Abstract
【解決手段】本発明のトリュフ栽培用の培地は、米糠、ふすま、及びブドウ糖のうち少なくとも1つを含んだ糖類と、牛蒡、ナッツ類、及びベリー類のうち少なくとも1つを含んだ食用材料と、の混合物である。その混合物は、糖類の重量を1としたときに食用材料の重量が0.5〜5の比率になるように混ぜ合わせてなることを特徴とする。なお、食用材料は、粒状に粉砕された牛蒡であることが好ましい。この培地にトリュフ種菌を注入して培養させたトリュフ菌糸体及びトリュフ子実体を提供することができる。

米糠、ふすま、及びブドウ糖のうち少なくとも1つを含んだ糖類と、
牛蒡、ナッツ類、及びベリー類のうち少なくとも1つを含んだ食用材料と、
を、前記糖類の重量を1としたときに前記食用材料の重量が0.5〜5の比率になるように混ぜ合わせてなるトリュフ栽培用の培地。
本発明の実施例1の培養条件として、米糠0.5kg、牛蒡0.5kgと、を良く混ぜ合わせて培地とし、この培地にトリュフ菌子体10g、を注入してさらに混合し、無菌容器に移して無菌環境下のクリーンベンチ内に20℃の温度で60日間保管した。60日間の培養後、この培養物1gを試験管(20ml容量)に採取し、80%濃度のメタノール10mlを加えて、100〜105℃にて30分間ホットプレート中で加熱抽出した。この抽出液を取り出し、8000rpmで10分間、遠心分離操作を行って得られた上澄液を回収した。回収した上澄液は−20℃で冷凍保存した。
本発明の実施例2の培養条件として、培地を用意する際の米糠と牛蒡との重量を、米糠0.5kg、牛蒡1.0kgと、に設定した点のみが実施例1と異なり、それ以外の点は実施例1と同じであり、その説明は省略する。
本発明のトリュフ培養物の作用効果を確認するために従来方法でもトリュフ培養物を作成し、これを比較例とした。具体的には、比較例の培地は、米糠0.5kgと大鋸屑1.0kgとを混合させたものであり、それ以外の点は実施例1と同じであり、その説明は省略する。
ORAC(Oxygen Radical Absorbance Capacity:活性酸素吸収能力)は1992年に米国農務省(USDA)と国立老化研究所(National Institute on Aging)の研究者らにより開発された抗酸化力を示す指標である。このORACの分析法は、特に食品中の抗酸化力の分析に優れており、この分析法によるORACのデータベースは、野菜や果物などの食品素材や加工食品に至るまで充実している。
一定の活性酸素種を発生させ、それによって分解される蛍光物質の蛍光強度を測定し、経時的に減少する蛍光強度の曲線を描いた場合、この反応系に抗酸化物質が共存すると蛍光物質の蛍光強度の減少速度が遅延する。この測定原理に基づき、検体(もしくは標準物質)存在下での蛍光強度の曲線下面積AUC(Area Under the Curve)と、検体等が存在しない状態(ブランク状態)でのAUCとの差(net AUCと呼ぶ。)を算出する。検体のnet AUCについては、濃度既知の標準物質(Trolox 登録商標)のnet AUCに対する相対値を求める。その相対値を基にTrolox濃度に換算して検体の活性酸素吸収能力とする。なお、net AUCの単位は、μmol TE/gである。

- 米糠、ふすま、及びブドウ糖のうち少なくとも1つを含んだ糖類と、
牛蒡、ナッツ類、及びベリー類のうち少なくとも1つを含んだ食用材料と、
を、前記糖類の重量を1としたときに前記食用材料の重量が0.5〜5の比率になるように混ぜ合わせてなるトリュフ栽培用の培地。 - 前記食用材料が粒状に粉砕された牛蒡であることを特徴とする請求項1に記載のトリュフ栽培用の培地。
- 請求項1又は2に記載の培地にトリュフ種菌を注入して培養させたトリュフ菌糸体。
- 請求項1又は2に記載の培地にトリュフ種菌を注入して培養させたトリュフ子実体。
