウイルス株PR8(5HAU/ml)での刺激に応答して、血液または扁桃由来の、免疫磁気的に精製したプラスマ細胞様BDCA−2+BDCA−4+DCによって、I型インターフェロンの分泌を阻害する。抗BDCA−2、インフルエンザウイルスおよび架橋mAbを含む培養物における、I型インターフェロン産生のレベル(図14、AC144+RamG1+FLU)は、インフルエンザウイルス単独(図14、FLU)またはアイソタイプコントロールmAb(抗サイトケラチンmAb CK3−11D5、IgG1)、インフルエンザウイルスおよび架橋mAb(図14、CK3+RamG1+FLU)を含む培養物よりずっと低い。
BDCA−2発現およびBDCA−4発現のプラスマ細胞様(plasmacytoid)DCを、PBMC(図14A)または扁桃細胞(図14B)から、抗BDCA−4(AD5−17F6)結合体化微小ビーズを用いた直接的な磁気標識およびMACSによる標識細胞の富化によって単離した。単離したBDCA−2発現およびBDCA−4発現のプラスマ細胞様DCを、24時間培地中で、a)IL−3単独(図14、コントロール);b)IL−3、抗BDCA−2 mAb(AC144、IgG1)およびラット抗マウスIgG1 mAb(図14、AC144+RamGl);c)IL−3、抗BDCA−2 mAb(AC144、IgG1)、ラット抗マウスIgG1 mAb、およびインフルエンザウイルス菌株PR8(図14、AC144+RamGl+FLU);d)IL−3およびインフルエンザウイルス菌株PR8(図14、FLU);e)IL−3、抗サイトケラチン mAb(CK3−11D5、IgG1)、ラット抗マウスIgG1 mAb、およびインフルエンザウイルス菌株PR8(図14、CK3+RamGl+FLU);およびf)IL−3、抗BDCA−4 mAb(AD5−17F6)、ラット抗マウスIgG1 mAb、およびインフルエンザウイルス菌株PR8(図14、17F6+RamGl+FLU)の存在下で培養した。
(BDCA−2は、リガンドをエンドサイトーシスし得るだけでなく、リガンドを抗原プロセシングおよびローディング区画に送達して、リガンドをCD4+クラスII制限T細胞に対して提示し得る。)
(材料および方法)
BDCA−2発現およびBDCA−4発現のプラスマ細胞様DCを、PBMCから、抗BDCA−4(AD5−17F6)結合体化微小ビーズを用いた直接的な磁気標識およびMACSによる標識細胞の富化によって単離した。単離したBDCA−2発現およびBDCA−4発現のプラスマ細胞様DCを、96ウェル平底マイクロプレート中IgG1 mAbs(0.2μg/ml)の存在下で、B13 T細胞クローンの4×104細胞/ウェルと共に共培養した(Lanzavecchiaら(1988)J.Exp.Med.167:345−352)。アッセイに使用したmAbは以下であった:AC144(抗BDCA−2、IgG1)、ZM3.8(抗ILT3、IgG1)およびCK3−11D5(抗サイトケラチン、IgG1)。48時間後、培養物を、(3H)チミジン(1μCi/ウェル)を用いてパルスし、そして取り込まれた放射能を、さらに16時間後に測定した。(3H)チミジン取り込み(cpm)を、培養物中の単離BDCA−2発現およびBDCA−4発現プラスマ細胞様DCの数に対してプロットした(図15)。

図16は、扁桃プラスマ細胞様CD123+ DCに対するBDCA−2およびBDCA−4の発現を示す。それぞれ、BDCA−2に対するFITC結合体化mAb(AC144)ならびにCD123およびBDCA−4に対するPE結合体化mAb(AD5−17F6)を用いた扁桃細胞の2色染色を示す。BDCA−2の発現がCD123brightプラスマ細胞様DCに対して制限され、一方、BDCA−4がまた数個の他の細胞に対して低いレベルで発現されることに注意する。
(BDCA−4 mAb(AD5−17F6)は、ニューロピリン1を認識する)
ニューロピリン1は、神経細胞ガイダンスを媒介するコラプシン/セマホリン(semaphorin)ファミリーに対するレセプターである。ニューロピリン1はまた、血管内皮増殖因子に対するアイソフォーム特異的レセプターとして内皮細胞および腫瘍細胞によって発現される。しかし、以前は、ニューロピリン1が血液および扁桃のプラスマ細胞様 DCに対して発現されること、およびニューロピリン1が少なくとも新鮮な非培養血液においてプラスマ細胞様DCに対して優れたマーカーを示すことは、知られていなかった。
ニューロピリン1を、抗BDCA−4 mAb AD5−17F6(抗NRP−1(ML))を用いて、トランスフェクトされていないPAE細胞(P)およびニューロピリン1をトランスフェクトしたPEA細胞(NP)の細胞溶解物から免疫沈降した(Sokerら(1998)Cell 92:735−745)。沈降したタンパク質を、SDS−PAGEおよびBDCA−4特異的mAb AD5−17F6(ML)またはニューロピリン1特異的mAb(Shay Soker、Children's Hospital、Boston、MA(S)より)を用いるウエスタンブロットにより分析した。
(ポリI:Cでの刺激に応答した精製BDCA−2+BDCA−4+BDCによるI型インターフェロン(IFN−α)の産生は、抗BDCA−2 mAbを用いてBDCA−2を誘発することによって阻害される)

CD11c-CD123brightプラスマ細胞様DCを、BDCA−4微小ビーズを用いてヒト末梢血単核球から分離した。CD11c-C123brightプラスマ細胞様DC(1×106細胞/ml)を、10μg/mlのAC144 mAbまたはマウスIgG1 mAb(CF6B、抗TPO)を用いて、RPMI、10% FCS、l0mM HEPES、50μM 2−ME、20μg/ml ジェネティシン中、37℃で30分間インキュベートした。20μg/mlのヤギ抗マウスIgG(Chemicon International)を添加し、そして細胞を、再び37℃で30分間インキュベートした。これらの細胞を、20μgのポリI:C(Sigma)の有りまたは無しで、37℃で24時間培養した。細胞上清を収集し、そしてIFN−α濃度を、ELISA(Endogen)によって決定した。アッセイの感度は、3pg/mlである。

(核酸配列およびアミノ酸配列)
BDCA−2をコードするcDNAを、COS細胞における発現クローニングによって得た。図5は、BDCA−2のアミノ酸配列(発現される6個のエキソンを有するアイソフォーム)を示す。BDCA−2は、C型レクチンII型膜タンパク質である。このようなレクチンは、例えば、Batesら(1999)J.Immunol.163:1973−1983に記載される。既知のC型レクチンに対するBDCA−2の比較を、実施例20に示す。
(BDCA−2およびBDCA−2スプライス改変体のエキソン/イントロン構造)
BDCA−2スプライス改変体に関する情報を、開始コドンの前のmRNA配列に相補的なプライマー(順方向プライマー:5'−TTGAAAGAACCACACCCCGAAAGT(配列番号7))および終止コドンの後ろのmRNA配列に相補的なプライマー(逆方向:5'−TAGCTTTCTACAACGGTGGATGCC(配列番号8))を用いたプラスマ細胞様DC由来のRT−PCR増幅、生じるフラグメントのプラスミドへのクローニング、および挿入物の配列決定によって得た。結果を図20に示す。比較に関して、マウス樹状細胞関連C型レクチン2(デクチン−2(Dectin−2))のスプライス改変体を、図21に示す。

(BDCA−3タンパク質分析)
BDCA−3発現HD−MY−Z細胞を、BDCA−3発現をアップレギュレートするために10ng/ml PMA(Sigma)および0.5mg/ml イオノマイシンを用いて24時間刺激した。3×107個のPMA/イオノマイシン刺激HD−MY−Z細胞を、1mg/ml Sulfo−NHS−LC−ビオチン(Pierce)を用いた15分間4℃でのインキュベーションによって表面ビオチン化し、そして2回洗浄した。細胞を、10% スクロースおよびプロテイナーゼインヒビター(フェニルメチルスルホニルフルオライド、ペプスタチンA、ロイペプチン、アプロチニン(Servaより))を補充した50mM Tris−HCl pH 8.0中に再懸濁し、そして0℃で超音波処理した(5×4秒、70% アウトプット)。超音波処理した細胞を、900×g、4℃で10分間遠心分離して、核およびインタクトな細胞を除去した。この上清を、30,000×g、4℃で2時間遠心分離して、精製細胞膜を得た。膜を、プロテイナーゼインヒビターおよび1% NP−40を補充した50mM Tris−HCl(pH8.0)、150mM NaCl中で1時間0℃でインキュベートすることによって溶解した。不溶性膜フラグメントを、30,000×g、4℃で遠心分離することによって除去した。上清に対して、MnCl2およびCaCl2を、各々1mMの終濃度で添加した。溶解物を、ConA Sepharoseカラム(1ml)上で吸着させ、そして結合タンパク質を、10mlの溶出緩衝液(0.5M D(+)マンノース、20mM Tris−HCI(pH7.4)、0.5M NaCl、1% NP−40)を用いて溶出し、そしてCetriprep−10遠心分離濃縮器(Amicon)を用いて1mlの容積に濃縮した。
