本発明は、一般的な表面洗浄と汚染除去のほか、広い種類の化学的及び生物学的兵器原料を含む汚染除去のための使用に適した過酸水溶液を効果的に生成するペルヒドロラーゼ酵素、過酸化水素源及びエステル基質を含む安定な組成物に関する。
過酸は汚染除去/消毒剤として広く受け入れられている。最も顕著には、過酢酸(「ペルオキシ酢酸」又は「PAA」として知られている)は広く使用され、特にバイオハザードの汚染除去のほか、食品産業と病院で使用される製品の滅菌用に使用される。PAAはまた、洗浄剤組成物とパルプの漂白のような工業プロセスにおいて漂白用化学物質としても使用できる。PAAは、使用後に比較的わずかな処理の問題しか起こさない。なぜなら汚水処理プラントで容易に分解される化合物に分解されるからである。これは微生物に対する活性の広いスペクトラムをもち、低温ですら効果をもつ。
本発明は、水溶液中で過酸を酵素的に生成し、広い範囲の汚染除去の方法と用途で使用できる安定な組成物、系及び用具一式を提供する。
本発明は水溶液中で過酸を酵素的に生成し、広い範囲の汚染除去法と用途で使用できる安定な組成物、系、用具一式を提供する。本発明は、一部、アシル基転移酵素のようなペルヒドロラーゼ酵素、過酸化水素源及びプロピレングリコールジアセテートのようなエステル基質を含む組成物が使用前に極めて安定であるという驚くべき発見に基づいている。実際に、これら3化合物からなる組成物は少なくとも7日、約14日、約30日、約60日またはそれより長く安定であることができ、ここで安定性とは水に稀釈したとき有効量の過酸を生成する性能に対応する。
II.定義
例えば、「熱的安定性」とは、本明細書で使用される場合、上昇した温度で物質または組成物がその機能を維持する(つまり、分解しない)性能をいう。安定性の実質的な変化は、選択された環境条件が欠けたときに存在する活性と比較して、少なくとも定量を受ける機能の半減期の少なくとも約5%またはそれより高い又は低い増大又は減少(殆どの実施態様で、増大が好ましい)により根拠付けられる。

VII.殺菌、消毒及び/または汚染除去の方法と用途

Claims (35)
- ペルヒドロラーゼ酵素、過酸化水素源及びエステル基質を含む組成物であって、前記組成物は水と混合されたときに過酸を生成し、また前記組成物は、少なくとも約7日の期間、過酸生成活性を保持する。
- 請求項1の組成物であって、当該エステル基質は液体であり、当該ペルヒドロラーゼ酵素と当該過酸化水素源は、当該液体中の固体懸濁物であるものである組成物。
- 請求項1の組成物であって、当該過酸は過酢酸、過ノナン酸、過プロピオン酸、過ブタン酸、過ペンタン酸及び過ヘキサン酸から選択されるものである組成物。
- 請求項4の組成物であって、当該過酸は過酢酸である組成物。
- 請求項1の組成物であって、当該エステル基質はプロピレングリコールジアセテートであるものである組成物。
- 請求項1の組成物であって、当該ペルヒドロラーゼ酵素はSEQID NO:1に表わされたアミノ酸配列を含むものである組成物。
- 請求項1の組成物であって、当該ペルヒドロラーゼ酵素はSEQID NO:1のS54V変異種であるものである組成物。
- 請求項1の組成物であって、当該過酸化水素源は過炭酸ナトリウム、過ホウ酸ナトリウム及び尿素過酸化水素から選択される過酸化水素生成化合物であるものである組成物。
- 請求項8の組成物であって、当該過酸化水素源は過炭酸ナトリウムであるものである組成物。
- 請求項1の組成物であって、当該組成物は、重量で約0.001%から約1%のペルヒドロラーゼ酵素、重量で約35%から約45%の過炭酸ナトリウム、及び重量で約55%から約65%のプロピレングリコールジアセテートを含む。
- 請求項1の組成物であって、当該過酸化水素源は酸化酵素とその基質を含む酵素的過酸化水素生成系であるものである組成物。
- 請求項11の組成物であって、当該酸化酵素はグルコース酸化酵素、ソルビトール酸化酵素、ヘキソース酸化酵素、コリン酸化酵素、アルコール酸化酵素、グリセロール酸化酵素、コレステロール酸化酵素、ピラノース酸化酵素、カルボキシアルコール酸化酵素、L-アミノ酸酸化酵素、グリシン酸化酵素、ピルビン酸酸化酵素、グルタミン酸酸化酵素、サルコシン酸化酵素、リジン酸化酵素、乳酸酸化酵素、バニリル(vanillyl)酸化酵素、グルコレート酸化酵素、ガラクトース酸化酵素、ウリカーゼ、シュウ酸酸化酵素、キサンチン酸化酵素から選択されるものである組成物。
- 請求項1の組成物であって、さらにプロテアーゼ、セルラーゼ、アミラーゼ、及び微生物の細胞壁を分解する酵素から選択される少なくとも一つの追加の酵素を含む組成物。
- 請求項1の組成物であって、当該組成物はさらに少なくとも一つの界面活性剤を含むものである組成物。
- 請求項1から14のいずれかの請求項の組成物を水へ加え少なくとも約20分間混合することを含む汚染除去溶液を調製する方法。
- 汚染除去の方法であって、
(a) 請求項1から14のいずれかの請求項に従い組成物を与え
(b) 前記組成物を水に加え、混合し、それにより過酸水溶液を生成し
(c )前記溶液へ汚染物質を含む物品を暴露する
ことを含む方法。 - 請求項16の方法であって、当該組成物は水溶性容器に含まれているものである方法。
- 請求項18の方法であって、当該容器はポリビニルアルコールフィルムから作られている小袋である方法。
- 請求項16の方法であって、当該水は滅菌されているものである方法。
- 請求項16の方法であって、前記暴露は高温で前記溶液に前記物品を暴露することを含む方法。
- 請求項16の方法であって、前記汚染物質は、ボツリヌス毒素、炭疽菌毒素、リシン、サバ毒素、シガトキシン、テトロドトキシン、マイコトキシン及びそれらの組合せから選択される毒素を含むものである方法。
- 請求項16の方法であって、前記汚染物質は細菌、ウィルス、菌類、寄生虫、プリオン、及びそれらの組み合わせから選択される病原体を含むものである方法。

- 請求項16の方法であって、前記物品は、硬い表面、布地、食品、飼料、衣服、敷物、カーペット、織物、医療機器、動物用医療機器から選択されるものである方法。
- 請求項1から15のいずれかの請求項に従い安定な組成物を含む水溶性容器を含む系。
- 請求項25の系であり、前記容器は小袋、アンプルまたは球である系。
- 請求項26の系であり、当該水溶性容器は水溶性ポリマー物質ポリビニルアルコール、ポリエチレンオキサイド、セルロース由来の水溶性炭水化物、及びポリアクチドから作られているものである系。
- 請求項26の系を水に加え、混合することを含む汚染除去溶液を調製する方法。
- 請求項1から15のいずれかの請求項の安定な組成物を包装して含む汚染除去用用具一式。
- 請求項29の用具一式であって、当該組成物は水溶性容器に入れられているものである用具一式。
- 請求項30の用具一式であって、当該水溶性容器は小袋、アンプル、カプセル又は球であるものである用具一式。
- 請求項30の用具一式であって、当該水溶性容器はポリビニルアルコール、ポリエチレンオキサイド、セルロース由来の水溶性炭水化物及びポリラクチドから選択される水溶性原料から作られるものである用具一式。
- 請求項29の用具一式であって、さらに前記組成物を水と混合するために空の混合容器を含むものである用具一式。
- 請求項29の用具一式であって、さらに密封した容器に水を含み、当該水は滅菌されているものである用具一式。
- 請求項29の用具一式であって、さらに請求項16に従った方法で使用するための指図を含むものである用具一式。
