Abstract
Description
本出願は、2006年5月15日に出願された米国仮特許出願60/800,380号の優先権の利益を要求する。この出願は参照され、すべての目的のためのその全体が本明細書に組み入れられる。

本発明は一般に医薬品、磁性薬品キャリアおよび磁性微粒子に関する。本発明は、医薬品を含む磁性微粒子、並びにこれの製造方法および使用方法を提供する。
系統的な投与の後に、選択された解剖部位内に制限されて存在する能力がある医薬についての必要が認識されている。この必要に対応する既知の方法のうちの1つは、所望の解剖部位における医薬品の閉じ込めが、磁気によって援助されるように、磁気特性を医薬品に賦与することである。これは、微視的な粒子中において医薬品および磁性成分を組み合わせることによりしばしば行われる。
本発明は、微粒子の磁性キャリヤにおける、低い磁性および/または薬品保持の不利を克服し、かつ磁性医薬品の実際的な利用性を増加させることを目的とする。これは、医薬品に対して高い親和力がある物質で磁性成分をコーティングすることにより行われる。該物質は、医薬品とそのようなコーティングされた磁性成分と溶液中での相互作用により、医薬品と磁性成分とが微粒子の形態で共沈殿するような性質を有する。
ここで水性媒体中の該イオン化可能な有機化合物は、前記の電荷を有するポリマーの電荷とは反対の符号の電荷にイオン化可能であり、前記の磁性材料および有機化合物は合計で微粒子の乾燥重量の少なくとも50%を構成し、該微粒子は水中に分散可能である。典型的に、磁性微粒子の高分子電解質成分はポリマーであるが、微粒子に含まれる有機化合物はポリマーではない(非ポリマー)。好ましくは、前述の磁性材料および前述の有機化合物は合計で、微粒子の乾燥質量の少なくとも70%、あるいは少なくとも90%を構成する。

本発明は、医薬品物質、治療および診断に有用な物質、あるいは疾病予防に有用な物質、または顔料もしくは染料のような着色物質、種々の基体への印刷もしくは他の方法のイメージ形成物質などのような、ある有用性を有する有機物質(有機活性物質とも呼ばれる)と、磁性体を含有する磁性微粒子を含む組成物に関する。磁性体成分および医薬品成分または着色成分の微粒子中での組み合わせは有用である。それは微粒子への有機化合物の制御可能な堆積とその有用な積載をもたらし、磁場の印加または高周波数電磁場の印加により有機物質を調整し、粒子内に熱を発生させるか、または磁性成分の磁気特性を使用して、たとえばMRIのような磁気画像方法を使用して患者の体内における有機物質の定量と追跡を行うことができる。
別法として、微粒子形成の前にあるいはその形成の後に、粒子の医薬品、磁性、高分子電解質成分に静電気的に引きつけられる、脂質グループあるいは高分子電解質鎖のような「アンカー」基が加えられる。典型的には、分子量500−20,000のポリマー、あるいは好ましくは分子量500−50,000のポリ(エチレングリコール)が、粒子の質量の0.1%から20%、より好ましくは0.5から10%で使用される。

コンドロイチン硫酸Aの50mgが蒸留水の1mLに溶かされた。この溶液0.3mLに、68.4mg/mLの鉄を含んでいるコロイド状のγ酸化鉄が加えられ、混合物は15分間超音波で処理された。その後、pH7.4の、1.5MのNaClおよび0.2Mのヒドロキシエチルピペラジンスルホネート−Naを含んでいる溶液(10x HEPES−NS)の0.1mLが加えられた。混合物は、溶離剤として、0.15M NaCl−20 mM HEPES−Na、pH7.4を使用して、セファロースCL−4Bの上でクロマトグラフィで分離された。鉄コロイドを含んでいる、暗色の画分が採取され、0.22のμmフィルタで濾過して殺菌された。生成物は、鉄を5.6mg/mL、コンドロイチン硫酸Aを0.96mg/mL含んでいた。図3はこの例によって生成されたコロイド粒子の透過型電子顕微鏡視界から得られた粒度分布ヒストグラムを示す。
ポリエチレンイミンでコーティングされた酸化鉄磁性体コロイド
68.4mg/mLの鉄を含んでいるコロイド状のγ酸化鉄の0.3mLが、塩化水素酸でpH3.5に調節されたポリエチレンイミン(Polymine P(シグマ・ケミカル社))を44mg含んでいる水溶液の1mLに加えられた。混合物は15分間超音波で処理された。得られた溶液は、溶離剤として脱イオン水を使用し、セファロースCL−4Bの上でクロマトグラフィで分離され、0.22μmフィルタによって濾過されて殺菌され、1.54mg/mlの鉄の濃度の、ポリエチレンイミンでコーティングされた鉄コロイドを与えた。
磁性ドキソルビシン
例1により得られた、コンドロイチン硫酸でコーティングされた酸化第二鉄の20μLと、ドキソルビシン注射溶液USP(ドキソルビシンの2mg/mL)の30μLが、pH 7.4の、0.15M NaClおよび20mM HEPES−Na(Ix HEPES−NS)をさらに含んでいる水溶液、100μLの全容積中で混合された。微粒子が直ちに形成された。それらは、反応槽を磁石へ暴露することにより分離され、好適な量のIx HEPES−NS中で渦流により再懸濁された。微小球粒子は74.9%のFe2O3、16.0%のドキソルビシンを含み、残部はコンドロイチン硫酸であった。パーセンテージは乾燥質量の合計に基づく。図2は、この例によって生成された磁性ドキソルビシンの蛍光顕微鏡イメージを示す。マウス赤血球(大きな球体は直径でおよそ6nmである)は、粒径の比較評価のために加えられた。
磁性ドキソルビシン−異なる例
ド キソルビシン溶液の容積を20μLに減らして、例3の手続きが繰り返された。微粒子は、77.6%のFe2O3、13.0%のドキソルビシンを含み、残部はコンドロイチン硫酸であった。パーセンテージは乾燥質量の合計に基づく。
磁性デキスニグルジピン
例1によって得られたコンドロイチン硫酸でコーティングされた鉄コロイドの20μLが、渦流により2mg/mLのデキスニグルジピン(BYK Guldenファーマシューティカルズ)および16%のDMSOを含んでいる溶液の40μLと混合された。水と、10xHEPES−NSが、0.15M NaClおよび20mM HEPESに加えられ、100μLの最終体積とされた。2時間の後、微粒子は磁石によって分離され、Ix HEPE−NSの好適な容積中で再懸濁された。微粒子は次のものを含んでいた(乾燥質量基準):デキスニグルジピン46.1%。Fe2O3 48.3%、残部のコンドロイチン硫酸。
磁性メトトレキセート
例2のようにして得られたポリエチレンイミンでコーティングされた鉄コロイドの40μLが、メトトレキセート(アメトプテリン(Ametopterin)、(シグマ・ケミカル社))溶液(水中4mg/mL、pH 7.4)の30μLと混合された。微粒子は磁石によって分離され、Ix HEPES−NSの中で再懸濁された。微粒子は86.9mgのFe2O3および81.3mgのメトトレキセートを含んでいた。
磁性ドキソルビシンの生物活性
人間の表皮癌細胞(KB)を、37℃および5%のCO2で、10%のウシ胎児血清で補われたRPMI 1470培地において成長させた。細胞は、96−ウエル組織培養プレート中に、5,000/ウエルの濃度でおかれ、48時間、付着し順応させた。遊離のドキソルビシンおよび再懸濁された、上記の例3および4により得られた磁性ドキソルビシン微粒子は、薬/mlの0.01−10マイクログラムの濃度範囲をカバーするように、ウエルに加えられた。薬への19時間の暴露の後、細胞は72時間新鮮な培地に暴露された。また、細胞生存率は文献に記載されられるような従来のテトラゾリウム(MTT)検定を使用して分析された。図4Aに示されるように、磁性薬の細胞毒性は遊離の薬のそれに近似していた。図4Cに示されるように、コンドロイチン硫酸塩でコーティングを施した酸化第二鉄単独では細胞無毒性だった。
磁性デキスニグルジピンの生物活性
例7に記載されたように、細胞は育てられ配置された。0.03−15マイクログラム/mlのデキスニグルジピン濃度の範囲をカバーするように、遊離のデキスニグルジピン(溶液)あるいは例5のようにして得られた磁性デキスニグルジピンは、細胞へ加えられた。細胞生存率検定および他の検定条件は例7での通りだった。図4Bに示されるように、磁性デキスニグルジピンの細胞毒性は遊離の薬剤のものと同じである。
磁性メトトレキセートの生物活性
人間の肺小細胞がん細胞(NCI−H1048)は、例7に記載された検定のために調製された。細胞は、薬液の形態または、例6に記載されたようにして得られた磁性体形態のメトトレキセートに、様々な濃度(0.001−2マイクログラム/ml)で94時間暴露された。薬物曝露の後の細胞生存率はMTT検定を使用して決定された。図5に表示された結果は、磁性薬の細胞増殖抑制性の特性が遊離の薬剤のものに近似していることを示す。
硫酸デキストランでコーティングされたマグヘマイト
γ酸化鉄(マグヘマイト)は、等モルの量の塩化第一鉄・塩化第二鉄とアンモニアを含む水溶液からのアンモニア沈降反応と空気酸化により得られたコロイド状の物質を乾燥することにより得られた。マグヘマイトの104.1mgを、研和w/ガラス棒を使用して、水3 mL中の101.3mgの硫酸デキストラン(DSおよびシグマ・ケミカル社、生成物番号D5224)の溶液中で分散し、次に、先細のプローブを使用して10分間、超音波で処理された。Branson Sonifier 250を使用して、電力スケール20−22の設定で行った。反応混合物は、0.25インチx0.25インチのNeFeB磁石への接触によって磁気的に分離された。上澄みは回収され、およびペレットは、簡単な(1分)超音波処理によって水1mL中に再度分散された。分散は、5,000回/毎分の回転数で3分間、エッペンドルフ型遠心機で遠心分離され、また、上澄みは0.2マイクロメートルのポリエーテルスルホンフィルタを通された。この物質は16.8mg/mLの鉄を含んでいた。
クロファジミンを含む磁性微粒子
例10によって作られた硫酸デキストランでコーティングされたマグヘマイトの0.1mL、0.4mLの水、およびDMSOの中のクロファジミン(シグマ・ケミカル社)の5mg/mLの溶液の0.4 mLが、ガラス・バイアル中で混合された。対照として、0.4mLクロファジミン溶液が磁性体および/または高分子電解質のない状態で水の0.5mLと混合された。混合物に、例10でのように、5M NaClの0.03mLが加えられ、粒子状物質が磁石によって分離された。上澄みは廃棄された。また磁性ペレットは1mLの水中で再度分散された。再沈殿、磁気分離および再分散工程が繰り返された。また、粒子状物質は簡単な(1秒)超音波処理を使用して、1mLの水中に再度分散した。この生成物は、クロファジミンの0.58±0.02mg/mLおよび鉄の1.14±0.03mg/mLを含んでいた。薬/鉄比率はO.51+/−O.O3。
水中の硫酸デキストランでコーティングされたマグヘマイト、磁性クロファジミン微粒子およびクロファジミンの凝集安定性
例9および10により作られた生成物の粒径は、生成の1時間後、および周囲温度での20時間の貯蔵の後に準弾性光散乱法を使用して、決定された。次のサイズ・データはナノメートルでの、平均±SDであり、単一分散(ガウス)法を使用して決定された。ND−全体が沈降;測定できなかった。高分子電解質でコーティングされたマグヘマイトおよび磁性クロファジミン微粒子の両方のサイズは、観察の期間、安定していた。
アムホテリシンBを含んでいる磁性微粒子
殺菌性抗生物質アムホテリシンB(AMB;シグマ・ケミカル社(アメリカ)から得られた)は、2mg/mLで、80%(体積)のジメチルスルホキシド(DMSO)水溶液に溶かされた。HEPES−NS中に0.0566mgの鉄および0.0096mgのChSAを含むコンドロイチン硫酸A(ChSA)でコーティングされた酸化鉄磁性体コロイドのアリコートを、0.01−0.05mgのAMBを含んでいるAMB溶液のアリコートと一緒にし、0.11−0.15mLの最終容積とし、渦流で簡単に混合した。粒子状物質は、磁石への混合物の暴露(1.5分)によって分離された。また、上澄み流体は分光光度法を使用して、鉄、AMBおよびChSAについて分析された。粒子状物質中のAMBの取り込みは実際的に完了していた。鉄、AMBおよびChSAの残留量に基づいて、粒子組成が決定された(表2)。ここで磁性体(酸化第二鉄)と抗生物質(AMB)の合計は92.3%−93.6%だった。また、抗生物質の含量は粒子乾燥質量の27%−42%だった。
Claims (29)
- 微粒子を含む組成物であって、該微粒子は
(i) 磁性体;
(ii) 前記磁性体と会合されたポリマーであって、荷電を生じうるイオン化可能な官能基を有するポリマー;および
(iii)イオン化可能な有機化合物を含み、
ここで、水媒体中の前記のイオン化可能な有機化合物は、前記ポリマーの荷電の符号と反対の符号の荷電にイオン化可能であり、そして前記の磁性体と有機化合物は合計で微粒子乾燥質量の少なくとも50%を構成し、そして前記の微粒子は水に分散可能である。 - 前記の磁性体と有機化合物は合計で微粒子乾燥質量の少なくとも70%、または少なくとも90%を構成する、請求項1記載の組成物。
- 前記の有機化合物が微粒子乾燥質量の少なくとも10%を構成する、クレーム1あるいは2記載の組成物。
- 前記の有機化合物が水に貧弱に可溶性である、請求項1から3のいずれか1項記載の組成物。
- 前記の有機化合物が医薬品、染料あるいは顔料である、請求項1から4のいずれか1項記載の組成物。
- 前記の有機化合物が抗癌剤、抗菌薬、ドキソルビシン、メトトレキセート、デキスニグルジピン、クロファジミンあるいはアムホテリシンBである、請求項5記載の組成物。
- 前記の磁性体がフェライト、マグネタイトあるいはマグヘマイトである、請求項1から6のいずれか1項記載の組成物。
- 前記の有機物が医薬品で、前記微粒子がその表面に、微粒子の血液循環時間を拡張するのに有効なコーティングを有する、請求項5記載の組成物。
- 前記の微粒子がその表面に、親水性のポリマー、ポリ(アルキルエーテル)あるいはポリ(エチレングリコール)を含むコーティングを有する、請求項1から8のいずれか1項記載の組成物。
- 前記の微粒子がターゲット部位を含む、請求項1から9のいずれか1項記載の組成物。
- 前記の微粒子が、前記のコーティングに結合したターゲット部位を含む、請求項8または9記載の組成物。
- 前記の有機物が医薬品であり、微粒子は薬学的に受容可能な媒体中にある、請求項1から11のいずれか1項記載の組成物。
- 前記の微粒子が約10ナノメートルと約10マイクロメートルの間、あるいは約30ナノメートルと約3マイクロメートルの間の平均サイズを有する、請求項1から12のいずれか1項記載の組成物。
- 前記磁性体が強磁性、常磁性、または超磁性のナノ粒子の形態であり、該ナノ粒子が少なくとも1つの磁区を有する、請求項1から13のいずれか1項記載の組成物。
- 前記の磁性体の前記のナノ粒子が、約1nmから約100nmの間のサイズを有する、請求項14記載の組成物。
- 微粒子含有組成物の調製方法であって、液体媒体中に分散された磁性体であって該磁性体と会合されて該媒体中でイオン化されて電荷を有する荷電ポリマーを有する磁性体と、前記の荷電を有するポリマーの荷電と反対の符号の荷電を該媒体中で示すイオン化可能な有機化合物とを接触させ、微粒子を形成する工程を含む方法。
- 磁性体および前記の有機化合物が合計で微粒子乾燥質量の少なくとも50%を構成する、請求項16記載の方法。
- 磁性体および前記の有機化合物が合計で微粒子乾燥質量の少なくとも70%、または少なくとも90%を構成する、請求項16記載の方法。
- 前記の液体媒体が本質的に、前記の磁性体と会合していない荷電ポリマーを実質的に含まない、請求項16記載の方法。
- 前記の液体媒体が水を含む請求項16記載の方法。
- 前記の有機物が水に貧弱な可溶性ょ有する、請求項21記載の方法。
- 前記の接触が、水含有媒体中に分散された前記の磁性体と、水混和性の有機溶剤を含む媒体中の前記有機物質の溶液とを混合することを含む、請求項21記載の方法。
- 前記の有機化合物が医薬品であり、さらに前記微粒子を微粒子血液循環時間を延長するのに有効な物質でコーティングする工程を更に含む、請求項16記載の方法。
- さらに親水性のポリマーで前記微粒子をコーティングする工程をさらに含む、請求項16記載の方法。
- 前記のコーティングする工程が、微粒子と、疎水性の部分が結合された親水性のポリマーと接触させることを含む、請求項24記載の方法。
- 前記の親水性のポリマーがさらにターゲット部位を含む、請求項24記載の方法。
- 請求項12記載の組成物の有効量を患者に投与することを含む患者の治療方法。
- 医薬品としての、請求項1から15のいずれか1項記載の組成物の使用。
- 有機物が染料または顔料である、ペイント、インキあるいは着色媒体としての、請求項1から15のいずれか1項記載の組成物の使用。
