Abstract
【解決手段】検体中のグアイアコール産生菌の検出方法であって、次式:
[式中、Rは、-H、-OH、-C(O)H、-C(O)CH3、-COOH、C1-C3のアルキル基またはC1-C3のアルケニル基であり、該アルキル基およびアルケニル基は、-OH、-C(O)Hまたは-COOHで置換されていてもよい]で表される化合物の1種またはそれ以上を含有する好酸性菌用固体培地上で検体もしくはその希釈物を平板培養し、形成されたコロニーを検出することを特徴とする方法。

Description
で表される化合物の1種またはそれ以上を含む好酸性菌用固体培地上で検体もしくはその希釈物を培養し、形成されたコロニーを検出することを特徴とする、検体中のグアイアコール産生菌の検出方法を提供する。具体的には、従来の方法で必要とされていた前培養や定性分析のために必要な処理(ペルオキシターゼ法による呈色)を要せず、直接検出でき、かつ直接定量できる方法を提供する。好ましくは、化合物は、バニリン、バニリン酸、フェルラ酸、グアイアコール、4-ハイドロキシ-3-メトキシフェニルアクリルアルデヒド、4-ハイドロキシ-3-メトキシフェニルプロピオン酸、4-ハイドロキシ-3-メトキシフェニルメチルアルコール、メトキシハイドロキノン、4-ハイドロキシ-3-メトキシフェニルアセトアルデヒドおよび4-ビニルグアイアコールからなる群より選択される。さらに好ましくは、化合物は、バニリン、バニリン酸またはフェルラ酸である。
で表される、メトキシフェノール骨格を有する化合物の1種またはそれ以上を含有する好酸性菌用固体培地を用いる。
で表される、メトキシフェノール骨格を有する化合物の1種またはそれ以上を含有する好酸性菌用固体培地を含む。好酸性菌用固体培地は、滅菌されシャーレ中に作成された形で提供してもよく、混釈用にフラスコ、ボトルなどの中に作成された形で提供してもよい。メンブレンフィルター法用のキットは、メンブレンフィルターおよび濾過装置をさらに含んでいてもよい。キットはさらに、希釈列作成用のプレート、陽性対照用のグアイアコール産生菌および使用説明書を含むことができる。
バニリン、バニリン酸またはフェルラ酸を添加したYSG寒天培地を用いて、耐熱性好酸性菌(TAB)がコロニーを形成しうるか否かを調べた。使用した菌株は、A.acidoterrestris ATCC49025、A.acidiphilus JCM21417、A.acidocaldarius ATCC27009、A.pomorum JCM21459の4菌株である。このうち、A.acidoterrestris ATCC49025、A.acidiphilus JCM21417は、グアイアコールを産生する菌株として報告されている。
バニリン、バニリン酸またはフェルラ酸を添加したYSG寒天培地を用いて、培養温度によるA.acidoterrestris ATCC49025のコロニー検出率の相違を調べた。YSG培地にバニリン酸75ppmを加えて、実施例1と同様にして固体培地を作成し、同数のA.acidoterrestris ATCC49025を含む菌液を平板培地に塗沫して、45℃または30℃で培養した。対照として無添加のYSG培地を用いた。

で表される化合物の1種またはそれ以上を含む好酸性菌用固体培地上で検体もしくはその希釈物を培養し、形成されたコロニーを検出することを特徴とする、検体中のグアイアコール産生菌の検出方法。 - 好酸性菌用固体培地が、バニリン、バニリン酸、フェルラ酸、グアイアコール、4-ハイドロキシ-3-メトキシフェニルアクリルアルデヒド、4-ハイドロキシ-3-メトキシフェニルプロピオン酸、4-ハイドロキシ-3-メトキシフェニルメチルアルコール、メトキシハイドロキノン、4-ハイドロキシ-3-メトキシフェニルアセトアルデヒドおよび4-ビニルグアイアコールからなる群より選択される化合物の1種またはそれ以上を含有する、請求項1記載の方法。
- 好酸性菌用固体培地が、バニリン、バニリン酸またはフェルラ酸のいずれかを含有する、請求項1または2に記載の方法。
- 好酸性菌用固体培地が50ppm以上のバニリン酸、500ppm以上のバニリン、または25ppm以上のフェルラ酸を含有する、請求項1〜3のいずれかに記載の方法。
- 好酸性菌用固体培地が50ppm以上225ppm以下のバニリン酸を含有する、請求項1〜3のいずれかに記載の方法。

で表される化合物の1種またはそれ以上を含有する好酸性菌用固体培地からなる、グアイアコール産生菌検出培地。 - 好酸性菌用固体培地が、バニリン、バニリン酸、フェルラ酸、グアイアコール、4-ハイドロキシ-3-メトキシフェニルアクリルアルデヒド、4-ハイドロキシ-3-メトキシフェニルプロピオン酸、4-ハイドロキシ-3-メトキシフェニルメチルアルコール、メトキシハイドロキノン、4-ハイドロキシ-3-メトキシフェニルアセトアルデヒドおよび4-ビニルグアイアコールからなる群より選択される化合物の1種またはそれ以上を含有する、請求項15記載の培地。
- 好酸性菌用固体培地が、バニリン、バニリン酸またはフェルラ酸のいずれかを含有する、請求項15または16に記載の培地。
- 好酸性菌用固体培地が、50ppm以上のバニリン酸、500ppm以上のバニリン、または25ppm以上のフェルラ酸を含有する、請求項15〜17のいずれかに記載の培地。
- 好酸性菌用固体培地が、50ppm以上225ppm以下のバニリン酸を含有する、請求項15〜17のいずれかに記載の培地。
- 好酸性菌用固体培地が、50ppm以上75ppm以下のバニリン酸を含有する、請求項15〜17のいずれかに記載の培地。
