Description
第2に、標的抗原を発現する細胞は、免疫機能を残したまま、その抗原を発現することをやめることによってそのブロッキング因子の存在に適用できるようになりうる。この状況においては、単独の免疫抑制因子による持続式処置は無効となる。
以上に基づき、免疫応答を抑制できる追加の、且つ改善された因子、特に選択的抑制の可能な因子についてのニーズがあることが明らかである。本発明は、活性化ヒトCD4+ T−リンパ球上に局在するレセプターに対するリガンド(ACT−4−L)を提供することにより、このような及びその他のニーズをある程度満足せしめる。
(a)ATCC HB11483として寄託されたハイブリドーマHBL106により生産されるL106と命名されているモノクローナル抗体以外のものである;そして
(b)図10−1〜図10−2に示す完全アミノ酸配列以外のアミノ酸配列を有する。
本発明は更に、 ACT−4−L−h−1、好ましくはその細胞外ドメインに特異的に結合する抗体の如くの結合性成分を考慮する。
本発明は更に任意の上記の特異的結合性パートナーに特異的な結合性成分を提供し、その結合性成分は ACT−4レセプターポリペプチド以外のものであり、そして図10−1〜図10−2に示す完全アミノ酸配列を有するポリペプチドとは交差反応性でない。
適切には、この結合性成分は抗体結合性ドメインを含んで成る。
抗体はモノクローナル抗体であってよい。この結合性成分は抗体のFab 又はF(ab)'2を含んで成りうる。
ACT−4レセプターポリペプチドに対する特異的結合性パートナーを試験すべき物質と接触させる;そして
この特異的結合性パートナーとこの因子、即ち試験する物質との結合を検出する;
ことを含んで成る。
上記の適当な結合性成分を患者に投与する;そして
その患者を選定の抗原に暴露する;
ことを含んで成る。
本発明は更に活性化CD4+ T細胞を検出するためのキットを提供し、これは上記の ACT−レセプターに対する特異的結合性パートナーを含んで成る。
他方、本発明は ACT−4レセプター又はその特異的に結合性の類似体を、図10−1〜図10−2に示すアミノ酸配列を有するポリペプチド又は請求項1記載の特異的結合性パートナーの認識のための特異的結合性反応に利用することを提供する。
20種の天然アミノ酸についての略語は慣用の用法に従う(Immunology-A Synthesis, E. S. Golub & D. R. Gren編Sinauer Associates, Sunderland, MA. 第2版、1991)(引用することで本明細書に組入れる)。20種の慣用アミノ酸の立体異性体(例えばD−アミノ酸)、非天然アミノ酸、例えばα,α−ジ置換化アミノ酸、N−アルキルアミノ酸、乳酸及びその他の慣用的でないアミノ酸も本発明のポリペプチドにとって適切な成分である。慣用的でないアミノ酸の例には、4−ヒドロキシプロリン、γ−カルボキシグルミン酸塩、ε−N,N,N−トリメチルリジン、ε−N−アセチルリジン、O−ホスホセリン、N−アセチルセリン、N−ホルミルメチオニン、3−メチルヒスチジン、5−ヒドロキシリジン、ω−N−メチルアルギニン並びにその他の類似のアミノ酸及びイミノ酸(例えば4−ヒドロキシプロリン)が含まれる。
特異的な結合性は、二量複合体についての解離定数が≦1μM、好ましくは≦100nM 、そして最も好ましくは≦1nMであるときに存在しているといえる。
「患者」なる語はヒト及び脊椎対象体を含む。
I. ACT−4レセプターポリペプチド
本発明の一態様に従うと、活性化CD4+ T細胞の表層上のレセプター(ACT−4レセプターと呼ぶ)及びそのフラグメントが提供される。 ACT−4レセプターポリペプチドなる語は、一般に全長タンパク質及びそのフラグメントを包括するように用いられている。 ACT−4レセプターなる語は通常全長タンパク質のために確保されている。特性決定する最初の ACT−4レセプターのアミノ酸配列(以降 ACT−4−h−1)を図5−1〜図5−2に示す。接尾辞hはヒト起源を意味し、そして接尾辞−1は ACT−4−h−1が特性決定される最初の ACT−4であることを示す。 ACT−4レセプターなる語は図5−1〜図5−2に示す配列を有するタンパク質のみを意味するだけでなく、 ACT−4−h−1の対立形質型、非対立形質型及び高度同系の変異体、並びに任意のこれらの天然又は誘導型突然変異体をも意味する。
本発明は、 ACT−4レセプターポリペプチドに特異的に結合し、且つかかるポリペプチドと、少なくともある程度、非共有結合によって複合体を形成できるリガンドも提供する。 ACT−4−Lリガンドポリペプチドなる語は一般に全長タンパク質及びそのフラグメントを包括するように利用される。この語は通常は ACT−4レセプターポリペプチドに対する抗体を含まない。 ACT−4リガンドなる語は通常全長タンパク質を意味するのに用いる。リガンドは天然でも合成分子でもよく、そして可溶性形態でも、細胞表層に定着されていてもよい。
A.組換技術
図5−1〜図5−2に示す ACT−4−h−1のヌクレオチド及びアミノ酸配列、並びにその他の ACT−4レセプター変異体についての対応の配列は、全長 ACT−4レセプターポリペプチド配列のポリペプチド及びそのフラグメントの製造を可能にする。同様に、 ACT−4−L−h−1のアミノ酸配列及びその他の ACT−4−Lリガンドポリペプチドについての対応の配列は全長及びフラグメントリガンドポリペプチドの製造を可能にする。かかるポリペプチドは ACT−4又は ACT−4−Lをコードするポリヌクレオチド又はこれらのいづれかのフラグメント及び類似体の発現により原核又は真核宿主細胞において製造されうる。
天然 ACT−4レセプターポリペプチドはアフィニティークロマトグラフィーの如くの慣用の技術により単離される。例えば、ポリクローナル又はモノクローナル抗体を事前に精製しておいた ACT−4−h−1に対して生起させ、そして公知の技術によって適当なアフィニティーカラムに付加させる。例えばHudson & Hay, PracticalImmunolgy (Blackwell Scientific Publications, Oxfurd, UK, 1980) 第8章を参照のこと(引用することで本明細書に組入れる)。例えば、抗− ACT−4−h−1はそのFcドメインをホモ二価架橋剤、例えばジメチルピメリミデートと架橋させることを介してプロテインAセファロースカラムに固定化できる。
他方、 ACT−4又は ACT−4−Lポリペプチドは化学的な方法により合成できるか、又は翻訳を誘導するポリヌクレオチド鋳型を利用するインビトロ翻訳系により製造される。ポリペプチドの化学合成及びインビトロ翻訳のための方法は当業界に公知であり、そしてBerger & Kimmel, Methods in Enzymology、第 152巻、Gnide to Molecular Cloning Techniques Academic Press, Inc., San Diego, CA, 1987 に更に記載されている。
A. ACT−4又は ACT−4−L核酸のクローニング
実施例5は ACT−4−h−1と命名した ACT−4レセプターのcDNAクローンについての核酸配列データーを提供する。この配列は翻訳領域並びに3’及び5’フランキング領域の双方を含む。この配列データーはプローブをデザインするために利用でき、そのプローブによりその他の ACT−4レセプター遺伝子を単離する。これらの遺伝子には ACT−4−h−1をコードするヒトゲノム遺伝子、並びに高等哺乳動物由来のcDNA及びゲノムクローン、並びに対立形質及び非対立形質、並びにこれらの遺伝子の全ての天然及び誘導化突然変異体が含まれる。詳しくは、本願において開示する全ての ACT−4レセプターポリペプチドをコードする全ての核酸フラグメントを提供する。多くの種のゲノムライブラリーが市販されており(例えばClontech, Palo Alto, CA)、又は慣用の手順により新規に単離できる。cDNAライブラリーは最良には、 ACT−4−h−1を大量に発現する活性化CD4+ 細胞から調製する。
本発明のさらなる態様においては、 ACT−4及び ACT−4−Lポリペプチドに対する抗体が供給される。
抗体又は免疫グロブリンは典型的には、4種の共有結合されたペプチド鎖から構成される。たとえば、IgG 抗体は2種のL鎖及び2種のH鎖を有する。個々のL鎖はH鎖に共有結合されている。他方、個々のH鎖は、免疫グロブリンコンホーメーションとしても知られている“Y”配置を形成するために他に共有結合される。それらの分子のフラグメント、又はH又はL鎖のみでさえ、抗原を結合することができる。抗体、抗体のフラグメント、及び個々の鎖はまた、免疫グロブリンとしても本明細書において言及される。
ACT−4受容体、 ACT−4−Lリガンド又はそれらのいづれかの結合フラグメントのいづれかを結合する抗体は、種々の手段でより生成され得る。非ヒトモノクローナル抗体、たとえばネズミ、ラット、等のモノクローナル抗体の生成は良く知られており、そしてたとえば ACT−4受容体又はそのリガンド、又はそれらのいづれかの免疫原性フラグメントを含む調製物により動物を免疫化することによって達成され得る。特に、組換え ACT−4又は ACT−4−L遺伝子により安定してトランスフェクトされ、そして ACT−4受容体又はそれらに対するリガンドをそれらの細胞表面上に発現する細胞が特に免疫原として有用である。
(1)抗原を直接的に非共有結合しているか、
(2)CDR 領域に隣接しているか、
(3)さもなければ、CDR 領域(たとえばCDR 領域の約3Å以内に存在する)と相互作用するか、又は
(4)VL −VH 界面に関与していることが正当に予測される場合、マウス抗体からの同等の骨格アミノ酸により置換されるはずである。
ACT−4−Lリガンドポリペプチドに対する選択された抗体に対する同じか又は異なった結合特異性を有する抗体は、類似する方法により同定され得る。
L106抗体、又は ACT−4受容体に対するいづれか他の選択された抗体により結合されるエピトープは、 ACT−4受容体ポリペプチド、たとえば ACT−4−h−1からの異なったアミノ酸セグメントを含むフラグメントのファミリーを提供することによって決定される。個々のフラグメントは典型的には、少なくとも4,6,8,10,20,50又は 100個の隣接するアミノ酸を含んで成る。集合的には、ポリペプチドのファミリーは、完全な長さの ACT−4受容体ポリペプチドのアミノ酸配列の多く又はすべてを包含する。そのファミリーのメンバーは、たとえばL106抗体への結合について個々に試験される。試験下で抗体に特異的に結合できる最少フラグメントは、抗体により認識されるエピトープのアミノ酸配列を示す。類似するアプローチが、 ACT−4リガンドポリペプチドに対する抗体により結合されるエピトープをマッピングするために使用される。
本発明のもう1つの態様においては、 ACT−4受容体又はそれらのリガンドに対する抗体のフラグメントが供給される。典型的には、それらのフラグメントは、少なくとも 107M及びより典型的には108 又は 109Mの親和性を有する、 ACT−4受容体又はリガンドに対する特異的結合性を示す。抗体フラグメントは、別々のH鎖、L鎖Fab, Fab',F(ab')2, Fabc 及びFvを含む。フラグメントは、組換えDNA 技法により、又は損なわれていない免疫グロブリンの酵素的又は化学的分離により生成される。
上記に論ぜられた抗体及びそれらのフラグメントを生成するすべてのハイブリドーマ、トリオーマ及び他の細胞系は本発明において明白に包含される。それらは、L106マウス抗体を生成する、ATCC HB11483としてAmerican Type Culture Collection, Rockville, MD20852 にブダペスト条約に基づいて寄託されたハイブリドーマ系HBL106を含む。
ACT−4及び ACT−4−Lポリペプチドに対する抗体及びそれらの結合フラグメントは、好ましくは種々の組織に由来するヒト又は霊長類cDNAを含むcDNA発現ライブラリーをスクリーニングするために、及び構造的に関連する免疫交差反応性タンパク質をコードする、cDNA挿入体を含むクローンを同定するために有用である。Aruffo & Seed, Proc. Natl. Acad. Sci. USA 84 : 8573-8577 (1987) (引用により本明細書に組込まれる)を参照のこと。
ACT−4及び ACT−4−Lポリペプチド、そのフラグメント、類似体、抗体及びそれに対する抗−イディオタイプ抗体、並びに他の化学的又は生物学的物質は、そのリガンドに対する ACT−4の結合を阻止し又は増強するそれらの能力についてスクリーンされる。さらに、それらは代謝工程、たとえば ACT−4受容体又はそれらの表面に固定されている ACT−4−Lリガンドポリペプチドのいづれかを担持する細胞におけるDNA 合成又はタンパク質リン酸化を刺激し又は阻害するそれらの能力について試験される。
A.診断方法
ACT−4受容体又はそのmRNA、又は ACT−4−Lリガンド又はそのmRNAの変更された発生量又は機能的な突然変異に関連する免疫システムの疾患及び状態は、本発明のプローブ及び/又は抗体を用いて診断され得る。 ACT−4受容体又はmRNAの検出は、活性化されたCD4+ T−細胞の他の白血球サブタイプの区別を可能にする。たとえば、 ACT−4受容体は、 ACT−4受容体に特異的に結合する抗体又は ACT−4−Lポリペプチドを用いて検出され得る。
本発明のもう1つの観点においては、前記診断方法のための診断用キットが提供される。キットは、診断用試薬、たとえば ACT−4受容体及び ACT−4−Lリガンドに対するラベルされた抗体、及びラベルを検出するための試薬及び/又は装置を包含する容器を含んで成る。そのようなキットに通常見出される他の構成成分もまた、診断試験を実施するための教示と一緒に含まれ得る。
予防又は治療処理のために使用される医薬組成物は、活性治療剤、たとえば ACT−4受容体、 ACT−4−Lリガンド、それらのフラグメント及びそれらに対する抗体及びイディオタイプ抗体、並びに種々の他の成分を含んで成る。好ましい形は、投与及び治療用途の意図されたモードに依存する。組成物はまた、所望する配合に依存して、動物又はヒト投与のための医薬組成物を配合するために通常使用されるビークルとして定義される、医薬的に許容できる非毒性キャリヤー又は希釈剤を含むことができる。希釈剤は、組成物の生物学的活性に影響を及ぼさないように選択される。そのような希釈剤の例は、蒸留水、生理学的食塩水、リンガー液、デキストロス溶液、及びハンクス液である。さらに、医薬組成物又は配合物はまた、他のキャリヤー、アジュバント、又は非毒性、非治療性、非免疫原性安定剤及び同様のものを含むことができる。
治療方法は、ヒト又は動物、特に脊椎哺乳類における種々の疾病の処理のために上記治療剤を使用する。治療剤は、 ACT−4受容体、その結合フラグメント、 ACT−4−Lリガンド、その結合フラグメント、抗− ACT−4受容体及び抗− ACT−4−Lリガンド抗体、及びそれらに対する抗−イディオタイプ抗体、それらの抗体の結合フラグメント、それらの抗体のヒト適合化されたバージョン、及び前述の他の剤を含む。いくつかの治療剤は、 ACT−4受容体とそのリガンドとの作用を阻止〔又はアンタゴニストすることによって機能する。
治療用途においては、医薬組成物(たとえば、 ACT−4−h−1又は ACT−4−L−h−1に対する抗体を含んで成る)は、病状の進行及びその合併症を治癒し、特に抑制し、又は検出できるように遅延するのに十分な量で、所望しない免疫応答(たとえば移植片拒絶)をすでに有する患者にインビボ又はエクソビボで投与される。これを達成するための適切な量は、“治療的に有効な用量”又は“効果的用量”として定義される。
上記医薬組成物は、免疫システムのいくつかの疾病及び病状を処理するために適切である。
a.移植片拒絶
最近、組織及び器官、たとえば皮膚、腎臓、肝臓、心臓、肺、膵臓及び骨髄を移植するための手術技法の効能に関して相当の改良がなされて来た。たぶん、主な目立った問題は、移植された同種移植片又は器官に対して受容体における免疫耐性を誘発するための満足する剤の欠乏である。
本発明の治療剤についての関連する用途は、“移植片−対−宿主”疾患(GVHD)に関する免疫応答の調節である。GVHDは、免疫学的コンピテント細胞が同種受容体に移入される場合に生じる潜在的に致命的な疾病である。この情況においては、ドナーの免疫適格細胞が受容体における組織を攻撃する。皮膚、腸上皮及び肝臓の組織が時々、標的であり、そしてGVHDの進行の間、破壊され得る。この疾病は、免疫組織がたとえば骨髄移植において移植される場合に特に重大な問題を提供するが、しかし低い重症度のGVHDかまた、他の場合、たとえば心臓及び肝臓移植において報告されている。本発明の治療剤は、ドナーの白血球(特に活性化されたCD4+ T−細胞及びB−細胞)の活性化を阻止し又は排除するために使用され、それにより、宿主における標的細胞を溶解するそれらの能力を阻害する。
免疫抑制が所望される追加の情況は、自己免疫疾患、たとえばインスリン依存性糖尿病、多発性硬化症、スティッフマン症候群、リウマチ様関節炎、重症筋無力症及びエリテマトーデスの処理に存在する。それらの疾病においては、身体がそれ自体の抗原の1つに対しての細胞性及び/又は体液性免疫応答を発生せしめ、その抗原の破壊を導びき、そして潜在的に能力の劣る及び/又は致命的な結果を導びく。活性化されたCD4+ T−細胞は、多くの自己免疫疾患、たとえば糖尿病において主要な役割を演ずると思われる。
炎症は、食作用性白血球、たとえば顆粒球及び単球の流体の蓄積及び移動による毛細拡張の結果を表わす。炎症は、種々の感染に対して宿主を防御することにおいて重要であるが、しかしまた、炎症性疾患、たとえば過敏性ショック、関節炎、痛風及び虚血性再灌流の所望しない結果をも有する。活性化されたT−細胞は炎症において重要な調節役割を有し、食作用白血球を活性化するインターフェロンγ及びコロニー刺激因子を放す。
本発明はまた、感染剤に起因する疾病及び病状を防止し又は処理することにワクチンの効能を増強するための方法も提供する。CD4+ T−細胞及び/又はB−細胞を活性化する能力を有する治療剤(たとえば ACT−4又は ACT−4−Lリガンドに対する一定のモノクローナル抗体)が、選択された抗原を含むワクチンの投与の直前、それと同時又はその直後に投与される。治療剤は、選択された抗原に対しての免疫応答を高めるように作用する。それらの方法は、免疫欠損性疾病を有する患者において特に好都合である。
ACT−4−h−L−1に対する抗体はまた、HTLV−I−感染の細胞を殺すためにも有用である。上記のように、そのような細胞は、 ACT−4−h−1と同一であるか又はほぼ同一であるgp 34 抗原を発現する。それらの方法は通常、HTLV−I−感染の個人に対してインビボ又はエクスビボで実施される。しかしながら、その方法はまた、HTLV−I−感染のHIV 細胞をインビトロで殺すためにも効果的である。たとえば、その方法は、分析下での組織サンプルとの接触による感染から病院の労働者を保護するために特に有用である。
HIV ウィルスは、CD4受容体に結合することによってヒトCD4+ T−細胞を感染することが知られている。しかしながら、存在する生産性感染のためには、CD4受容体はCD4+ T−細胞の受容体上に存在するもう1つの受容体と相互作用すべきである。活性化されたT−細胞の表面上での受容体としての ACT−4の同定は、 ACT−4及び/又はそのリガンド ACT−4−LがCD4と相互作用し、そしてT−細胞のHIV 感染に寄与していることを示唆する。 ACT−4又は ACT−4−Lに対して標的化される、治療的有効量の治療剤は、HIV 感染を阻止し、そしてそれにより、AIDSを処理することに効果的であり得る。それらの治療剤はまた、HIV 感染されたCD4+ T−細胞をインビボ又はインビトロで殺害するためにも効果的であり得る。インビトロ方法は、上記のように偶発的な感染から病院労働者を保護することに利用される。
ACT−4−L−L−1及びその特異的結合類似体及びパートナー( ACT−4を包含する)は炎症性腸疾患に関連して使用され得ることが見出された。
従って、本発明はインビボ及びインビトロ診断方法、及び処理方法を提供する。
例1: ACT−4−h−1に対するモノクローナル抗体
マウスを、 PHA−形質転換T−リンパ芽球により免疫化した。免疫化されたマウスからの脾臓細胞を、SP2/O骨髄腫細胞により融合し、そしてT−細胞クローンに対して特異的な抗体を分泌するハイブリドーマを選択した。ハイブリドーマを限界希釈法によりクローン化した。得られたハイブリドーマの1つにより生成された、L106と称するモノクローナル抗体を、さらなる特徴化のために選択した。L106抗体は、IgG1イソタイプを有することが見出された。HBL106と称する、前記抗体を生成するハイブリドーマは、ATCC HB11483として寄託された。
抗体L106を含むサンプルが、L106抗体に結合する組織及び細胞タイプを同定するために、Fourth International Workshop and Conference on Human Leucocyte Differentiation Amtigens (Vienna 1989)で一定の関係者に利用された。その研究会からのデータは、Leukocyte Typing IV (ed. W. Knapp, Oxford U. Press, 1989) (引用により本明細書に組込まれる)に示され、そして付随するコンピューターデータベースはWalter R. Gilks, MRC Biostatistics Unit, Cambridge University, Englandから入手できる。この文献は、L106抗体が約50KDa のポリペプチドを結合することを報告する。
CD4+ T−細胞を、種々の活性化刺激に応答しての ACT−4−h−1受容体の発現について試験した。CD4+ T−細胞を、固相免疫吸着により末梢血液単核細胞から精製した。5×104 個のCD4+ T−細胞を、10%ヒト血清により補充されたRPMI培地を含むマイクロタイターウェルにおいて活性剤と共に培養した。3種の異なった活性剤を使用した:(1)5×104 個の照射された(3000ラド)単球、(2)PHA(1μg/ml)及び(3)テタヌストキソイド(5μg/ml)。 3H−チミジンを、収穫の12〜16時間前、培養物に添加した。収穫の後、Engleman et al., J. Immunol. 127 : 2124-2129 (1981) により記載されるようにして、細胞を、種々のラベルされた抗体(L106、抗−CD4及び抗−CD8)と共にインキュベートすることにより細胞表面抗原の発現について試験した。
ACT−4−h−1は種々の活性化刺激に応答して発現されるCD4+ T−細胞活性化抗原であることが結論づけられる。
ACT−4−h−1受容体のためのcDNAクローンを、Aruffo & Seed(前記)により最初に開発された、わずかに変性されたCOS 細胞発現システムを用いて単離した。RNA を、72時間PHA 活性化されたヒト末梢血液リンパ球から単離した。全RNA を TRI−試薬(Molecular Research Center)により抽出し、そしてポリ(A)+RNA をオリゴdT−磁気ビーズ精製(Promega)により単離した。cDNAを、スーパースクリプト逆転写酵素(Gibco/BRL)及びオリゴdTプライマーを用いて、Gubler & Hoffman, Gene 25 : 263-369 (1982)の方法により合成した。ブラント末端化されたcDNAを、非−自己−相補的BstX1アダプターに連結し、そしてSephacryl S−400 回転カラム上に通し、連結されていないアダプター及び小さなフラグメント(300塩基対以下)を除去した。
E1−27と称するクローンからの挿入体をpBluescript 中にサブクローンし、そしてALF スクエンサー(Pharmacia)に基づくT7ポリメラーゼ自動読取り配列を決定キット(Pharmacia)を用いて、ジデオキシ鎖終結方法により配列決定した。制限地図は、サブクローニングのためのいくつかの便利な部位を示した。5つのサブクローンをpBluescript において生成し、そしてM13前進性及び普遍的プライマーにより両鎖を配列決定した。
XbaI−HindIII フラグメントを、例4に記載の構造体から切出し、そして XbaI/HindIII −消化された pcDNA−I−neo (Invitrogen)中に挿入し、 ACT−4−h−1−neo と称する発現ベクターを生成した(図6)。このベクターをSf1により線状化し、3種の真核細胞系中にエレクトロポレートした。それらの細胞系は、SP2/O(Balb/O株に由来するマウス骨髄腫)、Jurkat(形質転換されたヒトT−細胞系)及び COS−7(付着性サル細胞系)であった。48時間の回収期間の後、形質転換された細胞を1mg/mlのG418(Gibco)において選択した。
同じ手段及び原理を用いて、 ACT−4−L−h−1を発現する安定した細胞系を得た(例10を参照のこと)。
ACT−4−h−1の細胞外ドメインがヒト免疫グロブリンの不変ドメインのN−末端にそのC−末端を通して連結されている安定した融合タンパク質を構成した。例4に記載される ACT−4−h−1をコードするベクターを SmaI及び NotIにより切断し、トランスメンブラン、原形質及び3’未翻訳領域を包含する、 SmaI部位の下流のすべての ACT−4−h−1配列を切出した。残る領域は、 ACT−4−h−1の可溶性細胞外部分をコードする(図8)。
ACT−4−h−1に対するリガンドを発現する細胞タイプを、プローブとして、例7に記載される ACT−4−h−1組換え免疫グロブリン融合タンパク質(この後、 ACT−4−h−1−Rgと称す)を用いて、流動細胞計測法と組合しての間接的染色法により同定した。結合された融合タンパク質を、フィコエリトリン接合の抗−ヒトIgを用いて検出した。それらの実験は、 ACT−4−h−1に対するリガンドが、Burkitt リンパ腫細胞系(Jiyoye) 、及び EBV−形質転換されたLCL 9037, 9059及びMSABを包含する、少数のB−細胞系上に低レベルで発現されたことを示した。それらの細胞系は3のMCF を伴って、5%の陽性を示した。表2を参照のこと。
リガンドのためのcDNAクローンを、Aruffo & Seed により最初に開発された、わずかに変性されたCOS 細胞発現システムを用いて単離した。RNA を、72時間 PMA/イオノマイシン−活性化されたヒト EBY−形質転換B細胞(細胞系MSAB)から単離した。全RNA を TRI−試薬(Molecular Research Center)により抽出し、そしてポリ(A)+RNA をオリゴdT−磁気ビーズ精製(Promega)により単離した。cDNAを、スーパースクリプト逆転写酵素(Gibco/BRL)及びオリゴdTプライマーを用いて、Gubler & Hoffman, Gene 25 : 263-369 (1982)の方法により合成した。
例9におけるクローン26からの挿入体を、HindIII 及び XbaIクローニング部位でpBluescript 中にサブクローン化した。クローンを、ALF スクエンサー(Pharmacia)に基づくT7ポリメラーゼ基礎の自動読取り配列決定キット(Pharmacia)を用いて、ジデオキシ鎖終結方法により配列決定した。3種のサブクローンをpBluescript において生成し、そしてM13前進性及び普遍的プライマーにより両鎖を配列決定した。クローン26のcDNA及び予測されるアミノ酸配列は図10−1〜図10−2に示されている。その予測されるアミノ酸配列により形成されるポリペプチドを、 ACT−4−L−h−1と称する。

- 請求の範囲第42〜46のいづれか1項記載の医薬組成物の有効量を投与することを含んで成る、患者における免疫応答を改良するための方法。
- 前記投与段階がエクスビボで実施される請求の範囲第47項記載の方法。
- 前記患者の免疫応答が抑制される請求の範囲第47又は48項記載の方法。
- 移植片拒絶、GVHD、自己免疫疾患、炎症、感染剤、HTVL感染された細胞又はHIV の処理に使用するための請求の範囲第47〜49のいづれか1項記載の方法。
- 炎症性腸疾患又は障害の処理に使用するための請求の範囲第47〜49のいづれか1項記載の方法。
- 免疫応答の改良に使用するための請求の範囲第1〜21のいづれか1項記載の特異的結合パートナー又は請求の範囲第22〜32のいづれか1項記載の結合成分。
- ACT−4を認識できる免疫調節剤のためのスクリーニング方法であって、
ACT−4受容体ポリペプチドのための特異的結合パートナーと試験されるべき材料とを接触せしめ;そして
前記特異的結合パートナーと試験下の材料との間の結合を検出することを含んで成る方法。 - その特異的結合パートナーにより ACT−4受容体ポリペプチドの認識に影響を及ぼすことができる免疫調節剤のためのスクリーニング方法であって、(a) ACT−4受容体ポリペプチドのための特異的結合パートナー、(b) ACT−4受容体又はその特異的結合類似体及び(c)試験下の剤を一緒に接触せしめ;そして成分(a)と(b)との間の結合を検出することを含んで成る方法。

前記選択された抗原に患者を暴露することを含んで成る方法。 - 活性化されたCD4+ T−細胞をモニターするための方法であって、
ACT−4受容体の細胞外ドメインに特異的に結合する ACT−4受容体ポリペプチドのための特異的結合パートナーと細胞とを接触せしめ;そして
前記活性化されたCD4+ T−細胞の存在を示すために前記特異的結合パートナーと前記細胞との間の特異的結合を検出することを含んで成る方法。 - 前記細胞が、患者からの組織サンプルを含んで成り、そして前記方法がインビトロで実施される請求の範囲第57項記載の方法。

- 請求の範囲第39項記載の宿主生物を培養することを含んで成る、請求の範囲第1〜21のいづれか1項記載の ACT−4受容体のための特異的結合パートナー又は請求の範囲第22〜32のいづれか1項記載の結合成分を生成するための方法。
- ACT−4受容体又はその特異的結合類似体の認識のために特異的結合反応への、図10−1〜図10−2に示されるようなアミノ酸配列を有するポリペプチド又は請求の範囲第1項記載の特異的結合パートナーの使用。
- 図10−1〜図10−2に示されるアミノ酸配列を有するポリペプチド又は請求の範囲第1項記載の特異的結合パートナーの認識のために特異的結合反応への ACT−4受容体又はその特異的結合類似体の使用。
- ACT−4と特異的に結合できる特異的結合剤である可溶性ポリペプチド。
- 図10−1〜図10−2のアミノ酸配列を有するが、但しそのトランスメンブランサブ配列を除く請求の範囲第66項記載のポリペプチド。
- トランスメンブラン配列によりリポソームの調製物の表面上に担持される、請求の範囲第1項記載の特異的結合パートナー又は図10−1〜図10−2に示される完全なアミノ酸配列を有する特異的結合パートナーポリペプチドを含んで成る組成物。
