- 【特許請求の範囲】 1.少なくとも一種の成長因子を補給した培地を含むことを特徴とする前駆細胞 の増殖を誘発するための組成物。 2.表皮細胞成長因子、アンフィレグリン、形質転換成長因子α及びインシュリ ン様成長因子−1の群から選ばれた成長因子を含む請求の範囲第1項に記載の前 駆細胞の増殖を誘発するための組成物。 3.試験管内で培養され、少なくとも一種の成長因子の存在下で、増殖し、分化 するように誘発される哺乳類から単離された細胞を含むことを特徴とする前駆細 胞。 4.細胞がマウス又はヒトから単離され、試験管内で培養され、表皮細胞成長因 子又は形質転換成長因子αの存在下で、増殖し、分化するように誘発される請求 の範囲第3項に記載の前駆細胞。 5.ヒトに移植された細胞が神経細胞の表現型の性質を発現し、宿主の神経組織 と神経連結体を形成する請求の範囲第3項に記載の前駆細胞。 6.神経伝達物質又は神経伝達物質合成酵素に対し免疫反応性を示す請求の範囲 第5項に記載の前駆細胞。 7.免疫反応性がGABA、物質P、又はグルタミン酸デカルボキシラーゼに対 して示される請求の範囲第6項に記載の前駆細胞。 8.低温保存される請求の範囲第3項に記載の前駆細胞。 9(a)哺乳類から細胞を単離する工程、(b)その細胞を成長因子を含む培地 に露出する工程、(c)その細胞を増殖するように誘発する工程、及び(d)そ の細胞を分化するように誘発する工程を含むことを特徴とする前駆細胞の試験管 内の増殖方法。 10.細胞をマウス又はヒトから単離し、そして表皮細胞成長因子又は形質転換 成長因子αを含む培地に露出する請求の範囲第9項に記載の前駆細胞の試験管内 の増殖方法。 11.細胞を、神経系を標的とする推定の治療薬の薬剤スクリーニングの目的で 使用する請求の範囲第9項に記載の前駆細胞の試験管内の増殖方法。 12.細胞を成長因子又は医薬組成物で誘発して現場で増殖、分化させることを 特徴とする前駆細胞の現場増殖方法。 13.成長因子がEGF又はTGFαを含み、前駆細胞に接近して移植される請 求の範囲第12項に記載の前駆細胞の現場増殖方法。 14.(1)試験管内の増殖及び分化、その後の移植、(2)試験管内の増殖、 その後の移植、更にその後の生体内の増殖及び分化、又は(3)試験管内の増殖 、移植及び生体内の分化にかけられた細胞を哺乳類に移植することを特徴とする 前駆細胞の生体内の移植方法。 15.表皮細胞成長因子又は形質転換成長因子の存在下で培養された細胞をマウ ス又はヒトに移植し、移植前駆細胞が組織移植片又は神経移植片として作用する 請求の範囲第14項に記載の前駆細胞の生体内の移植方法。 16.(1)試験管内の増殖及び分化、その後の移植、(2)試験管内の増殖、 その後の移植、更にその後の生体内の増殖及び分化、(3)試験管内の増殖、移 植及び生体内の分化、又は(4)生体内の増殖及び分化にかけられた生理学上有 効な数の前駆細胞を神経疾患又は神経変性疾患の治療を要する患者に投与するこ とを特徴とする神経疾患又は神経変性疾患の治療方法。 17.前駆細胞が異種供与体に由来する請求の範囲第16項に記載の神経変性疾 患の治療方法。 18.前駆細胞が自己供与体に由来する請求の範囲第16項に記載の神経変性疾 患の治療方法。 19.供与体が胎児、幼若体又は成体である請求の範囲第17項又は第18項に 記載の神経変性疾患の治療方法。 20.神経変性疾患がパーキンソン病、もしくはアルツハイマー病又はハンチン トン舞踏病を含む請求の範囲第16項、第17項又は第18項に記載の神経変性 疾患の治療方法。 21.神経変性疾患がバリスムス又はアテトーシスを生じる請求の範囲第16項 、第17項又は第18項に記載の神経変性疾患の治療方法。 22.神経疾患が精神分裂症、筋萎縮性側索硬化症、てんかん、脳性小児麻痺又 は卒中を含む請求の範囲第16項に記載の神経疾患の治療方法。 23.前駆細胞を中枢神経系中の特別な位置に投与する請求の範囲第16項に記 載の神経変性疾患の治療方法。 24.特別な位置が基底核、マイネルト基底核、黒質級密層、淡蒼球、視床下核 、線条、脳幹、海馬、大脳皮質、脊髄、小脳、網膜又は視索を含む請求の範囲第 23項に記載の神経変性疾患の治療方法。 25.前駆細胞にDM発現ベクターを移入し、得られる細胞を神経移植して神経 変性徴候を治療することを特徴とする前駆細胞中の成長因子、神経伝達物質及び 神経伝達物質合成酵素の発現方法。 26.前駆細胞をグリセロールを含む培地中で希釈し、約−80℃以下の温度で 凍結することを特徴とする前駆細胞の低温保存方法。 27.培地が10%のグリセロールを含む請求の範囲第26項に記載の前駆細胞 の低温保存方法。 28.懸濁培養液中の前駆細胞の連続永続化方法。 29.基質付着培養液中の前駆細胞の連続永続化方法。
