Description
本発明は、癌の治療のための遺伝子療法の分野に関する。一つの態様において、本発明は、インターロイキン-12(IL-12)を条件的に発現するための樹状細胞の遺伝子操作および治療のための細胞の使用に関する。別の態様において、本発明は、インターロイキン-12(IL-12)および/またはインターフェロン-アルファ(IFN-アルファ)を条件的に発現するための樹状細胞の遺伝子操作および治療のための細胞の使用に関する。

(e)IFN-γの対照レベルと試験レベルを比較し、ここで対照レベルに比べてIFN-γの発現、活性または両方の試験レベルの上昇は、それを必要としている該患者において治療法が有効であることを示す方法を提供する。

a. ヒト患者からインビトロで遺伝子操作したDC以外の免疫細胞またはTSCの投与前に得た、第一の生体試料中のインターフェロン-ガンマ(IFN-γ)の発現のレベルもしくは活性のレベルまたは両方を測定し、それにより対照レベルをもとめる段階;
b. 該患者に、インビトロで遺伝子操作したDC以外の免疫細胞またはTSCを腫瘍内投与する段階;
c. 該患者に、活性化リガンドの有効量を投与する段階;
d. 該患者から該活性化リガンドの投与後の時点で得た、第二の生体試料中のIFN-γの発現のレベルもしくは活性のレベルまたは両方を測定し、それにより試験レベルのデータをもとめる段階;および
e. IFN-γの対照レベルと試験レベルを比較し、ここで対照レベルに比べて試験レベルにおけるIFN-γの発現、活性、または両方のレベルの上昇を示すデータは、該患者において治療レジメンが有効であることを示す方法を提供する。
DC.RheoIL12はインビトロでリガンドRG-115830に応答して高レベルのIL-12p70を条件的に生成する
1.1 材料と方法
1.1.1 マウス
雌6〜8週齢のC57BL/6野生型およびC57BL/6-TgN(ACTbEGFP)1Osb/J EGFP Tgマウスを、Jackson Laboratory(Bar Harbor, ME)から購入し、マイクロアイソレーターケージ内で維持した。マウスを実験動物の適切な管理と使用のための推奨に従って扱った。
C57BL/6マウスと同系のB16黒色腫およびEL-4胸腺腫H-2b細胞株が以前に記載されている(Itoh et al., 1994)。細胞株をCM(10%熱不活化ウシ胎仔血清、100単位/mlペニシリン、100 μg/mlストレプトマイシン、および10mM L-グルタミンを補足したRPMI 1640;すべての試薬はInvitrogen, Carlsbad, CAから)中、5%CO2および37℃の加湿インキュベーター内で維持した。
DCを、以前に記載されているとおり(Tatsumi et al., 2003)、マウス骨髄から生成した。簡単に言うと、野生型またはEGFP TgマウスBMを、1000単位/ml組換えマウス顆粒球/マクロファージコロニー刺激因子および組換えmIL-4(Peprotech, Rocky Hill, NJ)を補足したCM中、37℃の加湿した5%CO2インキュベーター内で7日間培養した。次いで、CD11c+ DCを、特異的MACSTMビーズを製造者のプロトコル(Miltenyi Biotec, Auburn, CA)に従い用いて単離した。この様式で生成したCD11c+ DCは、形態学ならびにCD11b、CD40、CD80と、クラスIおよびII MHC抗原の同時発現に基づき>95%の純度であった。
対照アデノウイルスベクターrAd.Ψ5およびCMVプロモーターによって駆動されるmIL-12をコードするrAd.cIL12(Tatsumi et al., 2003)は、University of Pittsburgh Cancer Institute's Vector Core Facilityによって生成され、提供された。
第7日目の培養DCは未処理であったか、構成的(rAd.cIL12)もしくは誘導性(rAd.RheoIL12)プロモーターによって駆動されたマウスIL-12p70をコードする組換えAdで感染させたか、または一連のMOIにわたる模擬、対照ベクターrAdΨ5で感染させた。この後の様々な時点(0〜48時間)で、DCを活性化リガンド(10〜200μg/ml)非存在下または存在下でさらに24時間培養し、その後、特異的ELISAキット(BDPharMingen, San Diego, CA;低検出レベル=62.5pg/ml)を用いてIL-12p70分泌を分析した。いくつかの場合には、条件的サイトカイン産生のストリンジェンシーを識別するために、活性化リガンドであらかじめ処理した、rAd.RheoIL12で感染させたDC(すなわち、DC.RheoIL12)を洗浄してリガンドを除去し、対照培地中でさらに24時間培養し、その後、IL-12p70分泌を分析した。または、48時間後、感染させたDCを回収し、表現型およびIL-12p70の産生について、低検出レベルが62.5pg/mlの、ELISAキット(BD-PharMingen, San Diego, CA)を用いて分析した。
アデノウイルス感染したDCの表現型分析のために、マウス細胞表面分子(CD11b、CD11c、CD40、CD54、CD80、CD86、H-2Kd、I-Ad(すべてBD-PharMingenから))に対するPEまたはFITC結合mAbおよび適当なアイソタイプ対照を用い、フローサイトメトリー分析を、FACscan(Becton Dickinson, San Jose, CA)フローサイトメトリー測定器を用いて実施した。
1.2.1 Rheo-IL12で感染させたマウスBM由来DCは、インビトロでリガンド処理した場合、高レベルのIL-12p70を条件的に産生する
インビトロで遺伝子操作した樹状細胞の動物への腫瘍内投与
2.1 方法と材料
2.1.1 B16腫瘍モデル
第0日目に、B6マウスの右側腹部に1×105個のB16黒色腫細胞を皮下に注射した。第7日目に、腫瘍は約20〜30mm2のサイズに達し、マウスをPBSまたは全量50μlのPBS中1×106個の対照およびアデノウイルス導入(MOI=100)DCのi.t.注射により処置した。また、表示のとおり、マウスに200μg RG-115830(50μl DMSO中)とDMSO担体対照のi.p.注射も、DC投与後0時間、24時間または48時間の時点で開始して行った。開始後、マウスに合計5日連続でRG-115830をこの用量で1日1回腹腔内注射した。さらなる実験において、リガンド投与をDC注射の日に開始し、DC注射後の1、3または5日目に停止して、IL-12p70導入遺伝子プロモーションを早期停止すると、このアプローチの治療上の利益が低減するかどうかを確認した。すべての場合に、腫瘍サイズを3日または4日に1回評価し、ノギスで測定した最大の直交する直径の積をもとめることによりmm2で記録した。データは平均腫瘍面積±SDで報告する。すべての動物コホートは1群あたり5匹のマウスを含んでいた。
腫瘍接種の25日後に1群あたり2匹の処置マウスの脾臓から、磁気ビーズ細胞選別(MACSTM; Miltenyi Biotec)を用いて、プールしたCD8+ T細胞を>95%の純度まで単離し、次いで1×104個の放射線照射(10,000rad)したB16またはEL-4腫瘍細胞と共に同時培養(1×105個/ウェル)した。48時間のインキュベーション後、培養上清を回収し、検出下限が31.5pg/mlの市販のELISA(BD-PharMingen)を用いて、IFN-γ放出について分析した。データは三重複測定の平均±SDで報告する。
処置を完全無作為デザインで適用した3群以上によるすべての実験を、まず一元配置または二元配置の要因ANOVAで分析した。得られるPが<0.05である場合、必要に応じて特定の対の対比を不等分散についてのウェルチの補正によるT検定で分析した。データを分布特性についてチェックし、適当な変換を適用した。脾細胞由来T細胞からのIFN-γ産生の分析を、正確なクラスカル-ウォリス検定により行った。クラスカル-ウォリス検定のP値が<0.05の場合、演繹的対比をウィルコクソン検定により評価した。治療的単一腫瘍接種マウス処置モデルの分析を、混合線形モデルで行った。データを対数変換し、マウス内共分散を推定し、処置の固定効果を無作為マウス効果に対して調節した。1時点で群の対を比較するための生のP値を、ブーツストラップ法の標本再抽出により調節した。腫瘍拒絶率を、処置群、観察日、およびそれらの相互作用を組み込んだ、一般線型モデル(二項連結による)にあてはめた。
2.2.1 DC.cIL12単独またはRG-115830のi.p.投与と組み合わせてのDC.RheoIL12の腫瘍内投与は、定着したs.c.B16黒色腫病変の退行を促進する
B16黒色腫細胞(1×105個)を同系H-2b B6マウスの左側腹部にs.c注射し、定着させた。第7日目に、マウスを5匹ずつのコホートに無作為割り付けし、コホートの平均腫瘍サイズは約20〜30mm2であった。次いで、マウスにPBSまたは全量50μlのPBS中106個のDC(インビトロでrAd.Ψ5、rAd.cIL12またはrAd.RheoIL12により48時間あらかじめ感染させた)を腫瘍内注射した。また、マウスにDMSOまたはRG-115830(DMSO中)の腹腔内注射も、DC投与時(すなわち、処置の第1日目)またはDC投与後24時間もしくは48時間(すなわち、処置の第2日目)に行った。図3Aおよび3Bに示すとおり、マウスのRG-115830単独またはRG-115830非存在下でのDC.RheoIL12による処置では、いかなる治療上の利益も得ることはできなかった。顕著な対照をなして、DC.cIL-12またはDC.RheoIL-12+RG-115830(DC注射の24時間以内に提供し、5日間のリガンド投与も一緒に行った)で処置した腫瘍は、その後の3週間でサイズが縮小した。これらの治療は、腫瘍サイズ測定に基づき、統計学的に識別不能で、これらそれぞれの場合に100%(マウス5匹中5匹)の腫瘍退行率を示した。興味深いことに、RG-115830投与が腫瘍内DC注射の48時間後(この物質がインビトロでDC.RheoIL-12からのIL-12p70産生を効率的に促進しうる時点、図2B参照)まで遅れると、DC.RheoIL12に基づく治療は腫瘍成長速度のわずかな阻害をもたらした(第10日目以降のすべての時点でP<0.05)が、すべての動物は進行を示し、第30日目までに屠殺が必要であった(図3A)。これは、腫瘍内DC.IL12処置の治療上の利益は、主に早期の時点でのIL-12p70産生(おそらくは腫瘍病変および/または流入領域リンパ節内で起こっている)に決定的に依存していることを示唆している。
発明者らの以前の報告(Tatsumi et al., 2003)は、IL-12遺伝子のDCへの挿入は、インビボで、腫瘍微環境に注入した後のこれらの細胞の生存増強と、その結果としてのこれらの細胞が抗腫瘍CD8+ T細胞をクロスプライミングし、おそらくは循環中のエフェクターT細胞を腫瘍微環境に動員する能力を促進すると示唆した。したがって、次の試みは、DC注射の48時間後に開始した(RG-115830のi.p.投与により)DC-RheoIL12療法の失敗は、DCが腫瘍微環境にとどまることができないため、これらの細胞が腫瘍流入領域LNに移動することができないため、および/または特異的CD8+ T細胞が処置の結果としてクロスプライミングを受けられないためであるかどうかを識別することであった。図3Aに概略を示した実験を、DC生成のための骨髄供給源としてEGFP Tg (H-2b)マウスを用いた以外は、1コホートあたり2匹のマウスをDCの腫瘍内注射の72時間後に屠殺して繰り返した。腫瘍およびLNを切除し、組織切片を蛍光顕微鏡によるEGFP+ DCの分析のために調製した。残りのマウス3匹/コホートを第25日目まで経過観察し、第25日目の時点で屠殺し、プールした脾細胞をB16特異的CD8+ T細胞応答の分析のために単離した。
注入DCの生存度がリガンド注入のタイミングに明らかに依存していることを考慮して、発明者らは、RG-115830によって0時間の時点で活性化されたDC.RheoIL12を投与したマウスでは、それよりも後に活性化されたものに比べて、特異的CD8+ T細胞のクロスプライミングの程度がすぐれていると予測していた。興味深いことに、これは0時間(DC.RheoIL-12、第1日目〜第5日目)と48時間(DC.RheoIL-12、第3日目〜第7日目)のDC.RheoIL-12コホートを比べると確かに観察されたが、0時間と24時間(DC.RheoIL-12+L、第2日目〜第6日目)を比べた場合には観察されなかった(図5A)。事実、関連するB16と無関係のEL-4腫瘍標的に対するインビトロでの脾臓CD8+ T細胞応答(IFN-γ分泌)は、これらのコホート両方で同等であり、これらはそれぞれDC.cIL12で処置したマウスで検出されたものに近づいた。全体に、これらのCD8+ T細胞応答特性は、治療転帰に直接相関するようであった(図3A)。図5Aと同様、特異的B16腫瘍細胞と無関係のMC38に対するIFN-γ産生による脾臓T細胞応答は、治療転帰に相関していた。
治療効果に対するリガンドの投与経路/用量の比較
3.1 方法と材料
B16黒色腫を同系B6マウスの右側腹部に7日間、皮下に定着させた。第7日目に、106個のDC.SP1-IL12(図10の比較で特定された最適なスイッチ)を腫瘍内に(i.t.)注入した。活性化リガンド(RG-115932)を、i.p.注入、Labrasol中の経口胃管栄養法、または適宜に提供する薬物含有飼料のいずれかで、DC注入の前日に開始して(その後1日1回6日間)提供した。各コホートは5匹の動物を含み、腫瘍成長を3〜4日に1回モニターし、平均サイズ(直交する測定値の積に基づくmm2)で報告した。各コホートの処置を以下に記載する。
結果は、任意の用量または任意の経路でのリガンド単独の投与は、B16腫瘍成長にまったく影響がなかったことを示している(図6)。DC-SP1 i.t.治療はB16の成長を制御する上で有効であるが、リガンドが適用される場合に限り、リガンド投与のすべての経路はいくらかの有効性を生じる。i.p.投与したリガンドを用いてのDC-SP1 i.t.治療は、明らかなリガンド用量-腫瘍阻害反応パターンを示し、最適な抗腫瘍効果はリガンド用量>30mg/kg/日で得られた。DC-SP1 i.t.治療に30mg/kg/日の用量で適用したリガンドは、i.p.注入、経口胃管栄養法、または飼料混合物のいずれで投与しても同等に有効であった。試料中で提供したより高用量のリガンドは、有効性がいくらか低かった。R鏡像異性体(RG-115932)だけがRTSを活性化することができるため、ラセミ混合物を含む飼料は約20〜22.5mg/kg/日の活性鏡像異性体を提供するにすぎない。これに関して、飼料によるコホートの抗腫瘍効果は、10および30mg/kg/日i.p.のRG-115932用量群で観察された効果の間にはいるため、飼料からラセミ混合物AD(すなわち、約20〜22.5mg/kg/日の活性鏡像異性体RG-115932)を投与した動物で観察された腫瘍退行は、純粋なRG-115932で見られたi.p.用量反応と一致する。これらのデータは、リガンドの経口投与は治療効果を誘導する上で有効であることを示唆している。リガンドの経口投与の利用可能性は、患者に対する治療の負担を軽減するであろう。
本実施例において、樹状細胞を骨髄から単離し、図7に示すアデノウイルス作成物を形質導入し、同系頭蓋内GL261神経膠腫を有するマウスに遺伝子操作した樹状細胞を腫瘍内注入し;かつRG-115830を腹腔内注入した。図7は、マウス頭蓋内神経膠腫GL261にRTS制御下またはRTSなしでIL-12および/またはIFN-アルファをコードするポリヌクレオチドを形質導入した樹状細胞を腫瘍内注入した結果を示す。データは、RTSのRG-115830リガンドによる活性化を通じてのIFN-アルファおよびIL-12のリガンド誘導性発現により、IFN-アルファ発現だけの場合と比べて、50日目の時点でGL261神経膠腫マウスの75%の生存が促進されたことを示している。さらに、RheoSwitchおよびリガンドによって提供される対照は生存増強を促進した。
第III期および第IV期黒色腫の対象における、RTS制御下でhIL-12を発現するよう遺伝子操作したアデノウイルス形質導入自己樹状細胞の腫瘍内注入の安全性、耐容性、導入遺伝子機能、および免疫学的効果を、以下に記載するものなどの方法により評価する。
組換えDNAをエクスビボでのアデノウイルスベクター形質導入により樹状細胞(DC)に導入する。組換えDNAを用いて、腫瘍内に注入した未成熟樹状細胞からヒトIL-12(p70)を発現させ、これは腫瘍微環境においてDCの生存を延長させて成熟を刺激し、その結果、DCは続いて流入領域リンパ節に移動することになる。これによってTヘルパー細胞はTh1型へと分化する方向に偏り、腫瘍抗原でのクロスプライミングによって腫瘍特異的細胞毒性T細胞の活性化も起こる。
ヒト対象由来の樹状細胞をエクスビボで形質導入し、腫瘍に注入する。DCを、ウイルス形質導入の前に、生存度、純度(典型的には>80%の細胞がDC表現型を示す)無菌性、マイコプラズマおよびエンドトキシンについて特徴付ける。ウイルス形質導入の後、細胞を繰り返し洗浄して、いかなる吸収されていないウイルスも除去する。最後の洗液の上清を、残留ウイルスの含有量についてPCRで試験する。DCをエクスビボでアデノウイルスベクター(非組み込み型ベクター)によって形質導入しており、腫瘍内注入およびその後の流入領域リンパ節への移動後のDCの寿命は短いため、ウイルスDNAが任意の非標的細胞に組み込まれるとは予想されない。DCのアデノウイルス形質導入に用いるプロトコルは80〜90%の形質導入を達成すると予想され、非常に効率的であると考えられる。
インビトロで生成した樹状細胞の各ロットを、微生物混入物(好気性および嫌気性菌、真菌およびマイコプラズマ)、ならびにエンドトキシンの存在について試験し、表現型および機能について特徴付ける。対象に注入するすべての樹状細胞は新鮮で、凍結保存することはない。
本明細書において用いる活性化薬物を以下の製剤の任意の1つに製剤する:
(1)100%Labrasol;
(2)(a)メントール、(b)チモール、(c)ユーカリプトール、(d)アスパルテーム、(e)サッカリンナトリウム、(f)クエン酸、(g)ペパーミント香料、(h)クリーム香料、(i)labrasolを含む、Listerine風味のLabrasol(Latitude Pharmaceuticals Inc., USA);
(3)Miglyol 812およびphospholipon 90G(Latitude Pharmaceuticals Inc., USA);または
(4)Miglyol 812、phospholipon 90GおよびビタミンEトコフェリルポリエチレングリコールスクシネート(Latitude Pharmaceuticals Inc., USA)。
様々な濃度および特定のプロトコルが想定されうるが、患者を治療するための一例は、RTSの制御下でhIL-12(ヒトインターロイキン12)を発現するよう遺伝子操作した形質導入自己樹状細胞(AdDC)を滅菌食塩水中に5×107個の濃度で懸濁し、その腫瘍内注入を、経口活性化薬物(RG-115932)との組み合わせで受ける患者を含むことになる。
第1日目の入院患者来診:第1日目に、基準時理学検査(生命徴候、体重、およびECOG状態を含む)を行う。基準時血清化学、尿検査、および血液学(安全性プロファイル)のために、尿を集め、血液を採取する。インビトロで遺伝子操作した樹状細胞の腫瘍内注入の約3時間前に、各対象に活性化薬物(コホート1−0.01mg/kg、0.3mg/kg、1.0mg/kg、および3mg/kg)を食事の直後に投与する。活性化薬物およびその主な代謝物の1回投与薬物動態を評価するために、血液を指定の間隔(投与前、AD投与後0.5、1、1.5、2、4、6、8、12、16、および24時間)で1日目に採取する。各対象に、RTSの制御下でhIL-12を発現するよう遺伝子操作した、アデノウイルス形質導入自己樹状細胞の1回腫瘍内注入を5×107細胞の濃度で投与する。対象を局所注射部位反応および/または過敏反応について注意深くモニターする。第2から14日目の入院患者来診: 第2から14日目に、各対象に活性化薬物を食事の直後に投与する。生命徴候および有害事象を第2から14日目まで1日1回収集する。第4日目±24時間の時点で、腫瘍および/または流入領域リンパ節の生検試料を約50%の対象から取り出し、hIL-12および細胞免疫応答を測定する。第8日目に、体重を測定する。第8日目±24時間の時点で、腫瘍および/または流入領域リンパ節の生検試料を、第4日目に生検を実施していない対象から取り出し、hIL-12および細胞免疫応答を測定する。第4日目±24時間および第8日目±24時間の時点で、アデノウイルスおよび/またはRTS成分に対して可能な抗体および細胞免疫応答の検定のために血液を採取する。他のサイトカインの発現がhIL-12導入遺伝子での処置によって影響されるかどうかを調べるために、血清サイトカインプロファイルも得る。第8日目に、基準時血清化学、尿検査、および血液学(安全性プロファイル)のために、尿を集め、血液を採取する。第8日目に、活性化薬物およびその主な代謝物の定常状態の薬物動態/ADMEを評価するために、血液を指定の間隔(投与前、AD投与後0.5、1、2、4、6、8、12、16、および24時間)で採取する。
エンドトキシン試験を、細胞培養物において回収時と最終生成物の放出前に行う。許容されるエンドトキシンレベルは体重1kgあたり<5EUである。非形質導入および形質導入樹状細胞をさらなる分析のために凍結保存する。
a. 明確に関係しているとは、治験薬の投与から妥当な時間的配列に従い、治験薬に対する公知の反応パターンに従い、かつプロトコルに対して適当である場合は、治験薬停止(積極的脱攻撃)後の改善および反復曝露(積極的再攻撃)後の反応の再出現によって確認され、かつ対象の臨床状態の公知の特徴または他の治療によって妥当に説明ができない、有害事象である。
b. 多分関係しているとは、治験薬の投与からの時間的配列に従い、治験薬に対する公知の反応パターンに従い、かつプロトコルに対して適当である場合は、脱攻撃後の改善によって確認され、かつ対象の臨床状態の公知の特徴または他の治療によって妥当に説明ができない、有害事象である。
c. ひょっとすると関係しているとは、治験薬の投与から妥当な時間的配列に従い、治験薬に対する公知の反応パターンに従うが、対象の臨床状態または他の治療によって生じている可能性がある、有害事象である。
d. 無関係とは、原因が治験薬に無関係であることを示す十分な情報がある、有害事象である。以下の変数の2つ以上が無関係の有害事象に適用される:1. 有害事象が治験薬投与後の妥当な時間的配列に従わない、2. 有害事象が対象の臨床状態または他の治療によって容易に説明される、3. 有害事象が用量低減または治療の停止後に軽減されない(観察間隔内での有害事象の軽減を予想するのが妥当であると仮定して)。
1. 制限性の毒性がこれまでまったくなく、
2. 対象の疾患が安定しているか、または改善の臨床もしくは主観的徴候を示しており、かつ
3. RheoSwitch(登録商標) Therapeutic Systemのアデノウイルス成分に対する抗体または細胞免疫応答の証拠がない。

- 請求項1〜14のいずれか一項記載のベクターを含む、インビトロで遺伝子操作した樹状細胞。
- IFN-アルファの機能を有するタンパク質を条件的に発現するベクターをさらに含むインビトロで遺伝子操作した樹状細胞であって、該ベクターが遺伝子スイッチをコードするポリヌクレオチドを含み、該ポリヌクレオチドが、(1)プロモーターに機能的に連結された、リガンド依存的転写因子をコードする少なくとも1つの転写因子配列、および(2)該リガンド依存的転写因子によって活性化するプロモーターに連結された、IFN-アルファの機能を有するタンパク質をコードするポリヌクレオチドを含む、請求項18記載のインビトロで遺伝子操作した樹状細胞。
- 請求項18記載のインビトロで遺伝子操作した樹状細胞の集団を含む、薬学的組成物。
- 腫瘍内、腹腔内または皮下投与に適した、請求項20記載の薬学的組成物。
- インビトロで遺伝子操作した樹状細胞の集団が少なくとも104個の細胞を含む、請求項20または21記載の薬学的組成物。
- インビトロで遺伝子操作した樹状細胞の集団が少なくとも107個の細胞を含む、請求項20または21記載の薬学的組成物。
- 哺乳動物の腫瘍治療用の医薬の製造における、請求項18または19記載のインビトロで遺伝子操作した樹状細胞の集団の使用。
- 腫瘍が良性腫瘍である、請求項24記載の使用。
- 腫瘍が悪性腫瘍である、請求項24記載の使用。
- 腫瘍が黒色腫である、請求項24記載の使用。

- 治療が腫瘍成長を低減させる、請求項24〜36のいずれか一項記載の使用。
- 治療が腫瘍成長を阻害する、請求項24〜36のいずれか一項記載の使用。
- 患者におけるインビトロで遺伝子操作した樹状細胞に基づく治療レジメンの有効性を判定するための方法であって:
(a)それを必要としている患者からインビトロで遺伝子操作した樹状細胞の投与前に得た、第一の生体試料中のインターフェロン-ガンマ(IFN-γ)の発現のレベルもしくは活性のレベルまたは両方を測定し、それにより対照レベルをもとめる段階;
(b)それを必要としている患者に、請求項18または19記載のインビトロで遺伝子操作した樹状細胞を投与する段階;
(c)それを必要としている患者に、活性化リガンドの有効量を投与する段階;
(d)それを必要としている患者からインビトロで遺伝子操作したDCおよび活性化リガンドの投与後に得た、第二の生体試料中のIFN-γの発現のレベルもしくは活性のレベルまたは両方を測定し、それにより試験レベルをもとめる段階;および
(e)IFN-γの対照レベルと試験レベルとを比較し、ここでIFN-γの発現、活性、または両方の対照レベルに対する試験レベルの上昇が、それを必要としている該患者において治療レジメンが有効であることを示す段階
を含む方法。 - それを必要としている患者がヒト患者である、請求項39記載の方法。
- 患者が癌患者である、請求項39記載の方法。
- 癌患者が黒色腫患者である、請求項41記載の方法。
- IFN-γのレベルをELISAで測定する、請求項39記載の方法。
