Abstract
Description
この出願は、2007年7月12日に出願された、米国仮出願第60/929,775号(この内容は、その全体が参考として本明細書に援用される)への優先権およびその利益を主張する。
本発明は、炎症性筋障害の診断および進行の評価に用いることができる組成物および方法の分野にある。
本明細書に記載されかつ当該技術において標準的な手順を用いて末梢血単核細胞(PBMC)を含有する試料を得て、溶解し、抽出して、RNAを得てよい。アッセイは、マイクロアレイプレート製造業者により記載され、全般的にGreenbergら、Neurology59巻(8号):1170〜82頁(2002年)に以前に記載されたような標準的な手順を用いて行ってよい。あるいは、mRNAは、生検の間に得られた筋肉細胞から得て、分析できる。
多発性筋炎または皮膚筋炎の患者において変化したと本明細書において同定される全ての遺伝子は、実施例の部に記載されるAffymetrixのプレート上に存在した。原則的に、同じプレートを、PBMCまたは筋肉細胞からのmRNAの評価のために用いることができた。しかし、用いられるプレートは、数千の異なる遺伝子についての固定化オリゴヌクレオチドを含むので、この系は不便であり不必要に高価である。本明細書で議論される分析により適するプレートは、表2に列挙される遺伝子に焦点を当てることにより作製できる。よって、Affymetrixプレートに似ているが、ほんの少数(例えば5〜50または5〜100個)の別々のハイブリッド形成部位を有するプレートを、同じアッセイ条件下で用いてよい。このことは分析を単純化し、結果の一貫性をよりよく確認するための重複を含めることを可能にする。
上記の方法を用いてPBMCおよび筋肉細胞内の遺伝子発現レベルを決定することができるが、この分析を行う任意のその他の手順も、本発明に関連して用いることができる。例えば、PCR増幅を伴うかまたは伴わないDNAブロッティング技術は、遺伝子のレベルを定量するために用いてよい。ウェスタンブロットまたはイムノアッセイを用いて、遺伝子産物を定量してよく、酵素ベースのアッセイを用いてよい場合もある。発現のレベルは、免疫蛍光技術またはプロモーターに基づくレポーターアッセイにより評価することもできる。手順に必須の要素は、定量をどのように行うかではなく、むしろ、調査される特定の遺伝子と、これらの遺伝子がPM、DMまたはIBMの存在または状態に特徴的なレベルで発現されているかどうかを決定することである。
本明細書に記載されるアッセイは、PBMCまたは筋肉細胞が、多発性筋炎、皮膚筋炎または封入体筋炎の存在または進行を示す遺伝子発現プロファイルを有するかどうかを評価するように設計されている。このことは、これらの疾患を研究する科学者、および診断を行うかまたは治療計画もしくは薬物が有益な効果を有するかどうかを決定しようとする臨床医にとって、明らかに大きな価値がある。
研究の対象
65回のマイクロアレイ実験を、36名が炎症性筋障害を有する(12名がDM、11名がPMおよび13名がIBM)合計で56名の元々登録されていた対象からの血液試料に対して行った。さらなる対照群のために、我々は、非炎症性自己免疫性筋疾患である重症筋無力症の患者5名、遺伝子的に決定された筋疾患の患者3名(2型筋緊張性ジストロフィー2名およびミトコンドリア筋症1名)、ならびに12名の健常対象有志について研究した。DMの患者6名およびPMの患者2名は、一方は疾患が活動性のとき、他方は疾患が改善中の2回の異なる時点で行ったマイクロアレイ実験のための第2の血液試料を提供した。不応性DMの1名の患者は、ともに疾患が活動性のときの異なる時点での2つの試料を提供した。全ての患者は、DMまたはPMが確定的またはほぼ確定的(Hoogendijkら、Neuromuscul.Disord.14巻(5号):337〜45頁(2004年))、およびIBMが確定的またはほぼ確定的(Griggsら、Ann.Neurol.38巻(5号):705〜13頁(1995年))との調査基準を満たしていた。全身性エリテマトーデスの患者は除外した。DM(平均年齢47歳)およびPM(平均年齢54歳)の患者の臨床的特徴を表1に概説する。DMの2名およびPMの4名の6名の患者は、間質性肺疾患であった。これらのうち、DMの2名およびPMの1名の患者は、抗ヒスチジルトランスファーRNA(抗Jo−1)抗体をさらに有した。5名の男性および7名の女性であるIBMの患者は、平均年齢69歳であり、免疫調節薬の投与を受けている者はいなかった。採用時に健常な有志は、過去6カ月間にいずれの重篤な病気もなく、過去6カ月間にいずれの新しい投薬も開始せず、過去2カ月間に重篤な風邪、流感またはその他の感染がなかった。有志は、5名の男性および7名の女性からなり、平均年齢46歳(30〜62歳の範囲)であった。内部調査委員会が研究を承認した。書面のインフォームドコンセントを、全ての参加する患者および健常有志から得た。
我々は、DMおよびPMの患者を、活動性疾患の者(DMA;PMA)と改善中の疾患の者(DMI;PMI)とに分類した。以下の4つのうち3つを有する患者を、活動性と分類した:(1)症状の増加、(2)徒手筋試験における客観的脱力の増大、(3)高い血清クレアチンキナーゼ(CK)レベル、そしてもし1回を超える測定が可能であればそのレベルの増大、ならびに(4)治療している医師が患者の免疫治療を増大させる。筋炎における活動性疾患を定義するために、同様の特徴が以前に用いられた(Nagyら、Immunol.Lett.74巻(3号):207〜10頁(2000年))。DMおよびPMの患者を、遺伝子発現データの分析の前に、先を見越して活動性または改善中に分類した。MRC等級分けを用いる徒手筋試験を用いて強さを評価した。30の異なる筋肉群についての複合スコアを算出し、最大スコアは150であった。我々は、国際筋炎評価・臨床研究グループ(International Myositis Assessment and Clinical Studies group)により提案されるように、疾患活動性のさらなる指標として、筋炎治療意図活動性指数(Myositis Intention to Treat Activity Index)(MITAX)を用いた(Isenbergら、Rheumatology (Oxford)43巻(1号):49〜54頁(2004年))。MITAXは、筋肉、皮膚粘膜、胃腸、呼吸器および筋骨格の系統をみる多系統の評価ツールである。疾患活動性のこの指標について、良好な評価者間信頼性が報告されている。対の試料を有する9名の患者において、活発と改善中との間に8.5のMITAXスコアの平均的な低減がみられた。活動性のスコアは12〜13の範囲であり、改善中のスコアは2〜6の範囲であった。
我々は、患者および有志からの血液10mlを、EDTA含有試験管(57個の試料)、またはいくつかの場合においてPAX−Gene−RNA試験管に直接(8個の試料)回収した。EDTA含有試験管について、遠心分離の後に、血漿(上層)を赤血球細胞から1mmのところまで低く吸引し、500μlのバフィコートを、1.2mlのRNAlater溶液(Ambion、Austin、Texas)を予め充填した低温槽貯蔵試験管に、注意深く吸引して入れた。併せたバフィコートとRNAlaterを−20℃にて凍結した。RNAを「Ribo Pure」(Ambion、Austin、Texas)を用いてバフィコートおよびPAX−Gene RNA試験管から抽出した。RNA濃度は、分光光度計を用いて測定し、RNAの質を、1μgのRNAを1%アガロースゲル上を移動させることにより評価した。70〜120mgの重量の筋肉生検試料のRNAを、以前に記載されたようにして抽出した(Greenbergら、Neurology59巻(8号):1170〜82頁(2002年))。筋肉生検組織は、15名の患者(5名のDM、5名のPMおよび5名のIBM)から、活動性疾患のときに得て、これらの全ての患者は、活動性および改善中の時点で血液マイクロアレイ研究も受け、神経筋疾患を有さない5名の患者は診断生検を受けた。筋肉RNA抽出は、RiboPureを用いて、PBMC RNA抽出と同様にして行った。15の炎症性筋障害筋肉マイクロアレイ研究のうち、9つ(3名のDM、2名のPMおよび4名のIBM)は、発表に用いたデータの一部分を用いて以前に行って、この研究において再分析し、6つはこれらの研究について特に新しく行った。
マイクロアレイ研究を、以前に記載されたようにして、Affymetrix HG−U133Aマイクロアレイを用いて筋肉について行った(Greenbergら、Neurology59巻(8号):1170〜82頁(2002年))。PBMC試料は、Affymetrix HG−U133Aプラス2.0マイクロアレイおよびGeneChipオペレーティングシステム(GCOSv1.3)バージョン1.3を用いて処理した。
Affymetrix HG−U133プラス2.0 GeneChipは、63個の対照プローブセットを含む54,675個のプローブセットを有する。プローブセットのアノテーションは、NetAffx Analysis Center、バージョン3/9/2007から得た。発現レベルは、Bioconductor GCRMAパッケージ(http://www.bioconductor.org/download/oldrelease/bioc1.6/popular/gcrma.htmlで利用可能)に備えられたGC−Content Robust Multichip Analysis(GCRMA)を用いて算出した。このアルゴリズムは、実験における全てのアレイからのGC含量に基づく偏りと光学的ノイズ挙動とを計上することにより、改善された発現測定を行う(Wuら、J.Comput.Biol.12巻(6号):882〜93頁(2005年))。品質管理は、走査され再構築された画像を目視検査して、全体のアーチファクトを同定すること、および対照プローブセットを含む品質評価パラメータを注意深く評価することにより行った。全ての血液および筋肉のマイクロアレイデータは、この研究においてGCRMAを用いて一緒に分析した。
倍率変化の平均および90%信頼区間(CI)を、対照群と比較してそれぞれの疾患群について算出し、それに加えてWelchのt検定を用いて2群の比較のp値も算出した(表2)。我々は、p値<0.0001を必要とし、CIの下限>4.0として、著しく上方制御されている遺伝子を選択するために厳しい基準を用いた。遺伝子は、文献(10−12)および分子データベースの検索により、IFN−α/β誘発と同定された。
我々は、定量的リアルタイムPCRを、Primerソフトウェア(Whitehead Institute、Cambridge、MA)を用いて設計され、商業的に購入された(Operon Biotechnologies,Inc.Huntsville、AL)プライマーを用いて、18個の試料(4個のDMA、5個のDMI、4個のIBMおよび5個の健常有志)に対して、2つのIFN誘導遺伝子:IFIT1およびMX1について行った。用いたプライマーは、次のとおりであった:MxA:フォワード5’−CGGCTAACGGATAAGCAGAG−3’(配列番号1)およびリバース5’−ACCTACAGCTGGCTCCTGAA−3’(配列番号2);IFIT1:フォワード5’−AAAAGCCCAC−ATTTGAGGTG−3’(配列番号3)、およびリバース5’−GAAATTCCTGAAACCGACCA−3’(配列番号4)。
筋肉がマイクロアレイ研究に付された15名の患者(DM、PMおよびIBMの5名ずつ)の凍結筋肉切片を、ミクソウイルス耐性Aに対する抗体(抗MxA抗体;Department of Virology、University of Freiburg、GermanyのOtto Haller博士の好意)を用いて、以前に記載されたようにして染色し(Greenbergら、Ann.Neurol.57巻(5号):664〜78頁(2005年))、転写研究との相関関係について調べた。
血液インターフェロンα/β誘導遺伝子転写産物は、活動性DMおよびより低い程度で活動性のPMの患者からのPBMC中の全ての遺伝子のうちで最も上方制御される
活動性のDM(DMA)およびPM(PMA)についての転写産物発現レベルを健常対照と比較すると、インターフェロンα/βにより誘導される遺伝子が、測定したおよそ38,500個の転写産物のうちで、最も大きい倍率変化および最も高い統計学的有意性を有していた(p値<0.0001)(表2)。25個の最も高く上方制御された遺伝子のうち、少なくとも21個(84%)は、インターフェロンα/β誘導性であることが知られている。これらの遺伝子のうち、IBM、MGまたは遺伝子的に決定された筋疾患の患者において著しく上方制御されたものはなかった。上方制御の大きさは、全般的にDMにおいてPMにおけるよりも高かった。定量的RT−PCRは、インターフェロン誘導遺伝子Mx1およびIFIT1が、DMA血液中で高く上方制御されたことを示し、このことは、マイクロアレイデータからの我々の知見を支持した(表3)。0.99の3重の測定間の高い相関関係で、3重の試料の変動の平均係数は0.15であった。マイクロアレイデータとのRT−PCRデータの相関関係は優れていた(Mx1:R2=0.9889;IFIT1:R2=0.9978)。全体として、8名のDMA患者のうちの8名および7名のPMA患者のうち5名が、研究した全てのその他の50個の血液検体のものを超えるインターフェロンα/β誘導遺伝子の過剰発現のレベルを有した。
我々は、8個のDMA試料の転写プロファイルを11個のDMIと、そして別個に、7個のPMA試料を6個のPMIと比較した。疾患の改善とともに最も高く下方制御された遺伝子は、主にインターフェロンα/β誘導である(表2、右の列)。定量的RT−PCRは、同様に、DM患者における改善を確認した(表3)。2つの異なる時点にて活動性および改善中の疾患を有するDM(N=6)またはPM(N=2)の同じ患者からの対の試料において、1型インターフェロン誘導遺伝子は、再び、全ての遺伝子のうちで最も下方制御された。不応性DMである1名の患者について、対の検体において、多くの1型インターフェロン誘導遺伝子について全体的な変化はほとんど観察されなかった。
15名の患者(DM、PMおよびIBMの5名ずつ)について、我々は、U133A(筋肉プロファイリングのために用いられる)およびU133プラス2.0(血液プロファイリングのために用いられる)マイクロアレイの両方で共有される13,398個の遺伝子を用いて、血液遺伝子発現プロファイルを筋肉遺伝子発現と比較した。DM筋肉において、血液中で高く上方制御されていることを我々が見出したのと同じインターフェロンα/β誘導遺伝子の発現が著しく上方制御される。これとは対照的に、PMおよびIBMの筋肉において、インターフェロンα/β誘導遺伝子転写は、穏やかに増加しただけであった。これは、MxAのようなそれら自体がインターフェロンα/β誘導遺伝子を発現する浸潤性免疫系細胞に起因するのであろう。DMにおいて特に興味深いことに、血液と比較して筋肉において特定のインターフェロン誘導遺伝子が著しく過剰発現する。例えば、ISG15のDM筋肉発現は、正常の筋肉のおよそ570倍で、DM血液よりも約100倍高い。
以前に報告されたように、1つのインターフェロンα/β誘導遺伝子タンパク質MxAの過剰発現が、DMの筋肉に存在する(Greenbergら、Ann.Neurol.57巻(5号):664〜78頁(2005年))。今回の研究において、MxA転写産物レベルは、DMA血液(6.2倍)およびPMA血液(6.0倍)において同様に上昇したが、PM筋肉(2.5倍)と比較してDM筋肉(281倍)において、マイクロアレイ研究により、著しくより高かった。DM筋肉中のMxA転写産物の著しい富化は、DMおよびPMからの筋肉切片の比較における免疫組織化学によるMxAタンパク質の著しい富化を同様に伴う。5名のDM患者のうち4名において、MxA染色が、多くの筋繊維、特に筋束周辺の筋繊維に強く存在したが、PMの5名全ての患者およびIBMの5名において、MxA染色は、浸潤性免疫系細胞に限定されていた。MxA染色は、正常筋肉生検に存在しない。
我々の発見は、IBMの患者ではそうではないが、DMおよびPMの患者のほとんどにおいて、インターフェロンα/β誘導遺伝子の著しい過剰発現を特徴とする独特の血液遺伝子発現プロファイルがあることを示唆する。免疫抑制治療の間の臨床的な改善は、一般的に、これらの遺伝子の過剰発現の通常レベルに向けての低減を伴う。筋肉における遺伝子発現に関連するこれらの発見は、病原の仮説および診断用の使用の可能性がある血液バイオマーカーのための意味を有する。
Claims (22)
- 対象が多発性筋炎または皮膚筋炎に特徴的な遺伝子発現パターンを示すかどうかを決定する方法であって、該方法は、
a)該対象から末梢血単核細胞を含有する試験生物学的試料を得る工程、
b)インターフェロンアルファ誘導タンパク質27;インターフェロン誘発タンパク質44様;ラジカルS−アデノシルドメイン/CIG5;インターフェロン誘発タンパク質44;仮定上のタンパク質LOC129607;2’,5’−オリゴアデニル酸合成酵素1;上皮間質相互作用1;XIAP関連因子1;インターフェロン誘発テトラトリコペプチド5;2’−5’−オリゴアデニル酸合成酵素様;2’−5’−オリゴアデニル酸合成酵素3;インターフェロン誘発テトラトリコペプチド1;リン脂質スクランブラーゼ1;hectドメインおよびRLD5;インターフェロン誘導プロテインキナーゼ;TNFスーパーファミリー、メンバー10;グアニル酸結合タンパク質1;TNF,アルファ誘発タンパク質6;インターフェロン誘発テトラトリコペプチド3;不稔アルファモチーフドメイン9様;クロマチン修飾タンパク質5;ISG15ユビキチン様修飾因子;2’−5’−オリゴアデニル酸合成酵素2;インターフェロン誘発ヘリカーゼCドメイン1;およびミクソウイルス耐性1からなる群から選択される1つまたは複数の遺伝子の発現について該試料をアッセイする工程、
c)段落b)のアッセイにおいて決定された結果から、該遺伝子の1つまたは複数が1つまたは複数の対照試料よりも該試験試料において高く発現されることが示された場合に、該対象が多発性筋炎または皮膚筋炎に特徴的な遺伝子発現プロファイルを有すると結論付ける工程
を含む、方法。 - 前記試験生物学的試料が血液、血漿または血清の試料である、請求項1に記載の方法。
- 前記アッセイがマイクロアレイプレートを用いて行われる、請求項1に記載の方法。
- 前記遺伝子の少なくとも5個の発現が、1つまたは複数の前記対照試料と比較して前記試験試料において上昇する、請求項1に記載のアッセイ。
- 前記遺伝子の少なくとも10個の発現が、1つまたは複数の前記対照試料と比較して前記試験試料において上昇する、請求項1に記載のアッセイ。
- 前記遺伝子の少なくとも15個の発現が、1つまたは複数の前記対照試料と比較して前記試験試料において上昇する、請求項1に記載のアッセイ。
- 段落b)における前記遺伝子の全ての発現が、1つまたは複数の前記対照試料と比較して前記試験試料において上昇する、請求項1に記載のアッセイ。
- 多発性筋炎または皮膚筋炎の患者が、疾患の進行または疾患の寛解に特徴的な遺伝子発現パターンを有するかどうかを決定する方法であって、該方法は、
a)該患者から末梢血単核細胞を含有する試験生物学的試料を得る工程、
b)インターフェロンアルファ誘導タンパク質27;インターフェロン誘発タンパク質44様;ラジカルS−アデノシルドメイン/CIG5;インターフェロン誘発タンパク質44;仮定上のタンパク質LOC129607;2’,5’−オリゴアデニル酸合成酵素1;上皮間質相互作用1;XIAP関連因子1;インターフェロン誘発テトラトリコペプチド5;2’−5’−オリゴアデニル酸合成酵素様;2’−5’−オリゴアデニル酸合成酵素3;インターフェロン誘発テトラトリコペプチド1;リン脂質スクランブラーゼ1;hectドメインおよびRLD5;インターフェロン誘導プロテインキナーゼ;TNFスーパーファミリー、メンバー10;グアニル酸結合タンパク質1;TNF,アルファ誘発タンパク質6;インターフェロン誘発テトラトリコペプチド3;不稔アルファモチーフドメイン9様;クロマチン修飾タンパク質5;ISG15ユビキチン様修飾因子;2’−5’−オリゴアデニル酸合成酵素2;インターフェロン誘発ヘリカーゼCドメイン1;およびミクソウイルス耐性1からなる群から選択される1つまたは複数の遺伝子の発現について該試料をアッセイする工程、
c)段落b)のアッセイにおいて決定された結果から、該遺伝子の1つまたは複数がより早い時期に同じ患者から得られた1つまたは複数の試料よりも該試験試料において高く発現されることが示された場合に、該対象が疾患の進行に特徴的な遺伝子発現プロファイルを有すると結論付けるか、または
d)段落b)のアッセイにおいて決定された結果から、該遺伝子の1つまたは複数がより早い時期に同じ患者から得られた1つまたは複数の試料よりも該試験試料において低く発現されることが示された場合に、該対象が疾患の寛解に特徴的な遺伝子発現プロファイルを有すると結論付ける工程
を含む、方法。 - 前記生物学的試料が血液、血漿または血清の試料である、請求項8に記載の方法。

- 前記疾患が進行していると結論付けるためには、前記遺伝子の少なくとも10個の発現が前記試験試料中で上昇しなければならず、該疾患が進行していると結論付けるためには、該遺伝子の少なくとも10個の発現が該試験試料中で減少しなければならない、請求項8に記載のアッセイ。
- 前記疾患が進行していると結論付けるためには、前記遺伝子の少なくとも15個の発現が前記試験試料中で上昇しなければならず、該疾患が進行していると結論付けるためには、該遺伝子の少なくとも15個の発現が該試験試料中で減少しなければならない、請求項8に記載のアッセイ。
- 前記疾患が進行していると結論付けるためには、段落b)における前記遺伝子の全ての発現が前記試験試料中で上昇しなければならず、該疾患が進行していると結論付けるためには、段落b)における該遺伝子の全ての発現が該試験試料中で減少しなければならない、請求項8に記載のアッセイ。
- 一連の別々の固定化オリゴヌクレオチドを含むマイクロプレートであって、
a)該オリゴヌクレオチドの少なくとも5個が、インターフェロンアルファ誘導タンパク質27;インターフェロン誘発タンパク質44様;ラジカルS−アデノシルドメイン/CIG5;インターフェロン誘発タンパク質44;仮定上のタンパク質LOC129607;2’,5’−オリゴアデニル酸合成酵素1;上皮間質相互作用1;XIAP関連因子1;インターフェロン誘発テトラトリコペプチド5;2’−5’−オリゴアデニル酸合成酵素様;2’−5’−オリゴアデニル酸合成酵素3;インターフェロン誘発テトラトリコペプチド1;リン脂質スクランブラーゼ1;hectドメインおよびRLD5;インターフェロン誘導プロテインキナーゼ;TNFスーパーファミリー、メンバー10;グアニル酸結合タンパク質1;TNF,アルファ誘発タンパク質6;インターフェロン誘発テトラトリコペプチド3;不稔アルファモチーフドメイン9様;クロマチン修飾タンパク質5;ISG15ユビキチン様修飾因子;2’−5’−オリゴアデニル酸合成酵素2;インターフェロン誘発ヘリカーゼCドメイン1;およびミクソウイルス耐性1からなる群から選択される遺伝子配列とストリンジェントな条件下で特異的にハイブリッド形成し、
b)該遺伝子配列とストリンジェントな条件下で特異的にハイブリッド形成する該オリゴヌクレオチドが、その他の配列とストリンジェントな条件下でハイブリッド形成するいかなるオリゴヌクレオチドも含有しない該マイクロアレイプレート上の場所に固定化され、
c)該マイクロアレイプレートが、合計で500個以下の別々の固定化オリゴヌクレオチドを含む
マイクロアレイプレート。 - 段落b)の遺伝子から選択される遺伝子配列とストリンジェントな条件下で特異的にハイブリッド形成する少なくとも10個の別々の固定化オリゴヌクレオチドを含む、請求項15に記載のマイクロアレイプレート。
- 段落b)の遺伝子から選択される遺伝子配列とストリンジェントな条件下で特異的にハイブリッド形成する少なくとも15個の別々の固定化オリゴヌクレオチドを含む、請求項15に記載のマイクロアレイプレート。
- 段落b)の遺伝子の全てとストリンジェントな条件下で特異的にハイブリッド形成する別々の固定化オリゴヌクレオチドを含む、請求項15に記載のマイクロアレイプレート。
- 合計で100個以下の別々の固定化オリゴヌクレオチドを含む、請求項18に記載のマイクロアレイプレート。
- 段落b)の遺伝子以外の遺伝子とストリンジェントな条件下でハイブリッド形成するあらゆる別々の固定化オリゴヌクレオチドも含まない、請求項18に記載のマイクロアレイプレート。
- 対象の遺伝子発現プロファイルを決定する方法であって、
a)該対象から末梢血単核細胞を含有する試験生物学的試料を得る工程、
b)請求項15〜20のいずれか1項のマイクロアレイプレートを用いて該試験生物学的試料中の遺伝子発現をアッセイする工程
を含む、方法。 - 対象における多発性筋炎または皮膚筋炎を診断する方法であって、
a)該対象から血液または筋肉の試験生物学的試料を得る工程、
b)該試験生物学的試料をアッセイして、IFI44L遺伝子および/またはEPSTI1の発現レベルを決定する工程、
c)段落b)で決定される遺伝子発現レベルが、1つまたは複数の対照生物学的試料中よりも高い場合に、該対象が多発性筋炎または皮膚筋炎を有すると結論付ける工程
を含む、方法。
