マウスのウィンドウチェンバー・モデルを用い、SCK乳腺腫瘍について、三酸化砒素(ATO)および熱による照射治療後の腫瘍におけるアポトーシス誘発レベルを評価した。腫瘍の目視のために用いたウィンドウチェンバーは、A/Jマウスの皮膚襞の上に設置した。2×105個のSCK乳腺癌細胞を、この皮膚襞の中に注入した。腫瘍を7から9日間増殖させた。試験マウスでは、20Gy、8mg/kgのATOを照射し、2時間後、腫瘍を41.5℃で60分加熱した。コントロールマウスは、照射または加熱処理をせず、ATOの代わりにプラシーボを注入した。
ATO治療前後において、FSaIIマウス線維肉腫に対する、三酸化砒素(ATO)の効力を、ウィンドウチェンバーを通じて評価するために、nu/nuマウスを用いて長軸実験を行った。
本プロジェクトの目的は、モルフィン介在性アポトーシスの検出を可能とするインビボ動物モデルを開発することであった。モルフィンを含む薬剤の乱用は、培養細胞においてアポトーシスを誘発することが示されている。アポトーシスは、様々な複雑な経路を通じて起動され、カスパーゼという名前のシステインプロテアーゼの活性化をもたらす。アポトーシスは、薬剤の有害作用、例えば、モルフィンなどの薬剤の乱用の理解のために重要である。アポトーシスの検出のために複数の技術があるが、それらの技術は、一般に、細胞を、その天然の環境から取り出すことに限定される。この機械的な処理は、アポトーシスを誘発するので、陽性結果を誇張する可能性のあることが知られる。本実験の目的は、モルフィンによって誘発されるアポトーシス検出用のインビボモデルを開発することである。
Claims (40)

- 前記カスパーゼ親和性標識剤が、静脈内、血管内、腹腔内、硝子体内、眼球内、頭蓋内、胸膜内、胸腔内、筋肉内、肺内の注入、灌流、または洗浄投与によって前記動物に導入される、請求項1〜24のいずれか1項に記載の方法。
- 前記動物が哺乳動物である、請求項1〜25のいずれか1項に記載の方法。

- 包装材料、一つ以上のカスパーゼ親和性標識剤、注入用バッファー、および、インビボでアポトーシスレベルを定量するために該カスパーゼ親和性標識剤を使用するための説明書を含むアッセイキット。
- 前記一つ以上のカスパーゼ親和性標識剤が、請求項28〜38のいずれか1項に記載の化合物を含む、請求項39に記載のキット。
