Abstract

コポリペプチド安定化エマルション
マイクロスケール、サブマイクロスケール、およびナノスケールの半径を有する液滴からなるシンプルおよびダブルエマルションを作る本発明の基本的な処理に使用可能な多数の組成的変更の可能性がある。本発明の実施の形態にかかる処理の基本要素は、第1の液(例、水)、異なる第2の非混和性の液(例、油)、および少なくとも2つの液のうちの1つ、または両者に顕著な溶解性(例、水溶性)を有する両親媒性のコポリペプチドである。コポリペプチドは少なくとも溶解性のある1つの液に付加され、2液間の界面を乱す非熱エネルギを液とコポリペプチドの系に供給し、液滴の形成と準安定性エマルションの生成を介して不可逆で最終的な界面表面領域を成長させる。適用される非熱エネルギは多数の異なる形態で供給でき、機械的剪断流動や、超音波、電磁界および電磁波、重力、ならびに濃度勾配を介して、または伸長流を引き起こす圧力低下を介しての供給が含まれる。液間の界面が乳化の起因となる非熱エネルギによって伸長されると、毛細管の不安定性による界面の不安定となり、より大きな液滴または膜のより小さな液滴への破壊をもたらす。液の種類、および界面を安定化するコポリペプチドの組成および構造により、これらの小液滴が他の液相の内側液滴を含んだり(例、水/油/水ダブルエマルションの形成)、含まなかったりする。
テトラヒドロフラン(THF)は、使用前に、窒素下でアルミナを装填したカラムを通過させて乾燥した(A. B. Pangborn他、Organometallics 15、1996年、p.1518)。分子量は、Wyatt DAWN EOS光散乱検出器とWyatt Optilab DSPを装備したSSI社のポンプにおいて、ゲル透過クロマトグラフィ/光散乱(GPC/LS)の2段階を60℃で行って得た。分離は、0.1MのLiBrのDMFを溶剤液として使用し、ポリペプチドの濃度を約5mg/mlとし、105、104および103オングストロームのPhenomenex社の5μmカラムで達成した。赤外線スペクトルは、ポリスチレン膜を使って校正されたPerkin Elmer社のRX1フーリエ変換赤外分光(FTIR)分光光度計で記録した。水素核磁気共鳴(1H-NMR)スペクトルは、Bruker社のAVANCE 400MHzの分光計で記録した。脱イオン(DI)水は、Purelab社のOption 560逆浸透浄水器を使って浄化した。ミリポア水は、Millipore社のMilli-Q Biocel A10浄化装置から得た。シリコーン油(ポリ−(ジメチルシロキサン)またはPDMS)は、粘度が1cStから1000cSt(PDMSの異なる平均分子量に対応)の範囲で、Gelest Inc.社から供給された。
αアミノ酸−N−カルボン酸無水物NCAモノマは、既刊の文献の方法で合成された(H. R. Kricheldorf、α-Aminoacid-N-Carboxyanhydrides and Related Materials (Springer-Verlag、NY)、1987年)。すべてのブロックコポリペプチドは、(PMe3)4Coイニシエータを使って重合された(H. F. Klein、及びH. H. Karsch、Chem. Ber. 108、1975年、p.944)。得られたポリペプチドは、GPC、1H−NMR、および赤外線スペクトルで特徴付けした(T. J. ドライボックスにおいて、L−リシンNCA(10.00g、33mM)をTHF(200ml)に溶解し、プラスチックの栓で密封可能な500mlの平底フラスコに入れた。次に、一定量の(PMe3)4Co(THF中48mg/ml溶液の16ml)を注射器でフラスコに加えた。撹拌棒を入れてフラスコを密封し、45分間攪拌した。重合物から一定量(50μl)をGPC分析用に取り出した(Mn=11000、Mw/Mn=1.24)。L−ロイシンNCA(1.3g、8.2mM)とD−ロイシンNCA(1.3g、8.2mM)とをTHF(52ml)に溶解し、続いて重合混合物に加えた。16時間撹拌後、溶液をドライボックスから取り出し、減圧下でTHFを除去した。FTIR分析は、モノマの完全消費を示し、既知の結果と同様であった(V. Breedveld他、Macromolecules 37、2004年、p.3943)。
上記のポリ(NεCBZ−L−リシン)40−b−ポリ(rac−ロイシン)20を、トリフルオロ酢酸(TFA)(350ml)に溶解し、1リットルの平底フラスコに移して氷浴中に置いた。次に、HBr(酢酸に33%)を加え(40ml、131mM)、2時間撹拌反応させた。反応混合物にジエチルエーテルを加えて遠心分離し、脱保護された高分子を単離した。続いて、単離した高分子をDI水に溶解し、四ナトリウムEDTA(3mM、4日)、0.1MのHCl(2日)、DI水(1日)、0.1MのLiBr(2日)、DI水(2日)で、それぞれの溶液を1日3回変えながら、透析(6000〜8000MWCOの膜)した。透析した高分子を、凍結乾燥によって単離し、製品として乾燥した白粉(4.8g、70.2%)とした。FTIRと1H−NMRを行い、その結果は既知のものと同様であった(V. Breedveld他、Macromolecules 37、2004年、p.3943)。
K40rL10は、上述のように生成された。最初の断片(ポリCBZ−リシン)のGPC分析は、Mn=10500、Mw/Mn=1.20であった。脱保護された共重合体(150mg、1.3x10−2mM)を水に溶解し、125mlの平底フラスコに入れた。NaHCO3(162mg、19mM)を溶液に加えた。フルオレセインイソチオシアネート(FITC)(5mg、1.3x10−2mM)を乾燥DMSO溶液(1ml)に溶解し、高分子溶液に加えた。撹拌棒を入れ反応混合物を一晩攪拌した。高分子溶液を、DI水で3日間、水を1日3回変えながら、透析(6000〜8000MWCOの膜)した。透析した高分子を、凍結乾燥によって単離し、高分子鎖当たりおよそ1単位のフルオロセインを含んだ黄橙色の高分子(130mg、87%)とした。
凍結乾燥したK40rL10コポリペプチドを、まず、脱イオン水に所望の濃度で溶解した。ブロックコポリペプチドの濃度Cの範囲は、1.0x10−4mMから1.7mMであった。シリコーン油(10cSt、Gelest社PDMS)を、所望の連続相に対する油体積分率φ(0.01≦φ≦0.8)となるように加えた。シリコーン油と水の異なる濃度(10cStPDMSシリコーン油0.793g/ml対水1.0g/ml)により、管の頂部に栓が形成(Cyro−ETMで液滴粒度>300nm)された。手持ホモジナイザ(S8N-8G分散エレメント付きIKA Ultra-Turrax T8)または手持超音波ホモジナイザ(Cole-Palmer 4710シリーズモデルASIを35%から40%出力で)のいずれかを用いて剪断を適用し、予混合エマルションを生成した。この予混合エマルションを、75μmステンレススチール/セラミック相互反応チャンバを有し、入力空気圧p=130psiに設定したM-110S Microfluidizer(r) Processorを通過させた。エマルションをマイクロ流体ホモジナイザの出力部で集め、さらに5回マイクロ流体ホモジナイザを通過させ(合計N=6)、平均液滴半径<a>を低減しサンプルの単分散性を増した。コポリペプチド、水、ならびに大豆油およびオレイン酸メチルを含む10cStおよび100cStのシリコーン油以外の種々の油のダブルエマルションを形成した。Cyro−CTEMにより、次のコポリペプチド組成物でのダブルエマルションの形成を確認した。K20rL10、K40rL5、K40rL10、K40rL20、K40rL30、K40rL20、およびE40rL10(E=グルタミン酸)。
1.5mMのK40rL20エマルション(上述どおりに生成)を15mlのプラスチックの遠心分離チューブに入れ、IEC社HN-S卓上遠心分離機を使い、毎分3500回転で24時間、遠心分離した。0.5mmの栓が形成され、残余物と分離した。シリコーン油と水の異なる濃度(10cStPDMSシリコーン油0.973g/ml対水1.0g/ml)により、チューブの頂部に栓が形成(Cyro−ETMで液滴粒度>300nm)された。栓を他のサンプルから主要残余物として分離し、これらの一次残余物を、Beckman L8-55超高速遠心分離機を使い、毎分2万回転で4時間、さらに分別した。懸濁液の頂部に栓(Cyro−TEMで、液滴粒度範囲30nmから200nm)が、浮遊して残る二次残余物(Cyro−TEMで、液滴粒度範囲10nmから30nm)とともに形成された。
エマルション液滴の直径は、Photocor-FC基板とソフトウェアを使って動的光散乱法により測定した。サンプルを、強度の読みが1x105から6x105の範囲となるように、希釈した。それぞれの測定は、リニアなチャンネル間隔と調節可能なベースラインを用い、散乱角90°で500秒間行った。使用した適合方法は、データに適合して平均液滴半径を算出するよう調節可能なベースラインを用いたキュムラント分析である。
疎水性液をラベル付けするため、ピレンをシリコーン油成分に0.01Mの濃度で溶解し、量子ドット(Evident Technologies社、Type T2-MP 650nm Macoun Red InGaP/ZnS、アミン機能化)を水分に2μMの濃度で分散させた。エマルション生成のため、FITC−K40rL10(0.1mMの溶液150μl)をInGaP量子ドット(8μMの溶液50μl)と10cStのシリコーン油中のピレン(0.01Mのピレン溶液50μl)とともに混合した。混合物を、超音波ホモジナイザ(35%出力で)を10秒間使って乳化した。
0.1mMのFITC−K40rL10エマルション(φ=0.2、10cStシリコーン油)を、0.1mMのFITC−K40rL10ポリペプチド溶液800μlと10cStPDMSシリコーン油200μlとを混合し、続いて手持超音波ホモジナイザ(35%出力で)を使って10秒間乳化した。撮像の前に、0.1mMのFITC−K40rL10エマルション懸濁液の一定量を脱イオン(DI)水で10倍に希釈した。ガラススライド上にエマルションを一滴載せ、続いてカバーガラスを置いた。サンプルを、488nm(青色)アルゴンレーザ(JDS Uniphase)、561nm(緑色)ダイオードレーザ(DPSS: Melles Griot)、およびSpectra-Physics Millenia X 532nm(緑色)ダイオードポンプレーザおよびTsunami Ti(紫外線励起用の768nmに同調したサファイアピコ秒パルス赤外線レーザ)からなる2フォトンレーザセットアップを装備したLeica-SP MP共焦点多光子倒立顕微鏡を使って撮像した。
蛍光撮像の前に、エマルション懸濁液をDI水で10倍に希釈した。ガラススライド上にエマルションを1滴載せ、カバーガラスで覆った。サンプルを、Zeiss 200蛍光顕微鏡を使って撮像した。
それぞれのエマルションサンプルを、撮像の前に、DI水で10倍に希釈した。次に、各サンプルの一定量(5μl)をカーボングリッド上に置いた。カーボングリッドを、液体エサン中でのサンプルの自動ブロッティングとガラス化のため、Vitrobot (FEI)自動ガラス化装置に搭載した。グリッドを液体窒素下で保存し、次に、コールドステージを使って加速電圧120kVのPhillips Tecnai F20電子顕微鏡内に置いた。画像は、Leginonソフトウェアパーッケージと連動したTeitz SCX低速走査CCD検出器で得た。
本発明のいくつかの実施の形態において設計したブロックコポリペプチド界面活性剤は、ポリ(L−リシン・HBr)x−b−ポリ(rac−ロイシン)y、KxrLyの一般構造を有する。ここで、xは20から100の範囲であり、yは5から30の範囲である(図1A)。親水性のポリ(L−リシン・HBr)断片は、中性pHにおいて高電荷で、良好な水溶性を有し(E. Katchalski、及びM. SeIa、Synthesis and chemical properties of poly-alpha-amino acids。Advances in Protein Chemistry 13、1958年、p.243-492)、化学機能化のためのアミン基を豊富に保有している(P. Niederhafner、J. Sebestik、及びJ. Jezek、Peptide dendrimers。Journal of Peptide Science 11、2005年、p.757-788)。他の重合体の両親媒性物質の疎水性の断片と違い、ポリ(L−ロイシン)断片は、棒状のαへリックス配座を採用し、それにより強力な鎖間会合と一般的な有機溶剤への溶解性の低さを引き起こす(A. P. Nowak他、Rapidly recovering hydrogel scaffolds from self-assembling diblock copolypeptide amphiphiles。Nature 417、2002年、p.424-428)。KxLy(例、K60L20)の構造のブロック共重合体は、水中において強く会合し、疎水性の断片のまとめを介して膜を形成することを示した(E. P. Holowka、D. J. Pochan、及びT. J. Deming、Charged polypeptide vesicles with controllable diameter。Journal of the American Chemical Society 127、2005年、p.12423-12428)。従って、そのペプチドの性質が油層における鎖間水素結合を介して液滴界面の追加的な機械的安定性を可能にしながら(C. Lapp、及びJ. Marchal、Preparation De La Poly-D,L-Phenylalanine En Helice Par Polymerisation De La D,L-Benzyl-4 Oxazolidine Dione-2-5。Journal De Chimie Physique Et De Physico-Chimie Biologique 60、1963年、p.756-766)、その無秩序な鎖配座が溶解性を改善し(表1)(H. Stereospecificity of the Polymerization of D,L-Leu-NCA and D,L-VaI-NCA。Makromolekulare Chemie-Macromolecular Chemistry and Physics 182、1981年、p.3077-3098、J. W. Breitenbach、K. Allinger、及びA. Koref、Viskositatsstudien an Losungen von DL-Phenylalanin-Polypeptiden。Monatsh. Chem. 86、1955年、p.269)、表面活動の促進を図る(表1)ことから、ポリ(rac−ロイシン)に重点的に取り組んだ。
K40(rac−L)20のコポリペプチドを、所望の濃度(例、0.01mM<C<1.5mM)で超純水に溶解する。PDMSシリコーン油(10cSt)を加えて、所望の油対連続相の体積分率φ(0.05<φ<0.50)とする。手持ホモジナイザ(S8N-8G分散エレメントを装備したIKA Ultra-Turrax T8)を1分間使って、または手持超音波ホモジナイザ(Cole-Palmer 4710シリーズモデルASIを出力35〜40%で)を10秒間使って混合し、マイクロスケールのエマルション(つまり、「予混合」エマルション)を作る。このエマルションを、75μmステンレススチール/セラミック相互反応チャンバを装備し、入力空気圧p=130psiに設定したMicrofluidizer(r) Processor M-110Sを通過させる。装置の出力部でエマルションを集め、次に、マイクロ流体ホモジナイザを合計6回繰り返し通過させる。これにより、平均液滴半径<a>(例、単純エマルションの一重液滴、ならびにダブルエマルションの内側および外側液滴の)が減少し、エマルション内の液滴の単分散が増加する。同様の手順が、他のブロックコポリペプチド界面活性剤を使ったエマルションの生成に用いられた(表1、図7A〜図7C)。内側液滴の半径を外側液滴の半径で割って得られる比(「I/O比」という)は、異なる疎水性鎖長において比較的均一で、約0.5であった(表1、図7D)。リシンまたはロイシン領域が異なる親水性または疎水性の残基でそれぞれ置き換えられた他の両親媒性ブロックコポリペプチドでも、ダブルエマルションを形成することが見出された(図8A〜図8D)。異なるコポリペプチド界面活性剤の乳化能力を、安定したエマルションまたは安定したダブルエマルションをもたらさないホモポリペプチドK60(図9A、図9B)との対照で、安定性の劣るエマルションを形成するトルエンを使って定性的にも評価した。
物質
テトラヒドロフラン(THF)を、使用の前に、窒素下でアルミナを装填したカラムを通過させて乾燥した(A. P. Nowak他、Rapidly recovering hydrogel scaffolds from self-assembling diblock copolypeptide amphiphiles。Nature 417、2002年、p.424-428)。分子量は、Wyatt DAWN EOS光散乱検出器とWyatt Optilab DSPを装備したSSI社のポンプにおいて、ゲル透過クロマトグラフィ/光散乱(GPC/LS)の2段階を60℃で行って得た。分離は、0.1MのLiBrのDMFを溶剤液として使用し、ポリペプチドの濃度を約5mg/mlとし、105、104および103オングストロームのPhenomenex社の5μmカラムで達成した。赤外線スペクトルは、ポリスチレン膜を使って校正されたPerkin Elmer社のRX1フーリエ変換赤外分光(FTIR)分光光度計で記録した。水素核磁気共鳴(1H-NMR)スペクトルは、Bruker社のAVANCE 400MHzの分光計で記録した。脱イオン(DI)水は、Purelab社のOption 560逆浸透浄水器を使って浄化した。超純水(18MΩ)は、Millipore社のMilli-Q Biocel A10浄化装置から得た。シリコーン油(10cSt、ポリジメチルシロキサン、PDMS)は、Gelest Inc.から供給された。
αアミノ酸−N−カルボン酸無水物NCAモノマを、既刊の文献の方法で合成した(同文献)。得られたポリペプチドは、GPC、1H−NMR、および赤外線スペクトルを使って特徴付けした(同文献)。共重合体の組成物は、重水(D2O)中で記録した1H−NMRの積算値の分析により測定した。すべての組成物は、予測値の5%以内であった。測定した高分子鎖の長さ分布から、多分散性指数(Mw/Mn)は1.1から1.3の範囲であった。K60L20を、既刊の文献の方法で合成した(E. P. Holowka、D. J. Pochan、及びT. J. Deming、Charged polypeptide vesicles with controllable diameter。Journal of the American Chemical Society 127、2005年、p.12423-12428)。疎水性のポリ(ロイシン)断片の鎖配座は、円偏光二色性分光法を使って確認した(図14)。ここでは、ポリ(リシン)断片からの影響は、既述(A. P. Nowak他、Rapidly recovering hydrogel scaffolds from self-assembling diblock copolypeptide amphiphiles。Nature 417、2002年、p.424-428)のように、ポリ(ラセミ−リシン)断片を使って除去されている。
窒素で充満したグローブボックスにおいて、CBZ−L−リシンNCA(10g、33mM)をTHF(200ml)に溶解し、プラスチックの栓で密封可能な500mlの平底フラスコに入れた。次に、一定量の(PMe3)4Co(THF中48mg/ml溶液の16ml)を注射器でフラスコに加えた。撹拌棒を入れてフラスコを密封し、45分間攪拌した。重合溶液から一定量(50μl)をGPC分析用に取り出した(Mn=11000、Mw/Mn=1.24)。L−ロイシンNCA(1.3g、8.2mM)とD−ロイシンNCA(1.3g、8.2mM)とをTHF(50ml)に溶解し、続いて重合混合物に加えた。16時間撹拌後、FTIR分析は、モノマの完全消費を示し、既知の結果と同様でとなった(同文献)。
上記のポリ(Nε−CBZ−L−リシン)40−b−ポリ(rac−ロイシン)20溶液をドライボックスから取り出し、減圧下でTHFを除去した。次に、ブロックコポリペプチドをトリフルオロ酢酸(TFA)(350ml)に溶解し、1リットルの平底フラスコに移して氷浴中に置いた。次に、HBr(酢酸に33%)を加え(40ml、131mM)、2時間撹拌反応させた。反応混合物にジエチルエーテルを加えて(400ml)遠心分離し、脱保護された高分子を単離した。続いて、単離した高分子をDI水に溶解し、四ナトリウムEDTA水溶液(3mM、2日)、HCl水溶液(100mM、2日)、DI水(1日)、LiBr水溶液(100mM、2日)、最後にDI水(2日)で、それぞれの溶液を1日3回変えながら、4リットルの容器内で透析(6000〜8000MWCOの膜を使用)した。透析した高分子を、凍結乾燥によって単離し、製品として乾燥した白粉(4.8g、70%)とした。ブロックコポリペプチドにFTIRと1H−NMRを行い、その結果は既知のものと同様となった(同文献)。

疎水性相をラベル付けするために、ピレンを0.01Mの濃度でシリコーン油に溶解した。水性相をラベル付けするために、水溶性量子ドット(Evident Technologies社、Type T2-MP 650nm Macoun Red InGaP/ZnS、アミン機能化)を水性相に2μMの濃度で分散させた。エマルションを作るために、FITCでラベル付けされたK40(rac−L)10(0.1mMの溶液150μl)とInGaP量子ドット(8μMの溶液50μl)とを10cStのシリコーン油中のピレン(0.01Mのピレン溶液50μl)と混合した。混合物を、超音波ホモジナイザ(35%出力で)を10秒間使って乳化した。FITC−K60L20ブロックコポリペプチド界面活性剤についても同様の手順で行った。撮像の前に、カプセル化されなかった量子ドットを、脱イオン水で透析して除去した。

ブロックコポリペプチド界面活性剤をエマルションの生成に使用した。すべてのエマルションは、マイクロ流体ホモジナイザを使用し、通過回数N=6、ホモジナイザ入力空気圧p=130psi、ブロックコポリペプチド濃度C=1.0mM、および油体積分率φ=0.20の条件で生成した。直径(外側液滴の)および内側/外側直径比は、CTEM画像の少なくとも50液滴の測定値を平均して得た。臨界凝集濃度(CAC)は、ピレン蛍光を20℃で使用して測定した。水溶解限度は、各ペプチドの15mM溶液を、光学的に透明な溶液になるまで希釈して測定した。ブロック共重合体の溶解性は、PDMS中では無視できる程度であった。油/水界面張力データは、デュヌュイ(Du Nuoy)リング法で、10cStのPDMSおよびブロックコポリペプチド溶液(0.1mM、引き上げ速度=0.01mm/s、25℃)を使って測定した。

- 実質的に連続な液体媒体と、

外部表面を有する第1の液の外側液滴と、
該第1の液滴内に内部表面を有する第2の液の内側液滴であって、該第2の液が該第1の液に非混和性であり、該内側液滴および該外側液滴が両者間の境界表面領域に該第1の液の膜を有する内側液滴と、
該外側液滴の該外部表面に配置されるブロック共重合体の外層と、
該内側液滴および該外側液滴間の該境界表面領域に近接して該内側液滴の該内部表面に配置されるブロック共重合体の内層と、を含み、

- 前記外側液滴の前記第1の液が疎水性であり、さらにその中に、脂肪、脂質、ワックス、天然油、合成油、シリコーン油、揮発油、精油、香料、コレストロール、ステロイド、疎水性薬物分子、高分子、ブロック共重合体、ポリペプチド、ブロックコポリペプチド、ポリ−酸、ポリ−塩基、ミセル、量子ドット、ナノ粒子、ナノクラスタ、カーボンナノチューブ、フラーレン、磁性流体、サーモトロピック液晶、リオトロピック液晶、フッ素化液、臭素化液、植物由来物質、動物由来物質、バクテリア由来物質、およびその組み合わせ、の少なくとも1つを少なくとも混合または分散した請求項16に記載のエマルション。
- 液滴構造であって、
外部表面を有する第1の液の外側液滴と、
該第1の液滴内に内部表面を有する第2の液の内側液滴であって、該第2の液が該第1の液に非混和性であり、該内側液滴および該外側液滴が両者間の境界表面領域に該第1の液の膜を有する内側液滴と、
該外側液滴の該外部表面に配置されるブロック共重合体の外層と、
該内側液滴の該内部表面に配置されるブロック共重合体の内層と、を含み、
該ブロック共重合体が親水性高分子ブロックと疎水性高分子ブロックとを含み、それらが共同して該外側液滴の該外部表面の該内側液滴の該内部表面との合体の安定化と該液滴構造の他の液滴構造との合体の安定化に作用する液滴構造。 - 前記液滴構造の非変形な液滴直径で与えられる最大直径が約1000nm以下で、かつ約10nm以上である請求項19に記載の液滴構造。
- 前記液滴構造の非変形な液滴直径で与えられる最大直径が約250nm以下で、かつ約50nm以上である請求項19に記載の液滴構造。
- 前記ブロック共重合体の内層のブロック共重合体が前記ブロック共重合体の外層のブロック共重合体と実質的に同じ分子形態である請求項19に記載の液滴構造。
- 前記親水性高分子ブロックが約200Daから約3,000,000Daの範囲の分子量を有し、前記疎水性高分子ブロックが約200Daから約3,000,000Daの範囲の分子量を有する請求項19に記載の液滴構造。
- 前記内側液滴の半径を前記外側液滴の半径で除して与えられる比が約0.9以下で、かつ約0.05以上である請求項19に記載の液滴構造。
- 前記ブロック共重合体の内層が2つの区別可能に異なる型のモノマの重合から形成されるジブロック共重合体であり、前記ブロック共重合体の外層が該2つの区別可能に異なる型のモノマの重合から形成されるジブロック共重合体である請求項19に記載の液滴構造。
- 前記親水性高分子ブロックが主に親水性アミノ酸を含むポリペプチドブロックであり、前記疎水性高分子ブロックが主に疎水性アミノ酸を含むポリペプチドブロックである請求項19に記載の液滴構造。
- 前記ポリペプチドブロックの少なくとも1つが表面機能化をもたらす部分表面を含む請求項26に記載の液滴構造。
- 前記親水性高分子ブロックが複数の型の親水性アミノ酸を主に含むポリペプチドブロックであり、前記疎水性高分子ブロックが複数の型の疎水性アミノ酸を主に含むポリペプチドブロックである請求項19に記載の液滴構造。
- 前記親水性アミノ酸が、L−アルギニン、L−アスパラギン、L−アスパラギン酸、L−システイン、L−グルタミン酸、L−グルタミン、L−ヒスチジン、L−リシン、L−セリン、L−トレオニン、L−チロシン、D−アルギニン、D−アスパラギン、D−アスパラギン酸、D−システイン、D−グルタミン酸、D−グルタミン、D−ヒスチジン、D−リシン、D−セリン、D−トレオニン、D−チロシン、DL−アルギニン、DL−アスパラギン、DL−アスパラギン酸、DL−システイン、DL−グルタミン酸、DL−グルタミン、DL−ヒスチジン、DL−リシン、DL−セリン、DL−トレオニン、DL−チロシン、およびこれらの組み合わせからなる親水性アミノ酸の群から選択される請求項26に記載の液滴構造。
- 前記疎水性アミノ酸が、ラセミ−アラニン、ラセミ−グリシン、ラセミ−イソロイシン、ラセミ−ロイシン、ラセミ−メチオミン、ラセミ−フェニルアラニン、ラセミ−プロリン、ラセミ−トリプトファン、ラセミ−バリン、およびこれらの組み合わせからなる疎水性アミノ酸の群から選択される請求項26に記載の液滴構造。
- 前記疎水性アミノ酸が、ラセミ−アラニン、ラセミ−グリシン、ラセミ−イソロイシン、ラセミ−ロイシン、ラセミ−メチオミン、ラセミ−フェニルアラニン、ラセミ−プロリン、ラセミ−トリプトファン、ラセミ−バリン、およびこれらの組み合わせからなる疎水性アミノ酸の群から選択される請求項29に記載の液滴構造。
- 前記親水性アミノ酸および前記疎水性アミノ酸が、アラニン、アルギニン、アスパラギン、アスパラギン酸、システイン、グルタミン酸、グルタミン、グリシン、ヒスチジン、イソロイシン、ロイシン、リシン、メチオニン、フェニルアラニン、プロリン、セリン、トレオニン、トリプトファン、チロシン、バリン、およびこれらの組み合わせからなるアミノ酸の群から選択される請求項26に記載の液滴構造。
- 前記親水性アミノ酸がL−リシンであり、前記疎水性アミノ酸がラセミ−ロイシンである請求項26に記載の液滴構造。
- 前記ブロック共重合体が、KがL−リシンを表し、rLがラセミ−ロイシンを表し、xが約10から約200の範囲の整数であり、yが約3から約30の範囲の整数であるとき、KxrLyの式を満足する構造を有するブロックコポリペプチドである請求項19に記載の液滴構造。

- ナノ液滴構造であって、
外部表面を有する第1の液の外側液滴と、
該第1の液滴内に配置されて内部表面を有する第2の液の内側液滴であって、該第2の液が該第1の液に非混和性であり、該内側液滴および該外側液滴が両者間の境界表面領域に該第1の液の膜を有する内側液滴と、
該外側液滴の該外部表面に配置されるブロックコポリペプチドの外層と、
該内側液滴および該外側液滴間の該境界表面領域に近接して該内部表面に配置されるブロックコポリペプチドの内層と、を含み、

- 前記外側液滴の前記第1の液が疎水性であり、さらにその中に、脂肪、脂質、ワックス、天然油、合成油、シリコーン油、揮発油、精油、香料、コレストロール、ステロイド、疎水性薬物分子、高分子、ブロック共重合体、ポリペプチド、ブロックポリペプチド、ポリ−酸、ポリ−塩基、ミセル、量子ドット、ナノ粒子、ナノクラスタ、カーボンナノチューブ、フラーレン、磁性流体、サーモトロピック液晶、リオトロピック液晶、フッ素化液、臭素化液、植物由来物質、動物由来物質、バクテリア由来物質、およびその組み合わせ、の少なくとも1つを少なくとも混合または分散した請求項38に記載のナノ液滴構造。
- 前記外側液滴の前記第1の液が疎水性であり、さらにその中に、脂肪、脂質、ワックス、天然油、合成油、シリコーン油、揮発油、精油、香料、コレストロール、ステロイド、疎水性薬物分子、高分子、ブロック共重合体、ポリペプチド、ブロックポリペプチド、ポリ−酸、ポリ−塩基、ミセル、量子ドット、ナノ粒子、ナノクラスタ、カーボンナノチューブ、フラーレン、磁性流体、サーモトロピック液晶、リオトロピック液晶、フッ素化液、臭素化液、植物由来物質、動物由来物質、バクテリア由来物質、およびその組み合わせ、の少なくとも1つを少なくとも混合または分散した請求項39に記載のナノ液滴構造。
- 液体媒体と、
該液体媒体内に分散する複数のナノ液滴であって、該複数のナノ液滴のそれぞれが第2の液で包囲された第1の液の内側液滴を含み、該第1の液が該第2の液に非混和性であり、該第2の液が該液体媒体に非混和性であるナノ液滴と、を含み、
該複数のナノ液滴の外側液滴の非変形直径のアンサンブル平均が少なくとも約10nmで、かつ約300nm以下であるエマルション。 - 前記液体媒体が前記第1の液と同じ物質である請求項42に記載のエマルション。
- 前記複数のナノ液滴の表面および前記内側液滴の表面の少なくとも1つに吸着するブロック共重合体をさらに含む請求項42に記載のエマルション。
- 前記ブロック共重合体のそれぞれが親水性高分子ブロックと疎水性高分子ブロックとを含み、それらが共同して前記複数のナノ液滴または前記内側液滴の少なくとも1つの安定化に作用する請求項44に記載のエマルション。
- 前記複数のナノ液滴の表面に吸着する第1のブロック共重合体と前記内側液滴の表面に吸着する第2のブロック共重合体とをさらに含む請求項42に記載のエマルション。
- 前記第1および第2のブロック共重合体のそれぞれが親水性高分子ブロックと疎水性高分子ブロックとを含み、それらが共同して前記複数のナノ液滴または前記内側液滴の少なくとも1つの安定化に作用する請求項46に記載のエマルション。
- 前記第1および第2のブロック共重合体が実質的に同じ分子構造である請求項47に記載のエマルション。
- 実質的に連続な液体媒体と、
該実質的に連続な液体媒体内に分散する複数の液滴構造と、を含み、
該複数の液滴構造の各液滴構造が、
外部表面を有する液の液滴と、
該液滴の該外部表面に配置されるブロック共重合体の層と、を含み、
該ブロック共重合体が親水性高分子ブロックと疎水性高分子ブロックとを含み、それらが共同して該液滴構造の安定化に作用し、該複数の液滴構造の液が該実質的に連続な液体媒体に非混和性であるエマルション。 - 前記親水性高分子ブロックが主に親水性アミノ酸を含むポリペプチドブロックであり、前記疎水性高分子ブロックが主に疎水性アミノ酸を含むポリペプチドブロックである請求項49に記載のエマルション。
- 前記親水性高分子ブロックが主に親水性アミノ酸の複数の型を含むポリペプチドブロックであり、前記疎水性高分子ブロックが主に疎水性アミノ酸の複数の型を含むポリペプチドブロックである請求項49に記載のエマルション。
- 前記親水性アミノ酸および前記疎水性アミノ酸が、アラニン、アルギニン、アスパラギン、アスパラギン酸、システイン、グルタミン酸、グルタミン、グリシン、ヒスチジン、イソロイシン、ロイシン、リシン、メチオニン、フェニルアラニン、プロリン、セリン、トレオニン、トリプトファン、チロシン、バリン、およびこれらの組み合わせからなるアミノ酸の群から選択される請求項50に記載のエマルション。
- 前記各液滴構造の非変形な液滴直径に対応する最大直径が約300nm以下で、かつ約10nm以上である請求項52に記載のエマルション。

外部表面を有する液の液滴と、
該液滴の該外部表面に配置されるブロック共重合体の層と、を含み、
該ブロック共重合体が親水性高分子ブロックと疎水性高分子ブロックとを含み、それらが共同して該液滴構造の安定化に作用する液滴構造。 - 前記親水性高分子ブロックが主に親水性アミノ酸を含むポリペプチドブロックであり、前記疎水性高分子ブロックが主に疎水性アミノ酸を含むポリペプチドブロックである請求項54に記載の液滴構造。
- 前記親水性高分子ブロックが複数の型の親水性アミノ酸を主に含むポリペプチドブロックであり、前記疎水性高分子ブロックが複数の型の疎水性アミノ酸を主に含むポリペプチドブロックである請求項54に記載の液滴構造。
- 前記親水性アミノ酸および前記疎水性アミノ酸が、アラニン、アルギニン、アスパラギン、アスパラギン酸、システイン、グルタミン酸、グルタミン、グリシン、ヒスチジン、イソロイシン、ロイシン、リシン、メチオニン、フェニルアラニン、プロリン、セリン、トレオニン、トリプトファン、チロシン、バリン、およびこれらの組み合わせからなるアミノ酸の群から選択される請求項55に記載の液滴構造。
- 第1の液と第2の液を準備するステップであって、該第1の液が該第2の液に非混和性であるステップと、
該第1の液および該第2の液の少なくとも1つに選択した量のブロック共重合体を追加するステップと、
該第2の液に分散する該第1の液の複数の液滴を生成するように該第1の液を該第2の液に乳化するステップと、を含み、
該ブロック共重合体が該複数の液滴が合体しないよう安定化するエマルションの製造方法。 - 前記第1の液の前記複数の液滴の各液滴が、前記乳化のステップにおいて前記複数の液滴の生成と実質的に同時に作られる前記第2の液の内側液滴をその中に含む請求項58に記載のエマルションの製造方法。
- 前記第1の液の前記複数の液滴の各液滴が、前記乳化のステップにおいて前記複数の液滴の生成と実質的に同時に作られる前記第2の液の複数の内側液滴をその中に含む請求項58に記載のエマルションの製造方法。
- 前記複数の液滴があらかじめ選択されたアンサンブル平均直径を有するように加える前記ブロック共重合体の量を選択するステップをさらに含む請求項58に記載のエマルションの製造方法。
- 前記追加のステップにおいて前記複数の液滴があらかじめ選択されたアンサンブル平均直径を有するように前記ブロック共重合体の分子組成を選択するステップをさらに含む請求項58に記載のエマルションの製造方法。
- 第1の液および第2の液の少なくとも1つに界面活性剤を追加する、または該第1の液および該第2の液の少なくとも1つに界面活性剤前駆体を追加する、の少なくとも1つのステップと、
該第2の液に浸漬される該第1の液の複数の液滴を形成して単純エマルションをもたらすように該第1の液を該第2の液に乳化するステップであって、該第1の液が該第2の液に非混和性であるステップと、
前記界面活性剤および前記界面活性剤前駆体の少なくとも1つを第3の液に追加するステップと、
該単純エマルションの複数の液滴を形成してダブルエマルションをもたらすように該単純エマルションを該第3の液に乳化するステップであって、該第2の液が該第3の液に非混和性であるステップと、を含み、
該ダブルエマルションの複数の液滴のそれぞれがその中に少なくとも1つの該第1の液の液滴を含むエマルションの製造方法。 - 請求項58から63のいずれかの方法により製造されたエマルション。
