Description
本発明は、神経前駆細胞の試験管内の培養方法、及び組織移植片としてのこれらの細胞の使用に関する。一つの局面において、本発明は、分化するように誘発でき、かつ動物への神経移植に使用されて神経病、神経変性及び中枢神経(CNS)外傷の徴候を軽減し得る多数の非腫瘍形成性の神経前駆細胞の単離及び試験管内の永続化方法に関する。別の局面において、本発明は神経系を標的とする推定の治療薬の薬剤スクリーニングの目的の神経細胞の生産方法に関する。また、別の局面において、本発明は自家移植のための細胞の生産方法に関する。別の局面において、本発明は宿主中の前駆細胞の現場(in situ)増殖及び分化方法に関する。
発明の背景
神経系の発達は胎児発達の初期段階で始まる。新しい神経細胞の発生である神経発生は生後期間で初期に完結する。しかしながら、神経回路に係わるシナプス連結体は、シナプス可変性及び細胞死滅のために、個人の寿命中に連続的に変化する。
神経発達の第一工程は細胞誕生であり、これは前駆体又は前駆細胞が増殖し、分化する正確な時間上及び空間上の順序である。増殖細胞は神経芽細胞、グリア芽細胞及び基幹細胞を生じる。
第二工程は、前駆細胞が神経細胞及びグリア細胞になり、それらの最終の位置に移動する時の細胞型の分化及び移動の期間である。神経管に由来する細胞は中枢神経(CNS)の神経細胞及びグリアを生じ、一方、神経冠に由来する細胞は末梢神経系(PNS)の細胞を生じる。神経成長因子(NGF)の如き発達中に存在する或る種の因子は、神経細胞の成長を促進する。NGFは神経冠の細胞により分泌され、神経軸索の発芽及び成長を刺激する。
発達の第三工程は、細胞が特別な神経伝達物質の発現の如き特定の表現型の性質を獲得する時に起こる。この時点で、神経細胞はまたそれらの標的でシナプス化するプロセスを延長する。神経細胞は分化後に分裂しない。
最後に、選択的な細胞死滅が起こり、この場合、特定の細胞、繊維及びシナプス連結体の変性及び死滅が神経系の複雑な回路を“微細に同調する”。この“微細な同調”が宿主の寿命にわたって続く。寿命の後半に、老化、感染及びその他の未知の病因による選択的な変性が神経変性疾患をもたらし得る。
神経変性疾患の治療が、これらの疾患につき最大のリスクのある老人の増大する人口のために、近年の重大な関心事になりつつある。これらの疾患(アルツハイマー病、ハンチントン舞踏病及びパーキンソン病を含む)は、脳中の特定の位置における神経細胞の変性に関連しており、これがその脳領域を神経の信号発生に不可欠である神経伝達物質を合成し、放出することをできなくさせる。神経伝達物質の欠如から生じると考えられる別の疾患は精神分裂症である。
また、神経変性は、神経損失を生じる症状及び疾患を含む。これらの症状として、CNS外傷の結果として生じる症状(卒中及びてんかんを含む)が挙げられる。筋萎縮性側索硬化症及び脳性小児麻痺の如きその他の疾患がまた神経損失を生じる。脳の特定の領域間の相互作用、及びこれらの相互作用がそれらの機能の外向きの発現を生じる方法の理解の欠如のために、CNS変性の治療が妨げられる。複雑な細胞相互作用並びに挙動及び認識機能に関するそれらの効果の研究は、多数の細胞及びそれらが形成する複雑な神経回路のために一層困難にされる。
殆どのCNS療法の目的は、細胞変性のために失われる特別な化学的機能又は酵素活性を回復することである。
基底核として知られている脳領域における変性は、その正確な位置に応じて、種々の認識上及び運動上の徴候を有する疾患をもたらし得る。基底核は、線条(これは尾状核及び果核からなる)、淡蒼球、黒質、無名質、腹側淡蒼球、マイネルト基底核、腹側被蓋領域及び視床下核を含む多くの別個の領域からなる。
アルツハイマー病の場合、前脳及び大脳皮質の深部に達する細胞変性がある。加えて、更に精密に検査すると、基底核の或る領域、即ち、マイネルト基底核に局在化された変性は選択的に変性されていることが明らかである。この核は、通常、コリン作用性突起部を大脳皮質に送り、これらが記憶を含む認識機能に関与すると考えられる。
多くの運動不足は基底核中の変性の結果である。ハンチントン舞踏病は線条中の神経細胞の変性と関連しており、これが宿主に不随意の反射運動をもたらす。視床下核と称される小領域の変性はバリスムスと称される症状における極度の激しくあばれる運動と関連しており、一方、果核及び淡蒼球における変性は遅い身をよじる運動又はアテトーシスの症状と関連している。パーキンソン病の場合、変性が基底核のその他の領域である黒質の対の緻密層に見られる。この領域は、通常、ドーパミン作用性連結体を背部線条に送り、これらは運動を調節するのに重要である。パーキンソン病の療法はこの回路につきドーパミン作用活性を回復することに集中していた。
神経障害のその他の形態は、筋萎縮性側索硬化症及び脳性小児麻痺の如き神経変性の結果として、又は卒中及びてんかんの如きCNS外傷の結果として生じ得る。
現在まで、CNS機能障害の治療は主として医薬組成物の投与によるものであった。不運なことに、この種の治療は、薬剤を血液脳関門を横切って輸送する制限された可能性、及びこれらの薬剤が長期投与される患者により獲得される薬剤寛容性を含む多くの合併症を伴っていた。例えば、パーキンソン病の患者におけるドーパミン作用活性の部分的な回復は、血液脳関門を横断できるドーパミン前駆体であるレボドーパで達成された。しかしながら、患者はレボドーパの効果に対して寛容性になり、それ故、その効果を維持するために絶えず増加する投薬量が必要とされる。現在、アルツハイマー病に有効な医薬上の治療がない。
最近、神経組織移植の概念がパーキンソン病の如き神経病の治療に適用されていた。神経移植は一定の薬剤投与の必要を避け得るだけでなく、血液脳関門により生じる複雑な薬剤送出系の必要を避け得る。しかしながら、この技術にも同様の制限がある。第一に、その他の宿主からの分化細胞の細胞表面分子を有する移植に使用された細胞は宿主に免疫反応を誘発し得る。加えて、細胞は、それらが隣接細胞と正常な神経連結体を形成できるような発達の段階にある必要がある。これらの理由のために、神経移植に関する初期の研究は胎児の細胞の使用に集中した。ペーロウ(Perlow)らは、“Brain grafts reduce motor abnormalities p-roduced by destruction of nigrostriated dopamine system,”Science 204:643-647(1979)において化学的に誘発された黒質線状体の外傷を有する成体ラットへの胎児のドーパミン作用性神経細胞の移植を記載している。これらの移植片は宿主動物において良好な生存率、軸索外殖及びかなり減少された運動異常を示した。リンドバル(Lindvall)らは、“Grafts of fetal dopamine neurons survive and improve motor function in Parkinson’s Disease,”Science 247: 574-577(1990)において、ヒト胎児の中脳ドーパミン神経細胞の神経移植がドーパミンの合成及び貯蔵を回復でき、かつパーキンソン病を患う患者の硬直及び運動緩徐を低減できることを示した。また、フリード(Freed)らは、“Transplantation of human fetal dopamine cells for Parkinson’s Disease,”Arch.Neurol.47: 505-512(1990)において、胎児移植片を受けた患者の改善を示す。
上記文献は、哺乳類の胎児脳組織が即時の移植後に良好な生存特性を有することを開示している。胎児神経細胞の増大された生存能は、成体神経細胞よりも胎児神経細胞の酸素欠乏に対する低減された感受性のためであり、そしてまた胎児細胞における細胞表面マーカー(その存在は成体から移植組織の拒絶をもたらし得る)の欠如のためであると考えられる。しかしながら、脳は免疫学上特権的な部位と考えられるが、胎児組織の或る種の拒絶が生じ得る。それ故、胎児組織を使用する可能性は、別の宿主から単離された胎児組織の組織拒絶、及びその結果として生じる免疫抑制薬の必要性のためだけでなく、胎児組織を得る際の倫理上の問題のために制限される。しかしながら、新生児の脳組織は生存につき制限された能力を有し、成体哺乳類のCNS神経細胞は一般に脳への移植に対して生き残らない。
成体CNS神経細胞は神経移植に良い候補ではないが、成体末梢神経系(PNS)からの神経細胞は移植に対して生き残り、かつ宿主の神経組織を発達させる上で神経栄養作用及び神経膠栄養(gliotrophic)作用を与えることが示された。移植のための非CNS神経組織の一つの源は副腎髄質である。副腎のクロム親和細胞は神経冠のようなPNS神経細胞に由来し、シナプスを受容し、PNS神経細胞と同様の酵素タンパク質及びキャリヤーを生産する。これらの細胞は無傷の副腎髄質中で内分泌様式で機能するが、培養中にこれらの細胞はそれらの腺状表現型を失い、また或る種の成長因子及びホルモンの存在下の培養中に神経の特徴を発達させる(ノッター(Notter)ら著、“Neuronal properties of monkey adrenal medulla in vitro, Cell Tissue R-esearch 244: 69-76[1986])。哺乳類のCNSに移植された場合、これらの細胞は生存し、そしてかなりの量のドーパミン(これはCNSの隣接領域中のドーパミン受容体と相互作用し得る)を合成する。
米国特許第4,980,174号明細書において、ラットの前頭皮質への成体ラットの松果腺及び副腎髄質から単離されたモノアミンを含む細胞の移植は、宿主のうつ病の形態である学習された無力の軽減をもたらした。米国特許第4,753,635号明細書において、去勢ウシに由来するクロム親和細胞及び副腎髄質組織がラットの脳幹又は脊髄に移植され、そして移植された組織又は細胞が侵害受容体相互作用物質(即ち、ドーパミンの如きカテコールアミン)を放出するように誘発された場合に痛覚消失を生じた。副腎髄質細胞がヒトに自家移植され、生存し、徴候の軽度〜適度の改善をもたらした(ワッツ(Watts)ら著、“Adrenal-caudate transplantation in patients with Parkinson’s Disease(PD):1-year follow-up”, Neurology 39 Suppl. 1: 127[1989]、ハーティグ(Hurtig)ら著、“Post-mo-rtem analysis of adrenal-medulla-to-caudate autograft in a patient with Parkinson’s Disease”, Annals of Neurology 25: 607-614[1989])。しかしながら、副腎細胞は正常な神経表現型を得ず、それ故、シナプス連結体が形成される必要がある場合の移植片につきおそらく制限されて使用される。
神経移植のための組織のその他の源は細胞系からのものである。細胞系は、癌遺伝子による正常な細胞の形質転換(セプコ(Cepko)著、“Immortalization of neural cells via retrovirus-mediated oncogene transduction”, Ann.Rev.Neurosci. 12: 47-65[1989])又は試験管内の変性された成長特性を有する細胞の培養(ロネット(Ronnett)ら著、“Human cortical neuronal cell line: Establishment from a patient with unilateral megalencephaly”, Science 248:603-605[1990])のいずれかにより誘導される不死化細胞である。このような細胞は、多量で培養液中で増殖されて多重移植に使用し得る。幾つかの細胞系が化学処理後に分化して種々の神経の性質、例えば、神経突起形成、興奮性膜並びに神経伝達物質及びそれらの受容体の合成を発現することが示された。更に、分化後に、これらの細胞は無糸分裂性(amitotic)であり、それ故、非癌性であることが明らかである。しかしながら、これらの細胞が逆免疫反応を誘発する潜在性、細胞を不死化するためのレトロウイルスの使用、無糸分裂状態へのこれらの細胞の復帰の潜在性、及び正常な成長抑制信号に対するこれらの細胞の応答の欠如が細胞系を広範囲の使用に最適なものにしない。
神経移植のその他の方法は、遺伝子操作された細胞型又は遺伝子療法の使用を伴う。この方法を使用して、異種遺伝子又はトランスジーンが、特別な酵素活性の不十分である細胞に導入され、それにより活性を回復し得る。移入遺伝子を含む細胞が神経変性の部位に移植でき、神経伝達物質及び成長因子の如き生産物を与えることができ(ローゼンベルグ(Rosenberg)ら著、“Grafting genetically modified cells to the damaged brain: Restorative effects of NGF Expression”, Science 242: 1575-1578,[1988])、これらが変性の徴候の一部を軽減するように機能し得る。しかしながら、これらの細胞と免疫反応を誘発する恐れが依然としてあり、レトロウイルス媒介の移入はその他の細胞異常を生じ得る。また、レトロウイルス媒介の遺伝子移入により生産された細胞系は移植後にそれらの移入遺伝子を次第に失活することが示された(パルマー(Palmer)ら著、“Genetically modified skin fibroblasts persist long after transplantattion but gradually inactivate introduced genes”, Proc.Natl.Acad.Sci.88: 1330-1334頁[1991])。加えて、これらの細胞はまた宿主組織との正常な神経連結体を得られないことがある。
従来技術では移植片を宿主組織中に充分に組み込むことができないこと、及び移植に関して信頼できる源から無制限の量の細胞の利用可能性の欠如が、おそらく、神経移植の最大の制限である。
それ故、神経細胞の培養及び移植の従来技術の方法に付随した上記欠点に鑑みて、神経移植に関する無制限の数の細胞(これは神経細胞に分化できる)の信頼できる源に対する要望が当業界に依然として存することが明らかである。更に、異種供与細胞が免疫学上拒絶し得ると仮定すると、神経細胞に分化できる幼若体又は成体の脳からの自己前駆細胞の単離、永続化及び移植に対する要望がある。加えて、比較的非観血的様式、即ち、前駆細胞を神経細胞に現場で増殖、分化するように誘発することによる損傷された神経組織の回復に対する要望がある。
従って、本発明の主目的は、神経移植に関する無制限の数の細胞(これは神経細胞に分化できる)の信頼できる源を提供することである。
本発明の別の目的は、胚体、幼若体及び成体の脳からの神経前駆細胞の現場増殖、神経細胞へのこれらの前駆細胞の分化、そして異種宿主又は自己宿主への前駆細胞の神経移植のための方法を提供することである。また、前駆細胞は現場増殖、分化するように誘発でき、それにより移植の必要を避け得る。
本発明の更に別の目的は、神経系を標的とする推定の治療薬のスクリーニングの目的で神経細胞を生産する方法を提供することである。
発明の要約
本発明は、一局面において、少なくとも一種の成長因子、好ましくは表皮成長因子又はトランスフォーミング成長因子αを補給した培地を含む前駆細胞の増殖を誘発するための組成物を提供する。
また、本発明は、(a)哺乳類から細胞を単離する工程、(b)その細胞を成長因子を含む培地に露出する工程、(c)その細胞を増殖するように誘発する工程、及び(d)その細胞を分化するように誘発する工程を含む前駆細胞の試験管内の増殖及び分化の方法を提供する。前駆細胞の増殖及び永続化は懸濁培養液中、又は細胞を固定基質に付着させることにより行い得る。増殖及び分化は移植の前又は後に、試験管内条件又は生体内条件の種々の組み合わせで行うことができ、これらの組み合わせは(1)試験管内の増殖及び分化、その後の移植、(2)試験管内の増殖、移植、その後の生体内の更なる増殖及び分化、及び(3)試験管内の増殖、移植及び生体内の分化を含む。また、本発明は前駆細胞の現場増殖及び分化を提供し、これは移植を必要としないで宿主中で直接行い得る。
別の局面において、本発明は凍結(cryogenesis)による前駆細胞の保存を提供する。
また、本発明は、上記(1)〜(3)のいずれかで処理された前駆細胞の生体内の移植方法であって、少なくとも一種の成長因子で処理されたこれらの細胞を哺乳類に移植することを含む生体内の移植方法を提供する。
更に、本発明は、(1)〜(3)のいずれかで処理され、成長因子で処理され、神経細胞に分化するように誘発された前駆細胞を哺乳類に投与することを特徴とする神経変性疾患の治療方法を提供する。また、本発明は、哺乳類のCNS前駆細胞を現場で刺激して神経細胞に増殖、分化させることを特徴とする神経変性疾患の治療方法を提供する。
また、本発明は、成長因子、成長因子受容体、及びペプチド神経伝達物質の遺伝子生産物を発現でき、又は神経伝達物質(アミノ酸、生体アミン及び神経ペプチドに関する神経伝達物質を含む)の合成に関与する酵素を発現し得るベクターによるこれらの前駆細胞のトランスフェクション、及び神経変性の領域へのこれらの移入細胞の移植のための方法を提供する。
更に別の局面において、本発明は潜在的な神経治療医薬のスクリーニング方法を提供する。

発明の詳細な説明
本発明は、動物からの神経前駆細胞の単離、成長因子の存在下の試験管内又は生体内のこれらの細胞の永続化、神経細胞及びグリアへのこれらの細胞の分化、前駆細胞の凍結、及び神経変性疾患及び神経外傷を治療するための宿主中への神経移植のためのこれらの細胞の使用、並びに薬剤スクリーニング応用を提供する。これらの前駆細胞における増殖及び分化の誘発は、組織培養液中で、又は、適当な条件下で、宿主中で下記の組み合わせで付着細胞又は懸濁細胞を生育することにより行い得る。(1)試験管内の増殖及び分化、その後の移植、(2)試験管内の増殖、移植、その後の生体内の更なる増殖及び分化、(3)試験管内の増殖、移植及び生体内の分化、及び(4)in situの増殖及び分化。in situの増殖及び分化は、前駆細胞を医薬操作でin situ増殖させる非外科的方法を伴う。
前駆細胞は、それらの分化が必要とされるまで前駆細胞を抑制状態に維持する持続性抑制の影響下にあると考えられる。この持続性抑制の影響は可溶性因子であってもよい。何となれば、前駆細胞に逆に添加された神経溶解産物の分画は試験管内でそれらの増殖を抑制し得るからである。培養中に、これらの前駆細胞はこの同じ抑制の影響を受けなくてもよい。これらの細胞は増殖でき、そして最終的に分化され、それ故、非分裂性である神経細胞と違って、それらは無制限の数で生産でき、それ故、神経変性疾患を有する異種宿主及び自己宿主への移植に非常に適している。
前駆細胞は、制限なく分裂でき、そして特定の条件下で最終的に神経細胞及びグリアに分化する娘細胞を生産できる多能な基幹細胞を意味する。これらの細胞は異種宿主又は自己宿主への移植に使用し得る。異種は、前駆細胞が最初に誘導された動物以外の宿主を意味する。自己は、前駆細胞が最初に誘導された同じ宿主を意味する。
細胞は、あらゆる動物からの胚体、生後、幼若体又は成体の神経組織から得ることができる。あらゆる動物は、神経組織を含むあらゆる多細胞動物を意味する。更に詳しくは、あらゆる昆虫、魚、爬虫類、鳥、両生類又は哺乳類、等を意味する。最も好ましい供与体は哺乳類、特にマウス及びヒトである。
異種供与動物の場合、動物が安楽死され、そして滅菌操作を使用して関係のある脳及び特定の領域が除去し得る。特別に関係のある脳領域は、宿主の脳の変性領域につき機能を回復するのに利用できる前駆細胞を得ることができるあらゆる領域を含む。これらの領域として、大脳皮質、小脳、中脳、脳幹、脊髄及び脳室組織を含む中枢神経(CNS)の領域、並びに頸動脈小体及び副腎髄質を含む末梢神経系(PNS)の領域が挙げられる。更に詳しくは、これらの領域として、基底核中の領域、好ましくは尾状核と果核からなる線条、又は種々の細胞群、例えば、淡蒼球、視床下核、アルツハイマー病の患者で変性されることが知られている基底核、もしくはパーキンソン病の患者で変性されることが知られている黒質緻密層が挙げられる。この後者の細胞群は高レベルの色素であるメラニンの存在によるその暗色にちなんで名付けられ、それにより同定される。メラニンはチロシナーゼの作用によりチロシン又はドーパから誘導される。
ヒトの異種神経前駆細胞は、選択的人工中絶により得られた胎児組織、又は生後、幼若体もしくは成体の器官供与体に由来し得る。自己神経組織はバイオプシーにより、又は神経組織が特にてんかん手術、更に特別に一時的葉切除及び海馬切除中に除去される神経手術を受ける患者から得ることができる。
組織は、組織の連結性細胞外基質からの個々の細胞の単離により供与組織から得ることができる。単離は、トリプシン、コラゲナーゼ等の如き酵素による処理を含む既知の操作を使用して、又はブラント(blunt)装置によるような単離の物理的方法を使用することにより得ることができる。胎児細胞の単離は組織培地中で行うことができ、一方、幼若体細胞及び成体細胞の単離に好ましい培地は人工の脳脊髄液(aCSF)である。レギュラーaCSFは124ミリモルのNaCl、5ミリモルのKCl、1.3ミリモルのMgCl2、2ミリモルのCaCl2、26ミリモルのNaHCO3、及び10ミリモルのD-グルコースを含む。低Ca++aCSFは、MgCl2につき3.2ミリモルの濃度、そしてCaCl2につき0.1ミリモルの濃度以外は同じ成分を含む。
単離された細胞は、細胞代謝に必要とされる補充物質、例えば、グルタミン及びその他のアミノ酸、ビタミン、ミネラル及び有益なタンパク質、例えば、トランスフェリン等を含む細胞増殖を支持できるあらゆる既知の培地(MEM、DMEM、PRMI、F-12、等を含む)に入れることができる。また、培地は、酵母、細菌及び菌類による汚染を防止するための抗生物質、例えば、ペニシリン、ストレプトマイシン、ゲンタマイシン等を含んでいてもよい。或る場合には、培地はウシ、ウマ、ニワトリ等に由来する血清を含んでいてもよい。細胞に特に好ましい培地はDMEMとF-12の混合物である。
培養の条件は生理条件に近似すべきである。培地のpHは生理pHに近似すべきであり、pH6〜8が好ましく、pH7付近が更に好ましく、pH約7.4が特に好ましい。細胞は生理温度に近い温度で培養されるべきであり、30℃〜40℃が好ましく、32℃〜38℃が更に好ましく、35℃〜37℃が最も好ましい。
細胞は懸濁して、又は固定基質上で増殖し得るが、前駆細胞の増殖は多数の細胞を生じるために懸濁して行われることが好ましい。懸濁細胞の場合、フラスコが良く振とうされ、神経球体がフラスコの底隅部上に沈降される。次いで球体が50mlの遠心分離管に移され、低速で遠心分離される。培地が吸引され、細胞が成長因子と共に少量の培地中で再度懸濁され、細胞が機械的に単離される。次いで細胞がカウントされ、再度塗布される。
固定基質上の細胞の培養は、最早分裂できないが、移植に使用されてその他の神経標的と連結体を形成し得る最終的に分化された細胞への前駆細胞の分化をもたらし得る。また、分化は、細胞が単離及び増殖の再開を行わないで所定期間にわたって培養される場合には懸濁状態で誘発し得る。
増殖を誘発するのに使用し得る成長因子として、前駆細胞を増殖させるあらゆる栄養因子が挙げられ、これは細胞の表面の受容体に結合して細胞に栄養効果又は成長誘発効果を与えるあらゆる分子を含む。このような因子として、神経成長因子(NGF)、酸性及び塩基性の繊維芽細胞成長因子(aFGF、bFGF)、血小板由来成長因子(PDGF)、チロトロピン放出ホルモン(TRH)、表皮細胞成長因子(EGF)、EGF様リガンド、アンフィレグリン(amphiregulin)、形質転換成長因子α及びβ(TGFα、TGF β)、インシュリン様成長因子(IGF-1)等が挙げられる。
最も好ましい因子はEGF及びTGF αである。これらの成長因子は夫々53のアミノ酸及び50のアミノ酸の一本鎖ポリペプチドであり、これらはEGF受容体(EGFR)の結合につき競合する。EGF及びTGF αは細胞分化に影響することが実証された。TGF α免疫反応性(IR)、EGF-IR及びEGFR-IRだけでなく、TGF α及びEGFのmRNAが、げっ歯類及びヒトのCNS中で観察された。リガンド結合研究の結果は、げっ歯類の脳中の[125I]EGFの結合部位の数が妊娠中に増加し、成体よりも生後第1週で高く、これがCNS発達におけるこの因子の役割と合致することを実証した。EGFは、短いプロセスを有する星状細胞及び神経細胞を生じる線条からの多能の前駆細胞の増殖を誘発し、またTGF αは長い良く発達した神経突起を有する原形質性星状細胞及び神経細胞を生じる多能の前駆細胞の増殖を誘発する。使用される成長因子の濃度は1fg/ml〜1mg/ml、好ましくは20ng/mlである。
神経細胞及びグリア細胞は、多能前駆細胞の分化後に誘導し得る。細胞の分化は、増殖をもらたす生物イベントのカスケードを活性化する当業界で知られているあらゆる方法(イノシトールトリホスフェート及び細胞内Ca++の遊離、ジアシルグリセロールの遊離及びプロテインキナーゼC及びその他の細胞キナーゼの活性化、等を含む)により誘発し得る。フォルボールエステル、成長因子及びその他の化学シグナルによる処理だけでなく、イオン的に帯電された表面、例えば、ポリ−L−リシン及びポリ−L−オルニチン等の如き固定基質上の増殖が、分化を誘発し得る。また、分化は、成長因子の継続した存在下で、増殖の再開の不在下で細胞を懸濁させることにより誘発し得る。
特別な神経領域からの細胞が、滅菌操作を使用して動物から除去され、当業界で知られているあらゆる方法を使用して機械的に単離される。胚体又は生後まもなくの脳組織が培地中に直接単離され、一方、幼若体又は成体の脳組織が人工の脳脊髄液(CSF)中に単離される。単離された細胞が200〜2000rpm、更に特別には400〜800rpmの低速で遠心分離され、次いで培地中で再度懸濁される。細胞は約5 x 104〜2 x 105個の細胞/ml、好ましくは1 x 105個の細胞/mlで再度懸濁される。固定基質に塗布される細胞は、約2−3 x 103個の細胞/cm2、好ましくは2.5 x 103個の細胞/cm2で塗布される。
培地中の細胞懸濁液は前駆細胞の増殖を可能にするあらゆる成長因子で補給され、細胞を維持できるあらゆる受器、特に培養フラスコ、培養プレート又はローラーびん、更に特別には25cm2の培養フラスコの如き小さい培養フラスコ中で接種される。細胞は37℃のインキュベーター中で3〜4日以内に増殖し、増殖はその後のいずれかの時点で細胞の単離及び成長因子を含む新しい培地中の再懸濁により再開し得る。
基質の不在下で、細胞はフラスコの床から隆起し、懸濁状態で増殖し続けて未分化細胞の中空球体を形成する。試験管内で約3〜10日後、更に特別には試験管内で約6〜7日後に、増殖クラスター(神経球体)が2〜7日毎、更に特別には2〜4日毎に穏やかな遠心分離及び成長因子を含む培地中の再懸濁により飼育される。
試験管内で6〜7日後に、神経球体中の個々の細胞が、ブラント装置による神経球体の物理的単離により、更に特別には神経球体をピペット、特に火仕上げパスツールピペットですり砕くことにより分離し得る。単離された神経球体からの単細胞が成長因子を含む培地中で懸濁され、これらの細胞の一部が増殖し、主として未分化細胞を含む新しい神経球体を形成する。細胞の分化は、EGF、TGF α又は分化を維持できるあらゆる因子、例えば、塩基性繊維芽細胞成長因子(bFGF)等の継続した存在下で細胞を固定基質(ポリ−L−リシン又はポリ−L−オルニチン被覆フラスコ、プレート又はカバースリップを含む)に塗布することにより誘発し得る。また、分化は、増殖を再開しないで、EGF又はTGF αを含む成長因子の存在下で細胞を懸濁させることにより誘発し得る。
成長因子誘発分化後に生じる二つの型の細胞、即ち、神経細胞及びグリアにつき細胞分子を同定するために、既知の細胞マーカーに特異的な抗体を使用して免疫細胞化学が行われる。神経タンパク質又はグリアタンパク質に特異的な抗体がグリアから神経細胞の細胞特性又は表現型の性質を区別するのに使用し得る。特に、神経細胞に関するこれらの細胞マーカーとして、神経特異的エノラーゼ(NSE)及び神経フィラメントが挙げられ、グリアに関して、グリア線維性酸性タンパク質(GFAP)、ガラクトセレブロシド等が挙げられる。
また、神経伝達物質の抗体が神経細胞を同定するのに有益である。これらの抗体として、アセチルコリン(ACh)、ドーパミン(DA)、エピネフリン(E)、ノルエピネフリン(NE)、ヒスタミン(H)、セロトニン又は5−ヒドロキシトリプタミン(5-HT)、神経ペプチド、例えば、物質P(SP)、副腎皮質刺激ホルモン(ACTH)、バソプレシン又は抗利尿ホルモン(ADH)、オキシトシン、ソマトスタチン、アンギオテンシンII、ニューロテンシン、及びボンベシン、視床下部放出ホルモン、例えば、甲状腺刺激ホルモン放出ホルモン(TRH)及び黄体形成ホルモン放出ホルモン(LHRH)、胃腸ペプチド、例えば、バソアクティブ・インテスティナル・ペプチド(VIP)並びにコレシストキニン(CCK)及びCCK様ペプチド、オピオイドペプチド、例えば、β-エンドルフィンのようなエンドルフィン及びmet-エンケファリン及びleu-エンケファリンの如きエンケファリン、プロスタグランジン、アミノ酸、例えば、γ-アミノ酪酸(GABA)、グリシン、グルタメート、システイン、タウリン及びアスパルテート並びにジペプチド、例えば、カルノシンの抗体が挙げられる。
また、神経伝達物質合成酵素の抗体、例えば、GABAの合成に関与するグルタミン酸デカルボキシラーゼ(GAD)、ACh合成のためのコリンアセチルトランスフェラーゼ(CAT)、ドーパミンのためのドーパデカルボキシラーゼ(DDC)、NEのためのドーパミン−β−ヒドロキシラーゼ(DBH)、及び5-HTのためのアミノ酸デカルボキシラーゼ(AADC)が有益である。
また、神経伝達物質の失活に関与する酵素の抗体、例えば、AChを失活するアセチルコリンエステラーゼ(AChE)が有益であり得る。神経末端への神経伝達物質の再吸収に関与する酵素の抗体、例えば、DAに関してモノアミンオキシダーゼ(MAO)及びカテコール-o-メチルトランスフェラーゼ(COMT)、5-HTに関してMAO、及びGABAに関してGABAトランスフェラーゼ(GABA-T)がまた神経細胞を同定し得る。
神経細胞のその他のマーカーとして、神経伝達物質受容体、例えば、AChEニコチン受容体及びムスカリン受容体、アドレナリン作用性受容体α1、α2、β1及びβ2、ドーパミン受容体等の抗体が挙げられる。
高レベルのメラニンを含む細胞、例えば、黒質中に見られるような細胞が、メラニンの抗体を使用して同定し得る。
前駆細胞は、前駆細胞のみに見られるタンパク質に特異的に結合し、分化した細胞には結合しない抗体で同定し得る。特に、中間体フィラメントタンパク質の抗体、更に特別にラット401と称される抗体(これは神経上皮基幹細胞中に特別に見られる中間体フィラメントタンパク質であるネスチンに送られる)がある。
第一抗体に結合し、特別な酵素、同位体、粒子又は色素に接合されることによりその検出を可能にするあらゆる第二抗体又は異種抗免疫グロブリンが使用し得る。このような抗体はあらゆる動物例えば、ヤギ、ヒツジ、ウサギ、ブタ、ウマ、マウス、ラット等中で生じることができ、そして同じこのような変種のあらゆる異種動物中に生じた第一抗体に送られる。第二抗体は色素、例えば、フルオレセイン、ローダミン又はその他の色素に接合でき、125Iの如きあらゆる同位体で放射能標識でき、又はジゴキシンの如き放射能標識タンパク質に接合でき、又は顕微鏡で見られる粒子、例えば、免疫金粒子等に接合し得る。また、第二抗体は、基質と反応して目視可能又は検出可能な反応生成物を生成できる酵素、例えば、ホースラディッシュ−ペルオキシダーゼ及びアルカリ性ホスファターゼに接合し得る。特に有益な抗体は、フルオレセイン接合されたアフィニティークロマトグラフィーで純粋な(affinipure)ヤギ抗マウスIgG及びローダミン接合されたアフィニティークロマトグラフィーで純粋なヤギ抗ウサギIgGからなる第二抗体である。
細胞は、細胞が増殖し、分化でき、顕微鏡を使用して分析し得るあらゆる基質、例えば、培養プレート、ガラススライド、カバースリップ等で培養される。細胞は試験管内で6週まで、更に特別には14〜30日にわたって培養され、そしてアルコール又はアセトンの如き脱水溶液中、又は架橋アルデヒドの溶液中で定着し得る。次いで細胞は、同定すべき特別な細胞の特徴に送られる第一抗体を使用して染色され、続いて検出用の第二接合抗体と反応させられる。
移植細胞は、化学的外傷又は電気分解的外傷の結果として、神経領域の実験上の吸引の結果として、又は老化プロセスの結果として得られたものを含むあらゆる方法で得られた異常な神経学的徴候又は神経変性徴候を有するあらゆる動物に投与し得る。非ヒト動物において特に好ましい外傷は、6−ヒドロキシ−ドーパミン(6-OHDA)、1−メチル−4−フェニル−1,2,3,6−テトラヒドロピリジン(MPTP)、イボテン酸等で得られる。
免疫抑制薬は、起こり得る感染の結果として罹病率及び死亡率の或る種のリスクを有するので、親又は子孫からの組織を使用する自家移植又は異種移植のみがこれらの薬剤の不在下で行い得る。移植は両側で行うことができ、又はパーキンソン病を患う患者の場合には、最も冒された側の反対側で行い得る。手術は、特別な脳領域が特に定位のガイドで頭骨縫合に対して配置されるような様式で行われる。細胞が冒された神経領域、特に基底核、好ましくは尾状核及び果核、基底核又は黒質に送出される。細胞は、脳の周囲の領域の保全性を維持するあらゆる方法を使用して、好ましくは注射カニューレにより特別な領域に投与される。
特別な神経領域に投与された細胞は神経移植体を形成することが好ましく、この場合、細胞は隣接神経細胞と正常な神経連結体又はシナプス連結体を形成し、神経細胞の軸索のまわりに髄鞘を形成し得るグリア細胞との接触を保ち、神経細胞に栄養効果を与える。これらの移植細胞は連結体を形成する際に、それらは病気及び老化のために損傷された神経網を再度樹立する。
前駆細胞は、細胞の増殖及び分化を誘発するあらゆる成長因子又は医薬組成物を宿主に投与することにより現場で増殖し、分化するように誘発し得る。これらの成長因子として、当業界で知られているあらゆる成長因子(試験管内の増殖及び分化につき上記された成長因子を含む)、特にEGF及びTGF αが挙げられる。医薬組成物は、抑制作用を阻止し、かつ/又は前駆細胞を刺激して増殖させ、最終的に分化させるあらゆる物質を含む。成長ホルモンの投与は、注射カニューレ、成長ホルモン発現ベクターによる細胞のトランスフェクション、注射、所望の部位で物質を投与し得る定時放出装置、等を含むあらゆる方法により行い得る。医薬組成物は、注射カニューレ、注射、経口投与、定時放出装置、等を含むあらゆる方法により投与し得る。
生きている宿主中の移植片の生存は計算機体軸断層撮影(CAT又はCTスキャン)、核磁気共鳴もしくは磁気共鳴画像形成(NMRもしくはMRI)又は更に好ましくは陽電子射出断層撮影(PET)スキャンの如き種々の非観血的スキャンを使用して調べることができる。移植片生存の検死は、神経組織を除去し、冒された領域を肉眼で、又は更に好ましくは顕微鏡を使用して調べることにより行い得る。細胞は、光又は電子顕微鏡条件下で目視可能な染色剤、更に特別には神経細胞及びグリアに特異的である染色剤により染色し得る。神経細胞表面マーカーを同定するモノクローナル抗体、例えば、マウス神経細胞を同定するM6抗体が特に有益である。神経伝達物質を同定する抗体、特にγアミノ酪酸(GABA)及び物質P、並びに神経伝達物質の合成に関与する酵素、特に、グルタミン酸デカルボキシラーゼ(GAD)に関する抗体が最も好ましい。また、移植細胞は、トレーサー色素、例えば、ローダミン又はフルオレセイン標識微小球体、ファーストブルー、ビス−ベンズアミド又はレトロウイルス導入組織化学マーカー、例えば、βガラクトシダーゼを生産するlac Z遺伝子の先の取り込みにより同定し得る。
宿主の神経組織への移植片の機能的組み込みは、種々の機能を回復することに関する移植片の効果を調べること(内分泌機能、運動機能、認識機能及び感覚機能に関する試験を含むが、これらに限定されない)により評価し得る。使用し得る運動試験として、脳の変性側から離れる回転運動を定量化する試験、及び運動の遅さ、バランス、協調、運動不能又は運動の欠如、硬直及び振せんを定量化する試験が挙げられる。認識試験として、毎日のタスクを行う能力の種々の試験だけでなく、迷路パーフォーマンスを含む種々の記憶試験が挙げられる。
本発明の別の実施態様において、前駆細胞が宿主に移植され、そして(1)試験管内の増殖及び分化、その後の移植、(2)試験管内の増殖、移植、その後の生体内の更なる増殖及び分化、又は(3)試験管内の増殖、移植及び生体内の分化のいずれかによりその宿主中で増殖し、かつ/又は分化するように誘発される。また、細胞は、それらの増殖及び分化を誘発する成長因子又は医薬組成物による誘発により現場で増殖し、分化するように誘発し得る。それ故、この後者の方法は非外科的であり、こうして物理的介入及び異種細胞に対する免疫反応と関連する問題を回避する。あらゆる成長因子、特にEGF、TGF α、aFGF、bFGF及びNGFが使用し得る。これらの成長因子は当業界で知られているあらゆる方法で投与でき、この場合、因子は血液脳関門(BBB)を通過してもよく、またバイパスしてもよい。
因子に血液脳関門を通過させる方法として、因子のサイズを最小にすること、又は更に容易に通過し得る疎水性因子を用意することが挙げられる。
血液脳関門をバイパスする方法として、細胞それ自体が因子を生産するように成長因子をコードする遺伝子を含む発現ベクターによる前駆細胞のトランスフェクションが挙げられる。細胞は当業界で知られているあらゆる手段により移入でき、これらの手段として、プロトプラスト融合、電気穿孔法、リポフェクション(lipofection)等によるCaPO4トランスフェクション、DEAE-デキストラントランスフェクション、ポリブレントランスフェクションが挙げられる。
当業界で知られているあらゆる発現ベクターは、それが細胞中で活性であるプロモーター、並びに適当な終止信号及びポリアデニル化信号を有する限り、成長因子を発現するのに使用し得る。これらの発現ベクターとして、pSC11を含む組換えワクチンウイルスベクター、又は種々のウイルス、例えば、シミアンウイルス(SV40、即ち、pSV2-dhfr、pSV2neo、pko-neo、pSV2gpt、pSVT7及びpBABY)、ラウス肉腫ウイルス(RSV、即ち、pRSVneo)、マウス乳癌ウイルス(MMTV、即ち、pMSG)、アデノウイルス(pMT2)、単純ヘルペスウイルス(HSV、即ち、pTK2及びpHyg)、ウシパピローマウイルス(BPV、即ち、pdBPV及びpBV-1MTHA)、エプスタイン−バールウイルス(EBV、即ち、p205及びpHEBo)に由来するベクター又は当業界で知られているあらゆるその他の真核生物の発現ベクターが挙げられる。
前駆細胞の増殖、成長又は分化を誘発することが知られているあらゆる成長因子が前駆細胞中の発現に使用でき、NGF、FGF、EGF、TGF α等が挙げられる。また、細胞はあらゆる成長因子、例えば、bFGF、NGF、EGF等の受容体で形質転換されてもよい。加えて、細胞は、上記の神経伝達物質又は神経伝達物質合成酵素をコードするあらゆる遺伝子又は宿主中の発現が所望されるその他の遺伝子で移入されてもよい。
成長因子を移植の領域に与えるその他の方法として、周囲の細胞への因子の注入を与えることができるあらゆる装置の移植片に接近した脳への移植が挙げられる。
前駆細胞は、細胞を特別な低温保存剤を含む等張溶液、好ましくは細胞培地中で懸濁させることにより、当業界で知られているあらゆる方法により低温保存し得る。このような低温保存剤として、ジメチルスルホキシド(DMSO)、グリセロール等が挙げられる。これらの低温保存剤は、5〜15%、好ましくは8〜10%の濃度で使用される。細胞は-10℃〜-150℃、好ましくは-20℃〜-100℃、更に好ましくは-70℃〜-80℃に次第に凍結される。
試験管内で培養された前駆細胞は、潜在的な神経治療組成物のスクリーニングに使用し得る。これらの組成物は種々の投薬量で培養中の細胞に適当でき、細胞の応答が種々の期間にわたって監視される。細胞の物理的特性が細胞及び神経突起の成長を顕微鏡で観察することにより分析し得る。新たなレベル又は増大されたレベルのタンパク質、例えば、酵素、受容体及びその他の細胞表面分子、又は神経伝達物質、アミノ酸、神経ペプチド及び生体アミンの発現の誘発が、このような分子のレベルの変化を同定し得る当業界で知られているあらゆる技術で分析し得る。これらの技術として、このような分子の抗体を使用する免疫組織化学、又は生化学分析が挙げられる。このような生化学分析として、タンパク質アッセイ、酵素アッセイ、受容体結合アッセイ、酵素結合免疫吸着検定法(ELISA)、電気泳動分析、高速液体クロマトグラフィー(HPLC)による分析、ウェスタンブロット、及びラジオイムノアッセイ(RIA)が挙げられる。ノーザンブロットの如き核酸分析が、これらの分子、又はこれらの分子を合成する酵素をコードするmRNAのレベルを調べるのに使用し得る。また、これらの医薬組成物で処理された細胞が動物に移植でき、それらの生存、神経連結体を形成する能力、並びに生化学的特徴及び免疫学的特徴が上記のようにして調べられる。
以下の実施例は、本発明の好ましい実施態様を更に詳しく説明するために示される。しかしながら、それらは請求の範囲により特定される本発明の範囲を限定するものと見なされるべきではない。
実施例1
生後14日のCD1シロネズミの胚胎(チャールズ・リバー(Charles River))を断頭し、滅菌操作を使用して脳及び線条を除去した。組織をダルベッコの改良イーグル培地(DMEM)とF-12栄養源(ギブコ(Gibco))1:1混合物を含む無血清培地に火仕上げパスツールピペットで機械的に単離した。単離された細胞を800rpmで5分間遠心分離し、上澄みを吸引し、細胞をカウント用のDMEM/F-12培地中で再度懸濁させた。

幼若体及び成体のマウスの脳組織からの脳組織を除去し、500μmの部分に切開し、1.33mg/mlのトリプシン、0.67mg/mlのヒアルロニダーゼ、及び0.2mg/mlのキヌレン酸を含む低カルシウムの酸素化された人工の脳骨髄液(低Ca++aCSF)に直ちに移した。組織をこの溶液中で32℃〜35℃で90分間攪拌した。aCSFを注ぎ出し、5分間で新しい酸素化aCSFと交換した。組織を、0.7mg/mlのオボムコイドを含むDMEM/F-12/10%のホルモン溶液に移し、火仕上げパスツールピペットですり砕いた。細胞を400rpmで5分間遠心分離し、上澄みを吸引し、ペレットにした細胞をDMEM/F-12/10%のホルモン混合物中で再度懸濁させた。
実施例3
実施例1及び2のようにして調製した単離された細胞を96ウェル(100μl/ウェル)組織培養プレート中で約1個の細胞/ウェルに希釈してEGF誘発増殖のクローン性質を調べた。ウェル中の単細胞の存在を位相差顕微鏡で確認した。
実施例4
実施例1のようにして調製した2500の細胞/cm2を24ウェルのヌンクロン(Nunclon)(0.5ml/ウェル)培養皿中のポリ−L−オルニチン被覆(15μg/ml;シグマ)ガラスカバースリップに塗布した。培地は、グルコース(0.6%)、グルタミン(2μM)、重炭酸ナトリウム(3ミリモル)、及びHEPES(4−[2−ヒドロキシエチル]−1−ピペラジンエタンスルホン酸)緩衝液(5ミリモル)(グルタミン[ギブコ]を除いて全てシグマから入手した)を含むDMEM/F-12(1:1)を含む無血清培地であった。インシュリン(25μg/ml)、トランスフェリン(100μg/ml)、プロゲステロン(20nM)、プトレシン(60μM)、及び塩化セレン(30nM)を含む特定のホルモンミックス及び塩混合物(シグマ)を血清に代えて使用した。培養液は16〜20ng/mlのEGF(マウスの下顎腺から精製した、コラボラチブ・リサーチ(Collaborative Research))又はTGF α(ヒト組換え体、ギブコ)と一緒に上記培地を含んでいた。試験管内で10〜14日後に、培地(DMEMのみ+ホルモン混合物)及び成長因子を交換した。この培地の交換を2〜4日毎に繰り返した。試験管内で5日目の生存細胞の数を、カバースリップを0.4%のトリパンブルー(ギブコ)中で2分間インキュベートし、食塩加リン酸緩衝液(PBS、pH7.3)で洗浄し、ニコン・ジアフォット(Diaphot)倒立顕微鏡で色素を排除した細胞の数をカウントすることにより測定した。
実施例5
実施例1及び2のようにして調製した単離されたマウスの脳細胞(1 x 105個の細胞/ml)を、20ng/mlのEGF又はTGF αと一緒に実施例4に記載した培地中に懸濁させた。細胞をT25培養フラスコ中で接種し、湿度100%、95%の空気/5%のCO2の37℃のインキュベーター中に収容した。細胞は3〜4日以内に増殖し始め、基質の欠如のためにフラスコの床から隆起し、懸濁状態で増殖し続けて未分化細胞の中空球体を形成した。試験管内で6〜7日後に、増殖クラスター(神経球体)を2〜4日毎に穏やかな遠心分離及び上記添加剤を含むDMEM中の再懸濁により飼育した。
実施例6
試験管内で6〜7日後に、実施例5からの神経球体中の個々の細胞を、神経球体を火仕上げパスツールピペットですり砕くことにより分離した。単離された神経球体からの単細胞を、組織培養フラスコ中で20ng/mlのEGFと一緒にDMEM/F-12/10%のホルモン混合物中に懸濁させた。単離された細胞の一部が増殖し始め、主として未分化細胞を含む新しい神経球体を形成した。フラスコを良く振とうし、神経球体をフラスコの底隅部に沈降させた。次いで神経球体を50mlの遠心分離管に移し、300rpmで5分間遠心分離した。培地を吸引して除き、神経球体を、EGFを含む培地1ml中に再度懸濁させた。細胞を火仕上げパスツールピペットで単離し、40回すり砕いた。細胞20μlをカウント用に除去し、1:2に希釈したトリパンブルー20μlに添加した。細胞をカウントし、50,000個の細胞/mlで再度塗布した。細胞を神経球体の単離により増殖を再開しないで20ng/mlのEGF又はTGF αの存在下で培養フラスコ中に保つことにより、又はEGFもしくはTGF αの継続した存在下でポリ−オルニチンに塗布することにより細胞の分化を誘発した。
実施例7
間接免疫細胞化学を、試験管内でガラスカバースリップで14〜30日培養された実施例1及び2のようにして調製された細胞で行った。抗NSE(又は抗ネスチン)及び抗GFAP免疫細胞化学に関して、細胞を夫々PBS中4%のパラホルムアルデヒド及び95%のエタノール/5%の酢酸で定着した。30分間の定着期間後に、カバースリップをPBS(pH7.3)中で3回(毎回10分間)洗浄し、次いでPBS/10%の正常なヤギ血清/0.3%のトリトン(Triton)-X-100中の一次抗血清(NSE 1:300、ネスチン1:1500又はGFAP 1:100)中で37℃で2時間インキュベートした。カバースリップをPBS中で3回(毎回10分間)洗浄し、第二抗体(抗NSE又は抗ネスチンに関してヤギ抗ウサギローダミン及び抗GFAPに関してヤギ抗マウス−フルオレセイン、両方とも1:50)と共に37℃で30分間インキュベートした。次いでカバースリップをPBS中で3回(毎回10分間)洗浄し、水ですすぎ、ガラススライドに塗布し、フルオレセイン接合抗体に使用するのに好ましい封入剤であるフルオルセーブ(Fluorsave)を使用してカバースリップした。蛍光を検出し、ニコン・オプチフォット光学顕微鏡で撮影した。
実施例8
ラットをネムブタール(25mg/kg i.p)で麻酔し、アトロピンスルフェート(2mg/kg i.p.)を注射した。動物は線条のイボテネート外傷を受けた。外傷の7日後に、動物は定位制御のもとに実施例1又は2のようにして調製された細胞の注射を受けた。マイクロインジェクターにテフロンカテーテルを取り付けた21ゲージのカニューレで外傷領域に注射をした。注射した細胞をフルオレセイン標識微小球体で標識した。動物を外傷の前後、及び移植後の種々の間隔で挙動試験を行って種々の術後の時点で移植細胞の機能性を測定し、次いで殺し、4%の緩衝パラホルムアルデヒド、0.1%のグルタルアルデヒド及び5%の蔗糖溶液で4℃で心臓を通して(transcardially)潅流した。脳を液体窒素中で凍結し、使用するまで-20℃で貯蔵した。脳の部分をクリオスタット上で26μmに細断し、4%のパラホルムアルデヒド中で定着し、マウス神経細胞の染色用のM6モノクローナル抗体を使用して染色し、次いで第二の抗ラット・フルオレセイン接合抗体と反応させた。神経及びグリアの表現型をNSE及びGFAPの抗体による細胞の二重標識により同定した。
実施例9
実施例2に従って調製した細胞を試験管内でガラスカバースリップで14〜30日間培養し、次いでPBS中4%のパラホルムアルデヒドで定着した。30分の定着期間後に、カバースリップをPBS(pH7.3)中で3回(毎回10分間)洗浄し、次いで一次抗血清(10%の正常なヤギ血清/0.3%のトリトン-X-100を含むPBS中、抗GABA 1:3000、抗グルタミンデカルボキシレート1:500、及び抗物質P1:100)中で37℃で2時間インキュベートした。カバースリップをPBS中で3回(毎回10分間)洗浄し、第二抗体(1:50でフルオレセイン又はローダミンに接合されたヤギ抗ウサギ又はヤギ抗ヒツジ)と共に37℃で30分間インキュベートした。次いでカバースリップをPBS中で3回(毎回10分間)洗浄し、水ですすぎ、ガラススライドに塗布し、封入剤であるフルオルセーブを使用してカバースリップした。蛍光を検出し、ニコン・オプチフォット光学顕微鏡で撮影した。
実施例10
実施例2のようにして調製した細胞を300rpmで3分間遠心分離し、培地を吸引し、次いで細胞を、10%のグリセロールを含む温かいDMEM/F-12培地中に約1 x 105個の細胞/mlの濃度で再度懸濁させ、開放したリドを有する前冷却したスタイロフォームボックス中で2時間にわたって-20℃に徐々に凍結した。一旦凍結すると、細胞を-80℃に移した。クリオバイアルを37℃の水浴に入れることにより細胞を解凍し、穏やかな攪拌により混合し、等容積の温めた培地を15分間にわたって5分毎に細胞に添加し、次いで容積を10mlにした。次いで細胞を300rpmで3分間遠心分離し、上澄みを捨てた。細胞10μlを除去して生存度を測定し、細胞10μlを除去し、トリパンブルーで染色し、血球計数器でカウントした。細胞を100,000個の細胞/フラスコでDMEM/F-12/10%のホルモン溶液中で再度懸濁させ、T25培養フラスコ中で37℃で3〜4日間培養した。
実施例11
ルーチンの吸引流産に従って細胞を生後8週のヒトの胎児からの腹側中脳組織から得、これを滅菌回収装置に回収する。組織の2 x 4 x 1mmの片を切開し、実施例1のようにして単離する。次いで細胞を実施例5のようにして培養する。
実施例12
実施例11のようにして調製した細胞を神経変性疾患を有する血液−群のつり合った宿主への神経移植に使用する。手術を、BRW計算機断層撮影(CT)定位ガイドを使用して行う。患者にミダゾラムを静脈内投与して補給して局所麻酔を施した。患者はCTスキャンを受けて移植片を受ける領域の座標を確立する。注射カニューレは19ゲージの内部スタイレットを有する17ゲージのステンレス鋼の外部カニューレからなる。これを脳中に正確な座標に挿入し、次いで除去し、組織懸濁液30μlを予め装填した19ゲージの注入カニューレと交換する。カニューレを抜き取る際に、細胞を3μl/分の流量で徐々に注入する。多重定位ニードル通過を関係する領域に約4mm離して行う。患者を術後に出血又は水腫につきCTスキャンにより調べる。神経学上の評価を種々の術後の間隔でPETスキャンで行って移植細胞の代謝活性を測定する。
実施例13
神経組織をヒトの患者のバイオプシーにより脳室領域から得、実施例5のようにして培養する。
実施例14
実施例1又は2のようにして増殖した細胞を、bFGF受容体又はNGF受容体の遺伝子を含む発現ベクターで移入する。遺伝子を含むベクターDNAを40μg/mlの濃度まで0.1X TE(1ミリモルのトリス(Tris)pH8.0、0.1ミリモルのEDTA)で希釈する。DNA 220μlを使い捨ての無菌の5mlのプラスチック管中の2X HBS(280ミリモルのNaCl、10ミリモルのKCl、1.5ミリモルのNa2HPO4・2H2O、12ミリモルのデキストロース、50ミリモルのHEPES)250μlに添加する。2MのCaCl231μlを徐々に添加し、その混合物を室温で30分間インキュベートする。この30分間のインキュベーション中に、細胞を4℃で800gで5分間遠心分離する。細胞を氷で冷却したPBS20倍容中に懸濁させ、1 x 107個の細胞のアリコートに分け、これらを再度遠心分離する。細胞の夫々のアリコートをDNA-CaCl2懸濁液1ml中に再度懸濁させ、室温で20分間インキュベートする。次いで細胞を増殖培地中で希釈し、37℃で5%〜7%のCO2中で6〜24時間インキュベートする。細胞を再度遠心分離し、PBSで洗浄し、48時間にわたって増殖培地10mlに戻す。
実施例15
実施例14のようにして移入した細胞を実施例12のようにしてヒトの患者に移植する。
本発明が種々の特定の物質、操作及び実施例を参考にして本明細書に記載され、説明されたが、本発明はその目的のために選ばれた物質の特別な物質組み合わせ、及び操作に限定されないことが理解される。このような詳細の多数の変化が、当業者により認められるように、当然に含まれ得る。
Claims (2)
- 懸濁細胞培養液中の神経幹細胞の試験管内増殖方法であって、前記方法が、以下の工程:
(a)EGF及びTGFαからなる群から選択される一種以上の成長因子を含む初期培養培地を調製し又は提供する工程;
(b)工程(a)の培養培地に、哺乳類組織に由来し、無制限に分裂することができ、及び、神経細胞及びグリアに分化し得る娘細胞を生産できる少なくとも一種の神経幹細胞を含む細胞を懸濁することによって懸濁細胞培養液を調製する工程;
(c)前記培養培地中の神経幹細胞を増殖して神経細胞及びグリアに分化し得る神経幹細胞の娘細胞を含む神経球体を生産する工程;
(d)前記神経球体を個々の神経幹細胞の娘細胞に分離し、及び、前記個々の神経幹細胞の娘細胞を、EGF及びTGFαからなる群から選択される一種以上の成長因子を含む新しい培養培地に移して、前記神経幹細胞の娘細胞を増殖し、及び、神経球体をさらに生産する工程、
を含むこと特徴とする、方法。 - 請求項1に記載の方法によって調製される神経球体に由来する、試験管内で分化した神経細胞を製造する方法であって、
(e)前記神経球体を個々の神経幹細胞の娘細胞に分離する工程、及び
(f1)前記神経幹細胞の娘細胞を、ポリオルニチンに塗布する工程、又は、
(f2)前記神経幹細胞の娘細胞を、増殖を再開しないで懸濁液内で分化できるようにする工程、
によって前記娘細胞の分化を誘発することを含む方法。
