Description
本発明は、人工多能性幹(以下、iPSという)細胞の樹立効率の改善方法に関する。より詳細には、本発明は、体細胞の核初期化工程において低酸素条件下で細胞を培養することを含む、iPS細胞の樹立効率の改善方法に関する。
近年、マウスおよびヒトのiPS細胞が相次いで樹立された。Yamanakaらは、Fbx15遺伝子座にネオマイシン耐性遺伝子をノックインしたレポーターマウス由来の線維芽細胞に、Oct3/4, Sox2, Klf4及びc-Myc遺伝子を導入し強制発現させることによって、iPS細胞を誘導した(1, 2)。Okitaら(3)は、Fbx15よりも多能性細胞に発現が限局しているNanogの遺伝子座に緑色蛍光タンパク質(GFP)及びピューロマイシン耐性遺伝子を組み込んだトランスジェニックマウスを作製し、該マウス由来の線維芽細胞で上記4遺伝子を強制発現させ、ピューロマイシン耐性かつGFP陽性の細胞を選別することにより、遺伝子発現やエピジェネティック修飾プロファイルが胚性幹(ES)細胞とほぼ同等のiPS細胞(Nanog iPS細胞)を樹立することに成功した。同様の結果が他のグループによっても再現された(4, 5)。その後、c-Myc遺伝子を除いた3因子によってもiPS細胞を作製できることが明らかとなった(6)。

本発明の目的は、iPS細胞の樹立効率を改善する手段を提供することであり、それを用いた効率的なiPS細胞の製造方法を提供することである。
[1] iPS細胞の樹立効率の改善方法であって、体細胞の核初期化工程において低酸素条件下で細胞を培養することを含む、方法。
[2] 雰囲気中の酸素濃度が1-10%の範囲内である、上記[1]記載の方法。
[3] 雰囲気中の酸素濃度が1-5%の範囲内である、上記[2]記載の方法。
[4] 核初期化物質が、以下の物質、あるいはそれらをコードする核酸である、上記[1]-[3]のいずれかに記載の方法;
(i) Oct3/4及びKlf4、又は
(ii) Oct3/4及びc-Myc、又は
(iii) Oct3/4、Klf4及びSox2、又は

本発明は、体細胞の核初期化工程において低酸素条件下で細胞を培養することを含む、iPS細胞の樹立効率の改善方法を提供する。
本明細書において用語「低酸素条件」とは、細胞を培養する際の雰囲気中の酸素濃度が、大気中のそれよりも有意に低いことを意味する。具体的には、通常の細胞培養で一般的に使用される5-10% CO2/95-90%大気の雰囲気中の酸素濃度よりも低い酸素濃度の条件が挙げられ、例えば雰囲気中の酸素濃度が18%以下の条件が該当する。好ましくは、雰囲気中の酸素濃度は15%以下(例、14%以下、13%以下、12%以下、11%以下など)、10%以下(例、9%以下、8%以下、7%以下、6%以下など)、または5%以下(例、4%以下、3%以下、2%以下など)である。また、雰囲気中の酸素濃度は、好ましくは0.1%以上(例、0.2%以上、0.3%以上、0.4%以上など)、0.5%以上(例、0.6%以上、0.7%以上、0.8%以上、0.9%以上など)、または1%以上(例、1.1%以上、1.2%以上、1.3%以上、1.4%以上など)である。
哺乳動物由来(例、マウス、ヒト)の生殖細胞以外のいかなる細胞も、本発明においてiPS細胞作製のための出発材料として用いることができる。例えば、角質化する上皮細胞(例、角質化表皮細胞)、粘膜上皮細胞(例、舌表層の上皮細胞)、外分泌腺上皮細胞(例、乳腺細胞)、ホルモン分泌細胞(例、副腎髄質細胞)、代謝・貯蔵用の細胞(例、肝細胞)、境界面を構成する内腔上皮細胞(例、I型肺胞細胞)、内鎖管の内腔上皮細胞(例、血管内皮細胞)、運搬能をもつ繊毛のある細胞(例、気道上皮細胞)、細胞外マトリックス分泌用細胞(例、線維芽細胞)、収縮性細胞(例、平滑筋細胞)、血液と免疫系の細胞(例、Tリンパ球)、感覚に関する細胞(例、桿細胞)、自律神経系ニューロン(例、コリン作動性ニューロン)、感覚器と末梢ニューロンの支持細胞(例、随伴細胞)、中枢神経系の神経細胞とグリア細胞(例、星状グリア細胞)、色素細胞(例、網膜色素上皮細胞)、およびそれらの前駆細胞(組織前駆細胞)等が挙げられる。細胞の分化の程度に特に制限はなく、未分化な前駆細胞(体性幹細胞も含む)であっても、最終分化した成熟細胞であっても、同様に本発明における体細胞の起源として使用することができる。未分化な前駆細胞の例としては、たとえば神経幹細胞、造血幹細胞、間葉系幹細胞、歯髄幹細胞等の組織幹細胞(体性幹細胞)が挙げられる。
本発明において「核初期化物質」とは、体細胞からiPS細胞を誘導することができる任意の物質(単数又は複数)を指し、タンパク質性因子またはそれをコードする核酸(ベクターに組み込まれた形態を含む)、あるいは低分子化合物等のいかなる物質から構成されてもよい。核初期化物質がタンパク質性因子またはそれをコードする核酸の場合、好ましくは以下の組み合わせが例示される(以下においては、タンパク質性因子の名称のみを記載する)。
(1) Oct3/4, Klf4, c-Myc
(2) Oct3/4, Klf4, c-Myc, Sox2(ここで、Sox2はSox1, Sox3, Sox15, Sox17またはSox18で置換可能である。また、Klf4はKlf1, Klf2またはKlf5で置換可能である。さらに、c-MycはT58A(活性型変異体), N-Myc, L-Mycで置換可能である。)

従来iPS細胞の樹立効率が低いために、近年、その効率を改善する物質が種々提案されている。よって前記核初期化物質に加え、これら樹立効率改善物質を体細胞に接触させることにより、iPS細胞の樹立効率をより高めることが期待できる。
実験系としてNanogレポーターを持つマウスを使用した。NanogレポーターはBACPAC Resourcesより購入したBAC(bacterial artificial chromosome)のNanog遺伝子座に緑色蛍光蛋白質(EGFP)とピューロマイシン耐性遺伝子がくみこまれたもの [Okita K. et al., Nature 448, 313-317(2007)]を使用した。マウスNanog遺伝子はES細胞や初期胚といった分化多能性細胞において特異的に発現する遺伝子である。また、このレポーターが陽性となったマウスiPS細胞はES細胞とほぼ同等の分化能力を持つことがわかっている。このNanogレポーターを有するNanogレポーターマウス [Okita K. et al., Nature 448, 313-317(2007)]から得られたマウス胎仔線維芽細胞(MEF)および尾部線維芽細胞(TTF)に対して、レトロウィルスにより遺伝子導入を行うことでiPS細胞を樹立し、NanogレポーターによりEGFPが発現しているコロニー数を計測することによりiPS細胞樹立効率を評価した。
Oct3/4とKlf4の2遺伝子導入のみでも低酸素培養法の効果が得られるかどうかを検討するための実験を行った。実験は、実施例1と同じNanogレポーターマウス由来のMEFを用いた。実施例1と同様にしてレトロウイルス発現ベクター(pMXs-Oct3/4, pMXs-Klf4)をMEFに感染させた。感染後4日目に、1x105個のMEFを、あらかじめフィーダー細胞を播いておいた100mmディッシュに播いた。以後2日ごとにES細胞用培地の交換を行った。
実施例1及び2で樹立したMEF由来のiPS細胞における未分化マーカーの発現を、Rever Tra Ace kit(Takara)を使用してRT-PCR解析を行うことにより調べた。使用した各プライマーの配列を配列番号:1〜18に示す。またRT-PCRの結果を図3に示す。4遺伝子(Oct3/4, Klf4, Sox2, c-Myc)、3遺伝子(Oct3/4, Klf4, Sox2)、または2遺伝子(Oct3/4, Klf4)を導入し、低酸素濃度下(5%、1%)で培養して樹立したiPS細胞は、いずれも、ES細胞特異的発現遺伝子であるOct3/4、Sox2、Klf4、c-Myc、Nanog、Rex1、ECAT1を発現しており、その発現量はマウスES細胞(RF8)や過去に4遺伝子で樹立したiPS細胞 [20D17: Nature, 448, 313-317 (2007)]と同等であることが示された。また、導入したOct3/4遺伝子(Oct3/4(Tg))の発現は認められなかったことから、サイレンシングが起こっていることが示された。以上の結果より、低酸素条件下で樹立した細胞はiPS細胞であることが確認された。
低酸素濃度下(5%)、Oct3/4およびKlf4で樹立したマウスiPS細胞 (527CH5-2)を用いて、Cell, 126, 663-676 (2006) に記載の方法に従ってテラトーマを形成させた。具体的には1×106個のiPS細胞を免疫不全マウスの皮下に注射し、4週間後に出現したテラトーマを単離した(図4上)。各テラトーマを切り刻んで4%フォルムアルデヒドを含有するPBS(-)で固定した。パラフィン包埋組織をスライスし、ヘマトキシリン・エオシンで染色した。結果を図4下に示す。組織学的に見ると、腫瘍は複数の種類の細胞から構成されており、軟骨組織、内胚葉性上皮組織、筋肉組織、角化上皮組織が認められたことから、iPS細胞の多能性が証明された。
実施例1および2において、低酸素濃度下(5%)、2遺伝子、3遺伝子または4遺伝子導入により樹立したiPS細胞をICRマウス由来の胚盤胞にマイクロインジェクションした結果、アダルトキメラを作出した。結果を図5に示す。
iPS細胞株の核型は実施例1及び2における低酸素処理(521AH5-1及び527CH5-1)後のものであり、これらの細胞株は正常な核型を示した(図18)。
低酸素条件下(5%)での細胞培養時期および細胞培養期間がヒトiPS細胞の樹立効率に及ぼす影響について検討した。体細胞として成人皮膚由来線維芽細胞(adult HDF)を用いた。低酸素培養のタイムスケジュールを図6に示す。まず、Cell, 131, 861-872 (2007) に記載の方法に従い、レンチウイルスを用いて、マウスエコトロピックウイルスレセプターSlc7a1遺伝子をHDFに発現させた。これらの細胞(8×105個)に対して、Cell, 131, 861-872 (2007) に記載の方法に従い、全てヒト由来の4因子 (Oct3/4, Sox2, Klf4, c-Myc) または3因子 (Oct3/4, Sox2, Klf4) をレトロウイルスで導入した。ウイルス感染から6日後に細胞を回収し、フィーダー細胞上への蒔き直しを行った (4遺伝子導入の場合は1 x 105 個、3遺伝子導入の場合は5 x 105個/100 mmディッシュ)。フィーダー細胞にはマイトマイシンCで処理して、細胞分裂を止めたSNL細胞 [McMahon, A. P. & Bradley, A. Cell 62, 1073-1085 (1990)]を用いた。翌日から霊長類ES細胞培養用培地 (ReproCELL) に4 ng/mlのリコンビナントヒトbFGF(WAKO)を加えた培地で培養を行った。
実施例7で樹立した成人HDF由来のiPS細胞における未分化マーカーの発現を、Rever Tra Ace kit(Takara)を使用してRT-PCR解析を行うことにより調べた。使用した各プライマーの配列を配列番号:19〜36に、またRT-PCRの結果を図10に示す。4遺伝子(Oct3/4, Klf4, Sox2, c-Myc)または3遺伝子(Oct3/4, Klf4, Sox2)を導入し、低酸素濃度下(5%)で培養して樹立したiPS細胞は、いずれも、ES細胞特異的発現遺伝子であるOct3/4、Sox2、Klf4、c-Myc、Nanog、Rex1、GDF3およびESG1を発現しており、その発現量は、過去に4遺伝子で樹立したiPS細胞 [201B2: Cell, 131, 861-872 (2007)]と同等であることが示された。以上の結果より、低酸素濃度下で樹立した細胞はiPS細胞であることが確認された。
Oct3/4, Klf4, Sox2, c-Mycの4遺伝子を導入し、5%酸素濃度下で感染後7日目から40日目まで培養して樹立したヒトiPS細胞(70AH5-2、70AH5-6)をlow-binding dishに播き、Cell, 131, 861-872 (2007)に記載のように8日間培養し、胚様体(embryoid body:EB)を形成させた(100mmディッシュ)。胚様体を8日間培養後、内胚葉系細胞の分化マーカーであるα-fetoprotein (R&D systems)、中胚葉系細胞の分化マーカーであるsmooth muscle actin (DAKO)、Desmin (NeoMarkers)、Vimentin (Santa Cruz)、外胚葉系の分化マーカーであるβIII-tubulin (Chemicon)、GFAP (DAKO)の各抗体を用いた染色を行った。結果を図12に示す。染色によりそれらの発現が確認され、樹立されたヒトiPS細胞は三胚葉系への分化能を有することが確認された。
4遺伝子導入、5%酸素濃度下での培養により樹立したヒトiPS細胞(70AH5-2)を用いてテラトーマ形成能を調べた。ヒトiPS細胞(70AH5-2)を、リコンビナントヒトbFGF(4ng/ml)およびRhoキナーゼ阻害剤Y-27632(10μM)を含有する霊長類ES細胞培養用培地 (ReproCELL)中で培養した。1時間後、collagen IVで処理して細胞を採取後、遠心・回収し、Y-27632(10μM)を含有するDMEM/F12中に浮遊させた。コンフルエントになった細胞(100mmディッシュ)の1/4量をSCIDマウスの精巣内に注射した。9週間後、腫瘍を切り刻んで4%フォルムアルデヒドを含有するPBS(-)で固定した。パラフィン包埋組織をスライスし、ヘマトキシリン・エオシンで染色した。結果を図13に示す。組織学的に見ると腫瘍は複数の種類の細胞から構成されており、神経上皮組織、網膜上皮組織、骨様組織、平滑筋組織、及び内胚葉性上皮組織が認められたことから、iPS細胞の多能性が証明された。
annexin Vを使用したフローサイトメトリー解析により、低酸素培養は、マウスES細胞又は4因子を導入したMEFに対して保護作用を有さないことが確認された(図19)。さらに、低酸素培養は、マウスES細胞の増殖に対して効果を示さなかった(図20 a))。導入後1日目から4日目まで低酸素でインキュベーションしても、mockを導入したMEFの増殖に対しては有意な効果を有さなかったが、4因子を導入したMEFに対しては有意な効果を有していた(図20 b))。初期化プロセスにおける細胞の発現プロファイルを調べるために、マイクロアレイ解析及び定量的リアルタイムRT-PCRを行った。1日目から4日目まで低酸素及び正常酸素条件下にて培養した、4因子を導入したMEFのマイクロアレイ解析により、低酸素で処理した細胞では、ES細胞特異的遺伝子のうち73.2%(全1045遺伝子のうち765遺伝子)が上方制御されており、MEF特異的遺伝子のうち85.8%(全1142遺伝子のうち980遺伝子)が下方制御されていることが示された(図21 a)及びb))。さらに、定量的リアルタイムRT-PCR解析により、3日間低酸素処理すると、4因子を導入したMEFにおいて、内在性のOct3/4及びNanogの発現がそれぞれ、3.4倍及び2.1倍増加していることが確認された(図22 a)及びb))。
プラスミド導入によるiPS細胞の作製を、先に記載されているように行った(Okita, K, et al. Science 322, 949-953, (2008))。簡潔には、Nanog-GFP-IRES-Purorレポーターを有するMEFを、6ウェルプレートに、1.0x105細胞/ウェルになるように蒔いた (0日目)。1、3、5及び7日目に、細胞をpCX-OKS-2A及びpCX-c-Mycで形質導入し、9日目に、細胞をトリプシンで回収して、STOフィーダー細胞を蒔いておいた100mmディッシュに蒔き直した。25日目に、GFP陽性コロニーの数を数えた。低酸素処理のために、細胞を10日目から24日目まで、5%酸素下にて培養した。
いくつかの改変を加えて、先に記載されているように、piggyback (PB)転位による直接的な初期化を行った (Woltjen et al., Nature; 458:766-70, (2009))。簡潔には、Nanog-GFP-IRES-Purorレポーターを有するMEFを、6ウェルプレートに、1.0x105細胞/ウェルになるように蒔いた。24時間後、Fugene HD (Roche, Switzerland)を使用して、PB-TET-MKOS、PB-CA-rtTA Adv、及びPBトランスポゼース発現ベクターで細胞を形質導入した。24時間後、培地をドキシサイクリンを含む培地(1.5ug/ml)で置換した(0日目)。細胞を低酸素又は正常酸素条件下にて培養し、GFP陽性コロニーの数を12日目にカウントした。PB-TET-MKOS及びPB-CA-rtTA advは、Addgeneから得た(Addgeneプラスミド20910及び20959)。PBトランスポゼース構築物は、pBSII-IFP2-orf (Dr. Malcolm J. Fraser, Jr, University of Notre Dameから頂いた)からPCRで増幅し、CAGプロモーターで発現させる発現ベクター(pCX-EGFP)に挿入した。
Claims (6)
- 人工多能性幹細胞の樹立効率の改善方法であって、体細胞の核初期化工程において低酸素条件下で細胞を培養することを含む、方法。
- 雰囲気中の酸素濃度が1-10%の範囲内である、請求項1記載の方法。
- 雰囲気中の酸素濃度が1-5%の範囲内である、請求項2記載の方法。
- 核初期化物質が、以下の物質、あるいはそれらをコードする核酸である、請求項1から3のいずれかに記載の方法;(i) Oct3/4及びKlf4、又は
(ii) Oct3/4及びc-Myc、又は
(iii) Oct3/4、Klf4及びSox2、又は
(iv) Oct3/4、Klf4及びc-Myc、又は
(v) Oct3/4、Klf4、Sox2及びc-Myc。 - 核初期化工程において効率改善物質としてバルプロ酸が使用されるさらなる工程を含む、請求項1から4のいずれかに記載の方法。
- 低酸素条件下における体細胞の培養が、核初期化物質を接触させた後3日以上行われる、請求項1から5のいずれかに記載の方法。
