本出願は、各々を全ての目的のために参照によりその全体を本明細書に組み込む、先の仮特許出願である2006年7月19日に出願されたUSSN 60/832,213(Christopher A.Hunter「WSX−1/P28 AS A TARGET FOR ANTI−INFLAMMATORY RESPONSES」)、および2006年8月11日に出願されたUSSN 60/837,450(Christopher A.Hunter「WSX−1/P28 AS A TARGET FOR ANTI−INFLAMMATORY RESPONSES」)の優先権および恩典を主張する非仮特許出願である。

T細胞が病気の臨界的メディエーターである多数の炎症性疾患があり、多くの努力がT細胞応答を特異的に阻害する戦略の開発に焦点を当ててきた。例えば、炎症性腸疾患、クローン病、多発性硬化症、ブドウ膜炎、乾癬、関節炎、喘息、狼瘡および移植片拒絶は、全てT細胞が関与する疾患である。免疫応答は、強直性脊椎炎(ankalyzing sponadalitis)およびサルコイドーシスなどの種々の他の特発性疾患にも関連付けられてきた。これらの疾患の全てについて、新しい治療手段を開発する差し迫った要求がある。WSX−1がT細胞応答の阻害に重要であるという認識は、この受容体がこれらのタイプの免疫応答を妨げるための実行可能な標的を表すことを意味する。あるいは、この受容体の遮断は、例えば、ワクチン接種または癌のための免疫介在療法の間に、T細胞応答を増大させるのに用いることができる。加えて、WSX−1を発現するある種のタイプの腫瘍も、この受容体を介しての阻害性シグナル伝達に感受性であり得る。
p28は、単独またはWSX−1の可溶性形態と組み合わせてアレルギー、関節炎、炎症性腸疾患、ブドウ膜炎、ある種の癌、乾癬、狼瘡、多発性硬化症、および結核および肝炎などの慢性感染性疾患を含む多くの炎症性疾患を抑制するのに用いることができるが、これらの疾患に限定されない。同様の手法は、これらの観察結果からの有用な治療手段であると意味する。従って、内因性IL−27と相互作用し、およびgp130とのその相互作用を促進し、標的に対する負の効果を促進する、(例えば、免疫グロブリンとの)WSX−1融合タンパク質が有用である。治療分子は、二重の機能を有するように構築することもでき、例えば、そのような分子は、1つの鎖がCD4、CD8、CD11c等の、これら限定されない細胞特異的マーカーを認識することができ、他の鎖がWSX−1またはp28融合を含む抗体構造に基づくことができる。これは、非常に特異的な細胞型の標的化を可能とする。加えて、受容体生物学がトランスシグナリングのエレメントを包含するという認識は、IL−27またはp28(またはEBI3さえ)との複合体におけるgp130の可溶性形態は、WSX−1を通じての特異的シグナル伝達を促進するように作用することもできるという考えを導く。いずれかの特定のメカニズムに制限されることなく、本発明者らの現在のモデルにおいては、IL−27受容体の異なる受容体鎖は固有のシグナル伝達機能を有し、および別々のT細胞機能に影響し得る。この概念は、特定のサイトカインのT細胞産生に非常に特異的に影響する分子の設計を導く。本発明者らの仕事およびデータに専ら基づくと、本発明者らは、この手法を用いて、例えば、IL−2、IFN−γ、TNF−α、IL−6、IL−4、IL−13、IL−17およびIL−25、ならびにIL−10、IL−5、および/またはCD25を標的化することができることを示唆する。これらの全てはバイオテクノロジーにおける有効な薬物標的を表し、多くのT細胞介在炎症性疾患において重要である。図13のパネルA〜Cはいくつかの候補戦略を概説する。本明細書におけるより詳細な検討およびさらなる実施例には、本発明者らが別々の免疫機能を合理的に標的化するのを可能とするこの情報に基づいて考案することができる多くのさらなる手法がある。
本発明の1つの態様は、抗炎症活性を有する組成物を含む、新規ポリペプチドおよび複合体を含む組成物を提供する。
WSX−1、p28、EBI3、および/またはgp130の活性を調節する化合物は、例えば、炎症を治療し、またはそうでなければ炎症反応を調節するのに有用であり得る。従って、実施形態の1つの一般的なクラスは、可溶性WSX−1ポリペプチド、可溶性WSX−1/p28ポリペプチド複合体、可溶性WSX−1/IL−27ポリペプチド複合体、可溶性WSX−1/EBI3ポリペプチド複合体、可溶性gp130/p28ポリペプチド複合体、可溶性gp130/IL−27ポリペプチド複合体、または可溶性gp130/EBI3ポリペプチド複合体に結合もしくはその活性を調節する化合物を同定する方法を提供する。前記方法において、前記ポリペプチドまたは複合体を含む、生物学的または生化学試料を試験化合物と接触させる。前記ポリペプチドまたは複合体への試験化合物の結合、もしくは試験化合物による前記ポリペプチドまたは複合体の活性の調節は検出され、これにより前記ポリペプチドまたは複合体に結合もしくはその活性を調節する化合物を同定する。
可溶性WSX−1ポリペプチド、p28ポリペプチド、単離または組換え可溶性WSX−1/p28ポリペプチド複合体、単離または組換え可溶性WSX−1/EBI3ポリペプチド複合体、単離または組換え可溶性WSX−1/IL−27ポリペプチド複合体、単離または組換え可溶性gp130/IL−27複合体、単離または組換え可溶性gp130/p28ポリペプチド複合体、単離または組換え可溶性gp130/EBI3ポリペプチド複合体、またはその変異体に特異的に結合する抗体は、本発明の特徴である。このような抗体は、任意で、前記ポリペプチドまたはポリペプチド複合体の活性を、例えば、増強などの調節をする。1つの実施形態において、抗体はgp130/WSX−1/IL−27複合体に結合またはその活性を調節し、または細胞中で前記複合体の形成を調節する。例えば、抗体は前記複合体の半減期を増大させることができる。このような抗体は、ポリクローナル、モノクローナル、キメラ、ヒト化、単一鎖、Fab断片およびFab発現ライブラリーによって産生された断片を含むことができるが、これらに限定されない。このような抗体は、例えば、炎症性疾患の治療において用途を見出す。このような抗体を生じさせる方法は本明細書において記載される。
ポリクローナル抗体は、好ましくは、関連抗原およびアジュバントの多数回の皮下または腹腔内注射によって動物で産生させる。例えば、マレイミドベンゾイルスルホスクシンイミドエステル(システイン残基を通じての接合)、(リジン残基を介する)N−ヒドロキシスクシンイミド、グルタルアルデヒド、無水コハク酸、SOCl2、RおよびR’が異なるアルキル基であるR’N=C=NRなどの二機能性または誘導体化剤を用いて、例えばキーホールリンペットヘモシアニン、血清アルブミン、ウシチログロブリン、または大豆トリプシン阻害剤などの関連抗原を免疫化すべき種において免疫原性であるタンパク質へ接合(conjugate)接合させるのは有用であり得る。
モノクローナル抗体は、すなわち、前記集団を含む個々の抗体がモノクローナル抗体の産生の間に生起する可能な変異体を除いて同一であり、および/または同一エピトープに結合する、実質的に均質な抗体集団から得られ、そのような変異体は、一般的に少量存在する。従って、修飾語「モノクローナル」は、別々のまたはポリクローナル抗体の混合物ではない、抗体の特性を示す。
非ヒト抗体をヒト化するための方法は当分野で記載されてきた。好ましくは、ヒト化抗体は、非ヒトである原料からそれに導入された一つまたは複数のアミノ酸残基を有する。これらの非ヒトアミノ酸残基は、しばしば、「移入」残基を指し、これらは、一般的には、「移入」可変ドメインから取られる。ヒト化は、ヒト抗体の対応する配列にを超可変領域配列で置換することによって、Winterおよび共同研究者の方法(Jones et al.,Nature,321:522−525(1986);Riechmann et al.,Nature,332:323−327(1988);Verhoeyen et al.,Science,239:1534−1536(1988))の方法に従って本質的に行うことができる。従って、このような「ヒト化」抗体はキメラ抗体であり(米国特許第4,816,567号)、ここに、実質的に完全ヒト可変ドメイン未満が、非ヒト種由来の対応する配列に置換されている。実施においては、ヒト化抗体は、一般的にいくつかの超可変領域残基および、場合により、いくつかのFR残基がげっ歯類抗体の類似の部位の残基によって置換されるヒト抗体である。
ヒト化に対する代替法として、ヒト抗体を生じさせることができる。例えば、今や、免疫化に際して、内因性免疫グロブリン産生の非存在下でヒト抗体の十分なレパートリーを産生することができるトランスジェニック動物(例えば、マウス)を産生するのが可能である。例えば、キメラおよび生殖系突然変異体マウスにおける抗体重鎖接合領域(JH)遺伝子のホモ接合性欠失の結果、内因性抗体産生の完全な阻害がもたらされると記載されている。そのような生殖系突然変異体マウスへの、ヒト生殖系免疫グロブリン遺伝子配列(array)の移入の結果、抗原誘発に際してヒト抗体の産生がもたらされる。例えば、Jakobovits et al.,Proc.Natl.Acad.Sci.USA,90:2551(1993);Jakobovits et al.,Nature,362:255−258(1993);Bruggermann et al.,Year in Immuno.,7:33(1993);および米国特許第5,591,669号、第5,589,369号および第5,545,807号を参照されたい。
抗体断片の産生のために、種々の技術が開発されている。伝統的には、これらの断片は完全抗体のタンパク質消化を介して誘導された(例えば、Morimoto et al.,Journal of Biochemical and Biophysical Methods 24:107−117(1992)and Brennan et al.,Science,229:81(1985)を参照されたい)。しかしながら、これらの断片は、今や、組換え宿主細胞によって直接的に産生することができる。例えば、抗体断片は、先に議論した抗体ファージライブラリーから単離することができる。または、Fab’−SH断片は大腸菌(E.coli)から直接的に回収することができ、化学的にカップリングさせて、F(ab’)2断片を形成することができる(Carter et al.,Bio/Technology 10:163−167(1992))。もう1つの手法では、F(ab’)2断片を組換え宿主細胞培養から直接的に単離することができる。抗体断片の産生のための他の技術は当業者に明らかである。他の実施形態において、選択した抗体は単一鎖Fv断片(scFv)である。WO 1993/16185および米国特許第5,571,894号および第5,587,458号を参照されたい。抗体断片は、例えば、米国特許第5,641,870号に記載された「直鎖状抗体」であってもよい。そのような直鎖状抗体断片は単一特異性または二重特異性であってよい。
二重特異性抗体は、少なくとも2つの異なるエピトープについて結合特異性を有する抗体である。例示的な二重特異性抗体はWSX−1、p28、gp130、またはEBI3抗原の2つの異なるエピトープに結合することができる。他のそのような抗体はWSX−1、p28、gp130、またはEBI3の1つに結合することができ、さらに、WSX−1、p28、gp130、またはEBI3のもう1つ、もしくはT細胞表面マーカーに結合することができる。二重特異性抗体を用いて、薬物または細胞傷害薬物を、抗原を含む細胞に局所化してもよい。これらの抗体はWSX−1、p28、gp130、またはEBI3結合アーム、および薬物または細胞傷害薬物に結合するアームを保有する。二重特異性抗体は、全長抗体または抗体断片(例えば、F(ab’)2二重特異性抗体)として調製することができる。
本明細書において、前記方法で用いられ、または製品に含まれる抗体は、任意でWO 2004/032828および米国特許出願公開2006/0024295に記載されているように、薬物に接合される。本発明の抗体は、プロドラッグ(例えば、ペプチジル化学療法剤、WO 1981/01145を参照)を活性な抗癌剤または他の薬物に変換するプロドラッグ活性化酵素と接合させてもよい。例えば、WO 1988/07378、米国特許第4,975,278号、および米国特許出願公開2006/0024295を参照されたい。

当業者に理解されるように、本発明の成分の適当な用量(例えば、ポリペプチド、複合体、抗体等)は、一般には、同様な成分が単独で、または他の治療剤と組み合わせて投与される臨床的療法で既に使用されたものの程度である。用量の変動は、治療すべき疾患に依存して起こる可能性が高い。治療を投与する医師は、個々の患者についての適当な用量を決定することができる。前記成分と組み合わせて投与される市販されている第二の化合物についての調製および投与スケジュールは、製造業者の指示に従って用いることができるか、または技量がある実施者によって経験的に決定できる。

現在開発中であるか、または市販されているいくつかのサイトカインまたはサイトカイン特定アンタゴニストがある。
これらの製品(特に、IL−10)の多くは、炎症時にT細胞応答を直接的に標的とできない。WSX−1はT細胞によって発現されるので、この受容体を標的とする戦略は、現在用いられているまたは開発されつつある他の手法のいくつかよりもかなり特異的な効果を有すると予測される。加えて、現在の標的の多くはT細胞活性の下流の単一分子であり、他方、p28/WSX−1(および他の融合タンパク質および本明細書において記載された複合体)はこれらの因子(例えば、IL−2、IFN−γ、IL−4、IL−17、TNF、IL−6)の多くを直接的に標的とし、加えてIL−10の発現を増大させることができることにより治療効果を増幅させる。加えて、副作用が多くのサイトカイン(IL−2、IL−12、IFN)で認められているが、本発明者らは、組換えIL−27で処理されたマウスにおいて、いかなる臨床疾患の明らかな兆候も観察されなかった。

遺伝暗号の縮重のため、「サイレント置換」(すなわち、コードされたポリペプチドの改変をもたらさない核酸配列における置換)は、アミノ酸配列をコードするあらゆる核酸配列の言外の特徴である。同様に、アミノ酸配列中の1または限定数のアミノ酸が、高度に同様な特性を備えた、異なるアミノ酸で置換される「保存的アミノ酸置換」もまた、開示された構築体と高度に同様であるとして容易に同定される。各開示された配列のこのような保存的変異は、本発明の特徴である。
比較ハイブリダイゼーションを用いて、本発明の核酸の保存的変異を含む本発明の核酸を同定することができる。加えて、高、超高および超−超高ストリンジェント条件下で本発明の核酸にハイブリダイズする標的核酸は、前記核酸が天然型核酸以外である場合、本発明の特徴である。このような核酸の例は、本発明の所定の核酸配列と比較して、1または少数のサイレントまたは保存的核酸置換を備えるものを含む。
2以上の核酸またはポリペプチド配列との関係で用語「同一」または「同一性割合」とは、以下に記載する配列比較アルゴリズムの1つ(または当業者に入手できる他のアルゴリズム)を用いて、または目視検査によって測定して最大対応性について比較および整列させた場合に、同一であるかまたは同一であるアミノ酸残基またはヌクレオチドの特定の割合を有する2以上の配列またはサブ配列を指す。
一般に、本発明の、または本発明の方法または組成物で用いるポリペプチドをコードする核酸は、クローニング、組換え、インビトロ合成、インビトロ増幅および/または他の入手可能な方法によって作成することができる。本質的にいずれの核酸も、特注品、またはOperon Technologies Inc.(Alameda,CA)などの、種々の商業的供給源のいずれかから注文された標準品であり得る。加えて、種々の組換え方法を、本発明のポリペプチドをコードする発現ベクターを発現するのに用いることができる。核酸を作成するための組換え方法、発現された産物の発現および単離は周知であり、例えば、Sambrook,Ausubel,and Innis et al.(eds.)、PCR Protocols:A Guide to Methods and Applications,Academic Press Inc.,San Diego,CA(1990)に記載されている。
最近の研究は、実験的自己免疫脳炎などの自己免疫性に関連するTH−17細胞の発生に影響する事象に焦点を当ててきたが、TH−17エフェクター応答に拮抗するサイトカインについては比較的ほとんど知られていない。本明細書における実験は、トキソプラズマ原虫に慢性的に感染したインターロイキン(IL)−27R−欠損マウスが、CD4+T細胞依存性および顕著なIL−17応答に関連する、重度の神経炎症を発生したことを示す。インビトロでは、ナイーブ一次T細胞のIL−27処理は、STAT1に依存するが、IL−6シグナル伝達のSOCS3介在阻害から独立した、IL−6およびTGF−βによって誘導されたTH−17細胞の発生を抑制した。従って、TH−17細胞発生の強力な阻害剤であるIL−27は、これらの細胞によって介在される炎症病を治療するための有用な標的である。同様に、前記の融合タンパク質および複合体は、これらの細胞によって介在される炎症病を治療するための有用な手法を提供する。
トキソプラズマ症脳炎時のIL−27の産生
リアルタイムPCRを用いて、TE時のIL−27についての転写産物の相対的レベルを定量した。少量〜最小量のebi3(EBI3に対する遺伝子)およびIl27(p28に対する遺伝子)のmRNAが未感染脳において検出された。しかしながら、慢性的に感染したマウスからの組織においては、脳におけるebi3 mRNAの量の3倍より大きい増加、およびIl27についてのmRNA量の500倍より大きい増加があった(図1のパネルA)。樹状細胞およびマクロファージが末梢組織におけるIL−27の産生源であると考えられると仮定すれば14、TE時に検出された上昇したebi3およびIl27転写体は、これらの細胞のCNSへの移動による可能性がある。または、IL−27を産生することもできる常在脳細胞は、小膠細胞および脳常在単球を含む29。TEの間における、CNS中での常在細胞のサブセットである星状膠細胞の活性化、および感染応答においてサイトカインを産生する星状膠細胞の能力30は、星状膠細胞がIL−27の産生源であることを意味することを示唆する。しかしながら、野生型マウスからの一次星状膠細胞は基底レベルのebi3 mRNAを発現したが、LPS+IFN−γでの刺激応答において増加は観察されなかった(図1のパネルB)。対照的に、Il27 mRNAの量はLPSおよびIFN−γでの刺激後にほぼ2000倍に増加した。これらの観察結果は、同時に、IL−27の両成分がTEの間に局所的に産生され、および活性化された星状膠細胞がEBI3およびp28の両方を発現できることを示唆する。
IL−27に対する従前の焦点は、ナイーブT細胞のエフェクターTH1細胞への分化におけるそれに対する役割に関連していた14、31−34。しかしながら、Il27ra−/−マウスは、トキソプラズマ原虫で攻撃した場合には、頑強なTH1応答を生じることが判明したが、マウスは、重度の肝臓病変および肺病理26に伴うIFN−γおよびIL−226、27の過剰な産生によって特徴付けられる、急性致死CD4+T細胞依存性炎症病で死亡した。
IL−27Rの非存在下における顕著な感染誘導CNS炎症を仮定して、実験を行って浸潤性細胞の表現型を同定した。組織病理学に従い、慢性的に感染したマウスから単離したBMNCの分析は、Il27ra−/−脳から回収された細胞の数の顕著な増加(P≦0.05;図3のパネルA)、およびCD4+T細胞の数およびパーセンテージの有意な増加(P≦0.05)、ならびに回収されたCD8+T細胞の数を示した(図3のパネルB)。T細胞集団の組成の差に拘らず、野生型およびIl27ra−/−マウス由来の脳のT細胞分析は、CD44hiおよびCD62Llowの活性化された表現型を示した(図8)。さらに、双方の組のマウス由来の単球は、それらの表面での主要組織適合性複合体クラスIIの発現の増大によって特徴付けられる、活性化された表現型を示した(データは示さず)。しかしながら、常在小膠細胞(CD11bint、CD45int)の数の差はなかったが、IL−27Rの非存在下において浸潤性マクロファージ(CD11bhi、CD45hi)の数の有意な増加があった(P≦0.05;図3のパネルB)。
慢性的に感染したIl27ra−/−マウスにおける神経病態は欠陥があるTH1応答の結果ではないが、代わりに、CD4+T細胞によって介在されるという認識は、致死的病態におけるCD4+T細胞の最近記載されたTH−17サブセットの可能性を調べる判断に導いた。TH−17細胞は、CNS炎症のモデルにおける病気の発生に関与しているIL−17、IL−6およびTNFの産生によって特徴付けられるので3、これらのサイトカインについてのmRNA転写体の量は、慢性的に感染した野生型およびIl27ra−/−マウスの脳に由来するRNAを用いてリアルタイムPCRによって評価された。野生型およびIl27ra−/−双方のマウスは同量程度ののIl6(IL−6)およびTnf(TNF)を発現したが、Il17(IL−17)の転写産物はIl27ra−/−マウス由来の試料のみで検出された(図4のパネルA)。
インビトロでのTH−17細胞の分化に対する最近の報告は、ナイーブCD4+T細胞からのこれらの細胞の発生についてTGF−βおよびIL−6が必要であり、他方、IFN−γおよびIL−4の遮断はTH−17発生について好都合な環境を支持すると結論した3、11−13、40、41。従って、C57BL/6マウスの脾臓から単離されたナイーブCD4+およびCD8+T細胞をこれらの条件下で培養して、IL−17のT細胞産生を阻害するIL−27の能力を直接的に評価した。PMAおよびイオノマイシンでの刺激に続き、ほとんどすべてのT細胞はTNFを産生し、およびかなりの集団がIL−17およびTNFを同時発現した。先に示した研究と同様に、IL−27の添加はCD4+およびCD8+T細胞によるIL−17産生を効果的に阻害したが、これらの細胞によるTNFの産生を変化させなかった(図5のパネルAおよびB)。PMAおよびイオノマイシン刺激の非存在下では、IL−17産生細胞の割合および平均蛍光強度はより低かったが、IL−27は、依然としてこの活性の強力なアンタゴニストであった(図9)。
多数のグループによる最近の研究は、TH−17細胞の発生におけるIL−6についての役割を明らかにした11−13。IL−6およびIL−27は共有される受容体成分gp130を通じてシグナル伝達を行うため、IL−17の産生を阻害するIL−27の能力は、gp130結合および/またはシグナル伝達に対するIL−6との競合の結果であった可能性があった。この論点に取り組むために、野生型 C57BL/6マウスの脾臓から単離されたCD4+Tを用いて一連の実験を行った。これらの細胞を非分極条件下でIL−23またはTGF−βの存在下で抗CD3+抗CD28抗体で活性化した場合(抗IFN−γ、抗IL−4)、IL−17の顕著な分泌が起こり、およびIL−27の添加は用量依存的にIL−17の産生を阻害できた(図6のパネルAおよびB)。(抗IL−6抗体を用いる)培養条件におけるIL−6の続く中和の結果、従前の報告と同様にIL−17産生の減少がもたらされた11;しかしながら、これらの条件においてさえ、IL−27の添加は、依然として、用量依存的にIL−17のレベルを低下させ(図6のパネルAおよびB)、これは、密接に関連するサイトカインであるIL−27およびIL−6がTH−17細胞に対して対照的な効果を有することを示す。
IL−6およびIL−27は密接に関連するサイトカインであり、およびgp130介在シグナル伝達を共有するが、示されたデータは、これらがIL−17産生に対して反対の効果を有することを大いに示す。この結論は、IL−27由来の阻害シグナルがIL−27R特異的成分を介して介在され、およびIL−6がSTAT3を主に活性化するが、いくつかの研究はユニークなIL−27R鎖をSTAT1の活性化および転写因子T−betに続く誘導に繋げていることを意味する32、33。従って、IL−17産生を阻害するIL−27の能力がこれらの転写因子に関係したかを決定するために、Stat1−/−またはTbx21−/−(T−bet−欠損)マウス由来のCD4+T細胞を、IL−27の存在下または非存在下、TH−17誘導条件下で刺激した。これまでのように、IL−27添加の結果、野生型およびT−bet−欠乏CD4+T細胞によるIL−17産生の顕著な阻害がもたらされたが、効果はSTAT1の非存在下で折衷された(図7のパネルA)。これらのデータは、TH−17機能に拮抗するIL−27の能力における、STAT1についての主要な役割を同定する。
最近、IL−17、TNFおよびIL−6の産生に係るT細胞独自のサブセットが、多発性硬化症、炎症性腸疾患および関節リウマチのモデルにおいて観察された病態の発生に関連付けられている3−7。異常なTH−17応答が自己免疫性に関連付けられている一方で、これらは病原体クレブシエラおよびトキソプラズマの攻撃に対する急性耐性においても役割を有するが47−49、これらの状況においては、TH−17応答は自己免疫性に導かない。これらの結果の意味は、ほとんどのT細胞応答について調べているのと同様に、TH−17活性を適切に調節するためのメカニズムが存在することであり、今日、IFN−γおよびIL−4が、TH−17細胞に拮抗する他のTヘルパー細胞に必要であるという明らかな証拠がある3,13,40,41。
マウスおよび寄生虫
C57BL/6マウスはJackson laboratoriesから入手し、WSX−1−/−(Il27ra−/−)マウスはChristiaan Saris博士(Amgen Inc.)より提供された。I−Adと関連するニワトリオボアルブミンペプチド(OVA(323−339))に対してTCR特異的なトランスジェニックDO11.10マウス、Stat1−/−マウスおよびTbx21−/−マウスはPhillip Scott博士(University of Pennsylvania,Philadelphia,PA)より提供された。gp130Y757FおよびCreMMTVSocs3fl/flマウスは従前に記載されている43、45。マウスを、制度的ガイドラインに従って、ペンシルバニア大学の病理生理学部門において特異的病原体が無い施設に収容し、育種した。
慢性的に感染した野生型およびIl27ra−/−マウスからのBMNCの単離を、従前に記載されたプロトコルに従って行った30、60。細胞を、従前に記載されているようにエクスビボ表面染色および細胞内染色のために処理した27。細胞を、CD4、CD8、CD44、CD45、I−A/I−E(BD Pharmingen)、CD62LおよびCD11b(eBioscience)に対する抗体を用いて、表面に染色した。IL−17(BD Pharmingen)、IFN−γおよびTNF(eBioscience)に対する抗体を用いて、T細胞を細胞内染色した。試料をFACScaliberフローサイトメーター(Becton Dickenson)で獲得し、FloJoソフトウエア(TreeStar Inc.)を用いて結果を分析した。BMNCを、96ウェル丸底プレート(Costar)中で200μlの最終容量中のウェル当たり2×105細胞の最終密度で平板培養した。組換えmIL−27(100ng/ml;Amgen Inc.)、IL−23(10ng/ml;DNAX)または双方の存在下または非存在下で、細胞をSTAg(50μg/ml)の有無にかかわらず刺激した。上清を48時間後に収集し、IL−2、IFN−γ、IL−12、IL−17、TNFおよびIL−6のレベルをELISAによって測定した。酸化窒素(NO)レベルをグレイス(Greiss)アッセイの使用によって測定した。
全細胞RNAを、標準的な手法を用いて慢性的に感染した野生型およびIl27ra−/−マウス、ならびに感染していない野生型マウスの、灌流後ホモジナイズした脳から単離した後、記載されているようにcDNAに変換した26。加えて、全RNAを野生型 C57BL/6一次星状膠細胞培養から単離し、これを用いてcDNAを作成した。一次星状膠細胞を、従前に記載されているように、1〜3日齢マウスの脳から収穫し30、グリア線維性酸性タンパク質(GFAP)染色(抗マウスGFAP、BD Pharmingen)によって判断した星状膠細胞培養の純度は一貫して90%を超えていた。Qiagenから入手したプライマーを用いて、TNF、IL−6、IL−27p28およびEBI3の発現を決定し、Power Sybr green(登録商標)試薬(Applied Biosystems)を用いてAB7500速リアルタイムPCRマシーンで行った。Taqman(登録商標)プライマー、プローブ、およびApplied Biosystemsから入手した試薬を用いて、IL−17の発現を決定した。β−アクチンハウスキーピング遺伝子を双方の場合における規準化対照として用いた。
IL−17−産生CD4+およびCD8+T細胞を、他の文献等に記載されている11、40、方法を改変して産生させた。簡単に述べると、前記したマウスから単離した脾細胞をCD8+およびNK1.1+細胞について枯渇させて、CD4+T細胞を豊富化させ、またはそれらをCD4+およびNK1.1+細胞について枯渇させて、磁性ビーズ分離(Polysciences)によりCD8+T細胞を豊富化させた。細胞を5×106細胞/mlの密度にて96ウェルプレート(Costar)中で平板培養した。Tg CD4+T細胞を5μg/mlのOVAペプチドで刺激し、他方、他のT細胞を抗TCR抗体(抗CD3;1μg/ml;eBioscience)および抗CD28(1μg/ml;eBioscience)で刺激した。TH−17細胞の産生のために、培養に、組換えマウスIL−23(10ng/ml;DNAX)またはヒトTGF−β(1ng/ml;R&D)単独またはIL−6(10ng/ml;eBioscience)、TNF(10ng/ml;eBioscience)およびIL−1β(10ng/ml;BD Pharmingen)との組み合わせを補充した。加えて、抗IFN−γ(10μg/ml;クローンXMG1.2)および抗IL−4(10μg/ml;クローン11B11)を用いて、IFN−γおよびIL−4を培養中で中和した。規定される場合には、組換えIL−27(100ng/ml;Amgen)を加えた。組換えp28はeBioscienceによって提供され、規定される場合、100ng/mlの濃度で用いた。CD8+T細胞を3日目に収穫し、他方、CD4+T細胞は3日目に新鮮な培地および試薬を補足し、4日目に収穫した。双方の細胞型を、続いてPMAおよびイオノマイシン刺激+ブレフェルジンA(Sigma)の存在下または非存在下にて、細胞内IL−17、TNFおよびIFN−γにつき染色した。
独立スチューデントt検定を用いて有意差を決定し、0.05未満のP値は有意と考えられた。
全脾細胞を2匹のIl27ra−/−マウス(WSX−1 KO1およびWSX−1 KO2)から単離した。脾細胞からNK1.1+およびCD8+細胞を枯渇させて、CD4+T細胞を豊富化した。次いで、CD4+T細胞をCFSE(カルボキシフルオレセインスクシニミジルエステル)で標識し、抗CD3抗体および抗CD28抗体で刺激した。sIL−27R Fcタンパク質(可溶性WSX−1ポリペプチド、Amgen Inc.によって提供された、WSX−1細胞外ドメイン配列を含むFc融合タンパク質)の有無に関わらず、IL−27を、細胞を含むウェルに加えた。sIL−27R Fcタンパク質を細胞ウェルへの添加に先立ってIL−27と共に30分間インキュベートして、結合を促進した。細胞を37℃で48時間インキュベートした。上清をELISAアッセイで用いて、IL−2およびIFN−γの産生を測定した(図12)。
IL−10は、保護的T細胞応答と病理学的T細胞応答の間のバランスを調節する際に主な役割を有する。この活性と合致して、ミエロイド細胞ならびに種々のT細胞サブセットを含むこのサイトカインの多数の産生源がある。しかしながら、IL−10の生来の産生を調節する多くの経路があるが、適応応答によるその産生を支配する因子はあまり理解されていない。本実施例において提示した実験は、IL−27およびIL−6が種々のT細胞集団を誘導してIL−10を産生させることができることを明らかにする。この効果は、IL−27については転写因子STAT1およびSTAT3、ならびにIL−6についてはSTAT3に依存する。同時に、これらの実験は、免疫系が炎症応答を和らげるのを可能とする新規な経路を同定する。
IL−27はIL−10のT細胞産生を誘導する。
最近の研究は多数の炎症促進性サイトカインのT細胞産生を阻害するIL−27の能力を強調してきたが38、43、IL−27に対するさらなる標的を同定することができるスクリーニングが目的であった。従って、C57BL/6マウスからのナイーブCD4+T細胞を、アクセサリー細胞が存在下するIL−27の存在下または非存在下、非分極条件下で(α−IFN−γおよびα−IL−4)、抗TCR(α−CD3)およびα−CD28抗体で活性化した。細胞を3日間培養した後、げっ歯類Multi−Analyte Profile(RodentMAP(商標))を用いて、上清を67の分泌免疫産物のパネルについてアッセイした。従前の報告と合致して、IL−27のこれらの非分極培養物への付加は、TH1(IFN−γ)、TH2(IL−5)およびTH17(IL−17)応答に関連する多数のサイトカインの減少に導くだけでなく、GM−CSF、IL−1β、IL−3、MIP−1αおよび−βおよびリンホタクチンも含んだ(図14のパネルA)。加えて、IL−18、IL−6、IL−7を含むいくつかの他のサイトカインおよびMCP−1、MCP−3、M−CSF、MMP−9を含むケモカインはこの処理によって変化しなかった(表2)。しかしながら、最も顕著な結果は、IL−27が、これらの培養上清におけるIL−10のレベルの1000倍増加に導いたという観察結果であった(図14のパネルAおよび表2)。
前記した実験は、IL−27がIL−10のT細胞産生を増強できることを示した。これらの事象におけるIL−27Rの役割を評価するために、野生型またはIl−27ra−/−マウス由来の脾細胞を、IL−27の存在下または非存在下、非分極条件下で活性化し、IL−10をアッセイした。野生型マウス由来の細胞はIL−27に応答した増強されたレベルのIL−10を分泌したが、これはIl−27ra−/−マウス由来の培養では観察されなかった(図15のパネルA)。事実、これらの上清における均一基礎レベルのIL−10は野生型対照と比較して低下した。IL−27/IL−27Rがインビボにて炎症応答の調節に関与したか否かを評価するために、野生型およびIl−27ra−/−マウスをトキソプラズマ原虫38で慢性的に感染した実験系を用いた。これらの実験において、脳単核細胞(BMNC)調製物および慢性的に感染した野生型マウス由来の脾細胞の再刺激は、IL−10を産生するCD4+T細胞の存在を直接的にエクスビボにて明らかにした。対照的に、慢性的に感染したIl−27ra−/−マウスの脳および脾臓由来の細胞を用いた場合、IL−10において顕著な欠陥があった(図15のパネルB)。逆に、IL−27の存在下における野生型BMNCのSTAgでの再刺激の結果、IL−10の有意な増加があった(図15のパネルC)。これらの結果は、集合的に、インビトロでのIL−10のT細胞産生の促進における、およびトキソプラズマ原虫に関連する慢性感染誘導炎症の状況における、IL−27およびIL−27Rに対する顕著な役割を示唆する。
前記した実験は中性条件下で行ったが、トキソプラズマ原虫で感染させたマウスからのデータは、TH1支配的応答の間におけるIL−10プロデューサーの発生におけるIL−27についての役割を示す。従って、さらなるインビトロ実験を行って、T細胞活性化後のいずれの時点において、IT−10が産生されたかを決定し、およびTH1、TH2またはTH17細胞の発生に好都合な条件下で、IL−10を促進するIL−27の能力を評価した。IL−27に応答した4日間にわたるCD4+T細胞によるIL−10産生の分析は、活性化から48時間後に細胞がIL−10の産生を開始し、72時間においてピークとなったIL−10+細胞の数は96時間に渡って維持されたことを明らかにした(図16のパネルA)。加えて、細胞をCFSE標識して、IL−27によって生じたIL−10産生T細胞が活発に増殖しているか、またはT細胞の非複製集団の一部であるかを決定した。CFSE希釈によって示されるように、CFSE反応がはっきりしない細胞のみがIL−27に応答してIL−10を産生した。この知見は、T細胞がサイトカイン産生を獲得するのに増殖が必要である際のモデルと合致する44。また、これらの実験におけるIL−10産生のパターンは、最近活性化されたT細胞についてのIL−27Rの発現プロフィールに相関する45。
IL−27はTH1およびTH2条件下でIL−10の産生を促進でき、TH17分極に続いてIL−10+CD4+T細胞のかなりの割合があったが、これらのIL−10+細胞もこれらのTHサブセットに関連するシグネチャーサイトカイン(sigunature cytokine)を産生するか否かは明らかでなかった。従って、CD4+T細胞をTH1、TH2およびTH17条件下で刺激し、IL−10についての細胞内染色を、IFN−γ、IL−13またはIL−17各々と組み合わせた。TH1条件下で刺激した場合、IL−10産生T細胞の大部分もまた、IFN−γについて陽性に染色されたが、二重プロデューサーのこの集団は、依然としてまさにIFN−γを産生する細胞と比較して少数であった(図17のパネルA)。IL−27の添加はIFN−γ+細胞の数を低下させず、その代わりIFN−γ+IL−10+CD4+T細胞の割合の増加をもたらした。TH2条件下では、IL−10+細胞のほぼ50%がやはりIL−13を産生していた(図17のパネルA)。IL−27の添加はIL−10+細胞の割合を増大させ、IL−13+IL−10+細胞およびIL−13単一産生株数の同時低下を引き起こした。
TH17細胞は、これらの条件下でIL−27はIL−10を増強することができないことと組み合わせて、有意なレベルのIL−10を産生したという知見は、TGF−βまたはIL−6もまたこれらの事象の調節に関与し得ることを示唆した。CD4+T細胞に対するTGF−βの効果の調査は、IL−27、TGF−β単独とは異なり、IL−10の中程度の増加をもたらしたが、IL−27と組み合わせた場合、IL−10+細胞の割合の増加ならびにMFIの増加に導く相加的効果を有したことを明らかとした(図18のパネルAおよびB)。加えて、外因性TGF−βはIL−10産生を増加させないが、内因性TGF−βの中和は、IL−10産生を促進するIL−27の能力を排除しなかったが、IL−10+細胞の割合を中程度の低下に導いた(データは示さず)。
TGF−βは、IL−10産生単独を刺激するIL−27の能力を増強させることができるが、TH17条件下で存在するIL−10+T細胞の高い割合を説明できない。従って、IL−27と構造的相同性および受容体サブユニットを共有するタイプIのサイトカインであるIL−6もまた、IL−10の産生を促進できるかを決定するために、CD4+T細胞を非分極条件下でIL−6と共にインキュベートした。これらの実験においてTGF−βで見られたように、IL−6の添加の結果、IL−10の中程度の増大のみがもたらされた(図19のパネルAおよびB)。なお、TGF−βと組み合わせた場合、IL−6は相乗的に、IL−10+CD4+T細胞の大きな集団の出現を促進した。IL−10を正に調節するIL−27の能力はTH1分極条件下で最も明らかであったので、TH1およびTH2条件下でのIL−10産生に対するIL−6の効果を調べた。IL−27とは異なり、IL−6は続いてのTH1分極下でIL−10を増強させることができなかった(図21)。対照的に、TH2分化条件下で、IL−6の添加は、IL−10の産生レベルに対して相加的効果を有した(図21)。
CD4+T細胞における特異的STATタンパク質の活性化は、T細胞の別々のTH細胞系への分化に関連した寄与因子の1つであり、IL−27は、STAT1(および結果として、T−bet)、STAT3および、より程度は低いが、STAT4を含む多数のSTATタンパク質を活性化することが示されており49、50、他方、IL−6は主としてSTAT3および、程度はより低いが、STAT1を活性化する51、52。IL−27およびIL−6がSTAT1およびSTAT3を活性化する動態学を決定するために、精製されたCD4+T細胞を3時間にわたって各サイトカインで刺激し、これらの転写因子のリン酸化をモニターした。これらの実験は、CD4+T細胞がIL−6およびIL−27に応答してSTAT1およびSTAT3をリン酸化できたが、IL−6はIL−27よりも早い速度でこれを行うことができたことをを明らかにした(図19のパネルCおよびD)。最高数のP−STAT1+細胞はIL−27での刺激後30分まで見られなかったが、IL−6とIL−27応答のピーク間に、P−STAT1+細胞の割合の差はなかった。他方、IL−6はSTAT3リン酸化のそのように強いインデューサーであったので、刺激の5分後には、T細胞のほぼ90%はP−STAT3+であり、高レベルのP−STAT3は3時間にわたって維持された。対照的に、わずかな割合のCD4+T細胞がIL−27に応答してP−STAT3につき陽性に染色され、P−STAT3+細胞のこの集団は刺激から3時間後に維持されていなかった。
TH2細胞に関連するサイトカインとしてのIL−10の元の記載以来、今日、免疫応答の多くのクラス(TH1、TH2、TH17)の全体的阻害剤として作用するIL−10の多くの先天的および適合する産生源があることが認識されている。それにも拘わらず、T細胞はIL−10の主な産生源であるという初期の認識に拘わらず、これらのリンパ球におけるその発現を支配する因子について多くの疑問が依然として存在する。Trinchieriおよび同僚による初期の研究は、IL−12をIFN−γ/IL−10ダブルプロデューサー23、26の発生の駆動に関連付けており、これはTH1条件下で、STAT4がこれらの事象に関与することを示すデータによって本明細書に反復された観察結果である。さらに、IL−10の存在下におけるヒトおよびマウスT細胞の慢性的刺激は、高レベルのIL−10を分泌し、結腸炎17を軽減し得るTヘルパー細胞(Tr1)の集団の出現を導いた。同様に、デキサメタゾン+ビタミンD3での、インビトロでのナイーブヒトおよびネズミCD4+T細胞の反復刺激は、Treg細胞の集団によってIL−10産生を促進することが示されている55。対照的に、本明細書において提示された研究は、短期間の刺激後においてさえ、IL−27およびIL−6が種々の分極条件下でIL−10のT細胞産生を誘導できたことを明らかとする。この観察結果は、IL−10の産生を促進し、およびIL−6/IL−12ファミリーメンバーの炎症促進性および抗炎症性特性間の複雑な関係を補強する新しい経路を同定する。
マウスおよび寄生虫
C57BL/6、Balb/c、Stat4−/−およびTbx21−/−マウスはJackson laboratoriesから入手した。WSX−1−/−(Il27ra−/−)マウスはChristiaan Saris博士(Amgen Inc.)より提供された。Stat1−/−マウスはPhillip Scott博士(University of Pennsylvania,Philadelphia,PA)より提供された。Foxp3についての翻訳の部位においてノックインされたGFPレポーターを備えるマウスが従前に記載されており72、Laurence Turka博士(University of Pennsylvania)より提供された。制度ガイダンスに従い、ペンシルベニア大学の病理生物学部門における特異的病原体が無い施設にマウスを収容し、育種した。
CD4+およびCD8+T細胞を、磁気ビーズ分離(Polysciences)によって、CD8+およびNK1.1+細胞を枯渇させてCD4+T細胞を豊富化した、またはCD4+およびNK1.1+細胞を枯渇させて、CD8+T細胞を豊富化させた脾細胞およびリンパ節から単離した。細胞を、5×106細胞/mlの密度で、96ウェル丸底プレート(Costar)中で平板培養した。細胞を抗TCR抗体(α−CD3;1μg/ml;eBioscience)および抗CD28抗体(1μg/ml;eBioscience)で刺激した。IL−10の産生のために、T細胞培養に、組換えマウスIL−27(100ng/ml;Amgen)、もしくはヒトTGF−β(1ng/ml;R&D)単独またはIL−27との組み合わせを補充した。加えて、抗IFN−γ(10μg/ml;クローンXMG1.2)および抗IL−4(10μg/ml;クローン11B11)を用いて、IFN−γおよびIL−4を非分極培養中で中和した。いくつかの場合においては、T細胞を、TH1(10ng/ml組換えIL−12;eBioscience+10μg/ml α−IL−4)、TH2(8ng/ml組換えIL−4;eBioscience+10μg/ml α−IFN−γ)またはTH17(1ng/ml TGF−β;R&D,10ng/ml IL−6;eBioscience,+10μg/ml α−IFN−γおよびα−IL−4)条件下で培養した。CD8+T細胞を3日目に収穫する一方で、3日目のCD4+T細胞に新鮮な培地および試薬を補充し、4日目に収穫した。次いで、T細胞をPMAおよびイオノマイシン+ブレフェルジンA(Sigma)で再刺激した。フローサイトメトリー分析は、FACSCaliber(BD Biosciences)またはBDFACS CantoII(BD Biosciences)機器で行い、FlowJoソフトウエア(Tree Star Inc.)を用いて分析した。全ての抗体は、BD PharmingenまたはeBioscienceから購入した。GFPの細胞内染色のために、細胞をまずマウス抗GFP抗体(eBioscience)で染色し、続いてウサギ抗マウス−FITC抗体(Jackson Immunoresearch)で第二の染色を行った。
CD4+単離キット(Milltenyi)を用いて、CD4+T細胞をC57BL/6マウスから精製した。1×106の精製されたCD4+T細胞をIL−6またはIL−27と共に5、30、60または180分間インキュベートした。次いで、細胞を、2%パラホルムアルデヒドで、37℃で10分間固定した。次いで、固定後、細胞に90%メタノールを氷上で30分間浸透させ、続いてCD4、P−STAT1およびP−STAT3について染色した。STAT1およびSTAT3のリン酸化されたチロシン残基に対する抗体は、BD Pharmingenから購入した。
独立スチューデントのt検定を用いて有意差を決定し記載し、P値<0.05は有意と考えられた。

- 哺乳動物患者において炎症性疾患を治療する方法であって、前記患者に、可溶性WSX−1ポリペプチド、p28ポリペプチド、可溶性WSX−1/p28ポリペプチド複合体、可溶性WSX−1/EBI3ポリペプチド複合体、可溶性WSX−1/IL−27ポリペプチド複合体、可溶性gp130/IL−27複合体、可溶性gp130/p28ポリペプチド複合体、可溶性gp130/EBI3ポリペプチド複合体、p28ポリペプチドおよび可溶性WSX−1ポリペプチド、EBI3ポリペプチドおよび可溶性WSX−1ポリペプチド、IL−27および可溶性WSX−1ポリペプチド、可溶性gp130ポリペプチドおよびp28ポリペプチド、可溶性gp130ポリペプチドおよびIL−27、可溶性gp130ポリペプチドおよびEBI3ポリペプチド、およびその変異体からなる群より選択される、単離されたまたは組換え部分を投与することを含む、方法。
- 前記患者がヒトである、請求項21に記載の方法。
- 前記炎症性疾患が、免疫異常、感染、癌、アレルギー、関節炎、喘息、炎症性腸疾患、クローン病、ブドウ膜炎、乾癬、狼瘡、多発性硬化症、慢性感染性疾患、結核、強直性脊椎炎、移植片拒絶、サルコイドーシスおよび肝炎から選択される、請求項21に記載の方法。
- 前記方法が前記投与に先立って炎症性疾患の前記患者を診断することを含む、請求項21に記載の方法。
- 前記単離されたまたは組換え部分が形質転換増殖因子βと組み合わせて前記患者に投与される、請求項21に記載の方法。
- 哺乳動物患者において炎症性疾患を治療する方法であって、前記患者にgp130/WSX−1/IL−27複合体に特異的に結合し、または前記複合体の活性を調節する、または細胞中の前記複合体の形成を調節する部分を投与し、それにより前記疾患について前記患者を治療することを含む方法。
- 前記部分が、抗体、アンタゴニスト、アゴニスト、および活性調節因子から選択される、請求項26に記載の方法。
- 前記炎症性疾患が、免疫異常、感染、癌、アレルギー、関節炎、喘息、炎症性腸疾患、クローン病、ブドウ膜炎、乾癬、狼瘡、多発性硬化症、慢性感染性疾患、結核、強直性脊椎炎、移植片拒絶、サルコイドーシスおよび肝炎から選択される、請求項26に記載の方法。
- 可溶性WSX−1ポリペプチド、可溶性WSX−1/p28ポリペプチド複合体、可溶性WSX−1/IL−27ポリペプチド複合体、可溶性WSX−1/EBI3ポリペプチド複合体、可溶性gp130/p28ポリペプチド複合体、可溶性gp130/IL−27ポリペプチド複合体、または可溶性gp130/EBI3ポリペプチド複合体に結合し、またはその活性を調節する化合物を同定する方法であって、
(a.)前記ポリペプチドまたは複合体を含む生物学的または生化学試料を試験化合物と接触させることと、
(b.)前記試験化合物の前記ポリペプチドまたは複合体への結合、または前記ポリペプチドまたは複合体の前記試験化合物による前記活性の調節を検出し、それにより前記ポリペプチドまたは複合体に結合し、またはその活性を調節する前記化合物を同定することと、
を含む方法。 - 前記化合物が前記ポリペプチドまたは複合体によるT細胞応答の阻害を増強し、IL−2またはIL−17に対するアンタゴニスト活性を増強し、またはT細胞の増殖、生存、もしくはIL−2、IFN−γ、TNF−α、IL−6、IL−4、IL−13、IL−17、IL−25、IL−10、IL−5、またはCD25の発現を改変する、請求項29に記載の方法。
