Abstract
Description
−2001年、Su−Chun Zhang等の研究チームは、初めてヒト胚性幹細胞から神経前駆細胞及び各種の神経細胞への分化を誘導して発表した。
−2004年、アメリカ・コロラド大学のKimberley等は、ヒト胚性幹細胞をPA6細胞と共培養した後、グリア細胞株由来神経栄養因子(GDNF)を添加することにより、ドーパミン神経細胞の収率を高めることができたとの研究結果を発表した。
−2004年、BresaGen Inc.のSchulz等の研究チームは、胚性幹細胞集合体を他の誘導因子無しに、無血清培地で自然に浮遊培養してドーパミンを分泌しながら電気生理学的作用を有する神経細胞に誘導し、移植後、ドーパミンを発現する細胞を確認した。
−2004年、Sloan−Kettering InstituteのPerrier等は、さらに他の間質細胞であるMS5細胞を用いて、ヒト胚性幹細胞から神経前駆細胞塊を形成した後、様々な成長因及び分化誘導因子を添加することにより、神経細胞の70%以上をドーパミン神経細胞に分化するのに成功した。
−韓国の場合、ヒト胚性幹細胞からドーパミン神経細胞を分化させて発表した研究チームは多くないが、2004年Maria Infertility HospitalのSepill Park等は、ヒト胚性幹細胞から様々な分化誘導因子を処理して神経細胞を分化させ、20%近くまでドーパミン神経細胞が分化されたと発表した。しかし、分化された細胞の機能性や他のマーカーの確認が足りなく、他の研究結果に比べて収率に劣るという問題があり、移植後の生存率等は報告されていない。
現在まで、韓国内外の研究チームの多くの研究にもかかわらず、実際に、全体細胞から純粋なドーパミン神経細胞が得られる収率は、未だ低いレベルにとどまっており、移植後の生存率、機能性に劣るという問題等を有している。そこで、本発明者等は、胚性幹細胞から80%以上の高収率で神経前駆細胞、神経細胞、及び純粋なドーパミン神経細胞が得られる方法を定立し、神経前駆細胞段階の細胞を、継代培養により多量の前駆細胞、神経細胞、及びドーパミン神経細胞として供給可能にして、本発明を完成した。
既存の分化方法が、効率面において実用化し難い問題点を有していたので、本発明では、実用化が可能に、分化効率性と機能性、及び量的確保のための実用性を画期的に高めようとした。
(a)最小5日から最大21日間培養された胚様体のうち、嚢胞性の胚様体を除いた残りを、マトリゲル(Matrigel(登録商標))、ラミニン、またはL−ポリオルニチンがコートされた培養皿に付着した後、最小5日から最大7日間、0.5 X N−2添加剤を添加した培地で培養し、神経系以外の細胞の成長を抑制させる段階と、
(b)N−2及びbFGFを添加した培地で、最小3日から最大7日間増殖された胚様体由来細胞群からニューロン特異的な構造物を切除及び分離する段階と、
(c)前記切除、分離されたニューロン特異的な構造物から作られた球状神経前駆細胞を培養する過程において発生する線維芽細胞様細胞と嚢胞構造を除去し、球状神経前駆細胞内で増殖されたニューロン特異的な構造物のみを分離する段階と、
(d)前記(c)の過程を最小4回以上繰り返し、球状神経前駆細胞の純度を高め、球状神経前駆細胞を増殖する段階とを含む。
本発明で用いられる用語の「胚性幹細胞」とは、受精卵の発達過程中、胚盤胞段階の内細胞塊から得られ、特定の分化環境が与えられない限り、未分化状態で分裂し続けることができ、条件に応じて、殆ど全ての組織の細胞に分化することができる特性を有する細胞のことをいう。
本発明で用いられた前記切除方法は、球状神経前駆細胞からニューロン特異的な構造物のみを分離するものであって、これは、公知の様々な方法により行われ得る。本発明の具体的な実施において、好ましい前記切除方法は、機械的な方法、特にガラスパスツールピペットを用いて機械的に分離する方法である。前記ガラスパスツールピペットを用いた機械的な方法は、パスツールピペットの細い部分を加熱して引っ張ることにより、一端を髪の毛程度の厚さに作り、これを用いて顕微鏡下で所望の部位を切り出す方法である。
他の具体的な態様として、本発明は、上述した方法で作られた神経前駆細胞を分離し、1乃至2日間N−2及びbFGFを添加した培地で培養し、これを3乃至7日間N−2及びB27を添加した培地でさらに培養することにより、神経細胞への分化を誘導する方法に関する。
前記特定の培地は、ヒト胚性幹細胞培養用培地を用いることができ、好ましくは20%のノックアウト血清代替物(K/SR)、2mM L−グルタミン、1% 非必須アミノ酸、0.5% ペニシリン−ストレプトマイシン(P/S)または0.1mM β−メルカプトエタノールが含まれたDMEM/F12倍地を用いる。
前記方法により効率的に前記神経細胞からドーパミン神経細胞への分化を誘導することができ、前記培養培地は、ヒト胚性幹細胞培養用培地を用いることができ、好ましくは、20%のノックアウト血清代替物(K/SR)、2mM L−グルタミン、1% 非必須アミノ酸、0.5% ペニシリン−ストレプトマイシン(P/S)または0.1mM β−メルカプトエタノールが含まれたDMEM/F12を好適に用いることができる。
本発明者等は、本発明の方法でヒト胚性幹細胞を神経前駆細胞及び神経細胞に分化誘導する場合、神経前駆細胞特異的マーカーであるnestinの発現、ニューロン特異的なマーカーであるβIII−チューブリンとNeuNの同時発現、βIII−チューブリンとA2B5の同時発現、及びβIII−チューブリンとnestinの同時発現、ニューロン特異的な細胞膜の活動電位(−60mV〜20mV)の確認、βIII−チューブリンとシナプトフィジン(SYP)の同時発現、及び神経細胞培養液からドーパミンの分泌を確認することができる。
前記タンパク質に対する抗体または前記タンパク質をコードする遺伝子のmRNAレベルを測定するRT−PCR等の当業者によく知られた方法を用いて、本発明により分化誘導された神経前駆細胞または神経細胞の特性を確認することができる。特定細胞中に存在する前記特性が多いほど、神経前駆細胞または神経細胞としてさらに特徴付けられ得る。前記特性の3以上、好ましくは4以上、より好ましくは5以上を有する細胞及び/又はその子孫が好ましい。本発明の方法で分化誘導された細胞の約40%、60%、80%、90%、95%または98%以上が、目的とする特性を有する細胞であることが好ましく、これらの数値が大きいほどさらに好ましい。
他の態様として、本発明は、本発明の方法で分化誘導され、上記で提供される、ヒト胚性幹細胞から分化誘導された細胞及び/又はその子孫、具体的には、神経前駆細胞、神経細胞及び/又は機能性ドーパミン神経細胞を含む脳または神経系疾患に対する治療用組成物に関する。
前記脳または神経系疾患は、パーキンソン病、神経痛、関節炎、頭痛、統合失調症、てんかん、脳卒中、不眠症、認知症、うつ病、ジスキネジア、アルツハイマー病、レヴィー小体認知症、ハンチントン病、トゥレット・シンドローム、不安、学習及び記憶障害、神経変性病等の神経系の機能減少または異常により、脳または神経系に現れる疾患をいう。本発明の具体的な実施において、本発明の前記組成物が神経系疾患中の一つであるパーキンソン病の治療に有用に適用され得ることを確認した。
また、本発明の治療用組成物は、一般的な医薬品製剤の形態で使用され得る。非経口製剤としては、滅菌した水溶液、非水溶液、懸濁剤、乳剤、または凍結乾燥剤、経口投与の際は、錠剤、トローチ、カプセル、エリキシル、サスペンション、シロップ、またはウエハー等の形態で製造することができ、注射剤の場合は、単回投与アンプルまたは複数回投与の形態で製造することができる。また、本発明の治療用組成物は、薬学的に許容される担体と一緒に投与されてもよい。例えば、経口投与の際は、結合剤、滑剤、崩壊剤、賦形剤、可溶化剤、分散剤、安定化剤、懸濁化剤、色素剤、または香料等を用いることができ、注射剤の場合は、緩衝剤、保存剤、鎮痛剤、可溶化剤、等張剤、安定化剤等を混合して用いることができ、局所性投与用の場合は、基剤、賦形剤、滑剤、保存剤等を用いることができる。
以下、実施例を通じて本発明をより詳述する。これらの実施例は、単に本発明を例示するためのものであり、本発明の範囲がこれらの実施例により制限されるものではない。
未分化ヒト胚性幹細胞株(SNUhES1、SNUhES3、SNUhES16)の継代及び維持のために、未分化胚性幹細胞をマイトマイシン−Cが処理された支持細胞(マウス胎児線維芽細胞株、STO)上で培養し、培養液は、20% ノックアウト血清代替物(SR)、2mM L−グルタミン、0.4ng/ml 塩基性線維芽細胞増殖因子(bFGF)、1% 非必須アミノ酸(NEAA)、0.5% ペニシリン−ストレプトマイシン、0.1mM β−メルカプトエタノールが含まれたDMEM/F12倍地を用いた。7日間培養後、増殖された胚性幹細胞(図1のa)を200乃至300個の細胞を有する塊となるように機械的に分割した後、これをさらに支持細胞上で培養した。
2mg/mlのコラゲナーゼ(collagenaseIV;Invitrogen、Carlsbad、CA、USA)を37℃で40分間処理し、胚性幹細胞コロニーを切り離した後、これらを微生物培養用培養皿に入れ、bFGFを除去した胚性幹細胞培養培地で7乃至21日間浮遊培養し、胚様体(図1のb)を作った。胚様体のうち嚢胞性の胚様体(図2のb)を除去した後、残りの胚様体をマトリゲル(Matrigel(登録商標);BD、Franklin lakes、NJ、USA)が処理された培養皿に付着した後、1 X N−2添加剤が添加された胚性幹細胞培養培地で5日間培養し、神経前駆細胞を選択培養した。以降、1 X N−2添加剤及びbFGF(20ng/ml)が添加されたDMEM/F12倍地で4日間培養し、神経前駆細胞を増殖させた(図3)。その結果、神経前駆細胞の特異的マーカーであるnestinの発現を確認することができた。
選別された球状神経前駆細胞を機械的な方法で30乃至80個の小片に分けた後、マトリゲル(50ug/ml)が処理された35mmの培養皿に入れ、1乃至2日間1 X N−2、bFGF(20ng/ml)を添加した倍地で培養し、1 X N−2、1 X B27 (Gibco)を添加したDMEM/F12倍地で4日間培養し、神経突起を有する神経細胞への分化を誘導した(図1のf及び図4)。その結果、ニューロン特異的なマーカーであるβIII−チューブリンとNeuNの同時発現、βIII−チューブリンとA2B5の同時発現、βIII−チューブリンとnestinの同時発現を確認することができた。
分化した神経細胞が神経細胞の機能をうまく行うがどうかを調べるために、ニューロン特異的な電気生理学的性質と神経伝達物質の分泌に関与する物質を発現するかを観察した。
ニューロン特異的な電気生理学的性質と確認するために、“dialyzed”ホールセル記録法を用いて細胞膜の活動電位を測定し、その結果、本発明の方法で分化誘導された細胞が、ニューロン特異的な細胞膜の活動電位(−60mV〜20mV)を示すことを確認した(図5のa)。
また、シナプス形成に関与するニューロン特異的なタンパク質であるシナプトフィジン(SYP)抗体を用いて免疫蛍光染色法を行い、βIII−チューブリンとシナプトフィジン(SYP)の同時発現を確認した(図5のb)。
上記した結果から、本発明の分化誘導方法で分化誘導された細胞が神経細胞の特徴を有していることが確認された。
実施例1乃至3のような過程で生成された神経細胞にソニック・ヘッジホッグ((SHH)、200 ng/ml;R&D)、線維芽細胞増殖因子8(FGF8、100 ng/ml;Peprotech、Rocky Hill、NJ、USA)を処理して4日間培養した後、これにSHH、FGF8、及びアスコルビン酸(200uM;Sigma)を投与して6日間さらに培養することにより、ドーパミン神経細胞への分化を誘導した。その結果、ドーパミン神経細胞の特異的マーカーであるTHとβIII−チューブリンの同時発現が確認された(図6)。
ニューロン特異的なマーカーと各種のカテコールアミン神経のサブタイプ特異的なマーカーに対する抗体を用いて、免疫組織学的な方法で確認し、その結果、THとAADCの同時発現、THとEn1の同時発現、THとPNMTの同時発現の減少または不在、THとDbHの同時発現の減少または不在、THとGABAの同時発現の減少または不在が確認された(図7のa)。
また、ヒト胚性幹細胞から分化された細胞が、ドーパミン神経細胞の多くの特異的なマーカーを示すかをRT−PCRで確認し、神経前駆細胞(SNM)のmRNAを用いたRT−PCRの結果、Pax6、Sox1、及びNurr1の発現、Oct4、En1、Ptx3、及びDbHの減少または不在を確認し、本発明の方法で最終分化されたドーパミン神経細胞(DA)のmRNAを用いたRT−PCRの結果、Pax6、Sox1、Nurr1、En1、及びPtx3の発現、Oct4及びDbHの減少または不在を確認することができた(図7のa)。
本発明の方法で分化誘導された神経細胞がドーパミン神経細胞に分化される分化効率を比較するために、SHHとFGF8が処理された群(図8の左側及び右側a)と処理されていない群(図8の右側b)から得られた神経細胞を、ドーパミン神経細胞の特異的酵素に対する抗体(抗−TH抗体)を貼り付け、免疫組織学的染色後、共焦点顕微鏡を用いて、陽性で染色された細胞の比率を測定し、86%以上のドーパミン神経細胞への分化誘導率が示されることを確認し(図8の左側)、蛍光フローサイトメトリー(FACS)で、神経細胞のマーカーであるβIII−チューブリンとドーパミン神経細胞のマーカーであるTHを同時発現する細胞の比率を確認した結果、免疫組織学的染色結果に類似するように、神経細胞の84%以上がドーパミン神経細胞に分化されること(図8の右側)を確認した。上記した結果から、神経前駆細胞が神経細胞、特にドーパミン神経細胞に分化され、これを脳、神経系疾患の治療用組成物に製造できることが確認された。
7.1.免疫組織学的方法
試料を先ず順次に80%−、90%−、100%のエタノールでそれぞれ10分ずつ固定させた。10分間含水した後、リン酸緩衝液に保管し、または3%ホルマリン溶液に20分間処理後、リン酸緩衝液に保管した。抗体を処理する前に、試料を1日間ブロッキング溶液(2%ウシ血清アルブミン、Sigma)で処理し、1次抗体に1時間、2次抗体に1時間反応させた後、蛍光保存液で封じた後、共焦点顕微鏡(ニコン、日本)で観察した。
免疫学的染色のために用いられた1次抗体は、次の通りである。Mouse anti−human nestin(1:200)、mouse anti−βIII−tubulin antibody(1:100)、mouse anti−mammalian gamma−aminobutyric acid(GABA、1:100)、mouse anti−synaptophysin(1:100)、mouse anti−TH(1:200)、mouse anti−A2B5(1:100)、rabbit anti−phenylethanolamine N−methyl transferase(PNMT、1:200)、rabbit anti−aromatic amino acid decarboxylase(AADC、1:1000)、mouse anti−neuron−specific nuclear protein(NeuN、1:100)、sheep anti−dopamine hydroxylase(DBH、1:400)、mouse anti−human nuclei(1:50)、mouse anti−human mitochondria(1:50、以上、Chemicon、Temecula、CA、USA);goat anti−TH(1:200、Santacruz、Santa Cruz、California、USA);rabbit anti−TH(1:500)、rabbit anti−vesicular monoamine transporter 2(VMAT2、1:500、以上、Pel−Freez);mouse anti−En1 antibody(1:50、Developmental Studies Hybridoma Bank、Iowa City、IA. USA)。
転写レベルでの発現に対するRT−PCR分析を次のように行った。
RNAを製造者のマニュアルにより、RNAeasy Kit TM(Trizol(invitrogen))を用いて細胞から抽出した後、最終産物をDNaseで分解させ、汚染性ゲノムDNAを除去した。10mMトリス緩衝液(pH7.5)、10mM MgCl2及び5mM DDTを含有する緩衝液に、RNAガード(Pharmacia Upjohn)、DNAseI(PharmaciaUpjohn)及び前記抽出したRNAを入れ、37℃、30〜45分間反応を行った。結果物からタンパク質を除去するために、フェノールクロロホルム抽出を行い、3M酢酸ナトリウム、及び100%低温エタノールでRNAを沈殿させた。前記沈殿されたRNAペレットを70%エタノールで洗浄した後、これを風乾及びDEPC処理水に再懸濁した。
ヒト胚性幹細胞から分化された神経前駆細胞、神経細胞及びドーパミン神経細胞の比率をフローサイトメトリーで測定して分化効率を調べた。神経前駆細胞の検出のためにnestin抗体を、神経細胞の検出のためにはβIII−チューブリン抗体を、ドーパミン神経細胞の検出のためにはチロシン・ヒドロキシラーゼ(TH)抗体を用いた。各細胞は、リン酸緩衝液で洗浄後、37℃で5分間0.05%トリプシン/0.1%EDTA(invitrogen)を処理して単細胞化した。細胞表面抗原ではない場合は、perforationsolution(0.05%Triton X−100、1% BSA、0.1 M Phosphate buffered saline、pH 7.2)で5分間処理した後、1次抗体で30分間反応させ、蛍光標識された2次抗体で30分間反応させた後、リン酸緩衝液で洗浄した。これを、FACScan(BDBioscience、USA)を用いて、CellQuest pro program(BD Bioscience)で分析した。用いられた1次抗体は、次の通りである;mouseanti−human nestin antibody(1:50, Chemicon)、mouse anti−tubulin antibody(1:50, Chemicon)、及びrabbit anti−human TH antibody(1:200、Pel−Freez)。1次抗体の蛍光標識のために用いられた2次抗体は、次の通りである;AlexaFluor 488 donkey anti−mouse IgG及びAlexa Fluor 594 donkey anti−rabbit IgG(1:200、MolecularProbes)。
200−230gの雄Sprague−Dawleyラットを、パーキンソン病の症状を有するモデルに作製するのに用いた。実験群は、次のように3群に分けた:(i)正常群、(ii)パーキンソン病モデルのうち、細胞を処理しなかった群、(iii)パーキンソン病モデルのうち、胚性幹細胞から分化された細胞を処理した群。
モデルを作るために、神経毒素である6−hydroxydopamine(6−OHDA)hydrobromide(8μgfree base、0.2% ascorbic acidが含まれた2μl水溶液)を、内側前脳束(MFB)に次のような脳手術座標に従って注入した:MFB;ブレグマに対して前後−4.4mm、内外側1.2mm、及び硬膜に対して背腹側面−7.8mm。ノルアドレナリン作用性ニューロンの破壊を防ぐために、6−OHDAを注入する30分前に、12.5mg/kgのデシプラミンを腹腔内注射(i.p.)で処理した。6−OHDAを処理してから2週間後、ドーパミン受容体作用薬であるアポモルヒネ(0.1mg/kgi.p.in saline containing ascorbic acid at 2mg/ml)による旋回運動を観察した。1週間後、ヒト胚性幹細胞から分化された細胞をaccutaseで単一細胞化した後、1x105cells/μl濃度、4μlの体積で微量注射器(Hamilton社製)に連結された滅菌したステンレス鋼針(0.3mmO.D.)を用いて、動物モデルの同側性線条体に[AP;+0.2mm、M−L;3.0mm、D−V;4.5mm(2μl)及び5.5mm(2μl)]4分にわたって移植した。移植後4分間細胞が沈着される時間を与えてから、針を除去した。動物モデルは、移植24時間前、シクロスポリンA(CsA:10mg/kg、i.p.)を投与し、解剖するまで処理し続けた。行動検査は、forepawadjusting stepping test(図9、Chang JW.,et al.Biochemical and anatomical characterization of forepaw adjusting steps in rat models of Parkinson’s disease:studies on medial forebrain bundle and striatal lesions.Neuroscience.199988:617−628)及びdrug−inducedturning behavioral test(図10、Cho YH.,et al.Dopamine neurons derived from embryonic stem cells efficiently induce behavioral recovery in a Parkinsonian rat model.Biochem.Biophys.Res.Commun.2006341:6−12)を一緒に行い、移植後2、4、6及び8週の際に連続して行い(図9及び図10のa)、9週間後にアンフェタミン(3mg/kg i.p.)による旋回運動検査を行った(図10のb)。分化させた細胞を移植したモデル(hES)は、培地のみを入れたモデル(sham)と比較するとき、顕著にステッピングが向上し、回転数は減少した。
本実施例では、ヒト胚性幹細胞から本発明の方法で分化誘導したドーパミン神経細胞を、パーキンソン病の治療目的で移植したが、脳、神経系疾患として、消失した脳機能を復元させて疾病を治療することができるものであれば、これに制限されない。
移植10週後、動物モデルをPBSバッファに溶かした25%ウレタンで麻酔させた後、124mlの生理食塩水と250mlの冷えた4%パラホルムアルデヒドで心臓かん流させた。脳を摘出して、4%パラホルムアルデヒド溶液で固定させ、これを、30%スクロースで48時間処理して、凍結ガラス化を防止した後、凍ったまま、30μmの厚さに切り、組織サンプルを作った。脳切片は、rabbitanti−tyroxinehydroxylase(TH)、rabbitanti−βIII−tubulin、mouseanti−human nuclei、またはmouse anti−human mitochondriaの1次抗体で4℃で12時間処理し、AlexaFluor(登録商標)594 donkey anti−rabbit IgGとAlexa Fluor(登録商標)488 donkey anti−mouse IgGの2次抗体を連続的に処理し、4’,6−diamidino−2 phenylindole(DAPI)を含む包埋液で包埋した後、共焦点顕微鏡で観察した(図11)。
上記した結果から、本発明の方法で分化誘導したドーパミン神経細胞が移植後も生きており、ドーパミン神経細胞の特異的な抗体を発現しており、パーキンソン病の治療に有用であることが確認された。
Claims (19)
- (a)5乃至21日間培養された胚様体のうち、嚢胞性の胚様体を除いた残りを、マトリゲル、ラミニン、またはL−ポリオルニチンがコートされた培養皿に付着した後、5乃至7日間、0.5 X N−2添加剤を添加した培地で培養し、神経系以外の細胞の成長を抑制させる段階と、
(b)N−2及びbFGFを添加した培地で、3乃至7日間増殖された胚様体由来細胞群からニューロン特異的な構造物を切除及び分離する段階と、
(c)前記切除されたニューロン特異的な構造物から作られた球状神経前駆細胞を培養する過程において発生する線維芽細胞様細胞と嚢胞構造を除去し、球状神経前駆細胞内で増殖されたニューロン特異的な構造物のみを分離する段階と、
(d)前記(c)の過程を最小4回以上繰り返し、球状神経前駆細胞の純度を高め、増殖する段階とを含むヒト胚性幹細胞から、高収率及び高純度で、神経前駆細胞の分化を誘導し、これを増殖させる方法。 - (a)及び(b)段階の培地が、血清や血清代替物を排除した2mM L−グルタミン、1% 非必須アミノ酸、0.5% ペニシリン−ストレプトマイシン(P/S)または0.1mM β−メルカプトエタノールが含まれたDMEM/F12のいずれか一つである請求項1に記載の方法。
- (a)及び(b)段階の培地が、0.1mM β−メルカプトエタノールが含まれたDMEM/F12である請求項2に記載の方法。
- (b)または(c)段階の切除または分離方法が、ガラスパスツールピペットを用いた機械的な方法である請求項1に記載の方法。
- (d)段階において、(c)の過程を4回乃至10回繰り返すことである請求項1に記載の方法
- 請求項1に記載の方法で作られた神経前駆細胞を分離した後、1乃至2日間N−2及びbFGFを添加した培地で培養し、3乃至7日間N−2及びB27を添加した培地で培養することにより、神経細胞への分化を誘導する方法。
- 神経前駆細胞の分離の際に、化学的処理を介さず、ガラスパスツールピペットを用いた機械的な方法で球状神経前駆細胞を切除、分離することにより、細胞の生存率を高める請求項6に記載の方法。
- 請求項6に記載の方法で作られた神経細胞にSHH及びFGF8を処理し、2乃至4日間培養した後、これに、SHH、FGF8及びアスコルビン酸を投与してから、3乃至7日間追加培養することにより、ドーパミン神経細胞を誘導する方法。
- 請求項1乃至8のいずれか一項に記載の方法により分化された細胞及び/又は該細胞の子孫。
- 下記の特徴の4つ以上を有する請求項9に記載の細胞及び/又はその子孫:
(a)nestinの発現;
(b)βIII−チューブリンとNeuNの同時発現;
(c)βIII−チューブリンとA2B5の同時発現;
(d)βIII−チューブリンとnestinの同時発現;
(e)−60mA〜20mAのニューロン特異的な細胞膜の活動電位;
(f)βIII−チューブリンとシナプトフィジン(SYP)の同時発現;及び
(g)神経細胞培養液からドーパミンの分泌。 - 下記の特徴の5つ以上を有する請求項9に記載の細胞及び/又はその子孫:
(a)βIII−チューブリンとTH(tyrosine hydroxylase)の同時発現;
(b)THとAADCの同時発現;
(c)THとEn1の同時発現;
(d)THとPNMTの同時発現の減少または不在;
(e)THとDbHの同時発現の減少または不在;
(f)THとGABAの同時発現の減少または不在;
(g)神経前駆細胞(SNM)でのPax6、Sox1、及び/又はNurr1の発現;
(h)神経前駆細胞(SNM)でのOct4、En1、Ptx3、及び/又はDBHの発現の減少または不在;
(i)ドーパミン神経細胞(DA)でのPax6、Sox1、Nurr1、En1及び/又はPtx3の発現;及び
(j)ドーパミン神経細胞(DA)でのOct4及び/又はDBHの発現の減少または不在。 - 約80%以上の細胞が、下記の特徴を有する請求項11に記載の細胞及び/又はその子孫:
(a)βIII−チューブリンとTHの同時発現;
(b)THとAADCの同時発現;
(c)THとEn1の同時発現;
(d)ドーパミン神経細胞(DA)でのPax6、Sox1、Nurr1、En1及び/又はPtx3の発現;
(e)ドーパミン神経細胞(DA)でのOct4及び/又はDBHの発現の減少または不在。 - 前記遺伝子の発現の増加または減少は、前記遺伝子がコードするタンパク質に対する抗体及び/又はRT−PCRで検出可能である請求項9乃至12のいずれか一項に記載の細胞及び/又はその子孫。
- 請求項9乃至12のいずれか一項に記載の細胞及び/又はその子孫を含む脳または神経系疾患に対する治療用組成物。
- 前記神経系疾患が、パーキンソン病、神経痛、関節炎、頭痛、統合失調症、てんかん、脳卒中、不眠症、認知症、うつ病、ジスキネジア、アルツハイマー病、レヴィー小体認知症、ハンチントン病、トゥレット・シンドローム、不安、学習及び記憶障害、神経変性病よりなる群から選ばれるいずれか一つ以上である請求項14に記載の組成物。
- 前記神経系疾患が、パーキンソン病である請求項14に記載の組成物。
- 請求項13に記載の細胞及び/又はその子孫を含む脳または神経系疾患に対する治療用組成物。
- 前記神経系疾患が、パーキンソン病、神経痛、関節炎、頭痛、統合失調症、てんかん、脳卒中、不眠症、認知症、うつ病、ジスキネジア、アルツハイマー病、レヴィー小体認知症、ハンチントン病、トゥレット・シンドローム、不安、学習及び記憶障害、神経変性病よりなる群から選ばれるいずれか一つ以上である請求項17に記載の組成物。
- 前記神経系疾患が、パーキンソン病である請求項17に記載の組成物。
