Abstract
Description
本願は、「Engineering H−NOX Proteins for Therapeutic Nitric Oxide and Oxygen Delivery」(UC Case No.B06−084)という表題の、Michael A.Marletta,Stephen P.L.Cary,Elizabeth M.Boonによる、2006年5月22日に出願された米国仮特許出願第60/921,505号の利益を請求する。この仮特許は、2006年5月22日に出願された米国特許出願第11/440,588号から、2007年5月1日における仮特許出願に変更され、その開示は、その全体が本明細書中に参考として援用される。
本研究は、助成金番号DE−AC03−76SFに支援された。米国政府は、本出願にかかる任意の特許に権利を有し得る。
本出願はH−NOX蛋白、及び酸素を送達するためにそれらを使用する方法に関する。H−NOX蛋白はヒトに対して、そして獣医科用途に関しては、動物に対してO2を送達するための新しい治療ツールを与える。
現在の血液銀行システムは固有の危険性及び重篤な制限事項を有する。血液型判別の誤り、免疫原性、細菌物質の伝播、及びHIV−1及び肝炎等のウィルス感染が輸液患者に対して致命的な危険をもたらしている。更に又、ドナーの限定された入手性、特定の血液型の必要性、赤血球の短いシェルフライフ、及び冷蔵保存の必要性はすべて、患者に対する輸液の使用し易さを制限している。安定な血液代替物の開発は現在の血液銀行システムの危険性を排除し、そして大部分の環境において患者に対する輸液の使用し易さを向上させると考えられる。即ち、失血又は低酸素に起因する症状を軽減するための臓器及び組織への酸素(O2)の送達は主要な治療上の目標である。
恐らくは糸球体におけるNOの枯渇に起因して腎毒性をもたらす場合があり、これにより収縮及びその後の機能不全をもたらす。
本発明は野生型及び変異体のH−NOX蛋白がヘモグロビンよりも遥かに低値のNO反応性を有し、そしてこのため望ましいO2担体であることを意外にも発見したことに部分的には基づいている。所望により、H−NOX蛋白内に変異を導入してそのO2及びNOリガンド結合を改変することによりO2担体としてのH−NOX蛋白の使用を更に最適化することができる。
H−NOX蛋白がヘモグロビンよりもはるかに低値のNO反応性を有するという意外な発見に部分的には基づいている。この固有の低NO反応性(及び高いNO安定性)は、内因性NOによるH−NOX蛋白の不活性化の確率がより低値であり、そしてH−NOX蛋白による内因性NOの捕獲の確率がより低値であるという理由から、野生型及び変異体のH−NOX蛋白を血液代替物として望ましいものにしている。重要な点は、一部のH−NOX蛋白における遠位ポケットチロシンの存在(Pellicena,P.等(2004年8月31日)、“Crystal Structure of An Oxygen−Binding Heme Domain Related to Soluble Guanylate Cyclase,”Proc Natl.Acad Sci USA 101(35):12854−12859)は望ましくない高値のNO反応性を示唆しており、血液代替物としての使用を禁忌としている。例えば類推すれば、構造的に類似の遠位ポケットチロシンを有するMycobacterium tuberculosisのヘモグロビン蛋白はNOと極めて急速に反応し、そして感染宿主により生成された防御性NOを効果的に捕獲してこれを回避するためにMycobacteriumにより使用される(Ouellet,H.等(2002年4月30日)、“Trancated Hemoglobin HbN Protects Mycobacterium Bovis From Nitric Oxide,”Proc.Natl.Acad.Sci.USA 99(9):5902−5907)。しかしながら、本発明者等は意外にも、H−NOX蛋白は実際には、血液代替物としてのその使用を可能にするような、ヘモグロビンのものよりも遙かに低値のNO反応性を有することを発見した。
H−NOX蛋白ファミリーの概説

何れかの属又は種に由来するH−NOX蛋白を本明細書に記載する組成物、キット及び方法において使用できる。種々の実施形態において、H−NOX蛋白は哺乳類(例えば霊長類(例えば、ヒト、サル、ゴリラ、類人猿、キツネザル等)、ウシ、ウマ、ブタ、イヌ、又はネコ)、昆虫、酵母、又は細菌由来の蛋白であるか、又はそのような蛋白由来である。例示される哺乳類H−NOX蛋白は野生型のヒト又はラットの可溶性グアニレートサイクラーゼ(例えばβ1サブユニット)を包含する。H−NOX蛋白の例は野生型哺乳類H−NOX蛋白、例えばH.sapiens、M.musculus、C.familiaris、B.taurus及びR.norvegicus;及び野生型非哺乳類脊椎動物H−NOX蛋白、例えばX.laevis、O.latipes、O.curivatus及びF.rubripesを包含する。非哺乳類野生型NO結合H−NOX蛋白の例はD.melanogaster、A.gambiae及びM.sextaの野生型H−NOX蛋白を包含し;非哺乳類野生型O2結合H−NOX蛋白の例はC.elegansのgcy−31、gcy−32、gcy−33、gcy−34、gcy−35、gcy−36、及びgcy−37;D.melanogasterCG14885、CG14886、及びCG4154;及びM.sextaベータ3の野生型H−NOX蛋白を包含し;原核生物野生型H−NOX蛋白の例はT.tengcongensis、V.cholera、V.fischerii、N.punctiforme、D.desulfuricans、L.pneumophila1、L.pneumophila2、及びC.acetobutylicumを包含する。
本明細書において考察する通り、H−NOX蛋白は対応する野生型蛋白と比較してそのO2解離定数、酸素に関するkoff、ヘム自動酸化の速度、NO反応性、NO安定性又はこれらの2つ以上の何れかの組み合わせ改変する変異のような変異1つ以上を含有してよい。操作されたH−NOX蛋白のパネルは、本明細書に記載した、及び追加的には当該分野で知られた手法を用いて本明細書の教示を鑑みながら、ランダム変異誘発、その後、必要な、又は望ましい解離定数、解離速度、NO反応性、安定性、生理学的適合性、又はこれらの2つ以上の何れかの組み合わせに関して実験的にスクリーニングすることにより、形成してよい。或いは、変異誘発は、特定の領域、又はH−NOX蛋白の実験的に決定された、又は予測された三次元構造から明らかになる遠位ポケット残基のような残基(本明細書の図1A;及び例えば、特に野生型及び変異体のH−NOX蛋白の配列に関して参照により全体が本明細書に組み込まれるBoon,E.M.等(2005)、“Molecular Basis For NO Selectivity in Soluble Guanylate Cyclase,”Nature Chemical Biology 1:53−59を参照できる)又は配列アライメントから識別される進化的に保存されている残基(本明細書の図2〜4;及び例えば、特に野生型及び変異体のH−NOX蛋白の配列に関して参照により全体が本明細書に組み込まれるBoon,E.M.等(2005)、“Molecular Basis For NO Selectivity in Soluble Guanylate Cyclase,”Nature Chemical Biology 1:53−59を参照できる)に対して選択的にターゲティングできる。

本明細書に記載する通り、広範なNO及びO2解離定数、O2に関するkoff、NO反応性及び安定性を提供する多数の多様なH−NOX変異体蛋白が形成されている。作動可能な血液ガス担体を提供するために、H−NOX蛋白はヘモグロビンのような内因性O2担体を機能的に置き換えるか、補充するために使用してよい。従って、一部の実施形態においては、H−NOX蛋白はヘモグロビンのような内因性O2担体と比較して、同様の、又は向上したO2会合速度、O2解離速度、O2結合に関する解離定数、NO安定性、NO反応性、自動酸化速度、血漿中保持時間、又はこれらの2つ以上の何れかの組み合わせを有する。

本発明は更に本明細書に記載する変異体H−NOX蛋白の何れかをコードする核酸を特徴とする。本明細書においては、「核酸」とは、1本鎖又は2本鎖の何れかの形態における2つ以上のデオキシリボヌクレオチド及び/又はリボヌクレオチドを指し、そして特段の記載が無い限り、天然に存在するヌクレオチドと同様の態様において核酸にハイブリダイズする天然に存在するヌクレオチドの既知類縁体を包含する。一部の実施形態においては核酸は組み換え核酸である。「組み換え核酸」とは、目的の核酸の由来元の生物の天然に存在するゲノム内において目的の核酸にフランキングしている核酸(例えば遺伝子)1つ以上を含まない目的の核酸を意味する。一部の実施形態においてH−NOX核酸は、組み換え核酸がH−NOX蛋白(例えばH−NOX蛋白に由来する別のドメインを有するか有さないH−NOXドメイン)及びヒト血清アルブミンのような別の蛋白の全て又は部分を包含する融合蛋白をコードするように別の蛋白の全て又は一部分をコードする別の核酸に作動可能に連結している。従って用語は、例えばベクター内に、自律複製プラスミド又はウィルス内に、又は原核生物又は親核生物のゲノムDNA内に組み込まれた、又は他の配列から独立して別個の分子(例えばPCR又は制限エンドヌクレアーゼ消化により作成されたcDNA、ゲノムDNAフラグメント、又はcDNAフラグメント)として存在する組み換えDNAを包含する。
本明細書に記載する野生型又は変異体H−NOX蛋白は医薬組成物又は非医薬組成物の製剤のために使用してよい。後に考察する通りこれらの製剤は種々の治療及び産業上の用途において有用である。
当該分野で知られた何れかの適当な担体を本発明の医薬組成物において使用してよいが、担体の型は投与の様式により変動することになる。組成物は例えば静脈内、動脈内、膀胱内、吸入、腹腔内、肺内、筋肉内、皮下、気管内、経粘膜、眼内、髄腔内、又は経皮投与を包含する何れかの適切な投与方法のために製剤できる。皮下注射のような非経腸投与のためには、担体は例えば水、食塩水、アルコール、油脂、ワックス、又は緩衝液を包含してよい。経口投与のためには上記した担体の何れか、又は固体担体、例えばマンニトール、乳糖、澱粉、ステアリン酸マグネシウム、サッカリンナトリウム、タルク、セルロース、グルコース、スクロース、又は炭酸マグネシウムを使用してよい。生体分解性の微小球(例えばポリラクテートポリグリコレート)もまた担体として使用してよい。
本明細書に記載する野生型又は変異体のH−NOX蛋白の何れか(例えば単離又は精製されたH−NOX蛋白)又は医薬組成物を治療用途に使用してよい。このような用途のために、特定のH−NOX蛋白を、治療すべき特定の適応症に対して所望のO2会合速度、O2解離速度、O2結合に関する解離定数、NO安定性、NO反応性、自動酸化速度、血漿中保持時間、又はこれらの2つ以上の何れかの組み合わせに基づいて選択することができる。H−NOX蛋白は心臓血管疾患、神経学的疾患、腫瘍低酸素、失血、又は創傷を治療するために使用できる。例えばO2結合H−NOX蛋白は赤血球又は血漿エキスパンダーが現在使用されている大部分の状況において使用できる。特定すれば、H−NOX蛋白は外傷(例えば戦地、災害救援、又は事故)、出血、出血性ショック、外科処置(例えば腹部動脈瘤手術、整形外科的手術、例えば腰骨置換手術、又は大量の失血をもたらす何れかの他の手術)、血液希釈、及び血液延長使用(例えば補給自己献血)、及び血液のぼる量が損失しているか、O2運搬能力が低減している何れかの他の状況の治療のために赤血球代替物として使用できる。創傷修復用途の例は、放射線後の創傷の修復(例えば高圧酸素作用)、術後修復、糖尿病性潰瘍修復、及び熱傷を包含する。
本明細書に記載したH−NOX蛋白及び組成物は又、多くのインビトロ又は産業上の用途のために使用できる(特にインビトロ又は産業上の用途に関して参照により全体が本明細書に組み込まれる米国特許6,455,676参照)。このような用途に対して、特定のH−NOX蛋白は、上記した用途のために望ましいO2会合速度、O2解離速度、O2結合に関する解離定数、NO安定性、NO反応性、自動酸化速度、半減期、又はこれらの2つ以上の何れかの組み合わせに基づいて選択することができる。産業上の用途の種々の実施形態においてH−NOX蛋白はヘムに結合できるアポ蛋白、又は結合したヘムを有するホロ蛋白である。
本明細書に記載したH−NOX蛋白の何れか及び適当なパッケージを包含する製造物品及びキットも提供される。一部の実施形態においては、本発明は(i)H−NOX蛋白(例えば本明細書に記載した野生型又は変異体H−NOX蛋白又は本明細書に記載したその製剤)及び(ii)個体にO2を送達するためにキットを使用するための説明書を伴ったキットを包含する。種々の実施形態において、本発明は(i)H−NOX蛋白(例えば本明細書に記載した野生型又は変異体H−NOX蛋白又は本明細書に記載したその製剤)及び(ii)本明細書に記載した産業上の使用の何れかのためにキットを使用する(例えばレファレンス標準物質としてのそのようなレファレンス標準物質を必要とする分析用計器類のためのH−NOX蛋白の使用、インビトロにおけるO2レベルを維持または上昇させることによる細胞培養物中の細胞生育の増強、溶液へのO2の添加、又は溶液からのO2の除去)ための説明書を伴ったキットを特徴とする。
本発明は又本明細書に記載した変異体H−NOX蛋白の何れかの産生のための方法を提供する。一部の実施形態においては、方法は変異体H−NOX蛋白の産生に適する条件下に変異体H−NOX蛋白をコードする核酸を有する細胞を培養することを包含する。種々の実施形態において、変異体H−NOXはまた精製される(例えば細胞又は培養用培地からのH−NOX蛋白の精製)。

速度論的KD:koffのkonに対する比
速度論的KD値は本質的に、特にO2会合速度、O2解離速度、O2結合に関する解離定数、自動酸化速度、及びNO解離速度に関して参照により全体が本明細書に組み込まれるBoon,E.M.等(2005)、“Molecular Basis For NO Selectivity in Soluble Guanylate Cyclase,”Nature Chemical Biology1:53−59による記載の通り野生型及び変異体のH−NOX蛋白に関して測定した。
ヘムへのO2会合は20℃においてフラッシュフォトリシスを用いて測定した。極めて高速の重複組み換え動態の結果としてFeIIO2複合体をフラッシュオフすることは不可能であり;即ちFeIIO2複合体を560nmにおけるレーザー(Hewlett−Packard,Palo Alto,CA)を用いたフラッシュフォトリシスに付すことにより、5−配位のFeII中間体を生成し、これに分子状O2の結合を種々の波長において行った。蛋白試料は10mMジチオナイトを用いた嫌気的還元、その後のPD−10カラム(Millipore,Inc.,Billerica,MA)上の脱塩により行った。次に試料をサイズ100μL〜1mL及びパス長1cmの制御大気圧石英キュベット中50mMTEA、50mMNaCl、pH7.5緩衝液中で20μMヘムまで希釈した。COガスを10分間このキュベットのヘッドスペース上に流すことによりFeIICO複合体を形成し、その形成はUV可視光スペクトル分析(ソレー最大423nm)により確認した。次にこの試料は、なおCOガス1気圧下にある時点でフラッシュフォトリシス後にCO再結合動態を測定するために使用するか、又は、開封して空気中30分間攪拌することにより緩衝液に完全に酸素付与した後にフラッシュフォトリシスを行うことによりO2再結合事象を観察した。ヘムへのO2会合は時間に対して多数の波長においてモニタリングした。これらの軌跡をIgor Proソフトウエア(Wavemetrics,Inc.,Oswego,OR;最新2005バージョン)を用いて片指数にフィットさせた。この速度は観察波長とは独立していたが、O2濃度には依存しており、UV可視光スペクトル分析を常時使用することにより全ての複合体及び中間体を確認した(Cary 3K,Varian,Inc.,Palo Alto,CA)。一過性吸収データを特に計器類に関して参照により全体が本明細書に組み込まれるDmochowski,I.J.等(2000年8月31日)、“Enantioimeric Discrimination of Ru−Substrates by Cytochrome P450cam,”J Inorg Biochem.81(3):221−228に記載の機材を用いて収集した。機器は応答時間20nsであり、データは200メガサンプルs−1においてデジタル化した。
koffを測定するために、嫌気的50mMTEA、50mMNaCl、pH7.5緩衝液中に希釈した蛋白のFeIIO2複合体(5μMヘム)を種々の濃度のジチオナイト及び/又は飽和COガスを含有する同じ緩衝液(嫌気的)等容量と急速に混合した。データは20℃にセットしたNeslab RTE−100恒温槽を装着したHI−TECH Scientific SF−61ストップドフロー分光光度計上で獲得した(TGK Scientific LTD.,Bradford On Avon,United Kingdom)。ヘムからのO2の解離は437nm、即ちFeII−FeII−O2差スペクトル、又は425nm、即ちFeII−FeII−CO差スペクトルにおける吸光度の増分としてモニタリングした。最終軌跡を機器の一部であるソフトウエアを用いながら片指数にフィットさせた。各実験は最低6回実施し、得られた速度を平均した。測定した解離速度はジチオナイト濃度(100、50、25、10及び2.5mMジチオナイトを試験した)から独立しており、そして10mMジチオナイト存在下及び非存在下の両方において還元された物質種のためのトラップとしての飽和COからも独立していた。
速度論的KDは上記したkoffおよびkonの測定を用いてkonに対するkoffの比を計算することにより求める。
計算されたKD
計算されたKDを求めるために、上記の通り得られたkoff及び速度論的KDの値をグラフ化した。koff及び速度論的KDの間の直線関係は等式(y=mx+b)により定義した。次にkoff値を直線に外挿し、Excel:MAC2004(Microsoft,Redmond,WA)を用いながら計算されたKDを誘導した。測定されたkoffがない場合は、この外挿はKDにkoffを関連付ける方法を与える。
自動酸化速度を測定するために、蛋白試料を嫌気的に還元し、次に好気的な50mMTEA、50mMNaCl、pH7.5緩衝液中5μMヘムとなるまで希釈した。次にこれらの試料を37℃にセットしたNeslabRTE−100恒温槽を装着したCary3E分光光度計中でインキュベートし、周期的にスキャンした(Cary 3E、Varian,Inc.,Palo Alto,CA)。自動酸化速度は時間に対してプロットし、Excel:MAC2004(Microsoft,Redmond,WA)を用いながらフィットさせたFeIII−FeII差スペクトルにおける最大と最小の間及びの差から求めた。
NO反応性は緩衝液A中に2μMに産生された精製蛋白(TtWTHNOX、TtY140HHNOX及びHomo sapiensヘモグロビン(HsHb))及びBufferA(BufferA:50mM Hepes、pH7.5、50mM NaCl)中200μMに産生されたNOを用いて測定した。データは20℃にセットしたNeslabRTE−100恒温槽を装着したHI−TECHScientificSF−61ストップドフロー分光光度計上で獲得した(TGKScientific LTD.,Bradford On Avon,United Kingdom)。蛋白は迅速にNOと1:1比で混合し、その組み込み時間は0.00125秒とした。最大変化の波長を分光光度計の部分であるソフトウエアを用いて片指数にフィットさせ、NOによる酸化の律速段階を本質的に測定した。反応の最終生成物はHNOX蛋白については第2鉄−NOであり、HsHbについては第2鉄水和物であった
p50測定
所望により変異体又は野生型のH−NOX蛋白のp50値を、特にp50値測定に関して参照により全体が本明細書に組み込まれるGuarnone,R.等(1995年9月/10月)“Performance Characteristics of Hemox−Analyzer for Assessment of The Hemoglobin Dissociation Curve”、Haematologica80(5):426−430の記載の通り測定することができる。p50値はHemOx分析器を用いて求める。測定チャンバーは0%酸素において指導させ、そして緩徐に100%酸素となるまで漸次上昇させる。チャンバー内の酸素プローブが酸素飽和%を測定する。第2のプローブ(UV−Vis光)はヘモ蛋白(例えばヘムと複合体化しているH−NOXのような蛋白)のUV−Visスペクトルのアルファ及びベータピークに調節してある吸収の2波長を測定する。これらの吸収ピークはヘモ蛋白が酸素と結合するに従って直線的に増大する。次に未結合から100%結合までのパーセント変化を%酸素値に対してプロットすることにより曲線を作成する。p50はヘモグロビンの50%が酸素に結合する曲線上の点である。
所望により、剪断速度200/sでCPE−40コーンスピンドルとともにコーン/プレートレオメーター(DV−III型、Brookfield;Middleboro,MA)を用いながらH−NOX溶液の粘度を測定することができる。血液希釈酸素送達実験において投与した1〜4センチポアズ(cP)の粘度を有する溶液は安全であることが報告されている(特に粘度の測定に関して参照により全体が本明細書に組み込まれるWinslow,R.M.等(2004年10月)、“Comparison of PEG−Modified Albumin And Hemoglobin in Extreme Hemodilution in the Rat,”J Appl Physiol.97(4):1527−1534、米国特許6,974,795及び6,432,918)。従って、一部の実施形態においては、H−NOX蛋白溶液の年度は1〜4cPである。
