Description
クライティン-IIは、イクオリンおよびクライティン-Iと比較すると、蛋白質量あたりの全発光量はほぼ同一であるが、発光初速強度は約5倍である。すなわち、クライティン-IIのS/N比はイクオリンおよびクライティン-Iの5倍である。このことは、検出系においてクライティン-IIを使用する場合に、イクオリンおよびクライティン-Iより優れている可能性を示唆している。
(1)アポクライティン-IIタンパク質をコードするコドン最適化核酸。
(2)配列番号:1のアミノ酸配列を有するアポクライティン-IIタンパク質またはその変異体をコードする、前記第1項に記載のコドン最適化核酸。
(3)前記第1項または第2項に記載のコドン最適化核酸と、他のタンパク質をコードする核酸とが融合したコドン最適化核酸。
(4)CGCアルギニンコード化コドン、AACアスパラギンコード化コドン、GACアスパラギン酸コード化コドン、CAGグルタミンコード化コドン、GAGグルタミン酸コード化コドン、GGCグリシンコード化コドン、CACヒスチジンコード化コドン、ATCイソロイシンコード化コドン、CTGロイシンコード化コドン、AAGリジンコード化コドン、CCCプロリンコード化コドン、TTCフェニルアラニンコード化コドン、TCCまたはAGCセリンコード化コドン、TACチロシンコード化コドン、およびGTGバリンコード化コドンからなる群から選ばれた1種以上のコドンの数が、配列番号:2の野生型クラゲアポクライティン-IIタンパク質をコードする核酸よりも多い、前記第1項から第3項の何れか1項に記載のコドン最適化核酸。
(5)前記第1項から第4項の何れか1項に記載のコドン最適化核酸を用いて発現されたアポクライティン-IIタンパク質。
(6)前記第5項に記載のアポクライティン-IIを構成成分とするクライティン-II。
(7)アポクライティン-IIタンパク質をコードするコドン最適化核酸の位置が、哺乳類細胞において作動するプロモーターの制御調節下である、発現ベクター。

組換え発現ベクターを宿主細胞に導入する方法は、特に限定されるものではなく、前述あるいは後述の方法により可能である。また、コドン最適化アポクライティン-IIタンパク質を発現させる方法も特に限定されるものではなく、コドン最適化アポクライティン-IIタンパク質の発現を可能とするのに適する条件において宿主細胞を培養する方法などが挙げられる。
(iv)発現したコドン最適化アポクライティン-IIタンパク質を有意な量の他の細胞内タンパク質から精製する工程
前述の(iv)の工程の後、コドン最適化アポクライティン-IIタンパク質の純度は、70%以上、85%以上、さらには95%以上であることが好ましい。

実験例1 コドン最適化アポクライティン-II核酸の設計および化学合成
コドン最適化アポクライティン-II核酸の設計は、アポクライティン-IIのアミノ酸配列を変えることなく、ヒトにおける高頻度使用のアミノ酸コドンの選択のみならず、転写因子認識配列の不活性化配列へ変換、スプライシングの可能性のある部位除去、6塩基認識の制限酵素部位の削除、ループ構造を作るパリンドローム配列が形成しないように配列設計を行った(配列表3)。コドン
最適化アポクライティン-IIコドン使用頻度表を表2 に示した。野性型アポクライティン-IIコドン頻度表の表1にくらべ、明らかにコドン最適化アポクライティン-II核酸は、ヒト化コドンへ最適化設計されている。
最適化設計したコドン最適化アポクライティン-II核酸を、常法により化学合成法により合成した。pBlueScript SK(+)(Stratagene社)のEcoRI/SalI制限酵素サイトにクローン化し、pBlue-hCLIIプラスミドを構築した。コドン最適化アポクライティン-II核酸の確認は、塩基配列(ABI社製)をDNAシークエンサーにて確認した。
コドン最適化アポクライティン-IIタンパク質発現ベクターの構築は以下の通りである。先ず、動物培養細胞での新規発現ベクターpCMX-Linker、を構築した。具体的には、pCMX-GFP (Ogawa et al., (1995) PNAS. 92(25), 11899-11903記載)において、制限酵素部位であるAsp718/SalI部位に、化学合成したマルチクローニング配列をもつリンカーLinker F(5)A-Sal (5' GT ACC ACC ATG CTC GAG CTG CAG GAA TTC TCT AGA G 3')(配列番号7)、および Linker R(5)A-Sal (5' TC GAC TCT AGA GAA TTC CTG CAG CTC GAG CAT GGT G 3')(配列番号8)を挿入して新規発現ベクターpCMX-Linkerを構築した。すなわち、新規発現ベクターは、CMVのプロモーターに制御され、その下流にコザック配列、マルチクローニングサイト配列(Asp718I/ XhoI/ PstI/ EcoRI/ XbaI/ SalI/ EcoRV/ BamHI/ MscI/ NheI)を有する。
次に、新規発現ベクターpCMX-Linkerを用いたコドン最適化アポクライティン-IIタンパク質発現ベクターの構築は以下の通りである。 pBlue-hCLIIを常法により制限酵素EcoRI/SalIにて消化した後、pCMX-LinkerのEcoRI-SalI 部位に連結することによって、図1に示す発現ベクターpCMX-hCLIIを構築した。なお、DNA シークエンサー(ABI社製)により塩基配列を決定することにより、インサートDNAの確認を行った。また、コドン最適化アポクライティン-II核酸の対照となる野生型アポクライティン核酸を含むコントロールベクターであるpCMX-CLIIは、同様の方法により構築した。
(1)発現プラスミドの切断および精製
実施例2にて得られたpCMX-hCLIIプラスミドを用いて以下の実験を行った。大腸菌JM83内のpCMX-hCLIIプラスミドを、Endofree Plasmid Maxi kit (QIAGEN社製)を用いて精製した。1 μg/ μlの濃度になるよう滅菌水に溶解した。同様にして、pCMX-hCLII,および内部標準のためのホタルルシフェラーゼベクター(pGL-control:プロメガ社)を使用した。
(2)トランスフェクション
ヒト子宮頸部がん由来細胞株であるHeLa株を、10%牛胎児血清(インビトロジェン社)を含むDMEM培地(高グルコース含:和光純薬)にて培養し、2 x 105 細胞/ウエルにて6ウエルプレートに播種し、インキュベーター中37 ℃、5 % CO2にて培養した。24時間後、精製pCMX-hCLIIプラスミドをFuGene HD(ロッシュ社)トランスフェクションキットを用いて、HeLa細胞にトランスフェクション 実験に用いた。具体的には、100μl のDMEM培地に、アポクライティン-II発現ベクターと内部標準ホタルルシフェラーゼ発現ベクターそれぞれ1μl/μlと FuGene HD 6μlを加え、室温で15分間放置した。50μlのDNA−FuGene 複合体溶液を、6ウエルの細胞に添加した。24時間培養後、2mlの冷PBSで3回洗浄した。細胞に250μlの冷PBSを加え、細胞を回収した。回収した細胞は、超音波破砕装置で破砕し、測定用酵素溶液とした。
(1)内部標準用ホタルルシフェラーゼの測定方法:
実験例3で得られた測定用酵素溶液10μlを、50μl の酵素アッセイ用試薬(プロメガ社)に加え、発光反応を開始させた。発光活性は、発光測定装置(アトー社製:AB2200)で、10秒間での発光積算し、相対発光強度で表記した。
(2)発光活性測定:
実験例3で得られた測定用酵素溶液50μl を、1μl のメルカプトエタノール(和光純薬)、1μl のセレンテラジン(チッソ社製)、および10mMEDTA(和光純薬)を含む950μlの50mMTris−HCl(pH7.6)に加え、4oC で3時間静置し、クライティン-IIを再生させた。この再生溶液10μlに100μlの50mM CaCl2 を含む50mMTris−HCl(pH7.6)溶液を、インジェクションすることにより発光させ、発光測定装置(アトー社製:AB2200)で、10秒間での発光測定し、最大発光強度を相対発光値で示した。
その結果を表3にまとめた。内部標準で使用したホタルルシフェラーゼ活性より、活性化率を求めた結果、野生型アポクライティン-IIに比べ、本発明のコドン最適化クライティン-IIタンパク質は約15倍以上の活性を示すことが明らかとなった。
大腸菌においてコドン最適化アポクライティン-II核酸を発現させるために、出発となる発現ベクターpiP-H-L(6)を構築した。具体的には、特開2008-22848記載のpiP-(His6)HEベクターのHindIII-BamHIサイトに、化学合成したマルチクローニング配列をもつリンカーH/P/S/Xb/Xh/B:Linker-F(5' AG CTT CTG CAG GTC GAC TCT AGA CTC GAG G 3') (配列番号9)、及びH/P/S/Xb/Xh/B:Linker-R(5' GA TCC CTC GAG TCT AGA GTC GAC CTG CAG A 3') (配列番号10)を挿入し、piP-H-L(6)を構築した。基本ベクターpiP-H-L(6)は、大腸菌のリポプロテインのプロモーター、ラクトースのオペロンに制御され、分泌のためのOmpA配列、キレートゲルによる精製のための6個のヒスチジン配列、種々のマルチクローニング配列を有する。
基本ベクターpiP-H-L(6)を用いたコドン最適化アポクライティン-IIタンパク質発現ベクターの構築は以下の通りである。pBlue-hCLIIを鋳型としてPCRプライマーペア:hCLII-1N/HindIII(5' ggc aag ctt GTG AAG CTG GAC CCC GAC TTC 3')(配列番号11)及びhCLII-2C/XhoI(5' ggc CTC GAG TTA GGG GAC GAA GTT GCC GTA 3')(配列番号12)を用いて、PCRキット(日本ジーン社製)にてPCR(サイクル条件:25サイクル;1分/94℃、1分/50℃、1分/72℃)を行った。得られた断片をPCR精製キット(キアゲン社製)で精製し、制限酵素HindIII/XhoIで消化した後、piP-H-L(6)の制限酵素HindIII/XhoI部位に挿入することによって、図2に示す発現ベクターpiP-H-hCLIIを構築した。なお、DNA シークエンサー(ABI社製)により塩基配列を決定することにより、インサートDNAの確認を行った。
コドン最適化アポクライティン-IIタンパク質(以下「hCLII」と言うことがある。)を、大腸菌において発現させるため、実施例2で作製した組換えプラスミドpiP-H-hCLIIを用いた。常法により大腸菌WA802株に導入し、得られた形質転換株をアンピシリン(50μg/ml)を含有する10mlのLB液体培地(水1リットルあたり、バクトトリプトン10g、イーストイクストラクト5g、塩化ナトリウム5g、pH7.2)に植菌し、30℃で18時間培養を行った。次いで、その培養物を新たなLB液体培地400ml x 5本(総量2L)に添加して30℃で18時間培養を行った。培養後、菌体を遠心回収(5,000rpm、5分)し、集菌された培養菌体をタンパク質抽出の出発材料とした。
集菌した培養菌体を200mlの50mM Tris-HCl(pH7.6)で懸濁し、氷冷下で超音波破砕処理(ブランソン社製、Sonifier model cycle 250)を3分間、3回行い、その菌体破砕液を10,000 rpm(12,000×g)で4 ℃、20分間遠心した。得られた可溶性画分を、50mM Tris-HCl(pH7.6)で平衡化したニッケルキレートカラム(アマシャムバイオサイエンス社、カラムサイズ:直径2.5×6.5cm)に供しコドン最適化アポクライティン-IIタンパク質を吸着させた。500mlの50mM Tris-HCl(pH7.6)で洗浄後、0.1Mイミダゾール(和光純薬工業社製)によりコドン最適化アポクライティン-IIタンパク質を溶出した。精製蛋白質濃度を、ウシ血清アルブミン(ピアス社製)を標品としてBradford法にもとづく市販のキット(バイオラッド社製)を用いて決定したところ、2Lの培養菌体より45.7mgのコドン最適化アポクライティン-IIタンパク質を得た。SDS-PAGE分析の結果、純度は95%以上であった。
精製過程における、コドン最適化アポクライティン-IIタンパク質を構成成分とするクライティン-IIの発光測定は、10 mM EDTA を含む30mM Tris−HCl(pH7.6)990 μlに2-メルカプトエタノール(1μl)、エタノールに溶解した基質セレンテラジン(1μg/μl)を混合した後、ヒト型アポクライティン-II溶液10μlを添加し、氷上(4℃)で2時間放置した。再生ヒト型クライティン-II溶液10μlへ、50mMカルシウム溶液を100μl加えることにより発光反応を開始させた。発光活性は、発光測定装置(アトー社製:AB2200)で、10秒間での発光測定し、最大発光強度を相対発光値で示した(表4)。
Claims (17)
- アポクライティン-IIタンパク質をコードするコドン最適化核酸。
- 配列番号:1のアミノ酸配列を有するアポクライティン-IIタンパク質またはその変異体をコードする、請求項1に記載のコドン最適化核酸。
- 請求項1または2に記載のコドン最適化核酸と、他のタンパク質をコードする核酸とが融合したコドン最適化核酸。
- CGCアルギニンコード化コドン、AACアスパラギンコード化コドン、GACアスパラギン酸コード化コドン、CAGグルタミンコード化コドン、GAGグルタミン酸コード化コドン、GGCグリシンコード化コドン、CACヒスチジンコード化コドン、ATCイソロイシンコード化コドン、CTGロイシンコード化コドン、AAGリジンコード化コドン、CCCプロリンコード化コドン、TTCフェニルアラニンコード化コドン、TCCまたはAGCセリンコード化コドン、TACチロシンコード化コドン、およびGTGバリンコード化コドンからなる群から選ばれた1種以上のコドンの数が、配列番号:2の野生型クラゲアポクライティン-IIタンパク質をコードする核酸よりも多い、請求項1から3の何れか1項に記載のコドン最適化核酸。
- 請求項1から4の何れか1項に記載のコドン最適化核酸を用いて発現されたアポクライティン-IIタンパク質。
- 請求項5に記載のアポクライティン-IIを構成成分とするクライティン-II。
- アポクライティン-IIタンパク質をコードするコドン最適化核酸の位置が、哺乳類細胞において作動するプロモーターの制御調節下である、発現ベクター。
- アポクライティン-IIタンパク質をコードするコドン最適化核酸、および哺乳類細胞においてアポクライティン-IIタンパク質をコードするコドン最適化核酸の発現を制御する制御調節配列を有する、請求項7に記載の発現ベクター。
- アポクライティン-IIタンパク質をコードするコドン最適化核酸を有する組換え宿主細胞。
- 宿主細胞が哺乳類細胞である、請求項9に記載の組換え宿主細胞。
- 宿主細胞がヒト細胞である、請求項9に記載の組換え宿主細胞。
- 宿主細胞が非ヒト哺乳類細胞である、請求項9に記載の組換え宿主細胞。
- 下記(i)〜(iii)の工程を有するアポクライティン-IIタンパク質の製造方法。
アポクライティン-IIタンパク質をコードするコドン最適化核酸を、宿主細胞において作動するプロモーターの制御調節下に置いた組換え発現ベクターを調製する工程
(ii)該組換え発現ベクターを宿主細胞に導入する工程
(iii)当該宿主細胞を培養し、アポクライティン-IIタンパク質を発現させる工程 - アポクライティン-IIタンパク質をコードするコドン最適化された核酸と、当該核酸を発現し得る制御調節核酸とが結合した核酸を、宿主細胞に導入することを特徴とする、クライティン-IIの発光強度増強方法。
- 宿主細胞におけるクライティン-IIの発光強度を増強させるための、アポクライティン-IIタンパク質をコードするコドン最適化核酸の使用。
- アポクライティン-IIタンパク質を発現するように遺伝子操作された宿主細胞を、セレンテラジンまたはセレンテラジン誘導体と接触させ、次いで、発生した光量を測定することを特徴とする、活性化時の細胞内カルシウム流動の変化を仲介する受容体を阻害する化合物の能力測定方法。
- さらに、受容体阻害の能力を記録する、請求項16に記載の方法。
